Gegenstand des Hauptpatentgesuches ist ein Verfahren zur
Herstellung von 6-Methyl-8ss-(ureidomethyl)ergolen-Deriva- ten der Formel A, worin Rl und R2 Methyl oder Rl Wasser stoff und R2 n-Butyl bedeuten.
Die vorliegende Erfindung betrifft ein Verfahren zur Her stellung neuer Harnstoffderivate des 6-Methyl-8 P-methylergo- lens der Formel I, worin R Wasserstoff oder Methyl bedeutet, und ihrer pharmakologisch verträglichen Salze mit Säuren, und umfasst auch die nach dem Verfahren hergestellten Verbin dungen der Formel I und ihre Säureadditionssalze.
Erfindungsgemäss gelangt man zu den Verbindungen der Tormel I und ihren Säureadditionssalzen, indem man auf 6 Methyl-8ss-aminomethyl-ergolen (Formel II) eine Verbindung der Formel III, worin R obige Bedeutung hat, einwirken lässt und die so erhaltenen Verbindungen der Formel I gegebenenfalls anschliessend in ihre pharmakologisch verträglichen Salze mit Säuren überführt.
Nach einer bevorzugten Ausführungsform des Verfahrens wird die Verbindung der Formel II bei Raumtemperatur in einem geeigneten organischen Lösungsmittel, wie Methanol oder Dioxan, mit einer Verbindung der Formel III versetzt.
Man verwendet auf 1 Mol einer Verbindung der Formel II ca.
2 Mol einer Verbindung der Formel III. Die Isocyansäure wird in situ durch Zugabe von Chlorwasserstoffsäure zu einer wässrigen Lösung von Kaliumcyanat gewonnen.
Das resultierende Reaktionsgemisch wird anschliessend noch 10 Minuten bis 4 Stunden am Rückfluss gekocht und nach üblicher Weise aufgearbeitet. Die Verbindungen der Formel I werden durch Chromatographie und/oder Kristallisa tion des Rohproduktes rein erhalten, gegebenenfalls in Form ihrer Salze.
Das als Ausgangsprodukt verwendete 6-Methyl-8ss-amino- methyl-ergolen kann beispielsweise nach dem folgenden Ver fahren hergestellt werden:
Lysergol (Formel IV) wird unter Zusatz eines säurebinden den Mittels, wie Pyridin, in einem inerten organischen
Lösungsmittel, vorzugsweise Acetonitril, suspendiert und mit einer Lösung von Methansulfonsäurechlorid in einem inerten organischen Lösungsmittel, vorzugsweise Acetonitril, versetzt.
Die Temperatur soll dabei 300 nicht übersteigen. Man lässt 2
Stunden bei 25o ausreagieren, kühlt zur Aufarbeitung auf 200 ab, versetzt mit Methanol, giesst anschliessend auf Was ser und stellt die resultierende Lösung mit 2N Ammoniaklö sung alkalisch ein. Durch Weiterbehandlung der organischen
Phasen nach üblicher Weise erhält man die Verbindung der
Formel V als kristallines Produkt.
Die Verbindung der Formel V wird anschliessend durch
Umsetzung mit Alkaliazid in das Alkylazid der Formel VI übergeführt. Die Reaktion wird in einem Temperaturbereich von 70 bis 100 , vorzugsweise bei 800, vorgenommen. Als
Lösungsmittel sind Gemische von Dimethylformamid oder
Hexamethylphosphorsäuretriamid mit Wasser geeignet.
Das gewünschte Ausgangsmaterial der Formel II wird erhal ten, indem man die Azidoverbindung der Formel VI in einem geeigneten Lösungsmittel reduziert. Die Reduktion kann beispielsweise mit Lithiumaluminiumhydrid oder Natriumstan nit bei -10 bis +400 erfolgen; als Lösungsmittel verwendet man je nach Methode Äther, Tetrahydrofuran, Äthanol, Di oxan, Dimethylformamid oder deren Gemische.
Die erfindungsgemäss hergestellten Verbindungen der
Formel I sind bei Raumtemperatur kristalline Verbindungen, die mit anorganischen oder organischen Säuren beständige, bei
Raumtemperatur kristallisierte Salze bilden.
Die neuen Verbindungen der Formel I zeigen antihyperten sive Wirkung, wie an der wachen hypertonen Grollmannratte sowie am wachen hypertonen Goldblatthund mit Dosen von
0,05 bis 0,5 mg pro kg Körpergewicht des Testtieres festge stellt werden konnte. Ihre Anwendung bei Hypertonien jeglicher Genese ist aufgrund dieser Wirkung angezeigt. Die zu verwendenden Dosen variieren naturgemäss je nach Art der verwendeten Substanz, der Administration und des zu behandelnden Zustandes. Im allgemeinen werden jedoch befriedigende Resultate mit einer Dosis von 0,01 bis 2,5 mg pro kg Körpergewicht des Testtieres erreicht; diese Dosis kann nötigenfalls in 2-3 Anteilen oder auch als Retardform verabreicht werden.
Die Tagesdosis für grössere Säugetiere liegt bei etwa 1 bis 200 mg. Für orale Anwendungen enthalten die Teildosen etwa 0,5 bis 100 mg einer Verbindung der Formel I neben festen oder flüssigen Trägersubstanzen oder Verdünnungsmitteln.
Die neuen Verbindungen der Formel I können als Arzneimittel allein oder in entsprechenden Arzneiformen für orale, enterale oder parenterale Verabreichung verwendet werden.
In den nachfolgenden Beispielen, welche die Erfindung näher erläutern, ihren Umfang aber in keiner Weise einschränken sollen, erfolgen alle Temperaturen in Celsiusgraden.
Soweit die Herstellung der Ausgangsverbindungen nicht beschrieben wird, sind diese bekannt oder nach an sich bekannten Verfahren oder analog zu an sich bekannten Verfahren herstellbar.

Beispiel 1 6-Methyl-8 p-(ureidomethyl)ergolen
17,3 g (68,5 mMol) 6-Methyl-8ss-aminomethyl-ergolen werden in 250 ml Methanol gelöst, mit 137 ml 1N Salzsäure, 5,64 g (69,5 mMol) Kaliumcyanat und 10 ml Wasser versetzt.
Nach 2 Stunden Rückflussieren werden nochmals 75 ml 1N Salzsäure und 5 g Kaliumcyanat sowie 80 ml Wasser zugefügt.
Die Reaktionslösung wird noch weitere 2 Stunden auf Rückflusstemperatur gehalten, anschliessend auf Wasser gegossen und mit Methylenchlorid/Isopropanol 2 1 wie üblich extrahiert. Nach Verdrängen des Lösungsmittels verbleibt ein brauner Schaum, der durch Chromatographie an der 50fachen Menge Aluminiumoxid reines Produkt ergibt. Durch Kristallisation aus Äthanol erhält man die Titelverbindung vom Smp.

Beispiel 2 6-Methyl-8ss-(3-methylureidomethyl)ergolen
13 g (51,7 mMol) 6-Methyl-8P-aminomethyl-ergolen werden in 650 ml Dioxan gelöst und mit 5,79 g (101 mMol) Methylisocyanat versetzt. Nach 10 Minuten Kochen am Rückfluss wird am Rotationsverdampfer zur Trockene eingedampft.
Man erhält einen dunkelbraunen Schaum, der in 300 ml Äthanol gelöst und mit 59,2 ml 1N Salzsäure versetzt wird. Das resultierende kristalline Produkt wird nochmals aus Äthanol kristallisiert, man erhält die Titelverbindung in Form des Hydrochlorids vom Smp. 1881900, [a]D = + 116,90 (c = 1,02 in Methanol).
Das als Ausgangsmaterial verwendete 6-Methyl-8ss-amino- methyl-ergolen kann beispielsweise wie folgt hergestellt werden: a) 6-Methyl-8 p-mesyloxymethyl-ergolen
25 g (98,5 mMol) Lysergol werden in 150 ml Pyridin und 250 ml Acetonitril suspendiert und mit einer Lösung von 22,6 ml (290 mMol) Methansulfonsäurechlorid in 50 ml Ace tonitril versetzt. Die Temperatur soll bei der Zugabe 30O nicht übersteigen. Nach 2 Stunden Rühren bei 250 wird auf - 200 gekühlt, mit 200 ml Methanol versetzt und anschliessend auf Wasser gegossen. Es wird mit 2N Ammoniaklösung alkalisch gestellt und viermal mit 1 Liter Methylenchlorid/Methanol
3 1 extrahiert.
Die organischen Phasen werden zweimal mit Wasser gewaschen, mit Natriumsulfat getrocknet und am Wasserstrahlvakuum zur Trockene eingedampft. Entfärbung mit Aktivkohle und Kristallisation aus Methylenchlorid/Äthanol 2 1 ergibt ein kristallines Produkt mit uncharakteristischer Zersetzung bei 160 bis 1750, [Q1D20 = +83,60 (c = 0,5 in Methanol/Methylenchlorid 1: 1), [aj20 = + 60,10 (c = 1 in Dimethylformamid).
b) 6-Methyl-8 P-azidomethyl-ergolen
30 g (90 mMol) 6-Methyl-8ss-mesyloxymethyl-ergolen werden bei 800 in 300 ml Hexamethyl-phosphorsäuretriamid gelöst und mit einer Lösung von 29,4 g (450 mMol) Natriumazid in 100 ml Wasser versetz. Nach dreistündigem Rühren bei 800 wird auf 250 gekühlt und unter starkem Rühren auf 4 Liter Wasser gegossen. Der gebildete Niederschlag wird abfiltriert, der Filterkuchen in 1,5 Liter Methylenchlorid gelöst. Im Scheidetrichter wird vom vorhandenen Wasser abgetrennt und die Methylenchloridlösung mit Natriumsulfat getrocknet. Mit Aktivkohle wird die Lösung entfärbt und anschliessend filtriert.
Nach dem Eindampfen auf ein kleines Volumen bei 30O am Vakuum kristallisiert watteartiges Azid aus, Smp. 1911930 (Zers.), [a]D = + 86,50 (c = 0,5 in Dimethylformamid).
c) 6-Methyl-8ss-aminomethyl-ergolen
19,8 g (71 mMol) 6-Methyl-8ss-azidomethyl-ergolen werden in 340 ml Dimethylformamid gelöst und bei 0 bis 5O portionenweise mit einer Suspension von 160 g (710 mMol) Zinnchlorid in 1065 ml 2N Natronlauge versetzt. Nach 2 Stunden Rühren bei 250 wird filtriert. Das Filtrat und der Filterkuchen werden mit Methylenchlorid/Methanol 9 1 erschöpfend extrahiert.
Die organischen Phasen werden zweimal mit Wasser gewaschen, über Natriumsulfat getrocknet und mit Aktivkohle entfärbt. Eindampfen zur Trockene ergibt einen leicht violetten Schaum, aus dem durch Kristallisation aus Essigester/ Methanol/Methylenchlorid reines Produkt vom Smp. 2322340 (Zers., subl. HV), [a]B" = + 90,5O (c = 0,503 in Methanol) erhalten wird.
