Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Imidazolderivate und deren Säureadditionssalze, die wertvolle pharmakologische Eigenschaften besitzen.
Imidazolderivate der allgemeinen Formel I,

in welcher R1 eine Alkylgruppe mit 2-6 Kohlentoffatomen oder eine Cycloalkylgruppe mit 3-6 Kohlenstoffatomen R2 eine Methoxy-, Methyl-, Hydroxy- oder Methylsulfonylgruppe und R3 Chlor, oder, wenn R2 die Hydroxyl- oder Methylsulfonylgruppe ist, auch eine Methoxy- oder Methylgruppe oder Wasserstoff, bedeutet, und deren Säureadditionssalze sind bisher nicht bekannt geworden.
Von Verbindungen des Hauptpatentes Nr. 523 257 unterscheiden sich die neuen Imidazolderivate durch die Substituenten R1 und R3 in der allgemeinen Formel I. Die Eigenschaften der neuen Imidazolderivate, insbesondere die wertvollen pharmakologischen Eigenschaften, sind ähnlich wie diejenigen der Verbindungen des Hauptpatentes Nr. 523 257.
Zur Herstellung der neuen Imidazolderivate der allgemeinen Formel I und ihrer Säureadditionssalze kondensiert man erfindungsgemäss ein substituiertes Benzil der allgemeinen Formel II,

Betreffend Ausführung des Herstellungsverfahrens der neuen Imidazolderivate und deren Säureadditionssalze und auch in bezug auf deren Anwendung gilt sinngemäss das in der Beschreibung des Hauptpatentes Gesagte.
Die Beispiele 1-4 werden analog zuBeispiel 1 des Hauptpatentes hergestellt:
Beispiel 1
2-Tert.butyl-4 (5)-(p-methoxyphenyl)-5 (4) -(o-chlorphenyl)-imidazol, Smp. 188-1909 aus 13,7 g 4-Methoxy-2'-chlorbenzil und 4,73 Pivalaldehyd.
Beispiel 2 2-Tert.butyl-4(5)-(p-methoxyphenyl)-5(4)-(m-chlor- phenyl)-imidazol, Smp. 169-171 , aus 13,7 g 4-Methoxy-3'-chlorbenzil und 4,73 g Pivalaldehyd.
Beispiel 3
2-Tert.butyl-4(5)-(p-methoxyphenyl)-5 (4)-(p-chlorphenyl)-imidazol, Smp. 148-150o, aus 13,7 g 4-Methoxy-4/-chlorbenzil und 4,73 g Pivalaldehyd.
Beispiel 4
2-Isopropyl-4(5)-(p-methoxyphenyl)-5 (4)-(m-chlorphenyl)-imidazol, Smp. 167)170 , aus 13,7 g 4-Methoxy-3/-chlorbenzil und 3,96 g Isobutyraldehyd.
Beispiel 5
Das Gemisch aus 5,8 g (0,02 Mol) 4-Methylsulfonylbenzil, 1,7 g (0,02 mol) Pivalaldehyd, 10,0 g (0,13 Mol) Ammoniumacetat und 50 ml Eisessig wird während 15 Stunden unter Rückfluss gekocht und anschliessend unter kräftigem Rühren auf 150 g Eis gegossen. Die gelbe Lösung wird mit wässriger Ammoniaklösung schwach alkalisch gestellt und mit Äthylacetat extrahiert. Die organische Phase wird abgetrennt, über Natriumsulfat getrocknet und eingedampft. Der Rückstand wird aus Toluol umkristallisiert und am Hochvakuum bei 110 getrocknet, wobei das 2-Tert.butyl-4(5)-[p- (methylsulfonyl)- phenyl]-5 (4)-phenyl-imidazol vom Smp. 215-217 erhalten wird.
Analog erhält man:
Beispiel 6
2-Isopropyl-4(5)-[p-(methylsulfonyl)-phenyl]5(4)-phenyl-imidazol, Smp. 207-208 (aus Toluol), ausgehend von 5,8 (0,02 Mol) 4-Methylsulfonylbenzil und 1,4 g (0,02 Mol) Isobutyraldehyd.
Beispiel 7
Das Gemisch aus 4,5 g (0,02 Mol) 4-Hydroxybenzil, 1,72 g (0,02 Mol) Pivalaldehyd, 10,0 g (0,13 Mol) Ammoniumacetat und 60 ml Eisessig wird während 18 Stunden unter Rückfluss gekocht und anschliessend unter kräftigem Rühren in ein Gemisch aus 150 g Eis und 120 ml konz. wässriger Ammoniaklösung gegossen. Der Kristallbrei wird mit Äthylacetat extrahiert, die organische Phase mit gesättigter Natriumchloridlösung neutralgewaschen, mit Natriumsulfat getrocknet und eingedampft. Der Rückstand wird aus Äthylacetat-Petroläther kristallisiert und am Hochvakuum bei 110 getrocknet, wobei das 2-Tert.butyl-4(5)-(p-hydroxyphenyl)- 5 (4)-phenyl-imidazol vom Smp. 190-1920 erhalten wird.
Analog erhält man:

2-Tert.butyl-4(5)-(p-methoxyphenyl)-5 (4) (p-hydroxyphenyl)-imidazol, Smp. 216-218 aus Ather/Pentan; ausgehend von 5,1 g (0,02 Mol) 4-Hydroxy-4/-methoxybenzil und 1,72 g (0,02 Mol) Pivalaldehyd.
Beispiel 9 2-Tert.butyl-4(5)-(p-hydroxyphenyl)-5 (4)-(m-tolyl)- imidazol, Smp. 227-2280 aus Toluol/Cyclohexan; ausgehend von 4,8 g (0,02 Mol) 4-Hydroxy-3'-methylbenzil und 1,72 g (0,02 Mol) Pivalaldehyd.
Beispiel 10 2-Tert.butyl-4(5)-(p-hydroxyphenyl)-5(4)-(m-chlor- phenyl)-imidazol, Smp. 238-2400, aus Toluol/Cyclohexan; ausgehend von 5,2 g (0,02 Mol) 4-Hydroxy-3'-chlorbenzil und 1,72 g (0,02 Mol) Pivalaldehyd.
Die Ausgangsstoffe für die beiden zuletzt genannten Verbindungen erhält man wie folgt:
Das Gemisch aus 9,5 g 4-Methoxy-3'-chlorbenzil, 50 ml Eisessig und 100 ml 48% Bromwasserstoff wird 20 Stunden unter Rückfluss gekocht und anschliessend auf Wasser gegossen. Die so erhaltene Suspension wird mit Äther extrahiert.
Die organische Phase wird dreimal mit 2-n. Natriumcarbonatlösung gewaschen, über Natriumsulfat getrocknet und eingedampft. Der Rückstand wird aus Äther/Petroläther umkristallisiert. Das so gereinigte 4-Hydroxy-3'-chlor-benzil schmilzt bei 1541550.
Analog erhält man, ausgehend von 14,5 g 4-Methoxy-3'methyl-benzil das 4-Hydroxy-3'-methyl-benzil, Smp. 1201210 (aus Äther/Petroläther).
Beispiel 11
Das Gemisch aus 1,0 g 4-Methylsulfonyl-4'-methoxy-benzil, 0,3 g Pivalaldehyd, 2,0 g Ammoniumacetat und 30 ml Eisessig wird während 14 Stunden unter Rückfluss gekocht und anschliessend unter kräftigem Rühren auf 100 g Eis gegossen.
Die gelbe Lösung wird mit Ammoniak schwach alkalisch gestellt und mit Äthylacetat extrahiert. Die organische Phase wird abgetrennt über Natriumsulfat getrocknet und eingedampft. Der Rückstand wird aus Alkohol/Cyclohexan umkristallisiert und am Hochvakuum bei 110o getrocknet, wobei das 2-Tert.butyl-4(5)-[p-(methylsulfonyl)-phenyl]-5(4)-(p-meth- oxyphenyl)-imidazol vom Smp. 205-2070 erhalten wird.
Die Ausgangsstoffe erhält man wie folgt: a) 4'-Methylthio-2-(p-methoxyphenyl)-acetophenon
Das Gemisch aus 5D g Thioanisol, 92,5 g 4-Methoxyphenylessigsäurechlorid und 360 ml Tetrachloräthan wird innert 30 Minuten bei 0-5o portionenweise mit 69,4 g Aluminiumchlorid versetzt. Zunächst wird das Reaktionsgemisch 7 Stunden bei 0,5O gerührt, dann 10 Stunden bei 20-25o. Die schwarze Masse wird anschliessend auf ein Gemisch aus Eis und konz. Salzsäure gegossen, gut verrührt und über Nacht stehengelassen. Die organische, untere Phase wird dann abgetrennt und am Rotationsverdampfer eingedampft. Der Rückstand wird in kochendem Alkohol gelöst und unter Rühren langsam abgekühlt. Die Lösung wird vom sich zunächst ausscheidenden dunklen Ö1 abgetrennt.
Beim weiteren Abkühlen erhält man das kristalline 4'-Methylthio-2-(p-methoxyphenyl)-acetophenon vom Smp.

b) 4'-Methylsulfonyl-2-(p-methoxyphenyl)- acetophenon
Das Gemisch aus 5 g 4'-Methylthio.2-(p-methoxyphenyl)- acetophenon, 100 ml Eisessig und 10 ml 30% Wasserstoffperoxid wird 22 Stunden bei 20-25 gerührt und anschliessend auf 700 ml Wasser gegossen. Die weissen Kristalle werden abgenutscht und aus wenig Alkohol umkristallisiert, Smp.
162-1630.
c) 4-Methylsulfonyl-4'-methoxy-benzil
Das Gemisch aus 1,5 g 4'-Methylsulfonyl-2-(p-methoxyphenyl)-acetophenon, 0,6 g Selendioxid und 30 ml Eisessig wird 26 Stunden unter Rückfluss gekocht, anschliessend heiss filtriert und auf 800 ml Wasser gegossen. Die so erhaltenen Kristalle werden abgenutscht, getrocknet und aus Alkohol/ Pentan umkristallisiert. Das 4-Methylsulfonyl-4'-methoxybenzil schmilzt bei 131-1330.

The new imidazole derivatives differ from compounds of main patent no. 523 257 by the substituents R1 and R3 in general formula I. The properties of the new imidazole derivatives, in particular the valuable pharmacological properties, are similar to those of the compounds of main patent no. 523 257.

Example 5
The mixture of 5.8 g (0.02 mol) of 4-methylsulfonylbenzil, 1.7 g (0.02 mol) of pivalaldehyde, 10.0 g (0.13 mol) of ammonium acetate and 50 ml of glacial acetic acid is refluxed for 15 hours boiled and then poured onto 150 g of ice with vigorous stirring. The yellow solution is made slightly alkaline with aqueous ammonia solution and extracted with ethyl acetate. The organic phase is separated off, dried over sodium sulfate and evaporated. Example 7
The mixture of 4.5 g (0.02 mol) of 4-hydroxybenzil, 1.72 g (0.02 mol) of pivalaldehyde, 10.0 g (0.13 mol) of ammonium acetate and 60 ml of glacial acetic acid is refluxed for 18 hours boiled and then with vigorous stirring in a mixture of 150 g of ice and 120 ml of conc. poured aqueous ammonia solution. The crystal pulp is extracted with ethyl acetate, the organic phase washed neutral with saturated sodium chloride solution, dried with sodium sulfate and evaporated. The residue is crystallized from ethyl acetate-petroleum ether and dried in a high vacuum at 110, the 2-tert.butyl-4 (5) - (p-hydroxyphenyl) -5 (4) -phenyl-imidazole of melting point 190-1920 being obtained .

2-tert-butyl-4 (5) - (p-methoxyphenyl) -5 (4) (p-hydroxyphenyl) -imidazole, m.p. 216-218 from ether / pentane; starting from 5.1 g (0.02 mol) of 4-hydroxy-4 / methoxybenzil and 1.72 g (0.02 mol) of pivalaldehyde.
Example 9 2-tert-butyl-4 (5) - (p-hydroxyphenyl) -5 (4) - (m-tolyl) - imidazole, m.p. 227-2280 from toluene / cyclohexane; starting from 4.8 g (0.02 mol) of 4-hydroxy-3'-methylbenzil and 1.72 g (0.02 mol) of pivalaldehyde.
Example 10 2-Tert-butyl-4 (5) - (p-hydroxyphenyl) -5 (4) - (m-chlorophenyl) -imidazole, m.p. 238-2400, from toluene / cyclohexane; starting from 5.2 g (0.02 mol) of 4-hydroxy-3'-chlorobenzil and 1.72 g (0.02 mol) of pivalaldehyde.
The starting materials for the last two compounds mentioned are obtained as follows:
The mixture of 9.5 g of 4-methoxy-3'-chlorobenzil, 50 ml of glacial acetic acid and 100 ml of 48% hydrogen bromide is refluxed for 20 hours and then poured into water. The suspension obtained in this way is extracted with ether.
The organic phase is three times with 2-n. Washed sodium carbonate solution, dried over sodium sulfate and evaporated. The residue is recrystallized from ether / petroleum ether. The 4-hydroxy-3'-chlorobenzil purified in this way melts at 1541,550.
Analogously, starting from 14.5 g of 4-methoxy-3'methyl-benzil, 4-hydroxy-3'-methyl-benzil is obtained, melting point 1201210 (from ether / petroleum ether).
Example 11
The mixture of 1.0 g of 4-methylsulfonyl-4'-methoxy-benzil, 0.3 g of pivalaldehyde, 2.0 g of ammonium acetate and 30 ml of glacial acetic acid is refluxed for 14 hours and then poured onto 100 g of ice with vigorous stirring .
The yellow solution is made slightly alkaline with ammonia and extracted with ethyl acetate. The organic phase is separated off, dried over sodium sulfate and evaporated. The residue is recrystallized from alcohol / cyclohexane and dried in a high vacuum at 110o, the 2-tert-butyl-4 (5) - [p- (methylsulfonyl) phenyl] -5 (4) - (p-methoxyphenyl) -imidazole of m.p. 205-2070.
The starting materials are obtained as follows: a) 4'-Methylthio-2- (p-methoxyphenyl) acetophenone
The mixture of 5D g of thioanisole, 92.5 g of 4-methoxyphenylacetic acid chloride and 360 ml of tetrachloroethane is mixed with 69.4 g of aluminum chloride in portions within 30 minutes at 0-5o. First, the reaction mixture is stirred for 7 hours at 0.5 °, then for 10 hours at 20-25 °. The black mass is then added to a mixture of ice and conc. Poured hydrochloric acid, stirred well and left to stand overnight. The lower organic phase is then separated off and evaporated on a rotary evaporator. The residue is dissolved in boiling alcohol and slowly cooled while stirring. The solution is separated from the dark oil that initially separates out.
Upon further cooling, the crystalline 4'-methylthio-2- (p-methoxyphenyl) acetophenone of mp.

b) 4'-methylsulfonyl-2- (p-methoxyphenyl) acetophenone
The mixture of 5 g of 4'-methylthio.2- (p-methoxyphenyl) acetophenone, 100 ml of glacial acetic acid and 10 ml of 30% hydrogen peroxide is stirred at 20-25 hours for 22 hours and then poured into 700 ml of water. The white crystals are suction filtered and recrystallized from a little alcohol, mp.

c) 4-methylsulfonyl-4'-methoxy-benzil
The mixture of 1.5 g of 4'-methylsulfonyl-2- (p-methoxyphenyl) acetophenone, 0.6 g of selenium dioxide and 30 ml of glacial acetic acid is refluxed for 26 hours, then filtered hot and poured onto 800 ml of water. The crystals obtained in this way are filtered off with suction, dried and recrystallized from alcohol / pentane. The 4-methylsulfonyl-4'-methoxybenzil melts at 131-1330.
