Die vorliegende Erfindung betrifft ein neues Verfahren zur Herstellung von 7-Chlor-2,3-dihydro- 1-methyl-5- phenyl-lH-1,4-benzodiazepin bzw. von 7-Chlor-2,3-dihydro 5-phenyl-lH-1,4-benzodiazepin. Diese beiden Verbindungen besitzen antikonvulsive, muskelrelaxierende und tranquilisierende Eigenschaften. Sie wirken psychisch entlastend, stimmungsausgleichend und vegetativ regulierend.
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man eine Verbindung der Formel

worin R Wasserstoff oder Methyl bedeutet, in Gegenwart von Raney-Nickel oder Raney-Cobalt katalytisch hydriert. Die erhaltene Verbindung kann erwünschtenfalls in ein für pharmazeutische Zwecke brauchbares Säureadditionssalz übergeführt werden.
Die katalytische Hydrierung einer Verbindung der Formel I kann in einem inerten Lösungsmittel durchgeführt werden.
Beispiele derartiger inerter Lösungsmittel sind Alkanole, vorzugsweise Methanol oder Äthanol, Äther, vorzugsweise Dioxan, dem gegebenenfalls Wasser zugesetzt werden kann, oder Dimethylformamid. Wird die Reaktion nicht in einem Autoklaven durchgeführt,so wird zweckmässigerweise eine Temperatur zwischen 100C und 400C eingehalten. Wird bei höheren Temperaturen, beispielsweise 100 C gearbeitet, so wird die Umsetzung in einem Autoklaven durchgeführt. Eine besonders bevorzugte Reaktionstemperatur ist Raumtemperatur.
Die Hydrierung kann sowohl bei Normaldruck als auch bei erhöhtem Druck durchgeführt werden. Wird die Hydrierung bei erhöhtem Druck durchgeführt, so muss das Reaktionsgemisch ebenfalls in einen Autoklaven verbracht werden. Besonders bevorzugt wird die Hydrierung bei Normaldruck durchgeführt.
Die Verbindungen der Formel I, die als Ausgangsmaterial im erfindungsgemässen Verfahren dienen, werden aus 2 Methylamino-5-chlor-benzophenon bzw. 2-Amino-5-chlorbenzophenon durch Umsetzung mit einem Halogenacetonitril gewonnen. Als Halogenacetonitrile gelangen Chloracetonitril, Bromacetonitril und Jodacetonitril zur Verwendung. Bei diese Reaktion dient beispielsweise das Chloracetonitril nicht nur als Reaktionspartner sondern zweckmässigerweise auch gleichzeitig als Lösungsmittel. In einer bevorzugten Ausführungsform wird bei Verwendung von Chloracetonitril dem Reaktionsgemisch ein Alkalimetallbromid, vorzugsweise Natriumbromid, oder ein Alkalimetalljodid, vorzugsweise Natrium- oder Kaliumjodid zugesetzt; in einer besonders bevorzugten Ausführungsform setzt man Kaliumjodid zu.
Es können jedoch auch anderweitige inerte Lösungsmittel wie beispielsweise Dimethylformamid zugegen sein. Zweckmässigerweise wird diese Reaktion bei erhöhter Temperatur, vorzugsweise bei der Siedetemperatur des Reaktionsgemisches, durchgeführt.
Aus der U.S. Patentschrift No. 3 243 427 sind beispielsweise Verfahren zur Herstellung von 7-Chlor-2,3-dihydro-1 methyl-5-phenyl-lH-1,4-benzodiazepin und 7-Chlor-2,3 dihydro-5-phenyl-lH-1,4-benzodiazepin bekannt. Die Durch führung dieser Synthesemethoden hat gezeigt, dass diese Ver bindungen teilweise infolge Nebenreaktionen nicht in Ausbeu ten erhalten werden, die derartige Verfahren für die Gross technik sehr attraktiv erscheinen lassen. Teilweise beanspru cfien diese Verfahren einen kostspieligen apparativen Auf wand oder umfassende Vorsichtsmassnahmen infolge Verwen dung von toxischen, korrosiven oder zur Explosion neigenden
Reagenzien oder Reaktanten.
Derartige Nachteile werden durch das erfindungsgemässe Verfahren vermieden, das die angestrebten Verbindungen ausgehend von billigen Ausgangs materialien in einfacher Weise grosstechnisch zugänglich macht.
In den nachfolgenden Beispielen sind alle Temperaturen in
Grad Celsius (oC) angegeben.
Bespiel 1
Eine Suspension von 11,4 g (2-Benzoyl-4-chlor-N-methylanilino)-acetonitril und ca. 5 g Raney-Nickel in 200 ml Alkohol wird bis zum Abklingen der Wasserstoffaufnahme hydriert.
Der Katalysator wird abgetrennt und das Filtrat im Vakuum bei 40O zur Trockene eingedampft. Der Rückstand wird in 700 ml siedendem Petroläther (40-45O) teilweise gelöst. Das unlösliche Material (1,0 g) wird abgetrennt und das Filtrat mit 1 g Aktivkohle zum Sieden erhitzt und filtriert. Die Aktivkohle wird noch mit 200 ml siedendem Petroläther (40-45O) gewaschen. Das Filtrat wird auf ca. 100 ml eingedampft. Nach Stehen über Nacht wird das auskristallisierte 7-Chlor- 1- methyl-5-phenyl-2,3-dihydro- 1H-1,4-benzodiazepin abgesaugt, mit Petroläther (40-45O) gewaschen und getrocknet.

Das Ausgangsmaterial kann nach einer der vier folgenden Methoden hergestellt werden:
A) Eine Lösung von 123 g (0,5 Mol) 2-Methylamino-5chlor-benzophenon in 320 ml Chloracetonitril wird unter Feuchtigkeitsausschluss 20 Stunden unter Rückfluss gekocht.
Die dunkle Lösung wird in 1,5 Liter Eiswasser gegossen. Die Mischung wird bis zur schwach alkalischen Reaktion mit Natriumbicarbonat versetzt und mit einem Liter Essigester extrahiert. Die Essigesterlösung wird mit 750 ml Wasser gewaschen.
Die wässrigen Lösungen werden nochmals mit einem Liter Essigester extrahiert. Die Essigesterphasen werden vereinigt, über Magnesiumsulfat getrocknet, filtriert und im Vakuum bei 40O zur Trockene eingedampft. Das zurückgebliebene Öl (150 g) wird in 150 ml Cyclohexan-Methylenchlorid (1:3) gelöst und an 2,5 kg Kieselgel (Merck) chromatographiert.
Eluieren der Säule mit Cyclohexan-Methylencldorid (1:3) liefert nach Verwerfen der ersten Fraktionen (2-Benzoyl-4chlor-N-methylanilino)-acetonitril, das aus Äther umkristalli- siert wird, Smp. 68-690.
B) Eine Mischung von 24,6 g 2-Methylamino-5-chlorbenzophenon und 83 g Kaliumjodid in 150 ml absolutem Dimethylformamid wird unter Rühren und Feuchtigkeitsausschluss bei 1000 im Laufe von 2 Stunden tropfenweise mit 37,8 g Chloracetonitril versetzt. Die Mischung wird noch 7 Stunden bei 100o gerührt und dann im Vakuum bei 7.6o zur Trockene eingedampft. Der Rückstand wird mit einer Lösung von 80 g Natriumthiosulfat in 400 ml Wasser versetzt und das ausgefallene Produkt mit 2 x 300 ml Essigester extrahiert. Die Essigesterphasen werden mit 2 x 300 ml Wasser gewaschen, vereinigt, über Magnesiumsulfat getrocknet und im Vakuum bei 40O zur Trockne eingedampft. Der Rückstand wird in 50 ml Methanol gelöst.
Nach 20 Stunden Stehen bei 4O wird das auskristallisierte (2-B enzoyl-4-chlor-N-methylanilino)-aceto- nitril abgetrennt, mit wenig Methanol gewaschen und getrocknet, Smp. 68-690. Die Mutterlauge wird im Vakuum bei 40O vom Lösungsmittel befreit. Der Rückstand wird in 50 ml Methylenchlorid-Cyclohexan (3:1) gelöst und an 500 g Kieselgel (Merck) chromatographiert. Eluieren der Säule mit Methylenchlorid-Cyclohexan (3:1) liefert noch weiteres (2-Benzoyl 4-chlor-N-rnethylanilino) -acetonitril vom Schmelzpunkt 68 690.
C) Eine Mischung von 24,6 g 2-Methylamino-5-chlorbenzonphenon und 75 g Natriumjodid in 150 ml absolutem Dimethylformamid wird unter Rühren und Feuchtigkeitsausschluss bei 100 im Laufe von 2 Stunden tropfenweise mit 37,8 g Chloracetonitril versetzt. Die Mischung wird noch 7 Stunden bei 100O gerührt und dann im Vakuum bei 70O zur Trockene eingedampft. Der Rückstand wird mit einer Lösung von 80 g Natriumthiosulfat in 400 ml Wasser versetzt und das ausgefallene Produkt mit 2 x 300 ml Essigester extrahiert.
Die Essig esterphasen werden mit 2 x 300 ml Wasser gewaschen, vereinigt, über Magnesiumsulfat getrocknet und im Vakuum bei 40O zur Trockene eingedampft.. Der Rückstand wird in 100 ml
Methylenchlorid-Cyclohexan (3:1) gelöst und an 1 kg Kieselgel (Merck) chromatographiert. Eluieren der Säule mit Methylenchlorid-Cyclohexan (3:1) liefert (2-Benzoyl-4-chlor N-rnethylanilino)-acetonitril, Smp. 68690.
D) Eine Mischung von 12,3 g 2-Methylamino-5-chlorbenzo- phenon und 33,3 g Jodacetonitril in 50 ml absolutem Dimethylformamid wird unter Feuchtigkeitsausschluss 6 Stunden bei 1000 gerührt. Das Dimethylformamid wird im Vakuum bei 70O entfernt und der Rückstand mit einer Lösung von 25 g Natriumthiosulfat in 300 ml Wasser versetzt. Das ausgefallene Produkt wird mit 2 x 300 ml Essigester extrahiert. Die Essigesterphasen werden mit 2 x 300 ml Wasser gewaschen, vereinigt, über Magnesiumsulfat getrocknet und im Vakuum bei 40O eingedampft. Der Rückstand wird in 30 ml Methanol gelöst. Nach 20 Stunden Stehen bei 4 wird das auskristallisierte (2-Benzoyl-4-chlor-N-methylanilino)-acetonitril abgetrennt, mit wenig Methanol gewaschen und getrocknet, Smp.
68690. Die Mutterlauge wird im Vakuum bei 40O zur Trokkene eingedampft. Der Rückstand wird in 30 ml Methylenchlorid-Cyclohexan (3:1) gelöst und an 200 g Kieselgel (Merck) chromatographiert. Eluieren der Säule mit Methylenchlorid-Cyclohexan (3:1) liefert noch weiteres (2-Benzoyl-4 chlor-N-methylanilino)-acetonitril vom Smp. 68-690.
Beispiel 2
Eine Suspension von 8,5 g (2-Benzoyl-4-chlor-N-methylani- lino)-acetonitril und 4 g Raney-Kobalt in 200 ml Alkohol wird bei 600 und 30 atü hydriert. Der Katalysator wird abgetrennt und das Filtrat im Vakuum zur Trockene eingedampft. Der Rückstand wird in 30 ml Essigester gelöst und an 200 g Kieselgel (Merck) chromatographiert. Eluieren der Säule mit Essigester liefert 7-Chlor- 1-rnethyl-5-phenyl-2,3-dihydro- 1H- 1,4benzodiazepin, Smp. 100-101".
Beispiel 3
Eine Suspension von 2,6 g N-(2-Benzoyl-4-chlorphenyl)- glycinonitril und ca. 1 g Raney-Nickel in 100 ml Alkohol wird bis zum Abklingen der Wasserstoff-Aufnahme hydriert. Der Katalysator wird abgetrennt und das Filtrat im Vakuum zur Trockene eingedampft. Der Rückstand wird in 20 ml Essigester-Methanol (9:1) gelöst und an 50 g Kieselgel (Merck) chromatographiert. Eluieren der Säule mit Essigester-Methanol (9:1) liefert7-Chlor-5-phenyl-1,2-dihydro-3H-1,4-benzo- diazepin, Smp. 172-1730.
Das Ausgangsmaterial kann nach einer der drei folgenden Methoden hergestellt werden:
A) Eine Lösung von 23,1 g 2-Amino-5-chlorbenzophenon in 65 ml Chloracetonitril wird unter Feuchtigkeitsausschluss 2'/2 Std. unter Rückfluss gekocht. Die dunkelrote Lösung wird in 500 ml Eiswasser gegossen. Die Mischung wird bis zur schwach alkalischen Reaktion mit Natriumbicarbonat versetzt.
Der ausfallende Niederschlag wird abgetrennt und verworfen.
Das Filtrat wird mit 2 X 250 ml Essigester extrahiert. Die Essigesterphasen werden mit 100 ml Wasser gewaschen, ver einigt, über Magnesiumsulfat getrocknet, filtriert und im Vakuum zur Trockene eingedampft. Der Rückstand wird in
100 ml Cyclohexan gelöst. Nach 30 Minuten Stehen bei Raumtemperatur wird das auskristallisierte N-(2-Benzoyl-4 chlorphenyl)-glycinonitril abgetrennt, mit Cyclohexan gewa schen und aus Methanol umkristallisert; Smp. 174-1760.
B) Eine Mischung von 23,2 g 2-Amino-5-chlorbenzophenon und 83 g Kaliumjodid in 150 ml absolutem Dimethylformamid wird unter Rühren und Feuchtigkeitsausschluss bei 1000 im Laufe von 2l/2 Stunden mit 37,8 g Chloracetonitril versetzt.
Die Mischung wird noch 6 Stunden bei 100o gerührt und anschliessend im Vakuum bei 70O eingedampft. Der Rückstand wird mit einer Lösung von 80 g Natriumthiosulfat in 400 ml
Wasser versetzt und das ausefallene Produkt mit 2 X 400 ml Essigester extrahiert. Die Essigesterphasen werden mit 2 X 300 ml Wasser gewaschen, vereinigt, über Magnesiumsulfat getrocknet und im Vakuum bei 40O zur Trockene eingedampft.
Umkristallisieren des Rückstandes aus 400 ml Essigester
Cyclohexan (1:4) liefert N-(2-Benzoyl-4-chlorphenyl-)-gly.cinonitril, Smp. 174-1760.
C) Eine Mischung von 23,2 g 2-Amino-5-chlorbenzophenon und 33,3 g Jodacetonitril in 50 ml absolutem Dimethylformamid wird unter Feuchtigkeitsausschluss 6 Stunden bei 100 gerührt. Das Dimethylformamid wird im Vakuum bei 70O entfernt und der Rückstand mit einer Lösung von 25 g Natriumthiosulfat in 300 ml Wasser versetzt. Das ausgefallene Produkt wird mit 2 x 400 ml Essigester extrahiert. Die Essigesterphasen werden mit 2 x 200 ml Wasser gewaschen, vereinigt, über Magensiumsulfat getrocknet und im Vakuum bei 40O eingedampft. Umkristallisieren des Rückstandes aus 400 ml Essigester-Cyclohexan (1:4) liefert N-(2-Benzoyl-4chlorphenyl)-glycinonitril, Smp. 174-1760.
The present invention relates to a new process for the preparation of 7-chloro-2,3-dihydro-1-methyl-5-phenyl-1H-1,4-benzodiazepine or of 7-chloro-2,3-dihydro 5-phenyl -lH-1,4-benzodiazepine. These two compounds have anticonvulsant, muscle relaxing and tranquilizing properties. They have a psychologically relieving, mood-balancing and vegetatively regulating effect.
The inventive method is characterized in that a compound of the formula

where R is hydrogen or methyl, catalytically hydrogenated in the presence of Raney nickel or Raney cobalt. The compound obtained can, if desired, be converted into an acid addition salt which can be used for pharmaceutical purposes.
The catalytic hydrogenation of a compound of the formula I can be carried out in an inert solvent.
Examples of such inert solvents are alkanols, preferably methanol or ethanol, ether, preferably dioxane, to which water can optionally be added, or dimethylformamide. If the reaction is not carried out in an autoclave, a temperature between 100 ° C. and 40 ° C. is expediently maintained. If the work is carried out at higher temperatures, for example 100 ° C., the reaction is carried out in an autoclave. A particularly preferred reaction temperature is room temperature.
The hydrogenation can be carried out either under normal pressure or under increased pressure. If the hydrogenation is carried out at elevated pressure, the reaction mixture must also be placed in an autoclave. The hydrogenation is particularly preferably carried out at normal pressure.
The compounds of the formula I which serve as starting material in the process according to the invention are obtained from 2-methylamino-5-chlorobenzophenone or 2-amino-5-chlorobenzophenone by reaction with a haloacetonitrile. As haloacetonitriles, chloroacetonitrile, bromoacetonitrile and iodoacetonitrile are used. In this reaction, for example, the chloroacetonitrile serves not only as a reactant but also conveniently as a solvent at the same time. In a preferred embodiment, when chloroacetonitrile is used, an alkali metal bromide, preferably sodium bromide, or an alkali metal iodide, preferably sodium or potassium iodide, is added to the reaction mixture; in a particularly preferred embodiment, potassium iodide is added.
However, other inert solvents such as dimethylformamide can also be present. This reaction is expediently carried out at an elevated temperature, preferably at the boiling point of the reaction mixture.
From the U.S. Patent No. For example, 3,243,427 are processes for making 7-chloro-2,3-dihydro-1 methyl-5-phenyl-1H-1,4-benzodiazepine and 7-chloro-2,3-dihydro-5-phenyl-1H-1 , 4-benzodiazepine known. The implementation of these synthesis methods has shown that some of these compounds are not obtained in Ausbeu as a result of side reactions that make such processes appear very attractive for large-scale engineering. In some cases, these processes require expensive equipment or extensive precautionary measures due to the use of toxic, corrosive or explosive substances
Reagents or reactants.
Such disadvantages are avoided by the process according to the invention, which makes the desired compounds easily accessible on an industrial scale starting from cheap starting materials.
In the examples below, all temperatures are in
Degrees Celsius (oC) indicated.
Example 1
A suspension of 11.4 g of (2-benzoyl-4-chloro-N-methylanilino) acetonitrile and about 5 g of Raney nickel in 200 ml of alcohol is hydrogenated until the uptake of hydrogen has ceased.
The catalyst is separated off and the filtrate is evaporated to dryness in vacuo at 40 °. The residue is partially dissolved in 700 ml of boiling petroleum ether (40-45O). The insoluble material (1.0 g) is separated off and the filtrate is heated to boiling with 1 g of activated charcoal and filtered. The activated charcoal is washed with 200 ml of boiling petroleum ether (40-45O). The filtrate is evaporated to approx. 100 ml. After standing overnight, the 7-chloro-1-methyl-5-phenyl-2,3-dihydro-1H-1,4-benzodiazepine which has crystallized out is filtered off with suction, washed with petroleum ether (40-45O) and dried.

The starting material can be made using one of the following four methods:
A) A solution of 123 g (0.5 mol) of 2-methylamino-5chlorobenzophenone in 320 ml of chloroacetonitrile is refluxed for 20 hours with exclusion of moisture.
The dark solution is poured into 1.5 liters of ice water. Sodium bicarbonate is added to the mixture until it is weakly alkaline and the mixture is extracted with one liter of ethyl acetate. The ethyl acetate solution is washed with 750 ml of water.
The aqueous solutions are extracted again with one liter of ethyl acetate. The ethyl acetate phases are combined, dried over magnesium sulfate, filtered and evaporated to dryness in vacuo at 40 °. The remaining oil (150 g) is dissolved in 150 ml of cyclohexane-methylene chloride (1: 3) and chromatographed on 2.5 kg of silica gel (Merck).
Eluting the column with cyclohexane-methylene chloride (1: 3) gives, after discarding the first fractions, (2-benzoyl-4-chloro-N-methylanilino) acetonitrile, which is recrystallized from ether, mp 68-690.
B) A mixture of 24.6 g of 2-methylamino-5-chlorobenzophenone and 83 g of potassium iodide in 150 ml of absolute dimethylformamide is added dropwise with 37.8 g of chloroacetonitrile over the course of 2 hours while stirring and excluding moisture at 1000. The mixture is stirred for a further 7 hours at 100o and then evaporated to dryness in vacuo at 7.6o. A solution of 80 g of sodium thiosulfate in 400 ml of water is added to the residue, and the product which has precipitated is extracted with 2 × 300 ml of ethyl acetate. The ethyl acetate phases are washed with 2 × 300 ml of water, combined, dried over magnesium sulfate and evaporated to dryness in vacuo at 40 °. The residue is dissolved in 50 ml of methanol.
After standing at 40 for 20 hours, the (2-benzoyl-4-chloro-N-methylanilino) -acetonitrile which has crystallized out is separated off, washed with a little methanol and dried, melting point 68-690. The mother liquor is freed from the solvent in vacuo at 40 °. The residue is dissolved in 50 ml of methylene chloride-cyclohexane (3: 1) and chromatographed on 500 g of silica gel (Merck). Eluting the column with methylene chloride-cyclohexane (3: 1) gives further (2-benzoyl-4-chloro-N-methylanilino) -acetonitrile with a melting point of 68,690.
C) A mixture of 24.6 g of 2-methylamino-5-chlorobenzone phenone and 75 g of sodium iodide in 150 ml of absolute dimethylformamide is added dropwise with 37.8 g of chloroacetonitrile over the course of 2 hours while stirring and excluding moisture at 100. The mixture is stirred for a further 7 hours at 100 ° and then evaporated to dryness in vacuo at 70 °. A solution of 80 g of sodium thiosulfate in 400 ml of water is added to the residue, and the product which has precipitated is extracted with 2 × 300 ml of ethyl acetate.
The ethyl acetate phases are washed with 2 × 300 ml of water, combined, dried over magnesium sulfate and evaporated to dryness in vacuo at 40O .. The residue is dissolved in 100 ml
Dissolved methylene chloride-cyclohexane (3: 1) and chromatographed on 1 kg of silica gel (Merck). Eluting the column with methylene chloride-cyclohexane (3: 1) gives (2-benzoyl-4-chloro-N-methylanilino) -acetonitrile, m.p. 68690.
D) A mixture of 12.3 g of 2-methylamino-5-chlorobenzophenone and 33.3 g of iodoacetonitrile in 50 ml of absolute dimethylformamide is stirred at 1000 for 6 hours with exclusion of moisture. The dimethylformamide is removed in vacuo at 70 ° and the residue is treated with a solution of 25 g of sodium thiosulfate in 300 ml of water. The precipitated product is extracted with 2 × 300 ml of ethyl acetate. The ethyl acetate phases are washed with 2 × 300 ml of water, combined, dried over magnesium sulphate and evaporated in vacuo at 40 °. The residue is dissolved in 30 ml of methanol. After standing at 4 for 20 hours, the (2-benzoyl-4-chloro-N-methylanilino) acetonitrile which has crystallized out is separated off, washed with a little methanol and dried, mp.
68690. The mother liquor is evaporated to dryness in vacuo at 40O. The residue is dissolved in 30 ml of methylene chloride-cyclohexane (3: 1) and chromatographed on 200 g of silica gel (Merck). Eluting the column with methylene chloride-cyclohexane (3: 1) gives still further (2-benzoyl-4-chloro-N-methylanilino) -acetonitrile with a melting point of 68-690.
Example 2
A suspension of 8.5 g of (2-benzoyl-4-chloro-N-methylanilino) acetonitrile and 4 g of Raney cobalt in 200 ml of alcohol is hydrogenated at 600 and 30 atmospheres. The catalyst is separated off and the filtrate is evaporated to dryness in vacuo. The residue is dissolved in 30 ml of ethyl acetate and chromatographed on 200 g of silica gel (Merck). Elution of the column with ethyl acetate gives 7-chloro-1-methyl-5-phenyl-2,3-dihydro-1H-1,4benzodiazepine, mp 100-101 ".
Example 3
A suspension of 2.6 g of N- (2-benzoyl-4-chlorophenyl) glycinonitrile and approx. 1 g of Raney nickel in 100 ml of alcohol is hydrogenated until the absorption of hydrogen has ceased. The catalyst is separated off and the filtrate is evaporated to dryness in vacuo. The residue is dissolved in 20 ml of ethyl acetate-methanol (9: 1) and chromatographed on 50 g of silica gel (Merck). Eluting the column with ethyl acetate-methanol (9: 1) yields 7-chloro-5-phenyl-1,2-dihydro-3H-1,4-benzodiazepine, mp 172-1730.

A) A solution of 23.1 g of 2-amino-5-chlorobenzophenone in 65 ml of chloroacetonitrile is refluxed for 2½ hours with exclusion of moisture. The dark red solution is poured into 500 ml of ice water. Sodium bicarbonate is added to the mixture until it has a weakly alkaline reaction.
The resulting precipitate is separated off and discarded.
The filtrate is extracted with 2 × 250 ml of ethyl acetate. The ethyl acetate phases are washed with 100 ml of water, combined, dried over magnesium sulfate, filtered and evaporated to dryness in vacuo. The residue is in
100 ml of cyclohexane dissolved. After standing for 30 minutes at room temperature, the N- (2-benzoyl-4-chlorophenyl) -glycinonitrile which has crystallized out is separated off, washed with cyclohexane and recrystallized from methanol; M.p. 174-1760.
B) A mixture of 23.2 g of 2-amino-5-chlorobenzophenone and 83 g of potassium iodide in 150 ml of absolute dimethylformamide is admixed with 37.8 g of chloroacetonitrile over a period of 2 1/2 hours with stirring and exclusion of moisture at 1000.
The mixture is stirred for a further 6 hours at 100 ° and then evaporated at 70 ° in vacuo. The residue is with a solution of 80 g of sodium thiosulfate in 400 ml
Water was added and the product which had precipitated was extracted with 2 × 400 ml of ethyl acetate. The ethyl acetate phases are washed with 2 × 300 ml of water, combined, dried over magnesium sulfate and evaporated to dryness in vacuo at 40 °.
Recrystallize the residue from 400 ml of ethyl acetate
Cyclohexane (1: 4) provides N- (2-benzoyl-4-chlorophenyl -) - gly.cinonitrile, m.p. 174-1760.
C) A mixture of 23.2 g of 2-amino-5-chlorobenzophenone and 33.3 g of iodoacetonitrile in 50 ml of absolute dimethylformamide is stirred at 100 for 6 hours with exclusion of moisture. The dimethylformamide is removed in vacuo at 70 ° and the residue is treated with a solution of 25 g of sodium thiosulfate in 300 ml of water. The precipitated product is extracted with 2 x 400 ml of ethyl acetate. The ethyl acetate phases are washed with 2 × 200 ml of water, combined, dried over magnesium sulphate and evaporated in vacuo at 40 °. Recrystallization of the residue from 400 ml of ethyl acetate-cyclohexane (1: 4) gives N- (2-benzoyl-4chlorophenyl) -glycinonitrile, melting point 174-1760.
