House, Modem Synthetic Reactions, W.A.
Benjamin, Inc., New York (1965)].
RMN (CCl4): 1,0 (3H, r, J=7 cps); 2,0 (3H, s); 2,1-2,6 (6H, m); 2,9 (2H, d, J= 6 cps); 5,2 (2H, m) 6 ppm.
La 4-méthyl-2-chloro-2-(pentyn-2-yl)-cyclohexane-l 3-dione, utilisée comme produit de départ dans le procédé décrit ci-dessus, peut être préparée ainsi:
I) 4-méthyl-cyclohexane-1,3-dione
SM: 192(64); 177 (100); 163 (2'5); 136(17); 122 (31).
Il I) 4-méthyl-2-chloro-2-(pentyn-2-yl) -sTclohexane-1,3-dione
Swiss Patent No. 539593 relates to a new process for the preparation of cyclic ketones of formula:
EMI1.1
wherein R1 represents either an alkyl residue comprising from 1 to 6 carbon atoms, or an alkenyl or alkynyl residue comprising from 2 to 6 carbon atoms, and R2 represents a lower alkyl residue or hydrogen, said process being characterized in that which is treated with an inorganic or organic base, or a salt of an alkali metal acting as a base under the reaction conditions, a compound of formula:
EMI1.2
in which X represents a halogen atom, and Rl and R2 have the same meaning as indicated for formula I.
In particular, the process of said patent allows the preparation of 3-methyl-2- (cis-penten-2-yl) -cyclopenten-2-one, said product being known in the art of perfumery under the name of cis -jasmone.
We have now found that it is possible to prepare cis-jasmone, with better yields than those obtained by the method indicated above, by subjecting to partial hydrogenation the acetylenic product of formula I in which R represents a methyl residue and Rl represents the pentyn-2-yl radical.
The partial hydrogenation of the triple bond of 3-methyl-2 (pentyn-2-yl) -cyclopenten-2-one can be carried out according to one of the usual methods employed in chemistry to promote the transformation of a triple bond. into the corresponding double bond [see: HO House, Modem Synthetic Reactions, WA
Benjamin, Inc., New York (1965)]. Thus, said reduction is preferably carried out by means of a catalyst of the so-called Lindlar type [Helv. Chim. Acta, 35, 446 (1952)], consisting of a palladium catalyst partially poisoned with quinoline or lead acetate.
A subject of the present invention is therefore a process for the preparation of 3-methyl-2- (cis-penten-2-yl) -cyclopenten-2-one.
characterized in that treating with an inorganic or organic base, or an alkali metal salt acting as a base under the reaction conditions, a compound of formula II in which X represents a halogen, Rl denotes a pentyn2-yl radical and R denotes a methyl radical, and that the acetylenic product obtained is subjected to partial hydrogenation.
The invention is illustrated in more detail by the example which follows. In the said example the temperatures are indicated in degrees centigrade.
Example Pre; paration of 3-tmedyl-2- (cis-petté? 2-2-yl) -cyclopenten-2-one
a) 3 g (0.015 mole) of 4-methyl-2-chloro-2- (pentyn-2-yl) cyclohexane-l, 3-dione in 15 ml of dry xylene were refluxed in the presence of sodium carbonate anhydrous (1.6 g: 0.015 mol) until the evolution of gas ceases (3 hours). The reaction mixture was washed with water, dried over anhydrous magnesium sulfate, then the solvent was evaporated under reduced pressure. The following distillation afforded 3-methyl-2- (pentyn2-yl) -cyclopenten-2-one (dehydrojasmone); Eb. 80 90 / 0.001 Torr; 850 mg (35%). Said product showed a purity of 90%; the remaining 10% consisting of 5-methyl-2 (pentyn-2-yl) -cyclopenten-2-one.
Dehydrojasmone showed the following analytical characteristics:
IR (CCl4): 2205, 1700, 1650cm-1
NMR (CCl4): 1.07 (3H, t); 1.7-2.7 (9H, m; 2.17 inclusive: 3H, s); 2.91 (2H, s) 6 ppm
MS: m / e: 162 (76); 147 (100); 153 (8.5); 119 (18); 105 (60); 91 (44).
b) 260 mg (0.016 mol) of dehydrojasmone in 10 ml of hexane were subjected to partial hydrogenation in the presence of 10 mg of catalyst of the so-called Lindlar type. One equivalent of hydrogen was absorbed after a period of 15 minutes. After filtration and evaporation of the hexane under reduced pressure, the residue obtained was distilled off and provided a mixture (9: 1) of 3methyl-2- (cis-penten-2-yl) -cyclopenten-2-one and 5 -methyl-2- (cis penten-2-yl) -cyclopenten-2-one; Eb. 60-70 "/ 0.001 Torr; 250 mg (95%).
Further purification was accomplished by gas chromatography on a Carbowax 5%, 2.5m, 175 "column.
The product obtained showed the following spectral characteristics: IR (CHCl3): 1685, 1640 cm-l
NMR (CCl4): 1.0 (3H, r, J = 7 cps); 2.0 (3H, s); 2.1-2.6 (6H, m); 2.9 (2H, d, J = 6 cps); 5.2 (2H, m) 6 ppm.
4-Methyl-2-chloro-2- (pentyn-2-yl) -cyclohexane-l 3-dione, used as a starting material in the process described above, can be prepared as follows:
I) 4-methyl-cyclohexane-1,3-dione
This product was prepared according to the method described in Zh.
Ôbschch. Khim., 32, 3983 (1962) as reported in
Chem. Abstr., 58, 13808 f. However, the cyclization was carried out in benzene and not in methanol as described.
II) 4-methyl-2- (pentyn-2-yl) -cyclohexane-1,3-dione
3.7 g (0.025 mole) of 1-bromo-2-pentyne was added to an ice-cooled solution of 4-methyl-cyclohexane-1,3-dione (3.12 g; 0.025 mole) and potassium hydroxide (1.68 g; 0.030 mole) in 6 ml of water.
The reaction mixture was kept under stirring for 15 hours at room temperature, then 12 hours at 50 ".
Said mixture was then poured into 4N aqueous sodium hydroxide solution (20 ml) and then washed with ether to remove neutral compounds. The aqueous solution was acidified with hydrochloric acid solution (20 g hydrochloric acid in 20 g of ice) and extracted with ether. After evaporation of the volatile parts, a residue was obtained which on distillation afforded 4-methyl-2- (pentyn-2-yl) -cyclohexane-1,3-dione; Eb. 120-140 / 0.001 Torr; 3 g (65%).
MS: 192 (64); 177 (100); 163 (2'5); 136 (17); 122 (31).
II) 4-methyl-2-chloro-2- (pentyn-2-yl) -sTclohexane-1,3-dione
1.66 g (0.015 mole) of tert.-butyl hypochlorite was added under nitrogen for 15 minutes to a cooled to -15 / -20 solution of 4-methyl-2- (pentyn-2-yl ) -cyclohexane-1,3dione (2.88 g; 0.015 mol) in dry chloroform (15 mL). After the addition was complete, the solvent was evaporated under reduced pressure and thus obtained 3 g of the desired chloride as a yellowish oil.
