La présente invention a pour objet un nouveau procédé de préparation des composés de formule I:

Le procédé selon l'invention consiste à faire réagir un anhydride d'acide de formule:

dans laquelle Z est un groupe triméthoxy-3,4,5 cinnamoyle ou un groupe isobutyloxycarbonyle sur un composé de formule III:

où R a la même signification que dans la formule (I).
On donne ci-après divers exemples de mise en oeuvre du procédé selon l'invention.
Exemple 1:
(triméthoxy-3',4',5') cinnamoyl-l éthoxycarbonylméthyl-4 pipérazine (de No de code: 6753), préparé à partir de l'anhydride symétrique de l'acide triméthoxy-3,4,5 cinnamoique.
On porte 5 heures à reflux un mélange de 37,9 g (0 22 mole) d'éthoxycarbonylméthyl-l pipérazine et 45.8 g (0.1 mole) d'anhydride triméthoxy-3,4,5 cinnamolque dans 400 ml de tétrahydrofuranne. Après évaporation, on reprend le résidu dans 200 ml d'eau. puis amène la solution à un pH franchement basique à l'aide de KoCO3. On extrait à l'acétate d'éthyle. Après séchage et concentration, jusqu'à début de précipitation, on glace et filtre le précipité. On obtient ainsi 28 grammes de cristaux de point de fusion 99o C, avec un rendement de 71 O/o.
Exemple 2:
(1 riméthoxy-3 ',4'.5') cinnamoyl - I pyrrolidinocarbonyl métl-yl-4 pipérazine (de No de code: 67 350), préparé à partir de l'anhydride symétrique de l'acide triméthoxy-3,4,5 cinnamoique.
On porte 5 heures à reflux un mélange de 43 g (0.22 mole) de pyrrolidinocarbonylméthyl-l pipérazine et 45,8 g (0.1 mole) d'anhydride triméthoxy cinnamo;ique dans 400 ml de tétrahydrofuranne. Après filtration, on concentre la solution et recristallise dans 100 ml d'acétate d'éthyle.
On obtient ainsi, avec un rendement de 69 oit . 29 g de cristaux de point de fusion : 135 C.
Exemple 3:
(triméthoxy-3',4',5') cinnamoyl-l propylaminocarbonyl- méthyl-4 pipérazine (de N > de code: 68 111).
Ce composé peut être préparé à partir de l'anhydride symétrique de l'acide triméthoxy-3,4,5 cinnamoïque par le procédé décrit à l'exemple 2.
On obtient ainsi, avec un rendement de 61 0/e 25 g de cristaux de point de fusion: 175( > C.
Exemple 4:
(triméthoxy-3',4',5') cinnamoyl- 1 éthoxycarbonylméthyl-4 pipérazine (de No de code: 6753).
Ce composé peut être préparé à partir de l'anhydride mixte de l'acide triméthoxy-3.4,5 cinnamoïque et du chloroformiate d'isobutyle.
A une solution de 23,8 g (0,1 mole) d'acide triméthoxy-3, 4.5 cinnamoïque dans 300 ml d'acétone anhydre et en présence de 10,1 g (0,1 mole) de triéthylamine, on ajoute à -lOoC et en 15mn 13,6g (0,1 mole) de chloroformiate d'isobutyle. Après un contact à - 10 C de 30 mn, on ajoute, toujours à cette température. 100 ml d'une solution acétonique de 34,5 g (0,2 mole) d'éthoxycarbonylméthyl-l pipérazine. On maintient en contact à 200 C pendant 9 heures. On filtre le chlorhydrate de triéthylarnine formé et concentre la solution. Les cristaux produits sont cristallisés dans l'acétate d'éthyle.
On obtient par ce procédé, avec un rendement de 71 Oio, 28 g de cristaux de point de fusion: 99o C.
Exemple 5:
(triméthoxy-3',4',5') cinnamoyl-l pyrrolidinocarbonylméthyl-4 pipérazine (de No de code: 67 350).
Ce composé est préparé à partir de l'anhydride mixte de l'acide triméthoxy-3,4,5 cinnamoique et du chloroformiate d'isobutyle.
A une solution acétonique de 23,8 g (0,1 mole) d'acide triméthoxy-3,4,5 cinnamoique et en présence de 10,1 g (0 1 mole) de triéthylamine, on ajoute à - 10O C et en 15 mn 13,6 g (0,1 mole) de chloroformiate d'isobutyle. Après un contact de 30 mn à - 10 C, on ajoute 39.4 g (0,2 mole) de pyrrolidinocarbonylméthyl- 1 pipérazine en solution dans 100 mi d'acétone.
Après un contact de 8 heures à 200 C, on filtre le chlorhydrate de triéthylamine formé, et on évapore l'acétone. On reprend le résidu par 200ml d'eau, et extrait à l'acétate d'éthyle. Après séchage, on évapore jusqu'à début de cristallisation et glace. On filtre les cristaux produits.
On obtient ainsi, avec un rendement de 55 O/o, 23 g de cristaux de point de fusion: 1350 C.
Exemple 6:
(triméthoxy-3',4',5') cinnamoyl-l isopropylaminocarbon- ylméthyl-4 pipérazine (de No de code: 68 111).
Ce composé est préparé à partir de l'anhydride mixte de l'acide triméthoxy-3,4,5 cinnamoique et du chloroformiate d'isobutyle. selon le procédé décrit à l'exemple 5.
On obtient ainsi, avec un rendement de 61 /o . 25 g de cirstaux de point de fusion : 1750 C.
Les composés préparés selon l'invention exercent principalement des propriétés vasodilatatrices au niveau de la circulation coronaire et de la circulation périphérique.
Ils ont été comparés à des composés notoirement connus par leur activité dans le même domaine pharmacologique, à savoir: -- le chlorhydrate de butyl-2 [ (diéthylamino-2" éthoxy)-4'
diiodo-3',5' benzoyle]-3 benzofuranne. dit chlorhydrate
d'amiodarone.
- la 1 1-[4,4-di(fluorophényl)butyl]-4-[(2,6-diméthylanilino-
carbonylméthyl) méthyl] pipérazine, dite lidoflazine.
le 2,2',2'',2'''-(4,8-dipipéridinopyrimidino [5,4-d] pyr
imidine-2,6-diyldinitrilo) tétraéthanol, dit dipyridamole.
- l'ester éthylique de l'acide [[3-[2-(diéthylamino)éthyl]-4-
méthyl-2-oxo-2H-1-benzopyran-7-yl]oxy]acétique, dit ch rom omar.
- la N-(3,3 -diphénylpropyl)-a-méthylphénéthylamine, dite

a) Voie intraveineuse.
- la 6,7-diméthoxy-1-veratrylisoquinoline, dite papavérine,
et - la 2-éthyl-3-benzofuranyl-4-hydroxy-3,5-diiodophényl cé
tone, dite benziodarone.
1. - Propriétés vasodilatatrices au niveau de la circulation coronaire.
Les composés No 67 350, No 68 111 et No 6753 augmentent le débit coronaire et l'oxygénation du sang veineux coronaire, ce qui traduit une réduction de l'extraction de l'oxygène par le myocarde aussi bien par voie intraveineuse que par voie intraduodénale.
Les résultats obtenus avec ces composés et avec quatre des composés de référence précités sont répertoriés dans le .a- bleau I suivant qui indique également la dose léthale 50 o par voie intraveineuse (DL 50: i.v.) desdits composés.

II ressort du tableau I que le composé NO 67 350 administré à une dose égale à 3 Olo de la dose léthale exerce une action sur le débit du sinus veineux coronaire et sur l'oxygénation qui est sensiblement équivalente à celle obtenue à la suite de l'administration d'une dose égale à 6 O/o de la dose léthale de la lidoflazine et que le composé No 6753 manifeste une activité de l'ordre de celle de ladite lidoflazine.
Il cst vrai que, à une dose égale à 1 Oio seulement de la dose léthale, le dipyridamole exerce une action supérieure à celle du composé No 67 350 et à celle du composé No 6753 sur le débit des sinus veineux et une action voisine de celle du composé No 67 350 sur l'oxygénation, mais l'on sait que le dipyridamole présente l'inconvénient d'avoir des effets secondaires gênants en provoquant des maux de tête, des vertiges et des perturbations gastro-intestinales, alors que les composés No 67 350 et No 6753 sont dépourvus de tels effets.
En ce qui concerne le composé No 68 111, il ressort du tableau I qu'aucun composé antérieurement connu ne présente une efficacité aussi grande pour une fraction aussi fai ble fn 5 O/o) de la dose léthale.
Il apparait donc que, administrés par voie intraveineuse, les c.)lnposés No 67 350 et No 68 111 sont de loin préférables aux composés de référence et ce, en particulier, du fait de leur faible toxicité.
Si l'on se réfère maintenant au tableau Il, on constate que, hormis le dipyridamole, aucun des composés de référence n'a d'effet sur la circulation coronaire par voie intraduodénale.
L'activité du dipyridamole est à peu près équivalente à celle du composé No 67 350, pour une même dose d'administration, mais la toxicité du composé No 67 350 étant nettement inférieure à celle du dipyridamole, il est clair que le composé préparé selon l'invention est d'un intérêt plus grand que le composé de référence.
Pour ce qui est du composé No 68 111, il ressort du tableau II que, administré à une dose d'administration infiniment plus faible, il a une activité sensiblement égale à celle du dipyridamole, lequel, en outre, est beaucoup plus toxique.
La encore donc l'administration des composés No 67 350 et No 68 lit procure des résultats qui ne peuvent être atteints avec les composés antérieurement connus ou qui. s'ils peuvent l'être, s'accompagnent d'une toxicité non négligeable.
Il ressort enfin du tableau III qu'aucun des composes antérieurement connus n'exerce au niveau de la circulation périphérique une activité équivalente à celle des composés
No 67 350, No 68 111 et No 6753 pour une dose d'administration correspondant à une fraction aussi faible (0,020/o ou 0,04 O/o) de la dose léthale.
L'administration des composés selon l'invention est donc, dans ce cas aussi, préférable à celle des composés antérieurement connus.
Ces composés peuvent être administrés en thérapeutique humaine à des doses comprises entre 100 et 300 mg par jour.

Example 1:
(3 ', 4', 5 'trimethoxy) cinnamoyl-1-ethoxycarbonylmethyl-4-piperazine (Code No: 6753), prepared from the symmetrical anhydride of 3,4,5-trimethoxy cinnamoic acid.
A mixture of 37.9 g (0 22 mole) of 1-ethoxycarbonylmethyl piperazine and 45.8 g (0.1 mole) of 3,4,5-trimethoxy cinnamol anhydride in 400 ml of tetrahydrofuran is brought to reflux for 5 hours. After evaporation, the residue is taken up in 200 ml of water. then bring the solution to a clearly basic pH using KoCO3. Extracted with ethyl acetate. There is thus obtained, with a yield of 69 oit. 29 g of crystals of melting point: 135 C.
Example 3:
(3 ', 4', 5 'trimethoxy) cinnamoyl-1 propylaminocarbonyl-4-methyl-piperazine (of N> code: 68 111).
This compound can be prepared from the symmetrical anhydride of 3,4,5-trimethoxy cinnamoic acid by the method described in Example 2.
In this way, with a yield of 610 / e, 25 g of crystals with a melting point of 175 (> C.
Example 4:
(3 ', 4', 5 'trimethoxy) 1-cinnamoyl-4-ethoxycarbonylmethylpiperazine (Code No: 6753).
This compound can be prepared from the mixed anhydride of trimethoxy-3,4,5 cinnamoic acid and isobutyl chloroformate.
To a solution of 23.8 g (0.1 mol) of trimethoxy-3, 4.5 cinnamoic acid in 300 ml of anhydrous acetone and in the presence of 10.1 g (0.1 mol) of triethylamine, is added to -10oC and in 15mn 13.6g (0.1 mol) of isobutyl chloroformate. After contact at -10 ° C. for 30 minutes, one adds, still at this temperature. 100 ml of an acetone solution of 34.5 g (0.2 mol) of ethoxycarbonylmethyl-1 piperazine. Contact is maintained at 200 ° C. for 9 hours. The triethylamine hydrochloride formed is filtered off and the solution concentrated. The crystals produced are crystallized from ethyl acetate.

Example 5:

To an acetone solution of 23.8 g (0.1 mole) of 3,4,5-trimethoxy cinnamoic acid and in the presence of 10.1 g (0 1 mole) of triethylamine, one adds at - 10O C and in 15 min 13.6 g (0.1 mol) of isobutyl chloroformate. After contact for 30 min at −10 ° C., 39.4 g (0.2 mole) of 1-pyrrolidinocarbonylmethyl-piperazine in solution in 100 ml of acetone are added.
After contact for 8 hours at 200 ° C., the triethylamine hydrochloride formed is filtered off, and the acetone is evaporated off. This compound is prepared from the mixed anhydride of 3,4,5-trimethoxy cinnamoic acid and isobutyl chloroformate. according to the process described in Example 5.
We thus obtain, with a yield of 61 / o. 25 g of melting point: 1750 C.
The compounds prepared according to the invention mainly exert vasodilator properties at the level of the coronary circulation and of the peripheral circulation.
They were compared with compounds well known for their activity in the same pharmacological field, namely: - butyl-2 hydrochloride [(2-diethylamino-ethoxy) -4 '
3 ', 5' diiodo benzoyl] -3 benzofuran. said hydrochloride
amiodarone.

Compounds No 67 350, No 68 111 and No 6753 increase coronary flow and oxygenation of coronary venous blood, which translates to a reduction in the extraction of oxygen by the myocardium both intravenously and intraduodenally .
The results obtained with these compounds and with four of the aforementioned reference compounds are listed in the following table I which also indicates the lethal dose 50 o by the intravenous route (LD 50: i.v.) of said compounds.

2. - Vasodilator properties at the level of the peripheral circulation.
These properties have been studied by the intraarterial route at the level of the femoral artery in anesthetized dogs.
The results obtained with compounds No 67 350, No 68 111 and No 6753 and with three of the aforementioned reference compounds are listed in the following Table III.

It emerges from Table I that the compound NO 67 350 administered at a dose equal to 3 0 0 of the lethal dose exerts an action on the flow rate of the coronary venous sinus and on the oxygenation which is substantially equivalent to that obtained following l administration of a dose equal to 6 O / o of the lethal dose of lidoflazine and that compound No. 6753 shows an activity of the order of that of said lidoflazine.
It is true that, at a dose equal to only 1 Oio of the lethal dose, dipyridamole exerts an action greater than that of compound No. 67,350 and that of compound No. 6753 on the flow of the venous sinuses and an action similar to that. of compound No. 67,350 on oxygenation, but it is known that dipyridamole has the disadvantage of having annoying side effects by causing headaches, dizziness and gastrointestinal disturbances, whereas compounds No. 67 350 and No 6753 are devoid of such effects.
With regard to compound No. 68 111, it emerges from Table I that no previously known compound exhibits such high efficacy for such a low fraction (5 O / o) of the lethal dose.
It therefore appears that, administered intravenously, c.) Lnposed Nos. 67 350 and No. 68 111 are far preferable to the reference compounds and this, in particular, because of their low toxicity.
If we now refer to Table II, we see that, apart from dipyridamole, none of the reference compounds has an effect on the coronary circulation by the intraduodenal route.
The activity of dipyridamole is roughly equivalent to that of compound No. 67,350, for the same administration dose, but the toxicity of compound No. 67,350 being markedly lower than that of dipyridamole, it is clear that the compound prepared according to the invention is of greater interest than the reference compound.
With regard to compound No. 68 111, it appears from Table II that, administered at an infinitely lower administration dose, it has an activity substantially equal to that of dipyridamole, which, moreover, is much more toxic.
Again, therefore, the administration of compounds No. 67,350 and No. 68 gives results which cannot be achieved with the previously known compounds or which. if they can be, are accompanied by a non-negligible toxicity.
Finally, it emerges from Table III that none of the previously known compounds exerts in the peripheral circulation an activity equivalent to that of the compounds
No 67 350, No 68 111 and No 6753 for an administration dose corresponding to such a small fraction (0.020 / o or 0.04 O / o) of the lethal dose.
The administration of the compounds according to the invention is therefore, in this case also, preferable to that of the previously known compounds.
These compounds can be administered in human therapy at doses of between 100 and 300 mg per day.
