Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Verbindungen der allgemeinen Formel I (siehe Formelblatt), worin R1 für Wasserstoff, R2 für den 1-Naphthyl-, den 1-Methylpropyl- oder den 2-Methylpropylrest steht.
Erfindungsgemäss gelangt man zu den neuen Verbindungen der allgemeinen Formel 1, indem man 4 -Dimethyl-epipodo phyllotoxin-P-D-glucopyranosid (Formel II) mit einer Verbindung der allgemeinen Formel III, worin Rl und R2 obige Bedeutung haben. oder einem entsprechenden niederen Acetal in Gegenwart eines geeigneten Katalysators umsetzt.
Als Katalysatoren kommen beispielsweise Lewissäuren, wie wasserfreies Zinkchlorid, Sulfonsäuren, wie z. B. p-Toluolsulfonsäuren oder getrocknete Kationenaustauscher mit Sulfonsäuregruppen in der H--Form in Frage. Als Acetale von Verbindungen der allgemeinen Formel III kommen das Dimethylacetal, das Diäthylacetal, das cyclische Äthylenacetal in Frage.
Zur Erhöhung der Ausbeute bei der Kondensation kann das entstehende Reaktionswasser bzw. der entstehende niedere Alkohol durch azeotrope Destillation bei tiefer Temperatur oder aber durch einen Katalysator, der gegebenenfalls wasserbindende Eigenschaften hat, entfernt werden.
Das erfindungsgemässe Verfahren kann ausgeführt werden, indem man 4'-Dem ethyl-epipodophyllotoxin- B-D -glucopy ranosid in eine Verbindung der allgemeinen Formel III oder das entsprechende niedere Acetal gibt und den Katalysator hinzufügt. Hierbei wird unter Feuchtigkeitsausschluss und gegebenenfalls in Stickstoffatmosphäre gearbeitet. 4 -Demethyl epipodophyllotoxin-ss-D-glucopyranosid kann aber auch in einem trockenen inerten Lösungsmittel, z. B. Nitromethan, gelöst bzw. suspendiert und durch Zugabe einer Verbindung der allgemeinen Formel III oder des entsprechenden niederen Acetals und eines geeigneten Katalysators, gegebenfalls in Stickstoffatmosphäre, zur Reaktion gebracht werden. Die Kondensation verläuft im allgemeinen bei Temperaturen von 20 bis 350C und ist nach 0,5 bis 20 Stunden beendet.
Zur Isolierung nimmt man gewöhnlich das Reaktionsgemisch, gegebenenfalls nach Abfiltrieren des Katalysators, in einem mit Wasser nicht mischbaren Lösungsmittel, z. B. Chloroform, auf und schüttelt gegebenenfalls zur Entfernung wasserlöslicher Katalysatoren und Nebenprodukte mehrmals mit Wasser aus. Die organische Phase kann anschliessend getrocknet und im Vakuum eingedampft werden. Der Rückstand kann entweder durch Digerieren bzw. Macerieren mit Pentan, Petroläther oder Hexan oder direkt durch eine Vorchormatographie von einer überschüssigen Menge einer Verbindung der allgemeinen Formel III oder dem entsprechenden niederen Acetal bzw.
Ketal befreit werden. Das rohe Kondensationsprodukt kann dann in bekannter Weise durch Nachchromatographie und/ oder Umkristallisation bzw. Umfällen gereinigt werden.
Die neuen Verbindungen der allgemeinen Formel I besitzen in vitro eine hohe cytostatische Wirkung an Mastocytomzellen und Fibroblastenkulturen. Ferner zeichnen sie sich durch eine starke Wirksamkeit gegenüber experimentellen Tumoren, insbesondere der Mäuseleukämie L-1210, dem Sarkom 37 der Maus und dem Walker-Carcinom der Ratte aus.
Die neuen Verbindungen der allgemeinen Formel I können therapeutisch verwendet werden zur Bekämpfung pathologischer Prozesse, welche mit abnormer Zellvermehrung einhergehen, z. B. maligne Neoplasmen.
Die Dosierung der neuen Verbindungen der allgemeinen Formel I variiert ungefähr zwischen 1 und 500 mg pro Tag.
In den nachfolgenden Beispielen, welche die Ausführung des Verfahrens erläutern, den Umfang der Erfindung aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden. Die Schmelz- bzw. Zersetzungspunkte sind auf dem Kofler-Block bestimmt.

Beispiel 1
4-Demethyl-1-O-[4,6-O-(1-naphthyliden)-ss- D-glucopyranosyl]-epipodophyllotoxin
1,5 g trockenes 4 -Demethyl-epipodophyllotoxin-ss-D- glucopyranosid werden mit 30 ml 1-Naphthylaldehyd versetzt und nach Zugabe von 750 mg Zinkchlorid unter N2-Atmo- sphäre und Feuchtigkeitsausschluss 21/4 Std. bei 203 gerührt.
Danach wird die braune, trübe Lösung mit 300 ml Chloroform verdünnt und 1 mal mit 250 ml Wasser gewaschen. Die wässrige Phase wird 2 mal mit je 100 ml Chloroform nachgewaschen. Die vereinigten Chloroform-Auszüge wäscht man 3 mal mit je 50 ml Wasser aus, trocknet die organische Phase über Na2SO4 und dampft im Vakuum ein. Den Rückstand, ein rotbraunes Öl, chromatographiert man an 80 g Kieselgel Merck (Korngrösse 0,05-0,2 mm). Chloroform und Chloroform-Methanol-Gemisch (98:2) eluieren 1,052 g einheitliches Material (DC Kieselgelplatten und Chloroform + 6% Methanol als Fliessmittel). Das reine Material wird in 6 ml Aceton gelöst und die Lösung unter Rühren zu 120 ml Pentan getropft.
Dabei wird reines 4 -Demethyl-1-O-[4,6-O-(1-naph- thyliden)-ss-D-glucopyranosyl]-epipodophyllotoxin als amorphes Pulver vom Smp. 189-195 erhalten; [a]D3 = - 90,40 (c = 0,869 in Chloroform).
Beispiel 2
4 -Demethyl-1-O-[4,6-O-(2-methylbutyliden)-P-
D -glucopyranosyl]-epipodophyllotoxin
Zu einer Lösung von 16 ml 2-Methylbutyraldehyd in 160 ml Nitromethan setzt man 8 g trockenes 4'-Demethyl-epipodo- phyllotoxin-ss-D-glucopyranosyd und 400 mg p-Toluolsulfonsäure zu. Die Suspension wird unter N2-Atmosphäre und Feuchtigkeitsausschluss 1'/2 Std. bei Raumtemperatur gerührt.
Die nun klar gewordene Lösung verdünnt man mit 500 ml Chloroform und wäscht dreimal mit je 50 ml Wasser neutral.
Die organische Phase wird über Natriumsulfat getrocknet und im Vakuum eingedampft. Das Rohprodukt wird an 270 g Kieselgel Merck (Korngrösse 0,05-0,2 mm) chromatographiert, wobei mit Chloroform-Methanol-Gemisch (98:2) eluiert wird. Die so erhaltenen angereicherten Fraktionen werden nochmals an derselben Menge Kieselgel gereinigt, wobei mit Chloroform-Aceton-Gemisch (4:1) eluiert wird. Die Spitzenfraktionen werden in 10 ml Aceton gelöst und unter Rühren langsam zu 175 ml Pentan getropft. Man erhält reines 4 Demethyl-1-O-[4,6-O-(2-methylbutyliden)-ss-D-glucopyrano- syl]-epipodophyllotoxin als farbloses amorphes Pulver vom Smp. 153-1590; [a]D3 = - 103,3' (c = 1,036 in Chloroform).
Beispiel 3
4 -Demethyl-l-O-[4 ,6-O-(3-methylbutylide n)-B- D-glucopyranosylj-epipodophyllotoxin 1.5 g trockenes 4 -Demethyl-epipodophyllotoxin-ss-D- glucopyranosid werden in 60 ml Nitromethan suspendiert, 1,5 ml 3-Methylbutyraldehyd und 150 mg p-Toluolsulfonsäure zugegeben und bei Raumtemperatur sowie Luft- und Feuchtigkeitsausschluss 1 Std. gerührt. Die Reaktion wird dünnschichtchromatographisch auf Kieselgelplatten mit Chloroform-Methanol-Gemisch (94:6) verfolgt. Danach wird die gelbliche Lösung mit 450 ml Chloroform verdünnt und 3 mal mit je 25 ml Wasser neutral gewaschen. Die organische Phase wird über Natriumsulfat getrocknet und eingedampft. Den Rückstand chromatographiert man an 85 g Kieselgel Merck (Korngrösse 0,02-0,5 mm).
Elution zuerst mit Chloroform, iann mit Chloroform-Methanol-Gemisch (98,5:1,5) liefert ein inheitliches Material. Kristallisation aus Aceton-Äther liefert reines 4/-Demethyl-1 -0-[4,6-0-(3-methyl-butyliden)-B- D-glucopyranosyl]-epipodophyllotoxin als farblose Kristalle vom Smp. 191-196'; [a ]D22 = - 102,10 (c = 1,045 in Chloroform).
The present invention relates to a process for the preparation of new compounds of the general formula I (see formula sheet) in which R1 is hydrogen, R2 is the 1-naphthyl, the 1-methylpropyl or the 2-methylpropyl radical.
According to the invention, the new compounds of general formula 1 are obtained by mixing 4-dimethyl-epipodo phyllotoxin-P-D-glucopyranoside (formula II) with a compound of general formula III, in which R1 and R2 have the above meaning. or a corresponding lower acetal in the presence of a suitable catalyst.
As catalysts, for example, Lewis acids, such as anhydrous zinc chloride, sulfonic acids, such as. B. p-toluenesulfonic acids or dried cation exchangers with sulfonic acid groups in the H - form in question. The acetals of compounds of the general formula III are dimethylacetal, diethylacetal and cyclic ethylene acetal.
To increase the yield in the condensation, the water of reaction formed or the lower alcohol formed can be removed by azeotropic distillation at low temperature or by a catalyst which may have water-binding properties.
The process according to the invention can be carried out by adding 4'-dem ethyl-epipodophyllotoxin-B-D -glucopyranoside to a compound of the general formula III or the corresponding lower acetal and adding the catalyst. This is done with the exclusion of moisture and, if necessary, in a nitrogen atmosphere. 4 -Demethyl epipodophyllotoxin-ss-D-glucopyranoside can also be used in a dry inert solvent, e.g. B. nitromethane, dissolved or suspended and made to react by adding a compound of the general formula III or the corresponding lower acetal and a suitable catalyst, optionally in a nitrogen atmosphere. The condensation generally takes place at temperatures from 20 ° to 350 ° C. and is complete after 0.5 to 20 hours.
The reaction mixture is usually isolated, optionally after filtering off the catalyst, in a water-immiscible solvent, e.g. B. chloroform and, if necessary, shakes out several times with water to remove water-soluble catalysts and by-products. The new compounds of general formula I have a high cytostatic effect in vitro on mastocytoma cells and fibroblast cultures. Furthermore, they are distinguished by a strong effectiveness against experimental tumors, in particular the mouse leukemia L-1210, the sarcoma 37 of the mouse and the Walker carcinoma of the rat.
The new compounds of general formula I can be used therapeutically to combat pathological processes associated with abnormal cell proliferation, e.g. B. Malignant neoplasms.
The dosage of the new compounds of general formula I varies approximately between 1 and 500 mg per day.

1.5 g of dry 4-demethyl-epipodophyllotoxin-ss-D-glucopyranoside are mixed with 30 ml of 1-naphthylaldehyde and, after addition of 750 mg of zinc chloride, stirred for 21/4 hours at 203 under an N2 atmosphere and with exclusion of moisture.

Example 2
4 -Demethyl-1-O- [4,6-O- (2-methylbutylidene) -P-
D -glucopyranosyl] epipodophyllotoxin
8 g of dry 4'-demethyl-epipodophyllotoxin-ss-D-glucopyranoside and 400 mg of p-toluenesulfonic acid are added to a solution of 16 ml of 2-methylbutyraldehyde in 160 ml of nitromethane. The suspension is stirred for 1½ hours at room temperature under an N2 atmosphere and with exclusion of moisture.
The solution, which has now become clear, is diluted with 500 ml of chloroform and washed three times with 50 ml of water each time.
The organic phase is dried over sodium sulfate and evaporated in vacuo. The crude product is chromatographed on 270 g of Merck silica gel (particle size 0.05-0.2 mm), eluting with a chloroform-methanol mixture (98: 2). The enriched fractions obtained in this way are purified again on the same amount of silica gel, eluting with a chloroform-acetone mixture (4: 1). The top fractions are dissolved in 10 ml of acetone and slowly added dropwise to 175 ml of pentane with stirring. Pure 4 demethyl-1-O- [4,6-O- (2-methylbutylidene) -ss-D-glucopyranosyl] -epipodophyllotoxin is obtained as a colorless amorphous powder with a melting point of 153-1590; [a] D3 = - 103.3 '(c = 1.036 in chloroform).

4 -Demethyl-lO- [4, 6-O- (3-methylbutylide n) -B- D-glucopyranosylj-epipodophyllotoxin 1.5 g of dry 4-demethyl-epipodophyllotoxin-ss-D- glucopyranoside are suspended in 60 ml of nitromethane, 1, 5 ml of 3-methylbutyraldehyde and 150 mg of p-toluenesulfonic acid were added and the mixture was stirred at room temperature and with the exclusion of air and moisture for 1 hour. The reaction is followed by thin layer chromatography on silica gel plates with a chloroform-methanol mixture (94: 6).
