EMI1.2
<tb> formule <SEP> II:
:
<tb> <SEP> A1 <SEP> R1 <SEP> R4 <SEP> Ai
<tb> <SEP> A2 <SEP> oxo <SEP> A2
<tb> <SEP> R2 <SEP> R6 <SEP> Il
The present invention relates to a process for preparing substituted chromone-2-carboxylic acids.
The present invention relates to a process for preparing a compound of formula I:
EMI1.1
in which Rt, R ", R3, R4, R5 and R6 are the same or different and each represents a hydrogen or halogen atom or an alkyl, hydroxyl or alkoxy group, the alkyl or alkoxy groups possibly being substituted (they represent for example a hydroxyalkoxy, alkoxyalkoxy or carboxyalkoxy group), and X is a saturated or unsaturated, substituted or unsubstituted hydrocarbon chain which can be interrupted by a carbocyclic or heterocyclic ring, or by one or more oxygen atoms or carbonyl groups, as well as their pharmaceutically acceptable salts, characterized in that a compound of
EMI1.2
<tb> formula <SEP> II:
:
<tb> <SEP> A1 <SEP> R1 <SEP> R4 <SEP> Ai
<tb> <SEP> A2 <SEP> oxo <SEP> A2
<tb> <SEP> R2 <SEP> R6 <SEP> It
<tb> where R1, R2, R3, R4, R5, RÜ and X have the above meaning, each pair of groups Al, A2 represents the pair of groups - OH and - COCH, COCOOH or the chain - O - C (COOH) = CH - CO -, or a salt thereof, provided that at least one of the pairs of groups Al, A2 represents the pair of groups - OH and - COCH2COCOOH, and, where appropriate, one converts the compound of formula I obtained into a pharmaceutically acceptable salt thereof, or vice versa.
The cyclization can be carried out by heating, preferably in the presence of an acid, eg hydrochloric acid, and in a solvent which is inert under the reaction conditions, eg ethanol. The reaction can be carried out at a temperature of about 200 C to 1500 C. It will be understood that the group - COCHBCOCOOH can also exist in the corresponding tautomeric form.
- COCH = C (OH) COOH
It is therefore also possible to use the compound of formula II in this tautomeric form. The compound of formula II can also be used in the form of one of its salts, for example one of its alkali metal salts.
Compounds of formula II can be prepared by reacting a compound of formula III:
EMI1.3
with a compound of formula IV:
EMI1.4
and a compound of formula V: A-X'-B (V) in one or more steps to form a compound of formula VI:
EMI1.5
converting the compound of formula VI to a compound of formula VII:
EMI1.6
by reacting it with diethyl oxalate in known manner, then carefully treating the compound of formula VII, or its hydrolysis product of formula I, with dilute sodium hydroxide at about room temperature.
Rs, R2, R3, R4, R5, R6 and X in formulas III, IV,
VI and VII have the meanings above, A and B in formula V are the same or different and each of these groups can react with a - OH group to form an ether bond, and X1 in formula V is such that forms a group X with the remains of A and B.
Alternatively, the compound of formula II can be prepared by treating a compound of formula VIII:
EMI2.1
in which R1, R2, R3, R4, R5, Ra and X have the above meanings, and R represents an alkyl group, with an alkali under controlled conditions, for example by the extraction of a chloroform solution of a compound of formula VIII with an aqueous alkali.
The compounds of formula I, and intermediates therefor, can be isolated from the reaction medium in which they are produced by known techniques.
Compounds of formulas III, IV and V are known or can be prepared from known compounds using known techniques.
The compounds of formula I are useful for the treatment of allergic asthma. Pharmaceutically acceptable derivatives of compounds of formula I are their pharmaceutically acceptable salts, for example sodium salts.
However, it is also possible to esterify or amidify a compound (I).
When R1 to RG are carbon-containing substituents, preferably they contain 1 to 8 carbon atoms. When R1 to R6 are halogen atoms, they can for example be chlorine or bromine atoms,
X can be for example a hydrocarbon chain which can be substituted by a chlorine or bromine atom, a hydroxyl or alkoxy group having 1 to 6 carbon atoms or which can be interrupted by a carbocyclic ring having 5 or 6 members or a heterocyclic ring containing nitrogen or oxygen.
Preferred compounds of formula I are those of formula Ia:
EMI2.2
where X has the meaning above.
It is also preferred that X is a polymethylenic chain containing from 3 to 7 carbon atoms, said chain being optionally substituted with an - OH group.
The process according to the invention has the advantage of allowing the preparation of a pure product and of giving an acid and not an ester as product. Hydrolysis of an ester of a compound of formula I to an acid of formula I can lead to the opening of the chromone ring and contamination of the product with this open-core substance.
The following examples illustrate the invention. Parts are by weight.
Example 1:
Cyclization
1,3-bis [2- (3-carboxy-1, 3-d ioxo propyl) -3-hydroxy-phenoxy] -2 -hyd roxy pro pane
A solution of 0.5 part of disodium salt of 1,3 -bis (2-carboxychromon-5-yloxy) -2-hydroxypropane in 100 parts of 01N aqueous sodium hydroxide solution at 250 C in a solution was kept. water bath for 16 hours. The solution was then acidified with dilute hydrochloric acid in the presence of ether.
After stirring the etheric layer was separated, dried over sodium sulfate and evaporated to leave 0.6 part of 1,3-bis [2- (3-carboxyl-1,3-dioxopropyl ) -3 -hydroxyphenoxy] -2-hydroxypropane. 0.06 part of this bis-1,3-dioxo-acid was dissolved in 10 parts of ethanol containing 0.2 part of concentrated hydrochloric acid. After a few minutes 0.04 part of 1,3-bis (2-carb oxychromon-5-yloxy) -2-hydroxypropane precipitated as a colorless solid which melted at 216-2170 C with decomposition.
Example 2:
Cyclization
1,3-bist4- (3-carboxy-1, 3-dioxo pro pyl) -3-hydroxy-phenoxy] -2-hydroxypropane
A solution of 0.5 part of 1,3-bis (2-carboxychromon-7-yloxy) -2-hydroxypropane disodium salt was kept in 1000 parts of 0.3N aqueous sodium hydroxide at 250 C for 90 minutes. The solution was then acidified with dilute hydrochloric acid in the presence of ether. After stirring the ethereal layer was separated, dried over sodium sulfate and evaporated to give a yellow residue of 1,3 -bis [4- (3-carboxy-1,3 -dioxopropyl) -3-hydroxyphenoxy] -2-hydroxypropane. This yellow substance was dissolved in 10 parts of ethanol containing 0.2 part of hydrochloric acid.
When this solution was heated to 500 ° C, a white precipitate of 1,3-bis (2-carboxy-chromon-7-yloxy) -2-hydroxypropane was obtained. It was identified by comparing its infrared spectrum to that of an authentic sample.
Preparation
1,3-bis [2- (3-carboxy-1, 3-dioxopropyl) -3-hydroxy-phenoxy] -2-hydrnxyprnpane
A solution of 0.0006 part of 1,3-bis [2- (3-ethoxycarbonyl-1, 3 -dioxopropyl) -3-hydroxyphenoxy] -2-hydroxypropane in 3 parts of chloroform was extracted with a solution of 0.1N sodium hydroxide. The aqueous extract had the same ultraviolet spectrum as an aqueous alkaline solution of the compound prepared as in Example 1.
