Im Zuge der Bestrebungen, natürlich vorkommende Steroid-Alkaloide durch Abbau reaktionen für die Herstellung von pharmazeutisch wirksamen Pregnanderivaten nutzbar zu machen, wurde auch versucht, das In dolizidinringsystem der Solanidanalkaloide vom Typ des Solanidins oder Demissidins abzubauen.
Ein solcher Abbau musste zwangsläufig mit einer Öffnung der Ringe E und/oder F verbunden sein. Da bekannt war, dass der Ring F, noch mehr aber der Ring E des Indolizidinringsystems in den Solanidanalkaloiden sehr beständig ist, war es ausserdem erstrèbenswert, vor allem in den Ring E funktionelle Gruppen einzubauen, die solche Abbaureaktionen begünstigen.
Aus der Arbeit von K. Schreiber und C. Horstmann Chem. Ber. 99 (1966), Seite 3183 ff. ist bekannt, dass bei dem aus Demissidin durch Dehydrierung herstellbaren A 22/N-Imoniumsalz bei Oxydation mit Wasserstoffperoxyd in alkalischer Lösung der Ring F in 22,23-Stel- lung unter Bildung der (25 S)-3P-Hydroxy-22-oxo-22.23- - sccO-Sa-solanidansäure-(23) geöffnet wird. trber die Oxydation von Verbindungen, bei denen sich die Doppelbindung nicht im Ring R, sondern in Ring E befindet, ist jedoch noch nichts bekannt geworden.
Es konnte nun überraschenderweise gefunden werden, dass in A 20,22-ungesättigten Aminosteroiden die sich von Solanidanalkaloiden ableiten, bei Oxydation mit Wasserstoffperoxyd nicht Ring E an der Doppelbindung sondern Ring F in Stellung 22,23 oxydativ geöffnet wird, wobei die Struktur einer Laktamcarbonsäure gebildet wird. Gleichzeitig wird in Ring E und zwar in Stellung 20 eine Hydroxygruppe eingeführt.
Diese dabei entstehenden 20-hydroxylierten Seco Solanidansäuren haben sich in der Folge gegenüber weiteren Bemühungen, den Ring E zu öffnen, ungewöhnlich stabil erwiesen, so dass es notwendig war, für den weiteren Abbau zunächst die Konfiguration d!es Ringes E so zu ändern, dass eine Abbaumöglichkeit unter Ringöffnung gegeben ist.
Es konnte nun ferner gefunden werden, dass eine solche Strukturänderung des Ringes E erzielt werden kann, wenn man die Laktamgruppierung im Ring E unter Bildung eines tertiären Amins reduziert, welches verschiede- nen Abbaumöglichkeiten zugänglich ist. Als Reduktionsmittel dienen dabei komplexe Hydride des Aluminiums.
Gleichzeitig wird die Säuregruppe in eine primäre Alkoholgruppe übergeführt. Es werden reine, einheitliche Produkte in nahezu quantitativer Ausbeute erhalten, was bei der doppelten Reduktion an dem komplizierten Mo lekül ausgesprochen überraschend ist. Damit ist ein weiterer wichtiger Schritt zu einem für präparative bzw. für technische Zwecke brauchbaren Abbau der Alkaloide der Solanidanreihe erreicht worden, wobei die Verfahrensprodukte eine Sauerstoffunktion in Stellung 20 besitzen, die bei vielen therapeutisch wertvollen Pregnanderivaten vorkommt.

Die erhaltenen Verbindungen der Formel I, sind in Stellung 20 aktiviert und daher für weitere Abbaureaktionen besonders interessant. Aus der Tatsache, dass bei der Oxydation neben der Ringaufspaltung auch eine Hydroxygruppe im Ring E eingeführt wird, ist klar, dass es sich nicht um eine einfache Verlagerung der Doppelbin dung vor der Oxydation handeln kann.
Als Lösungsmittel für die Oxydation werden vorzugsweise Methanol und Äthanol verwendet, als Alkalien vorzugsweise Natrium- oder Kaliumhydroxyd. Die Reak tionstemperatur beträgt vorzugsweise 0 - 25 C.
Zur Aufarbeitung des Reaktionsgemisches nach der Oxydation empfiehlt es sich, zunächst das Lösungsmittel abzudestillieren und geringe Mengen alkaliunlöslicher Anteile durch Filtration oder Extraktion zu entfernen, worauf dl e C20-Hydwxyseco-solanidansäuren der Formel III durch Ansäuern, vorzugsweise mit Mineralsäure, gefällt wird. Die nach Filtration erhaltenen Säuren können gewünschtenfalls durch Umkristallisation noch weiter gereinigt werden.
Die erfindungsgemässe Reduktion kann entweder bei Zimmertemperatur oder erhöhter Temperatur, höchstens jedoch bei Siedetemperatur des als Reaktionsmedium dienenden Lösungsmittels durchgeführt werden, wobei die Temperatur auf die Reaktion beschleunigend wirkt.
Wird als Ausgangsmaterial eine Verbindung der Formel III eingesetzt, die in 3-Stellung eine acylierte OH Gruppe trägt, so wird während der Reduktion die Acylgruppe abgespalten und die Hydroxygruppe in Freiheit gesetzt. Als Reduktionsmittel ist neben Lithiumalumimumhydnd und Natriumaluminiumhydrid vor allem das Natriumdihydro-bis-(2-methoxy-äthoxy)-alanat hervorzu- heben.
Für die Durchführung der Reaktion empfiehlt es sich, eine Lösung der zu reduzierenden Verbindung der Formel III in einem gegen die Reduktionsmittel inertem Lösungsmittel langsam einer vorgelegten Lösung des Reduktionsmittels in dem gleichen Lösungsmittel zuzusetzen. Verläuft die Reaktion anfangs zu stürmisch, so muss nötigenfalls die überschüssige Reaktionswärme durch Kühlung abgeführt werden. Als geeignete Lösungsmittel können z.B. Diäthyläther, Dioxan, Tetrahydrofuran oder Benzol genannt werden.
Die Aufarbeitung des Reaktionsgemisches kann auf an sich übliche Weise erfolgen. In der Regel wird zunächst das überschüssige Reduktionsmittel durch Zusatz von Wasser zerstört. Nach Abtrennen von eventuell dabei auftretenden Niederschlägen kann der Alkohol der Formel I einfach durch Abdampfen des Lösungsmittels zur Trockne erhalten werden. Die Produkte sind dabei so rein, dass in vielen Fällen für die präparative Weiterverarbeitung ein Umkristallisieren nicht erforderlich ist.
Will man Verbindungen der Formel I herstellen, in denen A die Gruppe =C=O darstellt, so muss man von Verbindungen der Formel III ausgehen, in denen die Ketogruppe in 3-Stellung durch Ketalisieren geschützt ist, um einen Angriff durch das Reduktionsmittel zu verhindern. Die Ketalgruppe wird dann anschliessend wieder in die Ketogruppe rückverwandelt, was beispielsweise durch Einnvirkung von verdünnten Säuren möglich ist.
Die Ketalisierung gelingt zweckmässig durch Umsetzung mit Äthylenglycol mit Vorteil in einem organischen Lösungsmittel wie Benzol in Gegenwart von Spuren von Perchlorsäure. Gleichzeitig kann dabei eine Veresterung der Carboxylgruppe eintreten vor allem dann, wenn grössere Mengen Perchlorsäure als Katalysator verwendet werden.
Die Ausgangsprodukte der Formel II sind neu und werden vorzugsweise erhalten, wenn man die der Formel II entsprechenden, aber keine Doppelbindung in 22-Stellung aufweisenden Steroide, wie Demissidin oder Solanidin, durch Behandlung mit Quecksiltbersaizen dehydriert, aus dem resultierenden Isomerengemisch das schwerer lösliche 22/N-Imo niumsalz durch fraktionierte Kristallisation aus Methanol vom leichter löslichen 16/N Imonliumsalz abtrennt,
dieses 22/N-Imoniumsalz in alkalischem Milieu in Gegenwart eines Gemisches aus Wasser und einem mit Wasser mischbaren organischen Lösungsmittel in die entsprechenden in 22,23-ungesättig- ten Aminosteroide umwandelt und letztere durch Er sitzen auf deren Schmelzpunkt in die der Formel II umlagert, die laufend absublimiert bzw. abdestilliert werden.
Die nachfolgenden Beispiele sollen das erfindungsgemässe Verfahren näher erläutern, ohne es darauf zu beschränken.
Beispiel I
In eine Mischung von 4,0 1 Methanol mit 224 ml 30%igem Wasserstoffsuperoxyd und 224 mi 30g0iger Kalilauge werden unter Rühren 37,6 g Solanid-5, 20(22) -dien-3d-ol eingetragen. Man lässt bei Zimmertemperatur 30 Stunden nachreagieren und destilliert dann im Vakuum das Methanol ab. Die so eingedampfte Lösung wird mit 1.25 1 Wasser verdünnt, noch vorhandene basische oder neutrale Produkte werden durch Ausschütteln mit Methylenchlorid entfernt.
Aus der wässerigen Phase wird nach Abdestillieren von Methylenchloridresten im Vakuum mit 50%'iger Schwefelsäure 3-Hydroxy- -androst-5-en[16ss.17ss-b]-l'-[2"(S)-methyl- 3"- carboxy]propyl-4'-(R)-methyl-4'-hydroxy-pyrrolidon-5' in Form eines feinen Niederschiages ausgefällt. Man saugt ab, wäscht mit Wasser säurefrei und trocknet.
Die Ausbeute beträgt 61,2 ,lo der Theorie.
Nach Umkristallisation aus Methanol-Wasser zeigt die Säure folgende phykalische Konstanten: Fp.: 2420C [OC]D24 64,30 (Methanol)
Eine Lösung von 1.0 g 3-Hydroxy-androst-5-en-[l 6,- 1 '-[2"(S)-methyl-3"-carboxy]-propyl-4'%)-methyls -4'-hydroxy-pyrrolidon-5', gelöst in 50 ml absolutem Tetrahydrofuran wird mit 10 ml einer 70%igen benzolischen Lösung von Natriumdihydro-bis-(2-methoxy-äthoxy)-ala- nat versetzt, und die Mischung 6 Stunden unter Rückfluss erhitzt. Anschkiesseid wird überschüssiges Reduk tionsmittel durch vorsichtige Zugabe von Wasser zersetzt, die Mischung mit MgSO4 versetzt,
filtriert und der Niederschlag mit Tetrahydrofuran gewaschen. Abdampfen des Lösungsmittels im Vakuum liefert 0,87 g 3ss-Hydroxy- -androst- 5-en-[16p,17i3 - b]-l'-f2"(S) - methyl-4"-hydroxy] butyl-4'(R)-methyl-4'-hydroxy-pyrrolidin als farblosen, feinkristallinen und dünnschichtchromatographisch fast einheitlichen Rückstand, der zweimal aus Essigester umkristallisiert Wird: Man erhält so 0,58 g, das sind 62% der Theorie.
Es sind farblose Nadeln vom Fp. 142-1440C; [α]D20 = - 39,20 Methanol)
Beispiel 2
In einer Mischung von 3,2 1 Methanol mit 180 ml
30%igem Wasserstoffsuperoxyd und 180 ml 25%iger Kalilauge werden unter Rühren 30 g 5,x-Solanid-20(22-en- -3ss-ol eingetragen. Man lässt bei Zimmertemperatur 25 Stunden nachreagieren und dampft anschliessend im Vakuum das Methanol ab. Die zurückbleibende Lösung wird mit 11 Wasser verdünnt. Noch vorhandene basische oder neutrale Anteile werden durch Extraktion mit Methylenchlorid entfernt.
Aus der wässerigen Phase fällt man nach Abdestillieren von Methylenchloridresten im Vakuum mit 50%iger Schwefelsäure die 3ss-Hydroxy-5α- -androstan-[16ss,17ss-b]-1' [2''(3)-methyl-3''-carboxy]-pro- pyl4'(R)-methyl-4'-hydroxy-pyrrolidon-5' als körnigen Niederschlag. Dieser wird nach Absaugen mit Wasser säurefrei gewaschen und getrocknet. Die Ausbeute beträgt 85%, der Theorie.
Nach Umkristallisation aus Methanol zeigt die Säure folgende physikalische Konstanten: Fp.: 2630C [X]D23 = 23,8 (Methanol)
Eine Lösung von 4,0 g 3ss-Hydroxy-5cc-androstan- -[16ss(17S-b]- 1 '-{2"(S) - methyl - 3"- carboxy]-propyl-43(R)- methyl4'-hydroxy-pyrrolidon-5', gelöst in 160 ml absolutem Tetrahydrofuran und 240 ml absolutem Benzol, wird zu einer siedenden Lösung von 85 ml einer 70%gen benzolischen Lösung von Natn.umdihydro-bis2-me- thoxyäthoxy)-alanat in 200 ml absolutem Benzol lang sam zugetropft. Man erhitzt noch 1,5 Stunden unter Rückfluss,
zersetzt überschüssiges Reduktionsmittel durch Zugabe von 30%iger Natronlauge, trennt die organische Phase ab, wäscht dieselbe mit Wasser, trocknet mit NaCl und destilliert das Lösungsmittel zuletzt im Vakuum ab.
Man erhält das 3P-Hydroxy-S.a-androstan-[l 6p,179-b]-1'- methyl4" - hydroxy] - butyl4'(R) - methyl - 4'- hydroxy-pyrrolidin als farblosen feinkristall'inen Rückstand.
Nach Umkristallisation aus Essigester erhält man 3,2 g das ist eine Ausbeute von 85% der Theorie. Es sind farblose Nadeln mit Fp: 164 - 1670C und [ab22 = - 9,90 (Methanol).
Beispiel 3
Eine Lösung von 0,8 g des nach Beispiel 1 hergestellten 3-Hydroxy-androst-5-en - [16ss3,17-b]-1' - [2"(S)-me- thyl-3 -carboxy]-propyl-4'(R)-methyl-4'-hydroxy - pyrrol- idon-5', gelöst in einer Mischung von 36 ml absolutem Tetrahydrofuran und 24 ml absolutem Benzol wird unter Rühren zu einer siedenden Lösung von 10 ml einer 105to- Eigen benzolischen Lösung von Natriumdihydro-42-me- tioxy-äthoxy)-alanat in 20 ml absolutem Benzol langsam getropft.
Nach 2stündigem Erhitzen unter Rückfluss wird wie bei Beispiel 2 beschrieben aufgeaSitet, wobei man 0,67 g Rohprodukt erhält, das beim Umkristallisie- ren aus Essigester 0,55 g, das sind 73% der Theorie 3,B- -Hydroxy-androst-en-[16ss,17b]-1'- [2"(S) -methyl4" -hydroxy]-butyl-4'(R)-methyl-4'-hydroxy-pyrrolidn vom Schmelzpunkt 145 - 1460C; [α]D20 = - 40,00 (Methanol), liefert.
In the course of efforts to make naturally occurring steroid alkaloids by degradation reactions for the production of pharmaceutically effective pregnane derivatives useful, attempts were also made to break down the In dolizidine ring system of the solanidan alkaloids of the solanidine or demissidine type.
Such a breakdown had to be connected with an opening of the rings E and / or F. Since it was known that the ring F, but even more so the ring E of the indolizidine ring system in the solanidan alkaloids is very stable, it was also worthwhile to incorporate functional groups into the ring E in particular, which favor such degradation reactions.

It has now surprisingly been found that in A 20,22-unsaturated aminosteroids which are derived from solanidan alkaloids, when oxidized with hydrogen peroxide, ring F is not opened oxidatively at the double bond but ring F in position 22.23, forming the structure of a lactamcarboxylic acid becomes. At the same time, a hydroxyl group is introduced into ring E in position 20.
The resulting 20-hydroxylated seco solanidanic acids have subsequently proven to be unusually stable compared to further efforts to open the ring E, so that it was first necessary to change the configuration of the ring E so that for further degradation there is a possibility of dismantling by opening the ring.
It has now also been found that such a change in the structure of ring E can be achieved if the lactam grouping in ring E is reduced to form a tertiary amine which is accessible to various degradation options. Complex hydrides of aluminum are used as reducing agents.
At the same time, the acid group is converted into a primary alcohol group. Pure, uniform products are obtained in almost quantitative yield, which is extremely surprising given the double reduction on the complicated molecule. A further important step towards a degradation of the alkaloids of the solanidan series, which can be used for preparative or technical purposes, has been achieved, the process products having an oxygen function in position 20, which occurs in many therapeutically valuable pregnane derivatives.

To work up the reaction mixture after the oxidation, it is advisable to first distill off the solvent and remove small amounts of alkali-insoluble components by filtration or extraction, whereupon the C20-Hydroxyseco-solanidanoic acids of the formula III are precipitated by acidification, preferably with mineral acid. The acids obtained after filtration can, if desired, be further purified by recrystallization.
The reduction according to the invention can be carried out either at room temperature or at an elevated temperature, but at most at the boiling point of the solvent used as the reaction medium, the temperature having an accelerating effect on the reaction.
If the starting material used is a compound of the formula III which has an acylated OH group in the 3-position, the acyl group is split off during the reduction and the hydroxyl group is set free. In addition to lithium aluminum hydride and sodium aluminum hydride, sodium dihydro-bis- (2-methoxy-ethoxy) -alanate should be emphasized as a reducing agent.
To carry out the reaction, it is advisable to slowly add a solution of the compound of the formula III to be reduced in a solvent inert to the reducing agent to a solution of the reducing agent in the same solvent. If the reaction is too stormy at first, the excess heat of reaction must, if necessary, be dissipated by cooling. Suitable solvents e.g. Diethyl ether, dioxane, tetrahydrofuran or benzene can be mentioned.
The reaction mixture can be worked up in a conventional manner. As a rule, the excess reducing agent is first destroyed by adding water. After any precipitates that may occur have been separated off, the alcohol of the formula I can be obtained simply by evaporating the solvent to dryness. The products are so pure that in many cases recrystallization is not necessary for preparative further processing.
If you want to prepare compounds of the formula I in which A represents the group = C = O, one must start from compounds of the formula III in which the keto group in the 3-position is protected by ketalization in order to prevent attack by the reducing agent . The ketal group is then converted back into the keto group again, which is possible, for example, by the action of dilute acids.
The ketalization is conveniently achieved by reaction with ethylene glycol, advantageously in an organic solvent such as benzene in the presence of traces of perchloric acid. At the same time, esterification of the carboxyl group can occur, especially when larger amounts of perchloric acid are used as a catalyst.
The starting products of the formula II are new and are preferably obtained if the steroids corresponding to the formula II but not having a double bond in the 22-position, such as demissidine or solanidine, are dehydrated by treatment with mercury salts, the less soluble 22 / N-Imonium salt is separated from the more soluble 16 / N Imonlium salt by fractional crystallization from methanol,
this 22 / N-imonium salt is converted in an alkaline medium in the presence of a mixture of water and a water-miscible organic solvent into the corresponding aminosteroids in 22,23-unsaturated and the latter is rearranged into that of the formula II by sitting at their melting point, which are continuously sublimated or distilled off.
The following examples are intended to explain the process according to the invention in more detail without restricting it thereto.
Example I.
37.6 g of solanid-5, 20 (22) -dien-3d-ol are introduced into a mixture of 4.0 l of methanol with 224 ml of 30% strength hydrogen peroxide and 224 ml of 30% strength potassium hydroxide solution, with stirring. The reaction is allowed to continue for 30 hours at room temperature and the methanol is then distilled off in vacuo. The solution evaporated in this way is diluted with 1.25 liters of water, any basic or neutral products still present are removed by shaking with methylene chloride.
After distilling off residual methylene chloride in vacuo with 50% sulfuric acid, 3-hydroxy- androst-5-ene [16ss.17ss-b] -l '- [2 "(S) -methyl- 3" - carboxy] propyl-4 '- (R) -methyl-4'-hydroxy-pyrrolidone-5' precipitated in the form of a fine precipitate. It is filtered off with suction, washed acid-free with water and dried.
The yield is 61.2 lo of theory.
After recrystallization from methanol-water, the acid shows the following physical constants: Mp .: 2420C [OC] D24 64.30 (methanol)

Evaporation of the solvent in vacuo gives 0.87 g of 3ss-hydroxy- androst- 5-en- [16p, 17i3 - b] -l'-f2 "(S) - methyl-4" -hydroxy] butyl-4 '( R) -methyl-4'-hydroxypyrrolidine as a colorless, finely crystalline and almost uniform residue according to thin-layer chromatography, which is recrystallized twice from ethyl acetate: 0.58 g is obtained, that is 62% of theory.
They are colorless needles with a melting point of 142-1440C; [α] D20 = - 39.20 methanol)
Example 2
In a mixture of 3.2 l of methanol with 180 ml
30% hydrogen peroxide and 180 ml of 25% potassium hydroxide solution are introduced with stirring, 30 g of 5, x-solanide-20 (22-en--3ss-ol. The reaction is allowed to continue for 25 hours at room temperature and the methanol is then evaporated off in vacuo. The remaining solution is diluted with 11% water and any basic or neutral components still present are removed by extraction with methylene chloride.
After distilling off residual methylene chloride in vacuo with 50% sulfuric acid, the 3ss-hydroxy-5α-androstan- [16ss, 17ss-b] -1 '[2' '(3) -methyl-3' is precipitated from the aqueous phase. '-carboxy] -propyl4' (R) -methyl-4'-hydroxy-pyrrolidone-5 'as a granular precipitate. After suctioning off, this is washed acid-free with water and dried. The yield is 85% of theory.
After recrystallization from methanol, the acid shows the following physical constants: Mp .: 2630C [X] D23 = 23.8 (methanol)
A solution of 4.0 g of 3ss-hydroxy-5cc-androstane - [16ss (17S-b] - 1 '- {2 "(S) - methyl - 3" - carboxy] -propyl-43 (R) - methyl4 '-hydroxy-pyrrolidone-5', dissolved in 160 ml of absolute tetrahydrofuran and 240 ml of absolute benzene, is converted into a boiling solution of 85 ml of a 70% benzene solution of sodium (umdihydro-bis2-methoxyethoxy) alanate in 200 ml of absolute benzene was slowly added dropwise. The mixture is refluxed for a further 1.5 hours,
decomposes excess reducing agent by adding 30% sodium hydroxide solution, separating the organic phase, washing it with water, drying with NaCl and finally distilling off the solvent in vacuo.
The 3P-hydroxy-S.a-androstan- [l 6p, 179-b] -1'-methyl4 "- hydroxy] - butyl4 '(R) - methyl - 4'-hydroxypyrrolidine is obtained as a colorless, finely crystalline residue.
After recrystallization from ethyl acetate, 3.2 g are obtained, which is a yield of 85% of theory. There are colorless needles with mp: 164-1670C and [ab22 = - 9.90 (methanol).
Example 3
A solution of 0.8 g of the 3-hydroxy-androst-5-ene - [16ss3,17-b] -1 '- [2 "(S) -methyl-3-carboxy] -propyl prepared according to Example 1 -4 '(R) -methyl-4'-hydroxy-pyrrolidone-5', dissolved in a mixture of 36 ml of absolute tetrahydrofuran and 24 ml of absolute benzene, is stirred into a boiling solution of 10 ml of a 105to-own benzene Solution of sodium dihydro-42-metioxy-ethoxy) alanate in 20 ml of absolute benzene was slowly added dropwise.
