Gegenstand der vorliegenden Erfindung ist ein neuer Stamm von Mutterkorn Claviceps purpurea Tul., der Ergometrin produziert.
Wie bekannt, können die Stämme Claviceps purpurea Tul. in Abhängigkeit von ihren Erbeigenschaften Alkaloide wie Ergotamin, Ergometrin, Ergocryptin, Ergokristin usw. synthetisieren. Das Ergometrin findet breite Verwendung in der Geburtshilfe und Gynäkologie als hämostatisches Mittel. Es wird aus Hörnern von Mutterkorn erhalten, in denen es zusammen mit anderen Alkaloiden vorliegt. Unter Hörnern des Mutterkornes werden die hornförmigen Dauermyzele des Mutterkornpilzes verstanden, die sich nach der Infektion des Getreides aus dem Fruchtknoten bilden (siehe zum Beispiel Brockhaus ABC Biologie, Verlag Leipzig, 1967, Seite 568).
Ein Nachteil bisher bekannter Stämme von Mutterkorn ist, dass das Ergometrin in diesen in einem Gemisch mit anderen Alkaloiden enthalten ist, wobei der Gehalt der Mutterkornhörner an Ergometrin im allgemeinen nur 5 bis 20% der Summe aus allen darin enthaltenen Alkaloiden beträgt, was 0,01 bis 0,07%, bezogen auf das Trockengewicht des Rohstoffes entspricht.
Ziel der vorliegenden Erfindung war es daher, einen neuen Stamm von Mutterkorn Claviceps purpurea Tul.
zu entwickeln, der es ermöglicht, einen an Ergometrin angereicherten Rohstoff zu erzeugen, in dem Begleitalkaloide praktisch fehlen, wodurch der Rohstoff wesentlich leichter aufgearbeitet werden kann.
Gegenstand der vorliegenden Erfindung ist daher ein Stamm von Mutterkorn Claviceps purpurea Tul., der Ergometrin produziert und durch die folgenden Eigenschaften gekennzeichnet ist: auf organischem Würzeagar Nährboden bildet er wattenartiges lockeres Luft-Myzel weisser oder gelblichgrauer Farbe mit grosser Anzahl von Sporen, ein Myzel in Form. eine dichten glatten, oder welligen oder faltigen Filmes, färbt den Nährboden nicht; auf Waksman-Nährboden ein lockeres Luft-Myzel weisser oder grauer Farbe, es bildet eine nicht grosse Zahl von Sporen, der Myzelfilm ist weniger dicht als auf dem Würzeagar, lässt sich schwer vom Nährboden trennen, färbt den Nährboden nicht; auf mineralischem Winogradskiy-Nährboden und auf angefeuchtetem Zellgewebe ist kein Wachstum;
die Pilzsporen auf dem Würzeagar Nährboden von Waksam sind einzellig, oval, durchsichtig, ohne merkliche Struktur sie variieren in ihrer Grösse: Länge 4 bis 10 p Breite 2,5 bis 5i, der Stamm produziert das Alkaloid Ergometrin in einer Menge von 90 bis 100% von der Summe der Ergotalkaloide, was 0,15% des Trockengewichtes der Mutterkornhörner ausmacht.
Der oben genannte Winogradskiy-Nährboden hat die folgende Zusammensetzung: Bestandteil Gewichtsteile (NH}2SO4 0,2 KH2PO4 0.1 MgSO4 0,05 NaCl 0,2 FeSO4 0,04 MgCO3 Spur Wasser 100 Agar-Agar 1,5
Die Zusammensetzung des oben genannten Waksmann-Nährbodens war wie folgt: Bestandteil Gewichtsteile Glukose 1,0 Pepton 0,5 KH5PO4 0,1 MgSO4X7 H20 0,05 Wasser 100,0 Agar-Agar 1,5
Der oben erwähnte Würzeagar hatte die folgende Zusammensetzung: nicht gehopfte Bierwürze, mit Wasser verdünnt auf 70 Be (d.h. bis ein spezifisches Gewicht des verdünnten Materials von 1,05 erreicht ist). Von diesem Material wurden 100 Gew.Teile mit 1,5 Gew.Teilen Agar-Agar vermischt.
Der erfindungsgemässe neue Stamm von Mutterkorn ist in der Lage, das Alkohol Ergometrin in einer Menge von 90 bis 100%, bezogen auf die Summe der Ergoalkaloide zu produzieren, wobei dieses Alkaloid im all gemeinen 0,15% des Trockengewichtes der Mutterkornhörner ausmacht.
Bei Verwendung des erfindungsgemässen Stammes von Claviceps purpurea Tul. steigt die Ausbeute an Ergometrin, die produziert wird, bezogen auf die Ausbeute, die mit bisher bekannten Stämmen von Mutterkorn erreicht wurde, auf das Drei- bis Vierfache. Dadurch wird die Herstellungsweise des Alkaloides Ergometrin wesent lich vereinfacht. Der erfindungsgemässe Stamm aus Claviceps purpurea Tul. wurde aus der Natur-Mutterkornrasse durch Zuchtwahl erhalten.
Zum Züchten von Sporen des Stammes Claviceps purpureo Tul., der das Ergometrin produziert, wird ein Würzeagar-Nährboden der folgenden Zusammensetzung verwendet: nicht gehopfte Bierwürze mit einem spezifischen Gewicht von 1,05, Agar-Agar 2%.
Die Züchtung beträgt 21 bis 30 Tage bei einer Temperatur von 25 bis 270C.
Mit den erhaltenen Sporen wurde Roggen im Blütestadium, nämlich in dem Stadium des Ährenschiebens aus der Blattröhre durch künstliche Injektion der währen infiziert. Am 15. bis 20. Tag nach der Injektion sind auf dem Roggen die ersten Mutterkörner bereits sichtbar, deren Vollreife nach zwei Monaten erreicht war. Der grösste Gehalt an Ergometrin wurde zum Zeitpunkt der Voll reife des Mutterkornes festgestellt. Die Erfindung sei nun anhand eines Beispiels näher erläutert.
Beispiel
Eine Hornhälfte von Mutterkorn Claviceps purpurea Tul., die 0,09% Ergometrin enthält, unterzieht man einer Oberflächen sterilisation, schneidet in Stückchen und inoculiert auf Würzeagar-Nährboden. Nach 30 Tagen Wachstums wird die Kultur auf Würzeagar-Nährboden in die Matrizenkolben übergeimpft. Nach 30 Tagen Wachstums verwendet man die erhaltene Kultur zur Injektion von Roggen unter feldmässigen Bedingungen. Aus den ausgewachsenen Hörnern von Mutterkorn werden die besten nach den chemischen und biologischen Merkmalen ausgelesen. Man führt die Auslese in mehreren Stufen durch.
Im Ergebnis wurde nach diesem Verfahren ein Stamm von Mutterkorn Claviceps purpurea Tul. erhalten, der einen Ernteertrag an Hörnern von 120- 200 kg umgerechnet auf 1 Hektar mit einem Gehalt an Ergometrin von 0,15% vom Gewicht eines Mutterkornhornes (90100% von der Summe der Alkoloide) bringt
Die Kultur des Stammes von Mutterkorn Claviceps purpurea Tul. bleibt auf einem Würzeagar-Nährboden bei einer Temperatur von +1, +50C bis zu 6 Monaten in Dunkelheit und in Hörnern im lufttrocknen Zustand in Dunkelheit viel längere Zeit erhalten. Die Gewinnung von Ergometrin aus den Hörnern des Stammes von Mutterkorn Claviceps purpurea Tul., der das Ergometrin produziert, erfolgt nach den bekannten Verfahren.

When using the inventive strain of Claviceps purpurea Tul. the yield of ergometrine that is produced, based on the yield that was achieved with previously known strains of ergot, increases three to four times. This simplifies the production method of the alkaloid Ergometrin wesent Lich. The strain according to the invention from Claviceps purpurea Tul. was obtained from the natural ergot breed through selection.
