Die Erfindung betrifft ein Verfahren zur Herstellung neuer N-Phenäthylpiperidinderivate der Formel I, worin R1 für Wasserstoff, Halogen, eine niedere Alkyl-, Alkylthio- oder Alkoxygruppe, die Trifluormethyl-, Nitro-, Amino-, Cyan- oder Acylaminogruppe, R2 für eine 1 Naphthyl-, 1,2,3,4-Tetrahydro-5-naphthyl- oder 5-Indanylgruppe, die gegebenenfalls im aromatischen Ring durch Halogen, eine niedere Alkyl-, Alkylthio- oder Alkoxygruppe, die Trifluormethyl-, Nitro-, Amino-, Cyan- oder Acetaminogruppe substituiert sein können, und Rs für Wasserstoff oder eine niedere Acylgruppe stehen, wobei jedoch R2 nicht durch eine Aminogruppe substituiert sein darf, wenn R3 für eine niedere Acylgruppe steht, und ihrer Säureadditionssalze.
Erfindungsgemäss gelangt man zu den neuen Verbindungen der Formel I und ihren Säureadditionssalzen, indem man eine Verbindung der Formel II, worin R2 und Ra obige Bedeutung besitzen, in einem unter den Reaktionsbedingungen inerten Lösungsmittel und in Gegenwart eines säurebindenden Mittels mit Verbindungen der Formel III, worin R1 obige Bedeutung besitzt und X für einen sich unter den Reaktionsbedingungen unter Bildung von XH abspaltenden Rest steht, kondensiert und die erhaltenen Verbindungen der Formel I gewünschtenfalls in ihre Säureadditionssalze überführt.
Als unter den Reaktionsbedingungen inertes Lösungsmittel können z.B. aromatische Kohlenwasserstoffe wie Benzol, Toluol oder Xylol, Di(nieder)alkylamide niederer aliphatischer Monocarbonsäuren wie Dimethylformamid oder chlorierte aliphatische Kohlenwasserstoffe wie Chloroform oder Tetrachlorkohlenstoff verwendet werden.
Als säurebindende Mittel könnten z.B. Alkalimetallkarbonate wie Kalium- oder Natriumkarbonat, tertiäre Amine wie Triäthylamin oder Pyridin oder ein Überschuss der Verbindungen der Formel II verwendet werden. In den Verbindungen der Formel III bedeutet X beispielsweise Chlor, Jod oder den Säurerest einer organischen Sulfonsäure wie z.B. einen Alkylsulfonyloxy- oder Arylsulfonyloxyrest, vorzugsweise aber Brom. Zur Beschleunigung der Reaktion kann das Reaktionsgemisch erwärmt und / oder durchmischt werden, vorzugsweise arbeitet man bei leicht erhöhter Temperatur zwischen 30 und 900 und unter Rühren.
Praktisch kann das Verfahren z.B. ausgeführt werden, indem man die Verbindungen der Formel II u. das säurebindende Mittel, z.B. ein Alkalimetallkarbonat vorzugsweise Kaliumkarbonat, in Lösungsmittel wie Toluol oder Dimethylformamid einträgt und die Verbindung der Formel III bei Raumtemperatur oder leicht erhöhter Temperatur, z.B. zwischen etwa 30-700, allmählich unter Rühren zusetzt. Das Reaktionsgemisch wird anschl?essend z.B. unter Rühren erwärmt, vorzugsweise auf Temperaturen zwischen 30 und 900, und nach Beendigung der Reaktion (ca. lz; bis 5 Stunden) z.B. in Wasser gegossen.
Die Verbindungen der Formel I können auf übliche Weise aus dem Reaktionsgemisch isoliert und nach bekannten Methoden gereinigt werden.
Verbindungen der Formel Ia, worin R1I für Wasserstoff, Halogen, eine niedere Alkyl-, Alkoxy- oder Alkylthiogruppe, die Trifluormethyl-, Nitro-, Acetaminooder Cyangruppe R21 für eine l-Naphthyl-, 1,2,3,4-Te- trahydroS-naphthyl- oder 5-Indanylgruppe, die gegebenenfalls im aromatischen Ring durch Halogen, eine niedere Alkyl-, Alkoxy- oder Alkylthiogruppe, die Trifluormethyl-, Nitro-, Acetamino- oder Cyangruppe substituiert sein können, und R31 für eine niedere Acylgruppe stehen, können auch aus Verbindungen der Formel Ib, worin R11 und R21 obige Bedeutung besitzen und die einen Spezialfall der Verbindungen der Formel I darstellen, durch Acylierung nach bekannten Verfahren erhalten werden, z:
:B. mittels Säurehalogeniden oder Säureanhydriden.
Aus den freien Basen lassen sich in bekannter Weise ihre Salze mit Säuren herstellen und umgekehrt.
Die Verbindungen der Formel II sind neu und können hergestellt werden, indem man a) eine Verbindung der Formel IV, worin R2 obige Bedeutung besitzt und R4 für eine niedere Alkylgruppe, die Phenyl- oder Benzylgruppe steht, in saurem oder alkalischem Medium zu einer Verbindung der Formel lib, worin R2 obige Bedeutung besitzt, hydrolysiert, oder b) zur Herstellung von Verbindungen der Formel IIa, worin R211 für eine l-Naphthyl-, 1,2,3,4-Tetrahydro-5 -naphthyl- oder 5-Indanylgruppe, die gegebenenfalls im aromatischen Ring durch eine niedere Alkyl-, Alkylthiooder Alkoxygruppe, die Trifluormethyl-, Nitro-, Amino-, Cyan- od.
Acetaminogruppe substituiert sein kann, steht, eine Verbindung der Formel V, worin R2ll obige Bedeutung besitzt, hydrogenolytisch debenzyliert, oder c) zur Herstellung einer Verbindung der Formel IIc, worin R2lll für eine l-Naphthyl-, 1,2.3,4-Tetrahydro-5 -naphthyl- oder 5-Indanylgruppe, die gegebenenfalls im aroma tischen Ring durch eine niedere Alkyl-, Alkylthiooder Alkoxygruppe, die Trifluormethyl-, Nitro-, Cyanoder Acetaminogruppe substituiert sein können, und R11 obige Bedeutung besitzt, eine Verbindung der Formel Va, worin R2111 und R31 obige Bedeutung besitzen, hydrogenolytisch debenzyliert,
oder d) zur Herstellung einer Verbindung der Formel lid, worin R21 und Rsl obige Bedeutung besitzen, eine Verbindung der Formel IVa, worin R.,l, R31 und R4 obige Bedeutung besitzen, in saurem oder alkalischem Medium hydrolysiert.
Zur Hydrolyse nach Verfahren a) oder d) kann man beispielsweise konzentrierte Salzsäure verwenden und die Reaktion bei erhöhter Temperatur, z.B. bei Siedetemperatur des Reaktionsgemisches, ausführen.
Die Verbindungen der Formel IV können erhalten werden, indem man eine Verbindung der Formel VI, worin R4 obige Bedeutung besitzt, mit einer Verbindung der Formel VII, worin R obige Bedeutung besitzt, in einem unter den Reaktionsbedingungen inerten Lösungsmittel z.B. in einem niederen Alkanol wie Äthanol oder in einem aromatischen Kohlenwasserstoff wie Benzol oder Toluol, gegebenenfalls unter Zusatz katalytischer Mengen von Eisessig oder p-Toluolsulfonsäure zu Verbindungen der Formel VIII, worin R2 und R4 obige Be- deutung besitzen, umsetzt und diese mit komplexen Borhydriden wie Natrium- oder Kaliumborhydrid in einem Lösungsmittel, z.B. in einem niederen Alkanol wie Äthanol oder in einem cyclischen Äther wie Dioxan zu Verbindungen der Formel IV reduziert.
Die Verbindungen der Formel V können erhalten werden, indem man die Verbindung der Formel IX mit einer Verbindung der Formel VIIa, worin R211 obige Bedeutung besitzt, kondensiert und die entstandenen Verbindungen der Formel X, worin R211 obige Bedeutung besitzt, mit komplexen Borhydriden wie oben beschrieben reduziert.
Die Verbindungen der Formel IVa bzw. Va können aus den Verbindungen der Formel IVb, worin R' und R4 obige Bedeutung besitzen, bzw. Vb, worin R,XII obige Bedeutung besitzt, durch Acylierung nach bekannten Me thoden erhalten werden, z.B. mit Säurechloriden oder Säureanhydriden.
Die Verbindungen der Formel IVb, bzw. Vb stellen Spezialfälle der Verbindungen der Formel IV bzw. V dar.
Die durch R1 symbolisierten sowie die gegebenenfalls als Substituenten in R vorhandenen niederen Alkyl-, Alkoxy- oder Alkylthiogruppen sowie die durch R3 symbolisierte niedere Acylgruppe bestehen vorzugsweise aus 1 bis 4 Kohlenstoffatomen.
Soweit die Herstellung der Ausgangsverbindungen nicht beschrieben wird, sind diese bekannt oder nach an sich bekannten Verfahren bzw. analog zu den hier beschriebenen oder analog zu an sich bekannten Verfahren herstellbar.
Die Verbindungen der Formel I und ihre pharmakologisch verträglichen Säureadditionssalze besitzen bei geringer Toxizität interessante pharmakodynamische Eigenschaften und können daher als Heilmittel verwendet werden. Sie zeigen insbesondere analgetische Wirksamkeit, wie durch Versuche an der Maus (Phenylbenzochinonsyndrom und hot-plate ) gezeigt wurde. Die zu verwendenden Dosen variieren naturgemäss je nach der Art der Administration und des zu behandelnden Zustandes. Im allgemeinen werden jedoch bei Testtieren befriedigende Resultate mit einer Dosis von 5 bis 30 mg1kg Körpergewicht erhalten; diese Dosis kann nötigenfalls in 2 bis 3 Anteilen oder auch als Retardform verabreicht werden.
Für grössere Säugetiere liegt die Tagesdosis bei etwa 30 bis 150 mg. Für orale Applikationen enthalten die Teildosen etwa 10 bis 75 mg der Verbindungen der Formel I neben festen oder flüssigen Trägersubstanzen.
Als Heilmittel können die Verbindungen der Formel I bzw. ihre physiologisch verträglichen Säureadditionssalze allein oder in geeigneter Arzneiform mit pharmakologisch indifferenten Hilfsstoffen verabreicht werden.
In den nachfolgenden Beispielen, die die Erfindung näher erläutern, ihren Umfang aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden und sind unkorrigiert.
Beispiel I
4-(1 -Naphthylamino).1.phenäthylpiperidin
Zu einer auf 500 erhitzten Aufschlämmung von 22,8 g 4--(1-Naphthylamino)piperidin und 16,7 g Kaliumkarbonat in 100 ml Dimethylformamid wird ein Gemisch von 18,7 g Phenäthylbromid und 50 ml Dimethylformamid innerhalb 2 Stunden zugetropft. Anschliessend wird der Ansatz noch weitere 4 Stunden bei der gleichen Temperatur gerührt. Das ungelöste Material wird abfiltriert und das klare Filtrat im Vakuum zur Trockne eingeengt.
Durch Kristallisation aus 95%igem Äthanol/Äther erhält man die im Titel genannte reine Verbindung vom Fp. 93 950. Das in äthanolischer Lösung hergestellte Fumarat schmilzt bei 204-2060 (Zers.).
Beispiel 2
Analog wie in Beispiel 1 beschrieben kann auch 1 -Phenäthyl-4-(1,2,3,4-tetrahydro- 5- naphthylamino)piperidin-fumarat erhalten werden, Fp. 218-2200 (Zers.).
Beispiel 3
N-(1-Naphthyl)-N-(1 -phenäthyl-4-piperidyl)proplonamid
Zu einer Lösung von 15,5 g 4-(lJNaphthylamino)-l- -phenäthyl-piperidin (Herstellung siehe Beispiel 1) in 100 ml Benzol wird eine Lösung von 7,4 g Propionsäureanhydrid in 20 ml Benzol auf einmal zugegeben. Der Ansatz wird während 8 Stunden unter Rühren am Rückfluss zum Sieden erhitzt, nach dem Abkühlen auf 200 mit 25 ml 10%iger Natronlauge versetzt und während 30 Minuten bei Zimmertemperatur gerührt. Nach dem Abtrennen des Alkaliteils wird die organische Phase mit Wasser nachgewaschen, über Natriumsulfat getrocknet und im Vakuum zum braunen öl eingedampft. Durch Kristallisieren mit n-Hexan/Äther erhält man die im Titel genannte reine Verbindung vom Fp. 87-900.
Beispiel 4
N-(1 -Naphthyl)-N-(1 -phenäthyl-4-piperidyl)propionamid
Zu einer auf 550 erhitzten Aufschlämmung von 14,1 g öligem N-( 1 -Naphthyl)-N-(4-piperidyl)propionamid (hergestellt aus dem Fumarat, Fp. 195-1970 (Zers.) und 7,3 g Kaliumkarbonat in 75 ml Dimethylformamid) wird ein Gemisch von 9,3 g Phenäthylbromid in 25 ml Dimethylformamid innerhalb 1% Stunden zugetropft. Anschliessend wird der Ansatz noch weitere 3 Stunden bei gleicher Temperatur gerührt. Das ungelöste Material wird abfiltriert und das Filtrat im Vakuum zur Trockne eingeengt.
Durch Kristallisation mit n-Hexan/Äther erhält man die im Titel genannte reine Verbindung vom Fp. 87-900.
Beispiel 5
N-(l -Pkenäthyl-4-piperidyl)-N-(1,2,3,4-tetrakydro-5- -napAthyl)propionamid
Analog wie im Beispiel 4 beschrieben kann auch das N - (1 - Phenäthyl - 4 - piperidyl). N - (1,2,3,4 - tetrahydro- 5 -naphthyl)propionamid, Fp. 81-830, erhalten werden.
Die benötigten Ausgangsprodukte können folgender- massen erhalten werden:
Beispiel 6
4-(1-Naphthylamino)piperidin (für Beispiel I)
Eine Lösung von 6,1 g 4-(1.Naphthylamino)piperidin- -1-carbonsäureäthylester in 700 ml konz. Salzsäure wird während 24 Stunden unter Rühren am Rückfluss zum Sieden erhitzt. Die saure Lösung wird gekühlt, mit Natriumhydroxyd alkalisch gestellt und das abgeschiedene öl mit 1000 ml Benzol extrahiert. Die organische Phase wird 3mal mit je 150 ml Wasser gewaschen, über Natriumsulfat getrocknet und im Vakuum eingedampft, wobei die im Titel genannte Verbindung als viskoses öl zurückbleibt.
Das in äthanolischer Lösung hergestellte Hydrogenmaleinat schmilzt bei 168-1700 (Zers.).
Beispiel 7 4-(1,2,3,4-Tetrahydro-5-napAthylamino)piperidin (für Beispiel 2)
Die Titelverbindung wird analog wie in Beispiel 6 beschrieben erhalten, Smp. des Dihydrochlorids 289-2930 (Zers.).
Beispiel 8 N-(l -Naphthyl)-N-(4-piperidyl)propionamid (für Beispiel 4) a) 1 .Benzyl.4.(1 .naphthylimino)piperidin
Eine Lösung von 143,1 g 1 -Naphthylamin und 189,2 g 1-Benzylpiperidon(4) in 600 ml Toluol wird 30 Stunden unter Rühren am Rückfluss zum Sieden erhitzt, wobei das bei der Reaktion entstehende Wasser durch azeotrope Destillation kontinuierlich abgetrennt wird. Die heisse Lösung wird im Vakuum eingedampft und das Rohprodukt ohne weitere Reinigung weiterverwendet.
b) 1-Benzyl-4-(1-naphthylamino)piperidin
Zu einer Suspension von 37,5 g Natriumborhydrid in 250 ml 95%igem Äthanol wird eine Lösung von 340,6 g öligem 1-Benzyl-4-(1-naphthylimino)piperidin in 300 ml 95%igem Äthanol innerhalb 1 Stunde unter Kühlen zugetropft. Den Ansatz rührt man zunächst während 15 Stunden bei Zimmertemperatur, anschliessend wird 3 Stunden unter Rühren am Rückfluss zum Sieden erhitzt.
Das Reaktionsgemisch wird auf 200 gekühlt, mit 450 ml Methanol und 300 ml Wasser versetzt und der ausgefallene Niederschlag abfiltriert. Das Filtrat wird im Vakuum zur Trockne eingedampft, mit dem Filterrückstand vereinigt und das Ganze zwischen je 1000 ml Äther und Wasser verteilt. Die organische Phase wird mit Wasser nachgewaschen, über Natriumsulfat getrocknet und im Vakuum zum kristallinen Rückstand eingeengt. Nach dem Umkristallisieren aus Äthanol schmilzt die im Titel genannte Verbindung bei 96-980. Das in äthanolischer Lösung hergestellte Fumarat schmilzt bei 203-2060 (Zers.).
c) N-(1 -Benzyl-4-piperidyl)-N-( 1 -naphthyl)propionamid
Eine Lösung von 31,6 g 1 -Benzyl-4-(1 -naphthylami- no)piperidin und 26,0 g Propionsäureanhydrid in 150 ml Benzol wird während 18 Stunden unter Rühren am Rückfluss zum Sieden erhitzt. Nach dem Abkühlen auf 200 wird der Ansatz mit 150 ml 1 0%iger Natronlauge versetzt und während 1 Stunde bei Zimmertemperatur gerührt. Nach dem Abtrennen des Alkaliteils wird die organische Phase mit Wasser nachgewaschen, über Natriumsulfat getrocknet und im Vakuum zum öl eingeengt. Das in äthanolischer Lösung hergestellte und aus Methanol umkristallisierte Fumarat schmilzt bei 191-1930 (Zers.).
d) N-( 1 -Naphthyl) -N-(4-piperidyl)propionamid
Man löst 31,7 g öliges N-(l-Benzyl-4-piperidyl)-N-(l- -naphthyl)propionamid in 200 ml Eisessig, setzt 3 g Palladium-Kohle-Katalysator (10%) zu und hydriert 12 Stunden lang bei 200 und 3 atü Wasserstoffdruck. Anschliessend wird der Katalysator über Cellite abfiltriert und das Filtrat im Vakuum zur Trockne eingedampft. Den Rückstand verteilt man zwischen je 150 ml Chloroform und 1obiger Natronlauge. Die organische Phase wird abgetrennt, mit Wasser nachgewaschen, über Natriumsulfat getrocknet und im Vakuum zum öl eingedampft. Das in äthanolischer Lösung hergestellte Fumarat schmilzt bei 195-1970 (Zers.).
Beispiel 9 N-(4-Piperidyl)-N-(1,2,3,4-tetrahydro-5-naphthyl)- propionamid (für Beispiel 5)
Die Titelverbindung wird analog wie in Beispiel 8 beschrieben hergestellt. Smp. des Bis-naphthalin-1,5-disulfonat-dihydrats 235-2400 (Zers.).
N-(1 -Benzyl-4-piperidyl)-N-(1,2,3,4-tetrahydro-5 naphthyl)-propionamid-fumarat, Smp. 181 - 1830; Zers.); 1-l8enzyl4-(1 ,2,3 A-tetrahydro--i -naphthylamino)- -piperidin-fumarat. Smp. 220-2230 (Zers.).
Beispiel 10
4-(1-Naphthylamino)piperidin4 -carbonsiureäthyiester (für Beispiel 6) a) 4-(1-Naphthylimino)piperidin-1-carbonsäureäthylester
Eine Lösung von 71,5 g l-Naphthylamin und 85,5 g l-Äthoxycarbonyl-4-piperidon in 250 ml Toluol wird 16 Stunden unter Rühren am Rückfluss zum Sieden erhitzt, wobei das bei der Reaktion entstehende Wasser durch azeotrope Destillation kontinuierlich abgetrennt wird.
Die heisse Lösung wird im Vakuum eingedampft und das als Rückstand verbleibende öl ohne Reinigung als Rohprodukt weiter verarbeitet.
b) 4-(l -Naphthylamino)piperidin- 1 -carbonsäureäthyl- ester
Zu einer Suspension von 22,7 g Natriumborhydrid in 250 ml 95%igem Äthanol wird eine Lösung von 148 g des obigen rohen 4-( 1 -Naphthylimino)piperidin-l -carbon- säureäthylesters in 250 ml 95%igem Äthanol innerhalb von 60 Minuten unter Kühlen zugetropft. Anschliessend wird 3 Stunden unter Rühren am Rückfluss zum Sieden erhitzt. Das Reaktionsgemisch wird auf 100 gekühlt, mit 200 ml Wasser versetzt und filtriert, der Filterrückstand wird zwischen 1000 ml Benzol und 750 ml Wasser ausgeschüttelt.
Die organische Phase wird über Natriumsulfat getrocknet, filtriert und im Vakuum eingedampft, wobei die im Titel genannte Verbindung als kristalliner Rückstand verbleibt. Smp. 114-1150 nach Kristallisation aus abs. Äthanol.
Beispiel 11 4Xl 234-TetraXydro-s-naphthylamino)piperidin-l -carbonsäureäthylester (für Beispiel 7)
Analog wie in Beispiel 10 beschrieben wird die Titelverbindung vom Smp. 95-970 (Zers.) erhalten.
Beispiel 12 4-(5-lndanamino)-1-phenuthylpiperidin
Zu einer auf 500 erhitzten und zunächst während 15 Minuten bei dieser Temperatur gerührten Aufschlämmung von 4,2 g 4-(5-Indanamino)piperidinfumarat und 2,3 g Kaliumkarbonat in 50 ml Dimethylformamid wird ein Gemisch von 1,5 g Phenäthylbromid und 50 ml Dimethylformamid innerhalb 2 Stunden zugetropft. Anschliessend wird der Ansatz noch weitere 12 Stunden bei der gleichen Temperatur gerührt. Das ungelöste Material wird abfiltriert und das klare Filtrat im Vakuum zur Trockne eingeengt. Der Rückstand wird zwischen Chloroform und Wasser verteilt und der getrocknete Chloroformteil im Vakuum eingeengt. Das rötliche öl ergibt in äthanolischer Lösung ein kristallisiertes Fumarat, das nach Umkristallisieren aus abs. Äthanol bei 199-2010 schmilzt.

Die benötigten Ausgangsprodukte können folgendermassen erhalten werden:
Beispiel 20 4-(5-lndai2crmino)piperidin (für Beispiel 12)
Eine Lösung von 95,3 g 4-(5-Indanamino)piperidin-1 -carbonsäure-äthylester in 500 ml konz. Salzsäure wird während 16 Stunden unter Rühren am Rückfluss zum Sieden erhitzt. Die saure Lösung wird gekühlt, mit 30%iger Natronlauge alkalisch gestellt und das abgeschiedene öl mit insgesamt 2000 ml Chloroform extrahiert. Die organischen Phasen werden 5mal mit je 200 ml Wasser gewaschen, über Natriumsulfat getrocknet und im Vakuum eingedampft, wobei die im Titel genannte Verbindung als viskoses öl zurückbleibt.
Das in äthanolischer Lösung hergestellte Fumarat schmilzt nach Umkristallisieren aus Methanol bei 2362380 (Zers.).
Analog wie in Beispiel 20 beschrieben können auch folgende Verbindungen erhalten werden (Beispiel 21 bis 24).

Eine Lösung von 44,2 g 4-Brom-1-naphthylamin und 41,6 g l-Benzylpiperidon(4) in 350 ml Toluol und 5 ml Eisessig wird 4 Stunden unter Rühren am Rückfluss zum Sieden erhitzt, wobei das bei der Reaktion entstehende Wasser durch azeotrope Destillation kontinuierlich abgetrennt wird. Die Lösung wird im Vakuum eingedampft und das Rohprodukt ohne weitere Reinigung weiter verwendet.
b) 1 -Benzy1-4-(4-brom-1 -naphthylamino)piperidin
Zu einer Suspension von 8,0 g Natriumborhydrid in 100 ml 95%igem Äthanol wird eine Lösung von 85,0 g öligem 1 -Benzyl-4-(4-brom- 1 -naphthylimino)piperidin in 160¯ml 95t0igem Äthanol innerhalb 30 Minuten zugetropft. Anschliessend wird 2 Stunden unter Rühren am Rückfluss zum Sieden erhitzt. Der Ansatz wird im Vakuum zur Trockne eingedampft und zwischen 800 ml Chloroform und 350 ml Wasser verteilt. Die organische Phase wird mit Wasser nachgewaschen, über Natriumsulfat getrocknet und im Vakuum zu einem öligen Rückstand eingeengt.
Nach dem Kristallisieren mit Aceton wird das Rohprodukt aus abs. Äthanol umkristallisiert. Die im Titel genannte Verbindung schmilzt bei 134-136 (Zers.).
c) 4-(4-Brom- 1 -naphthylamino)piperidin
Man löst 18,3 g kristallisiertes 1 -Benzyl-4-(4-brom- 1 - -naphthylamino)piperidin in 150 ml Eisessig, setzt 2 g Palladium-Kohle-Katalysator (10So) zu und hydriert 9 Stunden lang bei 200 und 3 atü Wasserstoffdruck. Anschliessend wird der Katalysator über Cellite abfiltriert und das Filtrat im Vakuum zur Trockne eingedampft, worauf man die im Titel genannte Verbindung als öl erhält.
Beispiel 26 4-(5-lndanamino)piperidin-1-carbonsäureäthylester (für Beispiel 20) a) 445-Indanimino)piperidin-1-carbonsäureäthylester
Eine Lösung von 64,6 g 5-Indanamin und 83,0 g 1 Athoxyvarbonyl-4-piperidon in 300 ml Benzol wird 18 Stunden unter Rühren am Rückfluss zum Sieden erhitzt, wobei das bei der Reaktion entstehende Wasser durch azeotrope Destillation kontinuierlich abgetrennt wird. Die Lösung wird im Vakuum eingedampft und das als Rückstand verbleibende öl ohne Reinigung als Rohprodukt weiter verarbeitet.
b) 4-(5-Indanamino)piperidin- 1 -carbonsäureäthylester
Zu einer Suspension von 19,4 g Natriumborhydrid in 250 ml 95%igem Äthanol wird eine Lösung von 146,8 g rohem 4-(5-Indanimino)piperidin-1 -carbonsäureäthylester in 250 ml 95%igem Äthanol innerhalb 30 Minuten zugetropft. Anschliessend wird 14 Stunden unter Rühren am Rückfluss zum Sieden erhitzt. Das Reaktionsgemisch wird auf 200 gekühlt, mit 20 ml Aceton versetzt und im Vakuum zur Trockne eingeengt. Der Rückstand wird zwischen je 300 ml Chloroform und Wasser verteilt. Die organische Phase wird mit Wasser nachgewaschen, über Natriumsulfat getrocknet, filtriert und im Vakuum ein gedampft, wobei die im Titel genannte Verbindung mit Äther/Hexan aus dem öligen Rohprodukt kristallin erhalten werden kann.
Smp. 72-74 nach einmaligem Umkristallisieren aus Leichtbenzin.

The invention relates to a process for the preparation of new N-phenethylpiperidine derivatives of the formula I, wherein R1 is hydrogen, halogen, a lower alkyl, alkylthio or alkoxy group, the trifluoromethyl, nitro, amino, cyano or acylamino group, R2 is a 1 naphthyl, 1,2,3,4-tetrahydro-5-naphthyl or 5-indanyl group, optionally substituted in the aromatic ring by halogen, a lower alkyl, alkylthio or alkoxy group, the trifluoromethyl, nitro, amino , Cyano or acetamino groups can be substituted, and Rs stand for hydrogen or a lower acyl group, but R2 must not be substituted by an amino group if R3 stands for a lower acyl group, and their acid addition salts.

Solvents which are inert under the reaction conditions can e.g. aromatic hydrocarbons such as benzene, toluene or xylene, di (lower) alkylamides of lower aliphatic monocarboxylic acids such as dimethylformamide or chlorinated aliphatic hydrocarbons such as chloroform or carbon tetrachloride can be used.
Acid-binding agents could e.g. Alkali metal carbonates such as potassium or sodium carbonate, tertiary amines such as triethylamine or pyridine or an excess of the compounds of the formula II can be used. In the compounds of formula III, X is, for example, chlorine, iodine or the acid residue of an organic sulfonic acid such as e.g. an alkylsulfonyloxy or arylsulfonyloxy radical, but preferably bromine. To accelerate the reaction, the reaction mixture can be heated and / or mixed, preferably at a slightly elevated temperature between 30 and 900 and with stirring.
In practice the method can e.g. be carried out by the compounds of formula II u. the acid binding agent, e.g. an alkali metal carbonate, preferably potassium carbonate, in solvents such as toluene or dimethylformamide and the compound of formula III at room temperature or slightly elevated temperature, e.g. between about 30-700, gradually adding with stirring. The compounds of the formula II are new and can be prepared by a) a compound of the formula IV, wherein R2 has the above meaning and R4 is a lower alkyl group, the phenyl or benzyl group, in an acidic or alkaline medium to a compound of Formula lib, in which R2 has the above meaning, hydrolyzed, or b) for the preparation of compounds of the formula IIa, in which R211 is a 1-naphthyl, 1,2,3,4-tetrahydro-5-naphthyl or 5-indanyl group, which is optionally in the aromatic ring by a lower alkyl, alkylthio or alkoxy group, the trifluoromethyl, nitro, amino, cyano or.

For the hydrolysis according to process a) or d), for example, concentrated hydrochloric acid can be used and the reaction carried out at elevated temperature, e.g. at the boiling point of the reaction mixture.
The compounds of the formula IV can be obtained by treating a compound of the formula VI, in which R4 has the above meaning, with a compound of the formula VII, in which R has the above meaning, in a solvent which is inert under the reaction conditions, e.g. in a lower alkanol such as ethanol or in an aromatic hydrocarbon such as benzene or toluene, optionally with the addition of catalytic amounts of glacial acetic acid or p-toluenesulfonic acid to form compounds of the formula VIII, where R2 and R4 are as defined above, and these with complex borohydrides such as Sodium or potassium borohydride in a solvent, e.g. reduced in a lower alkanol such as ethanol or in a cyclic ether such as dioxane to give compounds of the formula IV.

The compounds of the formula IVa or Va can be obtained from the compounds of the formula IVb, in which R 'and R4 have the above meaning, or Vb, in which R, XII has the above meaning, by acylation according to known methods, e.g. with acid chlorides or acid anhydrides.
The compounds of the formula IVb and Vb represent special cases of the compounds of the formula IV and V, respectively.
The lower alkyl, alkoxy or alkylthio groups symbolized by R1 and any lower alkyl, alkoxy or alkylthio groups symbolized by R3 and the lower acyl group symbolized by R3 preferably consist of 1 to 4 carbon atoms.
If the preparation of the starting compounds is not described, they are known or can be prepared by processes known per se or analogously to those described here or analogously to processes known per se.
The compounds of the formula I and their pharmacologically acceptable acid addition salts have interesting pharmacodynamic properties with low toxicity and can therefore be used as medicaments. In particular, they show analgesic effectiveness, as was shown by experiments on mice (phenylbenzoquinone syndrome and hotplate). The doses to be used naturally vary depending on the type of administration and the condition to be treated. In general, however, satisfactory results are obtained in test animals with a dose of 5 to 30 mg / kg body weight; if necessary, this dose can be administered in 2 to 3 portions or as a sustained-release form.
For larger mammals, the daily dose is around 30 to 150 mg. For oral administration, the partial doses contain about 10 to 75 mg of the compounds of the formula I in addition to solid or liquid carrier substances.
The compounds of the formula I or their physiologically tolerable acid addition salts can be administered as medicaments alone or in a suitable medicinal form with pharmacologically inert auxiliaries.
In the following examples, which explain the invention in more detail but are not intended to restrict its scope in any way, all temperatures are given in degrees Celsius and are uncorrected.
Example I.
4- (1-naphthylamino) .1.phenethylpiperidine
A mixture of 18.7 g of phenethyl bromide and 50 ml of dimethylformamide is added dropwise over 2 hours to a suspension, heated to 500, of 22.8 g of 4 - (1-naphthylamino) piperidine and 16.7 g of potassium carbonate in 100 ml of dimethylformamide. The batch is then stirred for a further 4 hours at the same temperature. The undissolved material is filtered off and the clear filtrate is concentrated to dryness in vacuo.
Crystallization from 95% ethanol / ether gives the pure compound mentioned in the title with a melting point of 93,950. The fumarate produced in ethanolic solution melts at 204-2060 (decomp.).
Example 2
1-Phenethyl-4- (1,2,3,4-tetrahydro-5-naphthylamino) piperidine fumarate can also be obtained analogously to that described in Example 1, melting point 218-2200 (decomp.).
Example 3
N- (1-naphthyl) -N- (1-phenethyl-4-piperidyl) proplonamide
A solution of 7.4 g of propionic anhydride in 20 ml of benzene is added all at once to a solution of 15.5 g of 4- (lJnaphthylamino) -l- -phenethyl-piperidine (preparation see Example 1) in 100 ml of benzene. The batch is refluxed for 8 hours while stirring, after cooling to 200, 25 ml of 10% sodium hydroxide solution are added and the mixture is stirred at room temperature for 30 minutes. After the alkali has been separated off, the organic phase is washed with water, dried over sodium sulphate and evaporated to a brown oil in vacuo. Crystallization with n-hexane / ether gives the pure compound mentioned in the title with a melting point of 87-900.
Example 4
N- (1-naphthyl) -N- (1-phenethyl-4-piperidyl) propionamide
To a slurry heated to 550 of 14.1 g of oily N- (1-naphthyl) -N- (4-piperidyl) propionamide (prepared from the fumarate, m.p. 195-1970 (dec.) And 7.3 g of potassium carbonate in 75 ml of dimethylformamide) a mixture of 9.3 g of phenethyl bromide in 25 ml of dimethylformamide is added dropwise within 1% hours. The batch is then stirred for a further 3 hours at the same temperature. The undissolved material is filtered off and the filtrate is concentrated to dryness in vacuo.

N- (1 -Pkenethyl-4-piperidyl) -N- (1,2,3,4-tetrakydro-5-napAthyl) propionamide
Analogously as described in Example 4, N - (1 - phenethyl - 4 - piperidyl) can also be used. N - (1,2,3,4-tetrahydro-5-naphthyl) propionamide, m.p. 81-830.

4- (1-naphthylamino) piperidine (for example I)
A solution of 6.1 g of 4- (1.Naphthylamino) piperidine- -1-carboxylic acid ethyl ester in 700 ml of conc. Hydrochloric acid is refluxed with stirring for 24 hours. The acidic solution is cooled, made alkaline with sodium hydroxide and the separated oil is extracted with 1000 ml of benzene. The organic phase is washed 3 times with 150 ml of water each time, dried over sodium sulfate and evaporated in vacuo, the compound mentioned in the title remaining as a viscous oil.
The hydrogen maleate produced in ethanolic solution melts at 168-1700 (decomp.).
Example 7 4- (1,2,3,4-Tetrahydro-5-napAthylamino) piperidine (for Example 2)
The title compound is obtained analogously to that described in Example 6, melting point of the dihydrochloride 289-2930 (dec.).
Example 8 N- (1 -Naphthyl) -N- (4-piperidyl) propionamide (for example 4) a) 1.Benzyl.4. (1.naphthylimino) piperidine
A solution of 143.1 g of 1-naphthylamine and 189.2 g of 1-benzylpiperidone (4) in 600 ml of toluene is refluxed for 30 hours with stirring, the water formed in the reaction being continuously separated off by azeotropic distillation. The hot solution is evaporated in vacuo and the crude product is used further without further purification.
b) 1-Benzyl-4- (1-naphthylamino) piperidine
To a suspension of 37.5 g of sodium borohydride in 250 ml of 95% ethanol, a solution of 340.6 g of oily 1-benzyl-4- (1-naphthylimino) piperidine in 300 ml of 95% ethanol is added dropwise over the course of 1 hour while cooling . The batch is first stirred for 15 hours at room temperature, then it is refluxed for 3 hours with stirring.
The reaction mixture is cooled to 200 ml, 450 ml of methanol and 300 ml of water are added and the precipitate which has separated out is filtered off. c) N- (1 -benzyl-4-piperidyl) -N- (1-naphthyl) propionamide
A solution of 31.6 g of 1-benzyl-4- (1-naphthylamino) piperidine and 26.0 g of propionic anhydride in 150 ml of benzene is refluxed with stirring for 18 hours. After cooling to 200, 150 ml of 10% strength sodium hydroxide solution are added and the mixture is stirred at room temperature for 1 hour. After the alkali has been separated off, the organic phase is washed with water, dried over sodium sulfate and concentrated to an oil in vacuo. The fumarate produced in ethanol solution and recrystallized from methanol melts at 191-1930 (decomp.).
d) N- (1-naphthyl) -N- (4-piperidyl) propionamide
31.7 g of oily N- (l-benzyl-4-piperidyl) -N- (l- -naphthyl) propionamide are dissolved in 200 ml of glacial acetic acid, 3 g of palladium-carbon catalyst (10%) are added and the mixture is hydrogenated for 12 hours long at 200 and 3 atm hydrogen pressure. The catalyst is then filtered off through Celite and the filtrate is evaporated to dryness in vacuo. The residue is distributed between 150 ml of chloroform and the above sodium hydroxide solution. The organic phase is separated off, washed with water, dried over sodium sulfate and evaporated to an oil in vacuo. The fumarate produced in ethanolic solution melts at 195-1970 (decomp.).
Example 9 N- (4-piperidyl) -N- (1,2,3,4-tetrahydro-5-naphthyl) propionamide (for example 5)
The title compound is prepared analogously to that described in Example 8. Bis-naphthalene-1,5-disulfonate dihydrate 235-2400 (dec.).

4- (1-Naphthylamino) piperidine-4-carboxylic acid ethyl ester (for Example 6) a) 4- (1-Naphthylimino) piperidine-1-carboxylic acid ethyl ester
A solution of 71.5 g of l-naphthylamine and 85.5 g of l-ethoxycarbonyl-4-piperidone in 250 ml of toluene is refluxed for 16 hours with stirring, the water formed in the reaction being continuously separated off by azeotropic distillation .
The hot solution is evaporated in vacuo and the oil remaining as residue is processed further as a crude product without purification.
b) ethyl 4- (1-naphthylamino) piperidine-1-carboxylate
A solution of 148 g of the above crude 4- (1-naphthylimino) piperidine-1-carbonic acid ethyl ester in 250 ml of 95% ethanol is added to a suspension of 22.7 g of sodium borohydride in 250 ml of 95% ethanol over the course of 60 minutes added dropwise with cooling. The mixture is then refluxed for 3 hours while stirring. The reaction mixture is cooled to 100, mixed with 200 ml of water and filtered, the filter residue is shaken between 1000 ml of benzene and 750 ml of water.

Example 12 4- (5-indanamino) -1-phenutylpiperidine

A solution of 64.6 g of 5-indanamine and 83.0 g of 1 ethoxyvarbonyl-4-piperidone in 300 ml of benzene is refluxed for 18 hours with stirring, the water formed in the reaction being continuously separated off by azeotropic distillation. The solution is evaporated in vacuo and the oil remaining as residue is processed further as a crude product without purification.
b) 4- (5-indanamino) piperidine-1-carboxylic acid ethyl ester
To a suspension of 19.4 g of sodium borohydride in 250 ml of 95% ethanol, a solution of 146.8 g of crude 4- (5-indanimino) piperidine-1-carboxylic acid ethyl ester in 250 ml of 95% ethanol is added dropwise within 30 minutes. The mixture is then refluxed for 14 hours while stirring. The reaction mixture is cooled to 200, mixed with 20 ml of acetone and concentrated to dryness in vacuo. The residue is distributed between 300 ml of chloroform and water. The organic phase is washed with water, dried over sodium sulfate, filtered and evaporated in vacuo, whereby the compound mentioned in the title can be obtained in crystalline form from the oily crude product using ether / hexane.
