worin Rein Wasserstoffatom oder einen Acylrest bedeutet. Als Acylreste R kommen Reste in Frage, die sich von Säuren ableiten, die in der Steroidchemie üblicherweise für Veresterungen angewandt werden.
Bevorzugte Säuren sind organische Carbonsäuren mit bis zu 15 Kohlenstoffatomen, insbesondere niedere und mittlere aliphatische Carbonsäu- ren. Weiterhin können die Säuren auch ungesättigt, ver- zweigt, mehrbasisch oder in üblicher Weise, z.B. durch Hy- droxyl-, Aminogruppen oder Halogenatome, substituiert sein. Geeignet sind auch cycloaliphatische, aromatische, ge mischt aromatisch-aliphatische und heterocyclische Säuren, die ebenfalls in üblicher Weise substituiert sein können.
Als bevorzugte Säuren seien beispielsweise genannt: Essigsäure, Propionsäure, Capronsäure, Önanthsäure, Undecylsäure, Ölsäure, Trimethylessigsäure, Monochloressigsäure, Dichlor- essigsäure, Cyclopentylpropionsäure, Phenylpropionsäure, Phenylessigsäure, Phenoxyessigsäure, Dialkylaminoessigsäure, Piperidinoessigsäure, Bernsteinsäure, Benzoesäure u. a. Für die Herstellung wasserlöslicher Präparate kommen insbeson dere Ester anorganischer Säuren in Betracht, wie die der Schwefel- und der Phosphorsäure.
Die in 6,7-Stellung isomeren 4-Chlor-1α,2α;6ss,7B-di- methylen-3-keto-4-pregnene sind bereits im Hauptpatent Nr. 518 272 beschrieben. Sie besitzen starke gestagene und ovulationshemmende Wirksamkeit.
Es wurde nun gefunden, dass die neuen 4-Chlor- 1α,2α;6α,7α-dimethylen-3-keto-4-pregnene der Formel I in ihrer ovulationshemmenden Wirksamkeit die isomeren 6ss,7ss- Methylenverbindungen des Hauptpatents übertreffen.
Die Ovulationshemmung wurde wie üblich durch Tuben inspektion ermittelt. In der nachfolgenden Tabelle wird je weils der prozentuale Anteil der Rattenweibchen angegeben, bei denen nach subcutaner Applikation der genannten Tages dosis, jeweils eine Ovulation unterbleibt.

Aus dieser Tabelle ist ersichtlich, dass das 1,2α;6,7α- Bismethylensteroid 2) in wesentlich geringerer Dosierung ovulationshemmend wirkt, als das bekannte isomere 1,2α;6,7ss-Bismethylensteroid 1).
Aufgrund ihrer wertvollen pharmakologischen Eigen schaften können die neuen Wirkstoffe beispielsweise zur Konzeptionsverhütung und in der Gynäkologie Verwendung finden.
Das erfindungsgemässe Verfahren ist dadurch gekenn zeichnet, dass man die 1,2- und 6,7-Doppelbindung eines entsprechenden 3-Oxo-4-chlor-A1,4,6-steroids mit Dimethyl- methylensulfoniumoxid methyleniert und aus dem erhalte nen Isomerengemisch das 6α,7α-Isomere abscheidet. Das Methylenierungsprodukt besteht aus einem Isomerengemisch von 6ss,7ss- und 6α,7α-Methylenverbindung, aus dem man das 6α,7α-Isomere durch die üblichen Trennungsverfahren iso liert. Die Abtrennung wird beispielsweise durch Chroma tographieren und/oder fraktioniertes Umkristallisieren er reicht.
Je nach der Bedeutung, die R im Endprodukt haben soll, kann gegebenenfalls vor oder nach der Isomerentrennung das Reaktionsprodukt einer Veresterung oder Verseifung unter worfen werden.
Die Veresterung und die Verseifung können nach be kannten Methoden durchgeführt werden. Speziell genannt seien für die Veresterung die Umsetzung mit Säureanhy driden oder -chloriden in Gegenwart einer Säure oder der Umsatz der gewünschten Säure in Gegenwart von Trifluor- essigsäureanhydrid.
Die Verseifung wird beispielsweise mit Basen in alkoho lischer Lösung durchgeführt.
Das für die Methylenierung verwendete Dimethyl- methylensulfoniumoxid kann aus einem Trimethylsulfoxo- niumsalz, wie zum Beispiel dem Halogenid, Perchlorat oder Methylsulfat, in einem aprotonischen Lösungsmittel, wie Dimethylsulfoxid oder Dimethylformamid, mit einer wasser freien Base freigesetzt werden.
Zur Durchführung des er- findungsgemässen Verfahrens bringt man gewöhnlich das so erhältliche Dimethylmethylensulfoniumoxid ohne Isolierung bei Temperaturen von -10 bis 100 C, vorzugsweise bei Raumtemperatur, mit dem genannten 4-Chlor-1,4,6-trien- 3-keto-steroid zur Reaktion, wobei je nach den Reaktionsbe dingungen ein Isomerengemisch 6ss,7ss-Methylen- zu 6α,7α- Methylenverbindungen von 70:30 bis 80:20 entsteht. Beispiel 1 250 g Trimethylsulfoxoniumjodid werden in 2,5 Liter Dimethylsulfoxid unter Rühren und Stickstoffbegasung ge löst.
Nach Zugabe von 40 g Natriumhydroxidpulver rührt man 30 Minuten unter Stickstoffbegasung. 180 g 4-Chlor- 17-hydroxy-1,4,6-pregnatrien-3,20-dion werden eingetragen und 75 Minuten bei Raumtemperatur gerührt. Das Reak tionsgemisch wird in 35 Liter Eiswasser, dem 250 ml Essig säure zugesetzt sind, eingerührt, der Niederschlag abgesaugt, mit Wasser gewaschen und getrocknet. Das erhaltene Roh produkt (185 g) schmilzt bei 222 bis 228'C-, UV: a2,6 = 11700.
Nach Umkristallisation aus Methanol-Methylenchlorid (2:1) erhält man 90,9 g 4-Chlor-17-hydroxy-1α,2α;6,7- dimethylen-4-pregnen-3,20-dion; F. 260 bis 265'C. UV: a2,6=12960.
74 g dieser Substanz werden in 740 ml Eisessig suspen diert, 139 ml Essigsäureanhydrid und 6,55 g p-Toluolsulfon- säure zugegeben und 17 Stunden bei Raumtemperatur unter Stickstoffatmosphäre gerührt. Man fällt das Reaktionsgemisch in 15 Liter Eiswasser, saugt den entstandenen Niederschlag ab, wäscht mit Wasser neutral und trocknet. Das erhaltene rohe 17-Acetat wird in 400 ml Essigester suspendiert, 30 Minuten unter Rückfluss erhitzt, gekühlt, abgesaugt, mit Essigester gewaschen und getrocknet. Man erhält 73,2 g 4-Chlor-17-acetoxy-1α,2α;6,7-dimethylen-4-pregnen-3,20- dion; F. 269 bis 272 C, UV: e275=12950; [α]D=-19,4 (Chloroform).
Durch wiederholte fraktionierte Umkristallisation aus Methylenchlorid/Diisopropyläther erhält man 4-Chlor-17- acetoxy-1α,2α;6ss,7ss-dimethylen-4-pregnen-3,20-dion vom Schmelzpunkt 279 bis 280 C, UV: a276=15250; [α]D= <B>-77'</B> (Chloroform).
Aus den eingeengten Mutterlaugen erhält man durch weitere fraktionierte Kristallisation aus Essigester 4-Chlor- 17-acetoxy-1α,2α;6α,7α-dimethylen-4-pregnen-3,20-dion vom Schmelzpunkt 264 bis<B>266'C.</B> UV: E268 =10400; [α]D=+233 (Chloroform). Beispiel 2 2 g 4-Chlor-17-acetoxy-1α,2α;6α,7α-dimethylen-4-pregnen- 3,20-dion werden in 40 ml Methanol und 1 ml Tetrahydro- furan suspendiert, in die Suspension wird eine Lösung von 1,6 g Natriumhydroxyd in 5 ml Wasser zugegeben und unter Stickstoff 17 Stunden bei Raumtemperatur gerührt.
Das Reaktionsgemisch wird in 800 ml Eiswasser, das 5 ml Essig säure enthält, eingerührt. Der entstandene Niederschlag wird auf einer Nutsche abgesaugt, mit Wasser neutral gewaschen und bei 50 C im Vakuum getrocknet. Man erhält 1,6 g 4-Chlor-17-hydroxy-1α,2α;6α,7α-dimethylen-4-pregnen- 3,20-dion. Die durch Umkristallisation aus Essigester er haltene reine Verbindung schmilzt bei 269 bis<B>271'C,</B> UV: ±269 = 10300; [a] =<B>+269,5'</B> (Chloroform). Beispiel 3 2,5 ml Capronsäureanhydrid und 100 mg p-Toluolsulfon- säure werden unter Feuchtigkeitsausschluss verrührt.
Unter Stickstoff und Rühren gibt man 1 g 4-Chlor-17-hydroxy- 1α,2α;6α,7α-dimethylen-4-pregnen-3,20-dion und 2,5 ml Capronsäure hinzu. Nach 4 Stunden Reaktionszeit bei Raum temperatur fällt man in Eiswasser, das 5 g Natriumhydrogen- carbonat enthält. Man saugt den erhaltenen Niederschlag ab, wäscht ihn mit Wasser neutral und trocknet ihn im Vakuum bei 40 C.
Durch Behandeln des rohen Capronates mit Di- isopropyläther entfernt man die noch anhaftende Capron- säure. Man erhält 1 g 4-Chlor-17-hexanoyloxy-1α,2α;6α,7α- dimethylen-4-pregnen-3,20-dion. Nach Umkristallisation aus Methanol schmilzt die Substanz bei 194 bis 195 C, UV: a268=10600; [a]D=+206,2 (Chloroform).
