Weiterhin können die Säuren auch ungesättigt, verzweigt, mehrbasisch oder in üblicher Weise, z. B. durch Hydroxyl-, Aminogruppen oder Halogenatome, substituiert sein.
Geeig net sind auch cycloaliphatische aromatische, gemischt aro matischaliphatische und heterocyclische Säuren, die ebenfalls in üblicher Weise substituiert sein können. Als bevorzugte Säuren seien beispielsweise genannt: Essigsäure, Propion- säure, Capronsäure, Önanthsäure, Undecylsäure, Trimethylessigsäure, Monochloressigsäure, Dichloressigsäure, ssig- Cyclopentylpropionsäure, Phenylpropionsäure, Pheny säure, Phenoxyessigsäure, Dialkylaminoessigsäure, Piperidino- essigsäure, Bernsteinsäure, Benzoesäure u. a.
Für die Her- stellung wasserlöslicher Präparate kommen insbesondere Ester anorganischer Säuren in Betracht, wie die der Schwefel- und der Phosphorsäure.
Die in 6,7-Stellung isomeren 4-Chlor-1a,2a;6ss,7ss-di- methylen-3-keto-4-pregnene sind bereits im Hauptpatent Nr. 518 272 beschrieben. Sie besitzen starke gestagene und ovulationshemmende Wirksamkeit Es wurde nun gefunden, dass die neuen 4-Chlor-1a,2a; 6a,7a-dimethylen-3-keto-4-pregnene der Formel I in ihrer ovulationshemmenden Wirksamkeit die isomeren 6,8,7ss- Methylenverbindungen des genannten Hauptpatents über treffen.
Die Ovulationshemmung wurde wie üblich durch Tuben inspektion ermittelt. In der nachfolgenden Tabelle wird jeweils der prozentuale Anteil der Rattenweibchen ange geben, bei denen nach subcutaner Applikation der genannten Tagesdosis jeweils eine Ovulation unterbleibt.

Aufgrund ihrer wertvollen pharmakologischen Eigen schaften können die neuen Wirkstoffe beispielsweise zur Konzeptionsverhütung und in der Gynäkologie Verwendung finden.
Das erfindungsgemässe Verfahren zur Herstellung von Steroiden der Formel I ist dadurch gekennzeichnet, dass man an die 1,2- und 6,7-Doppelbindung eines entsprechenden 3-Oxo-4-chlor 41,6,6-steroids Diazomethan anlagert, aus dem Zwischenprodukt die ankondensierten Pyrazolinringe spaltet und aus dem erhaltenen Isomerengemisch das 6a,7a- Isomere abscheidet.
Das Methylenierungsprodukt besteht aus einem Isomeren- gemisch von 6ss,7ss- und 6a,7a-Methylenverbindung, aus dem man das 6a,7a-Isomere durch die üblichen Trennungsver fahren isolieren kann. Die Abtrennung wird beispielsweise durch Chromatographieren und/oder fraktioniertes Umkri stallisieren erreicht.
Je nach der Bedeutung, die R im Endprodukt haben soll, kann gegebenenfalls vor oder nach der Isomerentrennung das Reaktionsprodukt einer Veresterung oder Verseifung unter worfen werden.
Die Veresterung und die Verseifung können nach den bekannten Methoden durchgeführt werden. Speziell genannt seien für die Veresterung das Umsetzen mit Säureanhydriden oder -chloriden in Gegenwart einer Säure oder der Umsatz der gewünschten Säure in Gegenwart von Trifluoressigsäure- anhydrid.
Die Verseifung wird beispielsweise mit Basen in alkoholi scher Lösung durchgeführt. Eine bevorzugte Methylenierung beruht darauf, dass man auf das ungesättigte Keton Diazo- methan einwirken lässt und aus der erhaltenen 1,2;6,7-Bis- (diazomethylen)-Verbindung thermisch oder katalytisch Stickstoff abspaltet. Die thermische Abspaltung wird gewöhn lich bei etwa 200 bis 250 C, vorzugsweise im Hochvakuum, vorgenommen. Die katalytische #yrazolinspaltung erfolgt in der Regel mit sauren Katalysatoren, wie Fluorborsäure, Bor- trifluoridätherat oder Perchlorsäure, in organischen Lösungs mitteln, wie z. B. Aceton, vorzugsweise bei Raumtemperatur.
Die Pyrazolinspaltung kann ausserdem mit hochsiedenden Basen wie Chinolin oder Anilin in der Siedehitze erfolgen. Beispiel 1 7,5 g 4-Chlor-17-acetoxy-1,4,6-pregnatrien-3,20-dion werden mit einer ätherischen Diazomethanlösung (hergestellt aus 75 g Nitrosomethylharnstoff, 550 ml Äther und 400 ml 40 %iger Kalilauge) versetzt und 7 Tage in einem geschlosse nen Gefäss bei Raumtemperatur stehengelassen. Danach wird das überschüssige Lösungsmittel im Vakuum vorsichtig abgezogen. Die als Rückstand verbleibende Bis-pyrazolin- Verbindung wird in 300 ml Aceton gelöst und unter Rühren tropfenweise mit 15 ml 70%iger Perchlorsäure versetzt.
Nach beendeter Gasentwicklung wird in die 10fache Menge Eis wasser eingerührt, der ausgefallene Niederschlag wird ab filtriert, mit Wasser gewaschen und getrocknet. Das erhaltene Rohprodukt wird in 40 ml Essigester suspendiert, 20 Minuten am Rückfluss erhitzt, gekühlt, abgesaugt, mit Essigester ge waschen und getrocknet. Man erhält 7,2 g 4-Chlor-17-acet- oxy-la,2a;6,7-dimethylen-4-pregnen-3,20-dion.
Durch wiederholte Umkristallisation aus Methylenchlorid/ Diisopropyläther erhält man 4-Chlor-17-acetoxy-1a,2a;6ss,7ss- dimethylen-4-pregnen-3,20-dion vom Schmelzpunkt 278 bis 280 C.
Aus den eingeengten Mutterlaugen erhält man durch weitere fraktionierte Kristallisation aus Essigester 4-Chlor- 17-acetoxy-la,2a;6a,7a-dimethylen-4-pregnen-3,20-dion vom Schmelzpunkt 263 bis 265'C. Beispiel 2 2 g 4-Chlor-17-acetoxy-la,2a;6a,7a-dimethylen-4- pregnen-3,20-dion werden in 40 ml Methanol und 1 ml Tetra- hydrofuran suspendiert, in die Suspension wird eine Lösung von 1,6 g Natriumhydroxyd in 5 ml Wasser zugegeben und unter Stickstoff 17 Stunden bei Raumtemperatur gerührt. Das Reaktionsgemisch wird in 800 ml Eiswasser, das 5 ml Essig säure enthält, eingerührt.
Der entstandene Niederschlag wird auf einer Nutsche abgesaugt, mit Wasser neutral gewaschen und bei 50 C im Vakuum getrocknet. Man erhält 1,6 g 4-Chlor-17-hydroxy-la,2a;6a,7a-dimethylen-4-pregnen- 3,20-dion. Die durch Umkristallisation aus Essigsäure erhal tene reine Verbindung schmilzt bei 269 bis 271 C, UV: E269 = 10300; [a]D = + 269,5 (Chloroform). Beispiel 3 2,5 ml Capronsäureanhydrid und 100 mg p-Toluolsulfon- säure werden unter Feuchtigkeitsausschluss verrührt. Unter Stickstoff und Rühren gibt man 1 g 4-Chlor-17-hydroxy- la,2a;6a,7a-dimethylen-4-pregnen-3,20-dion und 2,5 ml Capronsäure hinzu.
Nach 4 Stunden Reaktionszeit bei Raum temperatur fällt man in Eiswasser, das 5 g Natriumhydrogen- carbonat enthält. Man saugt den erhaltenen Niederschlag ab, wäscht ihn mit Wasser neutral und trocknet ihn im Vakuum bei 40 C. Durch Behandeln des rohen Capronates mit Diiso- propyläther entfernt man die noch anhaftende Capronsäure.
Man erhält 1 g 4-Chlor-17-hexanoyloxy-la,2a;6a,7a-dimethy- len-4-pregnen-3,20-dion. Nach Umkristallisation aus Metha nol schmilzt die Substanz bei 194 bis 195'C, UV: E268 = 10600; [a]D = + 206,2 (Chloroform).
