beschrieben, worin R1 und R2 Wasserstoff oder bestimmte, unten genannte, organische Reste und R3 eine Hydroxylgruppe bedeutet. Diese Verbindungen können als Ausgangsstoffe für die Herstellung von entsprechenden Verbindungen der Formel I verwendet werden, worin R3 eine N-substituierte Carbamoyloxygruppe bedeutet. Die vorliegende Patentschrift betrifft aiese therapeutisch wertvollen Carbamoyloxyderivate.
Gegenstand der vorliegenden Erfindung ist daher ein Verfahren zur Herstellung von Verbindungen der Formel I

worin Rl und R2 für Wasserstoff, Niederalkyl, monooder dicyclisches Aryl oder Aralkyl stehen und R4 ein aliphatischer, aromatischer, araliphatischer oder heterocyclischer Rest ist, und ihrer Salze.
Vorzugsweise stehen Rt und R2 für Wasserstoff oder wenigstens einer dieser beiden Substituenten für Wasserstoff und der andere für Niederalkyl, monocyclisches Aryl oder Arylniederalkyl. Die Aryl- und Aralkylreste können am Ring unsubstituiert oder substituiert sein, beispielsweise durch niedere Alkyl- oder Alkoxygruppen, die Nitrogruppe und/oder Halogenatome. Als Aralkylgruppe ist in erster Linie eine Benzylgruppe, wie der Benzylrest zu nennen.
R4 ist besonders ein unsubstituierter oder substituierter, vorzugsweise durch eine oder mehrere Niederalkoxygruppen oder Halogenatome substituierter, gerader oder verzweigter Niederalkylrest, wie der Methyl-, Äthyl-, vor allem aber der ss-Chloräthylrest.
Die Salze der neuen Verbindungen sind Metallsalze, vor allem solche von therapeutisch anwendbaren Alkali- oder Erdalkalimetallen, wie Natrium, Kalium, Ammonium, Calcium, oder Salze mit organischen Basen, z. B. Triäthylamin, N-Äthyl-piperidin, Dibenzyl äthylendiamin, Procain. Ist die Gruppe R4 basisch, so können sich innere Salze bilden.
Die neuen Verbindungen weisen eine besonders gute antibakterielle Wirkung auf. Sie sind sowohl gegenüber grampositiven wie vor allem auch gegenüber gramnegativen Bakterien wirksam, z. B. gegen Staphylococcus aureus penicillinresistent, Escherichia coli, Klebsiella pneumoniae, Salmonella typhosa und Bacterium proteus. Sie können daher zur Bekämpfung von Infektionen, die durch solche Mikroorganismen hervorgerufen werden, verwendet werden. Besonders wertvoll sind Verbindungen, in denen der Acylrest in 7-Stellung ein Cyanacetylrest und R4 die ss-Chloräthylcarbamoylgruppe ist.
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man eine Verbindung der Formel II

worin R3 und R2 die für Formel I genannte Bedeutung haben, oder ein Salz davon mit einem Isocyanat der Formel III R4 -N = C = 0 III , worin R4 einen aliphatischen, aromatischen, araliphatischen oder heterocyclischen Rest bedeutet, umsetzt.
Die erhaltenen Verbindungen mit freier Carboxylgruppe in 4-Stellung können in ihre Metall-, wie Alkali- oder Erdalkalimetallsalze oder Salze mit organischen Basen übergeführt werden.
Die Umsetzung erfolgt im allgemeinen in einem inerten Lösungsmittel für die Reaktionspartner, vorzugsweise in Gegenwart einer tertiären Base, insbesondere eines Triniederalkylamins, wie z. B. im Schweizer Patent 448 092 beschrieben.
Die Ausgangsstoffe können nach dem im Schweizer Patent Nr. 480 365 beschriebenen Verfahren hergestellt werden.
Die neuen Verbindungen können als Heilmittel, z. B. in Form pharmazeutischer Präparate, Verwendung finden. Diese enthalten die Verbindungen in Mischung mit einem für die enterale, topicale oder parenterale Applikation geeigneten pharmazeutischen organischen oder anorganischen, festen oder flüssigen Trägermaterial.
Im folgenden Beispiel sind die Temperaturen in Celsiusgraden angegeben.
Beispiel
14,07 g Triäthylammoniumsalz roher Desacetyl-7-cyanacetylamino-cephalosporansäure (I) werden in 140 ml frisch entgastem Dimethylformamid aufgenommen, mit 16 ml Tributylamin und 108 ml einer 100/oigen Lösung von ss-Chloräthylisocyanat in Dimethylformamid versetzt und 4 Stunden bei 220 C gerührt Man dampft den Ansatz bei 0,1 mm Hg ein.
Der Rückstand wird in 100/oigem wässrigem Dikaliumhydrogenphosphat aufgenommen und mit Essigester ausgeschüttelt. Die wässrige Phase wird mit konzentrierter Salzsäure auf pH 2,0 gestellt, mit Kochsalz gesättigt und mit Essigester extrahiert. Der über Natri umsulfat getrocknete Extrakt gibt beim Eindampfen nahezu reine O-Desacetyl-O-(ss-chloräthylcarbamoyl)- 7-cyanacetylamino-cephalosporansäure (II) der Formel

Das Produkt wird durch Chromatographie an Silicagel gereinigt. Die dabei mit Essigester eluierbare Substanz kristallisiert aus Aceton-Äther (1:2); F. 147-150 (Zersetzung).
Sie hat folgende Eigenschaften: UV-Absorptionsspektrum in 0,1-n. Natriumbicarbonat: man 260 mu (E = 9200); Dünnschichtchromatographie an Silicagel [System 1: n-Butanol-Eisessig (10:1) gesättigt mit Wasser; System 2: n-Butanol-Pyridin-Eisessig-Wasser (38:20:8:30)].

In Tabelle 3 ist die minimale Hemmkonzentration (in 3'/mol) von O-Desacetyl-0-(ss-chloräthylcarbamoyl)- 7-cyanacetylamino-cephalosporansäure (II) und Cephalosthin (III) gegenüber Bakterien (24-stündige, unverdünnte Kulturen) angegeben.

In Tabelle 4 ist die in vivo-Wirkung der obigen Verbindungen II und III bei mit Staph. aureus oder Esch. coli infizierten Mäusen dargestellt. Die Zahlen geben die Menge des Antibiotikums in mg/kg Maus an, die bei subkutaner Anwendung bewirken, dass 40-500/0 der Tiere überleben. Nicht mit Antibiotikum behandelte Tiere sterben innert 1-2 Tagen. Pro Versuchs reihe wurden 10 Mäuse verwendet.

Die als Ausgangsmaterial verwendete Desacetyl-7-cyanacetylamino-cephalosporansäure kann wie folgt hergestellt werden:
5,10 g (15 mMol) 7-Cyanacetylamino-cephalosporansäure werden in 102 ml destilliertem Wasser aufgeschlämmt und unter Rühren tropfenweise mit 15 mg n-Natronlauge in das Natriumsalz übergeführt. Die resultierende klare Lösung wird mit 2,55 g Zell-Lyophilisat von Bacillus subtilis, ATCC 6633 (vgl.
Schweizer Patent Nr. 475 284) 3 Stunden bei Raumtemperatur unter Konstanthaltung des pH bei 7,3 gerührt. Die bei der enzymatischen Verseifung freiwerdende Essigsäure wird mittels eines automatischen Titrators durch Zugabe von n-Natronlauge neutralisiert; Verbrauch 14,6 ml (97,40/0 der Theorie). Am Schluss der Verseifungsreaktion gibt man 130 ml Methylenchlorid zu, schüttelt kräftig und trennt die Emulsion durch Zentrifugieren. Die überstehende klare wässrige Phase enthält die Desacetyl-7-cyanacetylamino-cephalosporansäure als Natriumsalz in praktisch quantitativer Ausbeute, wie die dünnschichtchromatographische Analyse zeigt.
Zur Isolierung der freien Säure trennt man die wässrige Phase ab, sättigt sie mit Natriumchlorid, säuert sie nach Kühlung auf 0 mit 400/einer Phosphorsäure auf pH 2,1 an und schüttelt nochmals mit eiskaltem 1-Butanol aus. Die Auszüge werden im Rotationsverdampfer bei 0,1 mm Hg rasch eingedampft. Aus dem anfallenden Trockenrückstand wird die Desacetyl-7-cyanacetylamino-cephalosporansäure durch Herauslösen mit Essigester von anorganischen Salzen getrennt.

Table 4 shows the in vivo activity of the above compounds II and III when used with Staph. aureus or Esch. coli infected mice. The deacetyl-7-cyanoacetylamino-cephalosporanic acid used as starting material can be prepared as follows:
