worin X Halogen mit einem Atomgewicht nicht grösser als 81 bedeutet, das dadurch gekennzeichnet ist, dass man an die 1,2 und 6,7-Doppelbindung eines entsprechenden 4-Halogen- d1,4,6-3-ketosteroids Diazomethan anlagert und aus dem entstandenen Zwischenprodukt die ankondensierten Pyrazo- linringe spaltet.
Die Durchführbarkeit des erfindungsgemässen Verfahrens war deshalb höchst überraschend, weil in 4-Stellung unsub- stituierte d1,4,6-3-Ketosteroide (X = H) nur an der d 1- Doppelbindung zu den entsprechenden 1,2-Methylenverbin- dungen reagieren, und das selbst bei grossen Überschüssen der methylenierenden Reagenzien (DBP 1 072 991, DAS 353, DBP 1 183 500).
Es war daher auch vom Fach mann nicht vorauszusehen, dass d1,4,6-3-Ketosteroide, die am Kohlenstoffatom C-4 durch Halogen, insbesondere durch Chlor oder Brom, substituiert sind (X = Halogen), in 1,2- und 6,7-Stellung gleichzeitig methyleniert werden. 1,2α;6,7ss- Bismethylenverbindungen sind bisher noch nicht beschrieben worden.
Die erfindungsgemässe Methylenierung besteht, wie ge sagt, Inder Anlagerung von Diazomethan an die 41- und 46-Doppelbindung und der nachträglichen Spaltung der Pyrazolinringe, die thermisch oder katalytisch erfolgen kann. Die thermische Spaltung wird vorzugsweise bei 220 bis 250 C und zweckmässigerweise im Hochvakuum und in Ge genwart eines inerten Schutzgases, wie Stickstoff, vorgenom men. Die katalytische Spaltung erfolgt insbesondere durch luorborsäure, Bortri- Einwirkung saurer Katalysatoren, wie Fluorborsäure, Bortri- fluoridätherat oder vor allem Perchlorsäure in organischen Lösungsmitteln, beispielsweise in Aceton, vorzugsweise bei Raumtemperatur.
Ausserdem kann die Spaltung durch Ver wendung hochsiedender Basen, wie zum Beispiel Chinolin oder Anilin, erfolgen.
Die als Ausgangsstoffe benutzten Steroide können an den Ringen C und D beispielsweise durch Halogen, Alkyl-, Hy- droxyl-, Acyl-, Alkyloxy- und/oder Acyloxygruppen substi tuiert sein, sie können Ketogruppen und/oder Doppelbin dungen enthalten, es dürfen in ihnen jedoch keine ungesättig ten Ketongruppierungen auftreten.
Die erfindungsgemäss herstellbaren 1,2α;6,7ss-Bismethy- lensteroide sollen entweder aufgrund ihrer wertvollen pharmakologischen Eigenschaften als Arzneimittel oder auf grund ihrer spezifischen Reaktionsfähigkeit als neuartige Aus gangsstoffe zur Herstellung wertvoller Hormonsubstanzen dienen. Beispielsweise besitzen die Bismethylenderivate der Hydroxyprogesteronester überraschend starke gestagene und ovulationshemmende Wirksamkeit bei nur geringer anti- androgener Nebenwirkung.
Die folgenden Tabellen zeigen die Überlegenheit der neuen Verbindungen am Beispiel des 4-Chlor-1,2α;6,7ss-bismethylen-44-pregnen-17α-ol-3,20- dion-17-acetats (III) im Vergleich mit den bekannten Ge- stagenen 6-Chlor-44,6-pregnadien-17α-ol-3,20-dion-17- acetat (I) und 17α-Äthinyl-d4-östren-17ss-ol-3-on-17-acetat (II).
Die gestagene Wirkung wurde im bekannten Clauberg- Test geprüft. In Tabelle I wird jeweils die zur Erzielung eines positiven Effekts nötige Mindestmenge (Schwellenwert) wiedergegeben.

0 Aus den Zusammenstellungen geht deutlich hervor, dass die neue Bismethylenverbindung III mindestens 3-bis 10mal stärker wirkt als die bekannten Gestagene I und Il.<B>35</B> Beispiel 1 1,5 g 4-Chlor -A1,4,6-androstatrien-17ss-ol-3-on-17-acetat werden mit einer ätherischen Diazomethanlösung (herge stellt aus 15 g Nitrosomethylharnstoff, 110 ml Äther und 80 ml 40%iger Kalilauge) versetzt und 7 Tage in einem ge schlossenen Gefäss bei Raumtemperatur stehengelassen. Da nach wird das überschüssige Lösungsmittel im Vakuum vor sichtig abgezogen. Die als Rückstand verbleibende Bis-pyra- zolin-Verbindung wird in 60 ml Aceton gelöst und unter Rüh ren tropfenweise mit 3 ml 70%iger Perchlorsäure versetzt.
Nach beendeter Gasentwicklung wird in die 10fache Menge Eiswasser eingerührt, der ausgefallene Niederschlag abfil triert, in Methylenchlorid aufgenommen, mit Wasser gewa schen und über Natriumsulfat getrocknet. Nach dem Ein dampfen zur Trockne wird der Rückstand an Silicagel chro- matographiert. Man erhält 350 mg 4-Chlor-1,2α;6,7ss-bisme- thylen-d4-androsten-17ss-ol-3-on-17-acetat vom Schmelz punkt 181,5 bis 182 C, nach Umkristallisieren aus Isopro- pyläther steigt der Schmelzpunkt auf 191,5 bis 193' C.
UV: 62,2 = 11800.
Beispiel 2 5,5 g 4-Chlor d1,4,6-pregnatrien-17α-ol-3,20-dion-17- acetat werden, wie in Beispiel 1 beschrieben, mit der ent sprechenden Menge ätherischer Diazomethanlösung zur Bis- pyrazolinverbindung umgesetzt. Nach saurer Spaltung und an- schliessender Chromatographie werden, umkristallisiert aus Essigester, 1,1 g 4-Chlor-1,2α;6,7ss-bismethylen-d4-pregnen- 17α-ol-3,20-dion-17-acetat vom Schmelzpunkt 277 bis 278 C (Zersetzung) erhalten, UV: E274 = 12000.
Beispiel 3 1,4 g 4-Brom -A 1,4,6-pregnatrien-17α-ol-3,20-dion-17- acetat werden wie in Beispiel 1 beschrieben mit ätherischer Diazomethanlösung umgesetzt, und die entstandene Bispy- razolinverbindung wird sauer gespalten. Das isolierte rohe 4 Brom-1,2a;6,7ss-bismethylen-d4-pregnen-17α-ol-3,20-dion- 17-acetat wird durch Chromatographie an Silicagel und Um kristallisieren gereinigt.
Beispiel 4 3 g 4-Chlor -A1,4,6-pregnatrien-17α-ol-3,20-dion-17- capronat werden analog Beispiel 1 umgesetzt und aufgear beitet. Man erhält 735 mg 4-Chlor-1,2α;6,7ss-bismethylen- d4-pregnen-17α-ol-3,20-dion-17-capronat als Öl.
Beispiel s Zu einer Lösung von 3,2 g 17,20;20,21-Bismethylen- dioxyd-d1,4,6-pregnatrien-3,11-dion in 50 ml Dimethyl- formamid wird bei -30 C innerhalb von 10 Minuten eine Lösung von 565 mg Chlor in 10,5 ml Proprionsäure unter Rühren langsam zugetropft. Das Reaktionsgemisch bleibt dann 2 Stunden bei -30 C stehen und wird anschliessend in Eiswasser gegeben. Der ausgefallene Niederschlag wird mit Methylenchlorid extrahiert, der Extrakt nacheinander mit Natriumsulfat-, Natriumhydrogencarbonatlösung und Wasser gewaschen und über Natriumsulfat getrocknet.
Nach Umkri stallisieren des Rohproduktes aus Essigester erhält man 1,3 g 4-Chlor-17,20;20,21-bismethylendioxy-d1,4,6-pregnatrien- 3,11-dion vom Schmelzpunkt 247 bis 249 C (Zersetzung).
Die Umsetzung des so erhaltenen d l#4,6-Steroids mit Diazomethan und Spaltung der Pyrazolinringe erfolgt analog Beispiel 1.
