Verfahren zur Herstellung neuer Furanoside
Die vorliegende Erfindung betrifft die Herstellung von D-Glucofuranosiden der Formel I

worin Rz einen gegebenenfalls Hydroxy- oder Niederalkoxy gruppen aufweisenden niederaliphatischen Kohlenwasser stoffrest, einen gegebenenfalls Niederalkylgruppen aufwei senden cycloaliphatischen Kohlenwasserstoffrest oder einen, im Phenylring gegebenenfalls substituierten Benzylrest dar stellt, R2 für den Acylrest einer organischen Carbonsäure steht, R3 Wasserstoff oder einen niederaliphatischen Kohlenwasserstoffrest darstellt, und jeder der Reste Rs und R6 einen, im Phenylring gegebenenfalls substituierten Benzylrest bedeutet, mit der Massgabe, dass, wenn R3 für einen nieder aliphatischen Kohlenwasserstoffrest steht, die Gruppen R1 und R3 zusammen mindestens 3 Kohlenstoffatome enthalten,
sowie von Salzen von eine salzbildende Gruppe aufweisen den Verbindungen der obigen Art.
Die obigen Verbindungen haben die Konfiguration der D-Glucofuranose; die verätherte Hydroxygruppe in 1-Stellung kann die a- oder die ss-Konfiguration einnehmen, und die erfindungsgemässen Verbindungen können in Form von reinen Anomeren oder als Anomerengemische vorliegen.
Den Rest R1 darstellende niederaliphatische Kohlenwasserstoffreste sind in erster Linie Niederalkylgruppen, können aber auch Niederalkenylgruppen sein; diese Reste, insbesondere eine Niederalkylgruppe, können eine, zwei oder mehrere Hydroxy- oder Niederalkoxygruppen als Substituenten ent halten. Cycloaliphatische Kohlenwasserstoffreste sind in erster Linie Cycloalkylgruppen, z.B. mit 38, vorzugsweise mit 5-6 Ringkohlenstoffatomen, sowie Cycloalkenylgruppen, z. B. mit 5-8, vorzugsweise mit 5-6 Ringkohlenstoffatomen; cycloaliphatische Kohlenwasserstoffreste können gegebenen falls Niederalkylgruppen als Substituenten enthalten. Substi tuenten des Phenylrings in gegebenenfalls substituierten Ben zylgruppen sind z.B.
Niederalkyl- und/oder verätherte oder veresterte Hydroxygruppen, wie Niederalkoxy- oder Nieder alkylendioxygruppen und/oder Halogenatome, sowie Pseudohalogen -gruppen, wie Trifluormethylgruppen.
Ein Acylrest R2 einer organischen Carbonsäure ist vorzugsweise der Rest einer aliphatischen, aromatischen oder araliphatischen Carbonsäure, wie einer Niederalkan- oder Niederalkencarbonsäure, z. B. Essig- oder Propionsäure, und insbesondere einer Niederalkandicarbonsäure, z.B. mit 2-7, vorzugsweise 3-6 Kohlenstoffatomen, sowie einer äquivalenten Niederalkendicarbonsäure, z. B. mit 4-7 Kohlenstoff atomen; solche Säuren sind z.B. Malon-, 2-Methyl-bernstein-, Glutar-, 3-Methylglutar-, 3-Äthylglutar-, Adipin- oder Pimelinsäure, in erster Linie Bernsteinsäure, sowie Malein oder Fumarsäure.
Ein niederaliphatischer Kohlenwasserstoffrest R3 ist in erster Linie ein Niederalkyl- oder Niederalkenylrest.
Ein gegebenenfalls im Phenylkern substituierter Benzylrest R5 oder R6 enthält als Substituenten die oben angeführten Gruppen und/oder Atome.
Vorstehend, wie nachfolgend mit dem Ausdruck nieder modifizierte Reste, Radikale oder Verbindungen enthalten in erster Linie bis zu 7, vorzugsweise bis zu 4 Kohlenstoffatome.
Niederalkylreste sind z.B. Methyl-, Äthyl-, n-Propyl-, Isopropyl-, n-Butyl-, Isobutyl-, sek.-Butyl-, tert.-Butyl-, n-Pentyl-, Isopentyl-, Neopentyl-, n-Hexyl-, Isohexyl-, n-Heptyl- oder Isoheptylreste, während Niederalkenylreste z.B. Allyl-, Methallyl- oder 2-Butenylreste bedeuten. Als Substituenten von solchen niederaliphatischen Kohlenwasserstoff-, insbesondere Niederalkylresten auftretende Niederalkoxygruppen sind z.B. Methoxy-, Äthoxy-, n-Propyloxy-, Isopropyloxy-, n-Butyloxy-, Isobutyloxy-, n-Pentyloxy oder n-Hexyloxygruppen zu nennen.
Hydroxy- und Niederalkoxy-substituierte niederaliphatische Kohlenwasserstoffreste sind in erster Linie Hydroxy- oder Niederalkoxy-niederalkylreste, in welchen die Hydroxy- oder Niederalkoxygruppen vorzugsweise durch mindestens 2 Kohlenstoffatome vom Sauerstoffatom, welches den in solcher Weise substituierten niederaliphatischen Rest trägt, getrennt sind, wie Hydroxymethyl-, 2-Hydroxyäthyl-, 2-Hydroxypropyl-, 3 -Hydroxypropyl-, 2-Methoxyäthyl-, 2-Äthoxyäthyl-, 2-Methoxypropyl-, 3-Methoxypropyl- oder 3-Äthoxypropylreste.
Cycloalkylreste sind z.B. gegebenenfalls durch Niederalkyl-, in erster LinieMethylgruppen, mono- oder polysubstituierte Cyclopropyl-, Cyclobptyl-, Cyclopentyl-, Cyclohexyloder Cyclohetpylreste, während es sich bei Cycloalkenylresten z.B. um gegebenenfalls entsprechend substituierte 2- oder 3-Cyclopentenyl-, 2- oder 3-Cyclohexenyl- oder 2-, 3- oder 4-Cycloheptenylreste handelt.
Benzylreste können im Phenylkern mono-, di- oder polysubstituiert sein, wobei bei Mehrfach substitution verschiedenartige Substituenten vorhanden sein können. Halogenatome sind in erster Linie solche mit einem Atomgewicht von 19 bis 80, d. h. Fluor-, Chlor- oder Bromatome. Im Kern monosubstituierte Benzylreste enthalten einen Substituenten, vorzugsweise in p-Stellung.
Die neuen Verbindungen besitzen wertvolle pharmakologische Eigenschaften. So weisen sie insbesondere eine antiinflammatorische Wirkung auf, wie sich im Tierversuch, z. B.
an der Ratte, zeigt. Ferner besitzen sie, z. B. im Tierversuch, wie z. B. an Meerschweinchen, eine antiallergische Wirkung.
Die neuen Verbindungen können daher als Antiphlogistika Verwendung finden. Die neuen Verbindungen sind aber auch wertvolle Zwischenprodukte für die Herstellung anderer nützlicher Stoffe, insbesondere von pharmakologisch wirk samen Verbindungen.

worin R'1 für einen Niederalkylrest steht, R'2 den Acylrest einer Niederalkandicarbonsäure, insbesondere den Succinylrest, bedeutet, R'3 ein Wasserstoffatom, einen Niederalkyloder den Allylrest darstellt, und jede der Gruppen R's und R'6 eine gegebenenfalls einen Niederalkyl-, insbesondere Methylrest, oder ein Halogen-, insbesondere ein Chloratom, vorzugsweise in p-Stellung, aufweisende Benzylgruppe darstellt, mit der Massgabe, dass, wenn R'3 einen Niederalkylrest darstellt, R'1 und Rt3 zusammen mindestens 3 Kohlenstoffatome enthalten, sowie Salze, insbesondere nichttoxi- sche Ammonium-, Alkalimetall- oder Erdalkalimetallsalze von Verbindungen,
in welchen R'2 den Acylrest einer Niederalkandicarbonsäure, insbesondere einen Succinylrest, darstellt.
Besonders ausgeprägte pharmakologische Wirkung der obigen Art zeigen die Mederalkyl-3-O-R"3-5,6-di-O-R-D- glucofuranoside, worin R"3 ein Wasserstoffatom oder eine Niederalkylgruppe darstellt und R für einen gegebenenfalls durch Halogen-, insbesondere Chloratome, vorzugsweise in 4-Stellung, substituierten Benzylrest steht, mit der Massgabe, dass, wenn R"3 für einen Niederalkylrest steht, diese zu sammen-mit dem Niederalkylrest des Substituenten in 1-Stellung mindestens 3 Kohlenstoffatome enthält, sowie deren 2-O-Succinylverbindungen und ihre Salze, z.B.
solche der obigen Art, und insbesondere das Äthyl-3-O-n-propyl 5,6-di-O-(4-chlorbenzyl)-D-glucofuranosid, sowie dessen 2-O-Succinylverbindung und ihre Salze, insbesondere solche der obigen Art, die an der Ratte bei intraperitonealer Gabe in Dosen von 0,03 g/kg bis 0,3 g/kg eine ausgesprochene antünflarumatorische Wirkung aufweisen.
Die erfindungsgemässen Verbindungen werden hergestellt, indem man eine D-Glucofuranose der Formel II

worin R3, Rs und R6 die oben gegebene-Bedeutung haben und R"1 eine reaktionsfähige veresterte Hydroxygruppe und R 2 einen Acyloxyrest, worin Acyl den Rest einer organischen Carbonsäure bedeutet, mit einem Metallderivat einer Verbindung der Formel R1-OH, worin R1 die oben gegebene Bedeutung hat, umsetzt.
Wenn erwünscht, kann in einer erhaltenen Verbindung ein ungesättigter niederaliphatischer Kohlenwasserstoffrest gesättigt, oder, wenn erwünscht, in einer erhaltenen Verbindung ein Acyloxyrest in 2-Stellung in eine freie Hydroxyoder in eine andere Acyloxygruppe übergeführt werden.
Eine reaktionsfähige veresterte Hydroxygruppe ist in erster Linie eine durch eine Halogenwasserstoffsäure veresterte Hydroxygruppe und R 1 steht daher in erster Linie für ein Halogen-, insbesondere ein Bromatom. Ein Metallderivat eines alkoholischen Reagens R1-OH ist z. B. die entsprechende Alkalimetall-, z.B. Natrium- oder Kalium-, oder Erdalkalimetall-, z. B. Magnesium- oder eine Silberverbindung. Die obige Reaktion wird vorzugsweise in Gegenwart eines Lösungsmittels durchgeführt, wobei das alkoholische Reagens ebenfalls als solches verwendet werden kann.
In einer erfindungsgemäss erhältlichen Verbindung kann ein ungesättigter niederaliphatischer Kohlenwasserstoffrest, wie ein Niederalkenyl-, z.B. Allylrest, z.B. durch Behandeln mit katalytisch aktiviertem Wasserstoff, z. B. Wasserstoff in Gegenwart eines Palladiumkatalysators, gesättigt werden.
In erfndungsgetnässen Verbindungen, in welchen R2 einen Acylrest darstellt, kann dieser z.B. durch Behandeln mit einem alkalischen Mittel, wie einem Erdalkali- oder Alkalimetallhydroxid oder -carbonat, oder Silberoxid, in wässrigem oder alkoholischem Medium durch Wasserstoff ersetzt oder durch Abwandeln von im Acylrest vorhandenen in andere Gruppen, wie z; B. einer Halogencarbonyl- in die freie Carboxygruppe, umgewandelt werden.
Verbindungen mit einem, salzbildende Gruppen, wie z. B. freie Carboxylgruppen, enthaltenden Acylrest R2 werden je nach Reaktionsbedingungen in freier Form oder in Form von Salzen erhalten, welche Formen in an sich bekannter Weise ineinander überführbar sind. Salze von Verbindun gen mit freier Carboxylgruppe sind z.B. Metallsalze, insbesondere Alkalimetall-, z.B. Natrium- oder Kaliumsalze, sowie Erdalkalimetall-, z. B. Magnesium- oder Calciumsalze, oder Ammoniumsalze, z. B. solche mit Ammoniak oder organischen Basen, wie Trialkylaminen, z.B. Trimethylamin oder Triäthylamin, insbesondere die nichttoxischen Salze der obigen Art. Solche Salze, besonders die Alkalimetallsalze der neuen Verbindungen, zeichnen sich durch eine gute Wasserlöslichkeit aus; Salze können auch zur Reinigung der freien Verbindungen dienen. Man erhält sie z.
B. durch Behandeln mit Metallhydroxyden oder -carbonaten oder mit Ammoniak oder Aminen, sowie mit geeigneten Ionenaustauschern.
Infolge der engen Beziehungen zwischen den neuen Verbindungen in freier Form und in Form ihrer Salze sind im vorausgegangenen und nachfolgend unter den freien Verbindungen oder den Salzen sinn- und zweckmässig gegebenenfalls auch die entsprechenden Salze bzw. freien Verbindungen zu verstehen.
Die neuen Verbindungen können als reine Anomere oder als Anomerengemische vorliegen. Letztere können auf Grund der physikalisch-chemischen Unterschiede der Bestandteile in bekannter Weise in die beiden reinen Anomeren aufgetrennt werden, beispielsweise durch Chromatographie und/ oder fraktionierte Kristallisation. Vorzugsweise isoliert man das wirksamere der beiden Anomeren.
Die oben beschriebenen Verfahren werden nach an sich bekannten Methoden durchgeführt, in Abwesenheit oder vorzugsweise in Anwesenheit von Verdünnungs- oder Lösungsmitteln, wenn notwendig, unter Kühlen oder Erhitzen, in einem geschlossenen Gefäss und/oder in einer Inertgas-, wie Stickstoffatmosphäre.
Die Erfindung betrifft auch diejenigen Ausführungsformen des Verfahrens, bei denen man einen Ausgangsstoff in Form eines unter den Reaktionsbedingungen erhältlichen rohen Gemisches oder in Form eines reaktionsfähigen Derivats einsetzt.
Die Ausgangsstoffe sind bekannt oder, wenn neu, können in an sich bekannter Weise hergestellt werden. So kann man sie z.B. erhalten, indem man in einer D-Glucofuranose die Hydroxygruppen in Stellungen 1 und 2 und, wenn erwünscht, in Stellungen 5 und 6 durch Einführen von Schutzgruppen, z. B. der Gruppe X, die insbesondere für eine Isopropyliden-, aber auch für eine Benzylidengruppe steht, abschirmt. Die Hydroxygruppe in 3-Stellung kann dann durch Behandeln mit einem reaktionsfähigen Ester einer Verbindung der Formel R3-OH, wie z. B. einem niederaliphatischen R3-Halogenid, z.B. R3-Chlorid oder R3-Bromid, sowie einer entsprechenden R3-Sulfonyloxyverbindung in.Gegenwart eines basischen Mittels, wie eines Alkalimetallhydroxids, z.B.
Natrium- oder Kaliumhydroxids, oder eines Alkalimetallcarbonats, z.B. Natrium- oder Kaliumcarbonats, veräthert werden. Diese Verätherung mit einem reaktionsfähigen Ester einer Verbindung der Formel R3-OH kann unter geeigneten Bedingungen, z.B. in Gegenwart von Silberoxid oder eines Alkalimetall-, z.B. Kaliumcarbonats, auch an der 3,5,6 Trihydroxyverbindung durchgeführt werden. Im so erhältlichen Zwischenprodukt können die Hydroxygruppen in 5und 6-Stellung selektiv, d.h. ohne Freisetzen der Hydroxygruppen in 1- und 2-Stellung, z.B. durch Behandeln mit einer Säure, wie 60%iger wässriger Essigsäure (z. B. bei 35 ) oder wässriger äthanolischer Salzsäure, freigesetzt und dann ihrerseits durch eine gegebenenfalls substituierte Benzylgruppe, z.
B. unter Verwenden von reaktionsfähigen veresterten, gegebenenfalls susbstituierten Benzylalkoholen, wie den entsprechenden Halogeniden, z. B. Chloriden oder Bromiden, sowie Sulfonyloxy-, z.B. p-Toluolsulfonyloxyverbindungen, in Gegenwart von basischen Mitteln, wie eines Alkalimetall-, z.B. Kaliumhydroxids, veräthert werden. Dieser Schritt kann auch stufenweise durchgeführt werden, indem sich die primäre Hydroxygruppe in 6-Stellung, z. B. beim Behandeln mit einer etwa äquivalenten Menge eines reaktionsfähigen Esters der Verbindung der Formel R6-OH in Gegenwart einer etwa äquivalenten Menge eines Alkalimetallhydroxids oder in Gegenwart von Silberoxid vor derjenigen in 5-Stellung, veräthern lässt.
Man kann auch in einer 5,6-Dihydroxyverbindung, die in 3-Stellung eine freie oder vorzugsweise eine verätherte Hydroxygruppe enthält, selektiv die 6-Hydroxygruppe, z.B. durch Behandeln mit einem geeigneten organischen Sulfonsäurehalogenid, wie p-Toluolsulfonylchlorid, verestern und durch Behandeln mit einem geeigneten basischen Mittel, wie einem Alkalimetall-niederalkoxid, wie Natriumäthoxid, die 5,6-Epoxyverbindung bilden; durch Aufspalten des Epoxids mittels eines Alkohols der Formel R6-OH in Gegenwart eines Umesterungskatalysators, z.B. einer Alkalimetall-, wie Natriumverbindung eines Alkohols der Formel R6-OH, erhält man die 5-Hydroxy-6 R6-O-Verbindung, in welcher sich in üblicher Weise die freie Hydroxygruppe in 5-Stellung selektiv, z.
B. durch Behandeln mit einem reaktionsfähigen Ester einer Verbindung der Formel R5-OH, in Gegenwart eines basischen Mittels, veräthern lässt.
In den nach den obgenannten Verfahren erhältlichen Ausgangsstoffen, in welchen R3 vorzugsweise für einen niederaliphatischen Rest steht, kann die Schutzgruppe für die beiden Hydroxygruppen in 1- und 2-Stellung durch Behandeln z.B. mit wässriger Salzsäure geöffnet werden. Die beiden Hydroxygruppen können dann durch Behandeln mit einem geeigneten Derivat einer organischen Carbonsäure, wie Essigsäure, z.B. einem Anhydrid, wie Essigsäureanhydrid, unter Bildung einer 1,2-Bis-O-acylverbindung, insbesondere 1,2-Bis-O-acetylverbindung, verestert werden; beim Umsetzen z. B. mit Brom in Eisessig erhält man eine als Ausgangsmaterial verwendbare 2-O-Acyl-, z.B. 2-O-Acetylverbin- dung, mit einer reaktionsfähigen veresterten Hydroxygruppe in 1-Stellung.
Auf irgendeiner geeigneten Stufe der oben beschriebenen Verfahren zur Herstellung der Ausgangsstoffe kann eine in 3-Stellung durch einen geeigneten 2-Alkenyl-, wie den Allylrest verätherte Hydroxygruppe, z. B. durch Umlagern der Doppelbindung mittels Behandeln mit einer geeigneten Base, wie einem Alkalimetall-, z.B. Kalium-tert.-butoxid, vorzugsweise in einem geeigneten Lösungsmittel, wie z.B.
Dimethylsulfoxid, und oxydativ-hydrolytischer Entfernung der 1-Niederalkenyl-, wie 1-Propenylgruppe, z. B. durch Behandeln mit Kaliumpermanganat, vorzugsweise in basischem Medium, wie äthanolischem Alkalimetallhydroxid, z.B.
Kaliumhydroxid, freigesetzt werden.
Die neuen Verbindungen oder ihre Salze können als Heilmittel, z.B. in Form pharmazeutischer Präparate, Verwendung finden, welche sie in Mischung mit einem für die Verabreichung, z.B. die enterale oder parenterale, sowie topicale Applikation geeigneten pharmazeutischen organischen oder anorganischen, festen oder flüssigen Trägermaterial enthalten. Für die Bildung desselben kommen solche Stoffe in Frage, die gegenüber den neuen Verbindungen inert sind, wie Wasser, Gelatine, Zucker, z.B.
Milchzucker, Glucose oder Fruktose, Stärken, wie Mais-, Weizen- oder Reisstärke, Stearinsäure oder Salze davon, wie Calciumoder Magnesiumstearat, Talk, pflanzliche Öle, Benzylalkohole, Gummi, Polyalkylenglykole, Propylenglykol oder andere bekannte Arzneimittelträger. Die pharmazeutischen Präparate können z.B. als Tabletten, Dragees, Kapseln oder in flüssiger Form als Lösungen, Suspensionen oder Emulsionen vorliegen. Gegebenenfalls sind sie sterilisiert und bzw.
oder enthalten Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer. Sie können auch noch andere therapeutisch wertvolle Stoffe enthalten. Die Präpa rate werden nach üblichen Methoden gewonnen.
Die Erfindung wird in den nachfolgenden Beispielen näher beschrieben. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel 1
Zu 2,4 g 1,2-Di-O-acetyl-3-O-n-propyl-5,6-di-O-(4 chlorbenzyl)-D-glucofuranose werden unter Eiskühlen 5 ml einer 33 %igen Lösung von Bromwasserstoff in Eisessig gegeben und das Gemisch während 21/2 Stunden bei Zimmertemperatur stehengelassen. Dann wird auf Eiswasser ausgegossen und mit Chloroform extrahiert. Der organische Extrakt wird mit Eiswasser und einer eiskalten wässrigen Natriumhydrogencarbonatlösung gewaschen, über Natriumsulfat getrocknet und unter vermindertem Druck bei einer Badtemperatur von 30-40 eingeengt.
Der Rückstand, enthaltend das 2-O-Acetyl-3-O-n-propyl-5,6-di-O-(4-chlorbenzyl)-D-glucofuranosyl-bromid, wird mit 50 ml einer 1n äthanolischen Lösung von Natrium äthanolat vermischt und während 16 Stunden bei Zimmertemperatur stehen gelassen, dann mit 2n wässriger Essigsäure neutralisiert und mit Chloroform extrahiert. Der organische Extrakt wird mit Eiswasser und einer eiskalten konzentrierten wässrigen Natriumhydrogencarbonatlösung gewaschen, über Natriumsulfat getrocknet und unter Wasserstrahlvakuum eingeengt. Der Rückstand wird unter Hochvakuum destilliert, wobei das Äthyl-3 -O-n-propyl-5 ,6-di-O-(4-chlor- benzyl)-D-glucofuranosid als schwach gelbes Öl bei 195 / 0,01 mm Hg erhalten wird; [a]D20 = - 17 f 10 (c = 1 in Chlo- roform).
Durch Elution mit einem 85:15-Gemisch von Chloroform und Essigsäureäthylester lässt sich das Anomerengemisch in die reinen Anomeren dünnschichtchromatographisch an Silicagel (RF 254 der Firma Merck, Darmstadt) auftrennen.
Das a-Anomere hat einen Rf-Wert von 0,50; [a]D20 = + 24 + 1" (c = 1 in Chloroform); und das ss-Anomere einen Rf-Wert von 0,24; [a]D20 = 45+10 (c = 1 in Chloroform).
Das verwendete Ausgangsmaterial kann wie folgt hergestellt werden:
Eine Lösung von 82 g 1,2-O-Isopropyliden-3-0-allyla-D-glucofuranose in 800 ml Äthanol wird in Gegenwart von 1 g eines 10%gen Palladium-auf-Kohle-Katalysators hydriert. Der Katalysator wird abfiltriert, das Filtrat eingedampft und der Rückstand am Hochvakuum entgast; man erhält so die 1 ,2-O-Isopropyliden-3 -O-n-propyl-a-D -gluco- furanose; [a]D20=49f 1" (c= in Chloroform); die ohne Reinigung weiterverarbeitet wird.
Eine Lösung von 32 g 1,2-O-Isopropyliden-3-O-n-propyla-D-glucofuranose in 100 ml absolutem Dioxan wird mit 87,5 g pulverisiertem Kaliumhydroxid versetzt. Unter Rühren wird dann eine Lösung von 131 g 4-Chlorbenzylchlorid in 50 ml absolutem Dioxan innerhalb von 30 Minuten zugetropft; dann lässt man während 5 Stunden bei 80" reagieren.
Das überschüssige 4-Chlorbenzylchlorid wird mit Wasserdampf abdestilliert und nach dem Abkühlen wird mit Chloroform extrahiert. Der organische Extrakt wird mit Wasser gewaschen, über Natriumsulfat getrocknet und unter vermindertem Druck eingedampft. Die 1,2-O-Isopropyliden-3-0-n- propyl-5 , 6-di-(4-chlorbenzyl)-a-D-glucofuranose wird durch Destillieren des Rückstandes als schwach gelbes Öl bei 230"/0,015 mm Hg erhalten; [a]D20 =-22+1 (c = 1 in Chloroform).
Eine Suspension von 5 g 1,2-O-Isopropyliden-3-O-n- propyl-5,6-di-O-(4-chlorbenzyl)-a-D-glucofuranose in einem Gemisch von 35 ml Eisessig und 17,5 ml 1n wässriger Schwefelsäure wird unter intensivem Rühren und in einer Stickstoffatmosphäre während 30 Minuten bei einer Innentemperatur von 80" erwärmt. Das Reaktionsgemisch wird auf 10 gekühlt, mit 2n wässriger Natriumhydroxidlösung auf pH 23 gestellt und anschliessend bei einer Badtemperatur von 35 eingedampft Das Konzentrat wird mit Äther extrahiert, die organische Phase mit einer gesättigten wässrigen Natriumhydrogencarbonatlösung und mit Wasser neutral gewaschen, über Magnesiumsulfat getrocknet und eingedampft.
Der Rückstand entspricht der 3-O-n-Propyl-5,6-di-0-(4- chlorbenzyl)-D-glucofuranose, die im Dünnschichtchromatogramm (Silicagel) im System Chloroform : Essigsäureäthylester (85: 15) einen Rf-Wert von 0,12 aufweist; F. 60 nach Umkristallisieren aus einem 1 :3-Gemisch aus Toluol und Hexan und Abkühlen auf -17".
Man kann die 3 -O-n-Propyl-5,6-di-O-(4-chlorbenzyl)- D-glucofuranose auch erhalten, indem man 15 g Äthyl-3-O n-propyl-5 ,6-di-O-(4-chlorbenzyl)-D-glucofüranosid mit
600 ml Eisessig und 600 ml Wasser behandelt, das Gemisch während 16 Stunden auf 70" erhitzt, die überschüssige Essigsäure abdestilliert, den wässrigen Rückstand mit Äther extrahiert und die Ätherlösung wie oben beschrieben aufarbeitet.
Eine Lösung von 12,5 g 3-O-n-Propyl-5,6-di-O-(4-chlor- benzyl)-D-glucofuranose in 13 ml absolutem Pyridin wird abgekühlt und mit 13 ml Essigsäureanhydrid versetzt und während 16 Stunden bei Zimmertemperatur stehengelassen.
Dann verdünnt man unter Kühlen mit 12 ml Wasser, dekantiert die wässrige Schicht vom abgeschiedenen Öl ab und nimmt letzteres in Äther auf. Die organische Phase wird mit 2n Salzsäure ausgeschüttelt und mit Wasser gewaschen, dann über Magnesiumsulfat getrocknet und eingedampft. Der Rückstand wird destilliert und die erwünschte 1,2-Di-O-ace tyl-3 -O-n-propyl-5 ,6-di-0-(4 -chlorbenzyl) -D-glucofuranose bei 240 /0,05 mm Hg als leicht gelbliches Öl gewonnen.
Weitere Verbindungen, die nach dem oben illustrierten Verfahren bei Auswahl der geeigneten Ausgangsstoffe hergestellt werden können, sind z.B.
Äthyl-3-O-methyl-5 ,6-di-O-benzyl-D-glucofuranosid, schwach gelbes Öl, Kp. 1800/0,02 mm Hg; [a]D20 = -31+1" (c = 1 in Chloroform); durch Elution mit einem 85:15-Gemisch von Chloroform und Essigsäureäthylester lässt sich das Anomerengemisch in die reinen Anomeren dünnschichtchromatographisch an Silicagel (RF 254 der Firma Merck, Darmstadt) auftrennen.
Das a-Anomer hat einen Rf-Wert von 0,45; [a]D20 = +34+ 10 (c = 1 in Chloroform); und das ss-Anomer einen Rf-Wert von 0,23; [a]D20 = -61fl" (c = 1 in Chloroform); sek.-Butyl-3-O-methyl-5,6-di-O-benzyl-D-giucofüranosid, schwach gelbes Öl, Kp. 200 /0,05 mm Hg; [a]D20 = -24rrl" (c = 1 in Chloroform); Äthyl-3 -O-methyl-5,6-di-O-(4-methylbenzyl)-D-gluco- furanosid, schwach gelbes Öl, Kp. 195 /0,03 mm Hg; a] 20 = - 14 i 10 (c = 1 in Chloroform); Äthyl-3 -O-methyl-5 ,6-di-0-(4 -chlorbenzyl)-D -glucofuranosid, schwach gelbes Öl, Kp. 1900/0,01 mm Hg;

Beispiel 2
Eine Lösung von 15 g Äthyl-3-O-methyl-5,6-di-O-benzyl D-glucofuranosid in 30 ml absolutem Pyridin wird mit 4,1 g fein pulverisiertem Bernsteinsäureanhydrid versetzt und das Reaktionsgemisch unter Rühren und Feuchtigkeitsausschluss bei 70 während 28 Stunden erhitzt. Der nach dem Eindampfen unter vermindertem Druck bei 55" erhaltene Rückstand wird mit 45 ml Eiswasser versetzt und während 5 Minuten geschüttelt. Man extrahiert dann mit Äther, wäscht die Ätherlösung fünfmal mit je 50 ml eisgekühlter 2n Salzsäure und 50 ml Eiswasser, trocknet die Ätherphase über Natriumsulfat und dampft unter vermindertem Druck ein.
Der Rückstand wird im Hochvakuum bei 20 vom restlichen Lösungsmittel befreit; man erhält das Äffiyl-2-O-(ss-carboxy-pro- pionyl)-3 -O-methyl-5 ,6-di-O-benzyl-D -glucofuranosid der Formel

Nach kurzem Stehenlassen einer Lösung der obigen Verbindung in einem Überschuss einer 1n wässrig äthanolischen Natriumhydroxidlösung wird der Ester quantitativ verseift.
Das erhaltene Äthyl-3-O-methyl-5,6-di-O-benzyl-D-gluco- furanosid kann dünnschichtchromatographisch charakterisiert werden; das a-Anomer hat einen Rf-Wert von 0,45; [a]D20 = + 34 ist 10 (c = 1 in Chloroform); und das ss-Anomer einen RfWert von 0,23; [a]D20 = -61*1' (c = 1 in Chloroform).
Eine Lösung des Produktes in Äther wird mit der berechneten Menge wässriger Natriumhydrogencarbonatlösung geschüttelt. Nach Abdampfen des Äthers im Vakuum verdünnt man mit Wasser, bis eine klare Lösung entsteht, welche lyophylisiert wird. Aus dem Lyophylisat lässt sich durch Verdünnen mit Wasser eine mindestens 10%ige wässrige Lösung des so erhaltenen Natriumsalzes des Äthyl-2-O-- carboxy-propionyl)-3 -O-methyl-5 -di-O-benzyl-D -gluco- furanosids herstellen.
Beispiel 3
Eine Lösung von 2,75 g Bernsteinsäureanhydrid in 20 ml absolutem Pyridin wird mit einer Lösung von 10 g Äthyl 3-O-n-propyl-5 ,6-di-O-(4-chlorb enzyl) -D-glucofuranosid in 20 ml absolutem Pyridin versetzt und bei 70 unter Feuchtigkeitsausschluss 48 Stunden gehalten. Der nach Eindampfen unter vermindertem Druck bei 55" erhaltene Rückstand wird mit 45 ml Eiswasser versetzt und 5 Minuten geschüttelt.
Anschliessend extrahiert man mit Äther, wäscht die Ätherphase fünfmal mit 50 ml eisgekühlter 2n Salzsäure und 50 ml Eiswasser und trocknet über Natriumsulfat. Nach dem Abestillieren des Äthers unter vermindertem Druck wird der Rückstand im Hochvakuum vom restlichen Lösungsmittel befreit, und man erhält das Äthyl-2-O-6B-carboxy-pro- pionyl)-3-O-n-propyl-5 ,6-di-O-(4-chlorbenzyl) -D-glucofuranosid der Formel

Anschliessend wird mit Wasser so lange verdünnt, bis eine klare Lösung entsteht (9,5%). Diese Lösung lässt sich lyophy lisieren, wobei das Natriumsalz des Äthyl-2-O-(ss-carboxy- propionyl)-3-O-n-propyl-5 ,6-di-O-(4-chlorbenzyl) -D-glucofuranosids als dickflüssiges Öl anfällt.
Beispiel 4
Eine Lösung von 2,55 g Bernsteinsäureanhydrid in 20 ml absolutem Pyridin wird mit einer Lösung von 10 g Äthyl-3 O-n-propyl-5 ,6-di-O-benzyl-D-glucofuranosid versetzt und während 48 Stunden bei 70 gehalten. Der durch Eindampfen im Vakuum bei 55" erhaltene Rückstand wird mit 45 ml Eiswasser während 30 Minuten verrührt, dann mit Äther extrahiert. Man wäscht die Ätherlösung fünfmal mit 50 ml eisgekühlter 2n Salzsäure und 50 ml Eiswasser, trocknet über Natriumsulfat und dampft unter vermindertem Druck ein.
Der Rückstand wird im Hochvakuum vom restlichen Lösungsmittel befreit; und man erhält das Äthyl-2-O-(ss-carb- oxypropionyl)-3 -O-n-propyl-5,6-di-O-benzyl-D-glucofura- nosid der Formel als dickflüssiges Öl.
Nach kurzem Stehenlassen einer Lösung der obigen Verbindung in einem Überschuss einer 1n wässrig-äthanolischen Natriumhydroxydlösung wird der Ester quantitativ verseift.
Nach dem Abdampfen des Äthanols wird mit Methylenchlorid extrahiert; das nach dem Verdampfen des Lösungsmittels erhältliche Äthyl-3-O-n-propyl-5,6-di-O-benzyl-D-gluco- furanosid wird durch Destillation des Rückstandes als schwach gelbes Öl bei 220 /0,03 mm Hg erhalten; [am20 = -24 +1" (c = 1 in Chloroform).
Eine Lösung des Produkts in Äther wird mit der berechneten Menge Natriumhydrogencarbonat in Wasser versetzt.
Das Gemisch wird unter vermindertem Druck bei 40 vom Äther befreit. Die wässrige Lösung kann lyophylisiert werden, und man erhält das Natriumsalz des Äthyl-2-O-6B-carboxy- propionyl)-3-O-n-propyl-5,6-di-O-benzyl-D-glucofuranosids als dickflüssiges Öl, das sich in Wasser bis zu einer Konzentration von 25% löst.

In a 5,6-dihydroxy compound which contains a free or preferably an etherified hydroxyl group in the 3-position, the 6-hydroxyl group, e.g. esterifying by treatment with a suitable organic sulfonic acid halide such as p-toluenesulfonyl chloride and by treatment with a suitable basic agent such as an alkali metal lower alkoxide such as sodium ethoxide to form the 5,6-epoxy compound; by splitting the epoxide using an alcohol of formula R6-OH in the presence of a transesterification catalyst, e.g. an alkali metal, such as sodium compound of an alcohol of the formula R6-OH, the 5-hydroxy-6 R6-O compound is obtained, in which the free hydroxy group in the 5-position is selectively in the usual way, e.g.
B. by treating with a reactive ester of a compound of the formula R5-OH, in the presence of a basic agent, can be etherified.
In the starting materials obtainable by the abovementioned processes, in which R3 is preferably a lower aliphatic radical, the protective group for the two hydroxyl groups in the 1- and 2-positions can be obtained by treating e.g. opened with aqueous hydrochloric acid. The two hydroxy groups can then be treated with a suitable derivative of an organic carboxylic acid such as acetic acid, e.g. an anhydride, such as acetic anhydride, to be esterified to form a 1,2-bis-O-acyl compound, especially 1,2-bis-O-acetyl compound; when converting z. B. with bromine in glacial acetic acid, a 2-O-acyl-, e.g. 2-O-acetyl compound with a reactive esterified hydroxyl group in the 1-position.
At any suitable stage of the processes described above for preparing the starting materials, a hydroxyl group etherified in the 3-position by a suitable 2-alkenyl, such as the allyl radical, e.g. By rearrangement of the double bond by treatment with a suitable base such as an alkali metal, e.g. Potassium tert-butoxide, preferably in a suitable solvent such as e.g.
Dimethyl sulfoxide, and oxidative-hydrolytic removal of the 1-lower alkenyl, such as 1-propenyl group, e.g. By treatment with potassium permanganate, preferably in a basic medium such as ethanolic alkali metal hydroxide, e.g.
Potassium hydroxide, are released.
The new compounds or their salts can be used as medicaments, e.g. in the form of pharmaceutical preparations which they can be used in admixture with an administration agent, e.g. the enteral or parenteral, as well as topical application contain suitable pharmaceutical organic or inorganic, solid or liquid carrier material. Substances which are inert towards the new compounds, such as water, gelatine, sugar, e.g.
Lactose, glucose or fructose, starches such as corn, wheat or rice starch, stearic acid or salts thereof such as calcium or magnesium stearate, talc, vegetable oils, benzyl alcohols, gums, polyalkylene glycols, propylene glycol or other known excipients. The pharmaceutical preparations can e.g. as tablets, dragees, capsules or in liquid form as solutions, suspensions or emulsions. If necessary, they are sterilized and / or
or contain auxiliaries such as preservatives, stabilizers, wetting agents or emulsifiers, salts to change the osmotic pressure or buffers. They can also contain other therapeutically valuable substances. The preparations are obtained according to the usual methods.
The invention is described in more detail in the following examples. The temperatures are given in degrees Celsius.
example 1
To 2.4 g of 1,2-di-O-acetyl-3-on-propyl-5,6-di-O- (4 chlorobenzyl) -D-glucofuranose, 5 ml of a 33% solution of hydrogen bromide in Glacial acetic acid was added and the mixture was left to stand at room temperature for 21/2 hours. It is then poured onto ice water and extracted with chloroform. The organic extract is washed with ice water and an ice-cold aqueous sodium hydrogen carbonate solution, dried over sodium sulfate and concentrated under reduced pressure at a bath temperature of 30-40.
The residue, containing the 2-O-acetyl-3-on-propyl-5,6-di-O- (4-chlorobenzyl) -D-glucofuranosyl bromide, is mixed with 50 ml of a 1N ethanolic solution of sodium ethanolate and left to stand for 16 hours at room temperature, then neutralized with 2N aqueous acetic acid and extracted with chloroform. The organic extract is washed with ice water and an ice-cold concentrated aqueous sodium hydrogen carbonate solution, dried over sodium sulfate and concentrated under a water jet vacuum. The residue is distilled under high vacuum, the ethyl 3-on-propyl-5, 6-di-O- (4-chlorobenzyl) -D-glucofuranoside being obtained as a pale yellow oil at 195 / 0.01 mm Hg ; [a] D20 = - 17 f 10 (c = 1 in chloro form).
By eluting with an 85:15 mixture of chloroform and ethyl acetate, the anomer mixture can be separated into the pure anomers by thin layer chromatography on silica gel (RF 254 from Merck, Darmstadt).
The a-anomer has an Rf value of 0.50; [a] D20 = + 24 + 1 "(c = 1 in chloroform); and the ss-anomer has an Rf value of 0.24; [a] D20 = 45 + 10 (c = 1 in chloroform).
The raw material used can be produced as follows:
A solution of 82 g of 1,2-O-isopropylidene-3-0-allyla-D-glucofuranose in 800 ml of ethanol is hydrogenated in the presence of 1 g of a 10% palladium-on-carbon catalyst. The catalyst is filtered off, the filtrate is evaporated and the residue is degassed in a high vacuum; 1, 2-O-isopropylidene-3 -O-n-propyl-α-D -gluco-furanose is obtained in this way; [a] D20 = 49f 1 "(c = in chloroform); which is processed further without purification.
A solution of 32 g of 1,2-O-isopropylidene-3-O-n-propyla-D-glucofuranose in 100 ml of absolute dioxane is mixed with 87.5 g of powdered potassium hydroxide. A solution of 131 g of 4-chlorobenzyl chloride in 50 ml of absolute dioxane is then added dropwise over the course of 30 minutes while stirring; then left to react for 5 hours at 80 ".
The excess 4-chlorobenzyl chloride is distilled off with steam and, after cooling, it is extracted with chloroform. The organic extract is washed with water, dried over sodium sulfate and evaporated under reduced pressure. The 1,2-O-isopropylidene-3-0-n-propyl-5, 6-di- (4-chlorobenzyl) -aD-glucofuranose is obtained by distilling the residue as a pale yellow oil at 230 "/ 0.015 mm Hg; [a] D20 = -22 + 1 (c = 1 in chloroform).
A suspension of 5 g of 1,2-O-isopropylidene-3-on-propyl-5,6-di-O- (4-chlorobenzyl) -aD-glucofuranose in a mixture of 35 ml of glacial acetic acid and 17.5 ml of 1N aqueous Sulfuric acid is heated with vigorous stirring and in a nitrogen atmosphere for 30 minutes at an internal temperature of 80 ". The reaction mixture is cooled to 10, adjusted to pH 23 with 2N aqueous sodium hydroxide solution and then evaporated at a bath temperature of 35. The concentrate is extracted with ether, the organic phase washed neutral with a saturated aqueous sodium hydrogen carbonate solution and with water, dried over magnesium sulphate and evaporated.
The residue corresponds to the 3-on-propyl-5,6-di-0- (4-chlorobenzyl) -D-glucofuranose, which in the thin-layer chromatogram (silica gel) in the chloroform: ethyl acetate (85:15) system has an Rf value of 0 , 12; F. 60 after recrystallization from a 1: 3 mixture of toluene and hexane and cooling to -17 ".
The 3 -On-Propyl-5,6-di-O- (4-chlorobenzyl) - D-glucofuranose can also be obtained by adding 15 g of ethyl-3-O n-propyl-5, 6-di-O- (4-chlorobenzyl) -D-glucofuranoside with
600 ml of glacial acetic acid and 600 ml of water are treated, the mixture is heated to 70 "for 16 hours, the excess acetic acid is distilled off, the aqueous residue is extracted with ether and the ether solution is worked up as described above.
A solution of 12.5 g of 3-on-propyl-5,6-di-O- (4-chlorobenzyl) -D-glucofuranose in 13 ml of absolute pyridine is cooled and mixed with 13 ml of acetic anhydride and added for 16 hours Left to stand at room temperature.
Then, while cooling, it is diluted with 12 ml of water, the aqueous layer is decanted from the separated oil and the latter is taken up in ether. The organic phase is extracted with 2N hydrochloric acid and washed with water, then dried over magnesium sulfate and evaporated. The residue is distilled and the desired 1,2-di-O-acetyl-3 -On-propyl-5, 6-di-0- (4-chlorobenzyl) -D-glucofuranose at 240 / 0.05 mm Hg as slightly yellowish oil obtained.
Further compounds which can be prepared by the process illustrated above with the selection of suitable starting materials are e.g.
Ethyl 3-O-methyl-5, 6-di-O-benzyl-D-glucofuranoside, pale yellow oil, b.p. 1800 / 0.02 mm Hg; [a] D20 = -31 + 1 "(c = 1 in chloroform); by elution with an 85:15 mixture of chloroform and ethyl acetate, the anomer mixture can be converted into the pure anomers by thin-layer chromatography on silica gel (RF 254 from Merck, Darmstadt ).

Example 2
4.1 g of finely pulverized succinic anhydride are added to a solution of 15 g of ethyl 3-O-methyl-5,6-di-O-benzyl D-glucofuranoside in 30 ml of absolute pyridine and the reaction mixture is stirred at 70 while with exclusion of moisture Heated for 28 hours. The residue obtained after evaporation under reduced pressure at 55 "is mixed with 45 ml of ice water and shaken for 5 minutes. It is then extracted with ether, the ether solution is washed five times with 50 ml of ice-cooled 2N hydrochloric acid and 50 ml of ice water, the ether phase is dried over Sodium sulfate and evaporated under reduced pressure.

A solution of the product in ether is shaken with the calculated amount of aqueous sodium hydrogen carbonate solution. After evaporation of the ether in vacuo, it is diluted with water until a clear solution is formed, which is lyophilized. An at least 10% aqueous solution of the sodium salt of ethyl 2-O- carboxy-propionyl) -3 -O-methyl-5-di-O-benzyl-D -gluco can be obtained from the lyophilisate by diluting it with water - make furanosids.
Example 3
A solution of 2.75 g of succinic anhydride in 20 ml of absolute pyridine is mixed with a solution of 10 g of ethyl 3-on-propyl-5, 6-di-O- (4-chlorobenzyl) -D-glucofuranoside in 20 ml of absolute pyridine added and kept at 70 for 48 hours with exclusion of moisture. The residue obtained after evaporation under reduced pressure at 55 "is mixed with 45 ml of ice water and shaken for 5 minutes.
It is then extracted with ether, the ether phase is washed five times with 50 ml of ice-cold 2N hydrochloric acid and 50 ml of ice water and dried over sodium sulfate. After the ether has been distilled off under reduced pressure, the residue is freed from the remaining solvent in a high vacuum, and the ethyl 2-O-6B-carboxy-propionyl) -3-on-propyl-5, 6-di-O is obtained - (4-chlorobenzyl) -D-glucofuranoside of the formula

Example 4
A solution of 2.55 g of succinic anhydride in 20 ml of absolute pyridine is mixed with a solution of 10 g of ethyl 3 O-n-propyl-5, 6-di-O-benzyl-D-glucofuranoside and kept at 70 for 48 hours. The residue obtained by evaporation in vacuo at 55 "is stirred with 45 ml of ice water for 30 minutes, then extracted with ether. The ether solution is washed five times with 50 ml of ice-cold 2N hydrochloric acid and 50 ml of ice water, dried over sodium sulfate and evaporated under reduced pressure .
The residue is freed from the remaining solvent in a high vacuum; and the ethyl 2-O- (ss-carbo-oxypropionyl) -3 -O-n-propyl-5,6-di-O-benzyl-D-glucofuranoside of the formula is obtained as a thick oil.
After a solution of the above compound is left to stand for a short time in an excess of a 1N aqueous-ethanolic sodium hydroxide solution, the ester is saponified quantitatively.
After the ethanol has evaporated, it is extracted with methylene chloride; the ethyl 3-O-n-propyl-5,6-di-O-benzyl-D-gluco furanoside, which is obtainable after evaporation of the solvent, is obtained by distilling the residue as a pale yellow oil at 220 / 0.03 mm Hg; [am20 = -24 +1 "(c = 1 in chloroform).
A solution of the product in ether is mixed with the calculated amount of sodium hydrogen carbonate in water.
The mixture is freed from the ether under reduced pressure at 40 °. The aqueous solution can be lyophilized, and the sodium salt of ethyl 2-O-6B-carboxy-propionyl) -3-on-propyl-5,6-di-O-benzyl-D-glucofuranoside is obtained as a viscous oil which dissolves in water up to a concentration of 25%.
