Beispiel d-Lysergsäure-N-n icotinoylpiperazid

Process for the preparation of d-lysergic acid-N-nicotinoylpiperazide
The invention relates to a process for the production of d-lysergic acid-N-nicotinoylpiperazide (formula I, see formula sheet) and its pharmacologically acceptable salts, and pharmaceutical preparations thereof.
According to the invention, the compound of the formula I and its acid addition salts are obtained by reacting a reactive derivative of the acid of the formula II with the compound of the formula III or an acid addition salt thereof in an organic solvent which is inert under the reaction conditions in the presence of a basic condensing agent, the so The resulting compound of the formula I is isolated from the reaction mixture and, if appropriate, subsequently converted into its pharmacologically acceptable salts with acids.
As reactive derivatives of the acid of formula II, e.g. the reaction product resulting from the reaction of the acid of the formula II with a chlorinating or brominating agent and an N-di (lower) alkyl-substituted acid amide of an aliphatic monocarboxylic acid having 1 to 3 carbon atoms, the acid chloride hydrochloride or the addition product of the acid of the formula II with Dicyclohexylcarbodiimide can be used. Chlorinating or brominating agents are e.g. Oxalyl chloride, thionyl chloride, phosgene, phosphorus oxychloride and phosphorus oxybromide are usable. As the N-di (lower) alkyl-substituted acid amide of an aliphatic monocarboxylic acid having 1 to 3 carbon atoms, e.g. Dimethylformamide or dimethylacetamide are used.

For salt formation, i.a. the following acids are used: hydrochloric acid, sulfuric acid, phosphoric acid, tartaric acid, maleic acid, methanesulfonic acid.
d-Lysergic acid-N-nicotinoylpiperazid is characterized by interesting pharmacological properties and can therefore be used as a remedy.
d-Lysergic acid-N-nicotinoylpiperazid shows a pronounced central circulatory regulating effect, as in circulatory tests on anesthetized animals (cat and dog) by lowering blood pressure, bradycardia, noradrenaline and the like.
Tyramine enhancement, carotid occlusion reflex inhibition as well as enhancement of isoproterenol tachycardia have been found, which makes its use e.g. in orthostasis and hypertension. For the above application, the dosage will depend on the form of administration and treatment desired.
However, good results are achieved with a dose of approximately 0.02-15 mg per kg of body weight of the test animal.
The average daily dose for larger mammals can be 1-100 mg, divided into 2 to 3 applications in doses of 0.3 - 50 mg.
Furthermore, it is characterized by good salidiuretic effectiveness, as could be shown by diuresis experiments on the conscious rat. Good results are achieved with a dosage of 1-3 mg per kg of body weight of the test animal, the dose to be administered being dependent on the desired effect and type of application.
The average daily dose for larger mammals can be 10-500 mg, divided into 2 to 3 applications in doses of 3-250 mg.
d-Lysergic acid-N-nicotinoylpiperazid is also characterized by a pronounced anti-inflammatory and edema-inhibiting effect, as was found in the carrageenoedema in rats. The dosage used will depend on the mode of administration and treatment desired. Good results are achieved with a dosage of approximately 1-10 mg per kg of body weight of the test animal.
The average daily dose for larger mammals can be 10-500 mg, divided into 2 to 3 applications in doses of 3-250 mg.
In addition, it shows CNS-dampening effects, as could be determined in the awake mouse in the observation test through inhibition in the climbing test and temperature reduction. The dosage will depend on the desired effect and type of application, but good results are achieved with a dosage of approximately 10-50 mg per kg of body weight of the test animal.
The average daily dose for larger mammals can be 10-500 mg, divided into 2 to 3 applications in doses of 3-250 mg.
The new compound of the formula I can be used as a medicament alone or in corresponding medicament forms for oral, enteral or parenteral administration. In order to produce suitable dosage forms, this is processed with inorganic or organic, pharmacologically indifferent auxiliary substances.
In the following example, which is intended to explain the invention in more detail, but in no way restrict its scope, the temperatures are given in degrees Celsius and are not corrected. If the preparation of the starting compounds is not described, these are known or can be prepared by processes known per se or analogously to processes known per se.

Example d-Lysergic acid-N-n icotinoylpiperazide
In a stirred, cooled to -150 mixture of 75 ml of abs. Dimethylformamide and 150 ml of abs. Acetonitrile, 12.6 g (0.1 m) of oxalyl chloride in 10 ml of abs. Acetonitrile was added dropwise, the temperature being kept constant.
The resulting crystalline precipitate is stirred for a further 5 minutes at -100. Then 27 g (0.1 m) of dry d-lysergic acid are introduced and, with strong cooling, 86 g (1 m) of piperazine anhydr. and stir vigorously at 00 for 4 hours. After extraction between 2N soda / 10% methanol in methylene chloride to a cell negative sample, drying of the combined organic phases and evaporation of the solvent, a polar crude base is obtained which is dissolved in 150 ml of abs. Pyridine is dissolved. After slowly introducing 16 g of nicotinoyl chloride HC1 with thorough stirring at room temperature, the reaction is over after 60 minutes.
Between 10% methanol / methylene chloride and 2N soda solution are worked up and, after drying and removal of the organic solvent by distillation, the crude base is obtained, which is immediately chromatographed on 50 times the amount of Alox II-III.
The pure amorphous d-lysergic acid-N-nicotinoylpiperazide is eluted with 0.5% methanol in methylene chloride, while a mixed fraction of 0.7% between the title compound and the d-isolysergic acid form is obtained, which mix in dimethylformamide / triethylamine at 1000 up to the equilibrium conc. from approx.
