Verfahren zur Herstellung eines neuen Lysergsäurederivates
Die Erfindung betrifft ein Verfahren zur Herstellung eines Lysergsäurederivates der Formel 1 (siehe Formelblatt) und dessen pharmakologisch verträglichen Salzen mit Säuren, wie auch pharmazeutische Zubereitungen der Verbindung der Formel I und ihrer Säureadditionssalze.
Erfindungsgemäss gelangt man zu der Verbindung der Formel I und ihren Säureadditionssalzen, indem man ein reaktionsfähiges Derivat der Säure der Formel II mit der Verbindung der Formel III oder einem Säureadditionssalz derselben in einem unter den Reaktionsbedingungen inerten organischen Lösungsmittel in Gegenwart eines basischen Kondensationsmittels umsetzt, die so entstandene Verbindung der Formel I in an sich bekannter Weise isoliert und gegebenenfalls anschliessend in ihre pharmakologisch verträglichen Salze mit Säuren überführt.
Als reaktionsfähige Derivate der Säure der Formel II können z.B. das bei der Umsetzung der Säure der Formel II mit einem Chlorierungs- oder Bromierungsmittel und einem N-di(nieder)-alkylsulbstituierten Säureamid einer aliphatischen Monocarbonsäure mit 1 bis 3 Kohlenstoffatomen entstehende Reaktionsprodukt, das Säurechlorid-hydrochlorid oder das gemischte Anhydrid mit Schwefelsäure oder Trifluoressigsäure verwendet werden.
Als Chlorierungs- oder Bromierungsmittel sind z.B.
Oxalylchlorid, Thionylchlorid, Phosgen, Phosphoroxichlorid und Phosphoroxibromid verwendbar. Als N-di (nieder)-alkylsubstituiertes Säureamid einer aliphatischen Monocarbonsäure mit 1 bis 3 Kohlenstoffatomen können z.B. Dimethylformamid oder Dimethylacetamid dienen.
Als Lösungsmittel sind beispielsweise Methylenchlorid, Chloroform, Acetonitril und Dimethylformamid oder Mischungen derselben verwendbar. Als basisches Kondensationsmittel eignen sich z.B. Pyridin, Triäthylamin oder Überschuss der Verbindung der Formel III.
Die erfindungsgemässe Umsetzung kann bei Temperaturen etwa -10 bis +200 durchgeführt werden, vorzugsweise bei OOC. Die Reaktionsdauer beträgt, je nach der verwendeten Reaktionstemperatur, etwa 1/2 bis 5 Stunden.
Die Aufarbeitung der Reaktionslösung kann beispielsweise durch Extraktion der Reaktionslösung mit wässriger 2 N Sodalösung und einem organischen Lösungsmittel erfolgen. Die Verbindung der Formel I kann z.B.
nach Trocknung und Verdampfen des organischen Lösungsmittels chromatographisch isoliert werden.
Das als Ausgangsprodukt verwendete Nicotinoylpiperazin (Formel III) kann erhalten werden, indem man eine Verbindung der Formel IV (siehe Formelblatt), worin Z eine Schutzgruppe bedeutet, z.B. die Tritylgruppe, mit einem reaktionsfähigen Derivat der Verbindung der Formel VI, z.B. dem Säurechlorid, in Gegenwart eines basischen Kondensationsmittels, z.B. Pyridin, umsetzt und aus dem erhaltenen Reaktionsprodukt anschliessend die Schutzgruppe abspaltet, z.B. mit methanolischer Salzsäure.
Die Verbindungen der Formel IV, worin Z eine für Aminogruppen an sich bekannte Schutzgruppe, wie z.B.
die Tritylgruppe, bedeutet, kann man erfindungsgemäss erhalten, indem man Piperazin in grossem Überschuss mit einem reaktionsfähigen Derivat einer Verbindung der Formel V, worin Z die obige Bedeutung hat, umsetzt und die so entstandenen Verbindungen der Formel IV in an sich bekannter Weise isoliert.
Die erfindungsgemäss hergestellte Verbindung der Formel I bildet mit anorganischen oder organischen Säuren beständige, bei Raumtemperatur meist kristallisierte Salze. Zur Salzbildung können u.a. folgende Säuren verwendet werden: Chlorwasserstoffsäure, Schwefelsäure, Phosphorsäure, Weinsäure, Maleinsäure, Methansulfonsäure.
d- Lysergsäure - N- nicotinoylpiperazid zeichnet sich durch interessante pharmakologische Eigenschaften aus und kann daher als Heilmittel Verwendung finden.
d-Lysergsäure-N-nicotinoylpiperazid zeigt ausgeprägte zentrale kreislaufregulierende Wirkung, wie in Kreislaufversuchen am narkotisierten Tier (Katze und Hund) durch Blutdrucksenkung, Bradycardie, Norandrenalin- und Tyramin-Verstärkung, Carotisocclusion-Reflexhemmung sowie Verstärkung der Isoproterenol-Tachycardie festgestellt wurde, was seine Anwendung z.B. bei Orthostase und Hypertonie nahelegt. Für die obengenannte Anwen dung wird die Dosierung davon abhängen, welche Verabreichungsform und Behandlung gewünscht wird. Gute Resultate werden jedoch erreicht mit einer Dosierung von ungefähr 0,02-15 mg pro kg Körpergewicht des Testtieres.
Die durchschnittliche Tagesdosis für grössere Säugetiere kann 1-100 mg betragen, verteilt auf 2- bis 3malige Anwendung in Dosen zu 0,3-50 mg.
Weiterhin zeichnet es sich durch gute salidiuretische Wirksamkeit aus, wie durch Diurese-Versuche an der wachen Ratte gezeigt werden konnte. Gute Resultate werden mit einer Dosierung von 1-30 mg pro kg Körpergewicht des Testtieres erreicht, wobei die zu verabreichende Dosis von der gewünschten Wirkung und Applikationsart abhängen wird.
Die durchschnittliche Tagesdosis für grössere Säugetiere kann 10-500 mg betragen, verteilt auf 2- bis 3malige Anwendung in Dosen zu 3-250 mg.
d-Lysergsäure-N-nicotinoylpiperazid zeichnet sich zudem durch ausgeprägte antiphlogistische und oedemhemmende Wirkung aus, wie am Carrageenoedem an der Ratte festgestellt wurde. Die angewendete Dosierung wird davon abhängen, welche Verabreichungsform und Behandlung gewünscht wird. Gute Resultate werden erreicht mit einer Dosierung von ungefähr 1-10 mg pro kg Körpergewicht des Testtieres.
Die durchschnittliche Tagesdosis für grössere Säugetiere kann 10-500 mg betragen, verteilt auf 2- bis 3malige Anwendung in Dosen zu 3-250 mg.
Ausserdem zeigt es ZNS-dämpfende Effekte, wie an der wachen Maus im Beobachtungstest durch Hemmung im Klettertest und Temperatursenkung festgestellt werden konnte. Die Dosierung wird von der gewünschten Wirkung und Applikationsart abhängen, gute Resultate werden jedoch erreicht mit einer Dosierung von ungefähr 10-50 mg pro kg Körpergewicht des Testtieres.

Die neue Verbindung der Formel I kann als Arzneimittel allein oder in ensprechenden Arzneiformen für orale, enterale oder parenterale Verabreichung verwendet werden.
In dem nachfolgenden Beispiel, welches die Erfindung näher erläutern, ihren Umfang aber in keiner Weise einschränken soll, erfolgen die Temperaturangaben in Celsiusgraden und sind nicht korrigiert. Soweit die Herstellung der Ausgangsverbindungen nicht beschrieben wird, sind diese bekannt oder nach an sich bekannten Verfahren oder analog zu an sich bekannten Verfahren herstellbar.

Beispiel d-Lysergsäure-N-nicotinoylpiperazid
In eine auf -100 gekühlte, gerührte Mischung von
10 ml absolutem Dimethylformamid und 20 ml absolu tem Acetonitril wird innerhalb 5 Minuten eine Lösung von 1,27 g (10 mMol) Oxalylchlorid in 5 ml absolutem Acetonitril zugetropft. In die entstandene Suspension trägt man rasch bei derselben Temperatur 2,68 g wasserfreie d-Lysergsäure ein, lässt anschliessend eine Lösung von
1,91 g (10 mMol) N-Nicotinoylpiperazin in 5 ml absolu tem Pyridin unter guter Kühlung zufliessen und rührt das Reaktionsgemisch noch 3 Stunden bei 00. Zur Aufarbei tung verdünnt man mit 100 ml Methylenchlorid und
50 ml wässriger 2 N Sodalösung, schüttelt und extrahiert nach Abtrennung der organischen Phase die wässrige Phase noch 3mal mit je 50 ml Methylenchlorid nach.
Nach Trocknen der vereinigten organischen Phase über Natriumsulfat und Verdampfen des Lösungsmittels im
Vakuum erhält man einen braunen Schaum, welcher an der 60fachen Mengen Aluminiumoxid der Aktivität II chromatographiert wird. Die Titelverbindung wird mit 0,5% Methanol in Methylenchlorid als gelblicher Schaum eluiert.
Maleinat
Aus absolutem Äthanol und der doppelt-molaren
Menge Maleinsäure nach Erwärmen. Smp. 198-200 (Zers.). oID20 = 120 (c = 1, Methylenchlorid/Metha- nol = 1:1).
Das als Ausgangsmaterial verwendete N-Nicotinylpiperazin kann beispielsweise wie folgt erhalten werden: a) N- Trityl-piperazin
Eine gerührte Suspension von 25,8 g (0,3 m) Piperazin techn. in 30 ml Acetonitril und 8,55 ml Eisessig (0,15 m) wird bei 50O durch Zugabe von 16 ml Wasser in eine Lösung überführt. Anschliessend tropft man 8,34 g (0,03 m) Tritylchlorid in 45 ml Aceton innert 5 Minuten zu und rührt bei dieser Temperatur kräftig während 3 Stunden. Zur Aufarbeitung verdünnt man mit halbgesättigter, wässriger Pottachelösung und extrahiert mehrmals mit Äther, wobei nicht umgesetztes Piperazin als 3.
Phase verbleibt und nach dem Abkühlen als kristallines Hexahydrat zurückgewonnen und wieder eingesetzt werden kann.
Die Ätherphase wird über Pottasche getrocknet. Nach Eindampfen des Lösungsmittels verbleibt ein kristalliner Rückstand, welcher sich im Dünnschichtchromatogramm mit 10% Methanol/Methylenchlorid; Kieselgel G als ¯95%in reines N-Trityl-piperazin erweist, das sofort weiterverarbeitet wird.
b) N-Nicotinoyl-N' -trityl-piperazin
8,7 g (¯26,5 mMol) des nach Stufe a) erhaltenen N-Tritylpiperazins werden in 30 ml abs. Pyridin aufgeschlämmt, bei 0 mit 8,7 g Nicotinsäurechlorid. HCI (48,8 mMol) versetzt und 1 Stunde bei Raumtemperatur gerührt. Die rötliche Suspension wird im Vakuum eingeengt und zwischen halbgesättigter, wässriger Pottaschelösung und Äther gründlich extrahiert. Die vereinigten organischen Phasen werden mit 1 N Salzsäure und anschliessend mit gesättigter Natriumbicarbonat-Lösung gewaschen, über Natriumsulfat getrocknet und eingedampft.
Es verbleibt eine beiger, kristalliner Rückstand von N-Nicotinoyl-N'-trityl-piperazin, der sofort weiterverwendet werden kann.
Aus 50 ml Methyläthylketon werden hellbeige Kristalle der Titelverbindung vom Smp. 230-2320 erhalten.
c) N-Nicotinoyl-piperazin
9,8 g des nach Stufe b) erhaltenen Rohproduktes (20 mMol) werden in 80 ml Methanol, enthaltend 60 mMol HC1-Gas, gelöst und 1l/2 Stunden bei 500 gerührt.
Nach Eindampfen des Lösungsmittels kristallisiert das Dihydrochlorid der Titelverbindung durch Aufnehmen des Rückstandes in abs. Äthanol. Smp. 2100 (Zers. im zugeschmolzenen Röhrchen). Das N-Nicotinoyl-piperazin erweist sich im Dünnschichtchromatogramm auf Kieselgel G mit 15% Methanol/Methylenchlorid als rein.
Process for the production of a new lysergic acid derivative

The reaction solution can be worked up, for example, by extracting the reaction solution with aqueous 2N soda solution and an organic solvent. The compound of formula I can e.g.
isolated by chromatography after drying and evaporation of the organic solvent.
The nicotinoylpiperazine (formula III) used as the starting product can be obtained by adding a compound of the formula IV (see formula sheet) in which Z is a protective group, e.g. the trityl group, with a reactive derivative of the compound of formula VI, e.g. the acid chloride, in the presence of a basic condensing agent, e.g. Pyridine, and then splitting off the protective group from the reaction product obtained, e.g. with methanolic hydrochloric acid.
The compounds of the formula IV in which Z is a protective group known per se for amino groups, e.g.
the trityl group, can be obtained according to the invention by reacting piperazine in large excess with a reactive derivative of a compound of the formula V, in which Z has the above meaning, and isolating the resulting compounds of the formula IV in a manner known per se.
The compound of the formula I prepared according to the invention forms stable salts with inorganic or organic acids which are mostly crystallized at room temperature. For salt formation, i.a. the following acids are used: hydrochloric acid, sulfuric acid, phosphoric acid, tartaric acid, maleic acid, methanesulfonic acid.
d- Lysergic acid - N- nicotinoylpiperazid is characterized by interesting pharmacological properties and can therefore be used as a remedy.
d-Lysergic acid-N-nicotinoylpiperazid shows a pronounced central circulatory regulating effect, as was found in circulatory tests on anesthetized animals (cats and dogs) by lowering blood pressure, bradycardia, norandrenaline and tyramine intensification, carotid occlusion reflex inhibition and intensification of isoproterenol tachycardia Application e.g. in orthostasis and hypertension.
