Verfahren zur Herstellung eines neuen Antibioticums
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von 11-Desacetoxy-wortmannin (Formel 1, siehe Formelblatt), dadurch gekennzeichnet, dass man den Stamm NRRL 3363 der Pilzspecies Penicillium funiculosum Thom oder seine Mutanten in einer Nährlösung züchtet und hier auf l-Desacetoxy-wortmannin aus der Nährlösung isoliert und reinigt.
Der neue, erfindungsgemäss verwendete Stamm S 3196 von Penicillium funiculosum Thom wurde aus einer in Clarksburg, Maryland (USA) gefundenen Erdprobe isoliert und eine Probe davon beim United States Department of Agriculture (Northern Utilization Research and Development Division), Peoria, III., USA, unter der Nummer NRRL 3363 deponiert.
Der neue Stamm der Pilzspecies Penicillium funiculosum Thom wächst auf einem Malz-Hefeextrakt-Agar bei 18-27 und bildet einen dichten grünen Konidienrasen mit leuchtend gelben Konidienträgern. Die Rückseite der Kolonie zeigt eine grüngelbe Verfärbung des Substrats.
Der Stamm entspricht in seiner Morphologie und Physiologie der Beschreibung von C. Thom, US.-Dept. Agr. Bur.
Anim. Ind. Bul. 118, p. 69, fig. 27 (1910) und ist im Handbuch K. B. Raper und Ch. Thom, A Manual of the Penicillia , The Williams and Wilkins Co., 1949, Baltimore, auf den Seiten 616 bis 620 eingehend beschrieben und illustriert.
Für das erfindungsgemässe Verfahren können auch Stämme verwendet werden, wie sie z. B. durch Selektion oder Mutation unter Einwirkung von Ultraviolett- oder Röntgenstrahlen oder durch Anwendung anderer Massnahmen, wie z. B. durch Behandlung von Laboratoriumskulturen mit geeigneten Chemikalien aus dem neuen Stamm der Pilzspecies Penicillium funiculosum Thom gewonnen werden können.
Der neue Stamm der Pilzspecies Penicillium funiculosum Thom lässt sich auf vielerlei Nährböden, die die üblichen Nährstoffe enthalten, züchten. So verwendet ein solcher Stamm die für kohlenstoffheterotrophen Organismen üblicherweise benutzten Nährstoffe, beispielsweise Glucose, Stärke, Dextrin, Lactose, Rohrzucker usw. als Kohlenstoffquelle, organische und anorganische, stickstoffhaltige Verbindungen, wie Pepton, Hefe- oder Fleischextrakte, Ammoniumsulfat, Ammoniumnitrat, Aminosäuren usw. als Stickstoffquelle sowie die üblichen Mineralsalze und Spurenelemente.
Eine Verfahrensform zur Herstellung des neuen Antibioticums besteht darin, dass ein flüssiges Nährmedium mit Mycel eines neuen Stammes von Penicillium funiculosum Thom beimpft und die Kultur 5 bis 10 Tage bei 27 inkubiert wird. Die Züchtung kann unter aeroben Bedingungen in einer Oberflächenkultur oder submers unter Schütteln oder in Fermentern mit Begasung durch Luft oder Sauerstoff unter Rühren erfolgen. Sobald eine maximale Menge an Antibioticum produziert worden ist, wird filtriert und das Antibioticum durch extraktive oder adsorptive Arbeitsmethoden auf an sich bekannte Weise aus dem Filtrat gewonnen.
Eine Methode, die sich als vorzugsweise geeignet erwiesen hat, ist die Extraktion des Filtrates mit Essigester, jedoch können auch andere organische Lösungsmittel, wie z. B. Benzol, Chloroform, Butylacetat, Methylenchlorid oder Butanol, verwendet werden. Anschliessend werden die Extrakte vom Lösungsmittel befreit, z. B. durch Destillation, und der Rückstand zur Isolierung des gewünschten Antibioticums auf chromatographischem Wege an adsorbierenden Mitteln, wie Tonerde, Kieselgel, Magnesiumsilicat und dergleichen oder mittels Gegenstromverteilung gereinigt.
Das ll-Desacetoxy-wortmannin besitzt interessante, pharmakologische Eigenschaften und soll therapeutische Verwendung finden. Insbesondere besitzt das 1 1-Desacetoxy- wortmannin eine sehr hohe fungistatische Wirkung gegen über verschiedenen Erregern von Pilzinfektionen. In vitro zeigt sich das 1 1-Desacetoxy-wortmannin besonders wirksam gegen Saccharomyces cerevisiae, Candida albicans, Aspergillus fumigatus, Trichophyton mentagrophytes, Histoplasma capsulatum.
1 1-Desacetoxy-wortmannin zeigt überdies eine ödem hemmende und entzündungshemmende Wirksamkeit, wie sie aus dem Carrageen-Ödem-Test, dem Carrageen-Granulom beutel-Test und dem CMC-Granulombeutel-Test an der wa chen Ratte hervorgeht.
Die zu verabreichende Dosis variiert naturgemäss je nach der Art der Administration und des zu behandelnden
Zustandes. Im allgemeinen werden jedoch befriedigende Re sultate mit einer Dosis von 1 bis 15 mg/kg Körpergewicht erhalten. Diese Dosis kann nötigenfalls in 2 bis 3 Anteilen oder auch als Retardform verabreicht werden. Für grössere Säugetiere liegt die Tagesdosis bei etwa 1 bis 50 mg. Für die orale Applikation enthält eine geeignete Verabreichungsform 1 bis 50 mg der Wirksubstanz, vermischt mit einem flüssigen oder festen Träger. Für die topicale Anwendung können Pulver oder Sprays oder Salben oder Tinkturen, die 0,1 bis 2 /O der Wirksubstanz enthalten, verwendet werden.
In dem nachfolgenden Beispiel, welches die Ausführung des Verfahrens erläutert, den Umfang der Erfindung aber in keiner Weise einschränken soll, erfolgen alle Temperaturangaben in Celsiusgraden und sind nicht korrigiert. Die Schmelz- bzw. Zersetzungspunkte sind auf dem Kofler-Block bestimmt.

Beispiel
In einem Fermenter werden 150 Liter einer Nährlösung, die pro Liter 20 g Cerelose; 2 g Malzextrakt (Schweiz. Ferment AG); 2 g Hefeextrakt (Gistex X II, Koniklijke Gist en Spiritusfabrik N.V., Holland); 2 g KH2PO4; 2 g MgSO4 7 H2O und entmineralisiertes Wasser enthält, mit einer Sporensuspension des Stammes NRRL 3363 von Penicillium funiculosum Thom beimpft und unter Belüftung (150 L Luft/Min.) und unter Rühren (100 U./Min.) 112 Stunden bei 27C inkubiert. Die Kulturbrühe wird filtriert und das Filtrat zweimal mit je 100 Liter Essigester extrahiert. Die organische Phase wird einmal mit 60 Liter Wasser gewaschen und am Vakuum auf 5 Liter eingeengt. Das Konzentrat wird über wasserfreiem Magnesiumsulfat getrocknet und eingedampft.
Der Rückstand wird dreimal zwischen Petroläther und Methanol-Wasser (9:1) verteilt. Die Methanolphase wird konzentriert und anschliessend dreimal mit Essigester extrahiert. Die Essigester-Lösungen werden einmal mit Wasser gewaschen, über wasserfreiem Magnesiumsulfat getrocknet und eingedampft. Der Extrakt wird an der 50fachen Menge Kieselgel chromatographiert. Mit Chloroform Methanol (99:1) wird 1 l-Desacetoxy-wortmannin eluiert, das nach zweimaliger Umkristallisation aus Methylenchlorid Äther bei 178-180 schmilzt.
Process for the manufacture of a new antibiotic

The new strain of the fungal species Penicillium funiculosum Thom grows on a malt yeast extract agar at 18-27 and forms a dense green conidia lawn with bright yellow conidia carriers. The reverse side of the colony shows a greenish-yellow discoloration of the substrate.
The strain corresponds in its morphology and physiology to the description of C. Thom, US.-Dept. Agr. Bur.

One form of procedure for producing the new antibiotic consists in inoculating a liquid nutrient medium with mycelium of a new strain of Penicillium funiculosum Thom and incubating the culture at 27 for 5 to 10 days. The cultivation can take place under aerobic conditions in a surface culture or submerged with shaking or in fermenters with aeration by air or oxygen with stirring. As soon as a maximum amount of antibiotic has been produced, it is filtered and the antibiotic is obtained from the filtrate in a manner known per se by extractive or adsorptive working methods.
One method which has proven to be particularly suitable is the extraction of the filtrate with ethyl acetate, but other organic solvents such as. B. benzene, chloroform, butyl acetate, methylene chloride or butanol can be used. The extracts are then freed from the solvent, e.g. B. by distillation, and the residue for the isolation of the desired antibiotic purified by chromatography on adsorbing agents such as clay, silica gel, magnesium silicate and the like or by means of countercurrent distribution.
The ll-desacetoxy wortmannin has interesting pharmacological properties and is said to find therapeutic use. In particular, the 11-deacetoxy wortmannin has a very high fungistatic effect against various pathogens of fungal infections. In vitro, the 11-deacetoxy-wortmannin is particularly effective against Saccharomyces cerevisiae, Candida albicans, Aspergillus fumigatus, Trichophyton mentagrophytes, Histoplasma capsulatum.
11-Desacetoxy-wortmannin also shows an edema-inhibiting and anti-inflammatory activity, as is evident from the carrageenan edema test, the carrageenan granuloma bag test and the CMC granuloma bag test on the awake rat.
The dose to be administered naturally varies depending on the type of administration and the type of treatment to be treated
