Verfahren zur Herstellung von Phenazin-Derivaten 1,6-Phenazindiol-5,10-dioxyd (Iodinin) ist eine be kannte Verbindung mit einem breiten Spektrum von an tibakteriellen Eigenschaften. Iodinin wurde zuerst aus Chromobacterium Iodinum isoliert. Die Verbindung wurde in der Folge durch Oxydation von 1,6-Dihy- droxyphenazin, welches man durch De-äthylierung von 1,6-Diäthoxyphenazin entsprechend der Wohl-Aue- Reaktion erhält, synthetisiert. Es konnte ein neues Ver fahren zur Herstellung von Analogen von Iodinin aus gehend von einem 1,6-Dialkoxyphenazin, beziehungswei se 1,6-Dibenzyloxyphenazin gefunden werden.

Der Ausdruck Alkyl bezieht sich auf geradkettige gesättigte Kohlenwasserstoffe mit 1 bis 7 Kohlenstoff atomen, wie Methyl, Äthyl, Propyl, n-Propyl, n-Butyl und dgl. Besonders bevorzugt sind geradkettige Alkyl reste mit 1 bis 3 Kohlenstoffatomen d.h. Methyl, Äthyl und Propyl.
Die Analogen von Iodinin werden durch Oxydation eines 1,6-disubstituierten Phenazinderivats der Formel I mit einem Hydroperoxyd hergestellt, wobei man eine Mischung eines N-Mono-oxyds der Formel II und eines N,N'-Dioxyds der Formel III erhält. Diese Mischung kann leicht durch fraktionierte Kristallisation oder Chroma tographie getrennt werden. Hydroperoxyde, die man zur Durchführung dieser Oxydation verwenden kann, sind Wasserstoffsuperoxyd oder Peroxysäuren, insbesondere die organischen Peroxysäuren, wie niedere Alkanoyl- peroxysäuren, z.B. Peressigsäure, Trifluorperessigsäure, Perpropionsäure usw. oder Perbenzoesäuren, z.B. Per benzoesäure, m-Chlorperbenzoesäure usw.
Die Oxyda tion mit einem Hydroperoxyd wird zweckmässigerweise in Gegenwart eines inerten organischen Lösungsmittels, wie einem Kohlenwasserstoff, z.B. Benzol, Toluol oder dgl. durchgeführt. Wenn Wasserstoffsuperoxyd verwendet wird, ist das bevorzugte Lösungsmittel Essigsäure oder eine Lösungsmittelmischung mit Essigsäure. Die Reak tion kann leicht bei Raumtemperatur durchgeführt wer den, wobei die Umsetzung in etwa 15 bis 20 Stunden beendet ist. Höhere oder niedere Temperaturen können ebenfalls angewendet werden mit entsprechender kürze rer oder längerer Reaktionszeit. Die Phenazine der For mel I sind bekannte Verbindungen oder Analoga von bekannten Verbindungen, die leicht mittels der Wohl- Aue-Reaktion zugänglich sind.
Die Ausgangsmaterialien im erfindungsgemässen Verfahren umfassen die sym metrischen Verbindungen, wie die NN-Dioxyde von 1,6-Dimethoxyphenazin, 1,6-Diäthoxyphenazin, 1,6-Di- -n-propyloxyphenazin, 1,6-Dibenzyloxyphenazin, und die unsymmetrischen Derivate, wie die NN-Dioxyde von 1-Methoxy-6-äthoxyphenazin, 1-Methoxy-6-n-propyloxy- phenazin, 1-Äthoxy-6-n-propyloxyphenazin, 1-Benzyloxy- -6-methoxyphenazin, 1-Benzyloxy-6-äthoxyphenazin, 1 -Benzyloxy-6-n-propyloxyphenazin.
Die erfindungsgemässe partielle Dealkylierung bzw. Debenzylierung von Verbindungen der Formel III wird, wie gesagt, durch Behandeln mit einem Halogenid eines Elements der Gruppe III des periodischen Systems, z.B. der Bromide und der Chloride, wie z.B. Bortrichlorid Aluminiumbromid, Aluminiumchlorid und dgl., durch geführt. Sie wird üblicherweise durch Behandeln der Ausgangsverbindung der Formel III mit einem der we niger aktiven Halogenide der Gruppe III oder alternativ dazu durch Anwendung von milderen Reaktionsbedin gungen, z.B. niederen Temperaturen, erreicht. Im allge meinen sind die Chloride der Elemente der Gruppe III weniger aktiv hinsichtlich der Ätherspaltung als die ent sprechenden Bromide und dementsprechend wird die par tielle Ätherspaltung vorzugsweise durch Verwendung eines Chlorids der Gruppe III, z.B. Aluminiumchlorid, bewirkt.
Die Reaktion wird zweckmässigerweise durch Zusatz einer Lösung eines Halogenids der Gruppe III, z.B. Aluminiumchlorid, in einem inerten organischen Lösungsmittel, wie Äther, Tetrahydrofuran, Dioxan und dgl., zu einer Lösung der Verbindung der Formel III in einem unpolaren Lösungsmittel, wie einem Kohlenwas serstoff, z.B. Benzol, Chloroform oder dgl.. unter An- wendung von Raumtemperatur durchgeführt. Die Reak tion ist üblicherweise bei Raumtemperatur in einigen Stunden beendet. Höhere oder tiefere Temperaturen können ebenfalls angewendet werden mit entsprechend niedriger oder höherer Reaktionsdauer.
Die Verbindungen der Formeln II und III sind wertvolle Ausgangsprodukte in der Synthese der Ver bindungen der Formel IV. Zusätzlich besitzen ge wisse dieser Verbindungen ausgeprägte antimikrobielle Aktivität mit breitem Wirkungsspektrum. Verbindungen der Formeln III und IV sind insbesondere aktiv gegen eine Vielzahl gram-positiver und gram-negativer Bakte rien, gegen Hefe, Schimmel, Pilze, Mycobakterien usw. Sie sind zum Beispiel wirksam gegen B. subtilis, E. coli, M. phlei. S. aureus, Ps. Aeruginosa, S. cerevi- siae, P. varioti und C. albicans. Dementsprechend kön nen diese Verbindungen als Germizide eingesetzt werden, insbesondere eignen sie sich als innere Germizide zur Behandlung von Infektionskrankheiten.
Die Verfahrensendprodukte können unter Verwen dung von Hilfsstoffen in übliche pharmazeutische Pro dukte überführt werden. Dazu können z.B. organische oder anorganische inerte Trägermaterialien dienen. Die pharmazeutischen Präparate können in fester Form oder in flüssiger Form vorliegen. Die pharmazeutischen Zu satzstoffe können Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel enthalten, Salze zur Veränderung des osmotischen Druckes und Puffer. Sie können auch ande re therapeutisch wirksame Materialien enthalten.
a) Das Lösungsmittel in 120 ml einer 5%-igen Per benzoesäurelösung in Chloroform wird durch wieder holten Zusatz von Benzol und teilweiser Verdampfung durch Benzol ersetzt. Die etwa 200 ml betragende Ben zollösung wird mit 2,4 g 1,6-Dimethoxyphenazin ver setzt und etwa 15 Stunden bei Raumtemperatur gerührt. Dabei ändert sich die Farbe der Lösung von gelb in rötliches Orange. Die Lösung wird sodann zweimal mit je 100 ml einer 5%-igen wässrigen Natriumcarbonatlö- sung extrahiert. Die Benzolschicht wird mit 100 ml Was ser gewaschen. Die vereinigten wässrigen Extrakte wer den zweimal mit je 100 ml und zweimal mit je 50 ml Chloroform extrahiert. Die vereinigten Chloroformextrak te werden mit 100 ml Wasser gewaschen.
Die vereinig ten Benzol- und Chloroformextrakte werden über Na triumsulfat getrocknet und zur Trockene eingedampft. Der Rückstand wird in etwa 50 ml Chloroform gelöst und an Kieselsäure chromatographiert, wobei 1,6-Di- methoxyphenazin-5,10-dioxyd vom Schmelzpunkt 190- 191 (Zers.) erhalten wird.
In analoger Weise können die folgenden Verbindun gen gewonnen werden: 1,6-Diäthoxyphenazin-5,10-dioxyd 1,6-Dipropyloxyphenazin-5,10-dioxyd 1,6-Dibenzyloxyphenazin-5,10-dioxyd 1,6-Diäthoxyphenazin-5-oxyd 1,6-Dipropyloxyphenazin-5-oxyd 1,6-Dibenzyloxyphenazin-5-oxyd. b) Zu einer Suspension von 4,5 g 1,6-Dimethoxy- phenazin in 500 ml Benzol setzt man eine Lösung von 20 g m-Chlorperbenzoesäure in 500 ml Benzol zu. Die Reaktionsmischung wird 15 Stunden bei Raumtempera tur gerührt. Die erhaltene rote Lösung wird zweimal mit je 100 ml einer 100%-igen Natriumcarbonatlösung gewaschen. Die wässrige Phase wird dreimal mit je 100 ml Chloroform extrahiert.
Die Chloroformextrakte werden mit den Benzolextrakten vereinigt, einmal mit Wasser gewaschen, über Natriumsulfat getrocknet u. ein gedampft. Der Rückstand wird neuerlich in Chloroform gelöst und an Kieselgel chromatographiert, wobei man eine Fraktion von reinem 1,6-Dimethoxyphenazin-5- -oxyd vom Schmelzpunkt 192 (Zers.) und eine zweite Fraktion von reinem 1,6-Dimethoxyphenazin-5,10-dioxyd vom Schmelzpunkt 190-191 (Zers.) erhält.
c) Zu einer Lösung von 50 mg 1,6-Dimethoxyphena- zin-5,10-dioxyd in 5 ml Chloroform setzt man eine Lö sung von 25 mg Aluminiumchlorid in 7 ml Äther zu. Die Reaktionsmischung wird 14 Stunden bei Raumtem peratur gerührt. Sodann verdünnt man die Reaktions mischung mit 30 ml Chloroform und wäscht mit 40 ml Wasser. Die Schichten werden getrennt, und die wäss- rige Schicht fünfmal mit je 30 ml Chloroform extrahiert. Die organischen Schichten werden vereinigt, mit etwas Wasser gewaschen, über Natriumsulfat getrocknet und zur Trockene eingedampft.
Der Rückstand wird in Chlo roform gelöst und an saurem Aluminiumoxyd chroma- tographiert. Nach Entfernung nicht umgesetzten Aus gangsmaterials durch Eluieren mit Chloroform wird die Kolonnenfüllung durch Zusatz einer geringen Menge Wasser deaktiviert und sechsmal mit je 30 ml Chloro form extrahiert. Die erhaltene rötliche Lösung wird zur Trockene eingedampft. Der so erhaltene Rückstand ist reines 1-Hydroxy-6-methoxyphenazin-5,10-dioxyd vom Schmelzpunkt 125-135 (Zers.).
<I>Beispiel 2</I> Zu einer Lösung von 272 mg 1,6-Dimethoxyphenazin- -5,10-dioxyd in 25 ml Chloroform setzt man eine Lösung von 133 mg Aluminiumchlorid in 7 ml Äther zu. Die Reaktionsmischung wird 15 Stunden bei Raumtempera tur gerührt. Sodann verdünnt man die Reaktionsmi schung mit 100 ml Chloroform und wäscht mit 150 ml Wasser. Die wässrige Schicht wird mit<B>100</B> ml und so dann achtmal mit je 50 ml Chloroform extrahiert. Die vereinigten organischen Phasen werden mit 100 ml Was ser gewaschen, über Natriumsulfat getrocknet und zur Trockene eingedampft. Die erhaltene Mischung wird durch Chromatographie an saurem Aluminiumoxyd ge trennt.
Nach Eluieren des Ausgangsmaterials wird das Kolonnenmaterial in 100 ml Chloroform aufgeschlämmt und durch Zusatz von 5 ml Wasser deaktiviert. Das Chloroform wird abdekantiert und das Aluminiumoxyd sechsmal mit je 75 ml Chloroform gewaschen. Die verei nigten organischen Lösungen werden zur Trockene einge dampft und ergeben 1-Hydroxy-6-methoxyphenazin- -5,10-dioxyd vom Schmelzpunkt 125-l35 (Zers.).
<I>Beispiel 3</I> Zu einer Suspension von 10 g 1-Methoxy-6-äthoxy- phenazin in 500 ml Benzol wird eine Aufschlämmung von 40 g m-Chlorperbenzoesäure in 300 ml Benzol zu gesetzt. Die erhaltene Suspension wird sodann 16 Stun den bei Raumtemperatur gerührt und die Reaktionsmi schung von ausgeschiedener m-Chlorperbenzoesäure fil triert. Das Filtrat wird zweimal mit je 150 ml 50%-iger Natriumcarbonatlösung gewaschen. Die wässrigen Schich ten werden weiterhin zweimal mit je 100 ml Chloroform extrahiert. Die vereinigten organischen Schichten werden über Natriumsulfat getrocknet und eingedampft.
Der Rückstand wird in Chloroform gelöst und an Kieselgel chromatographiert, wobei man 1-Methoxy-6-äthoxyphen- azin-5,10-dioxyd vom Schmelzpunkt l55-156 (Zers.), 1-Methoxy-6-äthoxyphenazin-5-oxyd und 1-Methoxy-6- -äthoxyphenazin-10-oxyd erhält.
In analoger Weise können folgende Verbindungen hergestellt werden: 1-Methoxy-6-propyloxyphenazin-5,10-dioxyd 1-Methoxy-6-benzyloxyphenazin-5,10-dioxyd 1-Äthoxy-6-propyloxyphenazin-5,10-dioxyd 1-Äthoxy-6-benzyloxyphenazin-5,10-dioxyd Die Umsetzung der obigen 5,10-Dioxyde mit einem Halogenid eines Elements der Gruppe HI des periodi schen Systems liefert sowohl 1-Hydroxy- wie auch 6- -Hydroxyverbindungen der Formel IV. Beispielsweise erhält man aus 1-Methoxy-6-äthoxyphenazin-5,10-dioxyd sowohl 1-Hydroxy-6-äthoxyphenazin-5,10-dioxyd als auch 1-Methoxy-6-hydroxyphenazin-5,10-dioxyd.
