L'invention concerne un procédé de préparation de composés nouveaux dérivés de l'acide phénoxy-alcoylcarboxylique, de formule générale:

Lorsque le composé obtenu est un acide, Y repré
sentant - OH peut être estérifié, suivant le procédé
classique d'estérification sulfurique, par un alcool comme
par exemple le méthanol et l'éthanol.
Les esters obtenus par le procédé selon l'invention,
Y représentant - OCH5 ou - OCH2 - CH3, sont trans
formés en amide par ébullition avec une amine aroma
tique ou aliphatique.
Les corps obtenus par ces procédés de préparation
sont particulièrement remarquables par leurs applications
thérapeutiques en médication neurotrope, anti-inflamma
toire, normo-lipémiante, par exemple.
L'invention est précisée par la description de la pré
paration de quelques-uns des corps chimiques nouveaux
à titre d'exemple.
1. Synthèse d'acides p-acylphénoxy-alcoyl-carboxyliques
ou p-formyl-phénoxy-alcoyl-carboxyliques:
0,6 mole de NaOH,
0,3 mole de p-hydroxy-benzaldéhyde ou de p-hy
droxy-acétophénone (ou de ses homologues supé
rieurs propiophénone et butyrophénone),
0,3 mole d'acide chloroacétique (ou 3-chloropropio
nique ou 4-chlorobutyrique) sont introduits dans un
ballon contenant 600 ml d'eau.
La solution est portée à ébullition à reflux pendant 8 heures. Après refroidissement, on acidifie à pH 2-3 par
HCI 12 N. L'acide précipite, il est essoré. Le produit obtenu est redissous dans l'éther et extrait par une solution aqueuse d'hydrogénocarbonate de sodium. La solution aqueuse est acidifiée. L'acide recristallise dans l'eau par refroidissement à température ambiante.
Le rendement moyen de cette opération est voisin de 65 /o.
Cette méthode de synthèse - permet d'obtenir cinq types de produits dérivant des acides suivants:
Les acides para-formyl-phénoxy-alcoylcarboxylique, para-acétyl-phénoxy-alcoylcarboxylique, para-propionyl- phénoxy - alcoylcarboxylique, para - butyryl - phénoxy - al- coylcarboxyflque et para-benzoyl-phénoxy-alcoylcarboxylique.
Dans lesdits acides, le terme alcoyl- est représenté par le chaînon
-CH2-, -CH2-CH2- ou - CHp - CHp - CHp -
Pour obtenir les homologues de ces cinq séries pour lesquels le chainon alcoyl est un chaînon iso-butyrique, on peut former in situ le Cl-C - (CH3)2 - CO -Y en présence de

On peut alors procéder comme suit: à 0,1 mole du composé carbonylé p-hydroxy-phénylé dans 100 ml d'acétone, on ajoute 0,5 mole de NaOH pulvérisé et on agite 15 à 30 minutes à l'ébullition. On introduit goutte à goutte une solution de 10 ml de CHAI, dans 20 ml d'acétone anhydre.
Après 4h d'ébullition, on concentre sous pression réduite et acidifie à pH 3 (HC1). On extrait à l'éther et, après les traitements de purification usuels, on obtient l'acide p-acylphénoxy-isobutyrique avec un rendement d'environ 50 O/o.

Les corps obtenus par le procédé selon l'invention, qui présentent une fonction acide, Y étant - OH, peuvent être transformés en esters de la façon suivante:
10 g d'un acide obtenu en I sont dissous dans 150 ml de méthanol, d'éthanol ou de propanol;
150 ml de benzène anhydre, et
1 ml de H±SO4 34 N sont ajoutés successivement dans un ballon. Après ébullition à reflux pendant 3 heures, l'azéotrope benzène-alcool est distillé jusqu'à ce que tout le benzène soit éliiiiné.
La solution alcoolique est ensuite concentrée sous vide. L'huile obtenue est reprise par 200 ml d'éther éthylique, la solution est lavée à l'eau séchée sur Na2S94.
Après concentration sous vide de la solution éthérée,
I'ester est obtenu sous forme d'huile jaune ou sous forme cristallisée suivant le cas.
A titre d'exemple:
L'ester éthylique de l'acide p-formyl-phénoxy-acétique est obtenu dans les conditions ci-dessus: après dissolution de l'acide dans l'éthanol absolu, on obtient des cristaux blanc jaunâtre d'ester dont le point de fusion est 45O C. Rendement 90 /o.
Les résultats analytiques sont les suivants:

L'ester éthylique de l'acide para-acétyl-phénoxy-isobutyrique est obtenu dans les conditions ci-dessus. On obtient un liquide dont le point d'ébullition est à 1330 C sous un vide de 0,05 Hg. Le rendement est de 60 à 70 /o.
3. Utilisation des esters obtenus par le procédé selon
l'invention pour leur transformation en amides
8 g d'un ester obtenu par le procédé selon l'invention sont dissous dans 25 ml environ d'une amine préalablement séchée sur potasse. La solution est portée à ébullition à reflux pendant 3 heures. L'amide cristallise généralernent par simple refroidissement ou par addition d'une faible quantité d'eau. La précipitation complète est obtenue par addition lente de 200 à 300 ml d'eau. La purification est obtenue par recristallisation dans un mélange alcool et eau.
A titre d'exemple:
a) Le pipéridine-amide de l'acide p-formyl-phénoxyacétique est obtenu par le traitement de son ester éthylique par la pipéridine, suivant le procédé ci-dessus; on obtient un produit cristallisé dont le point de fusion est à 960 C, soluble dans l'éther, l'alcool et la plupart des solvants organiques; insolubles dans l'éther de pétrole et l'eau.
Le rendement de la préparation est d'environ 60 /o.

b) Le pipéridine-amide de l'acide p-acétyl-phénoxyacétique est obtenu dans les conditions ci-dessus par un traitement de son ester éthylique avec la pipéridine. On obtient un produit cristallisé dont le point de fusion est à 970 C avec un rendement d'environ 60 /o. Ce produit est soluble dans l'alcool et la plupart des solvants organiques, insoluble dans l'eau, l'éther et l'éther de pétrole.
Les résultats micro-analytiques sont les suivants:

On chauffe 18 heures à reflux une suspension de 150g (1 mole) de para-hydroxy-propiophénone, 276 g (2 moles) de K2CO3 anhydre, 135 g (1,1 mole) de CeCH2CO2 et dans 3200ml d'acétone anhydre.
Après refroidissement, on filtre, concentre le filtrat sous vide et reprend l'huile résiduelle par 1000 mol d'éther; on traite cette phase organique par 2 fois 100 ml de NaOH 4 N, puis on la lave à l'eau jusqu'à neutralité; on sèche ensuite cette phase éthérée, la décolore et élimine le solvant sous vide; I'huile résiduelle est distillée: le parapropionyl-phénoxy-acétate d'éthyle bout à 1460 C/0,05
Torr.
Rendement: 78 O/o.
5. Préparation du p-butyl-phénoxyacétylpipéridine
On chauffe 48 heures à reflux une suspension de 164 g (1 mole) de parahydroxybutyrophénone, 276 g (2 moles) de K2C03 anhydre et 242 g (1,5 mole) de chloracétylpipéridine dans 3200 ml de butanone anhydre.
Après le traitement habituel, similaire à celui décrit dans a où l'on remplace l'éther par du chloroforme, on obtient un résidu qui cristallise peu à peu au froid. Recristallisé dans l'alcool à 95o C, le para-butyryl-phénoxyacétyl-pipéridine fond à 820 C.
Rendement: 58 /o.
Préparation
du p-benzoyl-phénoxyacétyl-morpholide
On chauffe 48 heures à reflux une suspension de 198 g (1 mole) de para-hydroxy-benzophénone, 276 g (2 moles) de K2CO3 anhydre et 245 g (1,5 mole) de chloracétylmorpholide dans 3200 ml de butanone anhydre. Après le traitement habituel on obtient un solide qu'on recristallise dans l'alcool à 95o C, F. 1090 C.
Rendement: 63 O/o.

4. Procédé suivant la revendication I et la sous-revendication 1, caractérisé par le fait que la p-hydroxyphénone est choisie parmi la p-hydroxy-acétophénone, la p-hydroxy-propiophénone, la p-hydroxy-butyrophénone et la p-hydroxy-benzophénone.
5. Procédé suivant la revendication I, caractérisé par le fait que l'on condense le p-hydroxy-benzaldéhyde ou une p-hydroxy-phénone avec l'acide p-chloropropionique ou l'acide v-chlorobutyrique.

Complete precipitation is obtained by slowly adding 200 to 300 ml of water. Purification is obtained by recrystallization from an alcohol and water mixture.

a) The piperidine-amide of p-formyl-phenoxyacetic acid is obtained by the treatment of its ethyl ester with piperidine, according to the above process; a crystalline product is obtained, the melting point of which is at 960 C, soluble in ether, alcohol and most organic solvents; insoluble in petroleum ether and water.
The yield of the preparation is approximately 60 / o.

b) The piperidine-amide of p-acetyl-phenoxyacetic acid is obtained under the above conditions by treatment of its ethyl ester with piperidine. A suspension of 150 g (1 mole) of para-hydroxy-propiophenone, 276 g (2 moles) of anhydrous K2CO3, 135 g (1.1 mole) of CeCH2CO2 and in 3200 ml of anhydrous acetone is heated for 18 hours at reflux.
