Die Oxydation eines Alkohols der Formel II zu einem Keton der Formel I kann so durchgeführt werden, dass man den Alkohol in einem ketonischen Lösungsmittel, wie z. B. Aceton, mit einer Chromat- lösung umsetzt. Die Umsetzung erfolgt bevorzugt bei Raumtemperatur; es können jedoch auch höhere und niedrigere Temperaturen angewendet werden, z. B. Temperaturen zwischen<B>0</B> und<B>100</B> 'C.
Die erfindungsgemäss erhältlichen Verbindungen der Formel I und ihre pharmakologisch verträglichen Säure additionssalze können als antimikrobielle Mittel verwen det werden, besonders gegen Bakterien, Fungi, pathoge ne Hefen und Protozoen, z. B. gegen Trichomonas vaginalis, Trichomonas foetus, Histomonas meleagridis, Endamoeba histolytica, Trypanosomen, z. B. T. cruzi, T. rhodesiense, T. congolense. Die Verbindungen der Formel I können dementsprechend z. B. als Germicide, Trichomonacide, Histomonacide, Trypanocide oder als Mittel zur Bekämpfung von Hefeinfektionen Verwen dung finden.
Für die Behandlung von Infektionskrankheiten, wie Trichomoniasis, Trypanosomiasis oder Histomoniasis können die erfindungsgemäss erhältlichen Verbindungen oral, parenteral oder topisch angewendet werden. Sie können in die üblichen pharmazeutischen Anwendungs formen übergeführt werden, z. B. durch Vermischung mit organischen oder anorganischen Trägermaterialien, wie Wasser, Gelatine, Laktose, Stärke, Magnesium- stearat, pflanzlichen Oelen, Gummi arabicum, Poly- alkylenglykolen, Vaseline. Die pharmazeutischen Präpa rate können in beliebiger Form vorliegen.
Sie können auch noch Hilfsstoffe, wie Konservierungs-, Stabili- sierungs-, Netz- oder Emulgiermittel oder auch Salze zur Veränderung des osmotischen Druckes enthalten. Sie können auch noch andere therapeutisch wertvolle Stoffe enthalten.
Säureadditionssalze von Verbindungen der Formel I können durch Umsetzung mit anorganischen Säuren, wie Salzsäure, Bromwasserstoffsäure, Schwefelsäure, oder mit organischen Säuren, wie Oxalsäure, Essigsäure, Milchsäure, Weinsäure, erhalten werden.
In den nachfolgenden Beispielen sind die Tempera turen in Celsiusgraden angegeben. <I>Beispiel<B>1</B></I> 8 g 1-(2-Nitro-l-imidazolyl)-3-chlor-2-propanol (er hältlich durch Umsetzung von 2-Nitroimidazol mit Epichlorhydrin) werden in 200 ml siedendem Aceton gelöst und die Lösung wird auf<B>29'</B> abgekühlt. Unter Rühren gibt man 10,0 ml einer Cr03-Lösung zu. (Die Cr03-Lösung enthält 266,7 g Cr03 und 230 ml konzen trierte H2S04, die mit Wasser auf 1 Liter verdünnt wurde. 1 ml dieser Lösung enthält somit 2,67 mMol Cr03, was äquivalent mit 4 mMol -CHOH- ist.) Die Temperatur steigt auf<B>36,5'</B> im Verlaufe von<B>15</B> Minuten und fällt dann ab. Je nach 20,<B>30</B> -und<B>50</B> Minuten (totale Reaktionszeit) werden 3 1/3 ml-Portio- nen von Cr03-Lösung zugefügt.
Das Reaktionsgemisch wird total<B>1,5</B> Stunden gerührt, dann werden 200m1 destilliertes Wasser zugefügt. Die Lösung wird auf 20' abgekühlt und auf ein pH von 7,0 neutralisiert durch langsame Zugabe von 9 ml konzentrierter Ammoniaklö sung. Die sich bildende feste Substanz wird filtriert und das Filtrat mit 700 ml Aethylacetat extrahiert. Der Aethylacetatextrakt wird gertocknet und zu einem Oel eingedampft. Beim Behandeln mit Aether verfestigt sich dieses Oel. Die feste Substanz wird in 300 ml siedendem Benzol gelöst und das Dekantat wird ungefähr eine Stunde stehen gelassen. Nach der Filtration erhält man Kristalle von 1-(2-Nitro-l-imidazolyl)-3- chlor-2-propa- non vorn Schmelzpunkt 91,5-9311

<I>Beispiel 2</I> Zu einer gerührten Lösung von 12,25 g 1-(2-Nitro- 1-imidazolyl)-3-methoxy-2-propanol (erhältlich durch Umsetzung von 2-Nitro-imidazol mit 1,2-Epoxy-3-me- thoxy-propan) in 300 ml Aceton fügt man bei 21' 15 ml einer Cr03-Lösung wie in Beispiel 1 beschrieben zu. Die Temperatur steigt dabei im Verlaufe von<B>10</B> Minuten auf<B>35'</B> und fällt dann wieder. Dann werden bei 20 und<B>35</B> Minuten (totale Reaktionszeit) 5-mI-Portionen von Cr03-Lösung zugefügt. Das Reak tionsgemisch wird total eine Stunde gerührt, dann in einem Eisbad abgekühlt und mit<B>300</B> ml destilliertem Wasser versetzt.
Die Lösung wird dann auf<B>181</B> abge kühlt und durch tropfenweise Zugabe von 8 ml konzen trierter Ammoniaklösung unter Rühren auf ein pH von 5 neutralisiert. Die trübe Lösung wird filtriert und das Filtrat mit 1200 ml Aethylacetat extrahiert. Die organi sche Schicht wird getrocknet, filtriert und das Filtrat im Vakuum zu einem Oel eingedampft. Nach Zugabe von 60 ml Aether verfestigt sich das Oel. Die Festsubstanz wird in einem Mörser mit Aether zerrieben, filtriert und aus<B>100</B> ml siedendem Benzol umkristallisiert.
Man erhält so 1-(2-Nitro-l-imidazolyl)-3-methoxy-2-propa- non als Kristalle vomSchmelzpunkt

For the treatment of infectious diseases such as trichomoniasis, trypanosomiasis or histomoniasis, the compounds obtainable according to the invention can be used orally, parenterally or topically. They can be converted into the usual pharmaceutical application forms, eg. B. by mixing with organic or inorganic carrier materials such as water, gelatin, lactose, starch, magnesium stearate, vegetable oils, gum arabic, polyalkylene glycols, petrolatum. The pharmaceutical preparations can be in any form.
They can also contain auxiliaries, such as preservatives, stabilizers, wetting agents or emulsifiers, or else salts for changing the osmotic pressure. They can also contain other therapeutically valuable substances.
Acid addition salts of compounds of the formula I can be obtained by reaction with inorganic acids, such as hydrochloric acid, hydrobromic acid, sulfuric acid, or with organic acids, such as oxalic acid, acetic acid, lactic acid, tartaric acid.
In the following examples, the temperatures are given in degrees Celsius. <I> Example<B>1</B> </I> 8 g of 1- (2-nitro-l-imidazolyl) -3-chloro-2-propanol (can be obtained by reacting 2-nitroimidazole with epichlorohydrin) dissolved in 200 ml of boiling acetone and the solution is cooled to <B> 29 '</B>. 10.0 ml of a Cr03 solution are added with stirring. <I> Example 2 </I> To a stirred solution of 12.25 g of 1- (2-nitro-1-imidazolyl) -3-methoxy-2-propanol (obtainable by reacting 2-nitro-imidazole with 1, 2-epoxy-3-methoxy-propane) in 300 ml of acetone is added at 21 '15 ml of a CrO3 solution as described in Example 1. The temperature rises in the course of <B> 10 </B> minutes to <B> 35 '</B> and then falls again. Then 5 ml portions of Cr03 solution are added at 20 and 35 minutes (total reaction time). The reaction mixture is stirred for a total of one hour, then cooled in an ice bath and mixed with 300 ml of distilled water.
The solution is then cooled to 181 and neutralized to a pH of 5 by adding 8 ml of concentrated ammonia solution drop by drop while stirring. The cloudy solution is filtered and the filtrate is extracted with 1200 ml of ethyl acetate. The organic layer is dried and filtered, and the filtrate is evaporated to an oil in vacuo. After adding 60 ml of ether, the oil solidifies. The solid substance is ground in a mortar with ether, filtered and recrystallized from <B> 100 </B> ml of boiling benzene.
Thus, 1- (2-nitro-1-imidazolyl) -3-methoxy-2-propanone is obtained as melting point crystals
