In den vorstehenden und den folgenden Definitionen steht niederes Alkyl stets für ein solches mit 1-4 Kohlenstoffatomen in gerader oder verzweigter Kette. ( < Nie- deres Acyl bedeutet einen Acylrest (organischen Säurerest) mit bis zu 4 Kohlenstoffatomen, vorzugsweise einen geradkettigen oder verzweigten Alkanoylrest entsprechender Kettenlänge
Entsprechend den oben gegebenen Definitionen kann R beispielsweise bedeuten Methyl, Äthyl, Propyl, Isopropyl, Butyl, Isobutyl, tert.Butyl, Benzyl, a- oder p- Phenyläthyl, , - oder y-Phenylpropyl. Verbindungen, in denen R Methyl oder Benzyl ist und namentlich solche, in denen R Wasserstoff darstellt, sind bevorzugt.
R' kann beispielsweise bedeuten Äthyl, Propyl, Isopropyl, Butyl, Isobutyl, sec. Butyl, geradkettiges oder verzweigtes Amyl (Pentyl), Hexyl, Heptyl oder Octyl; die den genannten Kohlenwasserstoffresten entsprechenden Reste mit einer äthylenischen Doppelbindung wie Allyl oder Crotyl, ferner solche Alkyle mit 2-8 Kohlenstoffatomen, die noch eine Mercaptogruppe tragen wie ,8-Mercaptoäthyl oder höhere Mercaptoalkyle. Ferner kann R' z.B. bedeuten y-Methoxypropyl, 8-Methoxy-n- -butyl, ss-Athoxyäthyl, y-Äthoxypropyl, b-Äthoxybutyl oder höhere Alkyloxyäthyle, -propyle oder -butyle sowie die entsprechenden Gruppen, die statt des Sauerstoffatoms ein Schwefelatom oder das Glied -SO- tragen.
Weiterhin kommen als R' in Frage Benzyl, a-Phenyl äthyl, B-Phenyläthyl, a-, - oder y-Phenylpropyl oder Phenylbutyle
Besonders bevorzugt sind im Sinne der Erfindung solche Verbindungen, die als R' einen cycloaliphatischen, gegebenenfalls mit Alkyl bzw. Alkoxy substituierten oder über Alkylen an das Stickstoffatom gebundenen Kohlenwasserstoffrest enthalten. Als solche Reste seien beispielsweise genannt Cyclopentyl, Cyclohexyl, Cycloheptyl, Cyclooctyl, Methylcyclohexyl, Äthylcyclohexyl, Propyl- und Isopropylcyclohexyl, Methoxycyclohexyl, Äthoxycyclohexyl, Propoxy- und Isopropoxycyclohexyl, wobei die Alkyl- bzw.
Alkoxygruppen in 2-, 3- oder vorzugsweise in 4-Stellung, und zwar sowohl in cis- als auch in trans-Position vorliegen können, Cyclohexylmethyl, a- oder 13-Cyclohexyläthyl, Cyclohexylpropyle, Endomethylencyclohexyl (2,2,1 .Tricycloheptyl), Endoäthylencyclohexyl (2,2,2.Tricyclooctyl), Endomethylencyclohexenyl, Endoäthylencyclohexenyl, Endomethylencyclohexylmethyl, Endoäthylencyclohexylmethyl, Endomethylencyclohexenylmethyl oder Endoäthylencyclohexenylmethyl, a- oder ;,8-Phenylcyclopropyl sowohl in der cis- als auch in der trans-Form.
Endlich sind noch heterocyclische Ringe als R' geeignet, welche ausser 4-5 Kohlenstoffatomen noch 1 Sauerstoff- oder Schwefelatom und bis zu 2 Doppelbindungen enthalten können und gegebenenfalls an das benachbarte Stickstoffatom durch eine Methylen-Gruppe gebunden sein können. Beispiele solcher heterocyclischen Ringe sind:

X ist definitionsgemäss eine einfache chemische Bindung oder ein Brückenglied aus 1-6 Kohlenstoff-Atomen, die einer Kohlenwasserstoffkette angehören und gegebenenfalls einer der Gruppen -O-, -S-, -SO- oder -SO2-. Diese zuletzt genannten Gruppierungen können sowohl die Kohlenwasserstoffkette unterbrechen als auch zwischen dieser und dem Phenylkern stehen. Die Gruppen -Ooder -S- können sich ausserdem auch zwischen der Kohlenwasserstoffkette und der Carbonylgruppe befinden.
Demgemäss kommen als X beispielsweise in Frage: -CH2-, -CH2-CH2-, -CH(CH3)-, -CH2-CH2-CH2-, -CH (CH3)-CH2-, -CH2-CH(CH3)-, -C(CH8), -CHACH2- CH2-CH2-, -CH(CH3)-CH2-CH2-, -CH2-CH(CH3)-CH2-, -CH2-CH2-CH(CH3) -CH(CH3)-CH(CHs) -, -C(CH,)2- -CH2-, -CH2-C(CHa)2-, -CH(C2H5)-, -C(C2H5)2-, -C(CH3)- (C2H5)- sowie entsprechende geradkettige oder verzweigte Pentylen- oder Hexylen-Brücken, ferner ungesättigte Glieder wie -CH=CH-, -CH2-CH=CH-, -CH=CH -CH2-, -C(CH3)=CH-, -CH=C(CH,)- oder höhere Glieder gleichen Aufbaus, die auch mehrere Doppelbindungen tragen können.
Beispiele für Glieder, die noch eine der oben genannten Heterogruppen enthalten, sind -0 -CH2-, -O-CH2-CH2-, -O-CH2-CH2-CH2-, -O-CH(CH3)-, CH2-O-, -S-CH2-, -S-CH2-CH2-, -CH2-S-CH2-, -S-CH (CH3)-, -SQ2-CH2-, -O-C(C2H5)2-.
Endlich kann dieses Brückenglied auch cyclisch aufgebaut sein und beispielsweise den l,l-Cyclopentyliden Rest

Y stellt Kohlenwasserstoffrest mit 1-4 Kohlenstoff Atomen dar, der geradkettig oder verzweigt sein kann.
Als BeisPiele seien die im vorigen Absatz für X definierten Glieder genannt soweit sie unter die allgemeine Definition von Y fallen.
Beispiele für die Substituenten Z, Z' und Z" sind Methyl, Äthyl, Propyl, Isopropyl, n-, Iso- oder tert.Butyl sowie die entsprechenden Alkoxy-Gruppen, Fluor, Chlor, Brom, Jod, Cyclohexyloxy, Cyclopentyloxy, Methyl-, Äthyl, Propyl- oder Butylmercapto, -sulfinyl oder -sulfonyl mit geradkettigen oder verzweigten Alkylresten, Benzyl, Phenyläthyl, Phenylpropyl, Acetyl, Propionyl, Butyryl, Acetoxy, Propionyloxy, Butyryloxy, Carbonmethoxy, Carboäthoxy, Carbopropoxy, Methylcarbamyl, Äthylcarbamyl, Dimethyl- oder Diäthylcarbamyl.
Der in der Formel mit =phenylen- bezeichnete Phenylenrest kann unsubstituiert oder vorzugsweise ein- oder mehrfach mit Halogen, niederem Alkyl oder niederem Alkoxy substituiert sein. Er kann die restlichen Teile des Moleküls in o-, m- oder p-Stellung zueinander tragen, wobei die pStellung bevorzugt ist.

einführen und die Verfahrensprodukte gegebenenfalls zur Salzbildung mit alkalischen Mitteln behandeln kann.
Je nach der Natur der Glieder Z, Z', Z", X und R' wird in einzelnen Fällen das genannte Verfahren für die Herstellung der unter die allgemeine Formel fallenden individuellen Verbindungen ungeeignet sein oder zumindest Vorkehrungen zum Schutz aktiver Gruppen notwendig machen. Derartige verhältnismässig selten auftretende Fälle können vom Fachmann unschwer erkannt werden, und es bereitet keine Schwierigkeiten, in solchen Fällen die entsprechenden Massnahmen zu treffen. So ist es unter Umständen erforderlich, anstelle von Z stehende Hydroxygruppen durch Veresterung oder Ver ätherung, an entsprechender Stelle stehende Carboxygruppen durch Veresterung oder eine zur Carbonamidgruppe in o-Stellung stehende Carboxygruppe auch durch Ringschluss zum Phthalimidderivat zu schützen. Ähnliches gilt in den Fällen, in denen R' eine Mercaptoalkylgruppe darstellt.
Die Ausführungsformen des Verfahrens gemäss der Erfindung können im allgemeinen hinsichtlich der Reaktionsbedingungen weitgehend variiert und den jewei- ligen Verhältnissen angepasst werden.
Beispielsweise können die Umsetzungen unter Verwendung von Lösungsmitteln, bei Zimmertemperatur oder bei erhöhter Temperatur durchgeführt werden.

Die nach dem Verfahren gemäss der Erfindung erhältlichen Benzolsulfonylharnstoff-Derivate stellen wertvolle Arzneimittel dar, die sich durch eine starke und vor allem lang anhaltende blutzuckersenkende Wirkung auszeichnen. Ihre blutzuckersenkende Wirkung konnte z.B. am Kaninchen dadurch festgestellt werden, dass man die Verfahrensprodukte in den üblichen Dosen von 400 mg/kg verfütterte und den Blutzuckerwert nach der bekannten Methode von Hagedorn-Jensen über eine längere Zeitdauer bestimmte.
So wurde z.B. ermittelt, dass der N-[4-(f3-Benzamido- äthyl)-benzolsulfonyl] -N' -cyclohexyl-harnstoff eine maximale Blutzuckersenkung (gemessen nach 6 Stunden) von 32% bewirkt. Nach 24 Stunden beträgt diese immer noch 32%. Durch Verabreichung von N-[4-(k-Benzami- doäthyl)-benzolsulfonyl] - N'- (4'-methylcyclohexyl)-harn- stoff wird der Blutzuckerspiegel maximal um 34% erniedrigt, nach 24 Stunden beträgt die Senkung noch 31% und nach 48 Stunden noch 20%. Demgegenüber bewirkte beim Vergleichsversuch der als orales Antidiabetikum bekannte und weltweit als Arzneimittel verwendete N -(4-Methyl-benzolsulfonyl)-N'-butyl-harnstoff in der oben angegebenen Dosierung zwar eine maximale Blutzukkersenkung von 40%, die aber nach 24 Stunden wieder auf 0 zurückgegangen war.
Nach Verabreichung der wesentlich niedrigeren Dosierung von 50 mg/kg Kaninchen, welche eine differenziertere Bestimmung der blutzuckersenkenden Wirksamkeit erlaubt und der therapeutischen Dosierung näherkommt, wurde gefunden, dass der neue N-[4-q-Benzamido-äthyl)-benzolsulfonyl] -N'-cyclohexylharnstoff nach 24 Stunden eine Blutzukkersenkung von 17% und nach 48 Stunden noch eine von 11% und der neue N-[4-P-Benzamidoäthyl)-benzol- sulfonyl]-N'-(4' - methylcyclohexyl)-harnstoff nach 24 Stunden sogar noch eine Senkung von 21% bewirkt, während die blutzuckersenkende Wirksamkeit des bekannten N-(4-Methyl-benzolsulfonyl)-N'-n-butylharnstoffes bei gleicher Dosierung nach 24 Stunden auf 0 abgesunken ist.
Weiterhin wurde beispielsweise ermittelt, dass 50 mg/kg des N-[4-(p-Phenoxy-acetamidoäthyl)-benzolsul- fonyl]-N'-cyclohexyl-harnstoffs beim Kaninchen nach 6 Stunden eine Blutzuckersenkung von 40 < 70 bewirkt, während der Blutzuckerspiegel bei Verabreichung des bekannten N - (4 - Methyl - benzolsulfonyl)-N'-n-butyl-harnstoffs um 30% vermindert wird.
Die starke Wirksamkeit der Verfahrensprodukte wird besonders deutlich, wenn man die Dosis verringert. Verabreicht man den N-[4-Q-Phenoxyacetamidoäthyl)-ben- zolsulfonyl]-N'-cyclohexyl-harnstoff oder den N-[4-(,8- -Phenylacetamidoäthyl) - benzolsulfonyl] - N' - cyclohexyl -harnstoff in Dosierungen von 1 mg/kg an Kaninchen, so ist immer noch eine deutliche Blutzuckersenkung festzustellen, während der bereits erwähnte N-(4-Methyl -benzolsulfonyl)-N'-n-butyl-harnstoff bei einer Dosis von weniger als 25 mg/kg am Kaninchen nicht mehr wirk samist.
Verabreicht man den N-[4- 3-Chlor-4-methyl-benz- amidoäthyl) - benzolsulfonyl] - N' - (4 - methyl-cyclohexyl) -harnstoff in einer Dosierung von 0,2 mg/kg, den N-[4 -(p- 3,5 - Dimethoxy-benzamidoäthyl) - benzolsulfonyl]-N' -cyclohexyl-harnstoff in einer Dosierung von 0,3 mg/kg oder den N-[4-(X-3,4-Dichlor-benzamidoäthyl)-benzolsul- fonyl]-N'-cyclohexyl-harnstoff in einer Dosierung von 0,3 mg/kg, den N-[4-(ss-2-Hydroxybenzamidoäthyl)-ben- zolsulfonyl]-N'-n-butylharnstoff in einer Dosierung von 2 mg/kg oder den N-[4-(P-2-Hydroxybenzamidoäthyl)- -benzolsulfonyl]-N'-cyclohexyl-harnstoff in einer Dosierung von 0.15 mg/kg an Kaninchen, so ist immer noch eine deutliche Blutzuckersenkung festzustellen.
In der folgenden Tabelle werden für eine Anzahl von Verbindungen, die Gegenstand der vorliegenden Erfindung sind, diejenigen Dosierungen in mg/kg angegeben, die bei Kaninchen nach oraler Gabe gerade noch eine eindeutig erkennbare Senkung des Blutzuckerspiegels bewirken (aSchwellendosis ). Zum Vergleich sei darauf hingewiesen, dass die Schwellendosis für den bekannten N-(4-Methyl-benzolsulfonyl)-N'-n-butyl-harn- stoff etwa 25 mg/kg beträgt.

Die beschriebenen Benzolsulfonylharnstoffe sollen vorzugsweise zur Herstellung von oral verabreichbaren Präparaten mit blutzuckersenkender Wirksamkeit zur Behandlung des Diabetes mellitus dienen und können als solche oder in Form ihrer Salze bzw. in Gegenwart von Stoffen, die zu einer Salzbildung führen, appliziert werden.
Zur Salzbildung können beispielsweise herangezogen werden: Alkalische Mittel, wie Alkali- oder Erdalkalihydroxyde, -carbonate oder -bicarbonate, aber auch organische Basen, insbesondere tertiäre Stickstoffbasen, vorausgesetzt, dass sie physiologisch verträglich sind.
Als medizinische Präparate kommen vorzugsweise Tabletten in Betracht, die neben den Verfahrenserzeugnissen die üblichen Hilfs- und Trägerstoffe, wie Talkum, Stärke, Milchzucker, Traganth oder Magnesiumstearat, enthalten.
Ein Präparat, das die beschriebenen Benzolsulfonylharnstoffe als Wirkstoff enthält, z.B. eine Tablette oder ein Pulver mit oder ohne die genannten Zusätze, ist zweckmässig in eine geeignet dosierte Form gebracht.
Als Dosis ist dabei eine solche zu wählen, die der Wirksamkeit des verwendeten Benzolsulfonylharnstoffs und dem gewünschten Effekt angepasst ist. Zweckmässig beträgt die Dosierung je Einheit etwa 0,5 bis 100 mg.
vorzugsweise 2 bis 10 mg, jedoch können auch erheblich darüber oder darunter liegende Dosierungseinheiten verwendet werden, die gegebenenfalls vor Applikation zu teilen bzw. zu vervielfachen sind.
Beispiel 1
N- [4-(Benamido-nwthytY ben;olsnlfony1\- N' - -cyclohexyl- harnstoff
2,2 g N- [4-(Aminomethyl)-benzolsulfonyl]-N'-cyclohexyl-harnstoff (Schmelzpunkt 1990) werden mit 1,1 g Pyridin und 1,0g Benzoylchlorid in 10ml Chloroform 6 Stunden auf 350 erhitzt. Beispiel 2 N-[4-(P-4' -ChEorbenzamido-äthyI!- bentolsulfonyl] -N' -eyclohexyl- harnstoff 3 g N- [4- (p - Aminoäthyl)-benzolsulfonyl]-N'-cyclohexyl-harnstoff (Schmelzpunkt 2030) werden mit 1,6 g p-Chlorbenzoylchlorid und 1,5 g Pyridin in 20 ml.
