Entsprechend den oben gegebenen Definitionen kann R beispielsweise bedeuten Methyl, Äthyl, Propyl, Isopropyl, Butyl, Isobutyl, tert.-Butyl, Benzyl, a- oder ss-Phenyläthyl, a-, ss- oder y-Phenylpropyl, Verbindungen, in denen R Methyl oder Benzyl ist und namentlich solche, in denen R Wasserstoff darstellt, sind bevorzugt.
R1 kann beispielsweise bedeuten Äthyl, Propyl, Isopropyl, Butyl, Isobutyl, sec.-Butyl, geradkettiges oder verzweigtes Amyl (Pentyl), Hexyl, Heptyl oder Octyl; die den genannten Kohlenwasserstoffresten entsprechenden Reste mit einer äthylenischen Doppelbindung wie Allyl oder Crotyl, ferner solche Alkyle mit 2-8 Kohlenstoffatomen, die noch eine Mercaptogruppe tragen wie ,6-Mercaptoäthyl oder höhere Mercaptoalkyle. Ferner kann R1 z. B. bedeuten d-Methoxypropyl, d-Methoxy- n-butyl, ss-Athoxyäthyl, d-Athoxypropyl, 8-Athoxybutyl oder höhere Alkyloxyäthyle, -propyle oder -butyle sowie die entsprechenden Gruppen, die statt des Sauerstoffatoms ein Schwefelatom oder das Glied -SO- tragen.
Weiterhin kommen als R1 in Frage Benzyl, a-Phenyl äthyl, fi-Phenyläthyl, a-, ss- oder 8-Phenylpropyl oder Phenylbutyle.
Besonders bevorzugt sind im Sinne der Erfindung solche Verbindungen, die als R1 einen cycloaliphatischen, gegebenenfalls mit Alkyl- bzw. Alkoxy substituierten oder über Alkylen an das Stickstoffatom gebundenen Kohlenwasserstoffrest enthalten. Als solche Reste seien beispielsweise genannt Cyclopentyl, Cyclohexyl, Cycloheptyl, Cyclooctyl, Methylcyclohexyl, Äthylcyclohexyl, Propyl- und Isopropylcyclohexyl, Methoxycyclohexyl, Äthoxycyclohexyl, Propoxy- und Isopropoxycyclohexyl, wobei die Alkyl- bzw.
Alkoxygruppen in 2-, 3- oder vorzugsweise in 4-Stellung, und zwar sowohl in cis- als auch in trans-Position vorliegen können, Cyclohexylmethyl, a- oder B-Cyclohexyläthyl, Cyclohexylpropyle, Endomethylencyclohexyl (2,2,1 -Tricyclo- heptyl), Endoäthylencyclohexyl (2,2,2-Tricyclooctyl), Endomethylencyclohexenyl, Endoäthylencyclohexenyl, Endomethylencyclohexylmethyl, Endoäthylencyclohexylmethyl, Endomethylencyclohexenylmethyl oder Endo äthylencyclohexenylmethyl, a- oder ss-Phenylcyclopropyl sowohl in der cis- als auch in der trans-Form.
Endlich sind noch heterocyclische Ringe als R' geeignet, welche ausser 4-5 Kohlenstoffatomen noch 1 Sauerstoff- oder Schwefelatom und bis zu 2 Doppelbindungen enthalten können und gegebenenfalls an das benachbarte Stickstoffatom durch eine Methylen-Gruppe gebunden sein können. Der in der Formel mit -phenylen- bezeichnete Phenylenrest kann vorzugsweise unsubstituiert oder einoder mehrfach mit Halogen, niederem Alkyl oder niederem Alkoxy substituiert sein. Er kann die restlichen Teile des Moleküls in o-, m- oder p-Stellung zueinander tragen, wobei die p-Stellung bevorzugt ist.
Die Herstellung der genannten Benzolsulfonylharnstoffe kann nach Methoden erfolgen, die allgemein für die Herstellung von Verbindungen dieser Klasse angewandt werden. So kann man in entsprechend substituierten Benzolsulfonylthioharnstoffen das Schwefelatom durch ein Sauerstoffatom ersetzen und die Verfahrensprodukte gegebenenfalls zur Salzbildung mit alkalischen Mitteln behandeln.
Je nach der Natur der Glieder X und Rt wird in einzelnen Fällen das eine oder andere der genannten Verfahren für die Herstellung der unter die allgemeine Formel fallenden individuellen Verbindungen ungeeignet sein oder zumindest Vorkehrungen zum Schutz aktiver Gruppen notwendig machen. Derartige verhältnismässig selten auftretende Fälle können vom Fachmann unschwer erkannt werden, und es bereitet keine Schwierigkeiten, in solchen Fällen einen anderen der beschriebenen Synthesewege erfolgreich anzuwenden.
Der Ersatz des Schwefelatoms durch ein Sauerstoffatom in den entsprechend substituierten Benzolsulfonylthioharnstoffen kann beispielsweise mit Hilfe von Oxyden oder Salzen von Schwermetallen oder auch durch Anwendung von Oxydationsmitteln, wie Wasserstoffperoxyd, Natriumperoxyd oder salpetriger Säure, ausgeführt werden. Die Thioharnstoffe können auch entschwefelt werden durch Eehandlung mit Phosgen oder Phosphorpentachlorid. Als Zwischenstufe erhaltene Chlorameisensäureamidine bzw. -carbodiunide können durch geeignete Massnahme wie Verseifen oder Anlagerung von Wasser in die Benzolsulfonylharnstoffe überführt werden.
Die Ausführungsformen des Verfahrens gemäss der Erfindung können im allgemeinen hinsichtlich der Reaktionsbedingungen weitgehend variiert und den jeweiligen Verhältnissen angepasst werden.
Beispielsweise können die Umsetzungen unter Verwendung von Lösungsmitteln, bei Zimmertemperatur oder bei erhöhter Temperatur, durchgeführt werden.
Die nach dem Verfahren gemäss der Erfindung erhältlichen Benzolsulfonylharnstoff-Derivate stellen wertvolle Arzneimittel dar, die sich durch eine starke und vor allem lang anhaltende blutzuckersenkende Wirkung auszeichnen. Ihre blutzuckersenkende Wirkung konnte z. B. am I(aninchen dadurch festgestellt werden, dass man die Verfahrensprodukte in den üblichen Dosen von 10 mg/kg verfütterte und den Blutzuckerwert nach der bekannten Methode von Hagedorn Jensen oder mit einem Autoanalyzer bestimmte.
So wurde beispielsweise ermittelt, dass der
N-[4-(ss-Carbobenzoxyamido-äthyl)-benzol sulfonyl]-N'-cyclohexyl-harnstoff eine Blutzuckersenkung von 30S, der N-[4-(ss-Carbomethoxyamido-äthyl)-benzol- sulfonyl] -N'-cyclohexyl-harnstoff eine Blutzuckersenkung von 37 % nach jeweils 3 Stunden bewirkt. Demgegenüber ist beim Vergleichsversuch der als orales Antidiabetikum bekannte und weltweit als Arzneimittel verwendete
N-(4-Methyl-benzolsulfonyl)-N'-butyl-harnstoff bei Dosierungen unter 25 mg/kg am Kaninchen unwirksam.
Die starke Wirksamkeit der Verfahrensprodukte wird besonders deutlich, wenn man die Dosis weiter verringert. Verabreicht man den N-[4-(ss-Carbobenzoxyamido-äthyl)-benzol- sulfonyl] -N'-cyclohexyl-harnstoff in einer Dosis von 0,2 mg/kg oder den N-4-(fl-Carbomethoxyamido-äthyl)-benzol- sulfonyl] -N'-(4-methyl-cyclohexyl)-harnstoff in einer Dosis von 0,3 mg/kg oder den N'-[4-(i3-Carbäthoxy-amido-äthyl)-benzol sulfonyl]-N'-(4-meíhyl-cyclohexyl) -harnstoff in einer Dosierung von 0,1 mg/kg, so ist immer noch eine deutliche Blutzuckersenkung festzustellen.
Die beschriebenen Benzolsulfonylharnstoffe sollen vorzugsweise zur Herstellung von oral verabreichbaren Präparaten mit blutzuckersenkender Wirksamkeit zur Behandlung des Diabetes mellitus dienen und können als solche oder in Form ihrer Salze bzw. in Gegenwart von Stoffen, die zu einer Salzbildung führen, appliziert werden. Zur Salzbildung können beispielsweise herangezogen werden: Alkalische Mittel, wie Alkali- oder Erdalkalihydroxyde, -carbonate oder -bicarbonate, aber auch organische Basen, insbesondere tertiäre Stickstoffbasen, vorausgesetzt, dass sie physiologisch verträglich sind.
Als medizinische Präparate kommen vorzugsweise Tabletten in Betracht, die neben den Verfahrenserzeugnissen die üblichen Hilfs- und Trägerstoffe, wie Talkum, Stärke, Milchzucker, Tragant oder Magnesiumstearat, enthalten.
Ein Präparat, das die beschriebenen Benzolsulfonylharnstoffe als Wirkstoff enthält, z. B. eine Tablette oder ein Pulver mit oder ohne die genannten Zusätze, ist zweckmässig in eine geeignete dosierte Form gebracht.
Als Dosis ist dabei eine solche zu wählen, die der Wirksamkeit des verwendeten Benzolsulfonylharnstoffs und dem gewünschten Effekt angepasst ist. Zweckmässig beträgt die Dosierung je Einheit etwa 0,5 bis 100 mg, vorzugsweise 2 bis 10 mg, jedoch können auch erheblich darüber oder darunter liegende Dosierungseinheiten verwendet werden, die gegebenenfalls vor Applikation zu teilen bzw. zu vervielfachen sind.

N-[4-(ss-Carbomethoxy-amido-äthyl)-benzol sulfonyl] -N'-isobutyl-harnstoff .

N-[4-(ss-Carbomethoxy-amido-äthyl)-benzol- sulfonyl]-N'-isobutylthioharnstoff werden in 10 ml ln NaOH gelöst. Nach Zusetzen von 2 ml H202 (30%) erhitzt man die Lösung 3 Minuten auf dem Dampfbad. Nach dem Erkalten wird mit 50 ml Wasser verdünnt und mit Salzsäure angesäuert.
Man erhält ein Kristallisat von N- [4-(p-Carbomethoxy-amido-äthyl) -benzol- sulfonyl]-N'-isobutylharnstoff, das nach dem Umkristallisieren aus Methanol bei 145 bis 1470 C schmilzt.
Beispiel 2 N-[4-(ÄCarbomethoxy-amido-äthyl)-benzol sulfonyl] -N'-Äphenyläthyl-harnstoff.
11 g N-[4-(ss-Carbomethoxy-amido-äthyl) -benzol- sulfonyl]-N'-ss-phenyläthyl-thioharnstoff, hergestellt aus
4-(Carbomethoxy-amido-äthyl)-benzolsulfon-amid und ss-Phenyläthylsenföl, Schmelzpunkt 122-124 C, werden in 100 ml 1n NaOH gelöst. Nach Zusatz von 5,4 g HgO rührt man unter Erhitzen auf etwa 500 C 4 Stunden.
Man saugt ab, säuert das Filtrat mit Salzsäure an, saugt ab, trocknet, behandelt mit Essigester und kristallisiert den erhaltenen Niederschlag von N-[4-(fl-Carbomethoxy-amido-äthyD-benzol- sulfonyl]-N'-ss-phenyläthyl-harnstoff aus Methanol um. (Schmelzpunkt 132-134 C.)
Analog dem vorigen Beispiel erhält man aus N-[4-(p-Carbomethoxy-amido-äthyl)-benzol sulfonyl] -N'-cyclooctyl-thioharnstoff vom Schmelzpunkt 132-1340 C (erhalten aus 4-(ss-Carbomethoxy-amido-äthyl)-benzolsulfonamid und Cyclooctylsenföl) durch Behandlung mit H202 in alkalischer Lösung den N-[4-(/3-Carbomethoxyamido-äthyl)-benzol- sulfonyl]-N'-cyclooctyl-harnstoff vom Schmelzpunkt 148-150 C aus Methanol.

The benzenesulfonylureas mentioned can be prepared by methods which are generally used for the preparation of compounds of this class.
