Verfahren zur Herstellung neuer Derivate der 7-Aminocephalosporansäure
Gegenstand der Erfindung ist ein Verfahren zur Herstellung neuer therapeutisch wirksamer Derivate der 7-Amino-cephalosporansäure der Formel I

worin R1 ein 5-gliedriger Heterocyclylrest ist, der mindestens 2 Heteroatome aus der Gruppe Stickstoff, Sauerstoff und Schwefel enthält und der in 5-Stellung unsubstituiert oder mercaptosubstituiert ist und in den übrigen Stellungen unsubstituiert oder durch Niederalkyl- oder Niederalkoxygruppen oder die Hydroxyl- gruppe substituiert ist und der mit einem direkt zwischen 2 Heteroatomen liegenden Kohlenstoffatom an das Schwefelatom der Mercaptoacetylgruppe gebunden ist, und worin R. eine freie oder durch eine Carbonsäure, Thiocarbonsäure oder Dithiocarbonsäure veresterte Hydroxylgruppe,
eine gegebenenfalls N-substituierte Carbamoyloxygruppe oder eine quaternäre Aminogruppe ist, und ihrer gegebenenfalls inneren Salze.
R1 ist vorzugsweise mindestens teilweise ungesättigt.
So ist R1 beispielsweise Imidazolyl(2), Imidazolinyl(2), Oxazolyl(2), Oxazolinyl(2), Thiazolyl(2), Thiazolinyl(2), 1,2,4-Triazolyl(3), 1,3 ,4-Dithiazolyl(2), oder 1,2,4 Thiadiazolyl(3). Die Reste können auch substituiert sein, insbesondere durch Niederalkyl, Niederalkoxy, Hydroxy, Mercapto, Halogen.
Eine veresterte Hydroxylgruppe R.2 leitet sich von einer Carbonsäure, Thio- oder Dithiocarbonsäure ab und ist vorzugsweise die Acetoxygruppe oder z. B. durch Niederalkyl-, Niederalkoxy- oder Niederalkylmercaptoreste, Halogenatome oder die Nitrogruppe substituierte mono- oder dicyclische Arylcarbonyloxy-, Arylcarbonylmercapto- oder Arylthiocarbonylmercaptogruppe, insbesondere die Benzoylmercaptogruppe. Als weitere Bei spiele für R., sind zu zu nennen:
: a) eine Carbamoyloxygruppe der Formel -O-CO-NH-RJ worin Rs ein aliphatischer, aromatischer, araliphatischer oder heterocyclischer Rest, besonders ein unsubstituierter oder substituierter, vorzugsweise durch eine oder mehrere Niederalkoxygruppen oder Halogenatome substituierter, gerader oder verzweigter Niederalkylrest, wie der Methyl-, Aethyl-, vor allem aber der ,ss-Chloräthyl- rest, ist; oder b) eine quaternäre Aminogruppe, in der das quaternäre Stickstoffatom z. B. Teil eines aromatischen Ringes, wie eines Chinolin-, Isochinolin- oder Pyrimidinringes, insbesondere aber eines unsubstituierten oder substituierten Pyridinringes, z.

worin R5 für Wasserstoff oder eine oder mehrere Niederalkyl-, Niederalkoxvcarbonyl-, Carbamoyl- oder Carboxylgruppen oder ein oder mehrere Halogenatome steht.
Die Salze der neuen Verbindungen sind Metallsalze, vor allem solche von therapeutisch anwendbaren Alkalioder Erdalkalimetallen, wie Natrium, Kalium, Ammonium, Calcium, oder Salze mit organischen Basen, z. B.
Triäthylamin, N-Aethyl-piperidin, Dibenzyläthylen diarnin, Procain. Ist die Gruppe R2 basisch, so können sich innere Salze bilden.
Die neuen Verbindungen weisen eine besonders gute antibakterielle Wirkung auf. Sie sind sowohl gegenüber gram-positiven wie vor allem auch gegenüber gramnegativen Bakterien wirksam, z. B. gegen Staphylococcus aureus penicillinresistent, Escherichia coli, Klebsiella pneumoniae, Salmonella typhosa und Bacterium proteus, wie sich auch im Tierversuch, z. B. an Mäusen, zeigt. Sie können daher zur Bekämpfung von Infektionen, die durch solche Mikroorganismen hervorgerufen werden, verwendet werden, ferner als Futtermittelzusätze, zur Konservierung von Nahrungsmitteln oder als Desinfektionsmittel. Besonders wertvoll sind Verbindungen, in denen R1 Imidazolyl(2), Imidazolinyl(2), Thiazolyl(2), Thiazolinyl(2) oder Triazolyl(2) und R2 die Acetoxygruppe, die ss-Chloräthylcarbamoyl- oder eine unsubstituierte oder, wie oben angegeben, substituierte Pyridiniogruppe ist.
Die neuen Verbindungen werden erhalten, wenn man eine Verbindung der Formel II

worin Z für einen Halogenacetylrest wie den Fluor-, Chlor-, Jod- oder vor allem Bromacetylrest steht und R.1 die oben angegebene Bedeutung hat, mit einer Mercaptoverbindung der Formel RI--SH worin R1 die oben genannte Bedeutung hat, umsetzt.
Man kann erhaltene Verbindungen, worin R., für die Acetoxygruppe steht, in Verbindungen, worin R2 für eine Hydroxylgruppe steht, überführen, indem man sie mit einem desacetylierenden Mittel behandelt. Verbindungen, worin R; für eine Hydroxylgruppe steht, können mit einer anderen Carbonsäure als Essigsäure oder mit einer Thio- oder Dithiocarbonsäure acyliert werden. Man kann erhaltene Verbindungen, in denen R, für eine Hydroxylgruppe steht, durch Umsetzung mit einem Isocyansäureester in Verbindungen überführen, worin R eine N-substituierte Carbamoyloxygruppe bedeutet.
Erhaltene Verbindungen, in denen Ra für eine Carbonsäure oder Thiocarbonsäure steht, können durch Umsetzung mit einem tertiären Amin, besonders Pyridin, in Verbindungen übergeführt werden, in denen R; eine quaternäre Aminogruppe bedeutet. Wenn erwünscht, kann man die erhaltenen Verbindungen in ihre Metall-, wie Alkali- oder Erdalkalimetallsalze, oder Salze mit Ammoniak oder organischen Basen überführen oder aus erhaltenen Salzen die freien Carbonsäuren oder gegebenenfalls inneren Salze bilden.
Die Umsetzung der Verbindung II, worin Z für eine Halogenacetylgruppe steht, mit der Mercaptoverbindung wird vorzugsweise in einem inerten Lösungsmittel für die Verbindung wie Methylenchlorid, Chloroform, Tetrachlorkohlenstoff, Tetrahydrofuran, Dioxan, Dimethylformamid oder Acetonitril und in Gegenwart eines halogenwasserstoffbindenden Mittels, z. B. einer schwachen anorganischen Base wie eines Alkalicarbonates, -bicarbonates oder -acetates oder eines tertiären Amins, vorzugsweise Aethyldiisopropylamin (Hünigbase) vorgenommen. Die Reaktion findet bei Zimmertemperatur innert einiger Stunden statt; sie kann, wenn erwünscht, auch bei niedrigerer oder leicht erhöhter Temperatur durchgeführt werden.
Vorzugsweise verwendet man solche Ausgangsstoffe, die zu den erwähnten besonders wirksamen Endprodukten führen.
Die als Ausgangsstoffe verwendeten Cephalosporinderivate sind bekannt oder können nach an sich bekannten Verfahren hergestellt werden.
Die neuen Verbindungen können als Heilmittel, z. B.
in Form pharmazeutischer Präparate, Verwendung finden. Diese enthalten die Verbindungen in Mischung mit einem für die enterale, topicale oder parenterale Applikation geeigneten pharmazeutischen organischen oder anorganischen, festen oder flüssigen Trägermaterial.
In den folgenden Beispielen sind die Temperaturen in Celsiusgraden angegeben.
Für 7-Aminocephalosporansäure wird im folgenden die Abkürzung 7-ACA gebraucht.
In der Dünnschichtchromatographie auf Silicagelplatten werden die folgenden Systeme verwendet: System 52 = n-Butanpl-Eisessig-Wasser (75:7,5:21) System 101 A = n-Butanol-Pyridin-Eisessig-Wasser (42:24:4:30).
Beispiel I
3,93 g (10 mMol) Bromacetyl-7ACA werden in 20 ml Methylenchlorid unter Zugabe von 3,45 ml (20 mMol) N-Aethyl-diisopropylamin gelöst. Dazu gibt man eine Lösung von 1,37 g (12 mMol) 2-Mercapto-1methylimidazol in 20 ml Methylenchlorid. Mit 10 ml Methylenchlorid wird nachgespült.
Nach 7 Stunden wird das Lösungsmittel im Vakuum entfernt und das resultierende Öl mit 10 ml Methanol und 1,65 ml Eisessig versetzt, wonach es nach kurzer Zeit fast vollständig durchkristallisiert. Man nutscht ab, wäscht mit Methanol nach, trocknet und erhält die praktisch reine 7-[l-Methyl-imidazolyl-(2)-mercapto- acetylamino-cephalosporansäure. F. 177-1790 (Zers.) in evakuierter Kapillare.
Die Substanz wird wie folgt in das kristalline Natriumsalz übergeführt: 2,9 g werden in 60 ml Methanol aufgeschlämmt und mit 3,75 ml 3-m. methanolischem Natrium-a-äthylhexanoat versetzt. Beim Rühren und kurzem Erwärmen im Wasserbad von 30 tritt vorübergehende Lösung und dann Kristallisation des Natriumsalzes ein. Das Gemisch wird im Vakuum auf ca. 15 ml Volumen eingeengt, eine Stunde bei 200 stehen gelassen, genutscht, mit einem Gemisch von Tetrahydrofuran und Methanol (1:1) gewaschen und im Vakuum getrocknet. Das Natriumsalz ist in Wasser leicht löslich und besitzt ein []r = +1070 + 10 (c = 1, HL4O), das UV-Spektrum (in Wasser) zeigt ein Maximum bei Z = 256 m,u; (e = 12 800). F.
150-1540 (unter Zersetzung).

Beispiel 2
3,93 g (10 mMol) Bromacetyl-7-ACA werden in 15 ml Methylenchlorid unter Zugabe von 3,45 ml (20 mMol) N,N-Diisopropyläthylamin gelöst. Dazu gibt man eine Lösung, die durch Auflösen von 1,23 g (12 mMol) 2-Mercaptoimidazolin in 20 ml Dimethylformamid und anschliessendes Verdünnen mit 10 ml Methylenchlorid hergestellt worden ist. Mit 5 ml Methylenchlorid wird nachgespült.
Nach 6 Stunden wird das Methylenchlorid im Wasserstrahlvakuum entfernt, dann destilliert man das Dimethylformamid bei einem Vakuum von ca. 0,3 mm und einer Temperatur von 30 möglichst vollständig ab.
Es resultieren 10,7 g dickes Öl. Durch Zugabe von 100 ml Tetrahydrofuran erhält man ein Präcipitat, das intensiv durchgeknetet wird. Man filtriert ab und zerreibt den Rückstand mit weiteren 50 ml Tetrahydrofuran, saugt ab und wäscht mit wenig Tetrahydrofuran und dann mit Hexan. Nach dem Trocknen im Vakuum erhält man 6,75 g eines gelblichen Pulvers.
Man löst 4,5 g dieses Rohproduktes in 13,5 ml Dimethylformamid und vcrdünnt mit 13,5 ml Methanol.
Die Lösung wird mit einer Spateispitze Norit (Aktivkohle) bei Raumtemperatur wiihrend 5 Minuten geschüttelt, dann durch eine Schicht liyflo Supercel (Diatomeenerde) filtriert und das Filter mit 6 ml eines Gemisches von Dimethylformamid und Methanol (1:1) gewaschen. Filtrat und Waschflüssigkeit werden vereinigt und mit 165 ml Tetrahydrofuran versetzt, wobei sich eine hellgelbe Fällung bildet. Man lässt 1 Stunde bei
200 stehen. Dann nutscht man ab und wäscht den Rückstand sehr gut nacheinander mit einem Gemisch von Methanol-Tetrahydrofuran (1 : 4,5), Tetrahydrofuran und zuletzt mit Hexan.
Nach dem Trocknen erhält man die 7- [Imidazolinyl(2)-mercaptoacetylamino] -cephalosporansäure als fast farbloses amorphes Pulver, das im Dünnschichtchromatogramm einheitlich ist.
RfÏs = 0,05; Rflel A = 0,25. Schmelzpunkt (evakuierte Kapillare): 213-2160 (Zersetzung). Die optische Dre hung [j , = + 143 + 2" (in Wasser, c = 0;5).
Beispiel 3
3,93 g Bromacetyl-7-ACA werden in 27 ml Methylenchlorid unter Zugabe von 3,45 ml N,N-Diisopropyl äthylamin gelöst. Dazu gibt man eine Lösung, die durch Auflösen von 1,43 g 2-Mereaptothiazolin in 2 ml Dimethylformamid und anschliessendes Verdünnen mit 8 ml Methylenchlorid hergestellt worden ist. Mit 10 ml Methylenchlorid wird nachgespült.
Nach 7 Stunden wird das Lösungsmittel zuerst im Wasserstrahlvakuum und dann bei ca. 0,3 mm (Wasserbad 30 ) entfernt. Man erhält 7g eines dickflüssigen öles.
6,3 g dieses Rohproduktes werden in 27 ml 10 o/oi ger Kaliumdihydrogenphosphatlösung aufgenommen.
Durch Zugabe von 0,3 ml 2-n. Sodalösung stellt man das pH auf 5 ein. Dann wird die Lösung in einen Scheidetrichter gegeben und zuerst mit 55 ml Methylenchlorid, dann mit 90 ml Aethylacetat extrahiert. Die organischen Phasen werden nacheinander zweimal mit je 45 ml Phosphatpuffer vom pH 5 gewaschen und verworfen. Dann werden alle wässrigen Phasen vereinigt und mit 180 ml Aethylacetat überschichtet, worauf man durch Zugabe von 11 ml 1-n. Salzsäure das pH auf 2,9 einstellt und die wässrige Phase mit Kochsalz sättigt. Es wird gut geschüttelt, die Phasen getrennt und die wässrige Phase noch zweimal mit je 90 ml Aethylacetat extrahiert. Die organischen Phasen werden nacheinander mit 45 ml gesättigter Kochsalzlösung gewaschen, mit Natriumsulfat getrocknet, vereinigt und im Vakuum zur Trockne eingedampft.
Der amorphe Rückstand kristallisiert nach Zugabe von 6 ml eines Gemisches aus Aethylacetat und Tetrahydrofuran (9 : 1) fast vollständig durch. Die Kristalle (1,76 g) werden abgenutscht und mit Aethylacetat gewaschen. Sie werden gereinigt, indem man sie in 15 ml Tetrahydrofuran löst, mit einer Spatelspitze Norit bei Raumtemperatur schüttelt, durch eine Hyflo-Supercel -Schicht filtriert, das Filter mit 5 ml Tetrahydrofuran wäscht, das Lösungsmittel im Vakuum entfernt und den amorphen Rückstand aus 5 ml eines Gemisches aus Aethylacetat und Tetrahydrofuran (9 :1) kristallisiert. Die Kristalle werden abgenutscht, mit Aetylacetat gewaschen und im Hochvakuum getrocknet.
Man erhält die reine 7-[Thiazoli nyl-(2)-mercaptoacetylaminoj -cephalosporansäure vom Schmelzpunkt (evakuierte Kapillare) : 131-138" (Zersetzung). Zwecks Herstellung des Natriumsalzes werden 1,6 g der Säure in 32 ml Methanol suspendiert und 2,1 ml 3-m. methanolisches Natriumäthylhexanoat zugegeben. Die so erhaltene Lösung engt man im Vakuum auf ein Volumen von 8-10 ml ein und lässt ca. 21/2 Stunden bei 200 stehen. Dann wird abgenutscht und die Kristalle des Natriumsalzes werden nacheinander mit 12 ml Methanol-Aethylacetat (1:1), 5 ml Aethylacetat und Hexan gewaschen. Schmelzpunkt des Natriumsalzes: 185-189" (unter Zersetzung).
Die optische Drehung [X]2',= +1200 + 1" (in Wasser, c = 1).
Rf5. > = 0,26; Ruf101 A = 0,55.
Beispiel 4
3,93 g Bromacetyl-7-ACA werden in 20 ml Methylenchlorid unter Zugabe von 3,45 ml Diisopropyläthylamin gelöst. Dazu gibt man eine Lösung, die durch Auflösen von 1,22 g 3-Mercapto-l ,2,4-triazol in 10 ml Dimethylformamid und Verdünnen mit 10ml Methylenchlorid hergestellt worden ist. Mit 10 ml Methylenchlorid wird nachgespült.
Nach 7 Stunden wird das Lösungsmittel zuerst im Wasserstrahlvakuum und dann bei ca. 0,2 mm (Wasserbad 30-40") abdestilliert und es bleiben 10,5 g eines bräunlichen Öles zurück. Dieses Rohprodukt wird in 20 ml einer 10 0/obigen Kaliumdihydrogenphosphatlösung aufgenommen. Durch Zugabe von 0,9 ml 2-n. Sodalösung wird das pH auf 5 eingestellt. Dann wird die Lösung im Scheidetrichter nacheinander mit 80 ml Methylenchlorid und 30 ml Aethylacetat extrahiert. Die organischen Phasen wäscht man zweimal mit je 20 ml Phosphatpuffer vom pH 5. Die vereinigten wässrigen Phasen werden hierauf mit 300 ml Aethylacetat überschichtet und das pH durch Zugabe von 1,0 ml konzentrierter Salzsäure auf 2,9 eingestellt. Dadurch entsteht ein farbloses Präcipitat, das beide Phasen erfüllt.
Der gesamte Inhalt des Scheidetrichters wird durch einen Glasfilter abgenutscht und der Rückstand mit 100 ml Aethylacetat nachgewaschen. Durch wiederholtes Zerreiben dieses Niederschlages mit Tetrahydrofuran (ca.
150 ml) lässt sich die weitgehend reine und kristallisierende 7-[1.2.4-Triazol yl-(3 )-mercaptoacetyl amino]- cepha- losporansäure herauslösen. Das beide Phasen umfassende Filtrat wird abgetrennt und daraus noch weiteres Rohkristallisat gewonnen.
2,5 g des vereinigten Rohkristallisates werden in 30 ml eines Gemisches von Tetrahydrofuran und Methanol (1:1) gelöst. mit einer Spatelspitze Norit bei Raumtemperatur kurz geschüttelt, durch eine Schicht Hyflo Supercel filtriert und das Filter mit 3 ml des obigen Gemisches und 5 ml Methanol nachgewaschen.
Dann engt man das Filtrat im Vakuum auf ca. 5 ml ein.
worauf es nach dem Animpfen sofort kristallisiert.
Schmelzpunkt: 149151 (Zersetzung).
Man suspendiert 1,55 g der Säure in 50 ml Methanol und gibt 1,85 ml 3-m. methanolisches Natriumäthylhexanoat dazu. Die klare Lösung wird im Vakuum auf ca. 10-12 ml eingeengt, wobei sich langsam Kristalline bilden. Man gibt noch 2.5 ml Tetrahydrofuran zu und lässt 2 Stunden bei -20" stehen. Dann wird abgenutscht. und der Rückstand nacheinander mit Methanol Tetrahydrofuran (1:1), Tetrahydrofuran, Aethylacetat und Hexan gewaschen.
Optische Drehung des reinen Natriumsalzes [R]20,= +1130 + 10; Schmelzpunkt: 220-2300 (un ter Zersetzung). Rf52 = 0,21: Rf101 A = 0,5.
Beispiel 5 393 mg (1,0 mol) Bromacetyl-7-ACA werden in 2,0 ml Methylenchlorid unter Zugabe von 258 mg N,N Diisopropyläthylamin gelöst. Dazu gibt man eine Lösung, die durch Auflösen von 180 mg 2,5-Dimercapto1,3,4-thiadiazol in 0,5 ml Dimethylformamid und Verdünnen mit 2,2 ml Methylenchlorid hergestellt worden ist. Mit 1 ml Methylenchlorid wird nachgespült.
Nach 6 Stunden wird das Lösungsmittel zuerst im Wasserstrahlvakuum und dann bei ca. 0,2 mm (Wasserbad 30 ) abdestilliert und es bleiben ca. 1,1 g gelbliches Ö1 zurück. Dieses Rohprodukt wird in 10 ml einer 10 0'0gen Kaliumdihydrogenphosphatlösung aufgenommen und durch Zugabe von 0,3 ml 2-n. Sodalösung auf pH 5 gebracht. Dann wird im Scheidetrichter nacheinander mit 20ml Methylenchlorid und 30 ml Aethylacetat extrahiert. Die organischen Phasen wäscht man nacheinander mit zweimal je 10 ml Phosphatpuffer pH 5. Dann werden die wässrigen Phasen vereinigt und mit 40 ml Aethylacetat überschichtet. Durch Zugabe von 2,7 ml 1-n. Salzsäure stellt man auf pH 2,9 ein und sättigt die wässrige Phase mit Kochsalz.
Nach kräftigem Schütteln werden die Phasen getrennt und die wässrige Lösung noch zweimal mit je 20 ml Aethylacetat nachextrahiert. Die 3 organischen Phasen werden nacheinander mit 8 ml gesättigter Kochsalzlösung gewaschen.
Dann wird mit Natriumsulfat getrocknet, filtriert und im Vakuum zur Trockene eingedampft. Der Rückstand kristallisiert aus einem Gemisch von Aethylacetat-Tetrahydrofuran (9:1).
Schmelzpunkt der 7-[5-Mercapto-1.3.4-thiadiazo lyl- (2)-mercapto-acetylamino]- cephalosporansäure: 130-142 ' unter Zersetzung (evak. Kapillare).
Rf =- ().36; Ruf"" - = 0,6.
Beispiel 6 11,75 g 3-(Desacetoxymethyl)-3-benzoylthiomethyl- 7-bromacetylamino-cephalosporansäure werden unter Zugabe von 8,75 ml N-Aethyl-diisopropylamin in 75 ml Dimethylformamid gelöst. Dazu gibt man eine Lösung von 3.58 g 2-Mercaptothiazolin in 25 ml Dimethylformamid und lässt unter Lichtausschluss bei Zimmertemperatur stehen. Nach 20 Stunden wird das Lösungsmittel im Vakuum (0,5-1 mm HG) grösstenteils abdestilliert und der resultierende ölige Rückstand in 250 ml Phosphatpuffer pH 6 und 250 ml Essigester aufgenommen.
Dann stellt man das pH der wässerigen Phase auf 6 und trennt die Phasen. Die organische Phase wird verworfen.
Die wässerige Phase wird mit Salzsäure auf pH 2,3 gestellt und mit Kochsalz gesättigt. Man extrahiert nacheinander mit 1000. 250 und 250 ml Essigester. Die Essigesterextrakte werden mit gesättigter Kochsalzlösung gewaschen. mit Natriumsulfat getrocknet und durch eine Säule von 75 g Silicagel filtriert. Die vereinigten Filtrate ergehell einen Trocloenrückstand von l1 g.
Dieser wird an einer Säule von 520 g Silicagel mit Chloroform und Clllorofoi'm-Aceton-Gemischen chromatographiert. Die Cli loroform-Aceton(9:1 )-Eluate liefern 7,6 g amorphe 3-(Desacetoxymethyl)-3-benzoylthiomethyl-7-[thiazolinyl(2)- mercaptoacetylamino] -cephalosporansäure, die sich mittels Natrium-a-äthylhexa- noat in das kristalline Natriumsalz der Formel

überführen lässt. Rf 52 -t 0,50. Im UV-Spektrum in Wasser sind Maxima bei i. - 240 nm (e - 19 200) und A = 274 nm ( 20000) vorhanden.
Die als Ausgangsmaterial verwendete 3-(Desacetoxymethyl)- 3-benzoylthiomethyl- 7-bromacetylaminocephalosporansäure kann sie wie folgt hergestellt werden:
Eine Lösung von 17,5 g 3-(Desacetoxymethyl)-3- benzoylthiomethyl)-7-amino-cephalosporansäure (vgl.
belg. Patent 650 444) und 12,5 ml Triäthylamin in 11 Dimethylformamid wird während einer Stunde in eine gut gerührte und bei 130 bis 15 gehaltene Lösung von 9,2 ml Bromacetylbromid in 100 ml Methylenchlorid eingetropft (Stickstoffatmosphäre). Man lässt die Temperatur während 1 1/2 Stunden langsam auf 10 steigen und hält sie dort noch während einer halben Stunde. Dann wird der grösste Teil der Lösungsmittel im Vakuum bei 0,5-1 mm Hg gegen einen Kühler von Trockeneis-Aceton abdestilliert. Das ölige Produkt wird auf einen Phosphatpuffer von pH 6 gegossen und mit 1 1 Essigester geschüttelt. An der Grenze der beiden Phasen entsteht ein Präcipitat, das durch Filtration oder Zentrifugieren abgetrennt wird.
Dann stellt man das pH der wässerigen Phase auf 2 ein, sättigt diese mit Kochsalz und trennt die organische Phase ab. Die wässerige Phase wird mit 600 und 400 ml Essigester nachextrahiert.
Nach dem Waschen mit gesättigter Kochsalzlösung werden die organischen Phasen über Natriumsulfat getrocknet und nacheinander durch eine Säule von 100 mg Silicagel filtriert. Die Filtrate werden im Vakuum zur Trockne eingeengt, der Rückstand mit 30 ml Aethanol versetzt und bei -203 auskristallisiert. Man erhält 7,8 g 3-(Desacetoxymethyl)-3-benzoylthiomethyl 7-bromacetylamino-cephalosporansäure vom Schmelz- punkt 137-138". Rf 52 = 0.55. Das Natriumsalz zeigt im U.V.-Spektrum in Wasser: A max 243 nm ( 16800) und
275 nm (F = 20 600).

Beispiel 7
2,95 g Natriumsalz der 7-[Thiazolinyl(2)-mercap toacetylamino]-cephalosporansäure (vgl. Beispiel 3) werden in 150 ml Wasser gelöst. Die Lösung wird auf 370 erwärmt und mittels 0,7 ml (),i-n. Natronlauge auf pH 7,5 eingestellt. Dann gibt man eine Aufschlämmung von 60 mg Acetylesterase (aus Bacillus subtilis ATCC 6633, vgl. engl. Patent 1 080 904) in 2 mol Wasser dazu und neutralisiert die gebildete Essigsäure laufend mit Hilfe eines automatischen Titrators mit 0,1-n. Natronlauge (Einstellung auf pH 7,3). Nach 4 Stunden ist die Reaktion beendet. Man stellt das pH auf 6,5 ein, filtriert die Lösung durch eine Glasfritte G4 und lyophilisiert.
Es resultieren 3,18 g eines gelblichen Harzes des Natriumsalzes von 7-[Thiazolinyl(2)-mercap- toacetylaminoj-0-desacetyl-cephalosporansaure.
Dieses Rohprodukt wird direkt in einem Gemisch von 30 ml absolutem Dimethylformamid und 8,4 ml Triäthylamin gelöst, mit 5,05 ml ,fS-Chloräthylisocyanat versetzt und während einer halben Stunde bei Raumtemperatur gerührt. Dann entfernt man das Lösungsmittel im Hochvakuum und verreibt den harzartigen, braunen Rückstand dreimal mit je 150 ml absolutem Aether. Der ätherunlösliche Rückstand wird in 75 ml 10 0/obigem Phosphorpuffer von pH 6,7 aufgenommen und mit 500 ml Essigester überschichtet. Durch Zugabe von 30 ml 2-n. Salzsäure stellt man das pH der wässerigen Phase auf 2,5 ein. Nach kräftigem Schütteln werden die Phasen getrennt und die wässerige Phase anschliessend nochmals mit 300 und 200 ml Essigester nachextrahiert.
Die organischen Phasen werden zweimal mit je 50 ml gesättigter Kochsalzlösung gewaschen, mit Natriumsulfat getrocknet und im Vakuum zur Trockne eingedampft. Das resultierende Harz kristallisiert nach Zugabe von wenig Aceton. Man nutscht ab und wäscht die Kristalle mit einem Gemisch aus gleichen Teilen Aceton und Aether. Das Rohkristallisat lässt sich weiter reinigen, indem man eine Suspension in 18 ml Methanol mit 1,2 ml einer 3-m. methanolischen Lösung von Natrium a-äthylhexanoat versetzt, das Gemisch mittels wenig Hyflo (Diatomeenerde) erklärt, im Vakuum auf ca.
8 ml einengt und bei -15 OC das reine Natriumsalz der 0-Desacetyl-0-(/s-Chloräthylcarbamoyl)-7-[thiazoli- nyl(2)-mercaptoacetylamino ] -cephalosporansäure auskristallisiert. Es besitzt im Ultraviolett-Spektrum ein A",\x bei 255 nm mit E = 10850 (in Wasser). Die optische Drehung [n]ll ist T 990 + 10. Es zeigt im Dünnschichtchromatogramm folgende Werte: für System 52 = 0,35, für System 101A = 0,55, und im System Essigester-Pyridin-Eisessig-Wasser (62:21:6:11) Rf = 0,34.

The salts of the new compounds are metal salts, especially those of therapeutically applicable alkali or alkaline earth metals, such as sodium, potassium, ammonium, calcium, or salts with organic bases, e.g. B.
Triethylamine, N-ethyl-piperidine, dibenzylethylene diamine, procaine. If the group R2 is basic, internal salts can form.
The new compounds have a particularly good antibacterial effect. They are effective against both gram-positive and especially against gram-negative bacteria, e.g. B. against Staphylococcus aureus penicillin-resistant, Escherichia coli, Klebsiella pneumoniae, Salmonella typhosa and Bacterium proteus, as also found in animal experiments, e.g. B. on mice shows. Compounds obtained in which R. is the acetoxy group can be converted into compounds in which R.sup.2 is a hydroxyl group by treating them with a deacetylating agent. Compounds in which R; stands for a hydroxyl group can be acylated with a carboxylic acid other than acetic acid or with a thio- or dithiocarboxylic acid. Compounds obtained in which R 1 is a hydroxyl group can be converted into compounds in which R is an N-substituted carbamoyloxy group by reaction with an isocyanic acid ester.
Compounds obtained in which Ra stands for a carboxylic acid or thiocarboxylic acid can be converted by reaction with a tertiary amine, especially pyridine, into compounds in which R; means a quaternary amino group. If desired, the compounds obtained can be converted into their metal salts, such as alkali or alkaline earth metal salts, or salts with ammonia or organic bases, or the free carboxylic acids or optionally internal salts can be formed from the salts obtained.
The reaction of the compound II, in which Z is a haloacetyl group, with the mercapto compound is preferably carried out in an inert solvent for the compound such as methylene chloride, chloroform, carbon tetrachloride, tetrahydrofuran, dioxane, dimethylformamide or acetonitrile and in the presence of a hydrogen halide binding agent, e.g. B. a weak inorganic base such as an alkali metal carbonate, bicarbonate or acetate or a tertiary amine, preferably ethyldiisopropylamine (Hünig base). The reaction takes place within a few hours at room temperature; it can, if desired, also be carried out at a lower or slightly elevated temperature.
It is preferable to use those starting materials which lead to the particularly effective end products mentioned.
The cephalosporin derivatives used as starting materials are known or can be prepared by processes known per se.
The new compounds can be used as remedies, e.g. B.
in the form of pharmaceutical preparations, use. These contain the compounds in a mixture with a pharmaceutical, organic or inorganic, solid or liquid carrier material suitable for enteral, topical or parenteral administration.
In the following examples the temperatures are given in degrees Celsius.
The abbreviation 7-ACA is used below for 7-aminocephalosporanic acid.
The following systems are used in thin-layer chromatography on silica gel plates: System 52 = n-butanol-glacial acetic acid-water (75: 7.5: 21) System 101 A = n-butanol-pyridine-glacial acetic acid-water (42: 24: 4: 30).
Example I.
3.93 g (10 mmol) of bromoacetyl-7ACA are dissolved in 20 ml of methylene chloride with the addition of 3.45 ml (20 mmol) of N-ethyl-diisopropylamine. A solution of 1.37 g (12 mmol) of 2-mercapto-1methylimidazole in 20 ml of methylene chloride is added. It is rinsed with 10 ml of methylene chloride.
After 7 hours, the solvent is removed in vacuo and the resulting oil is treated with 10 ml of methanol and 1.65 ml of glacial acetic acid, after which it crystallizes almost completely after a short time. It is filtered off with suction, washed with methanol, dried and the practically pure 7- [1-methyl-imidazolyl- (2) -mercapto-acetylamino-cephalosporanic acid is obtained. F. 177-1790 (decomp.) In evacuated capillary.
The substance is converted into the crystalline sodium salt as follows: 2.9 g are slurried in 60 ml of methanol and 3.75 ml of 3-m. methanolic sodium a-ethylhexanoate added. When stirring and briefly heating in a water bath of 30, a temporary solution and then crystallization of the sodium salt occurs. The mixture is concentrated in vacuo to a volume of about 15 ml, left to stand at 200 for one hour, suction filtered, washed with a mixture of tetrahydrofuran and methanol (1: 1) and dried in vacuo. The sodium salt is easily soluble in water and has a [] r = +1070 + 10 (c = 1, HL4O), the UV spectrum (in water) shows a maximum at Z = 256 m, u; (e = 12,800). F.

Example 2
3.93 g (10 mmol) of bromoacetyl-7-ACA are dissolved in 15 ml of methylene chloride with the addition of 3.45 ml (20 mmol) of N, N-diisopropylethylamine. A solution is added which has been prepared by dissolving 1.23 g (12 mmol) of 2-mercaptoimidazoline in 20 ml of dimethylformamide and then diluting with 10 ml of methylene chloride. It is rinsed with 5 ml of methylene chloride.
After 6 hours the methylene chloride is removed in a water jet vacuum, then the dimethylformamide is distilled off as completely as possible under a vacuum of about 0.3 mm and a temperature of 30.
10.7 g of thick oil result. Adding 100 ml of tetrahydrofuran gives a precipitate which is thoroughly kneaded. It is filtered off and the residue is triturated with a further 50 ml of tetrahydrofuran, filtered off with suction and washed with a little tetrahydrofuran and then with hexane. After drying in vacuo, 6.75 g of a yellowish powder are obtained.
4.5 g of this crude product are dissolved in 13.5 ml of dimethylformamide and diluted with 13.5 ml of methanol.
The solution is shaken with a spatula Norit (activated charcoal) at room temperature for 5 minutes, then filtered through a layer of Lioflo Supercel (diatomaceous earth) and the filter washed with 6 ml of a mixture of dimethylformamide and methanol (1: 1). The filtrate and washing liquid are combined and 165 ml of tetrahydrofuran are added, a pale yellow precipitate being formed. Leave for 1 hour
200 stand. It is then filtered off with suction and the residue is washed very well in succession with a mixture of methanol-tetrahydrofuran (1: 4.5), tetrahydrofuran and finally with hexane.
After drying, the 7- [imidazolinyl (2) -mercaptoacetylamino] -cephalosporanic acid is obtained as an almost colorless amorphous powder which is uniform in the thin-layer chromatogram.

Example 3
3.93 g of bromoacetyl-7-ACA are dissolved in 27 ml of methylene chloride with the addition of 3.45 ml of N, N-diisopropyl ethylamine. A solution is added which has been prepared by dissolving 1.43 g of 2-mereaptothiazoline in 2 ml of dimethylformamide and then diluting with 8 ml of methylene chloride. It is rinsed with 10 ml of methylene chloride.
After 7 hours, the solvent is removed first in a water jet vacuum and then at about 0.3 mm (water bath 30). 7 g of a thick oil are obtained.
6.3 g of this crude product are taken up in 27 ml of 10% potassium dihydrogen phosphate solution.
By adding 0.3 ml of 2-n. Soda solution adjusts the pH to 5. The solution is then poured into a separating funnel and extracted first with 55 ml of methylene chloride and then with 90 ml of ethyl acetate. The organic phases are washed twice in succession with 45 ml of phosphate buffer of pH 5 each time and discarded. Then all aqueous phases are combined and covered with 180 ml of ethyl acetate, whereupon by adding 11 ml of 1-n. Hydrochloric acid adjusts the pH to 2.9 and saturates the aqueous phase with sodium chloride. It is shaken well, the phases are separated and the aqueous phase is extracted twice more with 90 ml of ethyl acetate each time. The organic phases are washed successively with 45 ml of saturated sodium chloride solution, dried with sodium sulfate, combined and evaporated to dryness in vacuo.
The amorphous residue crystallizes almost completely after the addition of 6 ml of a mixture of ethyl acetate and tetrahydrofuran (9: 1). The crystals (1.76 g) are suction filtered and washed with ethyl acetate. They are purified by dissolving them in 15 ml of tetrahydrofuran, shaking them with a spatula tip of Norit at room temperature, filtering through a Hyflo-Supercel layer, washing the filter with 5 ml of tetrahydrofuran, removing the solvent in vacuo and removing the amorphous residue from 5 ml a mixture of ethyl acetate and tetrahydrofuran (9: 1) crystallized. The crystals are filtered off with suction, washed with ethyl acetate and dried under high vacuum.
The pure 7- [Thiazoli nyl- (2) -mercaptoacetylaminoj -cephalosporanic acid with a melting point (evacuated capillary): 131-138 "(decomposition) is obtained. To prepare the sodium salt, 1.6 g of the acid are suspended in 32 ml of methanol and 2 1 ml of 3 m methanolic sodium ethylhexanoate are added The solution obtained in this way is concentrated in vacuo to a volume of 8-10 ml and left to stand for about 21/2 hours at 200. It is then filtered off with suction and the crystals of the sodium salt are successively Washed with 12 ml of methanol-ethyl acetate (1: 1), 5 ml of ethyl acetate and hexane. Melting point of the sodium salt: 185-189 "(with decomposition).
The optical rotation [X] 2 ', = +1200 + 1 "(in water, c = 1).

Example 4
3.93 g of bromoacetyl-7-ACA are dissolved in 20 ml of methylene chloride with the addition of 3.45 ml of diisopropylethylamine. A solution is added which has been prepared by dissolving 1.22 g of 3-mercapto-1,2,4-triazole in 10 ml of dimethylformamide and diluting with 10 ml of methylene chloride. It is rinsed with 10 ml of methylene chloride.
After 7 hours, the solvent is first distilled off in a water jet vacuum and then at approx. 0.2 mm (water bath 30-40 ") and 10.5 g of a brownish oil remain. This crude product is dissolved in 20 ml of a 10% above potassium dihydrogen phosphate solution By adding 0.9 ml of 2N sodium carbonate solution, the pH is adjusted to 5. The solution is then extracted in a separating funnel with 80 ml of methylene chloride and 30 ml of ethyl acetate, and the organic phases are washed twice with 20 ml of phosphate buffer each time pH 5. The combined aqueous phases are then covered with a layer of 300 ml of ethyl acetate and the pH is adjusted to 2.9 by adding 1.0 ml of concentrated hydrochloric acid, resulting in a colorless precipitate which fills both phases.
The entire contents of the separating funnel are filtered off with suction through a glass filter and the residue is washed with 100 ml of ethyl acetate. By repeatedly triturating this precipitate with tetrahydrofuran (approx.
150 ml) the largely pure and crystallizing 7- [1.2.4-triazol yl- (3) -mercaptoacetyl amino] cephalosporanic acid can be dissolved out. The filtrate comprising both phases is separated off and further crude crystals are obtained therefrom.
2.5 g of the combined raw crystals are dissolved in 30 ml of a mixture of tetrahydrofuran and methanol (1: 1). shaken briefly with a spatula tip of Norit at room temperature, filtered through a layer of Hyflo Supercel and the filter was washed with 3 ml of the above mixture and 5 ml of methanol.
The filtrate is then concentrated in vacuo to about 5 ml.
whereupon it crystallizes immediately after inoculation.
Melting point: 149151 (decomposition).
1.55 g of the acid are suspended in 50 ml of methanol and 1.85 ml of 3-m are added. methanolic sodium ethylhexanoate to it. The clear solution is concentrated to approx. 10-12 ml in vacuo, with crystallines slowly forming. Another 2.5 ml of tetrahydrofuran are added and the mixture is left to stand for 2 hours at -20 ". Example 5 393 mg (1.0 mol) of bromoacetyl-7-ACA are dissolved in 2.0 ml of methylene chloride with the addition of 258 mg of N, N diisopropylethylamine. A solution is added which has been prepared by dissolving 180 mg of 2,5-dimercapto1,3,4-thiadiazole in 0.5 ml of dimethylformamide and diluting with 2.2 ml of methylene chloride. Rinse with 1 ml of methylene chloride.
After 6 hours, the solvent is first distilled off in a water jet vacuum and then at approx. 0.2 mm (water bath 30) and approx. 1.1 g of yellowish oil remain. This crude product is taken up in 10 ml of a 10 0'0gen potassium dihydrogen phosphate solution and by adding 0.3 ml of 2-n. Brought soda solution to pH 5. Then it is extracted successively in a separating funnel with 20 ml of methylene chloride and 30 ml of ethyl acetate. Example 6 11.75 g of 3- (desacetoxymethyl) -3-benzoylthiomethyl-7-bromoacetylamino-cephalosporanic acid are dissolved in 75 ml of dimethylformamide with the addition of 8.75 ml of N-ethyldiisopropylamine. A solution of 3.58 g of 2-mercaptothiazoline in 25 ml of dimethylformamide is added and left to stand at room temperature with exclusion of light. After 20 hours, the solvent is largely distilled off in vacuo (0.5-1 mm HG) and the resulting oily residue is taken up in 250 ml of phosphate buffer pH 6 and 250 ml of ethyl acetate.
The pH of the aqueous phase is then adjusted to 6 and the phases are separated. The organic phase is discarded.
The aqueous phase is adjusted to pH 2.3 with hydrochloric acid and saturated with sodium chloride. It is extracted successively with 1000, 250 and 250 ml of ethyl acetate. The ethyl acetate extracts are washed with saturated sodium chloride solution. dried with sodium sulfate and filtered through a column of 75 g silica gel. The combined filtrates have a trocloen residue of 11 g.
This is chromatographed on a column of 520 g of silica gel with chloroform and Clllorofoi'm-acetone mixtures. A solution of 17.5 g of 3- (desacetoxymethyl) -3-benzoylthiomethyl) -7-aminocephalosporanic acid (cf.
Belgian Patent 650 444) and 12.5 ml of triethylamine in 11-dimethylformamide are added dropwise over one hour to a well-stirred solution of 9.2 ml of bromoacetyl bromide in 100 ml of methylene chloride kept at 130 to 15 (nitrogen atmosphere). The temperature is allowed to rise slowly to 10 over 11/2 hours and is held there for a further half an hour. Most of the solvents are then distilled off from dry ice-acetone in vacuo at 0.5-1 mm Hg against a condenser. The oily product is poured onto a phosphate buffer of pH 6 and shaken with 1 liter of ethyl acetate. A precipitate is formed at the boundary between the two phases and is separated off by filtration or centrifugation.
The pH of the aqueous phase is then adjusted to 2, this is saturated with sodium chloride and the organic phase is separated off. The aqueous phase is extracted with 600 and 400 ml of ethyl acetate.
After washing with saturated sodium chloride solution, the organic phases are dried over sodium sulfate and successively filtered through a column of 100 mg of silica gel.
