Verfahren zur Herstellung von 3-substituierten Amino-1,2,3,4-tetrahydrocarbazolen
Die Erfindung betrifft ein Verfahren zur Herstellung von Carbazol-Derivaten, insbesondere von 3-substituierten Amino-1,2,3,4-tetrahydrocarbazolen mit pharmakologischen Eigenschaften. Die erfindungsgemäss hergestellten Verbindungen entsprechen der Formel

worin R eine niedrige Alkylgruppe, R1 und Rt Wasser stqff oder eine niedrige Alkylgruppe sind oder R und R1 zusammen mit dem Stickstoffatom, mit dem sie verknüpft sind, eine monocyclische, heterocyclische Gruppe bilden und R3 Wasserstoff, die Hydroxylgruppe oder eine niedrige Alkoxygruppe ist. Die heterocyclische Gruppe kann ausser dem angegebenen Stickstoffatom ein weiteres Stickstoff, Sauerstoff oder Schwefelatom enthalten.

umgesetzt wird, und dass dieses substituierte Phenylhydrazon zum entsprechenden Tetrahydrocarbazol-Derivat cyclisiert wird.
Das erfindungsgemässe Verfahren verläuft in bequemer Weise nach der Fischerschen Indol-Synthese, indem das entsprechende Phenylhydrazin und Keton zur Bildung der Phenylhydrazon-Vorstufe verwendet werden, welche durch Ringschluss, vorzugsweise in Gegenwart von Säure oder durch Erhitzen, das gewünschte Carbazol-Derivat liefert.
Das saure Cyclisierungsmittel kann jedes gewöhnliche anorganische Wasserstoffhalogenid, wie HCI, HBr oder HJ oder jede gewöhnliche Mineralsäure, wie H2SO4 oder HSPO4, oder eine organische Säure, wie Essigsäure, oder eine Lewis-Säure wie BF3 oder ZnC12, sein.
Die entstehenden 3-substituierten-3-Amino41,2,3,4-te- trahydrocarbazole können in Form jedes gewünschten Salzes angewendet werden. beispielsweise als Hydrochlorid, Hydrobromid, Maleat, Oxalat oder dergleichen. Obwohl pharmakologisch akzeptable Salze bevorzugt sind, sind auch andere Salze, die toxisch sein können, nützlich, z. B. bei der Abtrennung der gewünschten Verbindungen aus Mischungen und für andere Zwecke als pharmakologische Anwendung.
Wenn die erfindungsgemäss hergestellten Verbindungen in der Form von pharmakologisch anwendbaren Säureadditionssalzen vorliegen, dann sind sie wertvolle Analgetika.
Das folgende Beispiel ist eine bevorzugte Ausführungsform des erfindungsgemässen Verfahrens.
Beispiel
3-Methylamino-1,2,3,4-tetrahydrocarbazol Teil A. 4-MetS2ylaminocyclohexeznol
230 g p-Methylaminophenolsulfat wurden in 800 ml wässrigem Methanol (1: 3) aufgelöst. Die Hydrierung wurde bei 1000 und 105 kg/cm2 unter Verwendung von 1,0 g Rutheniumdioxyd als Katalysator durchgeführt. Die Hydrierung war nach ca. 2 Stunden vollendet. Der Katalysator wurde abfiltriert und der grösste Teil des Lösungsmittels wurde im Vakuum abdestilliert. Zu dem zurückbleibenden Sirup wurde 20%ige Natriumhydroxydlösung (200 ml) gegeben, und die entstehende Lösung wurde einen Tag lang mit CHCI3 kontinuierlich extrahiert.
Der Extrakt wurde getrocknet und konzentriert, und der Rückstand wurde im Vakuum destilliert und ergab 123 g eines Produktes vom Siedepunkt Kp.5 = 105 - 1100. Der Stoff wurde in wasserfreien Äther gegossen und bildete weisse Kristalle (108 g) vom Schmelzpunkt F. 78 - 793. In einem anderen Ansatz betrug der Smp. F. 88 -89 . Dieser unterschiedliche Schmelzpunkt kann auf unterschiedliche Mengenanteile des cis- und trans-Isomeren beruhen.

Gefunden: N 10,59 Teil B. 4-Methylamilocyclohexanon
Zu einer Mischung von 12,9 g (0,10 Mol) 4-Methylaminocyclohexanol und 33 g Kalium-tert.butoxyd in 300 ml trockenem Benzol wurde eine Lösung von 90 g Benzophenon in 200 ml Benzol gegeben. Die Mischung wurde bei Zimmertemperatur 3 Tage lang unter einer Stickstoffatmosphäre gerührt. Wasser (200 ml) wurde dann zugegeben, und die Mischung wurde mehrere Minuten lang heftig gerührt. Die Benzolschicht wurde 4mal mit je 100 ml 10%iger Salzsäure extrahiert. Die vereinigten wässrigen Lösungen wurden dann im Gegenstrom mit Äther extrahiert. Nachdem die wässrige Lösung im Vakuum auf etwa die Hälfte eingedampft worden war, wurde festes Kaliumcarbonat zugegeben, um die freie Base zu erhalten. Die Lösung wurde einen Tag lang mit CHCl. kontinuierlich erxtrahiert.
Trocknen und Eindampfen des Extraktes bei Atmosphärendruck, und anschliessende Vakuumdestillation ergaben 5,09 g einer farblosen Flüssigkeit vom Siedepunkt Kp.ls = 105 bis 110 : jCpI,Jf13 = 1715 cm-1.
Teil C. 3-Methylamino-1,2,3,4-tetrahydrocarbazol
Eine Lösung von 15,4 g (0,121 Mol) 4-Methylaminocyclohexanon und 13,5 g (0,125 Mol) Phenylhydrazin in 200 ml Benzol, das 0 1 g p-Toluolsulfonsäure enthält, wurde 7 Stunden lang unter Rückflusskühlung erhitzt, wobei das gebildete Wasser in einer Dean-Stark-Falle aufgefangen wurde. Nach Verdampfen des Benzols wurde der Phenylhydrazon-Sirup (24,7 g) in 300 ml Essigsäure aufgelöst, und die Lösung wurde 3 Stunden lang auf 90 - 950 erhitzt. Die Essigsäure wurde im Vakuum ab; destilliert und der Rückstand wurde mit verdünnter NaOH-Lösung gerührt. Extraktion mit CHCI3, Trocknen und Konzentrieren im Vakuum ergab 20,1 g eines schmierigen Feststoffs, F. 125 - 1290.
Zur Analyse wurde eine Probe aus Benzol-Äther (Aktivkohlebehandlung) umkristallisiert; F. 134 - 1350; acHel3 = 3480 (Indol NH), 3410 (w., CH2NH-), Spitzen bei 1630 und 1600 cm-'; lMe H 226 mp (± = 18500) und 280 inlk (± = 5460).

11,3 g 3-Methylamino-l ,2,3,4-tetrahydrocarbazol (F.
125 - 1290) wurden in Aceton gelöst. Die Lösung wurde mit Aktivkohle behandelt, und das geklärte Filtrat wurde mit 1 Mol Maleinsäure in einer möglichst kleinen Menge Aceton behandelt. Etwa die Hälfte des Volumens an Äther wurde hinzugegeben und nach wenigen Minuten erfolgte Kristallisation. Das Produkt wurde gesammelt, mit Aceton-Äther gewaschen, pulverisiert und über Nacht in der Abderhalden-Pistole bei 600 getrocknet; Ausbeute 13,2g, F. 159 - 1600; )Me H 224 mg (# = 25600) und 279m,i( = 6040).

Insgesamt betrifft diese Erfindung ein Verfahren zur Herstellung einer Reihe von 3 -su bstituierten-Amino- -1,2,3,4-tetrahydrocarbazolen, die durch die obige allgemeine Formel charakterisiert werden und die wertvolle Analgetika sind.

The process according to the invention proceeds in a convenient manner according to Fischer's indole synthesis, in that the corresponding phenylhydrazine and ketone are used to form the phenylhydrazone precursor which, by ring closure, preferably in the presence of acid or by heating, yields the desired carbazole derivative.
The acidic cyclizing agent can be any common inorganic hydrogen halide such as HCl, HBr or HJ or any common mineral acid such as H2SO4 or HSPO4, or an organic acid such as acetic acid, or a Lewis acid such as BF3 or ZnCl2.
The resulting 3-substituted-3-amino41,2,3,4-tetrahydrocarbazoles can be used in the form of any desired salt. for example as the hydrochloride, hydrobromide, maleate, oxalate or the like. Although pharmacologically acceptable salts are preferred, other salts which may be toxic are also useful, e.g. B. in the separation of the desired compounds from mixtures and for purposes other than pharmacological use.
If the compounds prepared according to the invention are in the form of pharmacologically usable acid addition salts, then they are valuable analgesics.
The following example is a preferred embodiment of the method according to the invention.

3-Methylamino-1,2,3,4-tetrahydrocarbazole Part A. 4-MetS2ylaminocyclohexeznol
230 g of p-methylaminophenol sulfate were dissolved in 800 ml of aqueous methanol (1: 3). The hydrogenation was carried out at 1000 and 105 kg / cm2 using 1.0 g of ruthenium dioxide as a catalyst. The hydrogenation was complete after about 2 hours. The catalyst was filtered off and most of the solvent was distilled off in vacuo. To the remaining syrup, 20% sodium hydroxide solution (200 ml) was added, and the resulting solution was continuously extracted with CHCl 3 for one day.
The extract was dried and concentrated, and the residue was distilled in vacuo and gave 123 g of a product with a boiling point of bp.5 = 105-1100. The substance was poured into anhydrous ether and formed white crystals (108 g) with a melting point of 78 - 793. In another batch the m.p. was 88 -89. This different melting point can be based on different proportions of the cis and trans isomers.

A solution of 90 g of benzophenone in 200 ml of benzene was added to a mixture of 12.9 g (0.10 mol) of 4-methylaminocyclohexanol and 33 g of potassium tert-butoxide in 300 ml of dry benzene. The mixture was stirred at room temperature for 3 days under a nitrogen atmosphere. Water (200 ml) was then added and the mixture was stirred vigorously for several minutes. The benzene layer was extracted 4 times with 100 ml of 10% hydrochloric acid each time. The combined aqueous solutions were then extracted countercurrently with ether. After the aqueous solution was evaporated to about half in vacuo, solid potassium carbonate was added to obtain the free base. The solution was washed with CHCl for one day. continuously extracted.
Drying and evaporation of the extract at atmospheric pressure and subsequent vacuum distillation gave 5.09 g of a colorless liquid with a boiling point of bp.ls = 105 to 110: jCpI, Jf13 = 1715 cm-1.
Part C. 3-Methylamino-1,2,3,4-tetrahydrocarbazole
A solution of 15.4 g (0.121 mol) of 4-methylaminocyclohexanone and 13.5 g (0.125 mol) of phenylhydrazine in 200 ml of benzene containing 0.1 g of p-toluenesulfonic acid was heated under reflux for 7 hours, the water formed was caught in a Dean-Stark trap. The benzene was evaporated, the phenylhydrazone syrup (24.7 g) was dissolved in 300 ml of acetic acid and the solution was heated to 90-950 for 3 hours. The acetic acid was removed in vacuo; distilled and the residue was stirred with dilute NaOH solution. Extraction with CHCl3, drying and concentration in vacuo gave 20.1 g of a greasy solid, mp 125-1290.
For analysis, a sample was recrystallized from benzene ether (activated carbon treatment); F. 134-1350; acHel3 = 3480 (indole NH), 3410 (w., CH2NH-), peaks at 1630 and 1600 cm- '; lMe H 226 mp (± = 18500) and 280 inlk (± = 5460).

About half the volume of ether was added and crystallization took place after a few minutes. The product was collected, washed with acetone-ether, powdered and dried overnight in the Abderhalden pistol at 600; Yield 13.2g, m.p. 159-1600; ) Me H 224 mg (# = 25600) and 279m, i (= 6040).
