Verfahren zur Herstellung von neuen Penicillinen
Das Hauptpatent bezieht sich auf ein Verfahren zur Herstellung von neuen Penicillinen, sowie deren Ester oder Salze, der Formel

das dadurch gekennzeichnet ist, dass man 6-Aminopenicillansäure oder einen Ester oder ein Salz derselben durch Umsetzung mit einer Verbindung der Formel R-COX, in welcher R einen organischen Rest, der eine Guanidinogruppe aufweist, und X Hydroxyl oder ein Halogen bedeuten, acyliert.
Diese Penicilline besitzen nicht nur eine ausgeprägte antibakterielle Aktivität gegen gram-negative Bazillen, sondern im Vergleich zu entsprechenden Penicillinen ohne Guanidinogruppe auch eine hohe in vivo Aktivität.
Es wurde nun gefunden, dass auch solche Penicilline eine gleiche Aktivität entfalten, welche als organischen Acylrest R-CO- einen solchen enthalten, welcher einer substituierten Isoxazolcarbonsäure entspricht, wobei der Substituent eine Guanidinogruppe aufweist.
Das erfindungsgemässe Verfahren zur Herstellung neuer Penicilline, die den genannten Acylrest einer Isoxazolcarbonsäure enthalten, ist dadurch gekennzeichnet, dass man 6-Aminopenicillansäure oder einen Ester oder ein Salz derselben durch Umsetzung mit einer substituierten Isoxazolcarbonsäure oder deren Halogenid acyliert, wobei diese Isoxagolcarbonsäure oder deren Halogenid einen eine Guanidinogruppe aufweisenden Substituenten besitzt.
Bevorzugte, einen eine Guanidinogruppe aufweisenden Substituenten besitzende Isoxazolcarbonsäurechloride sind solche der Formel

in welcher A einen Guanidinphenyl- oder Guanidinme thylphenyirest darstellt.
Vorzugsweise verwendet man zur Herstellung dieser Penicilline einen geeigneten Ester der 6-Aminopenicillansäure, und verfährt dann so, wie im Hauptpatent beschrieben ist.
Die Ausgangsstoffe der Formel R-CO-X, in welcher R-CO- nun der Acylrest einer eine Guanidino- gruppe aufweisenden Substituenten besitzenden Isoxa zolcarbonsäure ist, kann man in analoger Weise, wie es für die Herstellung der mit einem Guanidinoaryl- oder Ouanidinomethylarylrest substituierten organischen Säuren bzw. Säurehalogenide der genannten Formel im Hauptpatent beschrieben ist, herstellen. So erhält man z. B. aus 5-Methyl-3-(p-arninophenyl)- 4-isoxazocarbonsäure die 5-Methyl-3-(pguam.dinophenyl)- 4-isoxazolcarbonsäure.
Insbesondere erhält man nach dem dort beschriebenen Verfahren z. B. die 5-Methyl-3 -(4-guanidinophenyl) 4-isoxazolcarbonsäure die 5-Methyl-3 -(2-chlor-4-guanidinophenyl)4-isoxazolcarbonsäure, 5-Methyle3 -(4-guan,idinomethylphenyl)- 4-isoxazolcarbonsäure oder 5-Methyl-3-(2-chlor-4-guanidinomethylphenyl)-4-isoxazolcarbonsäure.
Durch Umsetzung dieser Säuren, oder deren Halogenide mit der 6-Aminopenicillansäure erhält man die gewünschten Penicilline.
Man verfährt beispielsweise so, dass man die Komponenten in Gegenwart eines Carbodiimids, z. B. 1,3 Dicylclohexylcarbodiimid oder 1,3-Diisopropylcarbodi- imid bei Raumtemperatur in einem Lösungsmittel vermischt. Der dabei entstandene Harnstoff wird abfil triert und der erhaltene Penicillinester abgetrennt.
Durch katalytische Hydrierung lässt sich der Ester leicht in die entsprechende freie Säure überführen, wobei man vorzugsweise z. B. einen Palladium-, Platinoder Rhodiumkatalysator, zweckmässig auf einem inerten Träger, wie Holzkohle, Bariumcarbonat oder Kie- selguhr, verwendet. Nach der Hydrierung kann die erhaltene Penicillansäure nach bekannten Methoden isoliert werden. Nach. dieser Methode erhältliche Penicilline sind z. B.
5-Methyl-3-(4-guanidinophenyl) 4-isoxazolyl-peniciilin, 5-Methyl-3-(2-chlor-4-guanidinophenyl)- 4-isoxazolyl-penidllin, 5-Methyl-3-(4-guanidinomethylphenyi)- 4-isoxazolyl-penicillin oder 5-Methyl-3-(2-chlor-4-guanidinomethylphenyi)- 4-isoxazolyl-penicillin.
Diese Penicilline können in ihre inneren Salze oder nach Umsetzung mit Säure in ihre entsprechenden Penicillin-Säuresalze übergeführt werden.
Diese neuen Penicilline und deren nicht toxischen Salze sind wertvolle antibakterielle Mittel, welche therapeutisch für Geflügel und andere Tiere, aber auch für Menschen verwendbar sind, insbesondere für die Behandlung von durch gram-positive oder gram-negative Bakterien hervorgerufene Infektionen. Sie lassen sich auch als Futterzusätze bei der Ernährung von Tieren verwenden.
Beispiel
Herstellung von 5-Methyl-3-(p-guanidino phenyl)-4-isoxazolyl-penicillin
Ein Gemisch von 400 mg 5-Methyl-3-(p-guanidinophenyl)4-isoxazolcarbonsäure und 4 ml Thionylchlorid wird während 15 Minuten unter Rühren auf Rückflusstemperatur erhitzt. Zur Entfernung überschüssigen Thionylchlorids wird das Reatk- tionsgemisch im Vakuum eingedampft. Als Rückstand erhält mann 5-Methyl-3 -(p-guanidinophenyl)-4-isoxazol- carbonsäurechlorid-Hydrochlorid.
Dieser Rückstand wird in 5 ml wasserfreiem Aceton gelöst und einer Lösung zugesetzt, welche 283 mg 6-Aminopenicillansäure in einer kleinen Menge gesättigter Natriumbicarbonatlösung enthält. Das Gemisch wird etwa 15 Minuten lang bei 0-5 C gerührt, wobei man das pH durch Zugabe wässriger Natriumcarbonatlösung im Bereich zwischen etwa 7 und 8 hält. Entstandener Niederschlag wird abfiltriert, während das Produkt gelöst bleibt. Das Filtrat wird im Vakuum eingeengt, bis das 5-Methyl-3-(p-guanidinophenyl)- 4-isoxazolyl-penicillin ausfällt, welches abfiltriert wird. Dieses Produkt hemmt das Wachstum von Staphylococcus aureus bei einer Konzentration von 3,9 mcg/ml.
Die 5-Methyl-3-(p-guanidinophenyl)- 4-isoxazolcarbonsäure erhält man durch Umsetzung von p-Nitrobenzaldehyd mit Hydroxylamin-Hydrochlorid und erhält dabei das p-Nitrobenzaldoxim, welches dann durch Behandlung mit HC1 und gasförmigen Chlor in das p-Nitrobenzohydroxamsäurechlorid und durch anschliessende Behandlung mit Äthylacetoacetat in Gegenwart einer starken Base in das Athyl-[5-methyl-3-(p-nitrophenyl)- 4isoxazolyl]-carboxylat übergeführt wird, das man mit Natriumhydroxyd zur entsprechenden freien Säure verseift.
Die erhakene Säure wird in Gegenwart eines Katalysators zur 5-Methyl-3-(p-aminophenyl)4-isoxazolcarbonsäure hydriert, aus welcher man durch Reaktion mit Benzylcyanamid in Gegenwart von Natriumhydroxyd die gewünschte Säure erhält. Diese Umsetzungen werden anschliessnd ausführlich beschrieben:
Einer Lösung von 50 g p-Nitrobenzaldehyd in 500 ml Pyridin werden unter Rühren 25,3 g Hydroxy lamin-HydrochloridE zugegeben. Die entstandene Lösung wird während etwa einer Stunde auf Dampfbad-Temperatur erhitzt. Dann wird die Lösung mit dem dreifachen Volumen Wasser versetzt, worauf man kühlt, bis das Produkt aus der Lösung auskristailisiert.
Das als kristallines Produkt erhaltene p-Nitrobenzaldoxim wird abfiltriert und aus einer Äthanol-Wasser Mischung umkristallisiert.
Ungefähr 52 g p-Nitrobenzaldoxim werden in 202 ml 8,3 n Salzsäure suspendiert. Nach Kühlung auf 0 C lässt man während etwa einer Stunde Chlorgas durch die Lösung hindurchperlen, wobei p-Nitroben zaldoximchlorid aus der Lösung auskristallisiert, welches abfiltriert wird.
Einer Lösung von 18,2 g Natriummethoxyd in 113 ml Methanol gibt man schnell 44 g Äthylacetoacetat zu, worauf man die Lösung etwa 10 Minuten unter Rühren stehen lässt und dann auf -250 C abkühlt.
Eine Lösung von 51 g p-Nitrobenzaldoximchlorid in 113 ml Methanol wird auf etwa 10 C gekühlt und der vorangehend erhaltenen Äthylacetoacetat-Lösung in solchen Portionen zugegeben, dass die Temperatur der Reaktionslösung durch Kühlung unterhalb 0 C gehalten werden kann. Das resultierende Gemisch wird während etwa 18 Stunden bei 250 C gerührt, worauf sich ein orangenfarbiger Niederschlag von Äthyl-[5-methyl-3 -(p-nitrophenyl)4-isoxazolyl] -carboxylat abscheidet, welcher abfiltriert, mit Methanol und Wasser gewaschen und durch Umkristallisieren aus Methanol-Äther gereinigt wird.
Eine Lösung von 43 g so erhaltenem Äthylester in 755 ml Methanol wird unter Stickstoff auf Rückfluss Temperatur erhitzt, worauf man 167 ml- 1 n wässrige Natriumhydroxydlösung zugibt und noch ungefähr 11/j9 Stunden auf Rückfluss-Temperatur hält. Dann kühlt man die Lösung auf 25 C, gibt 25 ml Eisessig zu und engt im Vakuum ein, bis sich das Produkt abscheidet Durch Zugabe von Wasser zum Reaktionsgemisch wird die Ausfällung des Produktes vervollständigt, welches durch Filtration aus der Lösung abgetrennt, gewaschen und dann getrocknet wird.
Durch Umkristallisieren aus Äthanol erhält man praktisch reine 5-Methyl-3 -(p-nitrophenyl)4-isoxazolcarbonsäure.
1 g Rhodium-Katalysator auf Holzkohle, welcher vorangehend mit Wasserstoff reduziert wurde, wird mit einer Lösung von 2,4 g vorstehend erhaltenen Säure in 250 ml Methanol versetzt, worauf das resultierende Gemisch unter steter Bewegung bei einem Wasserstoff Druck von 2,8 kg/cm2 hydriert wird, bis kein Wasserstoff mehr aufgenommen wird. Nach der Abtrennung des Katalysators durch Filtration und Entfernung des Lösungsmittels durch Eindampfen im Vakuum erhält man das Produkt als Rückstand. Dieser wird in Äther aufgenommen und mit Methanol angerieben, worauf man durch weitere Reinigung unter Extraktion mit Äthylacetat und Abdampfen des Lösungsmittels praktisch reine 5-Methyl-3-(p-aminophenyl3- 4-isoxazolcarbonsäure erhält.
Etwa 973 mg der so erhaltenen Aminosäure mischt man in 5 ml Äthanol mit 362 mg Benzoyicyanamid und dampft die Lösung auf Dampfbad-Temperatur zur Trockne ein. Zugabe von 5 mi Äthanol zum Rückstand und erneutes Verdampfen zur Trockne wird zweimal wiederholt. Dann wird der Rückstand des Reaktionsge- misches in 50/oiger wässriger Natriumbicarbonatlösung suspendiert, die Suspension während etwa einer Stunde gerührt und anschliessend zur Entfernung nicht umgesetzten Ausgangsmaterials filtriert.
Dem Filtrat gibt man etwa 250 mg Natriumhydroxyd zu und erhitzt die resultierende Lösung während etwa 15 Minuten unter Stickstoff auf Rückfiusstemperatur, worauf die Lösung mit Aktivkohle entfärbt, anschliessend mit etwa 750 mg Natriumhydiroxyd behandelt und während etwa 30 Minuten bei 25O C gerührt wird.
Das Produkt, die 5-MethylF3-(p-guanidinophlenyl)- 4-isoxazolcarbonsäure, wird durch Neutralisierung der alkalischen Lösung mit Eisessig ausgefällt, abfiltriert und aus heissem Wasser umkristallisiert.

Production of 5-methyl-3- (p-guanidino phenyl) -4-isoxazolyl-penicillin
A mixture of 400 mg of 5-methyl-3- (p-guanidinophenyl) 4-isoxazolecarboxylic acid and 4 ml of thionyl chloride is heated to reflux temperature for 15 minutes while stirring. The solution is then cooled to 25 ° C., 25 ml of glacial acetic acid are added and the mixture is concentrated in vacuo until the product separates out. The addition of water to the reaction mixture completes the precipitation of the product, which is separated off from the solution by filtration, washed and then dried becomes.
Recrystallization from ethanol gives practically pure 5-methyl-3 - (p-nitrophenyl) 4-isoxazolecarboxylic acid.

About 250 mg of sodium hydroxide are added to the filtrate and the resulting solution is heated to reflux temperature under nitrogen for about 15 minutes, whereupon the solution is decolorized with activated charcoal, then treated with about 750 mg of sodium hydroxide and stirred at 25O C for about 30 minutes.
The product, the 5-methylF3- (p-guanidinophlenyl) -4-isoxazolecarboxylic acid, is precipitated by neutralizing the alkaline solution with glacial acetic acid, filtered off and recrystallized from hot water.
