Das erfindungsgemässe Verfahren betrifft somit die Herstellung von Verbindungen der Formel It ebenso wie die Herstellung von Verbindun gen der Formei I. Der Einfachheit halber werden jedoch die Verbindungen der Formel I und der Formel It unter der Formel I zusammengefasst.
Das Verfahren kann wie folgt durchgeführt wer den: Verbindungen der Formel II und Verbindungen der Formel III, die sich ebenfalls in Enol-Form befin den können, werden zusammen in einem inerten orga nischen Lösungsmittel, beispielsweise einem Alkanol mit 1-5 Kohlenstoffatomen, wie Isopropanol, oder einem aliphatischen Keton, wie Aceton, umgesetzt. Das verwendete Lösungsmittel ist nicht kritisch. Die Um setzung erfolgt zweckmässigerweise bei Temperaturen zwischen 20 und 50 C, insbesondere zwischen 25 und 35 C. Die Reaktionszeit beträgt vorzugsweise 3 bis 48 Stunden.
Die so erhaltenen Säureadditionssalze von Verbindungen der Formel I mit Säuren der Formel HX werden auf an sich bekannte Weise isoliert, gerei nigt und gegebenenfalls in freie Basen überführt.
Die Ausgangsprodukte der Formel II sind be kannt oder können hergestellt werden, indem man Ver bindungen der Formel IV,

worin R obige Bedeutung besitzt, mit Chlor oder Brom in einem inerten organischen Lösungsmittel, beispiels weise Dichlormethan, Chloroform oder Tetrachlorkoh lenstoff, 1 bis 8 Stunden bei Temperaturen zwischen 0 und 50 C, vorzugsweise 20 und 35 C, behandelt. Die hierbei als Ausgangsprodukte verwendeten Verbindun gen der Formel IV sind bekannt oder in an sich be kannter Weise herstellbar.
Die Ausgangsprodukte der Formel II sind bekannt. Die erfindungsgemäss hergestellten Verbindungen der Formel I sind bei Zimmertemperatur kristalline, amorphe oder ölige Basen, die mit geeigneten organi schen oder anorganischen Säuren gut kristallisierte Säureadditionssalze bilden. Für die Salzbildung kom men als anorganische Säuren insbesondere die Chlor wasserstoffsäure, Bromwasserstoffsäure, Schwefelsäure oder Phosphorsäure, und als organische Säuren insbe sondere Bernsteinsäure, Benzoesäure, Essigsäure, p-Toluolsulfonsäure oder Benzolsulfonsäure in Be tracht.
Die erfindungsgemäss hergestellten Verbindungen der Formel I und ihre Säureadditionssalze zeichnen sich durch ausserordentlich günstige pharmakodynami- sche Eigenschaften aus. Insbesondere besitzen sie eine antidepressive Wirkung. Diejenigen Verbindungen der Formel I, worin n für 2 steht und ihre Säureadditions salze wirken ferner appetitzügelnd.
Die Tagesdosis an Verbindungen der Formel I soll für ihre Verwendung als Antidepressiva und gegebe nenfalls als Appetitzügler zwischen 35 und 750 mg be tragen. Diese Menge wird vorzugsweise in kleinen Dosen, beispielsweise zwischen 8 und 375 mg, mehr mals täglich oder in Retardform verabreicht werden.
Die Verbindungen der Formel I sowie ihre Salze können zusammen mit geeigneten festen oder flüssigen Trägerstoffen in eine für orale oder parenterale Verab reichung geeignete Form gebracht werden. <I>Beispiel 1</I> 2-Chloräthyl-3-hydroxy-3-phenyl-2,3,5,6- tetra-hydroimidazo[2,1-b]thiazol In einen Kolben, versehen mit einem Rührer und einem Tropftrichter, werden 60,0 g (0,3 Mol) 4-Chlor- butyrophenon und 250 ml Chloroform gegeben. Die erhaltene Lösung wird gerührt und tropfenweise mit einer Lösung von 48,0 g (16,0 ml, 0,3 Mol) Brom in 250 ml Chloroform versetzt, wobei die Zugabege schwindigkeit so gewählt wird, dass die Temperatur im Kolben 35 C nicht übersteigt.
Das Reaktionsgemisch wird anschliessend noch während einer Stunde gerührt und anschliessend das Lösungsmittel im Vakuum ver dampft. Der Rückstand (2-Brom-4-chlorbutyrophenon) wird in 150 ml Isopropanol gelöst und in einer Portion zu einem Gemisch von 30,6 g (0,3 Mol) 2-Imidazolin- thion und 500 ml Isopropanol gegeben. Die Reaktion ist exotherm. Man erhält zuerst eine Lösung und nach ungefähr einer Stunde einen festen Niederschlag. Nach 24stündigem Rühren bei Raumtemperatur wird abfil triert und 2-Chloräthyl-3-hydroxy-3- phenyl-2,3,5,6-tetrahydroimidazo [2,1-b]thiazol-hydrobromid vom Smp. 162-163 C erhalten. Nach Wiedererstarren schmilzt das Salz bei 208-209 C.
<I>Beispiel 2</I> 2-Chloräthyl-3-(4'-fluorphenyl)-3-hydroxy- 2,3,5,6-tetrahydroimidazo[2,1-b]thiazol In einen Kolben, versehen mit einem Rührer und einem Tropftrichter, werden 60,0 g (0,3 Mol) 4-Chlor- 4'-fluorbutyrophenon und 250 ml Chloroform gegeben. Die Lösung wird gerührt und tropfenweise mit einer Lösung von 48,0 g (16,0 ml, 0,3 Mol) Brom in 250 ml Chloroform versetzt, wobei die Zugabegeschwindigkeit so gewählt wird, dass die Temperatur im Kolben 35 C nicht übersteigt. Das Reaktionsgemisch wird anschlies- send noch während einer Stunde gerührt und das Lösungsmittel im Vakuum verdampft.

Nach 24- stündigem Rühren bei Raumtemperatur wird abfiltriert und 2-Chloräthyl-3-(4'-fluorphenyl) 3-hydroxy-2,3,5,6- tetrahydroimidazo [2,1-b]thiazol-hydrobromid vom Smp. 148-149 C erhalten. Nach Wiedererstarren schmilzt das Salz bei 221-223 C.
2-Chloräthyl-3-(4'-fluorphenyl)-3- hydroxy-2,3,5,6-tetrahydro-imidazo [2,1-b]thiazol-hydrobromid (0,1 Mol) wird in Wasser gelöst und die Lösung in einem Eisbad abgekühlt. Danach wird eine 5%ige wässrige Natriumcarbonat-Lösung (0,1 Mol Natrium- carbonat) zugesetzt und das erhaltene Gemisch bei Raumtemperatur während 12 Stunden gerührt. Der ge bildete Niederschlag wird abfiltriert und getrocknet. Die erhaltene Verbindung ist das 2-Chloräthyl-3-(4'-fluorphenyl) 3-hydroxy-2,3,5,6-tetrahydro- imidazo[2,1-b]thiazol.
<I>Beispiel 3</I> 2-Chloräthyl-3-3',4'-dimethylphenyl)-3-hydroxy- 2,3,5,6-tetrahydroimidazol[2,1-b]thiazol In einen Kolben, versehen mit einem Rührer und einem Tropftrichter, werden 60,0 g (0,3 Mol') 4-Chlor- 3',4'-dimethyl-butyrophenon und 250 ml Chloroform gegeben. Die Lösung wird gerührt und tropfenweise mit einer Lösung von 48,0 g (16,0 ml, 0,3 Mol) Brom in 250 ml Chloroform versetzt, wobei die Zugabege schwindigkeit so gewählt wird, dass die Temperatur im Kolben 35 C nicht übersteigt. <I>Beispiel 4</I> 2-Chloräthyl-3-hydroxy-3-phenyl-2,3,6,7- tetrahydro-5H-thiazolo[3,2-a]pyrimidin In einen Kolben, versehen mit einem Rührer und einem Tropftrichter, werden 60,0 g (0,3 Mol) 4-Chlor- butyrophenon und 250 ml Chloroform gegeben. Nach 24-stündigem Rühren bei Raumtemperatur wird abfil triert und 2-Chloräthyl-3-hydroxy-3-phenyl- 2,3,6,7-tetrahydro-5H-thiazolo [3,2-a]pyrimidin-hydrobromid vom Smp. 167-168 C erhalten. Nach Wiedererstarren schmilzt das Salz bei 210-211 C.
<I>Beispiel 5</I> 2-Chloräthyl-3-(4'-fluorphenyl)-3-hydroxy-2,3,6,7- tetrohydro-5H-thiazolo[3,2-a]pyrimidin In einen Kolben, versehen mit einem Rührer und einem Tropftrichter, werden 60,0 g (0,3 Mol) 4-Chlor- 4'-fluorbutyrophenon und 250 ml Chloroform gegeben. <I> Example 5 </I> 2-Chlorethyl-3- (4'-fluorophenyl) -3-hydroxy-2,3,6,7-tetrohydro-5H-thiazolo [3,2-a] pyrimidine In a flask , provided with a stirrer and a dropping funnel, are given 60.0 g (0.3 mol) of 4-chloro-4'-fluorobutyrophenone and 250 ml of chloroform. The solution is stirred and a solution of 48.0 g (16.0 ml, 0.3 mol) of bromine in 250 ml of chloroform is added dropwise, the rate of addition being chosen so that the temperature in the flask does not exceed 35.degree. The reaction mixture is then stirred for a further hour and the solvent is evaporated off in vacuo.
The residue (2-bromo-4-chloro-4'-fluoro-butyrophenone) is dissolved in 150 ml of isopropanol and added in one portion to a mixture of 30.6 g (0.3 mol) 3,4,5,6- Tetrahydro-2-pyrimidinthiol and 500 ml of isopropanol are added. The reaction is exothermic. A solution is obtained first and after about an hour a solid precipitate is obtained. After stirring for 24 hours at room temperature, it is filtered off and 2-chloroethyl-3- (4'-fluorophenyl) 3-hydroxy-2,3,6,7-tetrahydro-5H-thiazolo [3,2-a] -pyrimidine- hydrobromide of m.p. 185-186 ° C obtained.
