On peut effectuer cette réduction par hydrogénation en présence d'un métal tel que le nickel, le palladium ou le platine comme catalyseur.
On peut aisément préparer la 3-non-substituée-6(7)- aminobenzoxazine à partir des composés 3-substitués cor respondants selon des processus connus. On obtient par exemple de bons rendements par la scission hydrolytique du substituant 3-acyle avec l'acide chlorhydrique.
On peut préparer le composé II initial par une syn thèse qui part de la 3-acétyl-lH-3,4-dihydro-2,3-benzoxa- zine, que l'on nitre à basse température avec du nitrate de potassium et l'acide sulfurique. La réaction donne un mélange des isomères 6-nitro et 7-nitro-3-acétyl-lH-3,4- dihydro-2,3-benzoxazine que l'on sépare par cristallisa- tion fractionnée.
Par hydrolyse acide on peut convertir le composé obtenu en 6 ou 7-nitro-lH-3,4-dihydro-2,3-benzoxazine. On peut préparer les dérivés 3-alcoyle par réaction de ce dernier composé avec un halogénure d'alcoyle, ou un composé contenant une double ou une triple liaison, ou un mélange de formaldéhyde et d'acide formique.
L'acylation avec des anhydrides et des halogénures d'acide carboxylique produirait les 3-acylbenzoxazines correspondantes avec de bons rendements.
Une variante de la méthode de préparation du com posé II est représentée dans le schéma 1, ont les symboles R et 12s ont la signification ci-dessus.
Cette méthode part d'un a-a'-dibromo-o-xylène 4- substitué de formule M que l'on chauffe au reflux avec une quantité environ équimoléculaire de N-hydroxyuré- thane, en présence d'un excès d'un hydroxyde de métal alcalin. On utilise un alcanol inférieur anhydre comme solvant.
On scinde alors le groupe carbétoxy par scission hydrolytique, en utilisant un hydroxyde de métal alcalin, obtenant ainsi un mélange des deux isomères de formule O, ayant le radical R2 en position 6 ou 7. On conduit la séparation des isomères selon des techniques bien con nues, par exemple la cristallisation fractionnée.

Les composés de formule I, préparés par le procédé selon la présente invention, ont un intérêt pharmacolo gique, comme agents anti-inflammatoires.

On sépare le catalyseur par filtration, concentre le filtrat sous vide et cristallise le résidu dans l'éthanol.
On obtient 0,26 g (rendement de 79 0/0) de 3-acétyl-7-amino-lH-3,4- dihydro-2,3-benzoxazine qui fond à 143-1440 C.
<I>Préparation</I> <I>de la</I> 7-amino-lH-3,4-dihydro-2,3-benzoxazine On peut chauffer au reflux 2 g de 3-acétyl-7-amino- 1H-3,4-dihydro-2,3-benzoxazine dans 30m1 d'acide chlorhydrique à 18 0/0. Après refroidissement on récolte par filtration les cristaux de dichlorhydrate de 7-amino- 1H-3,4-dihydro-2,3-benzoxazine formés et les cristallise dans de l'éthanol à 80 0/0.
On isole la base libre par mise en contact d'une solu tion aqueuse du dichlorhydrate ci-dessus avec une solu tion aqueuse saturée de carbonates de sodium. On obtient 1,25 g (rendement de 800/a) qui fondent à 151-152 C. <I>Exemple 2:</I> <I>Préparation de la</I> 3-méthyl- 6-atnino-IH-3,4-dihydro-2,3-benzoxazine On hydrogène à la température ordinaire 2 g de 3-méthyl-6-nitro-lH-3,4-dihydro-2,3-benzoxazine dissous dans 70 ml d'éthanol, en présence de 0,8 g de palladium à 5 0/a sur du charbon comme catalyseur.
On filtre alors la suspension, la concentre sous vide jusqu'à 10 ml et refroidit sur de la glace. Il se forme des cristaux de 3 - méthyl - 6 - amino -lH-3,4-dihydro-2,3-benzoxazine que l'on récolte et recristallise dans l'éthanol. On obtient 1,15 g (rendement de 68 0/0) qui fondent à 117-118 C. <I>Exemples 3 à 6:</I> On prépare les composés suivants pratiquement selon le procédé décrit dans l'exemple 2.

Le rendement est de 86 0/0 ; elle fond à 155-156 C. La préparation des 6- (ou 7) nitrobenzoxazines de départ peut être réalisée comme suit <I>Préparation</I> <I>de la</I> 7-nitro-IH-3,4-dihydro-2,3-benzoxazine On additionne goutte à goutte une solution de<B>15,2</B> g de nitrate de potassium dans 90 ml d'acide sulfurique concentré, en agitant à 0 C, à une solution de 19,5 g de 3-acétyl-lH-3,4-dihydro-2,3-benzoxazine dans 90 ml d'acide sulfurique concentré. On maintient la tempéra ture à 0ç)C pendant 30 minutes puis à 200C pendant 30 minutes.
On verse le mélange sur de la glace pilée et extrait le précipité formé au chlorure de méthylène. On lave les extraits avec du bicarbonate de sodium aqueux saturé, sèche sur du sulfate de sodium et évapore à sec. On chauffe au flux le résidu pendant 5 minutes dans 175 ml d'éthanol, puis on additionne lentement, en agi- tant, un volume égal d'eau chaude et laisse refroidir le mélange à la température ordinaire.
Un solide se sépare, que l'on récolte par filtration et dissout dans 105 ml d'éthanol chaud. On additionne un volume égal d'eau et laisse refroidir la solution. Des cris taux jaune pâle se séparent qui consistent en 3-acétyl-7- nitro-lH-3,4-dihydro-2,3-benzoxazine. On obtient 7,6 g (rendement de 31 %) qui fondent à 194-197 C.
<I>Préparation de la</I> 3-acétyl- 6-nitro-IH-3,4-dihydro-2,3-benzoxazine On garde les eaux mères hydroalcooliques réunies dont on a séparé par cristallisation la 3-acétyl-7-nitro- 1 H-3,4-dihydro-2,3-benzoxazine pendant une heure à 0o C et récolte le précipité formé pour obtenir 3,35 g (13,5 %) de 3 - acétyl - 6 - nitro -1 H-3,4-dihydro-2,3-benzoxazine qui fond à 119-1200 C.
