Verfahren zur Herstellung von neuen Imidazolidinonderivaten
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Imidazolidinonderivate.
Imidazolidinonverivate der allgemeinen Formel I,

in welcher R eine niedere Alkylgruppe und n 2 oder 3 bedeutet, sowie ihre Additionssalze mit anorganischen oder organischen Säuren sind bis jetzt nicht bekannt geworden.
Wie nun gefunden wurde, besitzen solche Verbindungen, insbesondere das 1-[2-[4-(4,5-Dihydro-thieno[2,3-b] [l]benzothiepin-4-yl)-1-piperazinyl] äthyl] -3-methyl-2-imidazolidinon und das 1-[3-[4-(4,5-Dihydro-thieno [2,3-b] [1]benzothiepin-4-yl)-1-piperazinyl] propyl] -3-methyl-2-imidazolidinon, sowie ihre Salze wertvolle pharmakologische Eigenschaften und einen hohen therapeutischen Index. Sie wirken bei peroraler, rektaler oder parenteraler Verabreichung zentraldämpfend, z. B. vermindern sie die Motilität und potenzieren die Narkose. Sie weisen auch antiemetische, sympathicolytische, hypotherme und katatone Wirkung auf. Ferner wirken sie auch beim test de traction .
Diese Wirkungsqualitäten, welche durch ausgewählte Standardversuche [vgl. R. Domenjoz und W. Theobald, Arch. Int. Pharmacodyn. 120, 450 (1959) und W. Theobald et al., Arzneimittelforsch. 17, 561 (1967)] erfasst werden können, charakterisieren die Verbindungen als geeignet zur Behandlung von Spannungs- und Erregungszuständen und Hyperemesis.
R kann als niedere Alkylgruppe die Methyl-, äthyl, Propyl-, Isopropyl-, Butyl-, Isobutyl-, sek. Butyl-, tert. Butyl-, Pentyl-, 2-Methyl-butyl-, Isopentyl-, 2,2-Dimethyl-propyl-, 1-Methyl-butyl-, 1-Athyl-propyl-, 1,2-Dimethyl-propyl- oder die tert. Pentylgruppe sein.
Zur erfindungsgemässen Herstellung einer Verbin- dung der allgemeinen Formel I setzt man einen reaktionsfähigen Ester einer Verbindung der allgemeinen Formel II,

Geeignete reaktionsfähige Ester der Verbindung der allgemeinen Formel II sind beispielsweise Halogenide, wie Chloride oder Bromide, ferner Sulfonsäureester, z.B. der Methansulfonsäureester oder der o- oder p-Toluolsulfonsäureester.
Diese Ester werden mit dem 4-(1 -Piperazinyl)-4,5-dihydro- thieno[2,3-b] [1]benzothiepin vorzugsweise in Gegenwart eines Lösungsmittels umgesetzt. Geeignete Lösungsmittel sind solche, die unter den Reaktionsbedingungen inert sind, beispielsweise Kohlenwasserstoffe, wie Benzol oder Toluol, Halogenkohlenwasserstoffe, wie Chloroform, ätherartige Flüssigkeiten, wie Äther oder Dioxan, niedere Alkanone, wie Aceton oder Methyläthylketon, sowie Phosphorsäureamide, z.B. das Hexamethylphosphorsäuretriamid.
Bei der erfindungsgemässen Umsetzung von einem Moläquivalent reaktionsfähigem Ester mit einem Mol äquivalent freier Base wird ein Moläquivalent Säure abgespalten. Diese Säure kann an überschüssiges 4-(1-Piperazinyl)-4,5-dihydro- thieno[2,3-b] [1]benzothiepin oder auch an das dibasische Reaktionsprodukt gebunden werden. Vorzugsweise setzt man aber dem Reaktionsgemisch ein säurebindendes Mittel zu. Geeignete säurebindende Mittel sind beispielsweise Alkalimetallcarbonate, wie Natrium- oder Kaliumcarbonat, ferner tertiäre organische Basen, wie z.B. Pyridin, Triäthylamin oder insbesondere N,N-Diisopropyl-äthylamin.
Überflüssige tertiäre Basen können auch als Lösungsmittel eingesetzt werden.
Verwendet man bei der erfindungsgemässen Reaktion anstelle von 4-(1-Piperazinyl)-4,5-dihydro- thieno[2,3-b] [1]benzothiepin ein Alkalimetallderivat dieser Verbindung, z.B. ein Natrium-, Kalium- oder Lithiumderivat, so ist es vorteilhaft, die Reaktion in einem Kohlenwasserstoff, z.B.
in Benzol oder Toluol, durchzuführen.
Von den Ausgangsstoffen, die unter die reaktion fähigen Ester von Hydroxyverbindungen der allgemeinen Formel II fallen, sind das 1-(-Chlor-äthyl)- und das 1-(3-Chlor-propyl)-3-methyl-2-imidazolidinon bekannt. Weitere Verbindungen von diesem Typus können analog hergestellt werden. Die zweite Reaktionskomponente, das 4-(1 -Piperazinyl)-4,5-dihydrothieno[2,3-b] [1]benzothiepin, ist z.
B. wie folgt herstellbar: Man geht von 4-Chlor-4,5-dihydro-thieno[2,3-b] [1]benzothiepin aus und setzt dieses in Benzol mit 1-Piperazincarbonsäureäthylester zum 4-(4,5-Dihydro-thieno [2,3-b] [1]benzothiepin-4-yl)-1- piperazincarbonsäureäthylester um, den man mit Hilfe von Kaliumhydroxid in Äthanol hydrolisiert und decarboxyliere, oder man setzt zur erfindungsgemässen Herstellung einer Verbindung der allgemeinen Formel I eine Verbindung der allgemeinen Formel III,

Geeignete Alkalimetailderivate von Verbindungen der allgemeinen Formel III sind beispielsweise Natrium-, Kalium- oder Lithiumderivate. Als reaktionsfähige Ester des 4-Hydroxy-4,5-dihydrothieno[2,3-b] [1]benzothiepin eignen sich z. B. Halogenide, wie Chloride oder Bromide, weiter Sulfonsäureester, wie der Methansulfonsäureester oder der o- oder p-Toluolsulfonsäureester.
Die erfindungsgemässe Umsetzung der freien Basen der allgemeinen Formel III, oder ihrer Alkalimetallderivate, mit den reaktionsfähigen Estern des 4-Hydroxy-4,5-dihydro thieno [2,3-b [[1]benzothiepin können in denselben Lösungsmitteln vorgenommen werden wie zuvor erwähnt.
Werden für die Umsetzung die freien Basen eingesetzt, so können auch dieselben säurebindenden Mittel verwendet werden.
Die Ausgangsstoffe der allgemeinen Formel III sind in der Literatur beschrieben. Ferner ist das 4 thieno[2,3-b] [1]benzothiepin, das ein Beispiel für die zweite Reaktionskomponente ist, ebenfalls bekannt. Das entsprechende 4-Brom-4,5-dihydrothieno[2,3-b] [1]benzothiepin kann analog hergestellt werden.
Die nach dem erfindungsgemässen Verfahren erhaltenen Verbindungen der allgemeinen Formel I werden anschliessend gewünschtenfalls in üblicher Weise in ihre Additiossalze mit anorganischen und organischen Säuren übergeführt. Beispielsweise versetzt man eine Lösung einer Verbindung der allgemeinen Formel I mit einem organischen Lösungsmittel mit der als Salzkomponente gewünschten Säure oder mit einer Lösung derselben. Vorzugsweise wählt man für die Umsetzung organische Lösungsmittel, in denen das entstehende Salz schwer löslich ist, damit es durch Filtration abgetrennt werden kann. Solche Lösungsmittel sind z.B. Methanol, Aceton, Methyläthylketon, Ace ton-Äthanol, Methanol-Äther oder Äthanol-Äther.
Zur Verwendung als Arzneistoff können anstelle freier Basen pharmazeutische annehmbare Säureaddi tionssalze eingesetzt werden, d.h. Salze mit solchen Säuren, deren Anionen bei der in Frage kommenden Dosierung nicht toxisch sind. Ferner ist es von Vorteil, wenn die als Arzneistoffe zu verwendenden Salze gut kristallisierbar und nicht oder wenig hygroskopisch sind. Zur Salzbildung mit Verbindungen der allgemeinen Formel I können z.B. die Chlorwasserstoffsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Methansulfonsäure, Äthansulfonsäure, ss-Hydroxyäthansulfonsäure, Essigsäure, Apfelsäure, Weinsäure, Citronensäure, Milchsäure, Oxalsäure, Bernsteinsäure, Fumarsäure, Maleinsäure, Benzosäure, Phenylessigsäure, Mandelsäure und Embonsäure verwendet werden.
Die nachfolgenden Beispiele erläutern die Herstellung der neuen Verbindungen der allgemeinen Formel
I und von bisher nicht beschriebenen Zwischenproduk ten näher, sollen jedoch den Umfang der Erfindung in keiner Weise beschränken. Die Temperaturen sind in
Celsiusgraden angegeben.
Beispiel 1 a) 12,05 g (0,04 Mol) 4-(1 -Piperazinyl)-thieno [2,3-b] [1]benzothiepin und 9,15 g (0,048 Mol) 1 -(3-Chlor-propyl)-3- äthyl-2-imidazolidinon werden in 100 ml Diäthylketon gelöst und 11,0 g (0,08 Mol) Kaliumcarbonat zugegeben. Man kocht das Reaktionsgemisch 24 Stunden unter Rückfluss, giesst es auf 200 ml Eiswasser und fügt 50 ml 2-n. Natronlauge zu.
Die organische Phase wird abgetrennt und die wässrige alkalische Lösung mit Äther-Methylenchlorid (2:1) extrahiert. Man vereinigt die organischen Lösungen, wäscht sie mit Wasser und extrahiert sie mit 250 ml molarer Methansulfonsäurelösung. Dann stellt man den sauren Extrakt mit konz. Natronlauge alkalisch und schüttet die ausgefallene, rohe Base mit 200 ml Äther Methylenchlorid (2:1) aus. Der Extrakt wird mit Wasser gewaschen, über Magnesiumsulfat getrocknet und im Vakuum eingedampft.
Man löst den Rückstand in 50ml Aceton, fügt 150 ml abs. Äther zu und fällt mit ätherischer Salzsäure das Dihydrochlorid aus. Das Dihydrochlorid wird abfiltriert, mit Aceton und Äther gewaschen, im Vakuum getrocknet und aus 96 0/obigem Äthanol-Äther umkristallisiert. Das erhaltene, reine 1-[3-[4-(4,5-Dihydrothieno[2,3-b] [1] benzothiepin-4-yl)-1-piperazinyl] propyl] -3-äthyl-2-imidazolidinon schmilzt bei 175-177 . Ausbeute 17,5 g 82 /o der Theorie.
Das als Ausgangsstoff verwendete 1-(3-Chlorpropyl)-3 äthyl-2-imidazolidinon wird wie folgt hergestellt: b) Man vermischt 60,0 g (0,52 Mol) 3--2ithyl-2-oxa- zolidinon und 68,1 g (0,57 Mol) (3-Chlor-propyl)-iso- cyanat mit 4,8 g Lithiumchlorid und erhitzt das Gemisch 90 Minuten auf 175-180 . Dann kühlt man das Reaktionsgemisch ab und versetzt es mit 300 ml Chloroform. Die Chloroformlösung wird abgetrennt, dreimal mit je 30 ml gesättigter Natriumchloridsäure gewaschen, über Magnesiumsulfat getrocknet und im Vakuum eingedampft. Man destilliert den Rückstand im Hochvakuum und erhält das reine 1-(3-Chlor-propyl)3-äthyl-2-imidazolidinon vom Kp. 105-110 /0,01 Torr;
Ausbeute 91,6 g, 92,5 /o der Theorie.
Beispiel 2 a) Analog Beispiel 1 a) wird wie folgendes Endprodukt erhalten: Aus 10,6 g (0,035 Mol) 4-(l-Piperazinyl)-4,5-dihydro- thieno[2,3-b] [1]benzothiepin und 7,4 g (0,042 Mol) 1-(2-Chloräthyl)-3 äthyl-2-imidazolidinon das 1 - [2- [-(4,5-Dihydro-thieno [2,3-b] [1]benzothiepin4-yl)-1-piperazinyl] - äthyl] -3-äthyl-2-imidazolidinon vom Smp. 122-124 ; Ausbeute 12,7 g, 82 /o der Theorie; Dihydrochlorid Smp. 187-189 .
b) 115 g (1,0 Mol) 3-Äthyl-2-oxazolidinon und 116 g (1,1 Mol) (2-Chlor-äthyl)-isocyanat werden mit 10,0 g Lithiumchlorid vermischt und 2 Stunden auf 1800 erwärmt. Das Reaktionsgemisch wird abgekühlt, mit 300 ml Chloroform versetzt und das Gemisch dreimal mit je 30 ml gesättigter Natriumchloridlösung ausgeschüttelt. Man trocknet die organische Phase über Magnesiumsuifat und dampft sie im Vakuum ein. Der ölige Rückstand wird unter Hochvakuum destilliert.
Das erhaltene, reine 1-(2-Chloräthyl)-3-äthyl-2-imida- zolidinon siedet bei 95 /0,01 Torr; und24": 1,4913.
Ausbeute 147,2 g, 83 /o der Theorie.
Beispiel 3
Analog Beispiel 1 a) wird wie folgendes Endprodukt erhalten:Aus 12,8 g (0,04 Mol) 1-(4,5.Dihydro-thieno[2, 3-b] [1]- benzo-thiepin-4-yl)-piperazin und 9,8 g (0,048 Mol) 1-(2-Chloräthyl)-3butyl-2-imidazolidinon wird das 11- [2- [4-(4,5-Dihydro-thieno [2,3-b] [1] benzothiepin-4-yl)-1-piperazinyl] äthyl [-3-butyl-2-imidazolidinon gewonnen, dessen Dihydrochlorid-sesqui-hydrat bei 191-193 schmilzt.
Beispiel 4 a) Analog Beispiel 1 a) wird folgendes Endprodukt erhalten:
Aus 12,8 g (0,04 Mol) 1-(4,5-Dihydro-thieno [2,3-b] [1]benzothiepin-4-yl)-1-piperazin und 11,2 g (0,048 Mol) 1 -(2-Chloräthyl)-3n- hexyl-2-imidazolidinon das 1-[2-[4-(4,5-Dihydro-thieno [2,3-b] [1]benzothiepin-4-yl)-1-piperazinyl] äthyl] -3 -n-hexyl-2-imidazolidinon mit dem Schmelzpunkt von 93-950, Dihydrochlorid 192-195 .
Das als Ausgangspunkt eingesetzte 1-(2-Chlor äthyl)-3-n-hexyl-2-imidazolidinon wird auf folgendem Wege hergestellt: b) 77,6 g (0,5 Mol) n-Hexylamino-äthanol werden mit 65,0 g (0,55 Mol) Diäthylkarbonat und 1 g Natrium langsam auf Rückflusstemperatur erwärmt.
Der bei der Reaktion freiwerdende Äthylalkohol wird während eines Zeitraumes von zwei Stunden kontinuielich über eine Kolonne abdestilliert. Der Rückstand wird im Hochvakuum rektifiziert. Das reine 3-n-Hexyl2-oxazolidinon siedet bei 1000/0,005 Torr, nD20 1,4564.
c) 51,3 g (0,3 Mol) des gemäss b) erhaltenen Produktes werden mit 34,8 g (0,33 Mol) (2-Chloräthyl)isocyanat und 2,4 g Lithiumchlorid etwa 2 Stunden auf 1800 erwärmt. Nach dem Abkühlen wird das Reaktionsgemisch unter Zusatz von 30 ml gesättigter Nafti umchloridlösung mit 300 ml Chloroform ausgeschütw telt. Die organische Phase wird über Magnesiumsulfat getrocknet, das Lösungsmittel im Vakuum entfernt und der Rückstand im Hochvakuum rektifiziert. Das erhaltene reine 1-(2-Chloräthyl)-3-n-hexyl-2-imidazolidinon siedet bei 121-127 /0,02 Torr, nD25 = 1,4818.
Beispiel S
12,6 g (0,05 Mol) SChIor-4,5-dihydro- thieno [2,3-b]'[1]benzothiepin werden in 25 ml Hexamethyl-phosphorsäure-triamid gelöst. Man tropft zu dieser Lösung 25,4 g (0,1 Mol) 1-[2-(1-Piperazinyl)-äthyl] 3-butyl-2-imidazolidinon in 25 ml abs. Benzol, erwärmt die Lösung 20 Stunden auf 800, kühlt sie auf 200 ab und giesst sie auf Eis.
Man extrahiert das Reaktionsgemisch mit Essigsäure äthylester und schüttelt die organische Phase sechsmal mit 2-n. Salzsäure aus. Die klaren, sauren Lösungen werden mit konz. Ammoniak alkalisch gestellt, mit Essigsäureäthylester erschöpfend extrahiert, mit Wasser gewaschen und die organische Phase über Natriumsulfat getrocknet. Durch Zugabe von überschüssiger ätherischer Salzsäure fällt das 1-[-[4-(4,5-Dihydrothieno[2,3-b] [1]benzothiepin-4-yl)-1-pip erazinyl] - äthyl] -3-butyl-2-imidazolidinondihydrochlorid-sesquihydrat aus. Smp. 191-193 .
Das als Ausgangsstoff benötigte 1-[2-(1-Piperazinyl)-äthyl] -3butyl)-2-imidazolidinon wird wie folgt erhalten: b) 40,4 g (0,2 Mol) 1-(2-Chloräthyl)-3butyl-2-imidazolidinon werden mit 35,0 g (0,22 Mol) Carbäthoxypiperazin in 200 ml Diäthylketon gelöst und unter Zugabe von 55,3 g (0,4 Mol) fein gepulvertem Kaliumkarbonat 24 Stunden unter Rückfluss gekocht, abgekühlt und filtriert. Der Rückstand wird zweimal mit je 200 ml Chloroform ausgekocht und abfiltriert.
Die vereinigten Filtrate werden im Vakuum zur Trockne eingedampft, und der ölige Rückstand wird im Hochvakuum destilliert. Das reine 1- [2-(N-Carbäthoxy-piperazinyl)- äthyl] -3-butyl-2-imidazolidinon siedet bei 190-192"/0,01 Torr, nD24 = 1,4941.
c) 54,0 g (0,164 Mol) 1- [2-(N-Carbäthoxy-piperazinyl)- äthyl] -3-butyl-2-imidazolidinon werden mit einer Lösung von 60 g Kaliumhydroxid in 300 ml Äthylalkohol 16 Stunden lang unter Rückfluss getrocknet. Der ausgefallene Niederschlag wird abfiltriert und mit heissem Äthanol ausgewaschen. Die vereinigten Filtrate werden dann zur Trockne eingedampft. Der Rückstand wird in 300 ml Benzol und 100 ml Wasser aufgenommen, die wässrige Phase nach Abtrennen mit Kaliumcarbonat gesättigt und viermal mit Benzol ausgeschüttelt. Die vereinigten benzolischen Lösungen werden über Kaliumcarbonat getrocknet und im Vakuum vom Lösungsmittel befreit.
Der Rückstand wird im Hochvakuum destilliert, wobei das reine t-[2-(1-Piperazinyl)-äthyl]- 3-butyl-2-imidazolidinon bei 155-160 /0,01 Torr übergeht; nD24 = 1,5033.
Beispiel 6
Analog Beispiel 5 wird aus 12,6 g (0,05 Mol) 4-Chlor-4,5-dihydrothieno[2,3-b][1]benzothiepin und 28,2 g (0,1 Mol) 1 - [2-Piperazinyl)-äthyl]- 3-n-hexyl-2-imidazolidinon das 1- [2-[4-(4,5-Dihydro-thleno [2,3-b] [1] benzothiepin-4-yl)-1-piperaz:inylj-äthyl] - 3-n-hexyl-2-imidazolidinon erhalten, Smp. 92-95 ,Dihydrochlorid 192-195".
Das als Ausgangsstoffe benötigte Imidazolidinonderivat wird analog Beispiel 5 b) und c) erhalten.

However, an acid-binding agent is preferably added to the reaction mixture. Suitable acid-binding agents are, for example, alkali metal carbonates, such as sodium or potassium carbonate, and also tertiary organic bases, such as e.g. Pyridine, triethylamine or, in particular, N, N-diisopropylethylamine.
Unnecessary tertiary bases can also be used as solvents.
If, in the reaction according to the invention, an alkali metal derivative of this compound is used instead of 4- (1-piperazinyl) -4,5-dihydthieno [2,3-b] [1] benzothiepin, e.g. a sodium, potassium or lithium derivative, it is advantageous to carry out the reaction in a hydrocarbon, e.g.
in benzene or toluene.
Of the starting materials that fall under the reactive esters of hydroxy compounds of the general formula II, 1 - (- chloro-ethyl) - and 1- (3-chloro-propyl) -3-methyl-2-imidazolidinone are known. The starting materials of the general formula III are described in the literature. Furthermore, 4 thieno [2,3-b] [1] benzothiepin, which is an example of the second reaction component, is also known. The corresponding 4-bromo-4,5-dihydrothieno [2,3-b] [1] benzothiepin can be prepared analogously.
The compounds of general formula I obtained by the process according to the invention are then, if desired, converted into their additive salts with inorganic and organic acids in the customary manner. For example, a solution of a compound of the general formula I with an organic solvent is mixed with the acid desired as the salt component or with a solution thereof. For the reaction, preference is given to choosing organic solvents in which the salt formed is sparingly soluble so that it can be separated off by filtration. Such solvents are e.g. Methanol, acetone, methyl ethyl ketone, Ace ton-ethanol, methanol-ether or ethanol-ether.
For use as a drug, pharmaceutically acceptable acid addition salts can be substituted for free bases; Salts with acids whose anions are not toxic at the dosage in question. It is also advantageous if the salts to be used as medicinal substances are readily crystallizable and have little or no hygroscopic properties. The following examples explain the preparation of the new compounds of the general formula
I and intermediate products not previously described, are not intended to limit the scope of the invention in any way. Temperatures are in

Example 1 a) 12.05 g (0.04 mol) 4- (1-piperazinyl) -thieno [2,3-b] [1] benzothiepin and 9.15 g (0.048 mol) 1 - (3-chloro- propyl) -3-ethyl-2-imidazolidinone are dissolved in 100 ml of diethyl ketone and 11.0 g (0.08 mol) of potassium carbonate are added. The reaction mixture is refluxed for 24 hours, poured onto 200 ml of ice water and 50 ml of 2-n added. Caustic soda too.
The organic phase is separated off and the aqueous alkaline solution is extracted with ether-methylene chloride (2: 1). The organic solutions are combined, washed with water and extracted with 250 ml of molar methanesulfonic acid solution. Then you put the acidic extract with conc. Sodium hydroxide solution is alkaline and the precipitated, crude base is poured out with 200 ml of ether methylene chloride (2: 1). The extract is washed with water, dried over magnesium sulfate and evaporated in vacuo.
The residue is dissolved in 50 ml of acetone, 150 ml of abs. Ether closes and the dihydrochloride precipitates with ethereal hydrochloric acid. The 1- (3-chloropropyl) -3 ethyl-2-imidazolidinone used as starting material is prepared as follows: b) 60.0 g (0.52 mol) of 3--2ithyl-2-oxazolidinone and 68, 1 g (0.57 mol) of (3-chloro-propyl) isocyanate with 4.8 g of lithium chloride and the mixture is heated to 175-180 for 90 minutes. The reaction mixture is then cooled and 300 ml of chloroform are added. The chloroform solution is separated off, washed three times with 30 ml of saturated sodium chloride acid each time, dried over magnesium sulfate and evaporated in vacuo. The residue is distilled in a high vacuum and pure 1- (3-chloro-propyl) 3-ethyl-2-imidazolidinone with a boiling point of 105-110 / 0.01 Torr is obtained;
Yield 91.6 g, 92.5 / o of theory.
Example 2 a) Analogously to Example 1 a), the following end product is obtained: From 10.6 g (0.035 mol) 4- (1-piperazinyl) -4,5-dihydthieno [2,3-b] [1] benzothiepin and 7.4 g (0.042 mol) of 1- (2-chloroethyl) -3-ethyl-2-imidazolidinone 1 - [2- [- (4,5-dihydro-thieno [2,3-b] [1] benzothiepin4) -yl) -1-piperazinyl] -ethyl] -3-ethyl-2-imidazolidinone of melting point 122-124; Yield 12.7 g, 82 / o of theory; Dihydrochloride m.p. 187-189.
b) 115 g (1.0 mol) of 3-ethyl-2-oxazolidinone and 116 g (1.1 mol) (2-chloro-ethyl) isocyanate are mixed with 10.0 g of lithium chloride and heated to 1800 for 2 hours. The reaction mixture is cooled, 300 ml of chloroform are added and the mixture is extracted three times with 30 ml of saturated sodium chloride solution each time. The organic phase is dried over magnesium sulfate and evaporated in vacuo. The oily residue is distilled under high vacuum.
The pure 1- (2-chloroethyl) -3-ethyl-2-imidazolidinone obtained boils at 95 / 0.01 Torr; and 24 ": 1.4913.
Yield 147.2 g, 83% of theory.
Example 3
The following end product is obtained analogously to Example 1 a): From 12.8 g (0.04 mol) 1- (4,5.Dihydro-thieno [2,3-b] [1] -benzo-thiepin-4-yl ) piperazine and 9.8 g (0.048 mol) 1- (2-chloroethyl) -3butyl-2-imidazolidinone is the 11- [2- [4- (4,5-dihydro-thieno [2,3-b] [1] benzothiepin-4-yl) -1-piperazinyl] ethyl [-3-butyl-2-imidazolidinone obtained, the dihydrochloride sesqui hydrate of which melts at 191-193.

The 1- (2-chloroethyl) -3-n-hexyl-2-imidazolidinone used as a starting point is prepared in the following way: b) 77.6 g (0.5 mol) of n-hexylamino-ethanol are 65.0 g (0.55 mol) diethyl carbonate and 1 g sodium slowly heated to reflux temperature.
The ethyl alcohol released during the reaction is continuously distilled off over a column over a period of two hours. The residue is rectified in a high vacuum. The pure 3-n-hexyl2-oxazolidinone boils at 1000 / 0.005 Torr, nD20 1.4564.
c) 51.3 g (0.3 mol) of the product obtained according to b) are heated to 1800 for about 2 hours with 34.8 g (0.33 mol) (2-chloroethyl) isocyanate and 2.4 g lithium chloride. After cooling, the reaction mixture is extracted by shaking with 300 ml of chloroform with the addition of 30 ml of saturated Nafti chloride solution. 12.6 g (0.05 mol) of chloro-4,5-dihydthieno [2,3-b] '[1] benzothiepin are dissolved in 25 ml of hexamethylphosphoric acid triamide. 25.4 g (0.1 mol) of 1- [2- (1-piperazinyl) ethyl] 3-butyl-2-imidazolidinone in 25 ml of abs are added dropwise to this solution. The 1- [2- (1-piperazinyl) ethyl] -3butyl) -2-imidazolidinone required as starting material is obtained as follows: b) 40.4 g (0.2 mol) 1- (2-chloroethyl) -3butyl -2-imidazolidinone is dissolved with 35.0 g (0.22 mol) of carbethoxypiperazine in 200 ml of diethyl ketone and refluxed for 24 hours with the addition of 55.3 g (0.4 mol) of finely powdered potassium carbonate, cooled and filtered. The residue is boiled twice with 200 ml of chloroform each time and filtered off.

c) 54.0 g (0.164 mol) of 1- [2- (N-carbethoxy-piperazinyl) -ethyl] -3-butyl-2-imidazolidinone are refluxed with a solution of 60 g of potassium hydroxide in 300 ml of ethyl alcohol for 16 hours dried. The deposited precipitate is filtered off and washed out with hot ethanol. The combined filtrates are then evaporated to dryness. The residue is distilled in a high vacuum, the pure t- [2- (1-piperazinyl) ethyl] -3-butyl-2-imidazolidinone passing over at 155-160 / 0.01 Torr; nD24 = 1.5033.
Example 6
Analogously to Example 5, from 12.6 g (0.05 mol) of 4-chloro-4,5-dihydrothieno [2,3-b] [1] benzothiepin and 28.2 g (0.1 mol) 1 - [2 -Piperazinyl) ethyl] - 3-n-hexyl-2-imidazolidinone 1- [2- [4- (4,5-dihydro-thleno [2,3-b] [1] benzothiepin-4-yl) - 1-piperaz: inylj-ethyl] -3-n-hexyl-2-imidazolidinone obtained, m.p. 92-95, dihydrochloride 192-195 ".
The imidazolidinone derivative required as starting materials is obtained analogously to Example 5 b) and c).
