Verfahren zur Herstellung von neuen Illlidazolidinonderivaten Die vorliegende Erfindung bett ein Verfahren zur Herstellung neuer Imidazohtionderivate.

in welcher n 2 oder 3 bedeutet, sowie ihre Additionssalze mit anorganischen oder organischen Säuren sind bis jetzt nicht bekannt geworden.
Wie nun gefunden wurde, besitzen solche Verbindungen - insbesondere das 1- [2-[4-(4,5-Dihydro-thieno[2,3-b] [l]benzo-thiepin-4-yl)-1-piperazinyl] - äthyl] -3-methyl-2-imidazolidinon - sowie ihre Salze wertvolle pharmakologische Eigenschaften und einen hohen therapeutischen Index. Sie wirken bei peroraler, rektaler oder parenteraler Verabreichung zentraldämpfend, z. B. vermindern sie die Motilität und potenzieren die Narkose.
Ferner weisen sie auch antiemetische, sympathicolytische, hypotherme und katatone Wirkung auf. Wirkungsqualitäten, welche durch ausgewählte Standardversuche [vgl. R. Domenjoz und W. Theobald, Arch.
Int. Pharmacodyn. 120, 450 (1959)] erfasst werden können, charakterisieren die Verbindungen als geeignet zur Behandlung von Schizophrenie, Spannungs- und Erregungszuständen und Hyperemesis.
Zur erfindungsgemässen Herstellung einer Verbindung der allgemeinen Formel I setzt man einen reaktionsfähigen Ester einer Verbindung der allgemeinen Formel II,

in welcher n die unter Formel 1 angegebene Bedeutung hat, mit dem 4(1 Piperazinyl)-4,5-dihydro thieno [2,3-b] [1]benzothiepin, oder mit einem Alkalimetallderivat dieser Verbindung, um und führt gegebenenfalls das Reaktionsprodukt mit einer anorganischen oder organischen Säure in ein Additionssalz über.
Geeignete reaktionsfähige Ester der Verbindungen der allgemeinen Formel II sind beispielsweise Halogenide, wie Chloride oder Bromide, ferner Sulfonsäureester, z. B. der Methansulfonsäureester oder der o- oder p-Toluolsulfonsäureester.
Diese Ester werden mit dem 4-(1-Piperazinyl)-4,5-dihydro thieno [2,3-b] [1]benzothiepin vorzugsweise in Gegenwart eines Lösungsmittels umgesetzt. Geeignete Lösungsmittel sind solche, die unter den Reaktionsbedingungen inert sind, beispielsweise Kohlenwasserstoffe, wie Benzol oder Toluol, Halogenkohlenwasserstoffe, wie Chloroform, ätherartige Flüssigkeiten, wie Äther oder Dioxan, niedere Alkanone, wie Aceton oder Methyläthylketon, sowie Phosphorsäureamide, z.B. das Hexameth'ylphosphorsäuretri amid.
Bei der erfindungsgemässen Umsetzung von einem Moläquivalent reaktionsfähigem Ester mit einem Mol äquivalent freier Base wird ein Moläquivalent Säure abgespalten. Diese Säure kann an überschüssiges 4-(1-Piperazinyl)-4,5-dihydro thieno[2,3-b] [l]benzothiepin oder auch an das dibasische Reaktionsprodukt gebunden werden. Vorzugsweise setzt man aber dem Reaktionsgemisch ein säurebindendes Mittel zu. Geeignete säurebindende Mittel sind beispielsweise Alkalimetallcarbonate, wie Natrium- oder Kaliumcarbonat, ferner tertiäre organische Basen, wie z. B. Pyridin, Triäthylamin oder insbesondere N,N-Diisopropyl-äthylamin.
Überschüssige tertiäre Basen können auch als Lösungsmittel eingesetzt werden.
Verwendet man bei der erfindungsgemässen Reaktion anstelle von 4-(1 -Piperazinyl)-4,5-dihydrothieno [2,3-b] [1]benzothiepin ein Alkalimetallderivat dieser Verbindung, z. B. ein Natrium-, Kalium- oder Lithiumderivat, so ist es vorteilhaft, die Reaktion in einem Kohlenwasserstoff, z. B.
in Benzol oder Toluol, durchzuführen.
Von den Ausgangsstoffen, die unter die reaktionsfähigen Ester von Hydroxyverbindungen der allgemeinen Formel II fallen, ist das 1 (2-Chlor-äthyl)-3-methyl- 2-imid azolidinon bekannt. Weitere Verbindungen von diesem Typus können analog hergestellt werden. Die zweite Reaktionskomponente, das 4-(1-Piperazinyl)-4,5-dihydrothieno[2,3-b] [1]benzothiepin ist z. B. wie folgt herstellbar:
Man geht von 4-Chlor-4,5-dihydro thieno[2,3-b] [1] benzothiepin aus und setzt dieses in Benzol mit 1-Piperazincarbonsäureäthylester zum 4-(4,5-Dihydro-thieno [2,3-b] [1] benzothiepin-4-yl)-1-piperazin- carbonsäureäthylester um, den man mit Hilfe von Kaliumhy < froxid in Äthanol hydrolysiert und decarboxyiiert.
Nach einem zweiten erfindungsgemässen Verfahren setzt man eine Verbindung der allgemeinen Formel III,

in welcher n die unter Formel I angegebene Bedeutung hat, oder ein Alkalimetallderivat einer solchen Verbindung, mit einem reaktionsfährigen Ester des 4-Hydroxy-4,54hydro thieno[2,3-b] [1]benzothiepin um und führt gegebenenfalls das Reaktionsprodukt mit einer anorganischen oder organischen Säure in ein Additionssalz über.
Geeignete Alkalimetallderivate von Verbindungen der allgemeinen Formel III sind beispielsweise Natrium-, Kalium- oder Lithiumderivate. Als reaktionsfähige Ester des 4-Hydroxy-4,5-dihydrothieno [2,3-b] [1]benzothiepin eignen sich z.B. Halogenide, wie Chloride oder Bromide, weiter Sulfonsäureester, wie der Methansulfonsäureester oder der o- oder p-Toluolsulfonsäureester.
Die erfindungsgemässe Umsetzung der freien Basen der allgemeinen Formel III, oder ihrer Alkalimetallderivate, mit den reaktionsfähigen Estern des 4-Hydroxy-4,5-dihydrothieno [2,3-b] [1]benzothiepin können in denselben Lösungsmitteln vorgenommen werden wie beim ersten Verfahren.
Werden für die Umsetzung die freien Basen eingesetzt, so können auch dieselben säurebindenden Mittel verwendet werden.
Die Ausgangsstoffe der allgemeinen Formel III sind in der Literatur beschrieben. Ferner ist das 4-Chlor-4,5-dihydro-thieno [2,3-b] [1]benzothiepin, das ein Beispiel für die zweite Reaktionskomponente ist, ebenfalls bekannt. Das entsprechende 4-Brom-4,5-dihydrothieno[2,3-b] [1]benzothiepin kann analog hergestellt werden.
Die nach den erfindungsgemässen Verfahren erhaltenen VerbindUngen der allgemeinen Formel I werden anschliessend gewünschtenfalls in üblicher Weise in ihre Additionssalze mit anorganischen und organischen Säuren übergeführt. Beispielsweise versetzt man eine Lösung einer Verbindung der allgemeinen Formel I mit einem organischen Lösungsmittel mit der als Salzkomponente gewünschten Säure oder mit einer Lösung derselben. Vorzugsweise wählt man für die Umsetzung organische Lösungsmittel, in denen das entstehende Salz schwer löslich ist, damit es durch Filtration abgetrennt werden kann. Solche Lösungsmittel sind z. B.
Methanol, Aceton, Methyläthylketon, Aceton-Athanol, Methanol-Äther oder Äthanol-Äther.
Zur Verwendung als Arzneistoffe können anstelle freier Basen pharmazeutisch annehmbare Säureadditionssalze eingesetzt werden, d. h. Salze mit solchen Säuren, deren Anionen bei den in Frage kommenden Dosierungen nicht toxisch sind. Ferner ist es von Vorteil, wenn die als Arzneistoffe zu verwendenden Salze gut kristallisierbar und nicht oder wenig hygroskopisch sind. Zur Salzbildung mit Verbindungen der allgemeinen Formel I können z. B. die Chlorwasserstoffsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Methansulfonsäure, Äthansulfonsäure, ss-Hydroxyäthansulfonsäure, Essigsäure, Äpfelsäure, Weinsäure, Citronensäure, Milchsäure, Oxalsäure, Bernsteinsäure, Fumarsäure, Maleinsäure, Benzoesäure, Salicylsäure, Phenylessigsäure, Mandelsäure und Embonsäure verwendet werden.
Die neuen Wirkstoffe werden, wie weiter vorne erwähnt, peroral, rektal oder parenteral Patienten verabreicht. Die täglichen Dosen der freien Basen oder von pharmazeutisch annehmbaren Salzen derselben bewegen sich zwischen 2-800 mg. Geeignete Doseneinheitsformen, wie Dragees, Tabletten, Kapseln, Suppositorien oder Ampullen, enthalten vorzugsweise 2-200 mg eines erfindungsgemässen Wirkstoffes oder eines pharmazeutisch annehmbaren Salzes desselben.
Die nachfolgenden Beispiele erläutern die Herstellung der neuen Verbindungen der allgemeinen Formel I und von bisher nicht beschriebenen Zwischenprodukten näher, sollen jedoch den Umfang der Erfindung in keiner Weise beschränken. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel 1 a) 14,6 g (0,05 Mol) 1-(4,5-Dihydro-thieno [2,3-b] [1]benzo thiepin-4-yl)-piperazin werden in 100 ml Methyläthylketon gelöst. Zu dieser Lösung fügt man 13,8 g (0,1 Mol) Kaliumcarbonat und tropft zum Reaktionsgemisch bei 40-50 10,0 g (0,06 Mol) 1-(2rChlor-äthy1)-3-methyl-2-imidazoliditlon. Das Gemisch wird unter gutem Rühren 20 Stunden unter Rücktisiss gekocht. Das Rohprodukt kristallisiert lang sam aus. Man kühlt die Suspension auf -100 ab. Dann saugt man den Niederschlag ab, suspendiert ihn in Wasser, rührt die Suspension, nutscht das Rohprodukt ab und kristallisiert es aus viel Aceton um.
Das erhaltene 1-[2-[4-(4,5-Dihydro-thieno[2,3-b] [1] benzothiepin-4-yl)- 1 -piperazinyl] - äthyl] -3-methyl-2-imidazolidinon schmilzt bei 180-182".
14,0 g der erhaltenen Base werden in 100 ml heissem Äthanol gelöst und mit 2 Moläquivalent äthanolischer Salzsäure neutralisiert; man fügt 50ml Essigsäureäthylester zu und kühlt das Gemisch auf -5 . Das auskristallisierte Hydrochlorid wird abgesaugt, mit wenig kaltem Athanol gewaschen, bei 1000 im Vakuum getrocknet und aus abs. Äthanol umkristallisiert. Das Hydrochlorid des 1-[2- [4-(4,5-Dihydro-thieno[2,3-b] [1]benzothiepin-4-yl)- 1-piperazinyl] äthyl] -3-methyl-2-imidazolidinon schmilzt bei 2100.
Zur Bildung des Bisrmethansulfonat Salzes werden 10,7 g (0,025 Mol) der Base in einem Gemisch von 20ml Benzol und 20 ml Methyläthylketon heiss gelöst und mit einer Lösung von 4,8 g (0,05 Mol) Methansulfonsäure in 10 ml Methyläthylketon versetzt. Nach Zugabe von 50ml absolutem Diäthyläther wird das auskristallisierte Produkt abfiltriert und aus Äthylalkoholl Essigsäureäthylester umkristallisiert. Das reine 1- [2-[4-(4,5-Dihydro-thieno [2,3-b] [1]benzothiepin-4-yl)-1-piperazinyl] - äthyl] -3-methyl-2-imidazolidinon-bismethansulfonat schmilzt bei 182-184 .
Die Ausgangsverbindung, das 1-(4,5-Dihydro-thieno[2,3-b] [1] benzothiepin-4-yl)-piperazin, wird wie folgt hergestellt: b) 12,6 g (0,05 Mol) 4-Chlor-thieno[2,3-b][1]benzothiepin werden in eine Lösung von 23,6 g (0,15 Mol) 1-Piperazincarbonsäureäthylester in 20 ml Benzol eingetragen. Das Gemisch wird 5 Stunden unter Rückfluss gekocht, in 200ml Wasser und 5ml konz.
Natronlauge aufgenommen und mit Äther-Methvlen chlorid (2:1) extrahiert. Man wäscht die organische Phase viermal mit Wasser, trocknet sie über Magnesiumsulfat und dampft sie im Vakuum ein. Der erhaltene, rohe 4-(4,5-Dihydro-thieno[2,3-b] [1]benzothiepin-4-yl)-1-piperazin carbonsäureester schmilzt bei 118-1250, die reine Verbindung nach Umkristallisieren aus Isopropanol/Petroläther bei 136-1380.
c) 10,4 g (0,0278 Mol) des nach b) erhaltenen Esters werden in eine Lösung von 18 g (0,316 Mol) Kaliumhydroxid in 100 ml abs. Äthanol eingetragen.
Das Gemisch wird 12 Stunden unter Rückfluss gekocht, mit 50 ml Wasser verdünnt, das Äthanol im Vakuum abgedampft und der Rückstand mit Ather- Methylenchlorid (2:1) extrahiert. Man trocknet die organische Phase über Magnesiumsulfat und dampft sie im Vakuum ein. Der Rückstand wird aus Ligroin Essigsäureäthylester umkristallisiert, wonach das 1-(4,5-Dihydro-thieno[2,3-b] [1] benzothiepin-4-yl)-piperazin bei 126-128 schmilzt.
Das gleiche Endprodukt kann auch wie folgt erhalten werden: d) 10,6 g (0,05 Mol) 1-[2-(1-Piperazinyl)-äthyl]-3-methyl- 2-imidazolidinon werden in 50 ml absolutem Benzol gelöst und bei 0 tropfenweise mit einer Lösung von 3,2 g (0,05 Mol) Butyllithium in Hexan versetzt. Das Reaktionsgemisch wird auf Raumtemperatur gebracht und 1 Stunde bei 400 gerührt. Nach erneuter Abkühlung auf 0 wird eine Lösung von 12,6 g (0,05 Mol) 4-Chlor-4,5-dihydrothieno[2,3-b] [1]benzothiepin in 40ml absolutem Benzol im Verlaufe von 30 mini ten zugetropft. Das Reaktionsgemisch wird im Verlaufe von einer Stunde auf Raumtemperatur erwärmt und anschliessend 4 Stunden unter Rückfluss gekocht. Das Gemisch wird dann in 200 ml Eiswasser gegossen, mit 25 ml 2-n.
Natronlauge versetzt und mit Benzol ausgeschüttelt. Die vereinigten benzolischen Extrakte werden mit 200ml einer 1-mol. Methansulfonsäurelösung ausgeschüttelt, und die saure wässrige Phase wird dann mit konz. Natronlauge alkalisch gestellt und wiederum mit Benzol ausgezogen. Die erhaltenen benzolischen Extrakte werden mit Wasser gewaschen, über Magnesiumsulfat getrocknet und im Vakuum vom Lösungsmittel befreit.
Der erhaltene Rückstand wird aus Benzol/Petrol äther umkristallisiert. Das reine 1-[2-[4-(4,5-Dihydro-thieno[2,3-b] [1]benzothiepin-4-yl)-1-piperazinyl] äthyl] -3-methyl-2-imidazolidinon schmilzt bei 180-182 .
Zum gleichen Endprodukt führt ebenfalls die foIgende Umsetzung: e) 11,7g (0,05 Mol) 4Hydroxy-4,5-dihydro-thieno [2,3-b] [1]benzothiepin und 4,75 g (0,06 Mol) Pyridin werden in 50 ml absolu- tem Toluol gelöst und tropfenweise bei -100 mit einer Lösung von 5,7 g (0,05 Mol) Methansulfonsäurechlorid in 10 ml absolutem Toluol im Verlaufe von 30 Minuten versetzt. Das Gemisch wird 2 Stunden bei -10 gerührt und alsdann bei 0 einer Lösung von 21,2 g (0,1 Mol) 1 - [2-( 1 -Piperazinyi)-äthyl]- 3-methyl-2-imidazolidinon in 50 ml absolutem Benzol zugetropft. Das Reaktionsgemisch wird dann im Verlaufe von 1 Stunde auf Raumtemperatur erwärmt und anschliessend 4 Stunden unter Rückfluss gekocht.
Das Gemisch wird in 200 ml Eiswasser gegossen und wie unter b) beschrieben weiter aufgearbeitet. Das erhaltene 1-[2-[4-(4,5-Dihydrothieno[2,3-b] [1] benzothiepin-4-yl)-1-piperazinyl] - äthyl] -3-methyl-2-imidazolidinon ist mit dem unter a) und b) erhaltenen Produkt identisch und schmilzt bei 180-182".
Beispiel 2
25,0 g (0,1 Mol) 4-Chlor-4,5-dihydro-thieno[2,3-b] [1] benzothiepin werden in 50 ml Hexamethyl-phosphorsäuretriamid gelöst. Man tropft zu dieser Lösung 42,0 g (0,2 Mol) 1 [2-(1-Piperazinyl)-äthyl3-methyl-2-imidazolidinon in 50 ml abs. Benzol, erwärmt die Lösung 20 Stunden auf 800, kühlt sie auf 200 ab und giesst sie auf Eis.
Man extrahiert das Reaktionsgemisch mit Essigsäure äthylester und schüttelt die organische Phase sechsmal mit 2-n. Salzsäure aus. Die klaren, sauren Lösungen werden mit konz. Ammoniak alkalisch gestellt, die ausgefallenen Kristalle abgesaugt, mit Wasser gewaschen und aus viel Aceton umkristallisiert. Das erhaltene 1-[2-[4-(4,5-Dihydro-thieno[2,3-b] [1] benzothiepin-4-yl)-1-piperazinyl] -äthyl] 3-methyl-2-imidazolidinon schmilzt bei 180-1820.
Beispiel 3
9,5 g (0,03 Mol) 1-(4,5-Dihydro-thieno[2,3-b] [1] benzo- thiepin-4-yl)-piperazin werden mit 6,35 g (0,036 Mol) 1-(3 -Chlorpropyl)-3-methyl2-imidazolidinon und 8,3 g (0,06 Mol) Kaliumcarbonat 36 Stunden in 70 ml Diätltylketon unter Rückituss gekocht Dann giesst man das Reaktionsgemisch auf 200 ml Eiswasser, fügt 40 ml 2-n. Natronlauge zu und extrahiert das Gemisch mit Chloroform. Man wäscht den organischen Extrakt mit Wasser, trocknet ihn über Magnesiumsulfat und dampft ihn im Vakuum ein.
Die rohe Base wird in 50 ml Aceton gelöst und die Acetonlösung mit ätherischer Salzsäure bis zur kongosauren Reaktion versetzt. Man filtriert das rohe Dihy drochlorid, das ausfällt, ab, wäscht es mit Aceton sowie Äther und trocknet es im Vakuum. Umkristallisieren des Rohproduktes aus 96 0/obigem Äilanol-Äther liefert das reine 1-[3-[4-(4,5-Dihydro-thieno[2,3-b] [1] benzothiepin-4-yl)-1-piperazinyl] propyl]-3-methyl-2-imidazolidinon- dihydrochlorid vom Smp. 189-192".

These esters are reacted with the 4- (1-piperazinyl) -4,5-dihydro thieno [2,3-b] [1] benzothiepin, preferably in the presence of a solvent. Suitable solvents are those which are inert under the reaction conditions, for example hydrocarbons such as benzene or toluene, halogenated hydrocarbons such as chloroform, ethereal liquids such as ether or dioxane, lower alkanones such as acetone or methyl ethyl ketone, and phosphoric acid amides, e.g. the Hexameth'ylphosphorsäuretri amid.

If the free bases are used for the reaction, the same acid-binding agents can also be used.
The starting materials of the general formula III are described in the literature. Furthermore, 4-chloro-4,5-dihydro-thieno [2,3-b] [1] benzothiepin, which is an example of the second reactant, is also known. The corresponding 4-bromo-4,5-dihydrothieno [2,3-b] [1] benzothiepin can be prepared analogously.
The compounds of the general formula I obtained by the process according to the invention are then, if desired, converted into their addition salts with inorganic and organic acids in the customary manner. For example, a solution of a compound of the general formula I with an organic solvent is mixed with the acid desired as the salt component or with a solution thereof. For the reaction, preference is given to choosing organic solvents in which the salt formed is sparingly soluble so that it can be separated off by filtration. Such solvents are e.g. B.
Methanol, acetone, methyl ethyl ketone, acetone-ethanol, methanol-ether or ethanol-ether.
For use as drugs, pharmaceutically acceptable acid addition salts can be used in place of free bases; H. Salts with acids whose anions are not toxic at the dosages in question. It is also advantageous if the salts to be used as medicinal substances are readily crystallizable and have little or no hygroscopic properties. For salt formation with compounds of general formula I, for. B. the hydrochloric acid, hydrobromic acid, sulfuric acid, phosphoric acid, methanesulfonic acid, ethanesulfonic acid, β-hydroxyethanesulfonic acid, acetic acid, malic acid, tartaric acid, citric acid, lactic acid, oxalic acid, succinic acid, fumaric acid, maleic acid, benzoic acid, salicylic acid, phenyl acetic acid.
As mentioned above, the new active ingredients are administered orally, rectally or parenterally to patients. The daily doses of the free bases or pharmaceutically acceptable salts thereof range between 2-800 mg. Suitable dosage unit forms, such as dragees, tablets, capsules, suppositories or ampoules, preferably contain 2-200 mg of an active ingredient according to the invention or a pharmaceutically acceptable salt thereof.
The following examples explain the preparation of the new compounds of general formula I and of intermediates not previously described, but are not intended to restrict the scope of the invention in any way. The temperatures are given in degrees Celsius.
Example 1 a) 14.6 g (0.05 mol) of 1- (4,5-dihydro-thieno [2,3-b] [1] benzo thiepin-4-yl) piperazine are dissolved in 100 ml of methyl ethyl ketone. 13.8 g (0.1 mol) of potassium carbonate are added to this solution and 10.0 g (0.06 mol) of 1- (2r chloro-ethy1) -3-methyl-2-imidazoliditlone are added dropwise to the reaction mixture at 40-50. The mixture is boiled under back pressure for 20 hours while stirring well. The crude product slowly crystallizes out. The suspension is cooled to -100. The precipitate is then filtered off with suction, suspended in water, the suspension is stirred, the crude product is filtered off with suction and recrystallized from a large amount of acetone.
The 1- [2- [4- (4,5-dihydro-thieno [2,3-b] [1] benzothiepin-4-yl) -1-piperazinyl] -ethyl] -3-methyl-2-imidazolidinone obtained melts at 180-182 ".
14.0 g of the base obtained are dissolved in 100 ml of hot ethanol and neutralized with 2 molar equivalents of ethanolic hydrochloric acid; 50 ml of ethyl acetate are added and the mixture is cooled to -5. The crystallized hydrochloride is filtered off with suction, washed with a little cold ethanol, dried at 1000 in a vacuum and from abs. Recrystallized ethanol. The hydrochloride of 1- [2- [4- (4,5-dihydro-thieno [2,3-b] [1] benzothiepin-4-yl) -1-piperazinyl] ethyl] -3-methyl-2-imidazolidinone melts at 2100.
To form the bisrmethanesulfonate salt, 10.7 g (0.025 mol) of the base are dissolved in a mixture of 20 ml of benzene and 20 ml of hot methyl ethyl ketone and a solution of 4.8 g (0.05 mol) of methanesulfonic acid in 10 ml of methyl ethyl ketone is added. After adding 50 ml of absolute diethyl ether, the product which has crystallized out is filtered off and recrystallized from ethyl alcohol and ethyl acetate. The pure 1- [2- [4- (4,5-dihydro-thieno [2,3-b] [1] benzothiepin-4-yl) -1-piperazinyl] -ethyl] -3-methyl-2-imidazolidinone -Bismethanesulfonate melts at 182-184.
The starting compound, 1- (4,5-dihydro-thieno [2,3-b] [1] benzothiepin-4-yl) piperazine, is prepared as follows: b) 12.6 g (0.05 mol) 4-chloro-thieno [2,3-b] [1] benzothiepine are introduced into a solution of 23.6 g (0.15 mol) of ethyl 1-piperazine carboxylate in 20 ml of benzene. The mixture is refluxed for 5 hours, concentrated in 200ml water and 5ml.
Sodium hydroxide solution was added and extracted with ether-methylene chloride (2: 1). The organic phase is washed four times with water, dried over magnesium sulfate and evaporated in vacuo. The resulting crude 4- (4,5-dihydro-thieno [2,3-b] [1] benzothiepin-4-yl) -1-piperazine carboxylic acid ester melts at 118-1250, the pure compound after recrystallization from isopropanol / petroleum ether at 136-1380.
c) 10.4 g (0.0278 mol) of the ester obtained according to b) are in a solution of 18 g (0.316 mol) of potassium hydroxide in 100 ml of abs. Ethanol registered.
The mixture is refluxed for 12 hours, diluted with 50 ml of water, the ethanol is evaporated off in vacuo and the residue is extracted with ether-methylene chloride (2: 1). The organic phase is dried over magnesium sulphate and evaporated in vacuo. The residue is recrystallized from ligroin ethyl acetate, after which the 1- (4,5-dihydro-thieno [2,3-b] [1] benzothiepin-4-yl) piperazine melts at 126-128.
The same end product can also be obtained as follows: d) 10.6 g (0.05 mol) of 1- [2- (1-piperazinyl) ethyl] -3-methyl-2-imidazolidinone are dissolved in 50 ml of absolute benzene and at 0 a solution of 3.2 g (0.05 mol) of butyllithium in hexane is added dropwise. The reaction mixture is brought to room temperature and stirred at 400 for 1 hour. After cooling again to 0, a solution of 12.6 g (0.05 mol) of 4-chloro-4,5-dihydrothieno [2,3-b] [1] benzothiepin in 40 ml of absolute benzene is added dropwise in the course of 30 minutes . The reaction mixture is warmed to room temperature over the course of one hour and then refluxed for 4 hours. The mixture is then poured into 200 ml of ice water, with 25 ml of 2-n.
Sodium hydroxide solution was added and extracted with benzene. The combined benzene extracts are with 200ml of a 1-mol. Methanesulfonic acid solution extracted, and the acidic aqueous phase is then with conc. Sodium hydroxide solution made alkaline and again extracted with benzene. The benzene extracts obtained are washed with water, dried over magnesium sulfate and freed from the solvent in vacuo.
The residue obtained is recrystallized from benzene / petroleum ether. The pure 1- [2- [4- (4,5-dihydro-thieno [2,3-b] [1] benzothiepin-4-yl) -1-piperazinyl] ethyl] -3-methyl-2-imidazolidinone melts at 180-182.
The following reaction also leads to the same end product: e) 11.7 g (0.05 mol) of 4-hydroxy-4,5-dihydro-thieno [2,3-b] [1] benzothiepine and 4.75 g (0.06 mol ) Pyridine is dissolved in 50 ml of absolute toluene and a solution of 5.7 g (0.05 mol) of methanesulfonic acid chloride in 10 ml of absolute toluene is added dropwise at -100 over the course of 30 minutes. The mixture is stirred for 2 hours at -10 and then at 0 a solution of 21.2 g (0.1 mol) of 1 - [2- (1-piperazinyi) ethyl] -3-methyl-2-imidazolidinone in 50 ml absolute benzene was added dropwise. The reaction mixture is then warmed to room temperature over the course of 1 hour and then refluxed for 4 hours.
The mixture is poured into 200 ml of ice water and worked up further as described under b). The 1- [2- [4- (4,5-dihydrothieno [2,3-b] [1] benzothiepin-4-yl) -1-piperazinyl] -ethyl] -3-methyl-2-imidazolidinone is with identical to the product obtained under a) and b) and melts at 180-182 ".
Example 2
25.0 g (0.1 mol) of 4-chloro-4,5-dihydro-thieno [2,3-b] [1] benzothiepine are dissolved in 50 ml of hexamethylphosphoric triamide. 42.0 g (0.2 mol) of 1 [2- (1-piperazinyl) ethyl3-methyl-2-imidazolidinone in 50 ml of abs. Benzene, heat the solution to 800 for 20 hours, cool it to 200 and pour it onto ice.
The reaction mixture is extracted with ethyl acetate and the organic phase is shaken six times with 2-n. Hydrochloric acid. The clear, acidic solutions are mixed with conc. Made ammonia alkaline, suction filtered, washed with water and recrystallized from a lot of acetone. The 1- [2- [4- (4,5-dihydro-thieno [2,3-b] [1] benzothiepin-4-yl) -1-piperazinyl] ethyl] 3-methyl-2-imidazolidinone melts at 180-1820.
Example 3
9.5 g (0.03 mol) of 1- (4,5-dihydro-thieno [2,3-b] [1] benzothiepin-4-yl) piperazine are mixed with 6.35 g (0.036 mol) 1- (3 -Chlorpropyl) -3-methyl2-imidazolidinone and 8.3 g (0.06 mol) of potassium carbonate boiled under reflux for 36 hours in 70 ml of diethyl ketone. The reaction mixture is then poured into 200 ml of ice water, 40 ml of 2-n . Sodium hydroxide solution and the mixture extracted with chloroform. The organic extract is washed with water, dried over magnesium sulfate and evaporated in vacuo.
The crude base is dissolved in 50 ml of acetone and ethereal hydrochloric acid is added to the acetone solution until it reacts with Congo acid. The crude dihydrochloride that precipitates is filtered off, washed with acetone and ether and dried in vacuo. Recrystallization of the crude product from 96 0 / above Äilanol -ether gives the pure 1- [3- [4- (4,5-dihydro-thieno [2,3-b] [1] benzothiepin-4-yl) -1-piperazinyl ] propyl] -3-methyl-2-imidazolidinone dihydrochloride of m.p. 189-192 ".
