Verfahren zur Herstellung von Gonatrienen
Diese Erfindung betrifft ein Verfahren zur Herstellung von mit Östradiol verwandten Gonatrienen, welche östrogene, antilipämische Hypophysen-gonadotrophininhibierungs- oder andere Steroidhormon-Wirksamkeit haben und/oder welche Zwischenprodukte für die Herstellung anderer Gonatrienderivate sind.
Erfindungsgemäss werden hergestellt Oona- 1,3,5(10)- triene der Formel I

worin RO eine freie, verätherte oder veresterte Hydroxylgruppe, R1 einen Alkylrest mit 2 bis 4 Kohlenstoffatomen bedeuten, die Wasserstoffatome in den Stellungen 8, 9 und 14 und der Rest R1 in der trans-anti-trans Konfiguration stehen und die 16-Hydroxylgruppe in der trans-Stellung zu R1 steht.
Das erfindungsgemässe Verfahren zur Herstellung dieser Verbindungen ist dadurch gekennzeichnet, dass man eine Verbindung der Formel II

worin R2 einen Acylrest darstellt, die stereochemische Konfiguration die gleiche ist wie in Formel I und die 16,17-Epoxygruppe in trans-Stellung zu Rt steht, der Hydrolyse unterwirft.
Die Verbindungen der Formel I können durch Acylierung der 1 6-Hydroxylgruppe in die entsprechenden 1 6-Ester und verätherte oder veresterte 3-Hydroxylgruppen durch Hydrogenolyse bzw. Hydrolyse in die freie Hydroxylgruppe übergeführt werden.
R kann beispielsweise Methyl, Äthyl, Allyl, Methoxymethyl, Propyl, 2'-Tetrahydropyranyl, Benzyl oder ss-Chloräthyl oder sie kann Acetyl, Propionyl, Benzoyl oder Chloracetyl sein. R1 ist vorzugsweise Äthyl, kann aber n-Propyl, n-Butyl oder Isobutyl sein.
Erfindungsgemäss erhältliche Verbindungen, die in 3-Stellung eine Methoxymethoxy-, 2'-Tetrahydropyranyloxy- oder Acyloxygruppe aufweisen, können leicht hydrolysiert werden, beispielsweise mittels verdünnter Mineralsäure; Verbindungen, die in 3-Stellung eine Alkoxygruppe tragen, können beispielsweise mit einem Gemisch aus Bromwasserstoffsäure und Essigsäure unter Erhitzen auf Rückflusstemperatur hydrolysiert werden; Verbindungen, die in 3-Stellung eine Benzyl- oder Allylgruppe tragen, lassen sich durch Hydrogenolyse leicht in die 3-Hydroxyverbindungen überführen. Letztere können leicht, beispielsweise mittels Natriumhydroxyd und einem Dialkylsulfat, zu 3-Alkoxyverbindungen ver äthert oder, z. B. mittels eines Säureanhydrids in Pyridin, zu 3-Acyloxyverbindungen oder analogen Estern verestert werden.

In den obigen Strukturen I bis IV sind die 13ss- und 13a-Verbindungen, erläutert gemäss Horeau-Reichstein Convention [vgl. Fieser & Fieser, Steroide, Reinhold (1959), Seite 336], nicht getrennt unterschieden, und in diesen Strukturen hat eine feste Linie, die ein Atom oder eine Gruppe mit dem Steroidkern verbindet, keine Bedeutung hinsichtlich der Konfiguration, sondern wird allein verwendet, um die Stellung in dem Kern anzugeben, in welcher das Atom oder die Gruppe verbunden ist. Das Produkt einer Gesamtsynthese, die keine geeignete Trennungsstufe enthalten hat, wird die 13ssund 13a-Formen in äquimolekularem Gemisch oder racemischer Form enthalten.
Vorzugsweise ist das Aus gangsmaterial ein isoliertes 1 3ss-Enantiomeres. Die Erfindung schliesst in besonderer Weise die Verwendung von Enantiomeren mit der 13B-Alkylgruppe in Gegenwart oder Abwesenheit ihrer 13a-Alkyl-Enantiomere ein. Sie schliesst ein die isolierten 13ss-Alkylverbindungen und die 13ss-Formen im Gemisch mit den entsprechenden 13a-Formen, besonders racemische Gemische.
Die gemäss den Beispielen hergestellten Verbindungen waren Racemate und werden als 13p-Formen bezeichnet, und das (+ )- oder (dl)-Präfix der Horeau Reichstein-Convention wurde weggelassen.
Die erfindungsgemäss erhaltenen Verbindungen können in pharmazeutische Zubereitungen eingearbeitet werden.
Die genannten Verbindungen zeigen verschiedene Steroidhormonwirksamkeiten und/oder sind i;wischen- produkte für die Herstellung anderer aktiver Steroide.
So wurden in Standard-Untersuchungsverfahren die nachfolgenden Wirksamkeiten durch die unten angegebenen Verbindungen aufgezeigt: 13ss-Äthylgona- 1,3,5(10)-trien-3,16a,17ss-triol zeigt unterdrückte Östrogenwirksamkeit, anti-lipämische Wirk- samkeit und Hypophysen-gonadotrophin-Inhibierungswirksamkeit.
3,16a-Diacetoxy-13ss-äthylgona-1,3,5(10)-trien-17-on zeigt anti-lipämische Wirksamkeit.
13ss - Äthyl - 3 -methoxygona-l ,3,5(10)-trien- 16a,17ss- diol zeigt Hypophysen-gonadotrophin-Inhibierungswirksamkeit.
Beispiel 1 a) 3, l7-Macetoxy-l 3ss-äthyl-gona-l ,3,5(10),16-tetraen 3 -Acetoxy- 13ss-äthyl-gona- 1,3,5(10)-trien- 17-on (3,31 g) wurde in Isopropenylacetat (50 ml) gelöst, mit p-Toluolsulfonsäure (500 mg) behandelt und in einem kochenden Wasserbad über Nacht erhitzt. Das Lösungsmittel wurde teilweise abdestilliert, das Gemisch mit Äther verdünnt und gewaschen, getrocknet und zu einem braunen Öl (3,54 g) eingedampft. Dieses Produkt wurde chromatographiert über Florisil (200 g) unter Eluieren mit Benzol. Die zuerst anfallenden Fraktionen ergaben 3,17-Diacetoxy-13ss-äthyl-gona-1,3,5(10),16- tetraen (1,44 g), das als Ö1 erhalten wurde, welches langsam kristallisierte.
Die Umkristallisation aus Äthanol bei 50 C ergab dieses Diacetat als Plättchen; Schmelzpunkt 89 bis 89,50 C. (Gefunden: 75,25 % C; 7,34% H; die Bruttoformel C23H2804 erfordert 74,97 % C; 7,66 % H.) b) 3 -Acetoxy- 1 3ss-äthyl-gona- 1,3,5(1 0)-trien- 16a-ol-17-on
3,17 - Diacetoxy -13ss-äthyl-gona-1,3,5(10),16-tetraen (1,170 g) wurde in Natrium-Trockenäther (50 ml) gelöst und mit einer Lösung von Monoperphthalsäure in Äther (0,28 molar; 25 ml) behandelt. Man liess das Gemisch bei Zimmertemperatur drei Tage stehen, und es wurde dann mit eiskalter Natriumcarbonatlösung und Salzlösung gewaschen, eingedampft zur Bildung eines Öls (1,147 g).
Dieses Produkt wurde dann mit einem solchen vereinigt, das zu einem früheren Zeitpunkt aus einem kleinen Ansatz erhalten worden war (140 mg) und über Florisil (100 g) chromatographiert und mit Benzol und Benzol-Äther eluiert.
Aus den zuerst anfallenden Benzol-Ätherfraktionen wurde 3,17ss-Diacetoxy- 16a- 17a-epoxy- 13ss-äthyl- gona-1,3,5(10)-trien (575,4 mg) als ein Ö1 erhalten, welches bei Behandlung mit Äther sich verfestigte. Aus den späteren Benzol-Ätherfraktionen wurde ein Gemisch des obigen Epoxyds und nachfolgendes Ketol (52,2 mg) als weisser Feststoff erhalten. Aus den weiteren Benzol-Äther-, Äther- und Äther-Methanolfraktionen wurde 3 -Acetoxy- 13 ss-äthyl- gona-1,3,5(10)-trien-16a-ol-17-on (544,9 mg) erhalten.
Das erste Eluat wurde aus Äthanol auskristallisiert zur Bildung des Epoxyds (344,2 mg), Schmelzpunkt 170 bis 171,50 C, und wurde aus Äthanol umkristallisiert, Schmelzpunkt 172 bis 173,50 C. (Gefunden: 71,95 % C; 7,24 % H; die Bruttoformel Cg3H28OÏ erfordert 71,85 % C; 7,34 % H.) Das zweite Eluat wurde zur Bildung des Ketols (264,5 mg) aus wässrigem Alkohol umkristallisiert;
Schmelzpunkt 166 bis 166,50 C. (Eine Probe kristallisierte in eine isomorphe Form um, Schmelzpunkt 181,5 bis 1830C.) (Gefunden: 73,36% C; 7,53 % H; die Bruttoformel C2lY,;04 erfordert 73,66 % C; 7,66 % H.) c) 1 3fi-Äthyl-gona- 1,3,5(1 0)-trien-3 1 6o-diol- 1 7-on 3,17ss-Diacetoxy-16a-17a-epoxy-13,/3-äthyl- gona-1,3,5(10)-trien (406,5 mg) wurde in warmem Methanol (60 ml) und Aceton (15 ml) gelöst. Die Lösung wurde auf Zimmertemperatur gekühlt und mit 6n Schwefelsäure (12 ml) unter Schütteln behandelt. Man liess das homogene Gemisch bei Zimmertemperatur 6 Tage stehen und es wurde dann mit einem gleichen Volumen Äthylacetat verdünnt.
Das Gemisch wurde auf 0 C gekühlt und mit eiskalter wässriger Lösung von Natriumbicarbonat behandelt, bis es alkalisch war. Die wässrige Schicht wurde mit Äthylacetat (2 X 150 ml) extrahiert und die Extrakte zusammengegeben und der ursprünglich erhaltenen organischen Schicht zugegeben. Diese Lösung wurde mit Wasser gewaschen, getrocknet und eingedampft. Der zurückbleibende weisse Feststoff (409 mg) wurde aus Äthanol (10 ml) umkristallisiert und ergab 13ss-Athyl-gona- 1,3,5(10)-trien-3,16-diol-17-on (258,7 mg); Schmelzpunkt 198 bis 200,50 C (die Kristalle wurden undurchsichtig bei ungefähr 1400 C) und hatte, umkristallisiert aus Äthanol, einen Schmelzpunkt von 200 bis 202,50 C.
(Gefunden: 73,80% C; 7,95 % H; die Bruttoformel C39H2403. y2 C2H5OH erfordert 74,27 % C; 8,41% H.)
Beispiel 2 3,16a-Diacetoxy- 13ss-äthyl-gona-1,3,5(10)- trien-17-on a) 13ss-Äthyl-gona-1,3,5(10)-trien-3,16α-diol-17-on (47,1 mg) wurde in Pyridin (1 ml) und Essigsäureanhydrid (0,6 ml) gelöst und 72 Stunden bei Zimmertemperatur stehengelassen. Die Lösungsmittel wurden im Vakuum entfernt und das Produkt, aus Äthanol (2 ml) auskristallisiert, ergab 3,16α-Diacetoxy-1 3,1 6ct-Diacetoxy-13ss-äthyl- gona-1,3,5(10)-trien-17-on (54,2 mg); Schmelzpunkt 172 bis 1730 C.
b) 3-Acetoxy-13ss-äthyl-gona-1,3,5(10)-trien-16a-ol- 17-on (271,5 mg) wurde in Pyridin (2 ml) und Essigsäureanhydrid (1,2 ml) gelöst und die Lösung bei Zimmertemperatur 72 Stunden stehengelassen. Die Lösungsmittel wurden im Vakuum entfernt, der zurückbleibende Feststoff wurde aus Äthanol (10 ml) umkristallisiert und ergab 3,16-Diacetoxy- - 13ss - äthyl- gona-1,3,5(10)- trien-17-on (238,5 mg);
Schmelzpunkt 173 bis 1740 C, und aus Athanol umkristallisiert, Nadeln, mit einem Schmelzpunkt bei 173 bis 173,50 C. (Gefunden: 71,91 X C; 7,20% H; die Bruttoformel C2sHo805 erfordert 71,85% C; 7,34% H.)
Beispiel 3 13ss-Sithyl-3-methoxy-gona- 1,3,5(10)-trien-
16a-ol-17-on 1 7ss - Acetoxy -1 6a, 1 7a - epoxy- 1 3ss-äthyl-3 -methoxy- gona-1,3,5(10)-trien (3,6 g) in Methanol (500 ml) wurde mit 6n Schwefelsäure (75 cm3) behandelt und das Gemisch 4 Tage bei 250 C gerührt. Die Lösung wurde unter Vakuum auf ein kleines Volumen reduziert, Wasser zugegeben und das Gemisch in einem Eisbad gekühlt.
Der Niederschlag wurde filtriert, mit Wasser gewaschen und getrocknet und man erhielt die oben bezeichnete Verbindung (2,98 g); Schmelzpunkt 130 bis 1380 C, homogen durch Dünnschichtchromatographie; Infrarotabsorptionsmaxima bei 2,95, 3,45, 3,53, 5,76, 6,21, 6,37, 6,67 ,u.
Der Ausgangsstoff wurde wie folgt hergestellt: 13ss- Äthyl - 3 - methoxy - gona- 1,3,5(10) -trien-17-on (16,8 g) in Isopropenylacetat (200 ml) und p-Toluolsulfonsäure (3 g) wurde auf 1000 C 18 Stunden erhitzt, und dann wurde das Lösungsmittel allmählich während einer Zeitdauer von 6 Stunden auf ungefähr 75 ml herunterdestilliert. Das Gemisch wurde dann gekühlt, mit Äther (2:1) verdünnt und mit gesättigter Kaliumbicarbonatlösung und Salzlösung gewaschen, die organische Schicht getrocknet und eingedampft.
Der sich ergebende braune Feststoff wurde aus Äthanol (200 ml) auskristallisiert und ergab das 1 7-Acetoxy- 13 ss-äthyl-3 - methoxy-gona-1,3,5(10),16-tetraen (9,8 g); Schmelzpunkt 128 bis 1300 C und hatte, umkristallisiert aus Äthanol, den Schmelzpunkt 133,5 bis 134,50 C. (Gefunden: 77,65 % C; 8,45 % H; die Bruttoformel C2sH2gO3 erfordert 77,61 % C; 8,29 % H.) 17-Acetoxy-13ss-äthyl-3-methoxy-gona-1,3,5(10),16- tetraen (9,28 g) in trockenem Äther (400 ml) wurde mit einer 1n-Lösung von Monoperphthalsäure (100 ml) in Äther behandelt, und das Gemisch 5 Tage bei Zimmertemperatur stehengelassen.
Die Lösung wurde dann gewaschen, getrocknet, eingedampft und ergab ein gelbes Harz, welches mit Benzol (400 ml) behandelt und über eine Kolonne mit Florisil (80 g) geleitet wurde..
Man erhielt 17ss-Acetoxy-16α,17α-epoxy-13ss-äthyl-3- methoxy-gona-1,3,5(10)-trien (6,5 g).

Example 1 a) 3, l7-Macetoxy-l 3ss-ethyl-gona-l, 3,5 (10), 16-tetraene 3 -acetoxy- 13ss-ethyl-gona- 1,3,5 (10) -triene- 17-one (3.31 g) was dissolved in isopropenyl acetate (50 ml), treated with p-toluenesulfonic acid (500 mg) and heated in a boiling water bath overnight. The solvent was partially distilled off, the mixture was diluted with ether and washed, dried and evaporated to a brown oil (3.54 g). The first eluate was crystallized from ethanol to form the epoxide (344.2 mg), melting point 170 to 171.50 C, and was recrystallized from ethanol, melting point 172 to 173.50 C. The aqueous layer was extracted with ethyl acetate (2 x 150 ml) and the extracts combined and added to the originally obtained organic layer. This solution was washed with water, dried and evaporated. The remaining white solid (409 mg) was recrystallized from ethanol (10 ml) and gave 13ss-ethyl-gona-1,3,5 (10) -trien-3,16-diol-17-one (258.7 mg) ; Melting point 198 to 200.50 C (the crystals became opaque at approximately 1400 C) and had a melting point of 200 to 202.50 C when recrystallized from ethanol.

Example 2 3,16a-Diacetoxy-13ss-ethyl-gona-1,3,5 (10) -trien-17-one a) 13ss-Ethyl-gona-1,3,5 (10) -triene-3,16α ; -diol-17-one (47.1 mg) was dissolved in pyridine (1 ml) and acetic anhydride (0.6 ml) and allowed to stand at room temperature for 72 hours. The solvents were removed in vacuo and the product, crystallized from ethanol (2 ml), gave 3.16 α-diacetoxy-1 3.1 6ct-diacetoxy-13ss-ethyl-gona-1,3,5 (10) -triene -17-one (54.2 mg); Melting point 172 to 1730 C.
b) 3-Acetoxy-13ss-ethyl-gona-1,3,5 (10) -trien-16a-ol-17-one (271.5 mg) was dissolved in pyridine (2 ml) and acetic anhydride (1.2 ml ) and the solution was left to stand at room temperature for 72 hours. The solvents were removed in vacuo, the remaining solid was recrystallized from ethanol (10 ml) and gave 3,16-diacetoxy- - 13ss - ethyl-gona-1,3,5 (10) - trien-17-one (238, 5 mg);
Melting point 173 to 1740 C, and recrystallized from ethanol, needles, with a melting point of 173 to 173.50 C. (Found: 71.91 XC; 7.20% H; the gross formula C2sHo805 requires 71.85% C; 7, 34% H.)
Example 3 13ss-Sithyl-3-methoxy-gona- 1,3,5 (10) -triene-
16a-ol-17-one 1 7ss - acetoxy -1 6a, 1 7a - epoxy- 1 3ss-ethyl-3-methoxy- gona-1,3,5 (10) -triene (3.6 g) in methanol ( 500 ml) was treated with 6N sulfuric acid (75 cm3) and the mixture was stirred at 250 ° C. for 4 days. The solution was reduced to a small volume in vacuo, water added and the mixture cooled in an ice bath.
The precipitate was filtered, washed with water and dried to obtain the above-identified compound (2.98 g); Melting point 130 to 1380 C, homogeneous by thin layer chromatography; Infrared absorption maxima at 2.95, 3.45, 3.53, 5.76, 6.21, 6.37, 6.67, and the like.
The starting material was prepared as follows: 13ss-ethyl-3-methoxy-gona-1,3,5 (10) -trien-17-one (16.8 g) in isopropenyl acetate (200 ml) and p-toluenesulfonic acid (3 g ) was heated to 1000 C for 18 hours and then the solvent was gradually distilled down to approximately 75 ml over a period of 6 hours. The mixture was then cooled, diluted with ether (2: 1) and washed with saturated potassium bicarbonate solution and brine, the organic layer dried and evaporated.
The resulting brown solid was crystallized from ethanol (200 ml) to give the 17-acetoxy-13 ss-ethyl-3-methoxy-gona-1,3,5 (10), 16-tetraene (9.8 g) ; Melting point 128 to 1300 C and, recrystallized from ethanol, had a melting point of 133.5 to 134.50 C. (Found: 77.65% C; 8.45% H; the gross formula C2sH2gO3 requires 77.61% C; 8, 29% H.) 17-Acetoxy-13ss-ethyl-3-methoxy-gona-1,3,5 (10), 16-tetraene (9.28 g) in dry ether (400 ml) was with a 1N solution of monoperphthalic acid (100 ml) in ether and the mixture left to stand for 5 days at room temperature.
The solution was then washed, dried, evaporated to give a yellow resin which was treated with benzene (400 ml) and passed through a column of Florisil (80 g).
There was obtained 17ss-acetoxy-16α, 17α-epoxy-13ss-ethyl-3-methoxy-gona-1,3,5 (10) -triene (6.5 g).
