Verfahren zur Herstellung von Tetracosapeptid-Zink-Pyrophosphat-Komplexen mit adrenocorticotroper Wirkung
Im Hauptpatent Nr. 471 819 ist ein Verfahren zur Herstellung neuer Tetracosapeptide mit adrenocorticotroper Wirkung, nämlich des L-Seryl-L-tyrosyl-L-seryl -L-methionyl-L-glutamyl-L-histidyl-L-phenylalanyl-L-ar ginyl-L-tryptophyl-glycyl-L-lysyl-L-prolyl-L-valyl-glycyl -L-lysyl-L-lysyl-L-arginyl-L-arginyl-L-prolyl-L-valyl-L- -lysyl-L-valyl-L-tyrosyl-L-prolins (#1-24-Corticotropin) u. der entsprechenden Verbindung, die statt des Glutamylrestes den Rest des Glutamins aufweist, und ihrer Salze und Schwermetall-, insbesondere Zinkkomplexe, beschrieben.
Im USA-Patent Nr. 3 228 839 wurden Komplexe der obigen Tetracosapeptide mit schwerlöslichen Zinkverbin dungen, wie Zinkphosphat. Zinkhydroxyd und Zinkcarbonat, unter Schutz gestellt. Die genannten Komplexe, die pro Gramme ! Tetracosapeptid wenigstens 20, vorzugsweise 400-1000 Grammäquivalente Zink aufweisen, zeichnen sich durch gute Depotwirkung aus.
Es wurde nun gefunden, dass die klinische Anwen dung der Tetracosapeptidkomplexe wesentlich erleichtert und verbessert werden kann, wenn man die Komplexe nicht mit den genannten Zinkverbindungen, sondern mit Zinkpyrophosphat bi) det. Überraschenderweise erhäft man in diesem Falle den in Wasser schwer löslichen Te tracosapeptid-Zinksalzkomplex in Form einer Fällung von definierter, gleichmässiger Teilchengrösse. der die Herstellung stabiler Suspensionen gestattet. Bei Sterili sierung der Suspensionen (20 Min. 20 C) verändert sich die Teitchengrösse nicht.
. le nach den Fallungsbedingungen (Zinksalzkonzen- tration, Temperatur. pH) kann man die Teilchengrösse variieren, beispielsweise zwischen 1 und SOu Teilchendurchmesser. Vorzugsweise stellt man Fallungen mit einem Teilchendurchmesser zwischen 2 und 20pue her.
Nach dem erfindungsgemässen Verfahren wird der 1'etracosapeptid-Zinkpyrophosphatkomplex in einer wässrigen Lösung vom pH 5 bis 6,5, die mindestens 20 Grammäquivalente eines therapeutisch verwendbaren löslichen Zinksalzes pro Mol Tetracosapeptid enthält, mit einer therapeutisch verwendbaren Pyrophosphationen-haltigen wÏssrigen L¯sung. vorzugsweise mit einem geringen überschuss über die stöchiometrische Menge Nat@umpyrophosphat. geraut. Der Uberschuss betragt etwa 1-2%. Als lösliche Zinksalze sind z. B. zu nennen : Zinkchlorid. Zinksulfat und Zinkacetat. Wählt man als lösliches Zinksalz das Acetat, so wirkt das bei der Fäl- lung entstehende Natriumacetat als Puffer, der das pH der Suspension auf ca. 6,0 hält.
Dieses pH bleibt auch bei Sterilisation erhalten. Falls die das iöstiche Zinksalz enthaltende Lösung ein pH über 6.5 aufweist, wird mittels verdünnter Mineralsäure, vorzugsweise SalzsÏure. das gewünschte pH eingestellt.
Die Zinksalzkonzentration wird so gewählt, dass die Suspension pro Moi mindestens 20. vorzugsweise 3002000 Grammäquivalente Zink enthält. Die Tetracosapep- tidkonzentration wird entsprechend der gewünschten Do sisstärke gewahlt ; I mg Tetracosapeptid, intramuskulär injiziert, entspricht 100 I. E.
Die Fällung wird bei Temperaturen von 0-50=C, vorzugsweise bei Zimmertemperatur, unter Rühren durchgeführt.
Die neuen Komplexe können in Form pharmazeutischer Präparate, vorzugsweise als Suspensionen für die intramuskuläre oder subcutane Injektion, verwendet werden. Die Suspensionen können in Ampullen oder Vials aufbewahrt werden. Man kann sie aber auch jeweils vor Gebrauch aus lyophilisierte Komplexen (Trockenampul- len) herstellen. Falls die Komplexe lyophilisiert werden, setzt man der zu lyophilisierenden Suspension wie üblich ein Trägermaterial, z. B. Mannit. zu.
Die Suspensionen kiinnen in üblicher Weise sterili- siert werden. Man kann auch die für die Fällung beniitz- ten Lösungen keimfrei filtrieren und clie Suspension unter sterilen Bedingungen in Ampullen oder Vials abfüllen. Die Suspensionen können auch Konservierungs- mittel, Stabilisierungs-, Netz-oder Emufgiermittet oder andere therapeutisch wertvolle Stoffe enthalten.
Das in den Komplexen enthaltene Tetracosapeptid kann nach dem im Hauptpatent Nr. 471 819 oder in den französischen Patenten Nr. 85 518 oder Nr. 84475 beschriebenen Verfahren hergestellt werden.
Die Erfindung wird in den nachfolgenden Beispielen beschrieben.
Beispiel l 1, 0 ml einer wässrigen Lösung, die 0,001 g #1-24- Corticotropin, 0, 02 g Zinkacetat (Zn (CH3COO) 2, 2H2O), 0, 0015 g Natriumchlorid, 0,02 g Benzylalkohol und 0,005 ml 1-n. Salzsäure enthält, wird unter Rühren bei Zimmertemperatur mit 1, 0 ml einer 2, 16%igen wÏssrigen Lösung von Natriumpyrophospbat (Na4P2O7, 10H2O) versetzt.
Die bei dieser Umsetzung entstehende Suspension von Zinkpyrophosphat ist isotonisch und hat einen pH-Wert von 6,0. Die sterilisierte Suspension hat ebenfalls ein pH von 6,0. Die Teilchen haben grösstenteils einen Durchmesser von 3-4 u, Streubreite : 2-6 u. Die Suspension enthält pro ml 0, 5 mg (50 I. E.)'-24-Corticotropin, 6, 94 mg Zinkpyrophosphat, 7,47 mg Natriumacetat, 0,75 mg Na triumchlorid und 10, 0 mg Benzylalkohol.
Beispiel 2 1. 0 ml einer wässerigen Lösung, welche 0,001 PI-24¯ Corticotropin, 0,05 g Zinksulfat (ZnSO4, 7 H. O) und 0, 02 g Benzylalkohol enthält, wird unter Rühren mit 1, 0 ml einer 3,95% igen Lösung von Natriumpyrophosphat (Na4P2O7, 10H2O) versetzt.
Die bei dieser Umsetzung entstehende Zinkpyrophosphatsuspension hat einen pH-Wert von 5, 5. Die sterilisierte Suspension hat ein pH von 6, 0. Die Teilchen haben im Mittel einen Durchmesser von 6-7 p. I ml Suspension enthält 50 I. E. I-'4-Corticotropin.

In U.S. Patent No. 3,228,839, complexes of the above tetracosapeptides with sparingly soluble zinc compounds such as zinc phosphate. Zinc hydroxide and zinc carbonate, placed under protection. The complexes mentioned that are per gram! Tetracosapeptide have at least 20, preferably 400-1000 gram equivalents of zinc, are characterized by a good depot effect.
It has now been found that the clinical application of the tetracosapeptide complexes can be significantly facilitated and improved if the complexes are not bi) det with the zinc compounds mentioned, but with zinc pyrophosphate. Surprisingly, in this case, the water-sparingly soluble Te tracosapeptide zinc salt complex is obtained in the form of a precipitate of a defined, uniform particle size. which allows the production of stable suspensions. When the suspensions are sterilized (20 min. 20 C), the particle size does not change.
. Depending on the precipitation conditions (zinc salt concentration, temperature, pH), the particle size can be varied, for example between 1 and 50 μm particle diameter. Preference is given to producing precipitates with a particle diameter between 2 and 20 pue.
According to the inventive method, the 1'etracosapeptide zinc pyrophosphate complex is in an aqueous solution of pH 5 to 6.5, which contains at least 20 gram equivalents of a therapeutically usable soluble zinc salt per mole of tetracosapeptide, with a therapeutically usable pyrophosphate ion-containing aqueous solution. preferably with a slight excess over the stoichiometric amount of sodium pyrophosphate. roughened. The excess is about 1-2%. As soluble zinc salts, for. B. to be mentioned: zinc chloride. Zinc sulfate and zinc acetate. If acetate is chosen as the soluble zinc salt, the sodium acetate formed during the precipitation acts as a buffer, which keeps the pH of the suspension at about 6.0.
This pH is retained even after sterilization. If the solution containing the iöstiche zinc salt has a pH above 6.5, dilute mineral acid, preferably hydrochloric acid, is used. set the desired pH.
The zinc salt concentration is selected so that the suspension contains at least 20, preferably 3002000 gram equivalents of zinc per mol. The tetracosapeptide concentration is chosen according to the desired dose strength; I mg tetracosapeptide, injected intramuscularly, corresponds to 100 I.U.
The precipitation is carried out at temperatures of 0-50 ° C., preferably at room temperature, with stirring.
The new complexes can be used in the form of pharmaceutical preparations, preferably as suspensions for intramuscular or subcutaneous injection. The suspensions can be stored in ampoules or vials. However, they can also be prepared from lyophilized complexes (dry ampoules) before use. If the complexes are lyophilized, the suspension to be lyophilized is, as usual, a carrier material, e.g. B. mannitol. to.
The suspensions can be sterilized in the usual way. The solutions used for the precipitation can also be filtered sterile and the suspension can be filled into ampoules or vials under sterile conditions. The suspensions can also contain preservatives, stabilizers, wetting agents or emulsifying agents or other therapeutically valuable substances.
The tetracosapeptide contained in the complexes can be prepared according to the process described in main patent no. 471,819 or in French patents no. 85,518 or no.
The invention is described in the following examples.
Example l 1, 0 ml of an aqueous solution containing 0.001 g # 1-24 corticotropin, 0.02 g zinc acetate (Zn (CH3COO) 2, 2H2O), 0.0015 g sodium chloride, 0.02 g benzyl alcohol and 0.005 ml 1 -n. Contains hydrochloric acid, 1.0 ml of a 2.16% aqueous solution of sodium pyrophosphate (Na4P2O7, 10H2O) is added while stirring at room temperature.
The suspension of zinc pyrophosphate resulting from this reaction is isotonic and has a pH of 6.0. The sterilized suspension also has a pH of 6.0. The majority of the particles have a diameter of 3-4 microns, spread: 2-6 microns. The suspension contains 0.5 mg (50 I.E.) '- 24-corticotropin, 6.44 mg zinc pyrophosphate, 7.47 mg sodium acetate, 0.75 mg sodium chloride and 10.0 mg benzyl alcohol per ml.
Example 2 1. 0 ml of an aqueous solution which contains 0.001 PI-24¯ corticotropin, 0.05 g zinc sulfate (ZnSO4, 7 H. O) and 0.02 g benzyl alcohol is stirred with 1.0 ml of a 3, 95% solution of sodium pyrophosphate (Na4P2O7, 10H2O) added.
The zinc pyrophosphate suspension formed during this reaction has a pH of 5.5. The sterilized suspension has a pH of 6.0. The particles have an average diameter of 6-7 μm. I ml of suspension contains 50 I.E. 14-corticotropin.
