Process for the preparation of 7-alkyl-19-nor-steroids The invention relates to a process for the preparation of new 19-nor-steroids of the general formula which are unsubstituted in the 3-position
EMI0001.0005
wherein Y is an alkyl radical, R, an OH or O-acyl group, R. is a hydrogen atom or a saturated or unsaturated hydrocarbon radical having 1 to 4 carbon atoms.
The compounds prepared according to the invention come because of their gestagenic, anabolic. androgenic, estrogenic, gonad-inhibiting properties and the o, ula- tion-inhibiting properties are of particular importance. In particular, the compounds with an unsaturated alkyl radical in the 17-position exert a strong ovulation-inhibiting effect.
The new compounds are prepared according to the invention by using a compound of the formula
EMI0001.0027
in which X represents a halogen atom and Y, R, and R, have the above meaning and the 3-halogen substituent is split off therefrom by a reductive route.
If necessary, the obtained 17-etherified or esterified hydroxy group can be saponified to give the free 17-hydroxy compound and oxidized to the corresponding 17-keto compound.
alkylate by an addition reaction at the 17-keto group with a hydrocarbon metal compound, the corresponding 17- hydroxy-17x-alkyl compound being formed and / or the 17-hydroxyl group esterifying to the corresponding 17-ester.
The starting materials to be used in the process according to the invention are e.g. in i <steroids. Vol. 1, page 317 (1963) or can be prepared by esterification or etherification of the corresponding 17-HyclrotyverbindtinQen.
As examples of usable protecting groups for the 17-hydroxy group are to be mentioned: ether groups, e.g. the t-tityl ether group and the tetrahydropyranyl ether group or ester groups, e.g. the carbon acid ester group (carbonates), the trimethylacetyl or the benzoyl ester group.
The 3-halogen group is split off by reduction. This reduction can be achieved by treatment of the 3-halogen steroid with an alkali metal in the presence of a primary amine or liquid ammonia and also by reduction with an alkali metal and an alcohol, e.g. Natriun1 '!.:
@thanol or with Raney nickel in the presence of an alcohol, e.g. Ethkinol. An alkali metal in liquid ammonia is preferably used for this reduction.
The - \, - 7x-alkyl-17 "-hydroxy-19-androstene compound obtained by the process according to the invention can then be used in a manner known per se by oxidation and, optionally, by a subsequent addition reaction with a hydrocarbon metal compound can be converted into the corresponding 17-keto or 17-hydroxy- 17u-alkyl steroids.
The 17-hydroxyl group can e.g. According to the method of Oppena uer or oxidized with chromium trioxide s \, enl s.
