Verfahren zur Herstellung von neuen Piperidinderivaten
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Piperidinderivate mit wertvollen pharmakologischen Eigenschaften.
Es wurde überraschenderweise gefunden, dass Piperidinderivate der Formel 1:

in welcher R1 eine Alkylgruppe mit 7 bis 9 Kohlenstoffatomen, eine Phenylalkylgruppe mit höchstens 4 Kohlenstoffatomen im Alkylrest, eine 2-(N-Alkanoyl- anilino)-äthylgruppe mit höchstens 4 Kohlenstoffatomen im Alkanoylrest, den 2-Anilinoäthyl-, 2-(N-Allyl-anilino)-äthyl-, 2-Phenoxyäthyl-, 2-Benzoyläthyl- oder Cinnamylrest, und Ro eine niedere Alkylgruppe bedeutet, und ihre Additionssalze mit anorganischen und organischen Säuren wertvolle pharmakologische Eigenschaften, insbesondere analgetische und antitussive Wirksamkeit bei günstigem therapeutischem Index besitzen.
Die analgetische Wirksamkeit ist besonders ausgeprägt bei den Verbindungen, die in R1 einen Phenylkern enthalten. Antitussive Wirksamkeit liegt bei allen Verbindungstypen vor, von besonderem Interesse ist sie jedoch bei Verbindungen mit einer definitions- gemässen Alkylgruppe als Rr. Die Verbindungen der Formel I und ihre Säureadditionssalze eignen sich deshalb als Wirkstoffe für pharmazeutische Präparate zur Linderung und Behebung von Schmerzen verschiedener Genese oder des Hustenreizes. Ihre Anwendung kann oral, rektal oder parenteral erfolgen.
Die neuen Piperidinderivate der Formel I und ihre Säureadditionssalze lassen sich herstellen, indem man eine Verbindung der Formel II:

in welcher R2 die unter Formel I angegebene Bedeutung hat, mit einem reaktionsfähigen Ester einer Verbindung der Formel III: R1-OH (III) in welcher R1 die unter Formel I angegebene Bedeutung hat, umsetzt. Die Umsetzung erfolgt bei Raumtemperatur oder mässig erhöhter Temperatur in einem geeigneten organischen Lösungsmittel, wie z. B. Äthanol, Aceton, Äthylacetat oder Dimethylformamid. Gegebenenfalls wird die Umsetzung durch Zusatz säurebindender Mittel, wie z. B. Kaliumcarbonat, und/oder von Kaliumjodid beschleunigt. Als reaktionsfähige Ester eignen sich insbesondere Halogenwasserstoffsäureester, wie Bromide, Chloride und Jodide, weiter Arylsulfonsäureester, z.
B. p-Toluolsulfonsäureester.
Die nach dem erfindungsgemässen Verfahren erhaltenen Piperidinderivate der Formel I werden anschliessend gewünschtenfalls in üblicher Weise in ihre Additionssalze mit anorganischen und organischen Säuren übergeführt. Beispielsweise versetzt man eine Lösung des Piperidinderivates der Formel I in einem organischen Lösungsmittel, wie Diäthyläther, Methanol oder Äthanol, mit der als Salzkomponente gewünschten Säure oder einer Lösung derselben und trennt das unmittelbar oder nach Zufügen einer zweiten organischen Flüssigkeit, wie z. B. Diäthyläther zu Methanol, ausgefallene Salz ab.
Zur Verwendung als Wirkstoff für Arzneimittel können anstelle freier Basen pharmazeutisch annehmbare Säureadditionssalze eingesetzt werden, d. h. Salze mit solchen Säuren, deren Anionen bei den in Frage kommenden Dosierungen entweder keine oder erwünschte eigene pharmakologische Wirkung zeigen.
Ferner ist es von Vorteil, wenn die als Wirkstoffe zu verwendenden Salze gut kristallisierbar und nicht oder wenig hygroskopisch sind. Zur Salzbildung mit Piperidin- derivaten der FormelI kann z. B. Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Methansulfonsäure, Äthansulfonsäure, fl - Hydroxyäthansulfonsäure, Essigsäure, Weinsäure, Äpfelsäure, Citronensäure, Milchsäure, Bernsteinsäure, Fumarsäure, Maleinsäure, Benzoesäure, Salicylsäure, Phenylessigsäure, Mandelsäure, Embonsäure oder 1 ,5-Naphthalindisulfonsäure verwendet werden.
Die neuen Piperidinderivate der Formel I und ihre Salze werden peroral, rektal oder parenteral verabreicht.
Die täglichen Dosen von freien Basen oder pharmazeutisch annehmbaren Salzen derselben bewegen sich zwischen 1 und 100 mg für erwachsene Patienten. Geeignete Doseneinheitsformen wie Dragees, Kapseln, Tabletten, Suppositorien oder Ampullen enthalten vorzugsweise 0,5-50 mg eines Piperidinderivates der Formel I oder eines pharmazeutisch annehmbaren Salzes desselben Besonders für die Behandlung des Hustens kommen ferner auch Lutschtabletten sowie nichteinzeldosierte orale Applikationsformen, wie z. B. mit den üblichen Hilfsstoffen bereitete Hustensirups und Hustentropfen, in Betracht.
Das nachfolgende Beispiel erläutert die Herstellung der neuen Verbindungen der Formel I. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel
1,1 g 4-Allyl-isonipecotinsäure-äthylester werden mit 2,2 g 2-Phenyläthylbromid, 5 g Natriumcarbonat und 0,1 g Natriumjodid in 40 ml Aceton 18 Stunden unter Rückfluss gekocht. Hierauf wird das Reaktionsgemisch filtriert, das Filtergut mit Aceton nachgewaschen und das Filtrat eingedampft und im Hochvakuum destilliert.
Der 1-(2-Phenyläthyl)-4-allyl - isonipecotinsäure - äthyl- ester siedet unter 0,01 Torr bei 125-1300. Das daraus hergestellte Fumarat schmilzt bei 1380.

Der als Ausgangsstoff benötigte 4-Allyl-isonipecotinsäure-äthylester wird wie folgt hergestellt: a) In einem 750-ml-Vierhalskolben werden unter Stickstoff zu 22,8 g Brombenzol in 180 ml Äther unter Rühren 2,03 g in kleine Stücke zerschnittener Lithiumdraht zugegeben, wobei der Äther zu sieden beginnt. Nachdem die Reaktion nachgelassen hat, wird das Gemisch noch 21/2 Stunden unter Rückfluss gekocht. Zur erhaltenen Lösung von Phenyllithium werden 35,4 g Triphenylmethan in 150 ml abs. 1,2-Dimethoxyäthan auf einmal zugegeben, wobei sich die Lösung infolge Bildung des Triphenylmethyllithiums tiefrot färbt und leicht siedet.
Nach 20 Minuten Rühren bei Raumtemperatur werden 42,3 g 1 - Benzyloxy- carbonyl-isonipecotinsäure-äthylester (hergestellt durch Umsetzung von Isonipecotinsäure-äthylester mit Chlorameisensäure-benzylester in Gegenwart von In Natriumbicarbonatlösung) in 50 ml abs. Äther bei 280 zugegeben. Unter leichter Temperaturerhöhung entfärbt sich die tiefrote Lösung. Sie wird 10 Minuten bei Raumtemperatur gerührt und anschliessend mit 18,0 g Allylbromid in 40 ml abs. Äther auf einmal versetzt.
Das Gemisch wird 21/2 Stunden bei Raumtemperatur gerührt, wobei es sich gelblich färbt und Lithiumbromid ausfällt. Anschliessend wird das Reaktionsgemisch mit 40 ml Wasser zersetzt und im Rotationsverdampfer fast bis zur Trockne eingedampft. Der Rückstand wird in 50 ml Äther aufgenommen und die erhaltene Ather- lösung dreimal mit 2n Salzsäure ausgezogen. Hierauf wird die Ätherlösung getrocknet und eingedampft und der Rückstand über Nacht stehengelassen, wobei das Triphenylmethan auskristallisiert. Das gesamte Gemisch wird in kaltem Methanol suspendiert, das Triphenylmethan abgenutscht und das Filtrat eingedampft.
Der Rückstand wird im Hochvakuum destilliert, wobei der I-Benzyloxycarbonyl -4-allyl- isonipecotinsäure- äthylester unter 0,07 Torr bei 170-192 übergeht. b) 8,0 g 1-Benzyloxycarbonyl-4-allyl-isonipecotinsäure-äthylester werden mit 40 ml einer gesättigten Lösung von Bromwasserstoff in Eisessig und 9 ml abs.
Ather in einem 100-ml-Rundkolben mittels Magnetrührer 2 Stunden gerührt. Die am Anfang starke Kohlendioxidentwicklung lässt allmählich nach. Dann wird die Lösung im Rotationsverdampfer eingedampft und der Rückstand in 6n Salzsäure aufgenommen. Die salzsaure Lösung wird mit Äther gewaschen, dann unter guter Kühlung mit konzentrierter Ammoniaklösung alkalisch gestellt und mit Chloroform ausgezogen. Die Chloroformlösung wird getrocknet, eingedampft und der zurückbleibende 4-Allyl-isonipecotinsäure-äthylester sofort weiter umgesetzt.
Analog a und b lassen sich auch andere niedere Alkylester der 4-Allyl-isonipecotinsäure herstellen.

The new piperidine derivatives of the formula I and their salts are administered orally, rectally or parenterally.
The daily doses of free bases or pharmaceutically acceptable salts thereof range between 1 and 100 mg for adult patients. Suitable dosage unit forms such as dragees, capsules, tablets, suppositories or ampoules preferably contain 0.5-50 mg of a piperidine derivative of the formula I or a pharmaceutically acceptable salt thereof. Lozenges as well as non-individually dosed oral administration forms, such as, for. B. cough syrups and cough drops prepared with the usual excipients.
The following example explains the preparation of the new compounds of the formula I. The temperatures are given in degrees Celsius.

The ethyl 4-allyl-isonipecotinate required as starting material is prepared as follows: a) In a 750 ml four-necked flask, 2.03 g of lithium wire cut into small pieces are added under nitrogen to 22.8 g of bromobenzene in 180 ml of ether with stirring whereby the ether begins to boil. The reaction mixture is then decomposed with 40 ml of water and evaporated almost to dryness in a rotary evaporator. The residue is taken up in 50 ml of ether and the ether solution obtained is extracted three times with 2N hydrochloric acid. The ethereal solution is then dried and evaporated and the residue is left to stand overnight, the triphenylmethane crystallizing out. The entire mixture is suspended in cold methanol, the triphenylmethane is suction filtered and the filtrate is evaporated.
The residue is distilled in a high vacuum, the I-benzyloxycarbonyl -4-allyl-isonipecotinic acid ethyl ester passing over under 0.07 torr at 170-192. b) 8.0 g of 1-benzyloxycarbonyl-4-allyl-isonipecotinic acid ethyl ester are mixed with 40 ml of a saturated solution of hydrogen bromide in glacial acetic acid and 9 ml of abs.
The ether was stirred in a 100 ml round bottom flask using a magnetic stirrer for 2 hours. The initially strong carbon dioxide development gradually subsides. The solution is then evaporated in a rotary evaporator and the residue is taken up in 6N hydrochloric acid. The hydrochloric acid solution is washed with ether, then made alkaline with concentrated ammonia solution with good cooling and extracted with chloroform. The chloroform solution is dried, evaporated and the remaining 4-allyl-isonipecotinic acid-ethyl ester immediately reacted further.
Other lower alkyl esters of 4-allyl-isonipecotic acid can also be prepared analogously to a and b.
