Neues Verfahren zur Herstellung von 7-Aminocephalosporansäure Gegenstand des Hauptpatentes Nr. 433 316 ist ein Verfahren zur Herstellung von 7-Aminocephalosporan- verbindungen, das dadurch gekennzeichnet ist, dass man eine Verbindung der Formel II

worin Z eine Aminoschutzgnuppe und COOR eine veresterte Carboxylgruppe bedeutet,
mit halogenisieren- den Mitteln behandelt, das gebildete Imidhalogenid in einen Iminoäther überführt und, diesen verseift.
Aus dem erhaltenen 7-Aminocephalosporansäureester kann man die veresterte Carboxylgruppe freisetzen, um die freie 7 Aminocephalosporansl.äure zu erhalten. Nach der vorliegenden Erfindung wird nun N,N-Phthaloyl-ce- phalosporin C-dibenzhydrylester mit Phosphorpenta- chlorid umgesetzt, das Imidchlorid, z.
B. mit einem Niederalkanol, in einen Niederalkyliminoäther über geführt, dieser mit Wasser in saurem Medium ver seift und der erhaltene 7-Aminocephalosporansäure- benzhydrylester mittels Trifluoressigsäure in Gegenwart von Anisol gespalten.
Dieses Verfahren ist dadurch gekennzeichnet, dass man nach der Verseifung des 7-Aminocephalosporan- säure-benzhydrylesters die Trifluoressigsäure in einem polaren Lösungsmittel mit einem tertiären Amin in das Trifluoracetat überführt und die 7-Amino-cephalospo- ransäure aus der Lösung kristallisiert.
Als Amin kann beispielsweise ein Triniederalkyl- amin, vor allem Triäbhylamin oder Tri n-butylamin, verwendet werden. Das mit dem Amin gebildete Tri- fluoracetat soll in dem polaren Lösungsmittel löslich sein; geeignete Lösungsmittel sind z.
B. Dimethylforrn- amid, Acetonitril oder vorzugsweise ein Niederalkanol, vor allem Methanol. Die Kristallisation der 7-Amino- cephalosporansäure erfolgt vorzugsweise aus methano- lischer Lösung.
Die erhaltene kristallisierte 7-Amino- cephalosporansäure wird vorteilhaft nacheinander mit Methanol, Methylenchloridund Äther gewaschen, um sie von den anhaftenden, aus dem Reaktionsmedium stammenden Verunreinigungen (Spaltprodukte, Lö sungsmittel) zu befreien. Man erhält so die 7 Amino- cephalosporansäure ohne Umkristallisation in über 97 ö iger Reinheit.
Die drei Verfahrensstufen zur Gewinnung des 7- Aminocephalosporansäure-benzhydrylesters werden vor teilhaft wie folgt durchgeführt, um die Reaktion zu vereinfachen und die Reaktionszeit zu verkürzen: 1. Bei der Umsetzung des Phthaloyl-cephalosporin C-dibenzhydrylesters mit Phosphorpentachlorid in Gegenwart tertiärer Amine, z. B. Pyridin, in einem inerten Lösungsmittel, z.
B. Methylenchlorid, soll das Molverhältnis des Cephalosporinderivats zum Phos- phorpentachlorid und zum Amin etwa 1 : 3: 12 be tragen. Die Temperatur soll bei etwa -10 C gehalten werden, um die Bildung von Nebenprodukten zu ver- mindern. Die Reaktionszeit beträgt unter diesen Um ständen 30 bis 40 Minuten.
2. Um das Imidchlorid möglichst schnell und quan titativ in den Iminoäther überzuführen, verwendet man einen grossen überschuss an Alkohol, z. B. etwa 100 Mol Methanol pro Mol, und lässt zunächst bei tiefer Temperatur (etwa 30 Minuten bei -10 ), dann. etwa eine Stunde bei Zimmertemperatur reagieren.
3. Die Hydrolyse des Iminoäthers zum 7 Amino- cephalosporansäure-benzhydrylester wird vorteilhaft in Methylenchlorid-Wasser-Gemisch vorgenommen. In diesem Lösungsmittel ist sie in etwa 45 Minuten bei Zimmertemperatur beendet, wenn man als. Verseifungs- mittel verdünnte wässrige Salzsäure verwendet.
Die Verfahrensstufen, 1 bis 3 werden also alle im gleichen Reaktionsgefäss und mit Methylenchlorid als Hauptlösungsmittel vorgenommen. Nach der 3. Ver fahrensstufe wird das Reaktionsgemisch alkalisch ge macht (pH 8) und der 7-Aminocephalosporansäure- benzhydrylester aus der Methylenchloridschieht als Rohprodukt isoliert. Der rohe Ester wird direkt mit Trifluoressigsäure in Gegenwart von Anisol verseift und die 7-Aminocephalosporansäure wie oben angege ben isoliert.
In den nachfolgenden Beispielen sind die Tempera turen in Celsiusgraden angegeben.
<I>Beispiel 1</I> 26,3 g kristallisierter N-Phthalyl-cephalosporin C- di-benzhydrylester werden in 1 Liter abs. Methylen- chlorid gelöst,
mit 29 ml abs. Pyridin versetzt und auf -10 abgekühlt. Dann gibt man unter Rühren in nerhalb 2 Minuten 187 m1 einer l0proz. Phosphor- pentachloridlösung in abs. Methylenchlorid zu und rührt 1/2 Stunde, wobei die Temperatur die ganze Zeit auf -10 gehalten wird.
Hierauf gibt man zu der Lösung innerhalb 1/2 Minute rasch 200 ml eiskaltes,- abs. Methanol und hält die Temperatur weitere 30 Mnuten auf -10 . Man erwärmt im Wasserbad auf +20 und rührt bei dieser Temperatur 1 Stunde. Die Lösung wird dann unter Rühren rasch (innerhalb 1/2 Minute mit 500 ml einer eiskalten,
wässrigen Lösung von 35 ml 85proz. Orthophosphorsäure versetzt und 11/2 Stunden bei 20 gerührt. Man stellt mit etwa 530 ml 50proz. wässriger Trikaliumphosphatlösung auf pH- 8,0 und trennt die untere Methylenchloridschicht ab.
Die wässrige Oberphase wird mit 500 ml Methylen- chlorid nachextrahiert. Die vereinigten und über Na- triumsulfat getrockneten Methylenehloridextrakte geben beim Eindampfen im Vakuum 34 bis 40 g (je nach Trocknungsgrad) flüssigen Rückstand, der noch etwa 1/2 Stunde bei 0,1 mm Hg und Raumtemperatur ge- trocknet wird.
Dieser Rückstand enthält den 7-Amino- cephalosporansäure-benzhydrylester. Er wird in 15m1 Anisol aufgenommen und unter Kühlen mit 45 ml Tri- fluoressigsäure versetzt. Man beschleunigt das Lösen des Rückstandes durch kräftiges Bewegen und hält wäh rend 15 Minuten die Temperatur auf etwa 20 .
Dann wird die Lösung rasch zu 700 ml gerührtem, kaltem Methanol gegossen und gleichzeitig das pH (pH-Meter) der methanolischen Lösung durch Zugabe von etwa 135 ml Tri-n-butylamin auf den Wert 3,5 gesteilt. An schliessend stellt man das pH noch fein ein, indem man 0,3 ml-Proben mit je 4,5 ml Wasser verdünnt rund nun diese mit dem pH-Meter misst.
Die Lösung wird 2 bis 15 Stunden auf +5 bis +10 gekühlt und dann abfil triert. Der nacheinander mit Methanol, Methylenehlorid und Äther gewaschene Rückstand gibt beim Trocknen im Vakuum 6,68 g (= 815v, d. Th.) 7-Amino-cephalo- sporansäure.
UV-Absorptionsspektrum (in 0, ln Natriumbicarbo- nat): .?",;" 263 mg (a = 8000). [a] D = +118 (c = 1 in 0,5n Natriumbicarbonat). <I>Beispiel 2</I> 26,3 g kristallisierter N-Phthalylrcephalosporin C- di-benzhydrylester werden in 263 ml abs. Methylen- chlorid gelöst,
mit 23 ml abs. Pyridin versetzt und auf -10 abgekühlt. Dann gibt man unter Rühren inner halb 2 Minuten 150 ml einer 10proz. Phosphorpenta- chloridlösung in abs. Methylenchlorid zu und rührt 1/2 Stunde, wobei die Temperatur die ganze Zeit auf -10 gehalten wird:
Hierauf gibt man zu der Lösung inner halb 1/2 Minute rasch 100 ml eiskaltes, abs. Methanol und hält die Temperatur weitere 30 Minuten auf -10, . Man erwärmt im Wasserbad auf +20 und rührt bei dieser Temperatur 1 Stunde. Die Lösung wird dann unter Rühren rasch (innerhalb 1/2 Minute) mit 250 ml einer eiskalten, wässrigen 1n Salzsäure versetzt und 3/4 Stunden bei 20 gerührt.
Man versetzt mit 50 ml 50proz. wässriger Trikalium- phosphatlösung ,und stellt mit etwa 230 ml 2n wässriger Natronlauge auf pH 8,0. Aus dem zur Emulsion nei genden Gemisch trennt man mittels Zentrifuge die Methylenchloridschicht ab und extrahiert die wässrige Phase mit 200 ml Methylenchlorid nach.
Die vereinig ten und über Natriumsulfat getrockneten Methylen- chloridextrakte geben beim Eindampfen im Vakuum 34 bis 38 g (je nach Trocknungsgrad) flüssigen Rück stand, der noch Etwa 1/2 Stunde bei 0,1 mm Hg und Raumtemperatur getrocknet wird.
Der Rückstand ent hält den 7-Amino-cephalosporansäure-benzhydrylester. Er wird wie in Beispiel 1 in :die freie 7 Amino-cephalo- sporansäure übergeführt. Diese hat den gleichen Rein heitsgrad wie in Beispiel 1.
<I>Beispiel 3</I> 1 kg kristalliner N Phthalyl-cephalosporin C-di- benzhydrylester wird in 30 Liter abs. Methylenchlorid gelöst, mit 1,15 kg abs. Pyridin versetzt und auf -16 abgekühlt.
Dann gibt man unter Rühren innert 10 Minuten eine Lösung von 730 g Phosphorpenta- chlorid in 13 Liter Methylenchlorid zu, wobei die Tem peratur auf<B>-13'</B> ansteigt. Man rührt anschliessend 40 Minuten bei -12 . Hierauf gibt man zu der Lösung innerhalb 3 Minuten 7,5 Liter abs. Methanol (etwa -20 kalt), wobei die Temperatur des Reaktionsgemi- sches auf -10 steigt.
Man rührt dann 1/2 Stunde bei -10 , erwärmt mittels Wasserbad auf +20 und rührt dann eine weitere Stunde. Die Lösung wird ,unter Rüh ren mit 25 Liter 1n wässriger Salzsäure (etwa +10 ) versetzt und 3/4 Stunden bei 20 gerührt. Man ver setzt mit 1,
9 Liter 50 % iger wässriger Trikaliumphosphat lösung und stellt mit etwa 20 Liter 2n vwässriger Na tronlauge auf pH $,0.
Die Phasen werden getrennt und die wässrige Phase mit 12 LiterRTI ID="0002.0215" WI="25" HE="4" LX="1751" LY="2105"> Methylenchlorid nachextrahiert. Die vereinigten und über 2,5 kg Na triumsulfat getrockneten Methylenchloridextrakte geben beim Eindampfen im Vakuum 1,33 kg Rückstand.
Dieser wird in 570 ml abs. Anisol aufgenommen, auf -15 abgekühlt und unter Kühlen und Rühren wäh rend 7 Minuten mit 1,66 Liter Trifluoressigsäure ver setzt. Man lässt .die Säure insgesamt 1/2 Stunde einwir ken, wobei eine Temperatur von etwa 25 eingehalten wird.
In den letzten 5 Minuten fällt aus der bräunlichen Lösung fein verteilte 7-Amino-cephalosporansäure aus. Dann wird die Lösung rasch zu 20 Liter gerührtem, -101 kaltem Methanol gegossen unter gleichzeitiger Zugabe von 2,7 Liter Triäthylamin. Die Zugabege- schwind'igkeiten der Trifluoressigsäurelösung und des Triäthylamins sind .dabei so zu bemessen,
.dass das Tri- äthylamin im Methanol in keinem Moment als über schuss vorliegt. Mit weiteren 350 ml Triäthylamin wird hierauf das pH der Mischung auf 3,5 gestellt. Man lässt über Nacht bei 0 bis +5 stehen, filtriert und wäscht den Rückstand durch Suspendieren in folgenden Lösungsmitteln: 2mal in je 2 Liter Methanol, 2mal in je 2 Liter Methylenchlorid und 2mal in je 2 Liter Äther.
Nach Trocknen im Vakuum bei Raumtem peratur erhält man 271 g (= 81,5 % d. Th.) 7-Amino- cephalosporansäure. Reinheit<B>100%</B> nach UV-Absorp- tionsspektrum (in 0,1n Natriumbicarbonat, Am". = 261 mu (E <I>=</I> 8500). Spezifische Drehung: [a] D _ +l14 1 (c = 1, 0,5n Natriumbicarbonat).

where Z is an amino protective group and COOR is an esterified carboxyl group,
treated with halogenating agents, the imide halide formed is converted into an imino ether and saponified.
The esterified carboxyl group can be liberated from the 7-aminocephalosporanic acid ester obtained in order to obtain the free 7-aminocephalosporanic acid. According to the present invention, N, N-phthaloyl-cephalosporin C-dibenzhydryl ester is reacted with phosphorus pentachloride, the imide chloride, e.g.
B. with a lower alkanol, in a lower alkylimino ether passed over, this soaps ver with water in an acidic medium and the 7-aminocephalosporanic acid benzhydryl ester obtained is cleaved by means of trifluoroacetic acid in the presence of anisole.
This process is characterized in that, after the 7-aminocephalosporanic acid benzhydryl ester has been saponified, the trifluoroacetic acid is converted into the trifluoroacetate in a polar solvent with a tertiary amine and the 7-aminocephalosporanic acid is crystallized from the solution.
A tri-lower alkylamine, especially triabhylamine or tri n-butylamine, for example, can be used as the amine. The trifluoroacetate formed with the amine should be soluble in the polar solvent; suitable solvents are e.g.
B. dimethylformamide, acetonitrile or preferably a lower alkanol, especially methanol. The 7-aminocephalosporanic acid is preferably crystallized from methanolic solution.
The crystallized 7-aminocephalosporanic acid obtained is advantageously washed successively with methanol, methylene chloride and ether in order to free it from adhering impurities (cleavage products, solvents) from the reaction medium. The 7 aminocephalosporanic acid is thus obtained without recrystallization in a purity of over 97%.
The three process steps for obtaining the 7-aminocephalosporanic acid benzhydryl ester are carried out before geous as follows to simplify the reaction and to shorten the reaction time: B. pyridine, in an inert solvent, e.g.
B. methylene chloride, the molar ratio of the cephalosporin derivative to the phosphorus pentachloride and to the amine should be about 1: 3: 12. The temperature should be kept at about -10 C in order to reduce the formation of by-products. Under these circumstances, the reaction time is 30 to 40 minutes.
2. In order to convert the imide chloride into the imino ether as quickly and quantitatively as possible, a large excess of alcohol is used, e.g. B. about 100 moles of methanol per mole, and leaves first at low temperature (about 30 minutes at -10), then. react for about an hour at room temperature.
3. The hydrolysis of the imino ether to give 7 aminocephalosporanic acid benzhydryl ester is advantageously carried out in a methylene chloride-water mixture. In this solvent, it is finished in about 45 minutes at room temperature, if one is called. Saponifying agent used dilute aqueous hydrochloric acid.
Process steps 1 to 3 are all carried out in the same reaction vessel and with methylene chloride as the main solvent. After the 3rd process stage, the reaction mixture is made alkaline (pH 8) and the 7-aminocephalosporanic acid benzhydryl ester is isolated from the methylene chloride layer as a crude product. The crude ester is saponified directly with trifluoroacetic acid in the presence of anisole and the 7-aminocephalosporanic acid is isolated as indicated above.
In the following examples, the temperatures are given in degrees Celsius.
<I> Example 1 </I> 26.3 g of crystallized N-phthalyl-cephalosporin C-di-benzhydryl ester are dissolved in 1 liter of abs. Methylene chloride dissolved,
with 29 ml abs. Added pyridine and cooled to -10. Then, while stirring, 187 ml of a 10 per cent are added within 2 minutes. Phosphorus pentachloride solution in abs. Methylene chloride is added and the mixture is stirred for 1/2 hour, the temperature being kept at -10 all the time.
200 ml of ice-cold abs is then quickly added to the solution within 1/2 minute. Methanol and keeps the temperature at -10 for a further 30 minutes. The mixture is warmed to +20 in a water bath and stirred at this temperature for 1 hour. The solution is then rapidly stirred (within 1/2 minute with 500 ml of an ice-cold,
aqueous solution of 35 ml 85% Orthophosphoric acid was added and the mixture was stirred at 20 for 11/2 hours. One sets with about 530 ml 50 per cent. aqueous tripotassium phosphate solution to pH 8.0 and separates the lower methylene chloride layer.
The aqueous upper phase is extracted with 500 ml of methylene chloride. The combined methylene chloride extracts, dried over sodium sulfate, give 34 to 40 g (depending on the degree of dryness) of liquid residue on evaporation in vacuo, which is dried for about 1/2 hour at 0.1 mm Hg and room temperature.
This residue contains the 7-aminocephalosporanic acid benzhydryl ester. It is taken up in 15 ml of anisole and mixed with 45 ml of trifluoroacetic acid while cooling. The dissolution of the residue is accelerated by vigorous agitation and the temperature is kept at about 20 for 15 minutes.
The solution is then quickly poured into 700 ml of stirred, cold methanol and at the same time the pH (pH meter) of the methanolic solution is adjusted to a value of 3.5 by adding about 135 ml of tri-n-butylamine. The pH is then fine-tuned by diluting 0.3 ml samples with 4.5 ml of water each and then measuring them with the pH meter.
The solution is cooled to +5 to +10 for 2 to 15 hours and then filtered off. The residue, washed successively with methanol, methylene chloride and ether, gives 6.68 g (= 815%, d. Th.) 7-Amino-cephalosporanic acid on drying in vacuo.
UV absorption spectrum (in 0.1N sodium bicarbonate):.? ",;" 263 mg (a = 8000). [a] D = +118 (c = 1 in 0.5N sodium bicarbonate). <I> Example 2 </I> 26.3 g of crystallized N-phthalylrcephalosporin C-di-benzhydryl ester are dissolved in 263 ml of abs. Methylene chloride dissolved,
with 23 ml abs. Added pyridine and cooled to -10. Then 150 ml of a 10 per cent are added while stirring within 2 minutes. Phosphorus pentachloride solution in abs. Add methylene chloride and stir for 1/2 hour, keeping the temperature at -10 all the time:
Then add 100 ml of ice-cold abs to the solution within half a minute. Methanol and keeps the temperature at -10 for a further 30 minutes. The mixture is warmed to +20 in a water bath and stirred at this temperature for 1 hour. 250 ml of an ice-cold, aqueous 1N hydrochloric acid are then added to the solution quickly (within 1/2 minute) while stirring, and the mixture is stirred at 20 for 3/4 hours.
50 ml of 50 percent are added. aqueous tripotassium phosphate solution, and adjusts to pH 8.0 with about 230 ml of 2N aqueous sodium hydroxide solution. The methylene chloride layer is separated off from the mixture, which tends to form an emulsion, using a centrifuge and the aqueous phase is extracted with 200 ml of methylene chloride.
The combined methylene chloride extracts, dried over sodium sulfate, give 34 to 38 g (depending on the degree of dryness) of liquid residue on evaporation in vacuo, which is then dried for about 1/2 hour at 0.1 mm Hg and room temperature.
The residue contains the 7-amino-cephalosporanic acid benzhydryl ester. As in Example 1, it is converted into: the free 7-aminocephalosporanic acid. This has the same degree of purity as in Example 1.
<I> Example 3 </I> 1 kg of crystalline N phthalyl-cephalosporin C-dibenzhydryl ester is dissolved in 30 liters of abs. Dissolved methylene chloride, with 1.15 kg abs. Added pyridine and cooled to -16.
A solution of 730 g of phosphorus pentachloride in 13 liters of methylene chloride is then added with stirring over the course of 10 minutes, the temperature rising to -13 '. The mixture is then stirred at -12 for 40 minutes. 7.5 liters of abs are then added to the solution within 3 minutes. Methanol (about -20 cold), the temperature of the reaction mixture rising to -10.
The mixture is then stirred for 1/2 hour at -10, warmed to +20 using a water bath and then stirred for a further hour. The solution is mixed with 25 liters of 1N aqueous hydrochloric acid (about +10) while stirring and stirred at 20 for 3/4 hours. One sets with 1,
9 liters of 50% aqueous tripotassium phosphate solution and adjusts to pH $ 0.0 with about 20 liters of 2N aqueous sodium hydroxide solution.
The phases are separated and the aqueous phase is re-extracted with 12 liters of RTI ID = "0002.0215" WI = "25" HE = "4" LX = "1751" LY = "2105"> methylene chloride. The combined methylene chloride extracts, dried over 2.5 kg of sodium sulfate, give 1.33 kg of residue on evaporation in vacuo.
This is abs in 570 ml. Anisole added, cooled to -15 and while cooling and stirring during 7 minutes with 1.66 liters of trifluoroacetic acid ver sets. The acid is allowed to act for a total of 1/2 hour, a temperature of about 25 being maintained.
In the last 5 minutes, finely divided 7-amino-cephalosporanic acid precipitates out of the brownish solution. The solution is then quickly poured into 20 liters of stirred, -101 cold methanol with the simultaneous addition of 2.7 liters of triethylamine. The rates of addition of the trifluoroacetic acid solution and of the triethylamine are to be measured in such a way that
.that the triethylamine in the methanol is never present as an excess. The pH of the mixture is then adjusted to 3.5 with a further 350 ml of triethylamine. It is left to stand overnight at 0 to +5, filtered and the residue is washed by suspending it in the following solvents: 2 times in 2 liters of methanol each, 2 times in 2 liters of methylene chloride each time and 2 times in 2 liters of ether each time.
After drying in vacuo at room temperature, 271 g (= 81.5% of theory) of 7-aminocephalosporanic acid are obtained. Purity <B> 100% </B> according to UV absorption spectrum (in 0.1n sodium bicarbonate, Am ". = 261 mu (E <I> = </I> 8500). Specific rotation: [a] D _ + l14 1 (c = 1, 0.5N sodium bicarbonate).
