Verfahren zur Herstellung des neuen 1-Methyl-N-carbobenzoxy-dihydro-lysergamins Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung des neuen, in der Thera pie nützlichen 1-Methyl-N-carbobenzoxydihydrolyserg- amins und dessen Salze.
Im Hauptpatent sind neue 6-Methyl- und 1,6-Dimethyl-ergolinderivate, und zwar Dihydrolyserg- amin, 1-Methyl-dihydrolysergamin und deren in der Therapie nützliche Acylderivate beschrieben und ein Verfahren zur Herstellung der letzteren beansprucht.
Es wurde jetzt gefunden, dass ein neues 1,6-Dimethyl- ergolinderivat und zwar 1-Methyl-N-carbobenzoxy-di- hydro-lysergamin der Formel:

mit mit und dessen nicht toxische, pharmazeutisch verträgliche Salze, eine hohe biologische Wirkung, eine lange Wir kungsdauer und eine niedrige Giftigkeit aufweisen, und sie daher in der Therapie als Mittel gegen Allergie und Entzündungen besonders zu empfehlen sind.
Die erfindungsgemäss hergestellten Produkte stellen einen bemerkenswerten Fortschritt im Vergleich sowohl zu den im Hauptpatent beschriebenen Produk ten wie auch den in der Therapie für die gleichen Zwecke verwendeten Produkte dar. Das erfindungsgemässe Verfahren zur Herstellung des Lysergamins der genannten Formel ist nun dadurch gekennzeichnet, dass. 1-Methyl-dihydro-lyser- einem Reduktionsmittel reduziert wird, wel- gamid ches fähig ist, die Amidgruppe zu einer Amingruppe zu reduzieren, in Gegenwart eines Äthers, der gegen Reduktionsmittel beständig ist, als Lösungsmittel, und dass das so erhaltene 1-Methyl-dihydro-lysergamin mit Carbobenzoxychlorid umgesetzt wird.
Als Lösungsmittel der Äthergruppe eignen sich z. B. Tetrahydrofuran oder Äthylenglykoldimethyläther und als tert. Amin, in dessen Gegenwart die Acylierung in der Regel durchgeführt wird, Pyridin, Diäthylamilin oder Trimethylamin.
Es wird vorgezogen, die Kondensation zwischen -15 und +5 C durchzuführen und die Reaktion bei Raumtemperatur zu beenden. Dann wird zweckmässig wie folgt vorgegangen: Am Ende der Reaktion wird eine verdünnte alkali sche wässrige Lösung, wie 5-10 %ige wässrige Natrium- oder Kaliumhydroxydlösung bis zu einem alkalischen. pH-Wert hinzugefügt und mit einem Lösungsmittel der obgenannten Gruppe, das mit Was ser nicht mischbar ist, extrahiert.
Der organische Aus zug wird verdampft und aus dem Rückstand wird nach den bekannten Kristallisierungsverfahren aus einem organischen Lösungsmittel oder Chromatographie an einem orga- Tonerde und darauffolgende Eluierung nischen Lösungsmittel, das 1-Methyl-N-carbobenzoxy- dhydro-lysergamin abgetrennt.
Dieses Produkt kann in bekannter Weise durch Zufügen eines Säureäquivalents in seine nicht toxi schen, pharmazeutisch verträglichen organischen oder anorganischen Salze, wie Chlorhydrat, Sulfat, Phos phat, Tartrat, Maleat, Citrat, Ascorbat, Succinat, Sali- zylat und andere gewöhnlich in der Therapie verwen dete Salze umgesetzt werden. Das erhaltene Produkt ist ein farbloses, kristallines Pulver, beständig sowohl ge gen Licht wie auch gegen Wärme und ist in den übli chen organischen Lösungsmitteln und Säuren löslich.
Beim Menschen kann das 1-Methyl-N-carbobenz- oxy-dihydrolysergamin oder dessen Salz auf oralem Wege verabreicht werden. Ausserdem findet es beson dere Verwendung bei den folgenden klinischen Indika tionen: Migräne, Kopfschmerz, Trigeminusneuralgie, Allergie im allgemeinen und bei den Entzündungs krankheiten.
Die üblich verwendeten therapeutischen Zuberei tungen enthalten 1-Methyl-N-carbobenzoxy-dihydro-ly- sergamin oder ein Salz hievon mit einer gewissen Menge eines festen oder flüssigen Bindemittels.
<I>Beispiel</I> 1-Methyl-N-carbobenzoxy-dihydro-lysergamin 16 g vorsichtig erwärmtes 1-Methyl-dihydro-lyser- gamin werden in 80 ml wasserfreiem Pyridin gelöst. Der unter Rühren auf -10 C gekühlten Lösung wer den ziemlich schnell 18 ml 85 %iges in: 36 ml Chloro form verdünntes Carbobenzoxychlorid (in Toluol) zu getropft. Die gut gerührte Reaktionsmischung wird während des Zusatzes bei -10 C und dann nochmals für 10 Min bei der gleichen Temperatur gehalten; dann wird das Kühlen beendet und die Temperatur steigt in ca. 10 Min wieder auf Raumtemperatur an. Das Ganze wird mit 240 ml Chloroform verdünnt und dann schnell mit 80 ml 5 %iger Natriumhydroxydlösung, dann mit gesättigter Natriumbikarbonatlösung und am Ende mit Wasser gewaschen.
Die Chloroformlösung wird hierauf kurz über wasserfreiem Natriumsulfat ge trocknet und im Vakuum bei 40 C zur Trockene ein gedampft. Der ölige Rückstand wird in 160 ml Benzol aufgenommen und auf eine chromatographische Säule aus A1203 gebracht; die Säule wird dann mit weiteren 160 ml Benzol eluiert. Die vereinigten Eluate werden im Vakuum bei 40 C eingedampft, der ölige dicke Rückstand wird einer kleinen Menge an wasserfreiem Äther zugefügt; nach einiger Zeit erhält man eine Kri stallmasse, die mit wenig Benzol und Äthyläther gewa schen wird. Es werden 12 g weisse Kristalle erhalten, die bei 146-148 C schmelzen; [α]D28 = -7 2 .
Anstelle von Reinigen durch Chromatographie kann das Produkt durch Umkristallisation aus Benzol- äthyläther erhalten werden. Das Produkt kann nach bekannten Verfahren in seine Salze mit nicht toxischen organischen oder anorganischen Säuren, wie Chlorhy drat, Sulfat, Phosphat, Tartrat, Maleat, Citrat, Ascor- bat, Succinat, Salizylat und andere in der Therapie übliche Salze übergeführt werden.
