Verfahren zur Herstellung von Penicillinsalzen Die vorliegende Erfindung bezieht sich auf ein Verfahren zum Herstellen von Penicillinsalzen des 1-Benzamido-1-phenyl- 3 -piperidino - propans der Formel

in der HP die saure Form eines Penicillins bedeutet. Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man ein Penicillin oder ein Salz eines solchen mit einer Base mit 1-Benzamido-l-phe- nyl-3-piperidinopropan oder einem Säuresalz desselben in Lösung umsetzt.
Beispielsweise geht man dabei so vor, dass man ein Penicillin und 1-Benzamido-l-phenyl-3-piperi- dino-propan zusammen in einem Lösungsmittel in Lösung bringt, in dem das 1-Benzamido-l-phenyl-3- piperidino-propansalz des Penicillins verhältnismässig unlöslich ist, wobei das Penicillin und das 1-Benz- amido-l-phenyl-3-piperidino-propan in solchen Mengen in Lösung gebracht werden, dass sich wesentlich mehr 1-Benzamido-l-phenyl-3-piperidino-propan-Salz des Penicillins bildet, als das Lösungsmittel zu lösen vermag, so dass eine Fällung erhalten wird, worauf die Fällung abgetrennt wird.
Das neue Salz kann ferner auch durch doppelte Umsetzung hergestellt werden, indem ein Salz des Penicillins mit einer Base mit einem Säuresalz des 1-Benzamido-l-phenyl-3-piperidino-propans umgesetzt wird. Diese Umsetzung wird am besten in einem Lösungsmittel durchgeführt, in dem alle chemischen Bestandteile mit Ausnahme des 1-Benzamido-l-phe- nyl-3-piperidino-propan-Salzes des Penicillins löslich sind, so dass eine leichte Isolierung dieser Substanz möglich ist.
Als Ausgangsstoffe kommen sowohl die natürlichen Penicilline, wie z. B. Penicillin G, Penicillin F, Penicillin X usw., als auch die biosynthetischen Peni- cilline, wie z. B. Penicillin O, Penicillin V usw., in Frage.
Die erfindungsgemäss erhältlichen neuen Salze haben verschiedene Anwendungsgebiete. Sie sind durch die Fähigkeit gekennzeichnet, eine Lokalanästhesie von ausserordentlich langer Dauer zu erzeugen. Zum Beispiel kann das 1-Benzamido-l-phe- nyl-3-piperidino-propan-Salz des Penicillins G, wenn es in Form einer Lösung in einem pharmazeutisch brauchbaren Lösungsmittel zur Infiltrationsanästhe- sie verwendet wird, eine 5 bis 7 Tage dauernde Anästhesie erzeugen. Das neue Salz zeichnet sich ferner durch die Fähigkeit aus, einen therapeutisch wirksamen Penicillinblutspiegel für längere Dauer zu erzeugen.
Beispielsweise ergibt die intramuskuläre Verabreichung einer wässrigen Suspension des feinverteilten 1-Benzamido-1-phenyl-3-piperidino-pro- pan-Salzes des Penicillins G in einer Menge von etwa 300 000 Penicillineinheiten im allgemeinen einen Peni- cillinblutspiegel, der therapeutisch bis zu 48 Stunden wirksam ist. Die Injektion einer Suspension von

600000 Einheiten ergibt in den meisten Fällen Blutspiegel, die 72 oder sogar 96 Stunden wirksam sind.
Infolge der sehr geringen Löslichkeit der 1-Benz- amido-l-phenyl-3-piperidino-propan-Salze der Peni- cilline lässt sich das erfindungsgemässe Verfahren auch dazu anwenden, um Penicillin auf leichte und bequeme Weise aus seinen wässrigen Lösungen abzutrennen.
Beispielsweise beträgt die Löslichkeit des 1-Benzamido-1-phenyl-3 -piperidino-propan-Salzes des Penicillins G in Wasser nur etwa 0,15 %, so dass sich das Penicillin G praktisch quantitativ in Form dieses Salzes ausfällen lässt.
Wie zu erwarten ist, unterscheiden sich die Löslichkeiten des 1-Benzamido- 1-phenyl-3-piperidino-propan-Salzes verschiedener Penicilline etwas; im allgemeinen liegen sie jedoch in der gleichen Grössenordnung wie die des 1-Benz- amido-l-phenyl-3-piperidino-propan-Salzes des Penicillins G.
Beispiel I Penicillin-G-Salz des 1-Benzamido-1-phenyl-3- piperidino-propans.
Eine Lösung von 1,8 g 1-Benzamido-l-phenyl-3- piperidino-propan-hydrochlorid wird zu 2 g des Kaliumsalzes von Penicillin G zugegeben, die in 5 cm3 Wasser gelöst sind. Die Lösungen werden gründlich gemischt, und das Gemisch wird gekühlt. Es trennt sich ein Öl ab, das beim Aufbewahren in einem Kühlschrank langsam kristallisiert. Das Kristallisieren kann etwas beschleunigt werden, wenn man die Gefässwand gelegentlich mit einem Glasstab kratzt. Die erhaltenen Kristalle des 1-Benzamido-l-phenyl-3- piperidino-propan-Salzes des Penicillins G werden abfiltriert, mit destilliertem Wasser gewaschen und an der Luft getrocknet.
Beispiel 2 Penicillin-G-Salz des 1-Benzamido-l-phenyl-3- piperidino-propans.
9 g 1-Benzamido-l-phenyl-3-piperidino-propan- hydrochlorid werden in 180 em3 destilliertem Wasser gelöst. Die Lösung wird filtriert, und Unter Rühren werden 10 g Natriumpenicillin G zugegeben, die in 25 cm3 destilliertem Wasser gelöst sind. Es werden Impfkristalle des Penicillin-G-Salzes des 1-Benz- amido-l-phenyl-3-piperidino-propans aus einem früheren Herstellungsverfahren zugesetzt, um die Abscheidung der Verbindung als Öl zu verhindern.
Die ausgeschiedenen Kristalle des Penicillin-G-Salzes des 1-Benzamido-1-phenyl-3-piperidino-propans werden auf einem Büchner-Trichter gewonnen und mit 50 cm3 kaltem destilliertem Wasser gewaschen. Das kristalline Salz wird zuerst in einem staubfreien Raum an der Luft -und dann in Vakuum über wasserfreiem Calciumchlorid 48 Stunden lang getrocknet.
Die Ausbeute an Penicillin-G-Salz des 1-Benzamido-l- phenyl-3-piperidino-propans beträgt etwa 16 g oder 97,5% der Theorie, bezogen auf die Menge des ver- wendeten Penicillins G. Die Kristalle haben keinen bestimmten Schmelzpunkt, sondern zersetzen sich beim Erhitzen allmählich. Die Löslichkeit des Salzes in Wasser beträgt etwa 1,5 mg/cm3. Die mikrobiologisch bestimmte antibiotische Wirksamkeit beträgt etwa 978 Oxford-Einheiten pro mg.
Die LD.O beträgt etwa 4279 T 179 mg/kg, wenn es Ratten subkutan injiziert wird. (Die LD"O des 1-Benzamido-l-phenyl- 3-piperidino-propanhydrochlorids beträgt, wie gefunden wurde, etwa 112 mg/kg, wenn es Ratten subkutan verabreicht wird.) Beispiel 3 Penicillin-V-Salz des 1-Benzamido-1-phenyl-3- piperidino-propans.
3,5g 1-Benzamido-l-phenyl-3-piperidino-propan- hydrochlorid werden in Wasser gelöst. Dieser Lösung werden 3,5 g Penicillin V (Phenoxymethylpenicillin), in einer verdünnten Kaliumkarbonatlösung gelöst, zugesetzt. Impfkristalle des 1-Benzamido-l-phenyl- 3-piperidino-propan-Salzes des Penicillins V werden der Penicillin-V-Lösung zugesetzt, und die Lösung des 1-Benzamido-l-phenyl-3-piperidino-propan- hydrochlorids wird unmittelbar darauf zugesetzt.
Die erhaltenen Kristalle des Penicillin-V-Salzes des 1- Benzamido-l-phenyl-3-piperidino-propans werden abfiltriert, mit kaltem Wasser gewaschen und dann in einem staubfreien Raum an der Luft getrocknet. Die Ausbeute beträgt etwa 95 % der Theorie. Das Peni- cillin-V-Salz des 1-Benzamido-l-phenyl-3-piperidino- propans ist in Wasser etwas löslicher als das entsprechende Penicillin-G-Salz; sie beträgt nämlich etwa 6,13 mg/em3 Wasser.
Beispiel 4 Zu 10 Litern einer wässrigen Lösung von rohem Kaliumpenicillin G (200000 Einheiten pro cm3), wie sie zum Beispiel bei der industriellen Herstellung von Penicillin aus einer Pilzkultur erhalten wird, werden 15 Liter einer wässrigen Lösung von 1125 g 1 -Benzamido-1-phenyl-3-piperidino-propanhydro- chlorid zugesetzt, die Impfkristalle des Penicillin-G- Salzes des 1-Benzamido-1-phenyl-3-piperidino-pro- Pans enthält. Die Lösung wird auf etwa 0 gekühlt und gerührt, bis die Ausfällung vollständig ist und eine etwaige ölige Fällung kristallin geworden ist.
Das kristalline Penicillin-G-Salz des 1-Benzamido-l- phenyl-3-piperidino-propans wird dann abfiltriert.
Das neue Salz kann wie gesagt auch aus der sauren Form des Penicillins und der basischen Form des 1-Benzamido-1-phenyl-3-piperidino-propans hergestellt werden, z. B. wie folgt: Das 1-Benzamido-l- phenyl-3-piperidino-propan wird in einem organischen Lösungsmittel, wie z. B. Äther. Chloroform, 1,2-Dichloräthan, Methylendichlorid, Kohlensäure- diäthylester, oder einem Carbonsäurealkylester, wie z. B. Äthyl- oder Amylacetat, gelöst werden.
Diese Lösung wird dann mit einer Lösung gemischt, die ein Moläquivalent Penicillin enthält, das zuvor in bekannter Weise durch Einstellung des PH-Wertes einer wässrigen Penicillinlösung auf 2,0 bis 2,2 und

Extrahieren der sauren Lösung bei einer verhältnismässig niedrigen Temperatur zwischen etwa 0 und 10' C mit einem unmischbaren organischen Lösungsmittel, wie z. B. Äther, Amylacetat, Butylacetat, n--Butanol, Chloroform, Methylendichlorid oder 1,2- Dichloräthan, hergestellt worden war. Eine derartige Arbeitsweise ist im folgenden Beispiel beschrieben.
Beispiel 5 Eine Lösung von 12,6 g Kaliumpenicillin G in 100 cm3 destilliertem Wasser wurde auf etwa 5 gekühlt. Der gekühlten Lösung wurden etwa 100 cm" Amylacetat zugesetzt, und dann wurde eine ausreichende Menge verdünnter Schwefelsäure langsam und unter Rühren zugesetzt, um den pH-Wert der wässrigen Phase auf 2,2 zu bringen. Das Gemisch wurde gründlich gerührt und der Amylacetatteil abgetrennt. Die wässrige Phase wurde erneut zweimal mit jeweils 30 cm3 Amylacetat extrahiert.
Die vereinigten Amylacetatextrakte wurden dreimal nacheinander mit jeweils 30 cm3 destilliertem Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet und filtriert. Der filtrierten Amylacetatlösung wurde dann eine Lösung von 10 g 1-Benzamido-l- phenyl-3-piperidino-propan in 75 cm3 Chloroform zugesetzt. Nach gründlichem Mischen wurde die Lösung durch Eindampfen unter vermindertem Druck auf etwa die Hälfte ihres ursprünglichen Volumens gebracht und etwa 12 Stunden lang auf etwa - 101 gekühlt. Es schied sich eine kristalline Fällung des 1-Benzamido-l-phenyl-3-piperidino-propan-Sal- zes des Penicillins ab, die abfiltriert und mit Äther gewaschen wurde.
Die Ausbeute betrug 19,8 g oder etwa 97 /a der Theorie.
Wenn die erfindungsgemäss erhältlichen Salze als Mittel zur Herbeiführung einer länger andauernden Anästhesie dienen sollen, so werden sie vorzugsweise in Lösung verwendet. Zur Herstellung der Lösung kann ein Lösungsmittel, wie z. B. wässriges Polyäthy- lenglykol 200, verwendet werden. Beispielsweise kann eine Lösung für therapeutische Zwecke, die zur Herbeiführung einer lang andauernden Anästhesie geeignet ist, dadurch erhalten werden, dass man 0,045 g des 1-Benzamido-l-phenyl-3-piperidino-pro- pan-Salzes des Penicillins G in 1 cm3 70prozentigen wässrigen Polyäthylenglykol-200 unter sterilen Bedingungen löst.
Vor der Verwendung wird das Gemisch vorzugsweise leicht angewärmt, um sicherzustellen, dass das Salz vollständig gelöst ist. Suspensionen, die 1 oder 2 1/a des 1-Benzamido-I-phenyl-3- piperidino-propan-Salzes des Penicillins G enthalten, ergeben gleichfalls eine langanhaltende anästhesierende Wirkung.
Ein wässriges medizinisches Präparat, das bei intramuskulärer Injektion einen länger anhaltenden therapeutisch wirksamen Penicillinblutspiegel aufrechtzuerhalten vermag, besteht zum Beispiel aus einer Suspension, die etwa 300 bis etwa 700 mg des fein zerteilten Penicillin-G-Salzes des 1-Benzamido- 1-phenyl-3-piperidinopropans pro cm3 der Suspension enthält. Das Suspensionsmedium kann entweder Wasser oder ein Pflanzenöl sein oder ein anderes für parenterale Verabreichung geeignetes Medium, wie z.
B. Kohlensäurediäthylester. Die intramuskuläre Verabreichung von 1 cm3 der vorstehenden Suspension ergibt in den meisten Fällen einen therapeutisch wirksamen Penicillinblutspiegel von etwa 48- bis 96stündiger Dauer, und zwar in Abhängigkeit von der Konzentration und Menge der verabreichten Suspension.
Als Lösungsmittel kann man anstelle des obenerwähnten wässrigen Polyäthylenglykols-200 auch zum Beispiel wässriges Polyäthylenglykol-300, wäss- riges Polyäthylenglykol-400, wässriges Polypropylen- glykol-250 und dergleichen sowie andere für paren- terale Verwendung geeignete Lösungsmittel verwenden.
Es können auch andere therapeutisch wirksame Bestandteile den ein erfindungsgemäss erhältliches Salz enthaltenden medizinischen Präparaten zugegeben werden. Weiterhin können die das Salz enthaltenden Zubereitungen auch Suspensionsmittel, oberflächenaktive Mittel und dergleichen enthalten, wie sie üblicherweise zur Herstellung von Suspensionen von Prokain-Penicillin verwendet werden.
Das 1 -Benzamido-1-phenyl-3-piperidinopropan kann wie folgt hergestellt werden: Ein Gemisch aus 101,5.g ss-Piperidino-propiophe- nonhydrochlorid, das nach dem Verfahren von Adams [Organic Reactions, Bd. 1, S.329 (1942)] hergestellt worden war, 31,4 g Hydroxylaminhydrochlorid, 33,6 g Natriumbicarbonat und 200 cm3 Wasser wurde über Nacht bei Raumtemperatur abgestellt.
Dem Reaktionsgemisch wurde dann eine Lösung von 43 g Natriumhydroxyd in 43 cm3 Wasser zugesetzt, und nach dem Kühlen wurde das ausgefallene ss-Piperi- dino-propriophenonoxim abfiltriert. Nach dem Um- kristallisieren aus Methanol schmolz das ss-Piperidino- propiophenonoxim bei 152 bis 153 .
Ein- Gemisch aus 10 g Raney-Nickelkatalysator, 81,5 g f-Piperidino-propiophenonoxim, 200 cm3 Äthanol und 75 cm3 flüssigem Ammoniak wurde in einen Autoklaven gegeben und eine Stunde lang bei 70 bis 110 und einem Druck von etwa 100 kg/em-' hydriert. Das Reaktionsgemisch wurde von dem Katalysator abfiltriert, und das Filtrat wurde unter ver- . mindertem Druck fraktioniert. Das bei der Reaktion entstandene 1-Phenyl-3-piperidino-propylamin destillierte bei 133 bis 134 C und einem Druck von 1 mm Hg.
Ein Gemisch aus 24 g 1-Phenyl-3-piperidino-pro- pyl-amin, 24 cm3 Benzoylchlorid, 300 cm3 Benzol und 45 cm3 trockenem Pyridin wurde etwa 16 Stunden bei Raumtemperatur stehengelassen. Eine ausreichende Menge Äthyläther wurde dann zugesetzt, um das während der Reaktion entstandene 1-Benz- amido -1-phenyl- 3 - piperidino - propan-hydrochlorid vollständig auszufällen. Die Fällung wurde abfiltriert

und durch Umkristallisieren aus einem Gemisch von Isopropanol und Äther gereinigt.
Das so hergestellte 1-Benzamido-l-phenyl-3-piperidin-propanhydrochlo- rid schmolz bei etwa 188 bis 189 C.
Aus dem Salz kann die freie Base durch Behandlung einer wässrigen Lösung des Salzes mit einem Äquivalent Alkali und Isolieren der sich aus der Lö- sung abscheidenden freien Base erhalten werden. Die freie Base kann dann kristallisiert werden.

Instead of the aqueous polyethylene glycol-200 mentioned above, aqueous polyethylene glycol-300, aqueous polyethylene glycol-400, aqueous polypropylene glycol-250 and the like and other solvents suitable for parenteral use can also be used as solvents.
Other therapeutically effective constituents can also be added to the medicinal preparations containing a salt obtainable according to the invention. Furthermore, the preparations containing the salt can also contain suspending agents, surface-active agents and the like, as are customarily used for the preparation of suspensions of procaine-penicillin.
The 1-benzamido-1-phenyl-3-piperidinopropane can be prepared as follows: A mixture of 101.5 g of ss-piperidino-propiophenone hydrochloride, which is prepared by the method of Adams [Organic Reactions, Vol. 1, p. 329 (1942)], 31.4 g of hydroxylamine hydrochloride, 33.6 g of sodium bicarbonate and 200 cm3 of water were stored overnight at room temperature.

The free base can be obtained from the salt by treating an aqueous solution of the salt with one equivalent of alkali and isolating the free base which separates out from the solution. The free base can then be crystallized.
