und R6 Wasserstoff oder eine Alkylkette mit 1-4 C-Atomen, n gleich 1 oder 2 ist und R2, R3, R4 und R5 jeweils für Wasserstoff, Aikylgruppen mit 14 C-Atomen, Cl-, Hydroxy-, Niitro-, Amino-, Suifo-, Sulfonamid-, Carbon säureamid-, Acetamino- und Acetylgruppen stehen, wertvolle biologische Eigenschaften besitzen.
Die Verbindungen beeinflussen als solche oder in Form ihrer Salze mit nicht-toxischen anorganischen oder organischen Säuren Amöben und Tb-Infektionen günstig. im Vergleich dazu sind die bekannten nichtbasi- sehen Imide der Naphthalin-(1 (1, 4, 5, 8) tetracarbonsäure [D.R.P. 552 760; H. Vollmann, H. Becker, M. Corell, H. Streeck u. G. Langbein A.531, 1 (1937); H. E.
Fierz-David und C.Rossi, Helv. Chim. Acta 21, 1466 (1938); US 2 914 531] unwirksam.
Erfindungsgemäss werden die basisch alkylierten naphthalin-(1,4,5,8)-tetracarbonsäurediimide dadurch hergestellt, dass man ein Anhydrid der allgemeinen Formel

oder die entsprechende Tetracarbonsäure mit Aminen der allgemeinen Formel

umsetzt. Die Umsetzung kann in Gegenwart eines Lösungsmittels oder in der Schmelze erfolgen. Als beson- ders günstig hat sich die Verwendung von Toluol und Xylol als Lösungsmittel erwiesen. Bei der bei Siedetem- peratur rasch verlaufenden Reaktion lässt sich das ent stehende Wasser durch azeotrope Destillation am Wasserabscheider in einfacher Weise entfernen.
Die erfindungsgemäss hergestellten Substanzen zeichnen sich durch eine gute Wirkung auch gegenüber solchen Tuberkelbazillen aus, die gegen eine Behandlung durch Isonikotinsäurehydrazid (INH) resistent geworden sind.
Sie zeigen auch gegenüber anderen klinisch bedeutsamen Tuberkulostatika wie Streptomycin, p-Aminosalicylsäure und Thiosemicarbazon keine Kreuzresistenz.
In der folgenden Aufstellung sind die minimalen Hemmkonzentrationen von verschiedenen Verfahrensprodukten in Iml Eimedium, die für eine Unterdrückung des Wachstums der Mykobakterien erforderlich waren, im Vergleich zur Hemmkonzentration des Isonikotinsäurehydrazids aufgeführt. Die Versuche wurden an einem Laborstamm (262) durchgeführt, der ursprünglich aus Patientenmaterial isoliert worden ist.

Die Wirkung verschiedener Verfahrensprodukte an der Maus wurde in der folgenden Versuchsanordnung ermittelt:
Infiziert man Mäuse intravenös mit einer Keimmenge von etwa 107 des Mydobakterienstammes H37Rv, so gehen nicht behandelte Tiere etwa um den 20. Tag nach der Infektion zu 100% an einer Tuberkulose ein.
Jeweils 10 auf diese Weise infizierte Mäuse wurden zu einer Behandlungsgruppe zusammengefasst und mit einer Standarddosierung von 30 mg/kg Körpergewicht lmal täglich über einen Zeitraum von 4 Wochen bei 5 Applikationen pro Woche mittels Schlundsonne behandelt.
An den Stichtagen 15., 20., 25., 30., 35., 40. und 50.
Tag nach der Infektion wurde die Überlebenszahl jeder Mäusegruppe bestimmt. Bei verwertbarem Infektionsablauf lebten am 15. Tage alle Mäuse; am 25. Tag waren unbehandelte bzw. ohne Therapieeffekt behandelte Mäuse an einer Tuberkulose eingegangen.
Aus diesem Infektionsablauf resultiert die Berechnung des Überlebens-Index .
Die Summe der überlebenden Mäuse einer Behandlungsgruppe an den genannten Stichtagen wurde durch die Summe der überlebenden Mäuse der Infektionskontrolle dividiert. Bei sieben Werterhebungen ist 70 der grösste erreichbare Summenwert und 10 der niedrigste Summenwert für die Infektionskontrolle. Somit besteht 7 als Höchstwert für den Uberlebens-Index, und 1 bedeutet keine therapeutische Wirksamkeit; d. h., die Überlebenswerte der Behandlungsgruppe entsprechen im letzteren Falle denen der Infektionskontrolle. Bei normalem Infektionsablauf mit einem Summenwert um 20 für die Infektionskontrolle ergeben sich bei einem deutlichen Therapie effekt Index-Werte ab 2.
Die erfindungsgemäss hergestellten Substanzen erreichen in diesem Versuch die folgenden Werte: Verbindung Index

Wie zu erwarten, zeigten diese Substanzen bei mit gegen Isonikotinsäurehydrazid und Streptomycin resistenten Tuberkelbazillen infizierten Mäusen (Stamm 282a/59) ebenfalls eine Wirkung.
Bei einer Dosierung von 40 mg/kg wurden die folgenden Werte erhalten: Verbindung Index

Die überlegene Wirksamkeit eines der Erzeugnisse des erfindungsgemässen Verfahrens wurde auch in folgen der Versuchsanordnung nachgewiesen:
Mäuse wurden mit einem aus Patientenmaterial iso- lierten gegenüber Isonikotinsäurehydrazi ! d (INH) und Streptomycin (SM) therapeu-resistenten Mykobakterienstamm (Laborstamrn 282a/59) intravenös infiziert. Vom 1.
Tag nach der Infektion ab wurde an den werktagen einmal täglich innerhalb vier Wochen insgesamt 20mal - soweit die Tiere der Infektion nicht erlagen - behan- delt.
Die Dosierungen betrugen für Isonikotinsäurehydrazid und Streptomycin 40 mg/kg und für das Prüfpräparat Nr. 16 30 mg/kg. Isonikotinsäurehydrazid und das Prüfpräparat wurden oral, Streptomycin subcutan appli zielt. Die mit je 10 Mäusen zusammengestellten Versuchsgruppen wurden wöchentlich zweimal gewogen, das Durchschnittsgewicht pro Maus errechnet und zum Anfangsgewicht bei Versuchsbeginn, das als 100 %-Wert eingesetzt wurde, in Beziehung gesetzt. Die so erhaltenen Prozentwerte der Durchschnittsgewichte wurden mit der Anzahl der überlebenden Mäuse an den Stichtagen mul tipliziert. Die Werte stellen die Messpunkte für die Kurvenzüge der Abbildung dar.
Wie aus der Abbildung ersichtlich, zeigte die Therapie mit Isonikotinsäurehydrazid (1) und Streptomycin (2) keinen Effekt; gleichlaufend mit der Infektiionskontrolle (3) gingen die Mäuse an der Tuberkulose ein.
Mit dem Prüfapparat (4) gelang es, den Infektionsablauf deutlich zu verzögern, so dass gegen Versuchsende nach 3-wöchigem therapiefreiem Intervall - einem Zeitraum, in dem bei der angewandten Infektionsdosis unbehandelte Tiere an der Tuberkulose eingingen - noch 40 % der einge- setzten Tiere überlebten. (Kurve 5 zeigt die gesunde Kontrolle).
Beispiel 1
30 g Naphthalin-1,4,5,8-tetracarbonsäuredianhydrid werden in 250 ccm Toluol zum Sieden erhitzt. Man tropft 42 g ss-Diäthylamino-äthylamin ein und kocht am Wasserabscheider bis sich die berechnete Menge Wasser abgeschieden hat. Nach dem Erkalten wird mit verdünnter Essigsäure ausgeschüttelt, die essigsaure Lösung alkalisch gestellt und der ausfallende Niederschlag abgesaugt und aus Benzol umkristallisiert. Man erhält 19 g N,N' Bis-(ss-diäthylamino-äthyl)-naphthalin-1,4,58-tetracarbonsäurediimid in Form orangefarbener Kristalle, die bei 243-246 schmelzen.

Das als Ausgangsstoffe verwendete ss-Isobutylamino- äthylamin wird aus dem Kondensationsprodukt von Isobutyraldehyd und Äthylendiamin durch katalytische Hydrierung (Kp11 58-59 C) hergestellt.
Das als Ausgangsmaterial verwendete N-(2'-Amino äthyl)-3-azabicyclo-[3,2,2]-nonan wird auf folgendem Wege hergestellt: Durch Erhitzen von 35 g 3-Azabicyclo [3,2,2]nonan, (F. 190-195 C, hergestellt durch Ammoniakabspaltung aus 1,4-Bis-(aminomethyl)-cyclohexan mit AluminiumhydroxydkatalysaLtor in der Gasphase bei 3500 C) mit 13,5 g Glykolsäurenitril in 200 ml Äthanol und 50 ml Wasser erhält man 35,7 g N-Cyanmethyl-3azabicyclo-[3,2,2]nonan, Kp12, 1340 C, F. 500 C (roh).
Durch Reduktion dieser Verbindung mit 8 gLithium- aluminiumhydrid in ätherischer Lösung bei -5 C entstehen daraus 31,5 g N-(2'-Aminoäthyl)-3-azabicyclo [3,2,2]nonan, Kp12, 116-188 C.
Das als Ausgangsstoff verwendete N-(3'-Aminopropyl)-3-azabicyclo-[3,2,2]nonan, Kp12 126-128 C, wird durch Umsetzung von 3-Azabicyclo[3,2,2]nonan mit Acrylnitril zu N-Cyanäthyl-3-azabicyclo-[3,2,2]- nonan, Kp0,4 110 C, und nachfolgende katalytische Hydrierung mit Raney-Nickel gewonnen.
Das als Ausgangsstoffe verwendete ss-(3, 3-Dimethyl- pyrrolidino)-äthylamin ist auf folgendem Wege zugänglich: α,α-Dimethylsuccinimid, Kp12 136-140 C, f. 101 1020 C, wird katalytisch zu 3,3-Dimethylpyrrolidon, Kp12 120 C, reduziert und anschliessend mit Lithiumaluminiumhydrid zu 3,3-Dimethylpyrrolidon, Kp760, 108 bis 110 C, reduziert.
Durch Umsetzung von Glykolsäurenitril mit dieser Verbindung entsteht N-(Cyanmethyl)-3,3-dimethylpyrrolidon, Kp12, 840 C, das mit Lithiumaluminium- hydrid zu N-(2'-Aminoäthyl)-3,3-dimethylpyrrolidon, Kp12, 74 C, reduziert wird.
Beispiel 2
In analoger Weise, wie in Beispiel 1 angegeben, erhält man N,N'-Bis-(γ-hexamethylenimino-propyl)-naph- thalin-1,4,5,8-tetracarbonsäurediimid, F. 191-193 C und N,N'-Bis-(γ-[dicyclohexylmethylamino]-propyl)- naphthalin-1,4,5,8-tetracarbonsäureimid, F. 152 C.
Das als Ausgangsstoff verwendete y-Hexamethylen- imino-propylamin wurde auf folgende Weise hergestellt:
27 g Acrylnitril werden in eine Lösung von 50 g Hexamethylenimin in 200 ml Benzol eingetropft. Man kocht über Nacht unter Rückfluss und destilliert im Vakuum, Kp4, 95 C; man erhält 60 g ss-Hexamethyleniminopropionitril. Dieses Nitril wird in methanolischem Ammoniak bei 50 atü und 95 C in Gegenwart von Ranev Kobalt hydriert. Man erhält nach der Destillation bei 10 mm und 1170 C 50 g y-Hexamethylenimino-propyl- amin.

Die als Ausgangsstoffe benötigten Amine wurden folgendermassen hergestellt: γ-Piperidino-ss-dimethyl-propylamin und γ-Morpho- lino-ss-dimethyl-propylamin sind durch Überführung der entsprechenden Alkohole in die Chloride und Austausch des Chloratoms gegen Ammoniak zugänglich.
Beispiel 4 ss-Diäthylaminopropylamin wird in analoger Weise, wie in Beispiel 1 angegeben, mit Naphthalin-1,4,5,8- tetracarbonsäuredianhydrid umgesetzt. Man erhält N,N' Bis-[ss-diäthylamino-propyl]-naphthalin-1,4,5,8-tetracarbonsäurediimid, das bei 130-134 C schmilzt.
BeispielS
Naphthalin-1,4,5,8-tetracarbonsäuredianhydrid wird in 150 ml Toluol suspendiert und unter Rühren zum Sieden erhitzt. Man tropft 13 g -(ss'-Diäthylamino-äthoxy)-propylamin, gelöst in 50 ml Toluol, ein und kocht am Wasserabscheider, bis die Reaktion beendet ist. Nach dem Erkalten wird abgesaugt, der Rückstand in verdünnter Essigsäure gelöst und mit Ammoniak gefällt.
Der ausfallende Niederschlag wird mit Wasser gewaschen und lässt sich aus Toluol umkristallisieren. Man erhält 18 g N,N'-Bis-[γ-(ss'-diäthylamino-äthoxy)-propyl]-naph- thalin-1,4,5,8-tetracarbonsäurediimid, F. 80-84 C.

Verschiedene als Ausgangsstolle verwendete Amine wurden durch Addition von Acrylnitril an Phenyläthylamin, N-Benzyl-N-propylamin, N-Isobutyl-N-isopropylamin, N-Benzyl-N-methylamin und N-Benzyl-N-isobutylamin und anschliessende katalytische Hydrierung gewonnen.
Das als Ausgangsstoffe verwendete #-(N-Benzyl-N- methylamino)-α-methylbutylamin wurde durch Umsetzung des 1-Brom-3-acetylpropans mit N-Benzyl-Nmethylamin und anschliessende katalytische Hydrierung in Gegenwart von Ammoniak dargestellt. #-Piperidino- und Hexamethylenimino-butylamin, die als Ausgangs stoffe benötigt wurden, wruden aus #-Brombuttersäure- nitril und Piperidin bzw. Hexamethylenimin und anschliessende katalytische Hydrierung dargestellt.
Beispiel 6
Der benötigte Ausgangsstoff wurde folgendermassen hergestellt:
Man erwärmt 130 g Äthylendiamin in 200 ml Toluol zum Sieden und tropft unter Rühren 195 g 2-Bromhexan ein. Anschlicssend wird unter Rückfluss über Nacht gekocht, das Lösungsmittel abdestilliert und der Rückstand nach dem Erkalten in einem Überschuss an gesättigter wässriger Natronlauge aufgenommen. Die obere, der sich bildenden beiden Schichten wird abgetrennt und im Vakuum destilliert. Man erhält bei 14 mm und 9598 65 g ss-(1-Methyl-pentylamino)-äthylamin. Die Umsetzung von 65 g ss-(1-Methyl-pentylamino)-äthylamin mit Naphthalin-1,4,5,8-tetracarbonsäuredianhydrid analog Beispiel 1 ergibt 72 g N,N'-Bis-[ss-(hexyl)-(2)-amino) äthyl]-naphthalin-1,4,5,8-tetracarbonsäurediimid, F.

The N- (2'-amino ethyl) -3-azabicyclo- [3.2.2] -nonane used as starting material is prepared in the following way: By heating 35 g of 3-azabicyclo [3.2.2] nonane, (F. 190-195 C, prepared by splitting off ammonia from 1,4-bis (aminomethyl) cyclohexane with aluminum hydroxide catalyst in the gas phase at 3500 C) with 13.5 g of glycolonitrile in 200 ml of ethanol and 50 ml of water, 35, 7 g of N-cyanomethyl-3azabicyclo- [3.2.2] nonane, bp 12, 1340 C, m.p. 500 C (crude).
By reducing this compound with 8 g of lithium aluminum hydride in an ethereal solution at -5 C, 31.5 g of N- (2'-aminoethyl) -3-azabicyclo [3.2.2] nonane, bp12, 116-188 C.
The N- (3'-aminopropyl) -3-azabicyclo- [3.2.2] nonane, boiling point 126-128 C, used as the starting material is converted into by reacting 3-azabicyclo [3.2.2] nonane with acrylonitrile N-cyanoethyl-3-azabicyclo [3.2.2] nonane, boiling point 0.4 110 ° C., and subsequent catalytic hydrogenation with Raney nickel.

The γ-hexamethylene-imino-propylamine used as starting material was prepared in the following way:
27 g of acrylonitrile are added dropwise to a solution of 50 g of hexamethyleneimine in 200 ml of benzene. It is refluxed overnight and distilled in vacuo, boiling point 4, 95 ° C .; 60 g of β-hexamethyleneiminopropionitrile are obtained. The amines required as starting materials were prepared as follows: γ-piperidino-ß-dimethyl-propylamine and γ-morpholino-ß-dimethyl-propylamine can be obtained by converting the corresponding alcohols into the chlorides and replacing the chlorine atom with ammonia.

Example S.
Naphthalene-1,4,5,8-tetracarboxylic dianhydride is suspended in 150 ml of toluene and heated to the boil with stirring. 13 g of - (ss'-diethylamino-ethoxy) -propylamine, dissolved in 50 ml of toluene, are added dropwise and the mixture is boiled on a water separator until the reaction has ended. Various amines used as starting materials were obtained by adding acrylonitrile to phenylethylamine, N-benzyl-N-propylamine, N-isobutyl-N-isopropylamine, N-benzyl-N-methylamine and N-benzyl-N-isobutylamine and subsequent catalytic hydrogenation.
