Im Hauptpatent wird ein Verfahren zur Herstellung des neuen Äthylesters der 6,7-Diäthoxy-4-hydroxychi nolin-3-carbonsäure der Formel:

beschrieben, welches dadurch gekennzeichnet ist, dass man 1 ,2-Diäthoxy-4-nitrobenzol in Gegenwart eines Katalysators und eines Lösungsmittels reduziert, Di äthyläthoxymethylenmalonsäureester zu der Lösung des reduzierten Produktes gibt, das Lösungsmittel entfernt, ein die Temperatur regulierendes Medium zusetzt und danach dem Gemisch Wärme zuführt.
Die Erfindung betrifft nun ein Verfahren zur Herstellung von neuen niederen Alkylestern von
6,7-Di-(niederalkoxy)-4-hydroxychinolin
3-carbonsäuren der Formel

worin R und R1 je einen Alkylrest mit vorzugsweise 2 bis 4 Kohlenstoffatomen und R2 eine niedere Alkylgruppe bedeuten, wobei R, R1 und R2 nicht gleichzeitig Äthyl darstellen.
Diese neuen Verbindungen zeichnen sich durch das hohe Mass der chemotherapeutischen Wirksamkeit aus, die sie bei der Behandlung parasitärer Infektionen von Tieren zeigen, wenn sie in erheblich geringeren als den toxischen Mengen verabreicht werden. Sie haben sich bei oraler Verabreichung als überraschend erfolgreich bei der Bekämpfung der Coccidiose erwiesen.
Die Coccidiose, eine weitverbreitete und wirtschaftlich sehr nachteilige Geflügelkrankheit, wird von verschiedenen Coccidien, einschliesslich Eimeria tenella, Eimeria acervulina und Eimeria necatrix, verursacht.
Die neuen erfindungsgemäss erhältlichen Verbindungen sind hochwirksame Mittel zur Bekämpfung dieser Coccidien. Wenn sie einem ernährungsmässig ausgewogenen Futter in einer Menge von etwa 0,006 bis 0,1 Ges. % zugesetzt und so an mit Coccidien infizierte Hühner verabreicht werden, werden die Sterblichkeit und Morbidität beseitigt, die gewöhnlich bei durch Coccidien verursachten Krankheiten auftreten, und es wird eine günstige Wirkung auf Wachstum und Aussehen der Vögel ausgeübt.
Die Zubereitung dieser neuen Verbindungen in einer Form, welche eine leichte Verabreichung ermöglicht, kann ohne weiteres durch Zumischen zum Futter der Tiere gemäss obigen Angaben erfolgen. Die neuen Verbindungen können gewünschtenfalls in Tabletten, Gelatinekapseln, Suspensionen oder ähnliche Zubereitungen aus geeigneten Trägern und Zusatzstoffen, wie sie in der Pharmazie üblich sind, einverleibt werden.
Das Verfahren gemäss der Erfindung ist dadurch gekennzeichnet, dass man ein 1,2-Di-(niederalkoxy- 4-nitrobenzol reduziert, das erhaltene Reduktionsprodukt mit einem entsprechenden Di-(niederalkyl)-niederalkoxymethylenmalonsäureester in Gegenwart eines die Temperatur regulierenden Mediums in Berührung bringt und das Gemisch erhitzt.
Vorzugsweise reduziert man das 1,2-Di-(niederal- koxy)-4-nitrobenzol in Gegenwart eines Katalysators, wie Palladium-auf-Kohle, und eines Lösungsmittels, wie Äthanol. Als Di-(niederalkyl)-niederalkoxymethylenma lonsäureester kann man Diäthyläthoxymethylen- oder Dimethylmethoxymethylen-malonat zur Lösung des reduzierten Produktes geben, das Lösungsmittel entfernen und als temperaturregulierendes Mittel z. B. Dow- therm (eingetragene Marke) zugeben, worauf man dem Gemisch Wärme zuführt.
Andere niedere Alkylester können durch Verseifen eines erhaltenen niederen Alkylesters und anschliessende Behandlung der gebildeten Säure nach Fisher-Speier oder Schotten-Baumann hergestellt werden. Eine andere Methode zur Herstellung verschiedener niederer Alkylester ist die Umesterung, indem eine niedere Alkylgruppe gegen eine andere ausgetauscht wird.
Beispiel I Äthyl-6,7-dibutoxy-4-hydroxy-chinolin-3-carboxylat
In eine dickwandige Flasche werden 29, 48 g (0. 11 Mol) 1.2-Dibutoxy-4-nitrobenzol, 200 ml Äthanol und 3 g Palladium/Kohle gebracht. Die theoretische Wasserstoffaufnahme bei der Reduktion dieser Mischung in einer sogenannten Parr-Anlage wird in einer Stunde erzielt (3,15 kg,lcm).
Die Reduktion wird mit einer gleichen Menge der Nitroverbindung wiederholt. Die Reduktionsprodukte werden vereinigt, filtriert und der Katalysator mit Äthanol gewaschen. Das Filtrat wird zusammen mit 47,5 g Diäthyläthoxymethylenmalonat in einen 1Liter Dreihalskolben gebracht. Die Mischung wird auf dem Dampfbad erwärmt und das Äthanol abdestilliert. Es wird Dowtherm A zu (600 ml) zugegeben und die Mi- schung rasch zum Rückfluss erwärmt (2500 C). Nach 40 Minuten Kochen am Rückfluss wird die Mischung auf Raumtemperatur abgekühlt. Hexan wird zugesetzt.
Der kristalline Niederschlag wird abfiltriert und mit Hexan gewaschen. Man erhält eine 29 % ; ige Ausbeute an rohem Äthyl-6.7-dibutoxy-4-hydroxychinolin-3-carb oxylat (22,76 g), Fp. 260-285 C.
Dieses Produkt kann gewünschtenfalls unter Verwendung von Aktivkohle aus Dimethylformamid um kristallisiert werden, wobei 19 % Wi 6,7-Dibutoxy-4-hydroxy- chinolin-3-carboxylat, Fp. 270-284 C, erhalten werden.

Beispiel 2
Athyl-6,7-dipropoxy-4-hydroxychinolin-3 -carboxylat
In eine dickwandige Flasche werden 38 g (0,16 Mol) 1,2-Dipropoxy-4-nitrobenzol, 200 ml Äthanol und 3 g 5 Ó iges Pd-auf-Kohle gebracht. Die Mischung wird in einer Parr-Anlage bis zu einer Wasserstoffaufnahme von 3.92 kgXcm- (Theorie 3,99 kg/cm2) hydriert. Dies wird wiederholt, bis insgesamt 172 g (0,72 Mol) der Nitroverbindung reduziert sind. Der Katalysator wird abfiltriert, und die Äthanolfütrate werden für den nächsten Verfahrensschritt vereinigt.
Die Äthanollösung wird mit Diäthyläthoxymethylenmalonat (0,72 Mol) gemischt und der Alkohol durch Abdestillieren auf dem Dampfbad entfernt. Die Erwärmung wird 3 Stunden fortgesetzt. Dowtherm A (750 ml) wird zugegeben und die Mischung rasch zum Rückfluss erwärmt. Die Lösung wird 30 Minuten am Rückfluss gekocht und abkühlen gelassen. Der kristalline Niederschlag wird abfiltriert und mit Hexan gewaschen. Die Ausbeute an der gewünschten Verbindung beträgt 96 g (40 X, bezogen auf die Nitroverbindung).
Sie kann aus Dimethylformamid umkristallisiert werden (Fp. 271-273 C).

Beispiel 3
A. 0,24 Mol eines nach Beispiel 1 oder 2 erhaltenen Esters und 300 ml einer 1ORigen Natriumhydroxydlösung werden in einen l-Liter-Kolben gegeben. Die Mischung wird 2,5 Stunden am Rückfluss gekocht. Es wird Aktivkohle zugesetzt, die Mischung erhitzt und dann abkühlen gelassen. Die Mischung wird mit 200 ml l:l-Salzsäure (100 ml konzentrierte Salzsäure und 100 ml Wasser) angesäuert. Es bildet sich ein cremefarbener Niederschlag. Dieser wird abfiltriert, mit Wasser gewaschen und über Nacht bei 1100 C getrocknet. Dies ergibt die entsprechende 6,7 Dialkoxy-4-hydroxy-chinolin-3-carbonsäure.
Zu 0,2 Mol der in 1500 ml Benzol suspendierten Säure werden unter Rühren 15 mi (0,2 Mol) Thionylchlorid zugesetzt.
Die Mischung wird unter Rühren 8,5 Stunden zum Rückfluss erhitzt, bis sich kein HCl-Gas mehr entwickelt.
Nach einer schwachen Abkühlung wird der Feststoff gesammelt, gründlich mit Benzol und dann mit Äther gewaschen. Dies ergibt das Säurechlorid.
Eine Suspension von 54,5 g (0185 Mol) des Säurechlorides in 1500 ml Methanol wird unter Rückfluss auf dem Dampfbad 5,5 Stunden erwärmt. Die Reaktionsmischung wird heiss filtriert und das Filtrat abgekühlt. Der sich abscheidende Feststoff wird abfiltriert und mit Äther gewaschen. Durch Zugabe von Wasser zum ursprünglichen Filtrat wird weiterer Feststoff erhalten, der abgetrennt und getrocknet wird. Das rohe Produkt kann durch Umkristallisieren aus 3 Liter Dimethylformamid mit Aktivkohle gereinigt werden und ergibt den entsprechenden Methylester.
B. 0,02 Mol der Verbindung von Beispiel 1 oder 2 in 700 ml Methanol und eine kleine Menge p-Toluolsulfonsäure werden 7 Tage zum Rückfluss erhitzt. Die Reaktionsmischung wird heiss filtriert und der Feststoff mit Äther gewaschen. Dies ergibt nach dem Trocknen den entsprechenden Methylester. Durch Einengen der ursprünglichen Reaktionsmischung können weitere Mengen Feststoff erhalten werden.
Beispiel 4 Äthyl-6,7 - diisopropoxy-4-hydroxychinolin-3-carboxylat A. o-Diisopropoxybenzol
Stickstoff wird in langsamem Strom durch eine Lösung von 220 g (2 Mol) Benzcatechin in 90 ml Alkohol geleitet. Dies geschieht in einem mit Rührer, Rückflusskühler, Tropftrichter und Stickstoffzuleitung ausgerüsteten Dreihalskolben. Nach einigen Minuten Rühren werden 160 g (4 Mol) Natriumhydroxyd zugesetzt und weiter gerührt, bis praktisch das gesamte Natriumhydroxyd aufgelöst ist. Zu der gerührten dicken Suspension werden während eines Zeitraums von 2 Stunden 750 g (6 Mol) 2-Brompropan zugesetzt, wobei mit einem Dampfbad ein schwacher Rückfluss aufrechterhalten wird. Nach der Zugabe wird die Stickstoffzufuhr unterbrochen und weitere 18 Stunden unter Rückfluss gehalten. Die Mischung wird gründlich gekühlt, das Natriumbromid abfiltriert und mit Alkohol gewaschen.
Das mit der Waschflüssigkeit vereinigte Filtrat wird im Vakuum bis zur möglichst weitgehenden Entfernung des Alkohols eingeengt. Während des Einengens bildet sich eine weitere Menge Natriumbromid und wird entfernt. Insgesamt fallen 390 g Natriumbromid (95 % der Theorie) an. Der ölige Rückstand wird in 500 ml Äther gelöst und 4mal mit je 200 ml 10% Der Natriumhydroxydlösung und dann 2mal mit je 100 ml Wasser extrahiert. Die Ätherlösung wird über Magnesiumsulfat getrocknet und zuerst bei Normaldruck und dann im Vakuum eingeengt. Dies ergibt 82 g bernsteinfarbenes Öl (47 % der Theorie an Diisopropoxybenzol).
B. 1 2-Diisopropoxy-4-nitrobenzol o-Diisopropoxybenzol (140 g, 0,72 Mol) wird in einer Stunde unter Rühren tropfenweise zu einer Lösung von 90 ml konzentrierter Salpetersäure in 90 ml Wasser zugegeben. Die Temperatur wird durch wiederholte Anwendung eines Eisbades bei 20-250 C gehalten. Es wird weiter gerührt, bis die exotherme Reaktion abgeklungen ist. Die Reaktionsmischung wird mit einem gleichen Volumen Eis-Wasser verdünnt und das dunkle, ölige Produkt dreimal mit je 250 ml Chloroform extrahiert. Die vereinigten Chloroformextrakte werden 3mal mit je 100 ml Wasser extrahiert und über Magnesiumsulfat getrocknet. Es wird ein gleiches Volumen Tetrachlorkohlenstoff zugegeben und diese Lösung auf eine 60 X 600 mm-Aluminiumoxydsäule (Merck 71707, etwa 1,6 kg) gegossen.
Die Säule wird mit einer Mischung aus gleichen Teilen Chloroform und Tetrachlorkohlenstoff bei einer Strömungsgeschwindigkeit von 30 ml pro Minute eluiert. Nach Abnahme von etwa 3 Liter einer gelben Lösung wird das Eluat fast farblos, wobei am oberen Teil der Säule eine dunkelbraune Zone zurückbleibt. Durch Einengen des gelben Eluats im Vakuum werden 100 g (58 %) der gewünschten Nitroverbindung in Form eines gelben Öls erhalten.
C. Äthyl-6,7-diisopropoxy-4-hydroxychinolin-3 carboxylat
Eine Portion von 20 g (0,084 Mol) 1,2-Diisopropoxy-4-nitrobenzol wird über 2 g 10% dem Palladium-auf-Kohle als Katalysator in 200 ml Alkohol bei einem Anfangsdruck von 3,85 kg/cm2 hydriert.
Die Reduktion ist nach einem Druckabfall von 1,62 kg/ cm2 (87 S der Theorie) in 30 Minuten beendet.
Nach Abfiltrieren des Katalysators werden 18 g (0, 084 Mol) Diäthyläthoxymethylenmalonat zugegeben und die Lösung 2 Stunden zum Rückfluss erhitzt. Der nach Abdestillieren des Alkohols im Vakuum zurückbleibende bernsteinfarbene ölige Rückstand wird zu 900 ml Dowtherm A bei etwa 2450 C zugegeben. Diese Lösung wird 30 Minuten gekocht und dann über Nacht abkühlen gelassen. Das Rohprodukt wird abfiltriert, erst mit Dotherm , dann mit Benzol gewaschen und bei 800 C getrocknet. Es werden 9,7 g (35 S0) der gewünschten Verbindung in Form von hellbraunen Kri- stallen erhalten; Fp. 220-2280 C. Durch Umkristallisieren aus Dimethylformamid werden 6,4 g weisse Kristalle, Fp. 233-2340 C, erhalten.

Beispiel 5 Methyl- 6 ,7-diisopropoxy-4-hydroxychinolin-3 carboxylat 25 g (0,11 Mol) 1 2-Diisopropoxy-4-nitrobenzol (Beispiel 4, Teil B) werden über 4 g 5 %igem Palladium-auf-Kohle als Katalysator, der 50 % Wasser enthielt, in 200 ml Alkohol bei einem Anfangsdruck von 3,85 kg/cm2 hydriert. Die Reaktion ist nach einem Druckabfall von 1,62 kg/cm2 (87 % der Theorie) in 30 Minuten beendet. Der Katalysator wird abfiltriert, und die vereinigten Filtrate aus 2 derartigen Reduktionen werden mit 38 g (0,22 Mol) Dimethylmethoxymethylenmalonat in 200 ml Alkohol versetzt.
Diese Lösung wird 1,25 Stunden zum Rückfluss erhitzt und dann im Vakuum zu einem zähen Rückstand eingeengt.
Dieser Rückstand wird auf dem Dampfbad erhitzt und zu 1400 ml Dowtherm A zugegeben, die vorgängig auf 2450 C erhitzt worden waren. Die Lösung wird 20 Minuten gekocht und dann abkühlen gelassen. Das Rohprodukt wird abfiltriert, erst mit Dowtherm > , dann mit Aceton gewaschen und an der Luft getrocknet. Es werden 25 g (40 %) der gewünschten Verbindung erhalten; Fp. 242520 C. Durch Umkristallisieren von 52 g dieses Produktes aus 550 ml Dimethylformamid werden 43 g weisse Kristalle, Fp. 253-256 C, erhalten.

Coccidiosis, a widespread and economically very disadvantageous poultry disease, is caused by various coccidia including Eimeria tenella, Eimeria acervulina and Eimeria necatrix.

Ethyl 6,7-dipropoxy-4-hydroxyquinoline-3-carboxylate

Example 3
A. 0.24 mol of an ester obtained according to Example 1 or 2 and 300 ml of a 1ORigen sodium hydroxide solution are placed in a 1 liter flask. The mixture is refluxed for 2.5 hours.
