Verfahren zur Herstellung neuer Hydroxylamin-Verbindungen hs ist t) ekannt, ctaii man aus noneren und metteren Pflanzen, insbesondere Actinomyceten, eisenhaltige Wuchsstoffe, die Ferrioxamine, gewinnen kann. Von diesen wurde bisher Ferrioxamin B am eingehensten untersucht, ciocn war seine btruKtur nocn unMKannt (vgl. H. Bickel et al., Experientia 16, 129 ff. [1960]).
Es wurde nun gefunden, dass dem Ferrioxamin B die folgende Strukturformel zukommt :

Die Ferrioxamine stellen demnach Eisenkomplexe von Aminopentyl-hydroxycarbamyl-propionyl-amino- pentyl-hydroxycarbamyl-propionyl-aminopentyl-hydroxyl- aminen dar.
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von neuen Hydroxylaminverbindungen der Formel

worin Ri eine Schutzgruppe und R2 einen, Acylrest bedeuten, und ihrer Salze, gekennzeichnet durch die Verknüpfung von Bernsteinsäure mit dem Hydroxylamin der Formel II HN-(CH2) 5-NHOH, II unter Schutz von primären Aminogruppen, die nach erfolgter Bildung der entsprechenden Amidbindungen mit Ausnahme einer die N-Aminogruppe bildenden Gruppe freigesetzt werden, oder unter Schutz von Hydroxylaminogruppen in Form der Nitrogruppen, die nach erfolgter Bildung der entsprechenden Amidbindungen zu den Hydroxylaminogruppen reduziert werden und durch Acylierung der Hydroxylaminogruppe in 28 Stellung.
Die Schutzgruppe Ri ist in erster Linie ein Acylrest. Wenn Verbindungen der Formel I erwünscht sind, worin Ri für Wasserstoff steht, verwendet man als Schutzgruppe Ri eine abspaltbare Gruppe, insbesondere Carbobenzoxy, Phthaloyl, Trifluoracetyl, vor allem die tert.-Butyloxycarbonylgruppe, weiter z. B. den Toxyloder Tritylrest. Diese Schutzgruppen können in bekannter Weise durch Behandlung mit hydrolysierenden oder hydrogenolysierenden Mitteln abgespalten werden.
Unter Acylresten sind z. B. Alkanoylreste, insbesondere niedere Alkanoylreste, wie Formyl, Propionyl, Butyryl, vor allem Acetyl, ferner Aroyl-und Aralkanoylreste, beispielsweise Benzoyl, Naphthoyl, Phthaloyl, oder Phenylacetyl, zu verstehen. Als Metalle kommen vor allem solche von physiologischer Bedeutung, insbesondere Eisen, Kobalt, Magnesium, ferner auch Kupfer und Antimon, in Betracht.
Auf Grund ihrer Eigenschaften mit Metallen, z. B. den obengenannten, sehr stabile Komplexe zu bilden, besitzen die Verbindungen der Formel I wertvolle pharmakologische Eigenschaften. So verhindern sie z. B. die Ablagerung eisenhaltiger Pigmente im Ge webe bzw. bewirken sie in Fällen von Eisenablacening im Organismus eine Ausscheidung des Eisens, z. B. bei Hämochromatose und Hämosiderose sowie auch bei Lebercirrhose. Sie können auch zur Ausscheidung anderer Metalle aus dem Organismus verwendet werden, z. B. von Kupfer. Die Metallkomplexe dieser Verbinw dungen, in erster Linie die Eisen und Kobaltkomplexe, besitzen antianämische Eigenschaften und können dementsprechend als Heilmittel, beispielsweise bei Entblutungsanämie und Eisenmangelanämie, verwendet werden.
Auch als Ausgangs-oder Zwischenprodukte für die Herstellung anderer wertvoller Stoffe können die obengenannten Verbindungen dienen.
Die Herstellung des Ferrioxamins B beispielsweise ist aus folgendem Schema ersichtlich :

Die Überführung der Hydroxylaminverbindung in den Metallkomplex erfolgt durch Umsetzung der ersten mit einem entsprechenden Metallsalz, beispielsweise dem Salz einer Mineralsäure, wie einem Metallchlorid -sulfat oder-nitrat, oder dem Salz einer organischen Säure, wie einem Metallacetat oder-sulfonat, oder durch Umsetzung der ersten mit Metallalkoholaten, beispielsweise Metalläthylaten.
Die metallfreie Hydroxylaminverbindung wird nach den für die Herstellung von Säureamiden bekannten Methoden bei der Kondensation der obengenannten 5 Reste erhalten. So geht man beispielsweise von einem Pentan der Formel Rl-NH-CH ;-CHy-CHy-CH-CH-Z, worin Ri eine Acylgruppe oder der 2, 4-Dinitrophenyl- rest und Z. eine Hydroxylaminogruppe oder eine Nitrogruppe ist, und einem reaktionsfähigen Bernsteinsäure- derivat aus, wobei die Nitrogruppe vor der weiteren Ankondensation eines Bernsteinsäurerestes zur Hydroxylaminogruppe reduziert und die Aminogruppe vor der Ankondensation eines Bernsteinsäurerestes am Aminoende in Freiheit gesetzt wird.
Reaktionsfähige Derivate der Bernsteinsäure sind z. B. deren Hal'ogenide, Anhydride, Azide, Imide, Imidazolide und Ester, wie auch Lactone.
Falls die Kondensation an der freien Carboxylgruppe durchgeführt wird, bedient man sich vorzugsweise eines Kondensationsmittels.
Eine an der Kondensation nicht beteiligte freie Carboxylgruppe wird zweckmässig durch Veresterung, z. B. mit Methanol, Benzylalkohol, p-Nitrobenzylalkohol, geschützt.
Statt mit Bernsteinsäure oder ihren Derivaten kann man das genannte disubstituierte Pentan auch mit einem Kondensationsprodukt der Bernsteinsäure mit einem oder mehreren der weiteren die Gesamtkette zusam mensetzenden Reste kondensieren.
So kann man beispielsweise gemäss nachfolgendem Schema l-AcyIamino-5-nitro-pentan (Ia) zu 1-Acyl amino-5-hydroxylamino-pentan (Ib) reduzieren, dieses mit Bernsteinsäureanhydrid zu II kondensieren, dieses mit l-Amino-5-nitropentan zum 1- (Acylaminopentylhydroxycarbamyl-propionylamino)-5-nitro-pentan (III) verknüpfen, die Nitrogruppe zur Hydroxylaminogruppe reduzieren, die so erhaltene Verbindung (lIIa) entweder direkt mit 1- (Succinylamino)-5-acylhydroxylamino- pentan, vorzugsweise in Gegenwart eines Kondensationsmittels zu V kondensieren oder sie zunächst mit Bern- steinsäureanhydrid zu der Verbindung IV umsetzen und diese dann mit 1-Amino-5-acylhydroxylamino-pentan kondensieren :

Falls eine an der Hydroxylaminogruppe nicht acylierte Verbindung gewünscht wird, kann die Verbindung IIIa auch mit l-(Succinylamino)-5-nitro-pentan kondensiert und dann z. B. mit Zink und Säure reduziert werden.
Statt bei dem beschriebenen Verfahren das l-Acyl- amino-5-hydroxylamino-pentan zunächst mit Bernstein- säureanhydrid und dann mit l-Amino-5-nitro-pentan zu kondensieren, kann man es auch in einer einzigen Stufe mit l-(Succinylamino)-5-nitro-pentan zu III umsetzen.
Nach einer weiteren Ausführungsform des Verfahrens gemäss nachfolgendem Schema kann man beispielsweise l-Amino-5-nitro-pentan (I) mit Bernsteinsäure-anhydrid zum l-(Succinylamino)-5-nitro-pentan (IIa) kondensieren, dieses zum 1- (Succinylamino)-5-hydroxylamino- pentan (IIb) reduzieren, die Carboxylgruppe verestern, die so erhaltene Verbindung (IIc) mit der Verbindung IId, die, man durch Umsetzung von IIb beispielsweise mit Acetylchlorid oder Keten erhält, zu IVa kondensieren, die Ester-und O-Acylgruppe verseifen und die erhaltene Säure mit 1-Acylamino-5-hydroxylamino- pentan, vorzugsweise in Gegenwart eines Kondensations- mittels, zu Va kondensieren :

Je nach der Arbeitsweise erhält man die Verbindungen in Form von Basen oder ihren Salzen. Aus den Salzen können in an sich bekannter Weise die Basen gewonnen werden. Von letzteren wiederum lassen sich durch Umsetzung mit Säuren, die zur Bildung therapeu- tisch verwendbarer Salze geeignet sind, Salze gewinnen, wie z. Die verfahrensgemäss erhaltenen Komplex-Verbindungen. können in Form von pharmazeutischen Präpa- raten Verwendung finden. Diese enthalten die Verbindungen in Mischung mit einem für die enterale oder parenterale. Applikation geeigneten pharmazeutischen organischen oder anorganischen Trägermaterial.
In den nachfolgenden Beispielen sind die Temperaturen in Celsiusgraden angegeben.

13, 2 g 1-Amino-5-nitro-pentan (I) [hergestellt nach H. Bickel et al., Helv. Chim. Acta 43, 904 (1960)] werden in 1 Liter Aceton aufgenommen und mit 1 Liter 0, 1 n Natriumbicarbonatlösung versetzt. Hierauf gibt man unter Eiskühlung und kräftigem Rühren 17, 0 g Carbobenzoxychlorid, in 200 ml wasserfreiem Tetrahydrofuran gelöst, langsam zu und lässt 7 Stunden bei 0-'stehen. Nach beendeter Reaktion wird im Vakuum die Hauptmenge des Acetons abgedampft und die verbliebene wässrige Lösung fünfmal mit je. 100 ml Chloroform ausgeschüttelt.
Die mit wenig 2n Salzsäure, ge sättigter Natriumbicarbonatlösung und Wasser gewaschenen und über Natriumsulfat getrockneten Chloroformauszüge geben beim Eindampfen 25, 8 g farblose Substanz der Formel II vom F. 53 .

24, 0 g Substanz II werden in 400 ml feuchtem 1, 2 Dimethoxy-äthan gelyst und zusammen mit 60 g frisch bereittem Aluminiumamalgam 4 Stunden bei 0 ge- rüh, rt. 16 g Substanz III werden in 150 ml absolutem Pyridin gelöst, mit 6, 4 g Bernsteinsäureanhydrid versetzt und 15 Stunden bei 22 unter Stickstoffatmosphäre stehengelassen. Anschliessend dampft man das Pyridin ; im Vakuum ab. Der Rückstand IV (20, 5 g) wird aus Athanol-Diäthyläther umkristallisiert, F. 115-116 .
Durch Erwärmen mit Dicyclohexylcarbodiimid erhält man die Verbindung V, die nach dem Kristallisieren aus Diäthyläther bei 84-85 schmilzt.
In analoger Weise erhält man aus 1-Benzoylamino- 5-hydroxylamino-pentan das 1-Benzoylamino-5-(N- succinyl-hydroxylamino)-pentan vom F. 114-115 , eine in Athanol und Essigsäureäthylester leicht, in Diäthyl- äther schwer lösliche Verbindung, und daraus das N- (5' Benzoylamino-pentyl)-tetrahydro-3,6-dioxo-1, 2-oxazin vom F. 104-106 .

20, 0 g Substanz V werden zusammen mit 8, 0 g 5-Nitro-l-amino-pentan in 300 ml Tetrahydrofuran gelöst und unter Stickstoffatmosphäre 21/2 Stunden zum Sieden erhitzt. Nach dem Eindampfen im Vakuum erhält man 27, 1 g rohe Verbindung VI. Diese wird an 1 kg Silicagel chromatographiert. Man leitet einen langsamen Strom von Keten während einer Stunde in eine eisgekühlte Lösung von 3, 7 g Verbindung IX in 200 ml Tetrahydrofuran. Hierauf wird im Vakuum eingedampft, in 200 ml Essigsäure- äthylester aufgenommen und mit wenig 2n Salzsäure, 2n Natriumcarbonat und Wasser gewaschen.

Das in Absatz g) beschriebene Produkt IXa kann wie unter h) beschrieben weiter acetyliert und gemäss i) und k) decarbobenzoxyliert und desacetyliert werden.
Man erhält so das 30- (-Carboxyäthy !)-7, 18, 29-trihy- droxy-8, 11, 19, 22, 30-pentaoxo-1, 7, 12, 18, 23, 29-hexaazatriacontan.

280 mg Substanz XII werden in 30 ml 0, ln Na tronlauge gelöst, mit 350 mg Benzoylchlorid versetzt und 1/¸ Stunden bet 0 geschüttelt. 500 mg Substanz XII werden in 100 ml Wasser aufgenommen und mit 400 mg kristallwasserhaltigem Eisen-(III)-chlorid (FeCl3 # 6H2O) in 20 ml Wasser versetzt, wobei das Reaktionsgemisch sofort die für Ferrioxamin typische Farbe annimmt. Nach 15 Minuten Stehen schüttelt man dreimal mit je 20 ml Chloroform-Phenol (1 : 1) aus, wäscht die Extrakte mit wenig Wasser und versetzt sie mit 200 ml Diäthyläther,
Ferrioxamin-B-hydrochlorid worauf das Ferrioxamin-B-hydrochlorid (XIII) als amorpher, rotbrauner Niederschlag ausfällt.
Das mehr- mals mit Diäthyläther gewaschene Präparat stimmt in folgenden Eigenschaften mit authentischem Material überein : Absorptionsspektren iim UV, Sichtbaren und IR ; Papierchromatogramm, Elektrophorese, Antagonismus-Test mit Ferrimycinen sowie N- (2, 4-Dinitrophenyl)- Derivat [vgl. o)].

Eine Lösung von 225 mg Substanz XIII und 225 mg Natriumbicarbonat in 15 ml Wasser wird mit 225 mg 2, 4-Dinitro-fluorbenzol in 15 ml Athanol versetzt und 5 Stunden bei 23 ste, hengelassen. Anschliessend destilliert man das Athanol im Vakuum ab, extrahiert die wässrige Phase zweimal mit je 30 ml Diäthyläther und schliesslich viermal mit je 30 ml n-Butanol. Die mit Wasser gewaschenen und im Vakuum eingedampften Butanolauszüge enthalten den rotbraunen Komplex. Er wird in Aceton aufgenommen, die Lösung von Unlös- lichem abfiltriert und das Filtrat mit Diäthyläther versetzt.
Man erhält 176 mg amorphes, oranges Pulver (XIV), das nach dreimal wiederholter Fällung und Trocknung zur Analyse bei 70 und 0, 01 Torr über Phosphorpentoxyd folgende Zusammensetzung aufweist : C31h4@O12N8Fe

Auch nach UV-Absorptionsspektrum ist Produkt XIV identisch mit dem authentischen De, rivat von Ferri- oxamin B.
Beispiel 2 (Die in diesem Beispiel mit römischen Ziffern bezeichneten Produkte sind mit den in Beispiel 1 formel- mässig angegebenen identisch.) a) I # II
Zu einer Mischung von 4, 1 g 1-Amino-5-nitro- pentan-hydrochlorid, 20 ml Wasser und 1, 43 g Kaliumhydroxyd lässt man unter Rühren bei 0 4, 5 g Carbobenzoxychlorid und eine Lösung von 2 g Kaliumhydroxyd in 10 ml Wasser zutropfen, rührt nachher noch 10 Minuten weiter, säuert mit Salzsäure schwach an und extrahiert das Reaktionsgemisch mit Chloroform.
Der Chloroformextrakt wird mit Natriumbicarbonatlösung gewaschen, über Natriumsulfat getrocknet und im Vakuum eingedampft. Der Rückstand wird aus Di äthyläther-Petroläther kristallisiert, und man erhält das 1-Carbobenzoxyamino-5-nitra-pentan (II) vom F. 52 bis 53 ; Ausbeute 85%. b) II-, III
Eine Lösung von 2 g Verbindung II in 40 ml Athanol versetzt man mit einer Lösung von 3 g Ammo- niumchlorid in 30 ml Wasser und 15 Tropfen konzentriertem wässrigem Ammoniak und fügt innerhalb 5-10 Minuten unter Rühren 1, 5 g Zinkstaub zu.
Man rührt noch 15 Minuten weiter und achtet darauf, dass die Temperatur 30 nicht übersteigt. Das Reaktionsgemisch wird hierauf durch Celite filtriert, das Filtrat im Vakuum eingedampft, der Rückstand in 50 ml 2n Salzsäure gelöst und die Lösung mit Ather ausgeschüttelt.
Die wässrige Phase alkalisiert man sodann mit wässrigem Ammoniak, extrahiert sie mit Chloroform und engt den getrockneten Chlorformextrakt ein. Beim Versetzen des Rückstandes mit Ather kristallisiert das gebildete 1-Carbobenzoxyamino-5-hydroxylamino-pentan (III) aus ; F. 102-104 . Sein Hydrochlorid schmilzt bei 83 bis 85 ; Ausbeute 71 %. c) III IV
Die Mischung von 1, 85 g Verbindung III, 20 ml Pyridin und 1, 1 g Bernsto ! insäureanhydrid erwärmt man 11/2 Stunden auf 85 , dampft sodann das Pyridin im Vakuum ab und nimmt den Rückstand in Chloroform auf.
Die Chloroformlösung schüttelt man mit Kaliumbicarbonatlösung durch, säuert die letzte hierauf mit Salzsäure schwach an und extrahiert sie viermal mit Essigsäureäthylester. Nach dem Eindampfen des getrockneten Extraktes erhält man das 1-Carbobenzoxy- amino-5- (N-succinyl-hydroxylamino)-pentan (IV), welches nach dem Umlösen aus Essigsäureäthylester Methanol bei 115 schmilzt ; Ausbeute 70%. d) IV-. Die Mischung aus 2, 865 g Verbindung IV, 1, 88 g Dicyclohexyl-carbodiimid und 50 ml absolutem Tetra- hydrofuran lässt man 2 Tage bei Zimmertemperatur stehen, filtriert vom ausgefallenen Dicyclohexylharnstoff ab und dampft das Filtrat im Vakuum ein.
Den Rück- stand nimmt man in Essigsäureäthylester auf und lässt die Lösung im Kühlschrank stehen. Nach dem Abfiltrieren von wenig ausgefallenem Dicyclohexylharnstoff versetzt man das. Filtrat mit Äther, worauf sich das gebildete N- (5'-Carbobenzoxyamino-pentyl)-tetrahydro-3, 6 dioxo-1, 2-oxazin (V) in farblosen Nadeln abscheidet ; F. 84-85 , Ausbeute 70%.
Führt man die unter a-d angegebenen Umsetzungen ohne jeweilige Separation und Reinigung der erhaltenen Zwischenprodukte durch, so erhält man die Substanz V in einer Totalausbeute von 51 %. e) V o VI
Eine Lösung von 1, 2 g Verbindung V und 1 g l-Amino-5-nitro-pentan-hydrochlorid in absolutem Methanol versetzt man mit einer r Losung von 0, 32 g Natriummethylat in Methanol. Nach halbstündigem Erwärmen der Mischung auf ungefähr 50 filtriert man vom abgeschiedenen Natriumchlorid ab und dampft das Filtrat im Vakuum ein.
Der Rückstand wird aus Methanol-Essigsäureäthylester kristallisiert, und man erhält das l-Carbobenzoxyamino-5-[N-(5'-nitro-pentylw amino-succinyl)-hydroxylamino]-pentan (VI) vom F. 114 bis 116 ; Ausbeute 88%. f) VI-. VII
Die Lösung von 0,85 g Verbindung VI in 20 ml Äthanol versetzt man unter Rühren mit 15 ml 10%iger und mit einigen Tropfen wässrigem Ammoniak alkalisierter Ammoniumchloridlösung und 0, 5 g Zinkstaub, rührt 20 Minuten weiter, filtriert und dampft das Filtrat im Vakuum ein. Den Rückstand nimmt man in Äthanol auf, filtriert neuerlich und dampft das Filtrat zur Trockne ein.
Das erhaltene 1-Carbobenzoxyamino-5- [N- (5'-hydroxylamino-pentylamino-succinyl)-hydroxyl-amino]-pentan (VII) kann ohne Reinigung weiter verarbeitet werden.

Der nach f) erhaltene Rückstand wird in 10 ml absolutem Pyridin und 2 ml Essigsäureanhydrid gelöst und die Lösung 1 Tag bei Zimmertemperatur stehengelassen. Der nach dem Eindampfen erhaltene farblose Rückstand wird in Essigsäureäthylester aufgenommen und an Silicagel chromatographiert. Man eluiert mit Essigsäureäthylester-Methanol 1 : 1 und hydrogenolysiert die Hauptfraktion wie unter i) angegeben wurde. Die vom Katalysator befreite Lö- sung dampft man hierauf ein.

Der nach k) erhaltene Rückstand wird in Wasser aufgenommen, die Lösung durch Celite klar filtriert, das Filtrat mit eine, r Eisen-(III)-chloridlysung versetzt und die rotbraun gefärbte Mischung nach einstündigem Stehen dreimal mit Chloroform-Phenol 1 : 1 extrahiert.
Die vereinigten Extrakte wäscht man mit wenig Wasser, versetzt sie mit Diäthyläther, filtriert den ausgefallenen rotbraunen Niederschlag ab und wäscht ihn mit Di äthyläther. Das so erhaltene Ferrioxamin-B-hydrochlorid (XIII) ist mit authentischem Material identisch ; Totalausbeute der Stufen i-1 ungefähr 50%.

where Ri is a protective group and R2 is an acyl radical, and its salts, characterized by the linkage of succinic acid with the hydroxylamine of the formula II HN- (CH2) 5-NHOH, II under protection of primary amino groups, which after formation of the corresponding amide bonds with the exception of a group forming the N-amino group, or with the protection of hydroxylamino groups in the form of nitro groups, which are reduced to the hydroxylamino groups after formation of the corresponding amide bonds and by acylation of the hydroxylamino group in position 28.
The protective group Ri is primarily an acyl radical. If compounds of the formula I are desired in which Ri is hydrogen, the protective group Ri used is a removable group, in particular carbobenzoxy, phthaloyl, trifluoroacetyl, especially the tert-butyloxycarbonyl group, further z. B. the toxyl or trityl residue. These protective groups can be split off in a known manner by treatment with hydrolyzing or hydrogenolyzing agents.
Acyl residues include, for. B. alkanoyl radicals, in particular lower alkanoyl radicals, such as formyl, propionyl, butyryl, especially acetyl, also aroyl and aralkanoyl radicals, for example benzoyl, naphthoyl, phthaloyl, or phenylacetyl. Metals which are particularly suitable are those of physiological importance, in particular iron, cobalt, magnesium, and also copper and antimony.
Due to their properties with metals, e.g. B. to form the above-mentioned very stable complexes, the compounds of formula I have valuable pharmacological properties. So they prevent z. B. the deposition of ferrous pigments in Ge tissue or they cause in cases of iron ablacening in the organism an excretion of iron, z. B. in hemochromatosis and hemosiderosis as well as in liver cirrhosis. They can also be used to eliminate other metals from the body, e.g. B. of copper. The metal complexes of these compounds, primarily the iron and cobalt complexes, have anti-anemic properties and can accordingly be used as remedies, for example for bleeding anemia and iron deficiency anemia.
The abovementioned compounds can also serve as starting materials or intermediates for the production of other valuable substances.

The hydroxylamine compound is converted into the metal complex by reacting the first with a corresponding metal salt, for example the salt of a mineral acid, such as a metal chloride sulfate or nitrate, or the salt of an organic acid, such as a metal acetate or metal sulfonate, or by reacting the first with metal alcoholates, for example metal ethylates.
The metal-free hydroxylamine compound is obtained by the methods known for the preparation of acid amides in the condensation of the above-mentioned 5 radicals. For example, one starts from a pentane of the formula Rl-NH-CH; -CHy-CHy-CH-CH-Z, where Ri is an acyl group or the 2,4-dinitrophenyl radical and Z is a hydroxylamino group or a nitro group, and a reactive succinic acid derivative, the nitro group being reduced to the hydroxylamino group prior to the further condensation of a succinic acid residue and the amino group being set free prior to the condensation of a succinic acid residue at the amino end.
Reactive derivatives of succinic acid are z. B. their halides, anhydrides, azides, imides, imidazolides and esters, as well as lactones.
If the condensation is carried out on the free carboxyl group, a condensing agent is preferably used.
A free carboxyl group which is not involved in the condensation is expediently removed by esterification, e.g. B. with methanol, benzyl alcohol, p-nitrobenzyl alcohol, protected.
Instead of using succinic acid or its derivatives, said disubstituted pentane can also be condensed with a condensation product of succinic acid with one or more of the other radicals composing the entire chain.

The mixture is stirred for a further 15 minutes, making sure that the temperature does not exceed 30. The reaction mixture is then filtered through Celite, the filtrate is evaporated in vacuo, the residue is dissolved in 50 ml of 2N hydrochloric acid and the solution is extracted by shaking with ether.
The aqueous phase is then made alkaline with aqueous ammonia, extracted with chloroform and the dried chloroform extract is concentrated. When ether is added to the residue, the 1-carbobenzoxyamino-5-hydroxylamino-pentane (III) formed crystallizes out; F. 102-104. Its hydrochloride melts at 83 to 85; Yield 71%. c) III IV

A solution of 1.2 g of compound V and 1 g of 1-amino-5-nitro-pentane hydrochloride in absolute methanol is treated with a solution of 0.32 g of sodium methylate in methanol.
