Verfahren zur Herstellung von Dibenzo-cycloheptenen
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von 5H-Dibenzo [a, d] cycloheptenen und 5H- Dibenzo [a, d] 10, 11-dihydrocycloheptenen, welche in 5 Stellung mit einem Aminopropylidenradikal substituiert sind.
Die erfindungsgemäss erzeugten Verbindungen sind bei der Behandlung von geistigem Unwohlsein nützlich, da sie sich durch Antidepressant-Eigenschaften auszeichnen und zur Erhöhung der Gemütsverfassung oder zur seelischen Kräftigung verwendet werden können. Die zu diesem Zwecke verabreichte tägliche Dosis bewegt sich innerhalb 5 mg bis 250 mg, vorteilhafterweise in kleinern, innerhalb eines Tages verteilter Mengen. Diese Verbindungen werden zweckmässigerweise in Form ihrer sauren Salze verabreicht. Die Herstellung dieser sauren Salze wird gleichfalls von der vorliegenden Erfindung umfasst.
Die erfindungsgemäss erzeugten Verbindungen kön- nen durch die folgenden Formeln veranschaulicht werden :

R und R'stehen für Wasserstoff, Niederalkyl, Niederalkenyl, Cycloniederalkyl, Phenyl oder Benzyl oder bilden zusammen mit dem Stickstoffatom einen heterocyclischen Ring, der als weitere Heteroatome Stickstoff oder Sauerstoff aufweisen kann.
Das erfindungsgemässe Verfahren kann schematisch durch das folgende Fliessschema veranschaulicht werden, in welchem die punktierte Linie anzeigt, dass die Verbindung in den 10, 11-Stellungen gesättigt (10, 11 Dihydro) oder ungesättigt sein kann und wobei X, X', R und R'dieselbe Bedeutung haben wie oben angeführt.

Das erfindungsgemässe Verfahren geht von bekannten Ketonen aus, welche mittels eines in der Literatur beschriebenen Verfahrens hergestellt werden können.
Wie aus dem oben angeführten Fliessschema zu ersehen ist, beruht die erste Stufe des erfindungsgemässen Verfahren in der Kondensation eines 5H-Dibenzo [a, d] cyclohepten-5-ons mit einem Grignard-Reagens, welches vom Cyclopropylhalogenid abgeleitet wird ; bei dieser Kondensation bildet sich das entsprechende Cyclopropylcarbinol, z. B. das 5-Cyclopropyl-5-oxy-5H-dibenzo [ad] 10, 11-dihydrocyclohepten. Diese Reaktion wird auf typische Weise so durchgeführt, dal3 man Cyclopropyl bromid in Tetrahydrofurantropfenweise und ùnterRühren zu einer bestimmten Menge Magnesium in trockenem Tetrahydrofuran zusetzt, wobei sich das Cyclopropyl magnesiumbromid bildet.
Dann wird das Keton zum Cyclopropylmagnesiumbromid zugesetzt und das Reak- tionsgemisch wird während einer zweckmässigen Zeitspanne bei Rückfluss erhitzt, wonach das Cyclopropylcarbinol-Zwischenprodukt erhalten wird.
Die nächste Stufe des erfindungsgemässen Verfahrens umfasst die Umlagerung des Cyclopropylcarbinols zum entsprechenden y-Oxy-propylidencyclohepten, z. B. 5 (y-oxypropylíden)-5H-dibenzo [a, d] 10, 11-dihydrocyclo- hepten. Das y-Oxypropylidencyclohepten wird dann mit einem Halogenierungsmittel behandelt und liefert das entsprechende y-Halopropylidencyclohepten-Derivat, z. B. 5- (y-Bromopropyliden)-5H-dibenzo [a, d] 10, 11-dihy- drocyclohepten. Die oben beschriebenen Umlagerungs- und Halogenierungsstufen können gleichzeitig oder jede fur sich durchgeführt werden.
Durch Behandlung des Cyclopropylcarbinols mit einem Hydrohalogenid in Essigsäurelösung kann das entsprechende,-Halopro- pylidencyclohepten direkt in einer Stufe erhalten werden. Entsprechende Jod-Derivate, falls solche gewünscht werden, können aus dem Chlor-oder Brom-Derivat, durch Reaktion des letztern mit Natriumjodid in Aceton unter Rückfluss, gebildet werden.
Anderseits kann das y-Oxypropylidencycloheptan- Derivat als solches, durch Behandlung des Cyclopropylcarbinols mit einer Säure, welche eine andere als eine Halogensäure ist, isoliert werden. Zu diesem Zwecke eignet sich am besten Perchlorsäure in Dioxan, obschon auch andere Säuren wie Schwefelsäure, Phosphorsäure und p-Toluolsulfonsäure benützt werden kön- nen. Danach kann die y-Oxyverbindung in das entspre chende,-Halopropylidencyclohepten, durch Reaktion mit einem geeigneten Halogenierungsmittel, konvertiert werden ; zu diesen Zwecke eignet sich am besten Thionylchlorid oder Phosphortrichlorid, obschon andere Halogenierungsmittel ebenfalls verwendet werden können.
Schliesslich wird die so erhaltene y-Halogenpropylidencyclohepten-Verbindung in das gewünschte Aminopropyliden-Derivat durch ein Aminierungsverfahren konvertiert. Die Aminierung kann durch direkte Reaktion der Halogenverbindung mit einem Amin erfolgen, wobei die gewünschte Aminopropyliden-Verbindung anschliessend als Hydrochlorid isoliert wird. In einer andern Alternative kann das gewünschte Amin-Derivat durch eine Aminierungsreaktion, unter Verwendung von Kaliumphthalimid, und nachfolgender Umsetzung mit Hydrazin hergestellt werden.
Beispiel 1
5-Cyclopropyl-5-oxy-SH-dibenzo [a, d]
10, 11-dihydrocyclohepten
Eine 1 25-ml-Dreihalsflasche welche unter trockenem Stickstoff gekühlt wurde und welche mit einem Rührer, einem zusätzlichen Trichter und einem Konden- sator vom Ather-Typus versehen war, wurde mit 1, 7 g, das sind 0, 070 Mole, reinen Magnesium-Drehspänen und mit 15 ml trockenem Tetrahydrofuran (THF) gefüllt. Danach wurden 8, 5 g, das sind 0, 0702 Mole, Cyclopropylbromid in 15 ml trockenem THF tropfenweise und unter Rühren und bei einer genügenden Geschwindigkeit, um einen milden Rückfluss aufrecht- zuerhalten, zugegeben.
Ein leichtes Erwärmen und Rüh- ren während etwa 30 Minuten ist notwendig, um die Reaktion anlaufen zu lassen, wonach keine äussere Wärme mehr notwendig ist. Das Rühren und die Behandlung bei Rückfluss wird so lange fortgesetzt, bis sämt- liches Metall verschwunden ist. Das Reaktionsgemisch wird dann unterhalb dem Rückflusspunkt gekühlt, aber nicht so stark, um ein Ausfällen des Grignard-Reagens zu verursachen, und es werden 7, 3 g, das sind 0, 0351 Mole, 5H - Dibenzo [a, d] 10, 11-dihydrocyclohepten-5-on in 20 ml trockenem THF unter Rühren innerhalb von
15 Minuten zugegeben.
Das Reaktionsgemisch wird gerührt und während 6 Stunden bei Rückfluss behandelt, während welchen, zwecks Verfolgung der Reaktion, jedesmal 0, 5 ml dem Kolben entzogen und jede Stunde mit Hilfe einer Dünnschichtchromatographie (DSC) aufgearbeitet wurden. Diese DSC zeigt an, ob die Reaktion innerhalb einer Stunde beendet ist, in welchem Zustand alle entnommenen Proben spontan nach Stehenlassen kristallisieren. Nach diesem 6stündigen Reaktionsver- lauf wird das Reaktionsgemisch in einem Eisbad abgeschreckt und mit 45 ml gesättigter Ammoniumchlorid- lösung behandelt. Die Schichten wurden separiert und es wurde gerade so viel Wasser zugesetzt, um die festen Salze in der wässrigen Schicht zu lösen. Die letztere wurde dann mit zweimal 25 ml Ather extrahiert.
Die vereinigten organischen Schichten wurden mit 25 ml gesättigter Salzlösung gewaschen, über Magnesiumsulfat getrocknet und im Vakuum zur Trockne abgedampft, wobei 9, 88 g eines gelborange gefärbten Öls erhalten wurden. Nach Behandlung dieses Öls mit Aktivkohle in Ather und Kristallisieren aus Petroläther erhält man 5, 49 g, in 62, 5 % iger Ausbeute, rohes kristallines Cyclopropylcarbinol, welches sich in der DSC durch einen einzigen Fleck auszeichnet.
Ein weiteres Umkristallisieren liefert ein Material mit F = 72, 8-73, 8 C, A MeOH 2630, E 1 % cm 24. man Analyse, für CisHisO : berechnet : C 86, 36 H 7, 24 gefunden : C 86, 43 H 7, 40
Beispiel 2 5-Cyclopropyl-5-oxy-5H-dibenzo [a, d] cyclohepten
Gemäss des im einzelnen in Beispiel 1 beschriebenen Verfahrens und unter Verwendung gleichwertiger Mengen von 5H-Dibenzo [a, d] cyclohepten-5-on erhält man das entsprechende 5-Cyclopropyl-5-oxy-SH-dibenzo [a, d]cyclohepten.
Beispiel 3
5 (y-Bromopropyliden-SH-dibenzo [a, d] 10, 11- dihydrocyclohepten
1, 3 g, das sind 5, 19 Millimole, 5-Cyclopropyl-5oxy-SH-dibenzo [a, d] 10, 11-dihydro-cyclohepten werden in 20 ml Eisessigsäure in einer 100-ml-Flasche gelöst und die Lösung wird dann auf 10 C abgeschreckt. Danach wird die Lösung mit 10 ml einer 15 % igen Lösung von HBr in Essigsäure unter Rühren versetzt und das Reaktionsgemisch bei 10-15 C während 0, 5 Stunden gerührt. Nach Filtrieren in der Kälte des Reaktionsgemisches wird das kristalline Produkt an der Luft getrocknet und man erhält 730 ml, in 45 % iger Ausbeute, eines ersten Ertrages an Bromid mit F = 71, 0-71, 8 C.
Das Filtrat wird dann abgeschreckt und mit 40 ml Wasser verdünnt, wonach 880 mg eines zweiten, rohen Ertrages erhalten werden, welcher nach Umkristallisieren aus Petroläther, der eine kleine Menge Ather enthält, 780 mg an kristallinem Material mit F = 69, 2-71, 6 C ergibt. Das gesamte kristalline Produkt ergibt sich mit
1, 51 g, das ist in 92, 5 % iger Ausbeute, und welches Produkt die folgenden Eigenschaften aufweist : 2 Mlax H maux
2400, E. 1% cm 565; ? CHCL3 max 3,27, 3,35, 3,43, 6,1
6, 21 und 6, 34 u.

Beispiel 4 5- (y-Bromopropyliden-5H-dibenzo [a, d] cyclohepten
Unter Befolgung des im einzelnen in Beispiel 3 beschriebenen Verfahrens und unter Verwendung äquivalenter Mengen an 5-Cyclopropyl-5-oxy-5H-dibenzo- [a, d] cyclohepten gelangt man zum entsprechenden 5-(γ- Bromopropyliden-5H-dibenzo [a, d] cyclohepten.
Beispiel S 5- (y-Chloropropyliden)-5H-dibenzo [a, d] 10, 11- dihydro-cyclohepten
100 ml, das sind 0, 4 Millimole, 5-Cyclopropyl-5-oxy5H-dibenzo [a, d] 10, 11-dihydro-cyclohepten werden in 5 ml Eisessigsäure in einer 25-ml-Flasche, die einen magnetischen Rührer enthielt, gelöst und die Lösung wird dann auf 10 C gekühlt. Unter Rühren wird hernach 1 ml einer 15% igen HCl-Lösung in Essigsäure zugesetzt und das Reaktionsgemisch wird bei 10-15 C während 3 Stunden gerührt.
Es werden dann aliquote Mengen in bestimmten Zeitintervallen zur Untersuchung auf Dünnschicht-Chromatographieplättchen abgenommen, welche angibt, dal3 die Reaktion innerhalb von 5 Minuten beendet ist. Danach wird das Reaktionsgemisch zur Trockne im Vakuum bei Zimmertemperatur konzentriert und dreimal mit Benzol gespült. Der kristalline Rückstand wird dann mit Aktivkohle behandelt und aus Ather-Petroläther umkristallisiert und liefert ein kristallines Produkt mit den folgenden Eigenschaften : F = 83-84 C ; ;. 2400, E 1 % cm 516.

Beispiel 6 5- (y-Chloropropyliden-5H-dibenzo [a, d] cyclohepten
Unter Befolgung der im einzelnen in Beispiel 5 beschriebenen Verfahrensschritte und unter Verwendung äquivalenter Mengen an 5-Cyclopropyl-5-oxy-SH- dibenzo [a, d] cyclohepten gelangt man zum entsprechenden 5-(y-Chloropropyliden-SH-dibenzo [a, d] cyclohepten.
Beispiel 7 5- (y-Oxypropyliden-5H-dibenzo [a, d] 10, 11- dihydro-cyclohepten
500 mg 5-Cyclopropyl-5-oxy-SH-dibenzo [a, d] 10, 11- dihydro-cyclohepten in 15 ml Dioxan werden mit 9 ml 2m Perchlorsäure bei Zimmertemperatur während insgesamt 6 Stunden behandelt. Für DSC werden Proben nach 1, 3 und 6 Stunden entnommen und es wird un tersucht, ob die Reaktion bei 1 Stunde beendet ist und keinen weiteren Wechsel nach 6 Stunden ergibt.
Durch Aufarbeiten des Reaktionsgemisches erhält man ein rohes 01, welches nach Umkristallisieren 320 mg, das ist in 64 % iger Ausbeute, eines Alkohols mit den folgenden Charakteristiken ergibt : F = 89-90, 2 C ; A MeOH 2375, E 1% cm 5548 ; ? CHCl2 max 2, 73, 2, 9, 3, 25,
3, 31, 6, 2, 6, 34 und 9, 6, u.

Beispiel 8 5-(,-Oxypropyliden)-SH-dibenzo [a, d] cyclophepten
Gemäss dem im einzelnen in Beispiel 7 beschriebe nen Verfahren und unter Verwendung gleichwertiger Mengen von 5-Cyclopropyl-5-oxy-5H-dibenzo [a, d] cyclo- hepten erhält man das entsprechende ; 5-(y-Oxypropy- liden)-5H-dibenzo [a, d] cyclohepten.
Beispiel 9 5-(γ-Chloropropyliden-5H-dibenzo [a, d] 10, 11 dihydro-cyclohepten
Durch Behandlung einer Lösung von 50 mg von 5-(γ-Oxypropyliden-5H-dibenzo[a, d] 10, 11-dihydro-cyclo- hepten in 3 ml trockenem Benzol, welcher 1 Tropfen
Pyridin enthält, mit 65 mg Thionylchlorid in 2 ml trok kenem Benzol und unter Erhitzen bei Rückfluss während
3 Stunden auf einem Dampfbad erhält man ein kri stallines Produkt mit F = 78-79 C ; dieses erwies sich nach Behandlung mit DSC.und durch Vermischen mit einem Körper von F = 78-81 C als identisch mit einer
Verbindung, welche durch Behandlung von 5-Oxy-5- cyclopropyl-5H-dibenzo [a, d] 10, 11-dihydrocyclohepten mit HCl/Essigsäure erhalten wurde.
Beispiel 10 5-(γ-Chloropropyliden)-5H-dibenzo [a, d] cyclohepten
Gemäss dem im einzelnen in Beispiel 9 beschriebe nen Verfahren und unter Verwendung äquivalenter Men gen von 5- S-Oxypropyliden)-5H-dibenzo [a, d] cyclohep ten erhält man das entsprechende 5- (y-Chloropropy- liden-5H-dibenzo [a, d] cyclohepten.
Beispiel 11 5-( > -Dimethylaminopropyliden)-5H-dibenzo [a, d]
10, 11-dihydrocyclohepten-HC1
Eine Lösung von 100 mg, das sind 0, 372 Millimole, 5- ( ;-Chloropropyliden)-5H-dibenzo [a, d] 10, 11-dihydro- cyclohepten in 1 ml Benzol wird in einem dickwandi gen Pyrexrohr mit Dimethylamin bei 10 C gesättigt.
Dieses Rohr wird dann geschlossen und bei 95 C wäh- rend 18 Stunden stehengelassen ; dann wird es gekühlt und geöffnet. Die Benzollösung wird nacheinander mit 5 S iger Kaliumbicarbonatlösung, hernach mit Wasser und gesättigter Salzlösung gewaschen, über Magnesium sulfat getrocknet und zur Trockne im Vakuum abge dampft. Das zurückgebliebene 01 wird in Ather gelöst und mit mit HCl gesättigtem Ather behandelt.
Das er haltene Gemisch von 01 und Ather wird, zwecks Ent fernung des Überschusses an HCl, zur Trockne geblasen ; der Rückstand liefert nach Zerreiben mit Ather ein kristallines Material von 5- (y-Dimethyl-aminopro- pyliden)-5H-dibenzo [a, d] 10, 11-dihydrocyclohepten mit F = 190-192 C. A MPOH 2400, E 1 % cm 437.

Beispiel 12 5- (y-Dimethylaminopropyliden)-5H-dibenzo [a, d]- cyclohepten-HC1
Gemäss dem im einzelnen im Beispiel 11 beschriebenen Verfahren und unter Verwendung gleichwertiger Mengen von 5- (y-Chloropropyliden)-5H-dibenzo [a, d]- cyclohepten erhält man das entsprechende 5-(γ-Dimethyl- aminopropyliden)-5H-dibenzo [a, d] cyclohepten HC1.
Beispiel 13 5- (y-Methylaminopropyliden-5H-dibenzo [a, d]
10, 11-dihydrocyclohepten-HCl
Eine Lösung, bestehend aus 100 mg, das sind 0, 372 Millimole, von 5- (y-Chloropropyliden)-5H-dibenzo [a, d]10, 11-dihydrocyclohepten in 1, 5 ml Benzol wird mit Monomethylamin bei 10 C in einem dickwandigen Pyrexrohr gesättigt. Das Rohr wird dann geschlossen, bei 95 C während 18 Stunden stehengelassen und dann gekühlt und geöffnet. Die Benzollösung wird nacheinander mit 5 % iger Kaliumbicarbonatlösung, dann mit Wasser und gesättigter Salzlösung gewaschen, über Magnesiumsulfat getrocknet und im Vakuum zur Trockne abgedampft.
Das zurückgebliebene 01 wird dann in 3 ml Ather gelöst und mit 3 ml Ather, der mit HCl gesättigt wurde, behandelt. Das erhaltene Gemisch von Ather und 01 wird zur Trockne geblasen, um den Uberschuss an HCl zu entfernen. Nach Zerreiben des Rück- standes in Ather erhält man ein kristallines rohes Produkt mit F = 200-210 C, welches nach Umkristallisieren aus Äther-Äthanol Kristalle mit F = 213-215 C ergibt.

Beispiel 14 5- (y-Methylaminopropyliden)-5H-dibenzo [a, d]- cyclohepten-HCl
Dem im einzelnen in Beispiel 13 beschriebenen Verfahren folgend und unter Verwendung äquivalenter Mengen an 5- (y-Chloropropyliden)-5H-dibenzo [a, d] cyclohepten erhält man das entsprechende 5-(y-Methylamino- propyliden)-5H-dibenzo [a, d] cyclohepten-HC1.
Beispiel 15
Substituierte 5-Cyclopropyl-5-oxy-5H-dibenzo [a, d]
10, 11-dihydrocycloheptene
Unter Befolgung der im einzelnen in den oben angeführten Beispielen 1-14 beschriebenen Verfahren und unter Verwendung gleichwertiger Mengen der entsprechenden 5H-Dibenzo [a, d] 10, 11-dihydro-cyclohepten-5- one und 5H-Dibenzo-cyclohepten-5-one, die mit den Kernsubstituenten X und X'substituiert sind, welche X und X'dieselbe Bedeutung haben wie oben angeführt, gelangt man zu den entsprechenden substituierten 5 Cyclopropyl-5-oxy-5H-dibenzo [a, d] 10, 11-dihydro-cyclo- heptenen und 5-Cyclopropyl-5-oxy-5H-dibenzo [a, d]- cycloheptenen.
Beispiel 16
Substituierte 5-Cyclopropyl-5-oxy-5H-dibenzo- [a, d] cycloheptene
Gemäss den im einzelien in den oben angeführten Beispielen 1-15 angegebenen Verfahren und unter Verwendung äquivalenter Mengen der entsprechenden 5H-
Dibenzo [a, d] 10, 11-dihydrocyclohepten-5-one und 5H-
Dibenzo [a, d] cyclohepten-5-one die mit den Kernsubsti tuenten X und X'substituiert sind, erhält man Verbin dungen, die den in den obigen Beispielen gebildeten Aminopropyliden-Verbindungen entsprechen.
Beispiel 17 5- (y-Aminopropyliden)-5H-dibenzo [a, d] 10, 11 dihydro-cyclohepten-HCl
Eine Lösung von 1, 0 g, das sind 3, 19 Millimole, von 5- (y-Bromopropyliden)-5H-dibenzo [a, d] 10, 11-dihy- dro-cyclohepten in 25 ml absolutem Athanol wird in einem Cariusrohr mit wasserfreiem Ammoniak bei 0 C gesättigt. Das Rohr wird dann geschlossen und bei
100 C während 18 Stunden stehengelassen, dann ge kühlt und geöffnet. Die klare, hellbraune äthanolische

The present invention relates to a process for the preparation of 5H-dibenzo [a, d] cycloheptenes and 5H-dibenzo [a, d] 10, 11-dihydrocycloheptenes, which are substituted in the 5 position by an aminopropylidene radical.
The compounds produced according to the invention are useful in the treatment of mental malaise, since they are distinguished by antidepressant properties and can be used to increase the state of mind or to strengthen the soul. The daily dose administered for this purpose is within 5 mg to 250 mg, advantageously in smaller amounts distributed within a day. These compounds are conveniently administered in the form of their acid salts. The preparation of these acid salts is also encompassed by the present invention.

The process according to the invention is based on known ketones which can be produced by means of a process described in the literature.
As can be seen from the flow diagram above, the first stage of the process according to the invention is based on the condensation of a 5H-dibenzo [a, d] cyclohepten-5-one with a Grignard reagent which is derived from the cyclopropyl halide; in this condensation the corresponding cyclopropylcarbinol is formed, e.g. B. 5-Cyclopropyl-5-oxy-5H-dibenzo [ad] 10, 11-dihydrocycloheptene. This reaction is typically carried out in such a way that cyclopropyl bromide is added dropwise in tetrahydrofuran and with stirring to a certain amount of magnesium in dry tetrahydrofuran, the cyclopropyl magnesium bromide being formed.
The ketone is then added to the cyclopropylmagnesium bromide and the reaction mixture is refluxed for a convenient period of time after which the cyclopropylcarbinol intermediate is obtained.
The next stage of the process according to the invention comprises the rearrangement of the cyclopropylcarbinol to the corresponding γ-oxy-propylidenecycloheptene, e.g. B. 5 (y-oxypropylene) -5H-dibenzo [a, d] 10, 11-dihydrocyclo- heptene. The γ-oxypropylidenecycloheptene is then treated with a halogenating agent and provides the corresponding γ-halopropylidenecycloheptene derivative, e.g. B. 5- (γ-Bromopropylidene) -5H-dibenzo [a, d] 10,11-dihydrocycloheptene. The rearrangement and halogenation steps described above can be carried out simultaneously or individually.
By treating the cyclopropylcarbinol with a hydrohalide in acetic acid solution, the corresponding, -halopropylidenecycloheptene can be obtained directly in one step. Corresponding iodine derivatives, if desired, can be formed from the chlorine or bromine derivative by reacting the latter with sodium iodide in acetone under reflux.
On the other hand, the γ-oxypropylidenecycloheptane derivative can be isolated as such by treating the cyclopropylcarbinol with an acid which is other than a halogen acid. Perchloric acid in dioxane is best suited for this purpose, although other acids such as sulfuric acid, phosphoric acid and p-toluenesulfonic acid can also be used. Thereafter, the y-oxy compound can be converted into the corresponding, -halopropylidenecycloheptene, by reaction with a suitable halogenating agent; thionyl chloride or phosphorus trichloride are most suitable for these purposes, although other halogenating agents can also be used.
Finally, the γ-halopropylidenecycloheptene compound thus obtained is converted into the desired aminopropylidene derivative by an amination process. The amination can take place by direct reaction of the halogen compound with an amine, the desired aminopropylidene compound then being isolated as the hydrochloride. In another alternative, the desired amine derivative can be prepared by an amination reaction using potassium phthalimide and subsequent reaction with hydrazine.

After treatment of this oil with activated charcoal in ether and crystallization from petroleum ether, 5.49 g, in 62.5% yield, of crude crystalline cyclopropylcarbinol are obtained, which is characterized by a single spot in the DSC.

The corresponding 5-cyclopropyl-5-oxy-SH-dibenzo [a, d] cycloheptene is obtained according to the procedure described in detail in Example 1 and using equivalent amounts of 5H-dibenzo [a, d] cyclohepten-5-one.

5 (γ-Bromopropylidene-SH-dibenzo [a, d] 10,11-dihydrocycloheptene
1.3 g, that is 5.19 millimoles, 5-cyclopropyl-5oxy-SH-dibenzo [a, d] 10, 11-dihydro-cycloheptene are dissolved in 20 ml of glacial acetic acid in a 100 ml bottle and the solution is then quenched to 10 C. 10 ml of a 15% strength solution of HBr in acetic acid are then added to the solution while stirring and the reaction mixture is stirred at 10-15 ° C. for 0.5 hours.
