Verfahren zur Herstellung von kreislaufwirksamen Propenylaminen und Propylaminen
Die Erfindung betrifft ein Verfahren zur Herstellung von geeignet substituierten Propenylaminderivaten und Propylaminderivaten mit spezifischer Wirkung auf den Kreislauf.
F r die Synthese von Verbindungen z. B. vom Typ des 2-(1,1-Diphenyl-propyl-(3)-amino)-1-phenylpropan, das bereits als Therapeutikum zur Behandlung von Herz-und Kreislauferkrankungen eine broite Anwendung gefunden hat, sind zahlreiche Verfahren bekanntgeworden. Nach DAS 1. 111. 642 erfolgt die Herstellung des genannten Propylamins durch Kondensation von 1, 1-Diphenylpropylamin-(3) mit Phenylaceton und Hydrierung der SCHIFFschen Base, durch Reaktion von 1, 1-Diphenylpropylamin-(3) mit 1-Phenyl-2-brom- propan, durch Reaktion von 1, 1-Diphenylpropylbro- mid- (3) mit 1-Phenyl-2-aminopropan oder durch Kondensation von 1, 1-Diphenylpropionaldehyd mit 1-Phenyl-2-aminopropan und Hydrierung der SCHIFFschen Base.
Es ist ausserdem möglich, durch Grignardierung eines entsprechenden substituierten ¯-Amino-propionsÏureesters bzw.¯-Aminopropiophenons, darauffolgende Wasserabspaltung aus dem tertiären Aminoalko- hol und Hydrierung des Aminopropens zum 2- (l, l-Diphenyl-propyI- (3)-amino)-l-phenylpropan zu gelangen (DDR-Patentschrift 27 914, Kl. 12q, 32/21).
Weitere noch bekanntgewordene Verfahren (z. B. DAS 1.100. 031, DAS 1. 133. 395) zur Herstellung analoger Verbindungen haben unseres Wissens eine praktische Bedeutung bisher nicht erlangt.

Das Verfahren gestaltet sich insbesondere dadurch so ausserordentlich vorteilhaft, da die als Ausgangspro- dukte benötigten Aldehyde (IV) sehr leicht aus Benzophenon oder substituierten Benzophenonen und Acetylen und anscEliessende Umlagerung der so erhaltenen ¯thinylcarbinole in bekannter Weise und guter Ausbeute erhalten werden.
Als Amine der allgemeinen Formel III können z. B. verwendet werden : l-Phenyl-2-methylamino-propanol-(1) l-Phenyl-2-methylamino-propan
1-Phenyl-2-amino-propanol-(1) l-PhenyI-2-amijio-propan l-Cyclohexyl-2-methylam, ino-propan l-Phenyl-2-amino-äthan
Die. Kondensation der Aldehyde (IV) mit Ammen (III) kann in einem inerten Lösungsmittel oder auch ohne Lösungsmittel bei Zimmertemperatur oder erhöh- ter Temperatur vorgenommen werden, wobei die gebil deten ungesÏttigten SCHIFFschen Basen (im Falle R1 =H) in ausgezeichneter Ausbeute erhalten und durch ansdhliessende katalytische Hydrierung mit Edehne- tallkatalysatoren oder Raney-Nickel in einem für die Hydrierung geeigneten Lösungsmittel, z. B.
Alkohol,
Eisessig, oder auch durch Reduktion mit naszierendem Wasserstoff in Abhängigkeit von den Reduktionsbedin gungen (alkalisch bis sauer) in die entsprechenden Pro penylamine oder Propylamine übergeführt werden.
Es ist andererseits auch m¯glich, die Aldehyde (IV) in Gegenwart der Amine (III) direkt in einem f r die Hydrierung geeigneten L¯sungsmittel, z. B. Alkohol, mit Katalysatoren oder auch mit naszierendem Wasser stoff zu reduzieren, wobei man in einem Reaktionsgang -je nach Wahl der Reduktionsbedingungen) (alkalisch bis sauer)-zu den Propenylaminen oder Propylaminen gelant. Falls Ri eine Alkylkette bedeutet, ist mur diese Verfahrensweise anwendbar. Darüber hinaus s kann man natürlich die tertiären Amine durch Alkylie- rung der sekundären Propenylamine oder Propylamine in bekannter Weise herstellen.
Die auf dem beschriebenen Wege erhaltenen Verbindungen können mit Hilfe von anorganischen oder organ, ischen Säuren in an sich bekannter Weise in die entsprechenden nichttoxischen Salze übergeführt werden. Im Falle der tertiären Amine können z. B. mit Alkylhalogeniden auch die entsprechenden quatemier- ten Derivate gewonnen werden.
Die Verfahrensprodukte besitzen, wie zum Teil auch in der Literatur beschrieben, im Vergleich zu bekannten Kreislaufmitteln eine bedeutend intensivere Kreislaufwirkung und bewirken vor allem eine starke KoronargefÏsserweiterung und Koronardurchstr¯mung.
Sie sind aus diesem Grund wertvolle Arzneimittel oder Zwischenprodukte f r solche. Die in den Beispielen.angegebenen Temperaturen beziehen sich auf Celsiusgrade ( C).
Beispiel 1 2-(1, 1-Diphenyl-(1)-propenyl-(3)-amino-1-phenyl- propan a) 13, 5 g 1-Phenyl-2-aminopropan und 20, 8 g 1, 1-Diphenyl-acrolein werden unter Rühren 15 Minuten auf 60 erwärmt. Nach Abkühlen wird das Reak tionsgemisch unter gelinden Erwärmen in Petroläther gelöst und die SCHIFFsche Base zur Kristallisation gebracht. Schmp. 88 bis 89 ¯ C. Ausbeute 27,5g. b) 27,5g der unter a) erhaltenen SCHIFFschen Base werden in 150 ml Methanol in Gegenwart von Raney-Nickel als Katalysator in einer Wasserstoffatmosphäre bei 50 geschüttelt.
Nach beendeter Wasserstoffaufnahme wird vom Katalysator abgetrennt, mit HC1 angesäuert und das Lösungsmittel bis zur begin- nenden Kristallisation abdestilliert. Schmp. des Hydrochlorids 197 bis 198 , Ausbeute 25 g.
Beispiel 2 2-(1, 1-Diphenyl-(1)-propyl-(3)- amin)-1-phenylpropan
27, 5 g der im Beispiel 1 a) erhaltenen SCHIFFschen Base werden in 150 ml Methanol und 5 ml Wasser in Gegenwart von Platinoxyd als Katalysator in einer Wasserstoffatmosphäre geschüttelt. Nach Aufnahme eines Mols Wasserstoff wird die Hydrierung unterbrochen, vom Katalysator abgetrennt, mit HCl angesäuert und das Lösungsmittel bis zur beginnenden Kristallisation abdestilliert. Schmp. des Hydrochlorids 204 bis 206 C. Ausbeute 25, 8 g.
Beispiel 3 2-(1, 1-Diphenyl-propyl-(3)- amino)-1-hydroxy-1-phenyl-propan a) 15, 1 g 1-Phenyl-2-amino-propanol-(1) und 20, 8 g 1, 1-Diphenylacrolein werden unter Rühren 15 Min. auf 70¯C erwÏrmt. Nach Zugabe von Aceton wird die SCHIFFsche Base zur Kristallisation gebracht. Schmp. 126 bis 127 C. Ausbeute 26, 5 g. b) 26, 5 g der unter a) erhaltenen SCHIFFschen Base werden in 150 ml Methanol in Gegenwart von Platinoxyd als Katalysator in einer Wasserstoffatmo sphäre bei 50 C geschüttelt. Nachdem 1 Mol Wasser- stoff aufgenommen wurde, wird die berechnete Menge Salzsäure zur Bildung des Cblorhydrates zugesetzt und weiter hydriert.
Nach beendeter Wasserstoffaufnabme wird vom Katalysator abgetrennt und das Lösungsmittel bis zur beginnenden Kristallisation abdestilliert.
Schmp. des Chlorhydrates 232 bis 233 C. Ausbeute 24g.
Beispiel 4 2- (1, 1-Diphenyl- (1)-propenyl-3- methylamino)-1-phenyl-propan
14,9g 1-Phenyl-2-methylaminopropan werden mit Platinoxyd und 200 ml Methanol in einer Wasserstoffatmosphäre geschüttelt. Nachdem das Platinoxyd reduziert ist, gibt man tropfenweise 20, 8 g 1, 1-Diphenylacrolein, gelöst in 100 ml Methanol, zu. Nach Aufnahme von 1 Mol Wasserstoff wird wie im Beispiel 2 beschrieben aufgearbeitet. Schmp. des Hydrochlorids 179 bis 180¯C. Ausbeute 17 g.
Beispiel S
2- (l, l-Dipbenyl-propyl- (3)- methylamino)-1-phenylpropan
14, 9 g 1-Phenyl-2-mebhylaminopropan und 20, 8 g 1, 1-Diphenylacrolein werden wie im Beispiel 4 beschrieben bis zur Aufnahme von 1 Mol Wasserstoff hydriert. Nach Zugabe der berechneten Menge Salzsäure zu der Hydrierlösung wird bis zur Aufnahme von 2 Mol Wasserstoff weiter hydriert und vom Katalysator abgetrennt. Das Lösungsmittel wird abdestilliert un, der Rückstand aus Essigester kristallisiert. Schmp. des Hydrochlorids 156 bis 158 C. Ausbeute 15 g.
Beispiel 6 2- (1, 1-Diphenyl-propyl- (3)- methylamino)-l-cyclohexyl-propan
15, 5 g 1-Cyclohexyl-2-methylaminopropan und 20, 8 g 1, 1-Diphenylacrolein werden wie im Beispiel 5 beschrieben hydriert. Nach beendeter Wasserstoffaufnahme wird vom Katalysator getrennt, das Lösungsmittel abdestilliert und vom Rückstand die Base freigesetzt. Schmp. der Base 97 bis 99¯C. Ausbeute 16, 1 g.
Beispiel 7 2-(1, 1-Diphenyl-propyl-(3)-methylamino)-
1-hydroxy-l-phenylpropan
16, 5 g 1-Phenyl-2-methylamino-propanol und 20, 8 g 1,1-Diphenylacrolein werden wie im Beispiel 5 beschrieben hydriert und aufgearbeitet. Schmp. des Hydrochlorids 161 bis 163¯C. Ausbeute 14, 8 g.
Process for the production of propenylamines and propylamines with a circulation effect
The invention relates to a process for the preparation of suitably substituted propenylamine derivatives and propylamine derivatives with a specific effect on the circulation.
For the synthesis of compounds e.g. B. of the 2- (1,1-diphenyl-propyl- (3) -amino) -1-phenylpropane type, which has already been widely used as a therapeutic agent for the treatment of cardiovascular diseases, numerous processes have become known. According to DAS 1. 111.642, the said propylamine is prepared by condensation of 1,1-diphenylpropylamine- (3) with phenylacetone and hydrogenation of the SCHIFF's base, by reaction of 1,1-diphenylpropylamine- (3) with 1-phenyl- 2-bromopropane, by reaction of 1,1-diphenylpropylbromide- (3) with 1-phenyl-2-aminopropane or by condensation of 1,1-diphenylpropionaldehyde with 1-phenyl-2-aminopropane and hydrogenation of the SCHIFF base .
It is also possible by Grignardation of a corresponding substituted ¯-amino-propionic acid ester or ¯-aminopropiophenone, subsequent elimination of water from the tertiary amino alcohol and hydrogenation of the aminopropene to give 2- (l, l-diphenyl-propyI- (3) -amino ) -l-phenylpropane (GDR patent specification 27 914, class 12q, 32/21).
Other processes that have become known (e.g. DAS 1.100. 031, DAS 1. 133. 395) for the production of analogous compounds have not, to our knowledge, achieved any practical significance.

The process is particularly advantageous because the aldehydes (IV) required as starting products can be obtained very easily from benzophenone or substituted benzophenones and acetylene and subsequent rearrangement of the ¯thynyl carbinols thus obtained in a known manner and in good yield.

The. Condensation of the aldehydes (IV) with amines (III) can be carried out in an inert solvent or else without a solvent at room temperature or elevated temperature, the unsaturated SCHIFF bases formed (in the case R1 = H) being obtained in excellent yield and through Subsequent catalytic hydrogenation with noble metal catalysts or Raney nickel in a solvent suitable for the hydrogenation, e.g. B.
Alcohol,
Glacial acetic acid, or by reduction with nascent hydrogen, depending on the reduction conditions (alkaline to acidic), can be converted into the corresponding propenylamine or propylamine.
On the other hand, it is also possible to dissolve the aldehydes (IV) in the presence of the amines (III) directly in a solvent suitable for the hydrogenation, e.g. B. to reduce alcohol, with catalysts or with nascent hydrogen, in one reaction - depending on the choice of reduction conditions) (alkaline to acidic) - gelant to the propenylamines or propylamines. If Ri is an alkyl chain, this procedure can only be used. In addition, the tertiary amines can of course be prepared by alkylation of the secondary propenylamines or propylamines in a known manner.
The compounds obtained in the route described can be converted into the corresponding non-toxic salts with the aid of inorganic or organic acids in a manner known per se. In the case of the tertiary amines, for. For example, the corresponding quaternized derivatives can also be obtained with alkyl halides.
The products of the process have, as partly also described in the literature, a significantly more intensive circulatory effect compared to known circulatory agents and, above all, cause a strong coronary vasodilatation and coronary flow.
For this reason, they are valuable drugs or intermediates for them. The temperatures given in the examples relate to degrees Celsius (C).
Example 1 2- (1, 1-Diphenyl- (1) -propenyl- (3) -amino-1-phenyl-propane a) 13.5 g of 1-phenyl-2-aminopropane and 20.8 g of 1, 1- Diphenyl-acrolein is heated to 60 for 15 minutes while stirring. After cooling, the reaction mixture is dissolved in petroleum ether with gentle warming and the SCHIFF base crystallized. Melting point 88 to 89 ° C. Yield 27.5 g. b) 27.5 g of the SCHIFF base obtained under a) are shaken at 50 in a hydrogen atmosphere in 150 ml of methanol in the presence of Raney nickel as a catalyst.

Example 2 2- (1,1-Diphenyl- (1) -propyl- (3) -amine) -1-phenylpropane
27.5 g of the SCHIFF base obtained in Example 1 a) are shaken in 150 ml of methanol and 5 ml of water in the presence of platinum oxide as a catalyst in a hydrogen atmosphere. After one mole of hydrogen has been taken up, the hydrogenation is interrupted, the catalyst is separated off, acidified with HCl and the solvent is distilled off until crystallization begins. M.p. of the hydrochloride 204-206 C. Yield 25.8 g.
Example 3 2- (1, 1-diphenyl-propyl- (3) -amino) -1-hydroxy-1-phenyl-propane a) 15, 1 g 1-phenyl-2-aminopropanol- (1) and 20 , 8 g of 1,1-diphenylacrolein are heated to 70 ° C for 15 minutes while stirring. After adding acetone, the SCHIFF base is made to crystallize. Mp 126-127 C. Example 4 2- (1,1-Diphenyl- (1) -propenyl-3-methylamino) -1-phenyl-propane
14.9 g of 1-phenyl-2-methylaminopropane are shaken with platinum oxide and 200 ml of methanol in a hydrogen atmosphere. After the platinum oxide has been reduced, 20.8 g of 1,1-diphenylacrolein, dissolved in 100 ml of methanol, are added dropwise. After 1 mole of hydrogen has been taken up, it is worked up as described in Example 2. M.p. of the hydrochloride 179 to 180 ° C. Yield 17g.
Example p
2- (1,1-Dipbenyl-propyl- (3) -methylamino) -1-phenylpropane
14.9 g of 1-phenyl-2-mebhylaminopropane and 20.8 g of 1,1-diphenylacrolein are hydrogenated as described in Example 4 until 1 mol of hydrogen is absorbed. After the calculated amount of hydrochloric acid has been added to the hydrogenation solution, hydrogenation is continued until 2 moles of hydrogen have been absorbed and the catalyst is separated off. The solvent is distilled off and the residue is crystallized from ethyl acetate. Mp. Of the hydrochloride 156 to 158 C. Yield 15 g.
Example 6 2- (1,1-Diphenyl-propyl- (3) -methylamino) -l-cyclohexyl-propane
15.5 g of 1-cyclohexyl-2-methylaminopropane and 20.8 g of 1,1-diphenylacrolein are hydrogenated as described in Example 5. When the hydrogen uptake has ended, the catalyst is separated off, the solvent is distilled off and the base is liberated from the residue. M.p. of the base 97 to 99¯C. Yield 16.1g.
Example 7 2- (1,1-Diphenyl-propyl- (3) -methylamino) -
1-hydroxy-l-phenylpropane
16.5 g of 1-phenyl-2-methylamino-propanol and 20.8 g of 1,1-diphenylacrolein are hydrogenated and worked up as described in Example 5. M.p. of the hydrochloride 161 to 163¯C. Yield 14.8g.
