Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von 1,10-Trimethylen-8-methyl-1,2,3,4- -tetrahydropyrazino-[1 .2-a]-indolhydrochlorid. welches in der Medizin als Wirkstoff in psychotropen Präparaten Anwendung finden kann.
Das psychotrope Präparat, welches von uns bedingt Pyrazidol genannt wurde, übt eine pharmakologische Wirkung aus, die für psychotrope Präparate kennzeichnend ist.
Es potenziert zentrale Effekte (Stereotypie, Hyperthermie, Gruppentoxizität u.a.) von Phenamin und 5-Hydroxytryptophan und periphere Effekte (hypertensive Wirkung, Kontraktion der Nickhaut u.a.) von Adrenalin, Noradrenalin und Serotonin, sowie vermindert die hemmende Wirkung von Reserpin auf das Zentralnervensystem. Dem Wirkungscharakter nach kann das Präparat den Präparaten von antidepressiver Wirkung zugerechnet werden. Gleichzeitig damit übt das Präparat eine sedative Wirkung aus, vermindert die Körpertemperatur, potenziert die Wirkung von Narkotika, Analgetika und Lokalanästhetika. Diesen Merkmalen nach ist das neue Präparat den Neuroleptika ähnlich.
Zum Unterschied von den bekannten Antidepressanten (Imipraminium, Amintriptylinum übt das neue Präparat weder eine cholinolytische noch eine adrenolytische Wirkung aus.
Es ist ein Verfahren zur Herstellung von l,10-Trimethy len-8-methyl- 1,2,3 ,4-tetrahydropyrazino-[l ,2-a]-indol bekannt, welches darin besteht, dass N-Metallderivate von l-Keto -1,2,3,4-tetrahydro-6-methylcarbazol, zum Beispiel das N-Natriumderivat von 1 -Keto- 1,2,3 ,4tetrahydro- 6-methylcarba- zol mit < Z-Bromacetalen aliphatischer Aldehyde, z.B., mit Dibutylacetal des Bromessigsäurealdehyds, mit darauffolgender Behandlung der Reaktionsmasse mit Ammoniumsalzen organischer Säuren (Ammoniumacetat u.a.) unter Kochen in Essigsäure umgesetzt werden. Das sich infolge der Reaktion bildende 1,1 O-TrimethyIen-8-methylpyrazino-[l ,2-a]-indol kann dann der Reduktion mit Natrium in absolutem Alkohol unter Kochen unterworfen werden.
Die sich bildende Base von 1,1 0-Trimethylen-8-methyl- -1 ,2,3,4-tetrahydropyrazino-[l ,2-a]-indol wird gewöhnlich in einem organischen Lösungsmittel- (Äther, Benzol u.a.) gelöst und die erhaltene Lösung kann mit ätherischer oder alkoholischer Chlorwasserstofflösung behandelt werden.

194 827).
Der Nachteil des genannten Verfahrens besteht in der Kompliziertheit der Technologie, die durch die Notwendigkeit der Verwendung von Natrium in grossen Mengen hervorgerufen ist, was die Ausnutzung des genannten Verfahrens im Industriemassstab erschwert.
Das Ziel der vorliegenden Erfindung besteht in der Beseitigung des genannten Nachteils.
In Übereinstimmung mit diesem Ziel wurde die Aufgabe gestellt, durch die Veränderung der technologischen Vorgänge die Herstellungstechnologie zu vereinfachen, um die Venvendung des genannten Verfahrens im Industriemassstab zu ermöglichen.
Die Aufgabe wurde dadurch gelöst, dass man im Verfahren zur Herstellung von lfi10-Trimethylen-8-methyl-1,2,3,4- -tetrahydropyrazino-[1 2-a]-indol das 1-Keto-1 ,2,3,4-tetrahy- dro-6-methylcarbazol erfindungsgemäss mit einem alkalischen Mittel umsetzt, das sich bildende 1 ,2,3,4-Tetrahydro-6-methylcarbazolderivat mit Chlorazetonitril im einem organischen Lösungsmittel alkyliert,
das erhaltene l-Keto-1,2,3,4-te- trahydro-6-methyl-9-cyanmethylcarbazol der reduzierenden Zyklisierung unter Wasserstoffeinwirkung in Gegenwart von Hydrierungskatalysatoren in organischen Lösungsmitteln unter einem Druck von 30 bis 70 atm und einer Temperatur von 30 bis 60"C mit darauffolgender Isolierung des Endproduktes unterwirft.
Als alkalisches Mittel verwendet man vorzugsweise Natriumalkoholat oder Natriumhydrid.
Die Alkylierung ist vorzugsweise in Dimethylformamid oder Dioxan durchzuführen. Als Hydrierungskatalysator ist es zweckmässig, Raney-Nickel oder einen Adams-Katalysator zu verwenden. Es ist im allgemeinen notwendig, die Reduktionszyklisierung im Medium von niederen aliphatischen Alkoholen durchzuführen.
Das erfindungsgemässe Verfahren kann folgenderweise verwirklicht werden: Der heissen l-Keto-l,2,3,4-tetrahydro- -6-methylcarbazollösung in einem wasserfreien organischen Lösungsmittel (Dioxan, Dimethylformamid, Benzol u.a.) wird eine Suspension von Natriumhydrid in oben genannten Lösungsmitteln oder eine alkoholische Natriumalkoholatlösung zugegeben: als Natriumalkoholat können Natriummethylat, Natriumäthylat u.a. verwendet werden. Der Alkohol wird unter Vakuum bei einer Temperatur von 65 bis 70"C abgetrieben und zu der auf die Zimmertemperatur abgekühlten Reaktionsmasse wird unter Rühren die Chloracetonitrillösung im genannten organischen Lösungsmittel hinzugegossen. Das Gemisch wird bei Zimmertemperatur während 30 Minuten gerührt und auf Eis ausgegossen. Das ausgeschiedene öl kristallisiert allmählich aus.
Es wird 1 -Keto- 1,2,3 ,4-tetrahy- dro-6-methyl-9-cyanmethylcarbazol erhalten. Die Ausbeute beträgt 70-81 Gew.-%; Schmp. 139 bis 140"C (aus dem Gemisch von Methanol und Dichloräthan). Das genannte Produkt stellt ein weisses kristallines Pulver dar, das in Methanol, Äthanol, Benzol oder Äther gut löslich ist.
Das erhaltene 1 -Keto-l ,2,3,4-tetrahydro-6-methyl-9-cyan- methylcarbazol wird gewöhnlich in einem organischen Lösungsmittel gelöst; es ist zweckmässig, als organisches Lösungsmittel niedere aliphatische Alkohole zu verwenden.
Nachher gibt man der Lösung einen Hydrierungskatalysator, z.B. eine Raney-Nickel-Suspension, zu und führt die Hydrierung bei einem Druck von 50 bis 70 atm und einer Temperatur von 30 bis 60 C so lange durch, bis die Wasserstoffaufnahme aufhört. Der Prozess dauert gewöhnlich 5 bis 20 Stunden. Man filtriert die Reaktionsmasse, dampft den Alkohol ein und löst das zurückgebliebene dicke öl in einem organischen Lösungsmittel, z.B. in Benzol, Azeton, Äther.
Dann fällt man das Endprodukt mit der ätherischen oder alkoholischen Chlorwasserstofflösung.
Die Ausbeute an 1,1 0-Trimethylen-8-methyl-1 ,2,3,44etra- hydropyrazino-tl ,2-aJ-indolhydrochlorid beträgt 70-58 Gew.-46: Schmp. 260-261 0C (unter Zersetzung; in einer verlötenen Kapillare). Das Endprodukt stellt eine weisse kristalline Substanz dar, die in heissem Wasser, Alkoholen, Essigsäure löslich, in Äther, Benzol, Azeton unlöslich ist.
Das erfindungsgemässe Verfahren gestattet es, die Herstellungstechnologie zu vereinfachen, indem die unbequeme und feuergefährliche Stufe der Reduktion von 1,1 0-Trimethylen-8- -methylpyrazino-[1,2-a]-indol mit Natrium in Alkohol ausgeschlossen wird.
Das erfindungsgemässe Verfahren ermöglicht es, das Arzneipräparat im Industriemassstab herzustellen.
Zur Veranschaulichung der vorliegenden Erfindung werden folgende Beispiele zur Ausbildung der Verfahrens der Herstellung von 1.1 0-Trimethylen-8-methyl-1,2,3 ,4-tetrahy- dropyrazino-[1,2-a]-indol angeführt.
Beispiel I
Zu einer heissen Lösung von 39,8 g (0,2 Mol) 1-Keton -1,2,3,4-tetrahydro-6-methylcarbazol in 100 ml absolutem Dioxan werden 4,6 g (0,2 g-Atom) aus Natrium bereiteten Natriumäthylat zugegeben. Der Äthylalkohol wird unter Vakuum auf dem Wasserbad bei einer Temperatur von 35-400C abdestilliert, die Reaktionsmasse wird auf Zimmertempera tur abgekühlt > 1Pla und unter kräftigem Rühren tropfenweise eine Lösung von 18,8 g (0,25 Mol) Chlorazetonitril in 100 ml wasserfreiem Dioxan zugegossen. Das Gemisch wird bei Zimmertemperatur während 30 Minuten gerührt und auf Eis ausgegossen. Das ausgeschiedene öl kristallisiert nach einer Stunde aus.
Man erhält 35,5 g (70 Gew.-%) l-Keto-1,2,3,4- -tetrahydro-6-methyl-9-cyanrnethylcarbazol; Schmp. 139 l400C (aus dem Gemisch von Methanol und Dichlhoräthan).

In einen Autoklaven bringt man 35,5 g (0,15 Mol) 1-Keto - 1,2,3 ,4-tetrahydro-6-methyl-9-cyanmethylcarbazol, 200 ml Methylalkohol und 10 g einer Raney-Nickel-Paste ein. Die Hydrierung wird unter einem Druck von 30 atm und bei einer Temperatur von 500so lange durchgeführt, bis die Wasserstoffaufnahme aufhört (gegen 20 Stunden). Der Katalysator wird abgetrennt, der Alkohol eingedampft, das zurückgebliebene dicke öl in Äther gelöst und aus der mit Eis abgekühlten ätherischen Lösung das 1,10-Trimethylen-8-me- thyl-1,2,3,4-tetrahydropyrazino-[1,2-a]-indolhydrochlorid mit der ätherischen Chlorwasserstofflösung ausgefällt.
Die Aus beute von 1,1 0-Trirnethylen-8 -methyl-i 23 ,44etrahydropyra- zino-[l ,2-a]-indolhydrochlorid beträgt 27,5 g (70 Gew.-O/oP, Schmp. 260-261 0C (unter Zersetzung; in einer verlöteten Kapillare). C,5H 9N2CI.
Gefunden: C 68,83%; 68,50%; H 7,38%; 7,10%; N 10,57%; 10,31% Berechnet: C 68,56%; H 7,29%; N 10,66%.
Beispiel 2
Zu einer heissen Lösung von 39,8 g (0,2 Mol) l-Keto-1,2.- 3.4-tetrahydro-6-methylcarbazol in 100 ml wasserfreiem Dimethylformamid werden 4,6 g (0,2 g-Atom) von aus Natrium bereitetem Natriummethylat zugegeben. Der Methylalkohol wird unter Vakuum auf dem Wasserbad bei einer Temperatur von 65 bis 70"C abdestilliert und zu der auf Zimmertemperatur abgekühlten Reaktionsmasse tropfenweise eine Lösung von 18,8 g (0,25 Mol) Chloracetonitril in 10 ml Dimethylformamid zugegegossen. Das Verfahren wird analog zu Beispiel 1 bis zum Entstehen von l-Keto-1,2,3,4-tetrahy- dro-6-methyl;9-zyanmethylcarbazols durchgeführt.
Die Ausbeute an l-Keto-l ,2,3,4-tetrahydro-6-methyl-9-cyanmethyl- carbazol beträgt 41,3 g (81,3 Gew.-%), Schmp. 139-1400C.
In einen Autoklaven werden 41,3 g (0,17 Mol) l-Keto -1,2,3,4-tetrahy & -6-methyl-9-zyanmethylkarbazol. 300 ml Äthylalkohol, 40 g Raney-Nickel-Paste eingetragen. Die Hydrierung wird bei einem Druck von 70 atm und einer Temperatur von 500C durchgeführt (die Wasserstoffaufnahme hört nach 5 Stunden auf). Der Katalysator wird abgetrennt, der Alkohol eingedampft, das zurückgebliebene dicke öl in Azeton gelöst und aus der mit Eis abgekühlten ätherischen Lösung das 1,10-Trimethylen-S-methyl-1,2,3,4-tetrahydropy- razino-tl ,2-a]-indolhydrochlorid mit einer alkoholischen Chlorwasserstofflösung aus gefällt. Die Ausbeute an Endprodukt beträgt 33 g (88 Gew.- %); Schmp. 260-261 0C (unter Zersetzung; in einer verlöteten Kapillare).
Beispiel 3
Zu einer Suspension von 7,2 g (0,3 Mol) Natriumhydrid in 30 ml wasserfreiem Dimethylformamid wird tropfenweise die auf eine Temperatur von 700C erwärmte Lösung von 39,8 g (0,2 Mol) l-Keto-1,2,3,4-tetrahydro-6-methylcarbazol in 150 ml Dimethylformamid zugegossen. Das Reaktionsgemisch wird 30 Minuten bei Zimmertemperatur gerührt und nachher werden tropfenweise 18,8 g (0,25 Mol) Chloracetonitril hinzugefügt. Das Verfahren wird analog zu Beispiel 1 bis zum Entstehen von 1 -Keto-1 ,2,3,4-tetrahydro-6-methyl- -9-cyanmethylcarbazol durchgeführt. Die Ausbeute an l-Keto-1,2,3,4-tetrahydro-6-methyl-9-cyanmethylcarbazol beträgt 42,0 g (81,4 Gew.-%); Schmp. 139-1400C.
In einen Autoklaven werden 42 g (0,17 Mol) l-Keto-1,2,- 3,4tetrahydro-6-methyl-9-cyanmethylcarbazol, 300 ml Äthylalkohol und ein aus 0,1 g nach Adams hergestellter Platinoxid-Katalysator eingetragen.
Die Hydrierung und die Isolierung des Endproduktes werden unter den zu Beispiel 2 analogen Bedingungen durchgeführt.
Die Ausbeute an Endprodukt beträgt 32,8 g (87,8 Gew.- %), Schmp. 260-261 C (unter Zersetzung; in einer verlöteten Kapillare).
The present invention relates to a process for the preparation of 1,10-trimethylene-8-methyl-1,2,3,4-tetrahydropyrazino [1 .2-a] indole hydrochloride. which can be used in medicine as an active ingredient in psychotropic preparations.
The psychotropic preparation, which we conditionally called pyrazidol, has a pharmacological effect that is characteristic of psychotropic preparations.
It potentiates central effects (stereotypy, hyperthermia, group toxicity, etc.) of phenamine and 5-hydroxytryptophan and peripheral effects (hypertensive effect, contraction of the nictitating membrane, etc.) of adrenaline, noradrenaline and serotonin, as well as reducing the inhibitory effect of reserpine on the central nervous system. According to the nature of its action, the preparation can be assigned to preparations with an antidepressant effect. At the same time, the preparation has a sedative effect, reduces body temperature, and increases the effect of narcotics, analgesics and local anesthetics. According to these characteristics, the new preparation is similar to neuroleptics.
In contrast to the well-known antidepressants (Imipraminium, Amintriptylinum), the new preparation has neither a cholinolytic nor an adrenolytic effect.

Sodium alcoholate or sodium hydride is preferably used as the alkaline agent.
The alkylation should preferably be carried out in dimethylformamide or dioxane. It is expedient to use Raney nickel or an Adams catalyst as the hydrogenation catalyst. It is generally necessary to carry out the reduction cycling in the medium of lower aliphatic alcohols.

The yield of 1,1 0-trimethylene-8-methyl-1, 2,3,44-tetrahydropyrazino-tl, 2-aJ-indole hydrochloride is 70-58 wt. 46: mp. 260-261 ° C. (with decomposition; in a soldered capillary). The end product is a white crystalline substance that is soluble in hot water, alcohols, acetic acid, and insoluble in ether, benzene, acetone.
The process according to the invention makes it possible to simplify the production technology by excluding the inconvenient and flammable step of reducing 1,1 0-trimethylene-8-methylpyrazino [1,2-a] indole with sodium in alcohol.
The method according to the invention makes it possible to manufacture the medicinal product on an industrial scale.
To illustrate the present invention, the following examples are given to implement the process of preparing 1,1 0-trimethylene-8-methyl-1,2,3,4-tetrahydropyrazino [1,2-a] indole.
Example I.
To a hot solution of 39.8 g (0.2 mol) of 1-ketone -1,2,3,4-tetrahydro-6-methylcarbazole in 100 ml of absolute dioxane, 4.6 g (0.2 g atom) Sodium ethylate prepared from sodium was added.
