Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen heterocyclischen Guanidin Derivaten der Formel Ia bzw. Ib,

Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man Hydrazin-Derivate der Formel II

worin entweder Ru/ obige Bedeutung von Ru besitzt und Z für einen Alkoxy-, Alkylthio-oder einen gegebenenfalls substituierten 1-Pyrazolylrest steht oder R3, und Z zusammen für eine dritte Bindung zwischen dem Kohlenstoff- und dem Stickstoffatom stehen, umsetzt.
Der chemische Vorgang des erfindungsgemässen Verfahrens besteht darin, dass die Hydrazingruppe einer Verbindung der Formel II durch einen Guanylrest substituiert wird, welcher in der Reaktionskomponente der Formel II bereits vorgebildet ist. Diese Substitution kann sowohl am sekundären als auch am primären Stickstoffatom der Hydrazingruppe erfolgen, wobei eine Verbindung der Formel Ia bzw. eine Verbindung der Formel Ib gebildet wird; meist entsteht bei der Umsetzung ein Gemisch der beiden möglichen Isomeren, worin Verbindung Ia mengenmässig überwiegt.
Das Verfahren kann beispielsweise folgendermassen durchgeführt werden:
Man versetzt die Hydrazinverbindung II in Wasser oder in einem Gemisch von Wasser und einem niederen Alkanol mit einem anorganischen Salz eines O-Alkylisoharnstoffes oder eines S-Alkyl-isothioharnstoffes, beispielsweise mit OMethyl-isoharnstoffsulfat, S-Methyl- isotbioharnstoffsulfat, N,S - Dimethyl - isothioharnstoffjo- did usw. Das Gemisch wird während mehrerer Tage bei Raumtemperatur stehen gelassen und/oder während 2-3 Std. auf 50-100 erhitzt.
Nach einer Variante des Verfahrens erhitzt man die Verbindung der Formel II in Wasser, Chloroform usw., mit einem Salz, z. B. mit dem Sulfat, Nitrat usw., des l-Guanylpyrazols oder eines seiner einfachen Kern Substitutionsprodukte (wie z. B. des l-Guanyl-3,5-di- methylpyrazols) während mehrerer Stunden zum Sieden.
Nach einer weiteren Verfahrensvariante wird die Verbindung der Formel II mit Cyanamid, Methylcyanamid usw. in einem niederen Alkanol, wie Äthanol, während mehrerer Stunden zum Sieden erhitzt; vorzugsweise setzt man die Verbindung II in Form eines Säureadditionssalzes ein oder fügt der Reaktionslösung eine Säure bei.
Die entstandenen Guanidin-Derivate können als freie Basen oder in Form ihrer Salze isoliert und gereinigt werden; die Auftrennung von Isomerengemischen in ihre Komponenten Ia und Ib kann durch fraktionierte Kristallisation und/oder Adsorptionschromatographie erfolgen. Kristallisiert man z. B. ein Gemisch der Säureadditionssalze einer Verbindung Ia und einer Verbindung Ib in polaren Lösungsmitteln, wie niedere Alkanole oder Äthanol/Wasser, so isoliert man die reine Verbindung Ia in Form eines Salzes, während das entsprechende Salz der Verbindung Ib in der Mutterlauge verbleibt.
Die als Ausgangsprodukte verwendeten Hydrazinderivate der Formel II können beispielsweise folgendermassen erhalten werden:
Piperidin oder ein Mono- oder Disubstitutionsprodukt des Piperidins, wie z. B. 3-Methylpiperidin, 2 Äthylpiperidin, 2,6-Dimethylpiperidin usw., wird mit Äthylenoxyd hydroxyäthyliert, und die so erhaltene Verbindung in ein Derivat der Formel IV

übergeführt, das an Stelle der Hydroxylgruppe einen reaktionsfähigen Säurerest (X), wie insbesondere Chlor, Brom oder eine Benzol-, p-Brombenzol- oder -Toluol- sulfonyloxygruppe usw. aufweist.
Hieraus kann durch Umsetzung mit Hydrazin oder Hydrazinhydrat direkt eine Verbindung der Formel II gewonnen werden; eine andere Möglichkeit besteht in der Reaktion der Verbindung IV mit einem acylierten Hydrazin und anschliessender Verseifung der entstandenen Verbindung der Formel V.

Schliesslich kann Piperidin oder ein geeignetes Substitutionsprodukt des Piperidins auch mit Kaliumcyanid und Formaldehyd oder mit Brom- oder Chlor-acetonitril, in Gegenwart eines säurebindenden Mittels, umgesetzt werden, worauf das entstandene Cyanomethylderivat zum entsprechenden Aminoäthylderivat der Formel VI

reduziert wird; anschliessende Reaktion mit Chloramin liefert ebenfalls ein Hydrazinderivat der Formel II.
Die nach dem vorliegenden Verfahren hergestellten, bisher unbekannten heterocyclischen Guanidin-Derivate zeichnen sich durch wertvolle pharmakodynamische Eigenschaften aus. So bewirken sie an wachen hypertonen Ratten und Hunden bereits in niedrigen Dosen eine rasch einsetzende, ausgeprägte und anhaltende Blutdrucksenkung, welche auch bei peroraler Verabreichung zustande kommt, sowie eine davon unabhängige leichte Bradykardie; Blutdruck und Herzfrequenz normotoner Versuchstiere werden dagegen erst in einem wesentlich höheren Dosenbereich beeinflusst.
In ihrem Wirkungsmuster unterscheiden sich die neuen Verbindungen deutlich von den bisher bekannten Antihypertensiva. So geht ihrer blutdrucksenkenden Wirkung keine pressorische sympathomimetrische Phase voraus, wie dies bei andern Sympathicusblockern der Guanidinreihe der Fall ist. Gegenüber den Antihypertensiva der Reserpinreihe zeichnen sie sich durch eine nur geringfügige sedative Komponente aus. Die Verbindungen zeigen keine hemmenden Einflüsse auf Rückenmarks reflexe oder auf die Impulsübertragung in motorischen Nervenbahnen und greifen weder durch Monoaminoxydase- noch durch Decarboxylase-Hemmung in den Stoffwechsel des Dihydroxy-phenylalanins (DOPA) ein.
Sie weisen - insbesondere bei peroraler Verabreichung- eine sehr geringe Toxizität auf und werden auch in relativ hohen Dosen während längerer Zeit gut vertragen.
Die neuen Guanidin-Derivate sollen deshalb als Heilmittel Verwendung finden, insbesondere zur Behandlung der Hypertonie, wobei sie - als freie Basen oder in Form physiologisch verträglicher, wasserlöslicher Säureadditionssalze - allein oder in geeigneten Arzneiformen enteral oder parenteral verabreicht werden können.
In den nachfolgenden Beispielen erfolgen alle Temperaturangaben in Celsiusgraden, ohne Korrekturen.
Beispiel 1 N-[2-(cis-2,6-Dimethylpiperidino)-äthylamino]- guanidin und N-Amino-N-[2-(cis-2,6-dimethylpiperidino)-äthylj- guanidin
Eine Lösung von 1,71 g 1-(2-Hydrazinoäthyl)-cis2,6-dimethylpiperidin und 1,75 g l-Guanyl-3,5-dime- thylpyrazol-hydrochlorid in 30 ml Chloroform wird
1 Std. am Rückfluss zum Sieden erhitzt. Man dampft im Vakuum ein, löst den Rückstand in Wasser und schüttelt zur Entfernung des gebildeten 3,5-Dimethyl- pyrazols dreimal mit Methylenchlorid aus. Die wässerige Lösung wird hierauf im Vakuum eingedampft und getrocknet.
Aus dem erhaltenen Rohprodukt wird entweder nach a) die erste oder nach b) die zweite der im Titel genannten Komponenten isoliert: a) Das Rohprodukt wird in Methylenchlorid aufgenommen, wobei eine Lösung entsteht, die angeimpft und 5 Std. bei 0 stehen gelassen wird. Man filtriert die Kristalle ab, nimmt sie in etwa 15 mi Methylenchlorid auf und erhitzt die Suspension kurz zum Sieden; die ungelöste Substanz wird abfiltriert und im Vakuum ge trocknet.
Das so erhaltene N-[2-(cis-2,6-Dimethylpiperidino)-äthylamino] guanidin-monohydrochlorid wird aus Äthanol-Äther oder aus Methanol-Äther um kristallisiert; Smp. 153-1560. b) Man löst das Rohprodukt in wenig Äthanol, ver setzt mit 1 äquivalent einer äthanolischen Chlorwasser stofflösung, impft an und lässt im Kühlschrank stehen, wobei das
N-Amino-N-[2-(cis-2, 6-dimethylpiperidino)-äthyl]- guanidin-dihydrochlorid auskristallisiert; Smp. 221-2220 (aus Äthanol).
Das als Ausgangsprodukt benötigte 1-(2-Hydrazino äthyl)-cis-2, 6-dimethylpiperidin kann folgendermassen hergestellt werden: cis-2, 6-Dimethylpiperidin wird mit Äthylenoxyd in Gegenwart einer Spur Wasser während 18 Std. im Autoklav auf 1000 erhitzt. Das entstandene 1-(2-Hydr- oxyäthyl)-cis-2,6-dimethylpiperidin setzt man in abs.
Benzol bei Siedetemperatur und unter Rühren mit Thionylchlorid um, wobei 1-(2-Chloräthyl)-cis-
2, 6-dimethylpiperidin-hydrochlorid vom Smp. 177L1780 erhalten wird. Das Chloräthyl Derivat wird schliesslich in Chloroformlösung während mehrerer Stunden mit Hydrazinhydrat bei Siedetemperatur gerührt; nach dem Abkühlen trennt man die Chloroformschicht ab, extrahiert die wässerige Phase mit Chloroform, dampft die vereinigten, über Magnesiumsulfat getrockneten organischen Phasen ein und fraktioniert den Rückstand im Vakuum. Sdp. des 1 -(2-Hydrazinoäthyl)-cis- 2,6-dimethylpiperidins 128-1330/11 mm Hg.
Beispiel 2 N-Amino-N-[2-(cis-2, 6dimethylpiperidino)- äthyl]-guanidin
Eine Lösung von 0,83 g Cyanamid in 20 ml abs.
Äthanol wird bei Raumtemperatur mit 3,4 g 1-(2-Hydrazinoäthyl)-cis-2,6-dimethylpiperidin und anschlie ssend mit 0,55 ml konz. Schwefelsäure versetzt. Man erhitzt während 20 Std. auf 1050 (Ölbadtemperatur), kühlt auf 0 ab und filtriert das kristalline Hemisulfat der im Titel genannten Verbindung ab; Smp. 2542580.
Durch Zugabe eines halben Mol Schwefelsäure zu der heissen Lösung des Hemisulfates in Methanol wird das Sulfat erhalten. Man kühlt ab, filtriert das kristalline Salz ab und wäscht mit Methanol nach. Smp. 2782800.
Hydrochlorid: Smp. 1;68-1700 (Zers.), erhalten durch Behandlung des Hemisulfats mit einem durch Natronlauge aktivierten Anionenaustauscher und Versetzen der Base mit Chlorwasserstoff.
Beispiel 3 N-Amino-N-[2-(trans-2,6-dimethylpiperidino)- äthyl]-guanidin
Zu einer Lösung von 0,83 g Cyanamid in 20 ml Äthanol werden unter Rühren in einer Portion 3,4 g I-(2-Hydrazinoäthyl)-trans-2,6-dimethylp zugegeben, wobei leichte Erwärmung eintritt. Unter weiterem Rühren werden darauf innerhalb von 7 Min.
0,54 ml Schwefelsäure zugetropft, wobei ein Temperaturanstieg bis 490 erfolgt. Die Reaktionslösung wird anschliessend während 20 Std. am Rückfluss (Ölbadtemperatur 1050) erhitzt, wobei sich nach etwa 45 Min. das Hemisulfat der im Titel genannten Verbindung kristallin abscheidet. Man kühlt auf 00, filtriert und trocknet die Kristallmasse während 4 Std. im Vakuum bei 800.
Das erhaltene Hemisulfat wird in 100 ml Methanol unter Erwärmen gelöst; die methanolische Lösung wird durch hochgereinigte Fullererde filtriert, im Vakuum auf etwa 50 ml eingeengt, bei 400 mit 0,366 ml Schwefelsäure versetzt und auf 100 abgekühlt, wobei das Sulfat sich kristallin ausscheidet. Man filtriert ab und wäscht den Filterrückstand mit 10 ml eiskaltem Methanol nach. Nach Umkristallisieren aus Wasser/Äthanol schmilzt das Sulfat der im Titel genannten Verbindung bei 2860 unter Zersetzung.
Das als Ausgangsprodukt benötigte 1-(2-Hydrazino- äthyl)-trans-2,6-dimethylpiperidin wird - analog wie in Beispiel 1 beschrieben - aus trans-2,6-Dimethylpiperidin hergestellt; Sdp. 123-1250/11 mm Hg.
Beispiel 4
N-Amino-N-[2-(cis-2,6-dimethylpiperidino)- äthyl] -methylguam.din
Eine Lösung von 3,4 g 1-(2-Hydrazinoäthyl)-cis2,6-dimethylpiperidin in 40 ml Wasser wird in einer Portion mit 4,6 g N,S-Dimethyl-isothioh arnstoffj odid versetzt. Die Reaktionslösung wird 120 Std. bei Raumtemperatur stehengelassen und Idarauf im Vakuum bei 650 eingeengt. Der ölige Rückstand wird mit einem alkalischen Anionenaustauscher behandelt; die wässerige Lösung, welche die im Titel genannte freie Base enthält, wird konzentriert und der Eindampfrückstand 2mal mit abs. Äthanol im Vakuum abgedampft. Der ölige Rückstand wird mit 7,5 ml 15,5 % iger Schwefelsäure auf pH 6,0 eingestellt und die Lösung im Vakuum vollständig eingedampft.
Das Sulfat der im Titel genannten Verbindung konnte nicht kristallin erhalten werden.
Beispiel 5 N-Amino-N-[2-(cis-2, 6-dimethylpiperidino)- äthyl]-guanidin
Eine Lösung von 5,0 g 1-(2-Hydrazinoäthyl)-cis 2,6-dimethylpiperidin in 60 ml Wasser wird mit 4,4 g S-Methyl-isothioharnstoffsulfat versetzt; das Gemisch wird während 90 Std. bei Zimmertemperatur stehengelassen, wobei vollständige Lösung eintritt und Methylmercaptan entsteht. Zur Beendigung der Reaktion wird die Lösung während 2 Std. bei 500 erwärmt und anschliessend im Vakuum zur Trockne eingedampft. Der Rückstand wird mit Äthanol verrieben, wobei sich ein kristalliner Niederschlag ausscheidet; nach einmaligem Umkristallisieren aus Äthanol/Wasser schmilzt das Hemisulfat der im Titel genannten Verbindung bei 254-2560.
Dihydrobromid
2,0 g des obigen Hemisulfats werden in 10 ml Wasser gelöst und mit 30,0 g eines durch Natronlauge aktivierten Anionaustauschers behandelt. Die Lösung der erhaltenen Base wird eingeengt und unter Kühlen mit überschüssiger wässeriger Bromwasserstoffiösung versetzt. Hierauf dampft man im Vakuum zur Trockne ein und kristallisiert den Rückstand aus Äthanol; das erhaltene Dihydrobromid der im Titel genannten Verbindung schmilzt bei 172-1730.
Beispiel 6 N-AminN-[2-(2-äthylpiperidino)-äthyl]-gu anidin
Eine Lösung von 5,0 g 1-(2-Hydrazinoäthyl)-2 äthylpiperidin in 60 ml Wasser wird mit 4,4 g S-Methyl- isothioharnstoffsulfat versetzt; die Reaktionsmischung wird während 20 Std. auf 300 erwärmt, wobei vollständige Lösung eintritt und Methylmercaptan entsteht.
Darauf wird im Vakuum zur Trockne eingedampft und der Rückstand mit Äthanol verrieben, wobei sich ein kristalliner Niederschlag ausscheidet. Nach Umkristal lisieren aus Wasser/Athanol schmilzt das Hemisulfat der im Titel genannten Verbindung bei 259-2620 (Zers.).
Das Ausgangsprodukt kann z. B. folgendermassen hergestellt werden: a) 1 -(2-Hydroxyäthyl)-2-äthylpiperidin
45,2 g 2-l2ithylpiperidin, 17,6 g Äthylenoxyd und 1 ml Wasser werden im Autoklav während 18 Std. auf 1000 erhitzt. Nach Abkühlen wird die Reaktionslösung im Vakuum fraktioniert, wobei das 1-(2 Hydroxyäthyl)-2-äthylpiperidin zwischen 108-1100 bei 13 mm Hg überdestilliert. b) 1 -(2-Chlor äthyl) -2-äthylpiperidin
Zu einer Lösung von 52,7 g 1-(2-Hydroxyäthyl)-2 äthylpiperidin in 330 ml abs. Benzol werden unter Rühren bei 20-300 79,4 ml Thionylchlorid zugetropft. Darauf wird das Reaktionsgemisch langsam zum Sieden erhitzt und anschliessend 2 Std. unter weiterem Rühren auf Siedetemperatur gehalten.
Das Benzol und das überschüssige Thionylchlorid werden abdestilliert; der gelbe, ölige Rückstand wird auf Raumtemperatur abgekühlt und mit Äther verrieben, wobei sich das 1-(2-Chlor äthyl)-2-äthylpiperidin-hydrochlorid kristallisiert ausscheidet. Smp. 156-1570 nach Umkristallisieren aus Äthanol/Äther. c) 1 -(2-Hydrazinoäthyl)-2-äthylpiperidin
Zu einer Lösung von 200 ml Hydrazinhydrat wird bei Siedetemperatur unter Rühren innerhalb von 15 Min. eine Lösung von 21,2 g 1-(2-Chloräthyl)-2 äthylpiperidin-hydrochlorid in 39,8 ml Wasser zugetropft. Die Reaktionsmischung wird anschliessend während 20 Std. zum Sieden erhitzt, wobei sich eine ölige Ausscheidung bildet. Nach Abkühlen auf Raumtemperatur wird das Reaktionsgemisch mit 10,0 g Kaliumhydroxyd versetzt und portionsweise mit insgesamt 600 ml Äther extrahiert.
Die Ätherextrakte werden vereinigt, über Magnesiumsulfat getrocknet und eingedampft; der Rückstand wird im Vakuum fraktioniert, wobei das 1 -(2-Hydrazinoäthyl)-2-äthylpiperidin zwi- schen 136-1370 bei 13 mm Hg als farbloses Ö1 überdestilliert.
Beispiel 7 N-Amino-N-2-(3 -methyipiperidino)- äthyl]-guanadin
Zu einer Lösung von 0,83 g Cyanamid in 20 ml Äthanol gibt man bei Raumtemperatur und unter Rühren in einer Portion 3,1 g 1-(2-Hydrazinoäthyl)-3-methylpiperidin. Unter weiterem Rühren werden innerhalb von 7 Min. 1,08 ml konz. Schwefelsäure zugetropft, wobei ein Temperaturanstieg bis 530 erfolgt. Die Reaktionslösung wird anschliessend während 16 Std. am Rückfluss erhitzt. Nach Abkühlen auf 50 wird der kristalline Niederschlag abfiltriert und mit Äthanol nachgewaschen. Nach Umkristallisieren aus Wasser/Äthanol schmilzt das Sulfat der im Titel genannten Verbindung bei 246-2480 (Zers.).
Das als Ausgangsprodukt benötigte 1-(2-Hydrazino äthyl)-3-methylpiperidin kann wie folgt hergestellt werden:
3-Methylpiperidin wird mit Äthylenoxyd in Gegenwart einer Spur Wasser während 18 Std. im Autoklav auf 1000 erhitzt. Das entstandene 1-(2-Hydroxyäthyl)3-methylpiperidin setzt man in abs. Benzol bei Siedetemperatur und unter Rühren mit Thionylchlorid um, wobei 1-(2-Chloräthyl)-3-methylpiperidin vom Sdp.
820/14 mm Hg erhalten wird. Das Chloräthyl-Derivat wird schliesslich in Chloroform während mehrerer Stunden mit Hydrazinhydrat bei Siedetemperatur gerührt; nach dem Abkühlen trennt man die Chloroformschicht ab, extrahiert die wässerige Phase mit Chloroform, dampft die vereinigten, über Magnesiumsulfat getrockneten organischen Phasen ein und fraktioniert den Rückstand im Vakuum. Sdp. 122-1300/15 mm Hg.
Beispiel 8
N-Amino-N-[2-(4-methylpiperidino) äthyl]-guanidin
Zu einer Lösung von 0,83 g Cyanamid in 20 ml Äthanol gibt man unter Rühren bei Raumtemperatur in einer Portion 3,12 g 1-(2-Hydrazinoäthyl)-4-methylpiperidin. Unter weiterem Rühren werden innerhalb von 7 Min. 1,08 ml konz. Schwefelsäure zugetropft, wobei Erwärmung bis 530 erfolgt. Die Reaktionslösung wird anschliessend während 5 Std. am Rückfluss erhitzt. Nach Abkühlen auf 50 wird der kristalline Niederschlag abfiltriert und mit Äthanol gewaschen.
Das Sulfat der im Titel genannten Verbindung schmilzt bei 239-243 (Zers.).
Das als Ausgangsprodukt benötigte 1-(2-Hydrazino äthyl)-4-methylpiperidin wird - analog wie in Beispiel 6 beschrieben - aus 4^Methylpiperidin hergestellt. Sdp.
122-1260/14 mm Hg.
Beispiel 9
N-Amino-N-[2-(4-methylpiperidino) äthyl]-methylguanidin
Eine Lösung von 3,1 g 1 -(2-Hydrazinoäthyl)-4- methylpiperidin in 20 ml Wasser wird mit 2,3 g N,S- Dimethyl-isothioharnstoffjodid versetzt und 90 Std. bei Zimmertemperatur stehengelassen. Anschliessend dampft man die Lösung im Vakuum zur Trockne ein und behandelt den Rückstand mit einem alkalischen Ionenaustauscher. Das die im Titel genannte freie Base enthaltende Eluat wird eingeengt und unter Kühlen mit 2 Äquivalenten einer äthanolischen Chlorwasserstofflösung versetzt; die Lösung wird eingeengt und nach Zugabe von wenig Äther stehengelassen. Nach einiger Zeit beginnt das Dihydrochlorid der im Titel genannten Verbindung zu kristallisieren; Smp. 198-2030 (Zers.).
Das Salz ist schwach hygroskopisch.
Beispiel 10
N-Amino-N-[2-(4,4-dimehylpiperidino)- äthyl]-guanidin
1,25 g 1-(2-Hydrazinoäthyl)-4,4-dimethylpiperidin in 20 ml Wasser werden mit 1,1 g S-Methyl-isothioharn stoffsulfat versetzt. Das Gemisch wird 3 Std. auf 600 erwärmt, wobei vollständige Lösung eintritt und Methylmercaptan entsteht. Man lässt die Reaktionslösung über Nacht bei Zimmertemperatur sbehen, dampft anschlie ssend zur Trockne ein und verreibt den Rückstand mit abs. Äthanol. Das ausgeschiedene amorphe Hemisulfat der im Titel genannten Verbindung wird abfiltriert und aus Wasser/Abhanol kristallisiert; Smp. 282-2860 (Zers.). Das Salz ist schwach hygroskopisch.
Das als Ausgangsmaterial verwendete 1-(2-Hydra- zinoäthyl)-4,4-dimethylpiperidin wird - wie in Beispiel 6 beschrieben - aus 4,4-Dimethylpiperidin durch Hydroxyäthylierung mit Äthylenoxyd, Umsetzung mit Thionylchlorid und anschliessende Behandlung mit Hydrazinhydrat erhalten. Sdp. 139-142 /12 mm Hg.
Beispiel 11
N-Amino-N-[2-(4-äthyl-2-methylpiperidino) äthyl]-guanidin
Eine Lösung von 2,5 g 1-(2-Hydrazinoäthyl)-4 äthyl-2-methylpiperidin in 40 ml Wasser wird mit 2,2 g S-Methyl-isothioharnstoffsulfat versetzt. Das Gemisch wird während 85 Std. bei Zimmertemperatur stehen gelassen, wobei vollständige Lösung eintritt und Methylmercaptan entsteht.
