Verfahren zur Herstellung von N,N-aliphatisch disubstituierten
5-Dialkylsulfamyl-anthranil-säureamiden
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von N,N-aliphatisch substituierten 5-Dialkylsulfamyl-anthranilsäureamiden mit unsubstituierter oder acylierter 2-Aminogruppe und gegebenenfalls weiteren Substituenten im aromatischen Ring und gegebenenfalls ihrer Salze.
Substituenten der Carbamylgruppe sind insbesondere ein- oder zweiwertige niedere aliphatische Kohlenwasserstoffreste, die auch durch Heteroatome, wie Sauerstoff-, Stickstoff-oder Schwefelatome, unterbrochen und/oder durch funktionelle Gruppen, wie Hydroxy oder Aminogruppen, substituiert sein können.
Als einwertige Kohlenwasserstoffreste dieser Art seien beispielsweise genannt: niederes Alkyl, wie Methyl, Äthyl, n-Propyl, Isopropyl, gerades oder verzweigtes, in beliebiger Stellung gebundenes Butyl, Pentyl, Hexyl oder Heptyl, niederes Alkenyl, wie Allyl oder Methallyl. Entsprechende, durch die genannten Heteroatome unterbrochene Reste sind z. B.: niederes Alkoxyalkyl, Alkylmercaptoalkyl oder Mono- oder Dialkylaminoalkyl, wie 2-Methoxy-äthyl, 2-Äthoxy-äthyl, 3-Methoxypropyl, 2 Methyl-mercaptoäthyl, oder Dimethyl-, Methyl-äthyloder Diäthylamdno-äthyl oder -propyl; durch funktionelle Gruppen substituierte Reste sind z.
B. : niederes Hydroxy- oder Aminoalkyl, wie 2Hydroxy- oder -Amino- äthyl oder -propyl. Für zweiwertige aliphatische Reste der genannten Art stehen beispielsweise: niederes Alkylen, wie Butylen-(1,4), Pentylen-(1, 5), Hexylen-(1, 5), Hexylen-(1,6) oder Heptylen-(2,6), oder niederes Oxa-, Aza- oder Thiaalkylen, wie 3-Oxa-, 3-Aza- oder 3 Thiapentylen-(1,5) oder -hexylen-(1,6), 3-Methyl-, 3 Äthyl- oder 3-Propyl-3-aza-pentylen-(1,5) oder -hexy len-(1,6), oder 4-Methyl-4-aza-heptylen-(2,6), oder durch funktionelle Gruppen substituierte Reste dieser Art, wie 3-(ss-Hydroxyäthyl)
-3-aza-pentylen-(1 5).
Die in 5-Stellung der neuen Verbindungen befindliche Dialkylsulfamylgirippe enthält z. B. die oben genannten niederen Alkylgruppen. Die in 2-Stellung vorhandene Aminogruppe kann auch durch einen Acylrest substituiert sein.
Als Acylreste seien insbesondere Reste aliphatischer, araliphatischer, aromatischer Carbonsäuren genannt, z. B. Carbalkoxyreste, wie Methoxy- oder Äthoxycarbonyl oder Fettsäurereste, wie Niederalkanoylreste, z. B. Acetyl, Propionyl, Butyryl, Pivalyl, Valeroyl, Caproyl, Önanthoyl, Lauroyl oder Phenylniederalkanoylreste, wie Phenylacetyl oder Benzoyl. In den angeführ- ten Acylresten können Ringe aromatischen Charakters noch z. B. durch Niederalkyl oder -alkoxy, Halogen oder Trifluormethyl substituiert sein.
Die neuen Verbindungen können, wie gesagt, am aromatischen Ring, d. hi in 3-, 4 und/oder 6-Stellung substituiert sein. Als Substituenten für diese Stellungen kommen insbesondere Halogen-, wie Fluor- oder Chloratome, Trifluormethyl-oder Niederalkylgruppen, z. B. die genannten, oder Niederalkoxygruppen, wie Methoxy-, ithoxy-, Propoxy- oder Butoxygruppen, in Frage.
Die neuen Verbindungen besitzen wertvolle pharmakologische, insbesondere antiinflammatorische und antiallergische Eigenschaften. So unterdrücken sie z. B. an Ratte, Meerschweinchen und Kaninchen entzündliche Prozesse und allergische Reaktionen. Sie können dementsprechend als Antiphlogistika und Antiallergika verwendet werden. Ferner besitzen sie, z. B. im Tierversuch an der Ratte, eine blutdrucksenkende Wirkung und können als Hypotensiva verwendet werden. Sie sind auch wertvolle Zwischenprodukte für die Herstellung weiterer, insbesondere pharmakologisch anwendbarer Stoffe.

Das erfindungsgemässe Verfahren zur Herstellung der neuen Verbindungen ist dadurch gekennzeichnet, dass man eine entsprechende 5-DiaLkylsulfamyl-anthra- nilsäure mit freier oder acylierter 2-Aminogruppe, oder ein reaktionsfähiges funktionelles Säurederivat davon, mit einem sekundären, nur aliphatische Substituenten enthaltenden Amin umsetzt und, wenn erwünscht, erhaltene 2-Aminoverbindungen acyliert oder erhaltene 2-Acylaminoverbindungen hydrolysiert.
Reaktionsfähige funktionelle Säurederivate von 5 Dialkylsulfamyl-anthranilsäuren oder ihren Acylderivaten sind beispielsweise deren Halogenide, wie das Chlorid oder Bromid, reine oder gemischte Anhydride, z. B. gemischte Anhydride mit Kohlensäuremonoalkylestern, wie Kohlensäuremonoäthyl- oder -isobutylester, oder Anhydride, wie z. B. innere Anhydride von N-Acyl-5 (dialkylsulfamyl)-anthranilsäuren, d. h. entsprechende 6 (Diaikylsulfamyl)-4-H-3, 1-benzoxazin-4-one, wie z. B. innere Anhydride von N-Acetyl-5-(dialkylsulfamyl)anthranilsäuren oder 5-(Dialkylsulfamyl)-isatosäurean- hydride, oder innere Anhydride von N-(a-Hydroxyalkyl) 5-(dialkylsulfamyl)-anthranilsäuren, wie z. B. von N Hydroxymethyl-5-(dialkylsulfamyl)-anthranilsäuren, oder Ester, z. B.
Alkyl- oder Aralkylester, wie Methyl-, Äthyl- oder Benzylester, aktivierte Ester, wie Cyanmethylester, Amide oder Azide.
Die verfahrensgemässen Umsetzungen kann man in Anwesenheit eines Lösungs oder Verdünnungsmittels, in An- oder Abwesenheit von Reaktionsbeschleunigern, bei Raumtemperatur oder vorzugsweise bei erhöhter
Temperatur, falls erforderlich, in geschlossenem Gefäss unter Druck und/oder unter einem Inertgas, z. B. Stick stoff, durchführen. Gegebenenfalls verwendet man Kon densationsmittel, z. B., bei Verwendung der freien Säure,
Carbodiimide, wie Dicycloalkylcarbodiimide, oder bei
Verwendung von Säurehalogeniden basische Konden sationsmittel oder einen Überschuss an sekundärem Amin.
In den erhaltenen Verbindungen können nach bekannten Methoden weitere Substituenten eingeführt oder vorhandene abgewandelt oder abgespalten werden. So können erhaltene 2-Aminoverbindungen z. B. durch Umsetzung mit reaktionsfähigen funktionellen Deriva ten von Carbonsäuren, wie Anhydriden oder Halogeniden, acyliert oder erhaltene 2-Acylaminoverbindungen, insbesondere durch Umsetzung mit alkalischen Mitteln, hydrolysiert werden.
Die genannten Reaktionen kann man in üblicher Weise in An- oder Abwesenheit von Lösungs oder Ver dünnungsmitteln, sauren oder basischen Kondensationsmitteln und/oder Katalysatoren bei erniedrigter, gewöhnlicher oder erhöhter Temperatur, gegebenenfalls im geschlossenen Gefäss unter erhöhtem Druck und/ oder unter einer Inertgasatmosphäre durchführen.
Je nach den Verfahrensbedingungen und Ausgangsstoffen erhält man die Endstoffe in freier Form oder in Form ihrer Salze. So können beispielsweise basische, neutrale, saure oder gemischte Salze, gegebenenfalls auch Hemi-, Mono-, Sesqui- oder Polyhydrate davon erhalten werden. Die Salze der Endstoffe können in an sich bekannter Weise, z. B. mit Alkalien oder Ionenaustauschern, in die freien Basen übergeführt werden.
Von den letzteren lassen sich durch Umsetzung mit organischen oder anorganischen Säuren, insbesondere solchen, die zur Bildung therapeutisch verwendbarer Salze geeignet sind, Salze gewinnen. Als solche Säuren seien beispielsweise genannt: Halogenwasserstoffsäuren, Schwefelsäuren, Phosphorsäuren, Salpetersäure, Per chiorsäure; aliphatische, alicyclische, aromatische oder heterocyclische Carbon- oder Sulfonsäuren, wie Ameisen-, Essig--, Propion-, Bernstein-, Glykol-, Milch-, Sipfel-, Wein-, Zitronen-, Ascorbin-, Malein-, Hydroxy malein- oder Brenztraubensäure;
Phenylessig-, B enzoe-, p-Amino-benzoe-, Anthranil-, p-Hydroxy-benzoe-, Salicyl-, p-Aminosalicyl- oder Embonsäure, Methansulfon-, 22ithansulfon-, Hydroxyäthansulfon-, Äthylensulfonsäure; Halogenbenzolsulfon-, Toluolsulfon-, Naphthalinsulfonsäuren oder Sulfanilsäure; Methionin, Tryptophan, Lysin oder Arginin.
Diese oder andere Salze der neuen Verbindungen, wie z. B. die Pikrate, können auch zur Reinigung der erhaltenen Basen dienen, indem man die Basen in Salze überführt, diese abtrennt und aus den Salzen wiederum die Basen freimacht.
Die Ausgangsstoffe sind bekannt oder, falls ; sie neu sind, lassen sie sich nach an sich bekannten Methoden herstellen. Beispielsweise kann man sie zum Teil nach den oben beschriebenen Verfahren unter sinngemässer Abänderung der Arbeitsweise oder der Wahl der Ausgangsstoffe gewinnen.
Die neuen Verbindungen können als Heilmittel, z. B. in Form pharmazeutischer Präparate, Verwendung finden, welche sie oder ihre Salze in Mischung mit einem für die enterale, parenterale oder topicale Applikation geeigneten pharmazeutischen organischen oder anorganischen, festen oder flüssigen Trägermaterial enthalten.
Die neuen Verbindungen können auch in der Veterinär medizin, z. B. in einer der oben genannten Formeln oder in Form von Zusatzmitteln für Tierfutter verwendet werden.
In den folgenden Beispielen sind die Temperaturen in Celsiusgraden angegeben.
Beispiel 1
3,2 g Anhydro- [N-acetyl-5- (dimethylsulfamyl) anthranilsäurej [6-(Dimethylsullamyl)-2-methyl-4H-3, 1 - benzoxazin-4-on] werden in Gegenwart von 10 ml N Methylpiperazin während 16 Stunden unter Rückfluss gekocht. Man dampft im Vakuum ein und nimmt den Rückstand in 2n Salzsäure auf. Nach etwa 30 Minuten wird die salzsaure Schicht mit Ammoniak alkalisch gestellt und mit Methylenchlorid extrahiert. Der Rückstand wird aus Äthanol-Petroläther umkristallisiert. Man erhält so das 5-(Dimethylsulfamyl)-anthranilsäure-(N'methyl-piperazid) der Formel

in Form weisser Kristalle vom F. 151-1520. Durch Umsetzen desselben mit alkoholischer Salzsäure erhält man das Monohydrochlorid vom F. 280-2820.
Die als Ausgangsmaterial verwendete Anhydro-[N acetyl-5-(diraethylsulfamyl)-anthranilsäure] kann folgendermassen hergestellt werden:
4,5 g 5-(Dimethylsulfamyl)-anthranilsäure werden in Gegenwart von 20 ml Acetanhydrid 2 Stunden auf 100-1050 erhitzt. Man kühlt auf Zimmertemperatur ab und filtriert die ausgeschiedenen Kristalle, die bei 186 bis 1880 schmelzen, ab. Das so erhaltene Rohprodukt kann für die oben erwähnte Reaktion direkt verwendet werden.
Beispiel 2
2 g 5-(Dimethylsulfamyl)-isatosäureanhydrid werden in Gegenwart von 20 ml N-Methylpiperazin 16 Stunden unter Rückfluss gekocht. Man dampft dann im Vakuum ein, nimmt den Rückstand in 2n Salzsäure auf und extrahiert mit Essigester. Die salzsaure wässerige Schicht wird mit Ammoniak alkalisch gestellt und mit Methylenchlorid extrahiert. Der feste Rückstand wird aus Äthanol-Petroläther umkristallisiert. Man erhält so das 5 (DimethylsulfamyD-anthranilsäure-(N'-methyl-piperazid) vom F. 151-1520, das mit dem im Beispiel 1 beschriebenen Produkt identisch ist.
Beispiel 3
2 g 5 -(Dimethylsulfamyl)-is atosäureanhydrid werden in Gegenwart von 20 ml Morpholin während 16 Stunden unten Rückfluss gekocht. Man dampft dann im Vakuum ein, versetzt mit Wasser und Eisessig, bis das Reaktionsgemisch sauer ist, neutralisiert mit Natriumhydrogencarbonat und extrahiert mit Essigester. Der Essigester¯Rückstand wird aus Äthanol-Petroläther umkristallisiert.
Man erhält so das 5-(Dimethylsulfamyl)- anthranilsäuremorpholid der Formel

Das in diesem Beispiel als Ausgangsmaterial verwendete 5-(Dimethylsulfamyl)-isatosäureanhydrid wird folgendermassen hergestellt:
15 g 2-Amino-5- (dimethylsulfamyl)- benzoesäure werden in Gegenwart von 90 ml Chlorameisensäure- äthylester 40 Stunden unter Rückfluss gekocht. Man kühlt ab und filtriert die ausgeschiedenen Kristalle vom F. 215-2160 ab, die wie oben sofort weiter verarbeitet werden.
In analoger Weise wie in den Beispielen 1 bis 3 beschrieben, kann man die folgenden Verbindungen erhalten: a) 5-(Dimethylsulfamyl)-anthranilsäure-[N'- (ss-hy- droxyäthyl)-piperazid], F. 98-100 . Das Monohydrochlorid schmilzt bei 245-2470. b) 5-(Diäthylsulfamyl)-anthranilsäure- (N'-methyl- piperazid), F. 141450. Das Hydrochlorid schmilzt bei 2650. c) 5-(Dimethylsulfamyl) - anthranilsäure - (N'- äthyl - piperazid), F. 150-1510. Das Hydrochlorid schmilzt bei 266-2670. d) 5- (Dimethylsulfamyl)-anthranilsäure-piperidid, F.
153-1540 e) 5- (Dimethylsulfamyl) - anthranilsäurediäthylamid, F. 1251270. f) 5-(Dimethylsulfamyl)-anthranilsäure-[N-äthyl - N (ss-dimethylamino-äthyl)-amid], F. 137-1380. g) 5-(Dimethylsuffamyl)-anthranilsäure - di - (ss - hy - droxyäthyl)-amid], F. 126-1270.
Beispiel 4
Eine Lösung von 10 g 5-(Dimethylsulfamyl)-anthra- nilsäure-(N'-methyl-piperazid) und 3 ml absolutem Pyridin in 100 ml absolutem Dioxan wird auf 150 abgekühlt und tropfenweise mit einer Lösung von 2,65 g Acetylchlorid in 50 ml absolutem Dioxan versetzt. Man rührt 1 Stunde bei Zimmertemperatur und 1 Stunde bei 500. Man dampft dann die Reaktionssuspension im Vakuum ein. Der Rückstand wird in Wasser gelöst, mit Natriumhydrogencarbonat neutralisiert und mit Chloroform extrahiert. Der Chloroformrückstand liefert nach dem Umkristallisieren aus Äthanol-Petroläther das 5 (Dimethylsullamyl)-2-acetylamino-benzoesäure-(N' - me - thyl-piperazid) der Formel

Beispiel 5
Eine Lösung von 10 g 5-(llimethylsulfamyl)-anthra- nilsäure-(N'-methyl-piperazid) und 3 ml absolutem Pyridin in 100 ml absolutem Dioxan wird auf 150 abgekühlt und tropfenweise mit einer Lösung von 6 g p Chlorbenzoylchlorid in 50 ml absolutem Dioxan versetzt. Man rührt 1 Stunde bei Zimmertemperatur und 1 Stunde bei 500. Nach dem Abkühlen filtriert man ab, extrahiert die feste Substanz mit Chloroform in Gegenwart von 2n Natronlauge. Nach dem Trocknen der Chloroformlösung über Magnesiumsulfat und Abdampfen des Lösungsmittels wird der Rückstand aus Äthanol Methylenchlorid umkristallisiert.
Man erhält so das 5 (Dimethylsulfamyl)-2-(p-chlor - benzoylami.no) - benzoe - säure- (N'-methyl-piperazid) der Formel
