Process for the preparation of new aminoalkyl-substituted 5-membered heterocycles and their salts
EMI1.1
The invention relates to a method of heterocycles of the general formula
EMI1.2
as well as their salts with physiologically compatible inorganic or organic acids.
In the above general formula, R and R2, which can be the same or different, denote straight-chain or branched alkyl radicals with 1-6 carbon atoms, optionally substituted by hydroxyl groups, or alkenyl radicals with 2-6 carbon atoms, cycloalkyl radicals with 6-8 carbon atoms, aralkyl radicals with 7- 9 carbon atoms or aryl radicals with 6-8 carbon atoms or together with the nitrogen atom a 5- to 7-membered heterocyclic ring optionally interrupted by a further hetero atom and / or substituted by lower alkyl radicals or hydroxyl groups;
reg is a hydrogen atom, a straight-chain or branched alkyl radical with 1-8 carbon atoms, optionally substituted by hydroxyl groups and / or by a dialkylamino group, or an alkenyl radical with 2-6 carbon atoms, a cycloalkyl radical with 6-8 carbon atoms, an aralkyl radical with 7-9 carbon atoms, an aryl group with 6-8 carbon atoms or an acyl group;
R4 is a hydrogen atom, a straight-chain or branched alkyl radical with 1-8 carbon atoms, optionally substituted by hydroxyl groups, or an alkenyl radical with 2-6 carbon atoms, a cycloalkyl radical with 6-8 carbon atoms, an aralkyl radical with 7-9 carbon atoms, an aryl radical with 6-8 Carbon atoms or, together with Rg and the nitrogen atom, a 5- to 7-membered heterocyclic ring which is optionally interrupted by a further heteroatom and / or substituted by lower alkyl radicals or hydroxyl groups; A is a straight-chain or branched, saturated or unsaturated, divalent acyclic hydrocarbon radical having 1-6 carbon atoms and X is an oxygen or sulfur atom or the imino group.
The compounds prepared according to the invention are therefore derivatives of 1,3,4-oxadiazole, 1,3,4-thiadiazole and 1,2,4-triazole and its tautomeric forms. The 5- to 7-membered heterocyclic rings mentioned are in particular the pyrrolidine, piperidine, morpholine, piperazine and azepine rings; the acyl radicals mentioned in the definition of Rg are preferably radicals of aliphatic or aromatic carboxylic acids and ring-substituted aromatic sulfonic acids.
According to the invention, the compounds are prepared by methods known from the literature. The following procedure gave the best yields:
Implementation of an imino ether of the general formula
EMI1.3
in which Ri, Rg and A have the meanings given, Alk is a lower alkyl radical and Hal is a halogen atom, with a compound of the general formula
<Desc / Clms Page number 2>
EMI2.1
in which Rg, R4 and X have the meanings given. The reaction is preferably carried out in the presence of an organic solvent, for example in the presence of ethanol or pyridine, at elevated temperatures, preferably at the boiling point of the solvent used, optionally in the presence of a customary hydrogen halide binding agent.
The compounds prepared by the above process can, if appropriate, subsequently be converted into their acid addition salts with physiologically compatible inorganic or organic acids using known methods. Physiologically acceptable acids are, for example, hydrochloric acid,
EMI2.2
Tartaric acid, succinic acid, suitable.
If the process according to the invention gives four compounds in which R3 represents a hydrogen atom, these can optionally be acylated subsequently by known methods, for example by reaction with a halide or anhydride of the corresponding aliphatic or aromatic carboxylic acid. If R, is the radical of a ring-substituted aromatic sulfonic acid, this is preferably introduced by reaction with the appropriately substituted benzenesulfonic acid chloride in an inert solvent in the presence of a hydrogen halide binding agent, for example an inorganic or tertiary organic base.
The new compounds have valuable therapeutic properties, and they are particularly effective
EMI2.3
individual substituents one or the other effect predominates. Thus, the compounds in which X is a sulfur atom or the imino group act in particular cough-inducing without simultaneous analgesic and antipyretic activity.
Compounds in which X signifies an oxygen alom have a good analgesic effect without simultaneous coughing. Compounds in which X is a sulfur atom and Ra is the residue of a ring-substituted aromatic sulfonic acid have a blood sugar lowering effect.

; 46.2 g (0.2 mol) ß-diethylamino-propionic acid iminomethyl ether dihydrochloride, obtained from ß-diethylamino-propionitrile by addition of methanol in dioxane with simultaneous introduction of the stoichiometric amount of HCl, are in 120 ml of absolute ethanol with 18.2 g (0, 2 mol) thiosemicarbazide heated to reflux for 2-3 h.
The solvent is distilled off, the evaporation residue is taken up in 100 ml of water, made alkaline with NaCOg and exhaustively extracted with chloroform. The crude product obtained from the dried chloroform is recrystallized from ethyl acetate while using decolorizing charcoal.
Yield ze F. = 158-1600 C.
Example 2: 2-Acetamido-5-dimethylaminomethyl-1,3,4-thiadiazole hydrochloride.
16 g (0.1 mol) of 2-amino-5-dimethylaminomethyl-1, 3, 4-thiadiazole are refluxed in 100 ml of glacial acetic acid with 20 g (0.2 mol) of acetic anhydride for 2 h. The acetic acid and acetic anhydride are then distilled off. The crude reaction product is taken up in ethanol and converted with the equivalent amount of ethanolic hydrochloric acid into the hydrochloride, which crystallizes out when the solution cools. After recrystallization from methanol, colorless crystals with a melting point of F. = 238-240 ° C. are obtained in a yield of 71%.
Example 3: 2-Benzamido-5-dimethylaminomethy1-1, 3, 4-thiadiazole hydrochloride.
3.2 g (0.02 mol) of 2-amino-5-dimethylaminomethyl-1,3,4-thiadiazole are dissolved in 10 ml of pyridine, and 5.6 g (0.04 mol) of benzoyl chloride are added. The reaction solution is heated to 45-50 ° C. for a few hours. The reaction product precipitates out during the reaction. After recrystallization from ethanol, colorless crystals are obtained in a yield of 74% with a decomposition point of 260-261'C.
EMI2.5
5 hours of reaction between 20-50 C, the reaction mixture is acidified with hydrochloric acid, evaporated to dryness and the dry residue is extracted with hot ethanol. After NaCl has been filtered off with suction, the 2- (o-methoxybenzenesulfonamido) -5-diethylaminomethyl-1,3,4-thiadiazole hydrochloride crystallizes out on cooling the solution.
After recrystallization from water, 65% of theoretical pure product, decomposition point 236-238 ° C., are obtained.
