Verfahren zur Herstellung eines Arylalkanons
Diese Erfindung betrifft Verfahren zur Herstellung chemischer Verbindungen, die ein Kohlenstoffskelett enthalten, das dem der Steroide verwandt ist.
In den vergangenen 20 Jahren waren viele Untersuchungen mit der Synthese der Steroide beschäftigt, einer Gruppe von Substanzen, die für die Medizin von grosser Wichtigkeit ist. Die meisten der industriellen Forschungen waren auf ihre Teil-Synthese aus natürlich vorkommenden Steroiden, die leicht als Ausgangsmaterialien verfügbar sind und die Herstellung von Analogen und Derivaten durch Abänderung der in der Natur gefundenen Steroid-Strukturen, gerichtet.
Es kamen einige Total-Synthesen der Steroide von grossem Scharfsinn und akademischem Verdienst zustande, aber es waren zahlenmässig relativ wenige und im Anwendungsbereich beschränkte. Typisch für diese Synthesen ist die grosse Zahl der Einzelstufen, die nötig sind, hauptsächlich wegen der offenkundigen Unübersichtlichkeit, die die Stereochemie einschliesst.
Viele der Einzelstufen, die bei solchen Synthesen benutzt wurden, verwendeten bekannte Reaktionen unter Standardbedingungen, und in erster Linie ist es wesentlich in der Wahl des Ausgangsmaterials, welcher die Anwendung einer Kombination von Stufen in einer bestimmten Reihenfolge folgt, dass ein neues Konzept gefunden wird; jedoch ist die Zahl der alternativen Stufen und Stufenkombinationen, die dem Steroidchemiker verfügbar sind, so gross, dass er, wenn er einmal die Gesamtanlage einer erfolgreichen Mehrstufensynthese sieht, es ihm oft möglich ist durch Anwendung weiteren Scharfsinns, Zwischenprodukte bei der Synthese auf solche Weise abzuändern, dass andere Steroide oder Steroidanaloge erreicht werden.
Solch eine Abänderung, obgleich auf einem besonderen Zwischenprodukt beruhend, nimmt folglich die Anregung von der Gesamtanlage, die durch die ursprüngliche erfinderische Reihenfolge gezeigt wird, wobei die Abänderung in Endprodukten, die gleich sind oder verschieden, von denen der ursprünglichen Synthesen, resultiert.
Es verhält sich also so, dass die Konzeption einer erfolgreichen mehrstufigen Steroidsynthese in jede neue Stufe in der Synthese einen Schritt an erfinderischem Verdienst einbringt, unabhängig von und in Unterstützung zu irgendeiner erfinderischen Konzeption, welche diese besondere Stufe aufweisen kann, betrachtet in Isolierung von anderen Stufen. Die vorliegende Erfindung betrifft eine besondere Stufe in einer neuen Totalsynthese von Steroiden.
Ein neuer Weg zur Totalsynthese von Steroidhormonen hat eine Anzahl von wertvollen Merkmalen.
Dieser Weg hat den Vorteil, dass die zur Bildung des Ringsystems eines Steroids wesentlichen Kohlenstoffatome alle in einer frühen Stufe bei der Synthese vorhanden sind, wobei die frühe Skelettbildung bei der Ausführung einer Hoch-stereospezifischen Synthese mithilft. So wird eine eckständige Methyl-Gruppe, gebunden in 13-Stellung in dem Steroidmolekül in einer frühen Stufe erzeugt, wobei die Herstellung eines ungünstigen Verhältnisses von Stereoisomeren vermieden wird, die allgemein auftritt, wenn Versuche gemacht werden diese Methyl-Gruppe in einer späteren Stufe einzuführen. Mit diesem neuen Weg können analoge Hormone leichter erreicht werden, die nicht oder be stenfalls nicht ohne ernsthafte Schwierigkeiten aus Natürlich-Steroiden verfügbar sind.
So wurde ein Zugang zu Steroiden verschiedener Arten verschafft, die einen Sauerstoff-Substituenten in der ll-Stellung haben; ferner ermöglicht der neue Weg die Synthese von Steroiden, die eine Vielzahl von Substituenten in der 3-Stellung haben. Der neue Weg macht ebenso die Herstellung von homologen Steroiden möglich, die eckständige Gruppen, grösser als Methyl in der 13 Stellung haben.
Letztlich schafft der Weg neue Totalsynthesen von Östron und Östradiol, zwei wertvollen therapeutischen Substanzen. Diese können leicht durch die neuen Synthesen mit guter Gesamtausbeute und durch eine effektvoll kleinere Zahl von Stufen als Gesamtsynthesen bisher verlangt haben, erreicht werden. Der neue Weg wird im folgenden für eine Totalsynthese von Östron dargelegt, wobei die römischen Ziffern Überweisungszeichen zu auf dem beiliegenden Zeichnungsblatt wiedergegebenen Strukturformeln sind.
Die bekannte Verbindung 3-m-Methoxyphenylpropan-l-ol ()wird zu dem entsprechenden Bromid (11) umgewandelt, welches zur Kondensation mit Natrium-acetylid in flüssigem Ammoniak oder anderen geeigneten Lösungsmitteln veranlasst wird unter Bildung des Acetylens (III). Diese Verbindung, deren geeigneten Lösungsmitteln veranlasst wird under Mannich-Reaktion unterworfen, wobei Formaldehyd (oder ein Formaldehyd-Polymerisat) und Diäthylamin verwendet wird, um das acetylenische Amin (1V) zu ergeben.
Die nächste Stufe ist die Hydratisierung der acetylenischen Bindung unter geeigneten Bedingungen, beispielsweise mit wässrigem Quecksilber-II-Sulfat und Schwefelsäure, um das 3-Ketoamin (V) zu ergeben.
Dieses 3-Ketoamin, welches leicht Diäthylamin, sogar bei Destillation ausscheidet, sein Ausscheidungsprodukt, das Vinyl-keton (Vp oder ein Gemisch von beiden wird mit 2-Methylcyklohexan-1,3-dion (VII) unter Rückfluss in Benzol, das Pyridin enthält, kondensiert. Das Produkt dieser Michael-Kondensation ist das dicyklische Triketon (VIII), welches das ganze KohlenstoffskeIett eines l9-nor-D-homo-Steroids enthält.
Rückflussbehandlung dieses Triketons in Xylol, in Gegenwart von Triäthylammoniumbenzoat als Katalysator, bringt eine innere Aldolkondensation und Dehydratisierung, welche das tricyklische Diketon (IX) bildet, das Ungesättigtheit in dem neugebildeten Ring aufweist, welcher dem Ring C bei der 8 (14) Stellung des Endsteroids entspricht. Dieses tricyklische Diketon ist ein Gemisch von Enantiomeren, welches ein asymmetrises Zentrum bei Cig gegenwärtig hat.
Nur die 13-beta-Verbindung wird dargestellt, obgleich dies so verstanden werden muss, dass das Gemisch eine gleiche Menge der 13-alfa-Verbindung wie die folgenden Verbindungen enthält; Auflösung zur Bil dung eines 13-beta-Enantiomers kann bei irgendeiner geeigneten Stufe erreicht werden.
Die katalytische Hydrierung des tricyklischen Diketongemisches sättigt den Ring C und ergibt ein öliges Gemisch reduzierter tricyklischer Diketone (X), in welchen zwei weitere asymmetrische Zentren als C8 und C14 erscheinen. Die Cyklodehydratisierung dieses Gemisches mit methanolischer Chlorwasserstoffsäure bei Zimmertemperatur folgt, wobei der Ring B gebildet wird und sich das tetracyklische Keton (X1) ergibt. Dieses Keton, das mit 51% Ausbeute aus Diketon (IX) erhalten wird, ist (+)-D-homo-9(1 1)-De- hydro-östron-methyläther, in der richtigen stereochemischen Struktur bei C8 und C14.
Die Reduktion des Keton (XI) mit Kalium und Ammoniumchlorid in flüssigem Ammoniak ergibt (+ )-D-homo-östradiol-methyläther (XII), das ein 4. asymmetrisches Zentrum bei C9, wobei der eingeführte Wasserstoff wieder stereochemisch richtig für die Dhomo-östron-Reihen ist und ein 5. Zentrum als Cl7a enthält. Durch Oxydation mit Bromsäure in Aceton wird der Diol-äther (XII) in (+)-D-homo-östron- methyläther (XII1) umgewandelt. Letztlich, wenn der ketonische Äther (MII) mit Benzaldehyd in Gegenwart einer Base kondensiert wird, wird ein Benziliden Derivat (XIV) erhalten, das identisch ist mit der Substanz, die zu Östron (XV) nach Johnson er al., J.
Amer.Chem.Soc., 1952, 74, 2832 durch Ozonolyse zu Homomarrianol-Säure-methyläther, umgewandelt wurde, durch Pyrolyse des Bleisalzes dieser Säure und Demethylierung. Östron kann leicht zu östradiol (XVI) reduziert werden.
Es wurde ferner gefunden, dass ähnliche Reaktionsfolgen durchgeführt werden können, in welchen statt 2-Methylcyklohexan-l,3-dion (VII) das entsprechende Cyklopentan-Derivat (XXV) verwendet wird, um das dicyklische Triketon Michael-Addukt (XXVI) zu bilden, welches das ganze Kohlenstoffskelett eines 19-nor-Steroids enthält; dieses führt unmittelbar zu (+)-Östradiol-3-methyläther (XXIN und (* )-Östron-methyläther (XXX), um so die Notwendigkeit zu vermeiden den Ring D zu kontrahieren.
(t ) -Östron-methyläther kann durch Demethylierung zu (+- )-Östron (XV) umgewandelt werden.
Eine Variante des neuen Weges welche die Synthese von Östron und Östradiol weiterhin noch verkürzt, wird durch die folgende Synthese dargestellt.
Das 3-Ketoamin (V), sein Ausscheidungsprodukt das Vinylketon (VI) oder ein Gemisch von beiden wird mit 2-Methylcyklopentan-1,3-dion (XXV) unter Rückfluss mit methanolischer Kaliumhydroxidlösung kondensiert. Das erhaltene Michaeladdukt (XXVI) wird unter Rückfluss in trockenem Benzol behandelt, welches Toluol-p-sulfonsäure enthält, unter Entfer- nung von Wasser, so wie es gebildet wird und ergibt das tetracyklische Keton (XXVII), welches in 8,9- und 14, 15-Stellung ungesättigt ist und ein äquimolekulares Gemisch oder ein Racemat der 13 alfa und 13 beta Enantiomere ist. Nur die 13 beta-Verbindungen werden in der Strukturformel gezeigt.
Die Hydrierung des Ketons (XV : (XXVII) unter Ver- wendung eines Raney-Nickel Katalysators von ge mässigter Aktivität geht selektiv mit der Bildung von 8-Dehydro-östron-methyläther (XXVIII) vor sich, wobei das Wasserstoffatom bei der 14-Stellung in trans Stellung zu der 13-Methylgruppe eingeführt wird. Die Reduktion von 8-Dehydroketon (XXVIII) mit Kalium in flüssigem Ammoniak und darauffolgender Zugabe von Ammoniumchlorid ergibt dann (+)-Östradiol- methyläther (XXIX), das anschliessend durch Oxydation mit Chromsäure in (+ ) -Östron-methyläther (XXX) umgewandelt wird.
Die vorliegende Erfindung wird repräsentiert in den obigen Mehrstufensynthesen von Östron bei der Herstellung von dicyklischen Triketonen (VIII) und (XXVI), welche Schlüsselzwischenprodukte in den Synthesen sind. Die Erfindung schafft zum ersten Mal eine dicyklische Verbindung, die die Ringe A und D und das ganze Kohlenstoffskelett eines natürlichen 19-Nor-steroids oder D-homo-Steroids enthält, und welche in eine tetracyklische Verbindung von steroider Struktur und Konfiguration umwandelbar ist.
Die vorliegende Erfindung betrifft nun ein Verfahren zur Herstellung eines Arylalkanons der Formel Ar.CR2.CR2.CH2.CO - CH2.CH2 - CR1-CO.R2, worin
Z Ar eine in der o-Stellung unsubstituierte Arylgruppe, R und R2 Wasserstoff oder einen organischen Rest, R1 einen aliphatischen Rest und Z eine veresterte Carbonsäuregruppe oder eine os-Ketogruppe ist, wobei R2 und Z zusammen mit je ihrem benachbarten Kohlenstoffatom auch einen 6- oder 5-gliedrigen carbocyclischen Ring bilden können.
Das Verfahren ist dadurch gekennzeichnt, dass man eine Verbindung der Formel Ar.CR2.CR2.CH2- CO.Vi, worin Vi eine Vinylgruppe ist, mit einer Verbindung der Formel R2-CO-CHR1-Z kondensiert. Die Vinylgruppe kann in situ aus einer p-substituierten Äthylgruppe gebildet werden.
Der Rest R1, welcher bei einer Steroidsynthese letztlich den eckständigen Substituenten in der 13 Stellung bildet, kann gesättigt oder ungsättigt sein und hat vorzugsweise weniger als 20 und vielfach weniger als 10 Kohlenstoffatome; er ist beispielsweise eine Methyl, - Äthyl-, oder Isopropyl-, Allyl-, But-2-enyloder 2-Methylprop-2-enyl-Gruppe. Eine Alkyl-Gruppe von weniger als 6 Kohlenstoffatomen ist sehr geeignet.
Vorzugsweise ist die Gruppe Z eine alfa-ketonische, das heisst es ist ein organischer Substituent, dessen Kohlenstoffatom beim Befestigungspunkt der einer ketonischen Carbonyl-Gruppe ist. So kann Z eine Acetyl- oder eine substituierte Acetyl-Gruppe sein. Die Gruppe R2 kann eine Alkyl-Gruppe sein, so dass der Rest R2COCR1Z derjenige eines beta-Diketons sein kann. Vorzugsweise ist Z ein alfa-ketonischer Substituent, der mit R2 verbunden ist, so dass die Gruppe R2-C-C-Z sowohl einen 6- oder einen 5gliedrigen carbocyklischen Ring umfasst. Verbindungen, in welchen der Ring 6-gliedrig ist, sind Zwischenprodukte, die zur späteren Umwandlung in Verbindungen der D-homosteroid-Reihen geeignet sind, während jene, in welchen der Ring 5-gliedrig ist Zwischenprodukte für die Steroiden selbst sind.
Vorzugsweise ist die Gruppe R2-C-C-Z eine 1,3-Dioxocyklohexyl- Gruppe, die in der 2-Stellung durch die Gruppe R1 und durch den Rest des Moleküls substituiert ist oder die entsprechende 1,3 -Dioxocyklopentyl-Gruppe ohne zusätzliche Substituenten im Hexan- oder Pentan Ring. Wenn R2 und Z nicht zusammengeschlossen sind, sind die Verbindungen für spätere Stufen der Synthese brauchbar, in welcher der Ring D eines Steroids oder homo-Steroids nachher zugeführt wird oder aus den Substituenten R2 und Z entwickelt wird; in diesem Falle kann R2 beispielsweise eine Alkoxycarbonylmethyl- oder 2-Alkoxycarbonyläthyl-Gruppe sein.
Wenn Z nicht alfa-ketonisch ist, ist es eine Cyanooder eine veresterte Carboxysäure-Gruppe; so kann Z eine Ethoxycarbonyl- oder Methoxycarbonyl Gruppe sein.
Wenn eine oder beide R-Gruppen in einer Gruppe CR2 ein organischer Substituent ist, so wie beispielsweise Methyl-, Äthyl- oder eine andere Alkyl-Gruppe von weniger als 5 Kohlenstoffatomen, können Steroide mit 6- oder 7-Alkyl-Gruppen oder anderen organischen Substituenten unter Verwendung der Verbindungen, hergestellt werden. Vorzugsweise ist jede Gruppe R Wasserstoff.
Die Gruppe Ar soll wenigstens eine ortho-Stellung, frei von Substituenten haben, so dass die Cyklodehydratisierung zur Bildung des Ringes B gegebenenfalls durchgeführt werden kann. Dies ist so zu verstehen, dass bei der Herstellung eines Steroids eine Aryl-Gruppe keine anderen benachbarten Substituenten enthalten wird, die eventuelle Cyklodehydratisierung unmöglich machen würden. Die Aryl-Gruppe ist vorzugsweise eine Phenyl-Gruppe, die frei von Substituenten sein kann; sie kann auch einen oder mehrere Substituenten enthalten. Vorzugsweise enthält sie einen weiteren Substituenten in der 3-Stellung (das ist sozusagen die Stellung 3, beides hinsichtlich zur CR2-Gruppe und zur Aufzählung der Kohlenstoffatome in der End-Steroid-Struktur).
Dieser 3 Substituent ist geeigneterweise eine Hydroxy-, Acyloxy-(beispielsweise Acetoxy), Alkoxy- (beispielsweise Methoxy), Nitro-, Amino-, Monoalkylamino- oder Dialkylamino- (beispielsweise Diäthylamino) Gruppe.
Es wurde gefunden, dass die leichte Durchführung des eventuellen B-Ringschlusses der Produkte des erfindungsgemässen Verfahrens beeinflusst wird durch die Natur des Substituenten, der in der Gruppe Ar vorhanden ist und dass eine folgende Cyklisierung leichter durchzuführen ist, wenn die Gruppe Ar einen Substituenten enthält, entweder in der m-Stellung oder anderswo, welcher die o-Stellung aktiviert, bei welcher die Cyklisierung abläuft.
Wo ein Produkt des erfindungsgemässen Verfahrens für eine Synthese derart wie sie beschrieben ist, zu verwenden ist, in welcher die B-Ring-Cyklisierung bewirkt wird, dann kann es, wenn nicht ein geeigneter Substituent in einer späteren Stufe eingeführt wird, wünschenswert sein, den Gebrauch solcher Verbindungen mit solchen Ar Gruppen zu vermeiden, die keine Substituenten haben, welche die o-Stellung aktivieren und mit welchen die Cyklisierung deshalb schwieriger ist. Jene Substituenten, welche später die Durchführung des B-Ringschlusses leicht bewerkstelligen, sind m-Substituenten (para zur Stellung des Ringschlusses), die Gruppen sind, die in elektrophiler aromatischer Substitution einen aromatischen Ring aktivieren und vorherrschend o- und p-dirigierend sind; beispielsweise eine Alkoxyoder Hydroxy-Gruppe.
Produkte des erfindungsgemässen Verfahrens, in welchen Ar einen aktivierenden Substituenten enthält und Z ein alfa-ketonischer Substituent, verkettet mit R2, ist, so dass die Gruppe R2-C-C-Z entweder einen 6- oder 5-gliedrigen carboxylischen Ring umfasst, können unmittelbar unter geeigneten Säurebedingungen zu tetracyklischen Dienen cyklisiert werden, aus welchen Steroide hergestellt werden können und sind daher von besonderem Wert.
Die Vinyl-ketone und Vinylketonderivate, die verwendet werden können, sind z.B. jene, die in der Britischen Patentschrift Nr. 975.591 beschrieben sind und umfassen Verbindungen, in welchen die Gruppe Vi eine p-Diorganoaminoäthylgruppe (besonders eine p Dialkylaminoäthylgruppe, beispielsweise eine p-Di- äthylaminoäthylgruppe) ist, solch eine Gruppe mit einem Aikylhalogenid (vorzugsweise ein Alkyl-iodid, beispielsweise Methyliodid) quarternisiert ist und eine f3-Chlor- oder p-Bwm-äthylgruppe ist.
Die Reaktion kann unter Bedingungen, die für eine Michaelkondensation geeignet sind, durchgeführt werden. So kann die Kondensation durchgeführt werden durch Zusammenbringen der beiden Reagenten in Lösung in Gegenwart einer Base, beispielsweise Pyridin, Triäthylamin, Diäthylamin, Natriumhydroxid oder Uatriummethoxid und durch Erhitzen, sofern dies erforderlich ist, durchgeführt werden. Die Art und Menge, der bei der Kondensationsreaktion verwendeten Base ist abhängig von den im Einzelfall verwendeten Reagenten. Wo das verwendete Vinyl-ketonderivat ein Ketoamon ist und Dialkylamin bei der Reaktion ausgeschieden wird, kann sich der Zusatz von Base erübrigen. Wo andererseits das verwendete Vinylketonderivat ein Chlor- oder Brom-keton ist, wird es üblicherweise erforderlich sein, vermehrte Mengen Base zu verwenden.
Sofern die Verbindung R2COCHR1Z eine 2-Alkylcyklopentan-1,3-dion ist, ist die Gruppe Vi der Verbindung, welche mit dieser kondensiert ist, vorzugsweise eine Vinyl-Gruppe und das Dion wird im Überschuss der molekularäquivalenten Menge verwendet.
Bei der Infrarot-Spektroskopie wurde beobachtet, dass in einigen Fällen die Triketonprodukte der Kondensationsreaktion das achte Triketon augenscheinlich im Gleichgewicht mit einer oder beiden möglichen adolisierten Formen enthält.
Die Verbindungen der Erfindung sind verwendbar bei der Totalsynthese der Steroide, wie dies oben dargestellt und in den Einzelheiten in der Britischen Patentschrift Nr. 975.591 beschrieben ist.
Die Erfindung wird weiter durch die folgenden Beispiele dargestellt, in welchen alle Temperaturen in Celsius, Drücke in mm Quecksilber bezeichnet sind; Infrarot-Absorptionsdaten beziehen sich auf die Lage der Maxima in cm-l und Ultraviolett-Absorptionsdaten beziehen sich auf die Maxima in mll mit Zahlen in Klammern, welche die molekularen Extinktionskoeffizienten bei diesen Wellenlängen angeben. Die in diesen Beispielen beschriebenen Verbindungen sind ihrer Art nach labil, gewöhnlich nicht kristallisationsfähig und schwierig zu reinigen, weil sie bei der Destillation dazu neigen sich der Umkehrung der Michael-Kondensationsreaktion zu unterwerfen.
Beispiel 1
9 g eines Gemischs von 1-Diäthylamino-6-m-me- thoxyphenylhexan-3-on (V) und 6-m-Methoxyphenylhex-l-en-3-on (VI), das wie in Beispiel 1 der Beschreibung der Britischen Patentschrift Nr. 975.591 beschrieben, erhalten wurde, wurde mit 4 g 2-Methyl cyklohexan-1,3-dion (VII) in 46 ml Benzol, das 3,5 ml Pyridin enthielt, 15 Stunden lang unter Rückfluss gehalten. Die gekühlte Lösung wurde gewaschen und getrocknet. Nach Entfernung des Lösungsmittels ergab sich 8,2 g Triketonaddukt 2-(6'-m-Methoxy phenyl-3'-oxohexyl) -2-methylcyklohexan -1,3-dion (VIII) mit einer Infrarot-Absorption von 1700, 1710, 1715, welches bei 180 - 185a/0,003 mm mm destilliert werden konnte.
Beispiel 2
19 g eines Gemisches von l-Diäthylamino-6-Phenylhexan-3-on und 6-Phenylhex-l-en-3-on, das wie in Beispiel 5 der Beschreibung der Britischen Patentde mit 8,4 g 2-Methylcyklohexan-1, 3-dion in 97 ml schrift Nr. 975.591 beschrieben, erhalten wurde, wur Benzol, das 7,4 ml Pyridin enthielt, 15 Stunden lang unter Rückfluss gehalten. Die gekühlte Lösung wurde gewaschen und getrocknet. Nach Entfernen des Lösungsmittels ergab sich 28 g rohes Triketonaddukt 2-Methyl-2-(6'-phenyl-3'-oxohexyl) cycklohexan-1,3- dion.
Beispiel 3
6 g eines Gemisches von l-Diäthylamino-6-m-methoxy-phenylhexan-3-on (V) und 6-m-Methoxyphenylhex-l-en-3-on (VI), verwendet als Ausgangsmaterial wie in Beispiel 1, wurde mit 2,8 g 2-Methylcyklopen tan-l, 3-dion (XXV) in 20 ml 0, 12einer trockener methanolischer Caliumhydroxydlösung 12 Stunden lang unter Rückfluss gehalten. Das meiste Methanol wurde unter reduziertem Druck entfernt und 50 ml eines Gemisches von gleichen Volumteilen Benzol und Äther wurde zugefügt, das Gemisch mit Wasser, Alkali und Salzsäure im Wechsel gewaschen und getrocknet.
Nach Verdampfung des Lösungsmittels ergaben sich 6, 7 g Triketonaddukt 2-6'-m-Methoxyphenyl-3'- oxohexyl)-2-methylcyklopentan-1, 3-dion (XXVI) als ein viskoses braunes Harz, das nachfolgend zu dem tetracyklischen Dien (+) 8,14-Bisdehydro-östron- methyläther (XXVII) umgewandelt wurde.
Beispiel 4
30 g 2-Äthylcyklopentan-1, 3, 4-trion-hydrat (Schmelzpunkt 65 - 690, Koenigs und Hopmann, Ber., 1921, 54, 1343) wurde in 200 ml Äthanol und 100 ml Wasser gelöst. Zu dieser Lösung wurde tropfenweise im Verlauf einer Stunde eine Lösung von 21 g Semicarbazid-hydrochlorid und 28,2 g Natriumacetat in 200 cc Wasser unter kräftigem Rühren zugefügt. Das ausgeschiedene Semicarbazon wurde dann abgefiltert, mit Methanol gewaschen und durch Rühren in, unter Rüclcfluss gehaltenem Methanol gereinigt. Nach Filtern ergab sich ein blasscremefarbiges Pulver, Schmelzpunkt 179- 1820.
34 g des ausgeschiedenen Semicarbazons wurden in einer Lösung von 34 g Caliumhydroxyd in trockenem Äthylenglykol bei 1300 gelöst, das Gemisch 1 Stunde lang auf 1600 erhitzt und danach 30 Minuten lang auf 1800. Das Glykol wurde durch Destillation bei 0,01 mm entfernt, der zurückbleibende Festkörper in 150 ml Wasser gelöst und die Lösung mit Salzsäure auf Kongo-Rot sauer gemacht, über Nacht auf 0 gekühlt und abgefiltert. Nach Umkristallisieren des Rückstandes aus Wasser ergaben sich 10 g 2-Äthylcyklopentan-1,3-dion, Schmelzpunkt I800 mit Sublimation.
5,25 g eines Gemischs von l-Diäthylamino-6-m- methoxy-phenylhexan-3 -on (V) und 6-m-Methoxyphenylhex-l-en-3-on (VI), wie in Beispiel 1 verwendet, wurde mit 3,3 g 2.Äthylcyklopentan-1,3-dion in einer trockenen 0,1 2%gen methanolischen Caliumhydroxydlösung 18 Stunden lang unter Rückfluss gehalten. Die sich ergebende Lösung wurde abgefiltert, zur Trockne verdampft und der Rückstand in Äther gelöst.
Die Ätherlösung wurde geeignet gewaschen mit Alkali, Salzsäure und Wasser in Wechsel, getrocknet und verdampft u. ergab als Rückstand das Triketonaddukt 2-(6'-m-Methoxyphenyl-3'-oxohexyl)-2-äthyl- cyklopentan-1,3-dion (7,1 g) als Harz das danach zu dem tetracyklischen Dien ( )-8,14-Bisdehydro-18- homo-östron-methyläther umgewandelt wurde.
Beispiel 5
50 g Methylisobutyl-keton und 160 g Diäthyloxalat werden einer eiskalten Lösung von 23 g Natrium in 350 ml trockenem Äthanol unter heftigem Rühren hinzugefügt, das Gemisch dann 30 Minuten unter Rückfluss gehalten, in Eis gekühlt und dann 55 ml wässrige Schwefelsäure (50%) schnell hinzugegeben.
Nach 15 Minuten Stehenlassen wurde Natriumsulfat abgefiltert, mit Äthanol gewaschen und die Waschlaugen dem Filtrat zugefügt, das dann unter reduziertem Druck zur Trockne verdampft Äthyl-4-isopropyl2, 3, 5-trioxocyklopentyl-glyoxylat als ein nicht kristallisierbares Öl zurückliess. Dieses öl wurde mit 1500 ml 2N-Salzsäure 1 Stunde lang gekocht und die heisse Lösung von dem zurückbleibenden teerigen Material dekantiert. Die dekantierte Lösung schied nach Kühlen Kristalle aus, welche abgefiltert und aus wässrigem Äthanol umkristallisiert wurden und ergab 2 Isopropylcyklopentan-1, 3, 5-trion mit einem Schmelzpunkt von 109 - 1120.
25 g des so erhaltenen Trions wurden nach einem Verfahren behandelt, analog dem, welches für die entsprechende 2-Äthyl-Verbindung in Beispiel 4 beschrieben ist und ergab 8,9 g 2-Isopropylcyklopentan 1,3 -dion mit einem Schmelzpunkt von 1460.
6 g eines Gemisches von 1 -Diäthylamino-6-phenyl- hexan-3-on (V) und 6-Phenylhex-l-en-3-on (VI) (siehe Beispiel 2) wurde mit 3 g 2-Isopropylcyklopentan-1, 3dion kondensiert, wobei ein ähnliches Verfahren verwendet wurde, wie dieses für die Michael-Kondensation im Beispiel 4 beschrieben ist. Das entsprechende Triketonaddukt, 7,2 g 2-Isopropyl-2-(6'-m-methoxy phenyl-3 -oxohexyl) -cyklopentan- 1,3-dion wurde als ein nichtkristallisierbares Harz erhalten, das nachfolgend zu dem tetracyklischen Dien (+ )-8,14-Bisde- hydro-18, 18-bishomo-östron-methyläther umgewandelt wurde.
Beispiel 6
6-m-Methoxyphenylhex-l-en-3-on, das eine kleine Menge von 1 .Diäthylamino.6.m.methoxyphenylhexan- 3-on (6 g, das Material hergestellt durch die langsame Destillation der letzteren Substanz) enthielt, wurde mit 3,5 g 2 - Methylcyklopentan - 1,3-dion in 10 cc 0,1 2%igem wasserfreiem methanolischem Caliumhydroxid 10 Stunden lang unter Rüclrfluss gehalten.
Das Reaktionsgemisch wurde, wie in Beispiel 3 beschrieben, bearbeitet und ergab das rohe Triketon addukt, 8 g 2-(6'-m-M : (6' -m-Methoxyphenyl-3 -oxohexyl) -2- methylcyklopentan-1,3-dion. Davon wurde eine kleine Menge bei 0,2 mm Druck zur Analyse destilliert. (Die Analyse ergab: 72,3% C, 7, 45SO H; die Bruttoformel C19H24O4 erfordert 72,1% C und 7,65% H).
Beispiel 7
Ein Gemisch von 3,3 g rohes 6-Phenylhex-l-en-3on (erhalten durch langsame Destillation des Ketoamins l-Diäthylamino-6-phenylhexan-3-on), 1,5 g 2 Methylcyklopentan-1,3-dion und 5,8 ml 0,12%ges wasserfreies methanolisches Kaliumhydroxid wurde 5 Stunden lang unter Rückfluss erhitzt.
Das Lösungmittel wurde dann unter reduziertem Druck entfernt und der Rückstand mit Wasser behandelt und aus Äther extrahiert. Die gewaschenen und getrockneten Ätherextrakte wurden verdampft u. hinterliessen das rohe Triketonaddukt, 6 g 2-Methyl 2-(6'-phenyl-3 '-oxohexyl) -cyclopentan-l, 3-dion als ein rötlich-gelbes Harz; Infrarot-Absorption: 3500, 2940, 1720, 610, damit die Gegenwart von irgendeiner Aldol Form im Gleichgewicht mit Triketon anzeigend.
Beispiel 8 1,0 g 6-m-Nitrophenylhex-l-en-3-on (eine kleine Menge 1 - Diäthylamino-6-m-nitrophenylhexan-3 - on enthaltend und hergestellt, wie in Beispiel 7 beschrieben in der Britischen Patentschrift Nr. 975.591) wurde in 10 ml wasserfreiem 0,1%igem methanolischem Caliumhydroxid mit 1,5 g 2-Methylcyklopentan-1, 3dion 12 Stunden lang unter Rüclcfluss gehalten. Die Lösung wurde gekühlt, in Wasser gegossen und aus Äther extrahiert. Die Extrakte wurden mit Natriumbicarbonatlösung gewaschen, getrocknet und verdampft und es blieben 1,3 g eines Harzes zurück.
Nach Zugabe von Äthanol kristallisierte dieses: Auskristallisiert aus Äthanol ergab das Addukt 2-(6'-m-Nitro phenyl-3 -oxohexy1) - 2 - methyl-cyklopentan -1,3 - dion mit einem Schmelzpunkt von 81 bis 83" und einer Infrarot-Absorption von: 1755, 1725, 1715, 1530, 1365 (die ersten drei Werte stellen Carbonyl-Gruppen, die anderen eine Nitro-Gruppe dar).
Beispiel 9
Zu 2,3 g von rohem Ketoamin l-Diäthylamino-6- (m-methoxyphenyl) -hexan-3 -on (das Material wurde durch Hydratisierung des acetylenischen Amins erhalten, wie dies in Beispiel 1 der Beschreibung der Britischen Patentschrift Nr. 975.591 beschrieben ist) wurde 1 g 2-Methylcyklohexan-1,3-dion, 1 mol Pyridin und 12 ml Benzol zugegeben und das Gemisch 15 Stunden lang unter Rückfluss gehalten. Das Gemisch wurde gekühlt und das nicht in Reaktion gebrachte Dion abgefiltert, dem Filtrat ein wenig Äther zugegeben und die ätherische Lösung mit Säure, dann mit Wasser gewaschen und getrocknet.
Die Verdampfung des Lösungsmittels ergab als Rückstand 1,7 g des Rohaddukts 2- (6'-m-Methoxyphenyl-3'-oxohexyl)-2-me thylcyklohexan-1,3-dion, das die gleiche Infrarot-Absorption hatte, wie das Produkt des Beispiels 1.
Beispiel 10
Zu einem Gemisch von 0,625 g 1-Chlor-6-(m-methoxyphenyl-hexan-3-on (erhalten wie in Beispiel 4 der Beschreibung der Britischen Patentschrift Nr.
975.591), 0,47 g 2-Methylcyklohexan-1, 3-dion und 0,06 mm term Butylalkohol wurde langsam 0,4 ml Triäthylamin zugeführt. Das Gemisch wurde dann 10 Minuten lang auf 600 erhitzt, gekühlt und Äther zugegeben: Die Lösung wurde mit verdünnter Schwefelsäure, danach mit einer Ammoniumsulfatlösung gewaschen und getrocknet. Die Verdampfung ergab das Addukt 2- (6'-m-Methoxyphenyl-3'-oxohexyl-2-me- thylcyklohexan-1,3-dion als ein Harz.
Das harzige Produkt wurde in 15 ml Benzol gelöst, welches 0,4 g Toluol-p-sulfonsäure enthielt, und die Lösung wurde 1 Stunde lang unter Rückfluss gehalten, dann gekühlt und Äther zugegeben. Die gewaschene und getrocknete Lösung wurde zu einem Harz verdampft, welches in 5 ml Benzol aufgenommen wurde und an einer 10 g basischen Tonerdekolonne absorbiert wurde. Das Eluieren mit einem Gemisch von Benzol und Leichtpetroleum ergab eine Fraktion, welche zu einem kristallinen Produkt verdampft wurde. Das Auskristallisieren aus Äthanol ergab (+)-8,14- Bisdehydro-D-homo-Östron-methyläther (0,072 g) mit einem Schmelzpunkt von 135- 1370.
Beispiel 11
0,05 g Natrium wurde 15 ml 0,12%iger methano lischer Caliumhydroxidlösung zugegeben. Zu dieser Lösung wurde 0,9 g 1 -Brom-6-(m-methoxyphenyl) - hexan-3-on (erhalten wie beschrieben in Beispiel 4 der
Beschreibung der Britischen Patentschrift Nr. 975.591) in 5 Methanol und 0,4 g 2-Methylcyklopentan 1,3-dion zugefügt u. das Gemisch 6 Stunden lang un ter Rückfluss gehalten. Nach Behandlung wie in Bei spiel 3 wurde das Rohaddukt 2-(6'-m-Methoxyphenyl 3'-oxohexyl)-2-methylcyklopentan- 1,3-dion als ein gelbes Harz erhalten.
Zum Beweis, dass dieses Produkt das erwartete Addukt war, wurde dieses in 25 ml Benzol mit 0,5 g Toluol-p-sulfonsäure unter Benutzung der Dean und Stark-Apparatur 30 Minuten lang unter Rückfluss gehalten. Das Produkt wurde gekühlt und mittels Äther bearbeitet und ergab ein rotes Harz, welches leicht kristallisierte, wenn es mit authentischem Muster des erwarteten Produktes zusammengebracht wurde. 0,28 g des rohen Festkörpers wurde aus Methanol auskristallisiert und ergab reinen (+)-8, 14-Bisdehydro- östronmethyläther mit einem Schmelzpunkt von 115 1160.
Process for the preparation of an arylalkanone
This invention relates to methods of making chemical compounds containing a carbon skeleton related to that of steroids.
In the past 20 years there has been a great deal of research into the synthesis of steroids, a group of substances that are of great importance for medicine. Most of the industrial research has been directed to their partial synthesis from naturally occurring steroids, which are readily available as starting materials, and the preparation of analogs and derivatives by altering the steroid structures found in nature.
There were some total syntheses of steroids of great acumen and academic merit, but they were relatively few in number and limited in scope. Typical of these syntheses is the large number of individual steps that are necessary, mainly because of the apparent confusion involved in stereochemistry.
Many of the single steps that have been used in such syntheses have used known reactions under standard conditions, and it is primarily in the choice of starting material, followed by the use of a combination of steps in a particular order, that a new concept is found; however, the number of alternative steps and step combinations available to the steroid chemist is so great that, once he sees the overall design of a successful multistep synthesis, he is often able, with the use of further acumen, to modify intermediates in the synthesis in such a way that other steroids or steroid analogs are achieved.
Such a modification, while based on a particular intermediate, thus takes the inspiration from the overall scheme shown by the original inventive sequence, the modification resulting in end products that are the same or different from those of the original syntheses.
It is thus the case that the conception of a successful multi-step steroid synthesis brings a step of inventive merit into each new step in the synthesis, independent of and in support of any inventive concept which this particular step may have, considered in isolation from other steps . The present invention relates to a particular stage in a new total synthesis of steroids.
A new route to the total synthesis of steroid hormones has a number of valuable features.
This approach has the advantage that the carbon atoms essential for the formation of the ring system of a steroid are all present at an early stage in the synthesis, the early skeleton formation helping to carry out a highly stereospecific synthesis. Thus, a terminal methyl group attached at the 13-position in the steroid molecule is created at an early stage, avoiding the creation of an unfavorable ratio of stereoisomers which commonly occurs when attempts are made to introduce this methyl group at a later stage . With this new approach, analog hormones can be achieved more easily, which are not available from natural steroids or at least not without serious difficulties.
Thus, access to steroids of various types has been provided which have an oxygen substituent in the II-position; furthermore, the new route enables the synthesis of steroids which have a variety of substituents in the 3-position. The new way also enables the production of homologous steroids that have corner groups larger than methyl in the 13th position.
Ultimately, the path creates new total syntheses of estrone and estradiol, two valuable therapeutic substances. These can easily be achieved by the new syntheses with good overall yield and by an effectively smaller number of steps than overall syntheses have previously required. The new way is shown below for a total synthesis of oestrone, with the Roman numerals being reference symbols for the structural formulas shown on the attached sheet of drawings.
The known compound 3-m-methoxyphenylpropan-l-ol () is converted to the corresponding bromide (11), which is caused to condense with sodium acetylide in liquid ammonia or other suitable solvents to form acetylene (III). This compound, caused by the appropriate solvents, is subjected to the Mannich reaction using formaldehyde (or a formaldehyde polymer) and diethylamine to give the acetylenic amine (1V).
The next step is hydration of the acetylenic bond under suitable conditions, for example with aqueous mercury II sulfate and sulfuric acid, to give the 3-ketoamine (V).
This 3-ketoamine, which easily separates out diethylamine, even with distillation, its elimination product, vinyl ketone (Vp or a mixture of both, is refluxed with 2-methylcyclohexane-1,3-dione (VII) in benzene, which contains pyridine The product of this Michael condensation is the dicyclic triketone (VIII), which contains the entire carbon chain of a 19-nor-D-homo-steroid.
Refluxing this triketone in xylene, in the presence of triethylammonium benzoate as a catalyst, brings about an internal aldol condensation and dehydration, which forms the tricyclic diketone (IX), which has unsaturation in the newly formed ring, which is ring C at the 8 (14) position of the terminal steroid corresponds. This tricyclic diketone is a mixture of enantiomers which currently has an asymmetric center at Cig.
Only the 13-beta compound is shown, although it should be understood that the mixture contains an equal amount of the 13-alfa compound as the following compounds; Resolution to form a 13-beta enantiomer can be achieved at any suitable stage.
The catalytic hydrogenation of the tricyclic diketone mixture saturates the ring C and gives an oily mixture of reduced tricyclic diketones (X) in which two further asymmetric centers appear as C8 and C14. Cyclodehydration of this mixture with methanolic hydrochloric acid follows at room temperature to form ring B and yield the tetracyclic ketone (X1). This ketone, which is obtained with 51% yield from diketone (IX), is (+) - D-homo-9 (11) -dehydro-estrone methyl ether, in the correct stereochemical structure at C8 and C14.
The reduction of the ketone (XI) with potassium and ammonium chloride in liquid ammonia gives (+) -D-homo-estradiol-methyl ether (XII), which has a 4th asymmetric center at C9, whereby the introduced hydrogen is again stereochemically correct for the Dhomo oestrone series and contains a 5th center as Cl7a. Oxidation with bromic acid in acetone converts the diol ether (XII) into (+) - D-homo-estrone methyl ether (XII1). Ultimately, if the ketonic ether (MII) is condensed with benzaldehyde in the presence of a base, a benzilidene derivative (XIV) is obtained which is identical to the substance which is converted to oestrone (XV) according to Johnson er al., J.
Amer.Chem.Soc., 1952, 74, 2832 by ozonolysis to homomarrianol acid methyl ether, by pyrolysis of the lead salt of this acid and demethylation. Eestrone can easily be reduced to estradiol (XVI).
It has also been found that similar reaction sequences can be carried out in which instead of 2-methylcyclohexane-1,3-dione (VII) the corresponding cyclopentane derivative (XXV) is used to form the dicyclic triketone Michael adduct (XXVI) which contains the entire carbon skeleton of a 19-nor steroid; this leads directly to (+) - estradiol-3-methyl ether (XXIN and (*) -estrone methyl ether (XXX), in order to avoid the need to contract ring D.
(t) -Oestrone methyl ether can be converted to (+ -) -estrone (XV) by demethylation.
A variant of the new approach, which further shortens the synthesis of estrone and estradiol, is represented by the following synthesis.
The 3-ketoamine (V), its elimination product the vinyl ketone (VI) or a mixture of both is condensed with 2-methylcyclopentane-1,3-dione (XXV) under reflux with methanolic potassium hydroxide solution. The Michael adduct obtained (XXVI) is refluxed in dry benzene containing toluene-p-sulfonic acid, removing water as it is formed, and gives the tetracyclic ketone (XXVII), which is in 8,9- and 14, 15-position is unsaturated and is an equimolecular mixture or a racemate of the 13 alpha and 13 beta enantiomers. Only the 13 beta compounds are shown in the structural formula.
The hydrogenation of the ketone (XV: (XXVII) using a Raney nickel catalyst of moderate activity proceeds selectively with the formation of 8-dehydro-oestrone methyl ether (XXVIII), the hydrogen atom in the 14-position The reduction of 8-dehydroketone (XXVIII) with potassium in liquid ammonia and the subsequent addition of ammonium chloride then gives (+) - estradiol methyl ether (XXIX), which is then converted into by oxidation with chromic acid in the trans position to the 13-methyl group (+) Oestrone methyl ether (XXX) is converted.
The present invention is represented in the above multistep syntheses of estrone in the preparation of dicyclic triketones (VIII) and (XXVI) which are key intermediates in the syntheses. The invention provides, for the first time, a dicyclic compound which contains rings A and D and the entire carbon skeleton of a natural 19-norsteroid or D-homo-steroid, and which is convertible into a tetracyclic compound of steroid structure and configuration.
The present invention now relates to a process for the preparation of an arylalkanone of the formula Ar.CR2.CR2.CH2.CO - CH2.CH2 - CR1-CO.R2, in which
Z Ar is an aryl group unsubstituted in the o-position, R and R2 are hydrogen or an organic radical, R1 is an aliphatic radical and Z is an esterified carboxylic acid group or an os-keto group, where R2 and Z together with their adjacent carbon atoms also represent a 6- or 5-membered carbocyclic ring.
The process is characterized in that a compound of the formula Ar.CR2.CR2.CH2-CO.Vi, in which Vi is a vinyl group, is condensed with a compound of the formula R2-CO-CHR1-Z. The vinyl group can be formed in situ from a p-substituted ethyl group.
The radical R1, which ultimately forms the corner substituent in the 13-position in a steroid synthesis, can be saturated or unsaturated and preferably has fewer than 20 and in many cases fewer than 10 carbon atoms; it is, for example, a methyl, ethyl, or isopropyl, allyl, but-2-enyl or 2-methylprop-2-enyl group. An alkyl group of less than 6 carbon atoms is very suitable.
The group Z is preferably an alpha-ketonic, that is to say it is an organic substituent whose carbon atom at the point of attachment is that of a ketonic carbonyl group. Thus, Z can be an acetyl or a substituted acetyl group. The group R2 can be an alkyl group, so that the radical R2COCR1Z can be that of a beta-diketone. Preferably Z is an alpha-ketonic substituent attached to R2 such that the group R2-C-C-Z comprises both a 6- or a 5-membered carbocyclic ring. Compounds in which the ring is 6-membered are intermediates suitable for later conversion to compounds of the D-homosteroid series, while those in which the ring is 5-membered are intermediates for the steroids themselves.
The group R2-CCZ is preferably a 1,3-dioxocyclohexyl group which is substituted in the 2-position by the group R1 and by the rest of the molecule or the corresponding 1,3-dioxocyclopentyl group without additional substituents in the hexane or pentane ring. If R2 and Z are not joined together, the compounds are useful for later stages of the synthesis in which the ring D of a steroid or homo-steroid is subsequently added or is developed from the substituents R2 and Z; in this case, R2 can be, for example, an alkoxycarbonylmethyl or 2-alkoxycarbonylethyl group.
If Z is not alfa-ketonic it is a cyano or an esterified carboxy acid group; thus Z can be an ethoxycarbonyl or methoxycarbonyl group.
If one or both of the R groups in a group CR2 is an organic substituent, such as methyl, ethyl or another alkyl group of less than 5 carbon atoms, steroids with 6- or 7-alkyl groups or other organic Substituents using the compounds. Preferably each R group is hydrogen.
The group Ar should have at least one ortho position, free of substituents, so that the cyclodehydration to form the ring B can optionally be carried out. It is to be understood that in the manufacture of a steroid an aryl group will not contain any other adjacent substituents that would render possible cyclodehydration impossible. The aryl group is preferably a phenyl group, which may be free of substituents; it can also contain one or more substituents. It preferably contains a further substituent in the 3-position (that is, so to speak, the 3-position, both with regard to the CR2 group and to enumerate the carbon atoms in the final steroid structure).
This 3 substituent is suitably a hydroxy, acyloxy (e.g. acetoxy), alkoxy (e.g. methoxy), nitro, amino, monoalkylamino or dialkylamino (e.g. diethylamino) group.
It has been found that the ease of carrying out the eventual B-ring closure of the products of the process according to the invention is influenced by the nature of the substituent which is present in the group Ar and that a subsequent cyclization is easier to carry out if the group Ar contains a substituent, either in the m-position or elsewhere which activates the o-position at which the cyclization takes place.
Where a product of the process according to the invention is to be used for a synthesis as described in which the B-ring cyclization is effected, then, if a suitable substituent is not introduced at a later stage, it may be desirable to use the To avoid the use of such compounds with such Ar groups that have no substituents which activate the o-position and with which the cyclization is therefore more difficult. Those substituents which later easily accomplish the B-ring closure are m-substituents (para to the position of the ring closure), which are groups which activate an aromatic ring in electrophilic aromatic substitution and are predominantly o- and p-directing; for example an alkoxy or hydroxy group.
Products of the process according to the invention, in which Ar contains an activating substituent and Z is an alpha-ketonic substituent, linked to R2, so that the group R2-CCZ comprises either a 6- or 5-membered carboxylic ring, can be used directly under suitable acid conditions are cyclized to tetracyclic dienes from which steroids can be made and are therefore of particular value.
The vinyl ketones and vinyl ketone derivatives that can be used are e.g. those described in British Patent Specification No. 975,591 and include compounds in which the group Vi is a p -diorganoaminoethyl group (especially a p-dialkylaminoethyl group, e.g. a p-diethylaminoethyl group), such a group with an alkyl halide (preferably an Alkyl iodide, for example methyl iodide) is quaternized and is an f3-chlorine or p-Bwm-ethyl group.
The reaction can be carried out under conditions suitable for Michael condensation. For example, the condensation can be carried out by bringing the two reagents together in solution in the presence of a base, for example pyridine, triethylamine, diethylamine, sodium hydroxide or sodium methoxide, and by heating, if necessary. The type and amount of the base used in the condensation reaction depends on the reagents used in the individual case. Where the vinyl ketone derivative used is a ketoamone and dialkylamine is excreted in the reaction, the addition of a base may be unnecessary. On the other hand, where the vinyl ketone derivative used is a chloro- or bromo-ketone, it will usually be necessary to use increased amounts of base.
If the compound R2COCHR1Z is a 2-alkylcyclopentane-1,3-dione, the group Vi of the compound which is fused with this is preferably a vinyl group and the dione is used in excess of the molecular equivalent amount.
In infrared spectroscopy, it has been observed that in some cases the triketone products of the condensation reaction contain the eighth triketone apparently in equilibrium with one or both of the possible adolized forms.
The compounds of the invention are useful in the total synthesis of the steroids as set forth above and described in detail in British Patent No. 975,591.
The invention is further illustrated by the following examples, in which all temperatures are in Celsius, pressures in mm of mercury; Infrared absorption data relate to the location of the maxima in cm-l and ultraviolet absorption data relate to the maxima in mll with numbers in brackets indicating the molecular extinction coefficients at these wavelengths. The compounds described in these examples are inherently labile, usually incapable of crystallization and difficult to purify because they tend to undergo the reverse of the Michael condensation reaction during distillation.
example 1
9 g of a mixture of 1-diethylamino-6-m-methoxyphenylhexan-3-one (V) and 6-m-methoxyphenylhex-l-en-3-one (VI), which as in Example 1 of the description of the British Patent Specification No. 975,591, was refluxed with 4 g of 2-methyl cyclohexane-1,3-dione (VII) in 46 ml of benzene containing 3.5 ml of pyridine for 15 hours. The cooled solution was washed and dried. After removal of the solvent, 8.2 g of triketone adduct 2- (6'-m-methoxy phenyl-3'-oxohexyl) -2-methylcyclohexane -1,3-dione (VIII) with an infrared absorption of 1700, 1710, 1715, which could be distilled at 180-185a / 0.003 mm mm.
Example 2
19 g of a mixture of l-diethylamino-6-phenylhexan-3-one and 6-phenylhex-l-en-3-one, which as in Example 5 of the description of the British Patent with 8.4 g of 2-methylcyclohexan-1, 3-dione described in 97 ml of publication No. 975.591, benzene containing 7.4 ml of pyridine was refluxed for 15 hours. The cooled solution was washed and dried. Removal of the solvent gave 28 g of crude triketone adduct 2-methyl-2- (6'-phenyl-3'-oxohexyl) cyclohexane-1,3-dione.
Example 3
6 g of a mixture of 1-diethylamino-6-m-methoxyphenylhexan-3-one (V) and 6-m-methoxyphenylhex-1-en-3-one (VI), used as starting material as in Example 1, was used refluxed with 2.8 g of 2-methylcyclopene tan-1,3-dione (XXV) in 20 ml of 0.12 of a dry methanolic potassium hydroxide solution for 12 hours. Most of the methanol was removed under reduced pressure and 50 ml of a mixture of equal parts by volume of benzene and ether was added, the mixture washed with water, alkali and hydrochloric acid alternately and dried.
After evaporation of the solvent, 6.7 g of triketone adduct 2-6'-m-methoxyphenyl-3'-oxohexyl) -2-methylcyclopentane-1,3-dione (XXVI) resulted as a viscous brown resin, which is subsequently to the tetracyclic diene (+) 8,14-bisdehydro-estrone methyl ether (XXVII) was converted.
Example 4
30 g of 2-ethylcyclopentane-1, 3, 4-trione hydrate (melting point 65-690, Koenigs and Hopmann, Ber., 1921, 54, 1343) was dissolved in 200 ml of ethanol and 100 ml of water. To this solution, a solution of 21 g of semicarbazide hydrochloride and 28.2 g of sodium acetate in 200 cc of water was added dropwise over the course of one hour with vigorous stirring. The semicarbazone which separated out was then filtered off, washed with methanol and purified by stirring in refluxing methanol. Filtering gave a pale cream colored powder, melting point 179-1820.
34 g of the precipitated semicarbazone were dissolved in a solution of 34 g of potassium hydroxide in dry ethylene glycol at 1300, the mixture heated to 1600 for 1 hour and then to 1800 for 30 minutes. The glycol was removed by distillation at 0.01 mm, the remaining one Solids dissolved in 150 ml of water and the solution made acidic to Congo red with hydrochloric acid, cooled to 0 overnight and filtered off. After recrystallization of the residue from water, 10 g of 2-ethylcyclopentane-1,3-dione, melting point 1800 with sublimation were obtained.
5.25 g of a mixture of 1-diethylamino-6-m-methoxy-phenylhexan-3-one (V) and 6-m-methoxyphenylhex-1-en-3-one (VI) as used in Example 1 was used kept under reflux for 18 hours with 3.3 g of 2nd ethylcyclopentane-1,3-dione in a dry 0.1 2% methanolic potassium hydroxide solution. The resulting solution was filtered off, evaporated to dryness and the residue dissolved in ether.
The ether solution was suitably washed with alkali, hydrochloric acid and water alternately, dried and evaporated u. gave the triketone adduct 2- (6'-m-methoxyphenyl-3'-oxohexyl) -2-ethyl-cyclopentane-1,3-dione (7.1 g) as a resin which then gave the tetracyclic diene () -8 , 14-bisdehydro-18-homo-estrone methyl ether was converted.
Example 5
50 g of methyl isobutyl ketone and 160 g of diethyloxalate are added to an ice-cold solution of 23 g of sodium in 350 ml of dry ethanol with vigorous stirring, the mixture is then refluxed for 30 minutes, cooled in ice and then 55 ml of aqueous sulfuric acid (50%) quickly added.
After standing for 15 minutes, sodium sulfate was filtered off, washed with ethanol and the washing liquors were added to the filtrate, which then evaporated to dryness under reduced pressure, leaving ethyl 4-isopropyl2, 3, 5-trioxocyclopentyl glyoxylate as a non-crystallizable oil. This oil was boiled with 1500 ml of 2N hydrochloric acid for 1 hour and the hot solution was decanted from the tarry material that remained. The decanted solution separated after cooling, crystals, which were filtered off and recrystallized from aqueous ethanol and yielded 2 isopropylcyclopentane-1,3,5-trione with a melting point of 109-1120.
25 g of the trione thus obtained were treated by a process analogous to that described for the corresponding 2-ethyl compound in Example 4 and gave 8.9 g of 2-isopropylcyclopentane 1,3-dione with a melting point of 1460.
6 g of a mixture of 1-diethylamino-6-phenyl-hexan-3-one (V) and 6-phenylhex-l-en-3-one (VI) (see Example 2) was mixed with 3 g of 2-isopropylcyclopentane-1 , 3dione condensed using a method similar to that described for the Michael condensation in Example 4. The corresponding triketone adduct, 7.2 g of 2-isopropyl-2- (6'-m-methoxy phenyl-3-oxohexyl) -cyclopentane-1,3-dione was obtained as a non-crystallizable resin which was subsequently converted to the tetracyclic diene (+ ) -8,14-bisde- hydro-18, 18-bishomo-oestrone methyl ether was converted.
Example 6
6-m-methoxyphenylhex-l-en-3-one, which contained a small amount of 1 .diethylamino.6.m.methoxyphenylhexan-3-one (6 g, the material produced by the slow distillation of the latter substance) kept under reflux for 10 hours with 3.5 g of 2-methylcyclopentane-1,3-dione in 10 cc of 0.1 2% strength anhydrous methanolic potassium hydroxide.
The reaction mixture was processed as described in Example 3 and gave the crude triketone adduct, 8 g of 2- (6'-mM: (6'-m-methoxyphenyl-3-oxohexyl) -2-methylcyclopentane-1,3-dione A small amount of this was distilled at 0.2 mm pressure for analysis. (The analysis showed: 72.3% C, 7.45% H; the gross formula C19H24O4 requires 72.1% C and 7.65% H).
Example 7
A mixture of 3.3 g of crude 6-phenylhex-l-en-3-one (obtained by slow distillation of the ketoamine l-diethylamino-6-phenylhexan-3-one), 1.5 g of 2-methylcyclopentane-1,3-dione and 5.8 ml of 0.12% sat. Anhydrous methanolic potassium hydroxide was refluxed for 5 hours.
The solvent was then removed under reduced pressure and the residue treated with water and extracted from ether. The washed and dried ether extracts were evaporated u. The crude triketone adduct, 6 g of 2-methyl 2- (6'-phenyl-3'-oxohexyl) -cyclopentane-1,3-dione left behind as a reddish-yellow resin; Infrared absorption: 3500, 2940, 1720, 610, thus indicating the presence of any form of aldol in equilibrium with triketone.
Example 8 1.0 g of 6-m-nitrophenylhex-l-en-3-one (containing a small amount of 1-diethylamino-6-m-nitrophenylhexan-3-one and prepared as described in Example 7 in British Patent No. 975.591) was kept under reflux for 12 hours in 10 ml of anhydrous 0.1% strength methanolic potassium hydroxide with 1.5 g of 2-methylcyclopentane-1, 3dione. The solution was cooled, poured into water and extracted from ether. The extracts were washed with sodium bicarbonate solution, dried and evaporated, leaving 1.3 g of a resin.
After the addition of ethanol this crystallized: Crystallized from ethanol gave the adduct 2- (6'-m-nitrophenyl-3-oxohexy1) -2-methyl-cyclopentane -1,3-dione with a melting point of 81 to 83 "and a Infrared absorption of: 1755, 1725, 1715, 1530, 1365 (the first three values represent carbonyl groups, the others a nitro group).
Example 9
To 2.3 g of crude ketoamine 1-diethylamino-6- (m-methoxyphenyl) -hexan-3-one (the material was obtained by hydration of the acetylenic amine as described in Example 1 of British Patent Specification No. 975,591 1 g of 2-methylcyclohexane-1,3-dione, 1 mol of pyridine and 12 ml of benzene were added and the mixture was refluxed for 15 hours. The mixture was cooled and the unreacted dione filtered off, a little ether added to the filtrate and the ethereal solution washed with acid, then with water and dried.
Evaporation of the solvent gave 1.7 g of the crude adduct 2- (6'-m-methoxyphenyl-3'-oxohexyl) -2-methylcyklohexane-1,3-dione, which had the same infrared absorption as that, as a residue Product of example 1.
Example 10
To a mixture of 0.625 g of 1-chloro-6- (m-methoxyphenyl-hexan-3-one (obtained as in Example 4 of the description of British Patent No.
975.591), 0.47 g of 2-methylcyclohexane-1, 3-dione and 0.06 mm term butyl alcohol were slowly added to 0.4 ml of triethylamine. The mixture was then heated to 600 for 10 minutes, cooled and ether added: the solution was washed with dilute sulfuric acid, then with an ammonium sulfate solution and dried. Evaporation gave the adduct 2- (6'-m-methoxyphenyl-3'-oxohexyl-2-methylcyclohexane-1,3-dione as a resin.
The resinous product was dissolved in 15 ml of benzene containing 0.4 g of toluene-p-sulfonic acid and the solution was refluxed for 1 hour, then cooled and ether added. The washed and dried solution was evaporated to a resin which was taken up in 5 ml of benzene and absorbed on a 10 g basic alumina column. Eluting with a mixture of benzene and light petroleum gave a fraction which was evaporated to a crystalline product. Crystallization from ethanol gave (+) - 8,14-bisdehydro-D-homo-oestrone methyl ether (0.072 g) with a melting point of 135-1370.
Example 11
0.05 g of sodium was added to 15 ml of 0.12% strength methanolic potassium hydroxide solution. 0.9 g of 1-bromo-6- (m-methoxyphenyl) -hexan-3-one (obtained as described in Example 4 of
Description of British patent specification No. 975,591) in 5 methanol and 0.4 g of 2-methylcyclopentane 1,3-dione added u. the mixture was refluxed for 6 hours. After treatment as in Example 3, the crude adduct 2- (6'-m-methoxyphenyl 3'-oxohexyl) -2-methylcyclopentane 1,3-dione was obtained as a yellow resin.
To demonstrate that this product was the expected adduct, it was refluxed in 25 ml of benzene with 0.5 g of toluene-p-sulfonic acid using the Dean and Stark apparatus for 30 minutes. The product was cooled and ether processed to give a red resin which readily crystallized when placed with authentic specimens of the expected product. 0.28 g of the crude solid was crystallized from methanol and gave pure (+) - 8,14-bisdehydro-estronmethylether with a melting point of 115,160.
