Die vorliegenX Erfindung betrifft ein Verfahren zur HersteRung neuer Sulfonamilde der Formel I

worin RI und R2 niedere Alkylreste bedeuten, ins- besondere solche mit 1-6 C-Atomen, wie Athyl, n- Propyl, Isopropyl, Butyl oder Hexyl, vorzugsweise aber je eine Methylgruppe, sowie von Salzen solcher Sulfonamide.
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man den Hal-Substituenten einer Verbindung der Formel II

wo, rin Hal ein Halogenatom, insbesondere. ein Chloratom, und X eine freie oder geschützte Aminogruppe, z. B. eine Acylamino-oder Carbalkoxyaminogruppe, di'e Acetylamino oder Carbäthoxyaminogruppe, bedeutet, mittels Alkoholyse durch eine R20-Gruppe ersetzt und eine im Umsetzungsprodukt gegebenen- falls vorhandene Schutzgruppe eliminiert.
Um den Hal-Substituenten eines Pyrimidijns der Formel II durch einen R20-Rest zu ersetzen, geht man zweckmässig so vor, dass man das Pyrimidin der Formel II mit einem Alkalialkoholat des entsprechen- d'en Alkohols, z. B. mit einer methanolischen Lö sung von Natnummethylat, zum Sieden unter Rückfluss erhvtzt. Die Alkoholyseoperation kann auch unter Druck bei hoheren Temperaturen, z. B. bei etwa 120-130 C, vorgenommen werden. Eine gegebenenfalls vorliegende geschützte Aminogruppe kann vor oder nach der Alkoholyseoperation in die freie Aminogruppe übergeführt werden. So kann z.
B. eine N4-Acylgruppe leicht durch Erhitzen der N4- Acylverbindung mit verdünnten Säuren oder Alkalien, z. B. mit verdünnter Natronlauge, abgespalten werden. Führt man die Alkoholyse unter Druck bei höheren Temperaturen durch, so wird in der Regel eine gegebenenfalls vorhandene N4-Acylgruppe gleichzeitig abgespalten.
Die nach dem erfindungsgemässen Verfahren erhältlichen Sulfonanude der Formel I lassen sich sowohl mit stärkeren Säuren, wie Salzsäure oder Schwefelsäure, als auch mit Basen, wie Alkalihy- droxyden,-carbonaten oder-bicarbonaten, in Salze überführen.
Die Verfahrensprodukte, unter ihnen besonders das 4-Sulfanilamido -5,6-dimethoxy-pyrimidin, erweisen sich gegen Infektionen der verschiedensten Erreger, wie Staphylokokken, Pneumokokken. E. coli und Salmonella, als hoch aktiv. Sie zeichnen sich überdies durch langanhaltende Wirkung aus. Sie können als Heilmittel in Form pharmazeutischer Präparate Verwendung finden, welche sie oder ihre Salze in Mischung mit einem für die enterale oder parenteral!eApplikationgeeignetenpharmazeutischen, organischen oder anorganischen inerten Träger- material enthalten. Die pharmazeutischen Präparate können in fester Form oder in flüssiger Form vorliegen.
Gegeblenenfalls sind sie sterilisiert und bzw. oder enthalten Hilfsstoffe, wie Konservierungs-, Sta bilisierungs-, Netz-oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer.
Sie können auch noch andere therapeutilsch wertvolle Stoffe enthalten.
Die Ausgangsstoffe der Formel II können z. B. dadurch erhalten werden, dass man ein 4, 6-Dihalogen- 5-alkoxy-pyrimidin, insbesondere d'as 4, 6-DichWior-5- methoxy-pyrimidin, mit ei'nem Alkalisalz eines Ben- zolsulfonsäureamids kondensiert, das in p-Stellung eine freie oder geschützte Aminogruppa. aufweist, also beispielsweise mit Sulfanilamidnatrium, N4-Ace- tyl-sulfanilamidnatrium oder N4-Carbäthoxy-sulf- anilamidhatrium. Diese Kondensation wird zweck- mässig in Gegenwart eines Lösungs-bzw. Verd'ün nungsmittels,
wie Dimethylformamid, bei Temperaturen zwischen 80 und 120 C, insbesondere bei etwa 90-100 C, ausgeiihrt.
Die genannten 4, 6-Dihalogen-5-alkoxy-pyrimidine können ihrerseits z. B. wie folgt erhalten werden :
Ein Alkoxyessigsäuremethylester wird mittels Dimethylcarbonat in den entsprechenden Alkoxymalon säuredimethylester übergeführt und diser mit Ammoniak in das entsprechende Alkoxymalonsäurediamid umgewandelt. Dieses Diamid wird mittels Formamid in Gegenwant von Natriumäthylat zum 4, 6-Di hydroxy-5-alkoxy-pyrimidin cyclisiert, aus welchem durch Halogenierung mit einem Phosphoroxyhalo- genid, z. B. mit PhosphoroxycMomd, das 4, 6-Di halogen-5-alkoxy-pyrimidi'n erhalten werden kann.
Beispiel 1
15, 75 g reines 4-Sulfanilamido-5-methoxy-6- chlor-pyrimidin (0, 05 Mol) werden in eine e Lösung von 5, 75 g Natrium (0, 25 Grammatome) in 100m! absolutem Methanol eingetragen und im Autoklaven 4 Stunden auf 125 Cerhitzit. Nach Abkühlen wird das Lösungsmittel im Vakuum abgezogen und der Eindampfrückstand in 50 ml Wasser aufgenommen.
Nach Behandlung mit Tierkohle wird ein klares gerbas Filtrat erhalten, das beim Ansäuern mit Eis essig amorphes 4-Sulfanilamido-5, 6-dimethoxy-pyri- midin abscheidet. Ietzteres wird durch Anreiben rasch kristallin. Nach Filtration und Trocknen im Vakuum wird ein nahezu farbloses Produkt erhalten. Schmelzpunkt : 182-190 C.
Zur Reinigung wilrd dieses in etwa 150 ml Eisessig/Wasser (1 : 1) in der Hitze in Lösung gebracht, mit Tierkohle behandelt, rasch filtriert und die Lösung der Kristallisation überitassen. Man erhält 10, 6 g rein weisses 4-Sulfanilamido-5,6-dimethoxy-pyrimidin, Schmelzpunkt 190-194 C (Ausbeute 78,2% der Theorie).
Die rein weisse Verbindung ist in kaltem Wasser und Ather wenig löslich. Löslich ist sie etwas in Alkohol, besser in Aceton. In Alkalien sowie in Sodaund Bicarbonatlösung löst sie sich unter Salzbildung, desgleichen in stärkeren Mineralsäuren.
Das als Ausgangsstoff verwendete 4-SuEanil- amido-5-methoxy-6-chlor-pyrimidin kann wie folgt erhalten werden :
42, 7 g Sulfanilamidnatrium (0, 22 Mol) werden in kleinen Anteiiten in 140 ml absolutes Dimethylformamid eingetragen.DerresultierendeBrei wird unter Rühren auf 95 C erwärmt. Dann werden 19, 7 g (0, 11 Mol) 4, 6-Dichlor-5-methoxy-pyrimidin portio nenweise eingerührt. Die rasch eintretende Reaktion ist exotherm.
Es wird dafür Sorge getragen, dass die Temperatur im Reaktionsgemisch 90-100 C beträgb. Nach beendeter Zugabe der Pyrimiidinver- bindung wird weitere 5 Minuten unter Rühren bei 90 C gehalten. Dreistündiges Erhitzen auf dem siedenden Wasserbad unter Feuchtigkeitsausschluss vervollständigt den Umsatz. Das nach dibser Zeib im Vak*uum möglichst voHsständig abdestillierte Lo Lösungsmittel hinterlässt einen bräunlichen, kristalZisier- ten Rückstand, der, mit 60 ml Wasser verrieben, zum grossen Teil in Lösung geht.
Die Hauptmenge des im Überschuss eingesetzten Sulfanilamids scheidet sich nach kurzer Zeit ab und wird abgenutscht, mit wenig Wasser gewaschen und getrocknet.
Das. bräunlich gefärbte Filtrat wird mit Eisessig auf pH 6 angesäuert, wobei das 4-Sulfanilamido 5-methoxy-6-chlor-pyrimidin als zäher Honig aus- fällt. Dekantileren und Anreiben der Faltung mit wenig wässrigem Alkohol führt rasch zur Kristalli- sation. Ausb, eute : 29, 1 g (84% der Theorie) ; Schmelz- punkt : 189-198 C. Zur Reinigung werden 15 g dieses Produktes in 250 ml Eisessig/Wasser (1 : 1) in der Hitze geläst, 5 g Tierkohle zugesetzt und 10 Minuten im heissen Wasserbad belassen. Nach rascher Filtration kristallisiert 4-Sulfanilamido 5-methoxy- 6-chlor-pyrimidin in völlig farblosen Blättchen aus.
Beispiel 2
0, 35 g Natrium (etwa 0, 015 Grammatome) wer- den in 8 ml absolutem Methanol gelöst und 1, 07 g rohes 4- (N4-Acetylsulfanilamido)-5-methoxy-6-chlorpyrimidin (0, 003 Mol) eingetragen. Der Ansatz wird im Glasautoklaven 3 Stunden bei 115-125 C gehalten und der nach dieser Zeit durch Abdestillieren des Lösungsmittels gewonnene Eindampfrückstiand in 10 mu'tasser aufgenommen. Nach Behandeln mit Tierkohle im siedenden Wasserbad wird filtriert und mit 50% iger Essigsäure angesäuert ; dabei schei- deit sich 4-Su!]fanÜ!amido-5-6-dimethoxy-pyrimidin in amorpher Form ab.
Abdekantieren und Digerieren mit wenig wässrigem Alkohol führt rasch zur Kri stallisation. Das nach einer Stunde Stehen im Kühlschrank gesammelte Rohmaterial wiegt vakuumgetrocknet 760 mg (82, 5 % der Theorie). Schmelzpunkt 172-187 C.
Die in Mineralsäuren lbsliche Verbindung schmilzt nach zweimaligem Umkristallisieren aus Methanol/Wasser (4 : 1) unter Zusatz von Tierkohle bei 192-194' (korr.).
Das als Ausgangsstoff verwendete 4- (N4-Aoetyl- sulfanilamido)-5-methoxy-6-chlor-pyrimidin kann wie folgt erhalten werden :
2, 36 g N4-Acetylsulfanilamidnatrium (0,01 Mol) werden mit 7 ml absolutem Dimethylformamid zu einem Brei verrührt. Nach Erwärmen auf 95 C werden unter Rühren 0, 895 g 4, 6-Dichlor-5-meth oxy-pyrimidin (0, 005 Mol) portionsweise eingetragen und die Temperatur im Reaktionsgemisch zwischen 90-100 C gehalten. 5 Minuten nach beendeter Zugabe wird das dünnflüssig gewordene Gemisch unter Feuchtigkeitsausschluss 1 Stunde auf dem siedenden Wasserbad erhitzt, wobei der Grossteil des festen Materials in Lösung geht.
Das nach dieser Zeit i'm Vakuum vollständig abdestillierte Lösungsmittel hinterlässt einen sirupösen braunen Rückstand ; mit we, nig Wasser angerieben, scheidet dieser den Grossteil des im Überschuss eingesetzten N4-Acetylsulfanil- amids als farbloses Kristallisat ab. Dieses wird durch Filtration entfernt.
Das Filtrat wird mit 50 % iger Essigsäure auf pH 5 gebracht, wobei sich 4-(N4-Acetylsulfanilamido) 5-methoxy-6-chlor-pyrimidin als zähe, durch Anreiben rasch kristallisierende gummiartige Substanz abscheidet. Das abgenutschte, schwach gelbliche Produkt wiegt nach Trocknen im Vakuum 1, 65 g (92, 5 % der Theorie). Umkristallisation aus Methanol unter Kohlenzusatz führt zu einem rein weissen Produkt, das bei 210-212 C (198 C Umwandlung) schmilzt.
Beispiel 3
40 g 4-Sulfanilamido-5-äthoxy-6-chlor-pyrimidin werden in eine Lösung von 6, 9 g Natrium in 300 ml Methanol eingetragen. Das Reaktionsgemisch wird 3 Stunden unter Riickfluss gekochn. Dann werden 250 ml Methanol abdestilliert. Die verbleibende Lösung wird weitere 18 Stunden unter Rückfluss gekocht. Nach Eindampfen wird der Rückstand in 200 ml Wasser und 130 ml konzentrierter Salzsäure gelöst, miit Tierkohle filtriert und mit Ammoniak auf pH 5-6 gestellt. Es werden so 26 g 4-Sulfanilamido 5-äthoxy-6-methoxy-pyrimidin vom Schmellzpunkt 224-227 C erhalten. Nach Umkristallisation aus Acetonitril beträgt der Schmelzpunkt 228-229 C.
Das als Ausgangsstoff verendete 4-Sulfanil amido-5-äthoxy-6-chlor-pyrimidin kann wie folgt erhalten werden :
In 1030 g Dimethylformamid werden 320 g trok- kenes Sulfanilamidnatrium innerhalb einer halben Stunde eingetragen. Das Gemisch wird eine Stunde bei 100 C intensiv gerührt. Dann werden 160 g 4, 6 Dichlor-5-äthoxy-pyrimidin so eingetragen, dass die Temperatur bei 100 C bleibt. Das Reaktionsgemisch wird weitere 3 Stunden bei 100 C gerührt und'dann bei 65 C Badtemperatur vom Dimethylformamid im Vakuum möglichst vollständig befreit. Der Rück- stand wird bei 40 C in 600 ml Wasser gelöst und die Lösung mit etwa 45 ml In Salzsäure auf pH 7, 1 gestellt.
Nach zweistündigem Stehen bei 0-5 C kri stallisiert der tSberschuss an Sulifanilamid aus. Dieser wird abgenutscht, das hellgelbe Filtrat mit 300 ml Athylalkohol versetzt und mit Eisessig auf pH 5 eingelstellt. Man erhält so 217 g 4-Sulfanilamido-5 äthoxy-6-chlor-pyrimidin vom Schmelzpunkb 215 bls, 216 C (aus Dimethylformamid/Wasser).
Beispiel 4
40 g 4-Sulfanilamido-5-äthoxy-6-chlor-pyrimidin werden, wie vorstehend beschrieben, mit 6, 9 g Natrium in 300 ml absolutem Äthanol zum 4-Sulfanil- amido-5, 6-diäthoxy-pyrimidin umgesetzt. Schmelz- punkt : 173-174 C (aus Dimethylformamid/Wasser).
Beispiel S
40 g 4-Sulfanilamido-5-äthoxy-6-chlor-pyrimidin werden, wie vorstehend beschrieben, mit 6, 9 g Natrihum in 300 ml n-Propanol zum 4-Sulfanilamido-5 äthoxy-6-n-propoxy-pyrimidin umgesetzt. Ausbeute : 22 g ; Schmelzpunkt 162 C (aus Butylacetat).
Beispiel 6
Durch Umsetzung von 4-Sulfanilamido¯5-äthoxy- 6-cMor-pyrimidin mit einer Lösung von Natrium in Isopropanol auf die vorstehend beschriebene Weise erhäl, man 33 g rohes 4-SulfaniElamido-5-äthoxy-6- isopropoxy-pyrimidin vom Schmelzpunkt 178 bis 182 C. Nach Umlösen aus Butylacetat beträgt der Schmelzpunkt 181-183 C.

40 g of 4-sulfanilamido-5-ethoxy-6-chloropyrimidine are introduced into a solution of 6.9 g of sodium in 300 ml of methanol. This gives 26 g of 4-sulfanilamido 5-ethoxy-6-methoxy-pyrimidine with a melting point of 224-227 ° C. After recrystallization from acetonitrile, the melting point is 228-229 C.
The 4-sulfanil amido-5-ethoxy-6-chloro-pyrimidine terminated as starting material can be obtained as follows:
320 g of dry sodium sulfanilamide are introduced into 1030 g of dimethylformamide within half an hour. The mixture is stirred intensively at 100 ° C. for one hour. 160 g of 4,6 dichloro-5-ethoxypyrimidine are then added in such a way that the temperature remains at 100.degree. The reaction mixture is stirred for a further 3 hours at 100 ° C. and then freed as completely as possible from dimethylformamide in vacuo at a bath temperature of 65 ° C. The residue is dissolved in 600 ml of water at 40 ° C. and the solution is adjusted to pH 7.1 with about 45 ml of 1 n hydrochloric acid.
After standing for two hours at 0-5 C, the excess sulifanilamide crystallizes out. This is suction filtered, the light yellow filtrate is mixed with 300 ml of ethyl alcohol and adjusted to pH 5 with glacial acetic acid. This gives 217 g of 4-sulfanilamido-5 ethoxy-6-chloro-pyrimidine of melting point 215 to 216 ° C. (from dimethylformamide / water).
Example 4
40 g of 4-sulfanilamido-5-ethoxy-6-chloro-pyrimidine are, as described above, reacted with 6, 9 g of sodium in 300 ml of absolute ethanol to form 4-sulfanil-amido-5, 6-diethoxy-pyrimidine. Melting point: 173-174 C (from dimethylformamide / water).

40 g of 4-sulfanilamido-5-ethoxy-6-chloropyrimidine are, as described above, reacted with 6.9 g of sodium in 300 ml of n-propanol to give 4-sulfanilamido-5-ethoxy-6-n-propoxypyrimidine. Yield: 22 g; Melting point 162 C (from butyl acetate).
Example 6
By reacting 4-sulfanilamido-5-ethoxy-6-cMor-pyrimidine with a solution of sodium in isopropanol in the manner described above, 33 g of crude 4-SulfaniElamido-5-ethoxy-6-isopropoxy-pyrimidine with a melting point of 178 are obtained up to 182 C. After redissolving from butyl acetate, the melting point is 181-183 C.
