Verfahren zur Herstellung von hydrophilen Erzeugnissen von hohem Molekulargewicht aus Dextransubstanzen
Die Erfindung bezieht sich auf ein Verfahren zur Herstellung von hydrophilen Erzeugnissen von hchem Molekulargewicht durch Polymerisation von Dextransubstanzen, wobei Produkte entstehen, die je nach dem Ausmass de. Polymerisation und der Hy drophilität wertvolle Eigenschaften für verschiedene Verwendungszwecke aufweisen.
Das erfindungsgemässe Verfahren besteht darin, eine Hydroxylgruppe enthaltende Dextransubstanz mit einer bifunktionellen organischen Substanz zur Reaktion zu bringen, die Halogenatome und bzw. oder Epoxygruppen, z. B.

enthÏlt, welche mit den Hydroxylgruppen des Dextrans unter Bildung von Bindungen der Athertype reagieren, so ; dass ein Polymerisationsprodukt entsteht, welches mindestens zwei der Moleküle der Dextransubstanz untereinander mit ¯therbr cken gebunden, enthält.
Unter r ?Dextransubstanzen? werden hier Dextran, sowie auch hydrophile, Hydroxylgruppen enthaltende Dextranderivate verstanden. Beispielsweise können als Dextransubstanzen bei der Umse. tzung natürliches Doxtran oder teilweise depolymerisiertes Dextran oder neurale Hydroxylgruppen enthaltende, hydrophile Derivate von Dextran oder teilweise de, polymerisiertes Dextran, wie ¯thyl-, HydroxyÏthyloder 2-Hydroxypropyläther von Dextran oder Dex tranglyzeringlykosid oder Hydrodextran (d.
i. ein Dextran, dessen reduzierende Endgruppen zu Alko- holgruppen reduziert wurden) oder Hydroxylgruppen enthaltende, hydrophile Dextanderivato oder teilweise depolymerisiertes Dextran mit sauren oder basischen Gruppen, wie Carboxyl-, Sulfosäure, Aminooder substituierte Aminogruppen, wie etwa Carboxy- methyldextran oder Dextran verwendet werden, dessen n Endgruppen zu Carboxylgruppen oxydiert sind.
Oft können auch Fraktionen der vorerwähnten Dex transubstanzen vorteilhaft verwendet werden.
Beispiele geeigneter bifunktionoller Substanzen zur Ausführung des Verfahrens können in erster Finie sein : bifunktionelle Glyzerinderivate, wie Epichlor-, Dichlor-, Epibrom-und Dibromhydrin, femer 1,2,3,4-Diepoxybutan, DiepoxypropylÏther, Diepoxy propyläther von. Athylenglykol, Propylenglykol, Poly- äthylenglykolen und dgl. Verbindungen. Allgemein sind aliphatische Epoxyverbindungen mit Kohlenstoff, Wasserstoff, Sauerstoff und ohne dissoziierbare Gruppen für den angegebenen Zweck geeignet.
Die Reaktion wird zweckmässig in Anwesenheit eines Lösungsmittels durchgeführt, in welchem mindestens einer der reagierenden Bestandteile loslich ist, vorzugsweise Wasser, doch können ebenfalls organische Lösungsmittel, wie Dimethylformamid, ben tzt werden. Weiter erwies sich der Zusatz einer alkalisch reagierenden Substanz mit katalysierender Wirkung zweckmässig ; als Beispiele hierfür können Alkalioder Erdalkalimetallhydroxyde erwähnt werden. Andere Katalysatoren sind tertiäre und quaternäre Amine.
Bei der Reaktion werden zwei oder mehrere Moleküle der Dextransubstanzen zu grösseren Molekülen unter Bildung von Atherbrücken der Art -R-O-X-O-R-gebunden, wobei R Reste von Molekülen der Dextransubstanz und X ein Radikal in der Brücke ist, die von der bifunktionellen organi schen Substanz geschaffen wird. Wird ein bifunktionelles Glyzerinderivat, beispielsweise Epichlorhydrin verwendet, entsteht eine Br cke der Art R-O-CH2.
CH(OH)CH2-O-R. Werden, drei Dextranradikale miteinander durch zwei Brucken verbunden, so entspricht das Produkt der allgemeinen Type R-O X-O-R-O-X-O-R, wobei R die Radikale der Dextransubstanz und X das genannte Radikal in der Brücke darstellt.
Je nach den Reaktionsbedingungen, beispiels- weise das Molekulargewicht der als Ausgangsmaterial dienenden Dextransubstanz, sowie die Anteile der Reaktionsbestandteile, können Produkte von hohem Molekulargewicht mit sehr verschiedenen durch- schnittlichen Molekulargewichten und in anderer Be ziehung verschiedenen Eigenschaften erzielt werden.
Beispielsweise ist es möglich, PolymerisationsproF dukte mit hohem Molekulargewicht zu erhalten, die wasserlöslich sind oder auch Produkte in Gelform.
Die auf diese Weise erhaltenen Gele bestehen aus dreidimensionalen makroskopischen Netzwerken von hauptsächlich a-1, 6 glukosidartig gebundenen Ketten von Glukoseresten, die untereinander durch Atherbrücken der vorgenannten Type-R-O-X-O-R-ge, bunden sind, wobei diese Substanzen im Lösungsr mittel unlöslich sind, aber das Lösungsmittel unter Quellen aufnehmen können.
Die Quellfähigkeit kann durch die Wassermenge in g ausgedrückt werden, die von 1 g des trockenen Gels absorbiert wird und die mit Wasserwiederauf nahmes bezeichnet werden kann.
Wenn die anderen Faktoren der Reaktion konstant gehalten werden, ist der Quellfaktor des Gels proportional dem Lösungsmittelgehalt, umgekehrt proportional dem Gehalt an bifunktioneller organischer Substanz und auch umgekehrt proportional dem Molekulargewicht der Dextransubstanz.
Als Beispiel soll angeführt werden, dass Dextran mit einem durchschnittlichen Molekulargewicht von 2000 in 60 /oiger wässeriger Lösung mit 10 /o Epi- chlorhydrin (bezogen auf die Menge an Dextran) zur Reaktion gebracht, kein Gel ergibt, wogegen Dextran mit einemDurchschnittsmolekulargewicht von 40. 000 in 50%iger Lösung durch Reaktion mit 4 /o Epi chlorhyclrin ein Polymerisationsprodukt in Gestalt eines Gels liefert
Wie vorhin angegeben, wird die Reaktion vor- zugsweise in Anwesenheit von Wasser und eines al- kalischen Katalysators durchgeführt, der zweckmässig in Wasser gelöst sein kann.
Allgemein soll jedoch die Wassermenge in der Reaktion so gering als m¯gglich gehalten werden, um unerwünschte Nebenreak- tionen zu vermeiden. Die Reaktion kann entweder ausgeführt werden mit Dextransubstanz in Lösung, welch letztere bei Verwendung von nativem Dextran sehr viskos sein kann, oder es kann die Reaktion mit der Dextransubstanz in Form von in Wasser gequol- lenen Teilchen ausgeführt werden. Bei Verwendung einer wässerigen Lösung soll die Konzentration der Dextransubstanz mindestens 5"/o, vorzugsweise 10 oder mehr Prozent, beispielsweise zwischen 10 und 70 /0 betragen.
Werden Halogenhydrine als bifunk- tionell organische Substanzen benützt, muss die Al- kalimenge zur Neutralisierung des sich bei der Reak- tion abspaltenden Halogenwasserstoffes ausreichen.
Die Reaktionstemperatur kann innerhalb weiter Grenzen und damit natürlich auch die Reaktionszeit verändert werden und diese Zeit ist von der gewählten Temperatur abhängig. Bezüglich der Reaktionsgeschwindigkeit empfiehlt sich ein Arbeiten bei nicht zu tiefer Temperatur. Um jedoch andererseits uner- wünschte Nebenreaktionen zu vermeiden, soll die Temperatur nicht zu hoch sein, ja es kann manchmal notwendig werden zu kühlen. Bei entsprechender Beachtung dieser Verhältnisse wird eine Reaktionstemperatur zwischen etwa 15 C und 60-90 C bevorzugt.
Je nach dem gewünschten durchschnittlichen Molekulargewicht und den sonstigen Eigenschaften des herzustellenden Produktes ist es möglich, die Mengenverhältnisse der Reaktionssubstanzen sowie die anderen Bedingungen derart zu wählen, dass das gewünschte Erzeuniss entsteht, wenn die Reaktion vollstÏndig durchgef hrt wird, oder es kann auch die Reaktion in einer Stufe unterbrochen werden, sobald ein Erzeugnis mit dem gewünschten durchschnittlichen Molekulargewicht entstanden ist.
In gewissen Fällen kann es zweckmässig sein, die Reaktion in zwei oder mehr Stufen, beispielsweise derartdurchzuführen, dass sic in einer Stufe unterbrochen wird, wenn die bifunktionelle Substanz nur teilweise umgesetzt wurde, beispielsweise wenn eine ihrer reaktiven Gruppen reagiert hat und an das werdende Polymer gebunden wurde, während die andere reaktive Gruppe noch nicht zur Umsetzung gelangt ist, worauf die Umwandlung in einer späteren Stufe vervollständigt wird. Es ist auch möglich, derart vor zugehen, dass die Dextransubstanz mit Mengen an bifunktioneller Substanz zur Reaktion gebracht wird, welche stufenweise, nacheinander zugesetzt werden, wobei sich das Durchschnittsmolekulargewicht des Erzeugnisses stufenweise erhöht.
Diese Vorgangs- weise ist besonders dann zweckmässig, wenn das Erzeugniss einer Reinigung unterworfen werden soll, bevor das Molekulargewicht und infolge desselben die Viskosität sehr gross geworden ist, welch letztere die Behandlung des Erzeugnisses erschwert.
Die Reinigung von erfindungsgemäss gewonne- nen Produkten von hohem Molekulargewicht kann, je nachdem die Erzeugnisse löslich oder nicht l¯slich sind, in verschiedenen Weisen ausgeführt werden.
Bei löslichen Erzeugnissen kann die Reinigung so- wohl als Fraktionierung des polydispersen Reaktionsproduktes in der üblichen Weise, etwa durch Fällung mit geeigneten Fällmitteln, wie Alkoholen oder Ketonen, beispielsweise Äthylalkohol, Isopropylalkohol oder Azeton bzw. anderen Substanzen durchgeführt werden, in welchen das Erzeugnis nur schwach lös- lich ist. Andere Verfahren zur Behandlung löslicher Reaktionsprodukte sind Extraktion mit Wasser enthaltenden Nicht-Lösungsmitteln oder Reinigung mit Hilfe von Ionenaustauschern oder Dialyse.
Unlösliche Produkte sollen zuerst zu einer solchen Teilchengrösse gemahlen werden, dass die Reinigung deshalb nicht schwierig wird, weil das Diffu sionsgleichgewicht zu langsam erreicht wird. Zu diesem Zweck werden die Produkte vorzugsweise zu einer Teilchengrösse entsprechend einem Teilchen durchmesser im Gebiet 0, 01-2, 0 mm gemahlen. Dio Reinigung kann alsdann auf einem Filter, in einer Zen, trifuge o. dgl. durch Waschen mit Wasser oder organischen Lösungsmitteln erfolgen. Wesentlich ist, dass etwas Wasser anwesend ist, weil ungequollenes Gel häufig mechanisch eingeschlossene Verunreini gungen enthält, die, wenn das Gel unter der Wirkung von Wasser quillt, aus diesem Gel durch Diffusion austreten können.
Werden bei der Polymerisation Produkte mit zu hohen durchschnittlichen Molekulargewichten erhal- ten, können diese einer teilweisen Depolymerisierung, z. B. durch schwache Hydrolyse, etwa mit Hilfe einer Säure, oder einer Alkoholyse unterworfen und auf diese Weise auf das erwünschte niedrigere durch schnittliche Molekulargewicht gebracht werden. Eine solche teilweise Hydrolysierung kann zweckmässig in 0. 1 normaler HCl-Lösung bei 80 C ausgeführt und unterbrochen werden, wenn die Depolymerisa- tion auf das gewünschte niedrigere durchschnittliche Molekulargewicht erreicht worden ist, wie dies bei der bekannten, teilweisen Depolymerisation von Dextran vor sich geht.
Als Beispiel soll ein teilweises Herabsetzen eines durchschnittlichen Molekularge wichtes von 100. 000 auf ein solches von 80. 000 er wähat werden, Natürlich können sämtliche Produkte von hohem Molekulargewicht sowie auch Gele auf das gewünschte Molekulargewicht durch Teilhydrolysierung gebracht werden.
Die erfindungsgemäss erzielten, hoch molekular- gewichtigen Produkte können, wenn sie löslich sind, beispielsweise durch Bestimmung ihres mittleren MoW lekulargewichtes (Mw) etwa durch Messungen der Lichtzerstreuung, gekennzeichnet werden. Manchmal ist es erforderlich, die Verteilung des Molekulargewichtes festzustellen. Es ist auch möglich, die Grenz viskosität (X7) ausgedrückt durch

festzustellen, wobei W sp die spezifische Viskosität (= relative Viskosität-1) und c die Konzentration des hochmolekulargewichtigen Produktes,, beispielsx weise ausgedrückt in g/100 cm3 L¯sung, ist, oder es kann die Viskosität einer gegebenen Konzentration in bezug auf Wasser festgestellt werden.
Sind die Produkte Gelee, dann können sic durch ihre Quellkraft (Wasserwiederaufnahme) gekennzeichnet werden, die etwa 1-50 g/g des trockenen Produktes ber trägt.
Wie bereits erwähnt, haben dis erfindungsgemäss erzeugten Produkte wertvolle Eigenschaften für ver schiedene Verwendungszwecke, z. B. k¯nnen die lös lichen Polymeren für therapeutische und pharmazeutische Zwecke Verwendung finden, beispielsweise als Plasmaersatz, als Mittel gegen Bildung von sog. nSludge in den Blutgefassen, für Untersuchungen auf verschiedene Durchlässigkeit, z.
B. für Proben der Nierenfunktion, als Mittel zum Wasserhalten, zum Stabilisieren von Suspensionen und zur Rege- lung der Viskosität von pharmazeutischen und cher motechnischen Präparaten. Da beim erfindungsge- mÏssen Verfahren von Dextransubstanzen von aus serordentlich verschiedenen Molekulargewichten, von sehr niedrigen bis zu ausserordentlich hohen Gewich- ten, ausgegangen werden kann, lassen sich Endpro- dukte mit sehr verschiedenen, je dem Anwendungs, gebiet entsprechenden Eigenschaften erzielen.
Da in den Dextransubstanzen die Glykosidbindungen zumeist 1 : 6-Bindungen sind, widerstehen diese der Hy drolyse verhältnismässig gut und werden durch Amylasen nicht zerstört.
Die Gele können als Sprengmittel fUr Tabletten verwendet werden, femer als wasserhaltende Abführ- mittel und als Füllmittel in pharmazeutischen und chemotechnischen Präparaten etc. Sic erwiesen sich besonders wertvoll zur Trennung von Substanzen von verschiedenen molekularen Abmessungen bei dem sogenannten molekularen ?Sieben?. Sie k¯nnen auch als Grundsubstanz für die Herstellung von Ionenaus tauschern dienen. Wenn sie saure oder basische Gruppen, z. B. Carboxylgruppen, SulfosÏuregruppen oder Aminogruppen enthalten, können sie unmittel- bar als sehr brauchbare Ionenaustauscher dienen.
Besonders wertvolle Erzeugnisse werden erhalten, wenn die Polymerisation so weit durchgeführt wird, dass das erhaltene Produkt-etwa nach Fraktionie ; rung und bzw. oder teilweise Depolymerisation-ein durchschnittliches Molekulargewicht innerhalb der Grenze, n von 2000-100. 000. 000, vorzugsweise zwischen 5000 und 10. 000. 000, beispielsweise zwischen 10. 000 und 1. 000. 000 hat.
Insbesondere bei der Herstellung von Produkten für medizinische und pharmazeutische Zwecke, beispielsweise kolloidale Injektionslösungen, kann die Polymerisation so ausgeführt werden, dass ein Pro- dukt mit einem mittleren Molekulargewicht-ge- wünschtenfalls nach Fraktionierung-zwischen 10. 000 und 500. 000, zweckmässig zwischen 20. 000 und 300. 000, beispielsweise zwischen 30. 000 und 200. 000 ensteht.
Je nach dem Verwendungszweck kann das Produkt in grösserem oder geringerem Masse einer Frak tionierung und einer ausreichenden Reinigung unter worfen werden, so dass ein Erzeugnis mit entsprechendem Molekulargewicht und für den beabsichtig- ten Zweck ausreichendem Reinheitsgrad gewonnen wird. Als Beispiel mag ein Produkt mit einem durch schnittlichen Molekulargewicht von ungefähr 70. 000 genannt werden.
Wenn die Produkte für die Zwecke von Injektionen für medizinischen Gebrauch, z. B. für kolloi- daleInjektionslösungen, z. B. als Plasmaersatz verwendet werden sollen, ist es wichtig, dass sie eine ge eignete Verteilung der Molekulargewichte aufweisen, die man, falls erforderlich, beispielsweise durch Fraktionierung erzielen kann, weil Substanzen mit sehr hohem Molekulargewicht unerwünschte Nebenreak- tionen hervorbringen können und weil im Produkt zurückbleibcnde Substanzen von zu niedrigem Mole- kulargewicht vermieden werden sollen, da nach In jektion zu kleine Moleküle aus dem Blut rasch aus- filtriert werden.
Produkte mit geeignetem Molekulargewicht, welche erfindungsgemäss aus kleineren Dextranmole- külen durch Synthese zu grösseren Molekülen erhalten wurden, haben im Vergleich zum üblichen Dextran mit gleichem Molekulargewicht gewisse Vorteile, welche einer anderen Form und geänderten Eigenschaften der Moleküle zuzuschreiben sind. So z.
B. bringen derartige synthetische Produkte eine geringere Agglutinierung der Blutkörperchen hervor und haben einen weniger ausgesprochenen Einfluss auf die Geschwindigkeit der Erythrozyten-Senkung. Im Vergleich zum blichen Dextran hat dieses Produkt auch einen anderen Einfluss auf die Blutkoagulation und veränderte serologischel Eigenschaften. Solche Produkte von ausreichend niedrigem Molekularge- wicht (beispielsweise 30. 000) haben femer die wichtige Eigenschaft, als Mittel gegen Bildung von sog.
Sludgep zu wirken, wenn sie in Blutgefässe injiziert werden. Weiter sind diese Produkte durch ihre geringe Toxizität gekennzeichnet.
Mittels der vorliegenden Erfindung ist es ebenfalls möglich, die wertlose niedermolekularen Fraktionen von Dextran, die als Abfall bei der üblichen Herstellung von Plasmaersatz aus Dextran anfallen, durch Kupplung derselben zu Molekülen der er wünschten Grosse in wertvolle Erzeugnisse umzu- wandeln.
Die Erfindung soU nun anhand der folgenden Beispiele näher beschrieben werden.
Beispiel 1
60 g Dextran vom durchschnittlichen Molekular- gewicht (Mw = 2000) wurden in 40 cm3 einer 5 n NaOH-Lösung gelöst. Nach Zusatz von 6 g Epichlor- hydrin wurde die Lösung 48 Stunden bei Zimmer- temperatur gerührt. Nach Neutralisierung und wie- derholten Umfällungen aus Athanol wurden 38 g eines Produktes von durchschnittlichem Molekular- gewicht von 4900 gewonnen.
Beispiel 2
Einer wässerigen Lösung von Dextran einer Grenzviskosität (n) = 0, 16 (Mw = 25. 000), die 36, 8 g Dextran je l00 cm3 Lösung enthält, wurden 5, 5 g Epichlorhydrin und 10 cm3 einer 33 /0igen Natrium hydroxydlösung zugesetzt. Bei Beginn des Versuches war die Temperatur 60 C, stieg jedoch rasch auf 88 C an. Nach einer Stunde wurde der Versuch durch Kühlung und Neutralisation abgebrochen. Das Produkt wurde durch mehrfache Umfällungen aus wässerigen Lösungen mit Athanol gereinigt. Es hatte eine Grenzviskosität von 0, 35 und ein durchschnittliches Molekulargewicht von 170. 000.
Beispiel 3
700 g Dextran der gleichen Art wie im vorstehen- den Beispiel wurden in Wasser auf eine Konzentra- tion von 34, 5g je lOOcm3 gelöst. Dieser Lösung wurden 80 g Epichlorhydrin und 80 cm3 einer 33 lo- igen Natriumhydroxydlösung zugesetzt. In 5 Min. stieg die Temperatur von 60 auf 91 C und wahrend der folgenden 10Min. sank die Temperatur auf 85 C.
Nach einer Stunde wurde der Versuch durch Abkühlung und Neutralisation unterbrochen, das Produkt durch zweimalige Umfällung aus Athanol gereinigt. Seine Grenzviskosität (4) war 0, 26. Das Produkt wurde durch schrittweisen Zusatz von Athanol fraktioniert, wobei die folgenden Fraktionen erhalten wurden : Fraktion Gewicht in g % (n) Mw

Beispiel 4
500g Dextran (Mw=40. 000) wurden in 1170cm3 einer n-Natriumhydroxydlösung gelöst und 100 g Epichlorhydrin zugef gt. Nach 2 Stunden hatte sich bei 45 C ein Gel gebildet, welches durch 24-stündi- ges Halten auf 45 C gehärtet wurde.
Nach Vermahlen auf eine Teilchengrösse zwischen 50 und 200 Maschen wurde das Gel mit Salzsäure neutralisiert, mit Wasser am Filter bis zur Salzreinheit gewaschen und in einem Ofen bei 80 C getrocknet. 560 g eines Produktes mit einer Wasserwiederaufnahme von 5 g/g des trockenen Produktes wurden gewonnen.
Beispiel 5
500g Dextran (Mw=40. 000) wurden in 2000cm3 einer n-Natriumhydroxydlösung gelöst und 100 g Epichlorhydrin zugef gt. Nach 4 Std. hatte sich bei 45 C ein Gel gebildet, das durch 24-stündiges Erhitzen auf 45 C gehärtet wurde. Nach Mahlen, Waschen und Trocknen ergaben sich 360 g eines Pro- duktes mit Wasserwiederaufnahme von 13 g/g des trockenen Produktes.
Beispiel 6
500g Dextran (M= 40. 000) wurden in 2000cmS einer n-Natnumhydroxydlösung gelöst und 185 g Epichlorhydrin zugefügt. Nach 2 Std. hatto sich bei 45 C ein Gel gebildet, das durch 24-stündiges Erhitzen auf 45 C gehärtet wurde. Nach Mahlen, Neutralisieren, Waschen und Trocknen ergaben sich 574 g eines Produktes mit Wasserwiederaufnahme von 7 g/g des trockenen Produktes.
Beispiel 7 500g Dextran (Mw = 1. 800. 000) wurden in 2000 cm3 einer n-Natriumhydroxydlösung gelöst und 100g Epichlorhydrin zugesetzt. Nach einer Stunde hatte sich bei 45 C ein Gel gebildet, das durch 24 stund. Erhitzen auf 45 C gehartet wurde. Nach Mahlen, Neutralisieren, Waschen und Trocknen ergaben sich 540 g eines Produktes mit Wasserwiederauf- nahme von 10 g/g des trockenen Produktes.
Beispiel 8
100 g Dextran (Mw = 1. 800. 000) wurden in 2000 cm3 einer 0, 5 n-Natriumhydroxydlösung gelöst und 100g Epichlorhydrin zugesetzt. Das Gemisch wurde 8 Std. auf 55 C erhitzt, wobei ein Gel entstand. Nach Mahlen, Neutralisieren, Waschen und Trocknen ergaben sich 60 g eines Produktes mit Wasserwiederaufnahme von 50 g/g des trockenen Produktes.
Beispiel 9
120 g Dextran (M, = 5000) wurden in 80 cm3 einer 5 n-Natriumhydroxydlösung gelöst und 24 g Epichlorhydrin zugesetzt. Nach 24 Std. hatte sich die Masse bei Zimmertemperatur zu einem Gel gefestigt, welches durch 24-st nd. Erhitzen auf 40 C gehärtet wurde. Nach Mahlen, Neutralisieren, Waschen und Trocknen ergaben sich 74 g eines Produktes mit Wasserwiederaufnahme von 9 g/g des trockenen Produktes.
Beispiel 10 1000g Dextran (Mw = 40. 000) wurden, in 1000 cm3 einer 4 n-Natriumhydroxydlösung gelöst und 550 g Epichlorhydrin zugesetzt. As indicated above, the reaction is preferably carried out in the presence of water and an alkaline catalyst, which can conveniently be dissolved in water.
In general, however, the amount of water in the reaction should be kept as low as possible in order to avoid undesired side reactions. The reaction can either be carried out with dextran substance in solution, which latter can be very viscous when using native dextran, or the reaction can be carried out with the dextran substance in the form of particles swollen in water. The reaction temperature can be changed within wide limits and thus of course also the reaction time and this time depends on the selected temperature. The purification of products of high molecular weight obtained according to the invention can be carried out in various ways, depending on whether the products are soluble or insoluble.
