B. 1-Amino-2-mercaptobenzol in Betracht.

Process for the preparation of new tetrahydrothiophene derivatives The present invention relates to a process for the preparation of new, valuable tetrahydrothiophene derivatives which have the formula
EMI0001.0004
correspond, in which A is an optionally further substituted phenylene radical and X is an: oxygen or sulfur atom or for the group
EMI0001.0006
is in which R represents a hydrogen atom or an organic substituent.
The invention accordingly relates to a method for the production of z. B. 2,5-di [-benzoxazolyl- (2 ')], 2,5-di [-benzthiazolyl- (2')] and 2,5-di [-benzimidazyl- (2 ')] tetrahydrothiophene compounds .
According to the invention, the new tetrahydrothiophene derivatives which correspond to the formula (1) given are obtained if o-aminobenzenes of the formula
EMI0001.0014
where A is an optionally substituted phenylene radical and X is an oxygen or sulfur atom or for the group
EMI0001.0015
is in which R is a hydrogen atom or an organic substituent, or its salts at higher temperatures with tetrahydrothiophene-2,5-dicarboxylic acid of the formula
EMI0001.0018
or with functional derivatives of this dicarboxylic acid.
According to a preferred embodiment, the reaction described above is carried out in the presence of catalysts. It is also possible, if necessary, to sulfonate the condensation products obtainable in this way, or in the case of the bisimidazyl-tetrahydrothiophenes
EMI0002.0002
optionally acylating, alkylating, alkenylating, oxyalkylating, aralkylating and / or converting them into salts or into quaternary ammonium compounds.
The two phenylene radicals A in the formula (1) given above can be a halogen atom, especially a chlorine atom, a low molecular weight alkyl or alkoxy group with 1-4 C atoms, especially a methyl or methoxy group, or a substituent, for example Have phenyl radical. These substituents are preferably in the 5- or 6-position of the benzoxazole, benzthiazole or benzimidazole radical.
In the case of the above-mentioned imidazole derivatives which correspond to the formula (1) wherein X is the group
EMI0002.0005
correspond, in which Y is hydrogen, chlorine, a low molecular weight alkyl or alkoxy group with at most 4 carbon atoms or a phenyl radical, and salts of these compounds, for. B. the 2,5 di [-benzimidazyl- (2 ')] - tetrahydrothiophene and the

correspond, where Y is hydrogen, chlorine, a low molecular weight alkyl or alkoxy group with at most 4 carbon atoms or a phenyl radical, z. B. the 2,5-di [-benzoxazolyl- (2 ') - tetrahydrothiophene and the 2,5-di- [benzoxazolyl- (2') tetrahydrothiophene, which each has a methyl group on the two benzene residues.
As starting materials in the preparation of the new tetrahydrothiophene derivatives of the formula (1) are, for. B. in the case of the imidazyl-tetrahydrothiophenverbindun gene o-diamines of the benzene series, whose one amino group is primary and the other amino group is at most secondary, z.
B. 1,2-diaminobenzene, 1- amino-2-monomethylamino-benzene, 1-methyl-3,4-diaminobenzene, 1-methoxy-3,4-diaminobenzene, isopropyl-o-phenylenediamine or 1-chloro 3,4-diamino-benzene. In the case of the oxazole-tetrahydrothiophene derivatives, for example, 1-oxy-2-
EMI0002.0018
is, R represents a hydrogen atom or an organic radical of any kind, z.
B. an acyl radical; in particular, it is an aliphati rule or araliphatic radical and preferably a low molecular weight alkyl, alkenyl or oxyalkyl radical, such as. B. the methyl, ethyl, isopropyl, allyl or oxyethyl radical.
The salts of the new imidazyl-tetrahydrothiophene derivatives can be derived from any inorganic or organic acids, eg. B. from sulfuric acid, hydrochloric or phosphoric acid, also from arylsulfonic acids, z. B. p-toluenesulfonic acid.
Among the imidazole compounds, for example, those of the formula 2,5-di [-benzimidazyl- (2 ') - tetrahydrothiophene, which has a methyl group on each of the two benzene radicals, are particularly valuable, and among the oxazole compounds, those of the formula aminobenzene and 1-oxy-2-amino-methylbenzenes such as 1-oxy-2-amino-4- or -5-methylbenzene, also 1-amino-2-oxy-5-methoxy-benzene, 1-amino-2- oxy-5-tertiary-butylbenzene and 1-amino-2-oxy-5-chlorobenzene, and in the case of the thiazole-tetrahydrothiophene derivatives, e.g. B. 1-amino-2-mercaptobenzene into consideration.
As further starting materials for the present process, tetrahydrothiophene-2,5-dicarboxylic acid or functional derivatives of this dicarboxylic acid, eg. B. esters or acid dihalides are used. Both the individual stereoisomeric forms and mixtures of different stereosomeric forms of tetrahydrothiophene-2,5-dicarboxylic acid or the functional derivatives can be used for the implementation.
Among the esters of this dicarboxylic acid, those with low molecular weight aliphatic alcohols are particularly suitable, for example the dimethyl ester or diethyl ester. The respective two starting materials, namely the aminobenzenes of the formula (2) given above and the tetrahydrothiophene-2,5-dicarboxylic acid or the functional derivatives of this dicarboxylic acid, are at least approximately in the theoretically correct quantitative ratio with one another for the reaction brought, d. H.
to 1 mole of dicarboxylic acid, 2 moles of an aminobenzene of the formula (2) or an amount which deviates only slightly therefrom are used. The implementation between the respective two components takes place by heating to higher temperatures, for example to about 90 to 260, optionally in an inert gas, eg. B. in a nitrogen stream. The reaction is preferably carried out in the presence of a catalyst. Suitable catalysts are, for. B. boric acid, sulfonic acid of the benzene series, such as p-toluenesulfonic acid, also polyphosphoric acids including pyrophosphoric acid and zinc chloride. The reaction is advantageously conducted in such a way that compounds of the formula (1) are formed immediately, d. H.
that the acylation of the o-amino compounds and the ring closure to the azole ring in question take place in one operation.
If boric acid is used as the catalyst, it is advantageously used in an amount of about 0.5 to 5%, based on the total weight of the reaction mass.
When using boric acid or, for example, p-toluenesulfonic acid, it is advantageous to use an inert high-boiling solvent, e.g. B. substitution products of benzene, such as monochlorobenzene, o-dichlorobenzene, trichlorobenzene, or higher-boiling hydrocarbons of the benzene series such as toluene, xylenes or p-cymene. It is expedient to work at boiling point in one of the solvents mentioned, to separate the water of reaction from the distilled solvent with the aid of a water separator and to return the dehydrated solvent to the reaction mixture.
The tetrahydrothiophene derivatives of the formula (1) can also be obtained by simply melting the two starting materials together in a zinc chloride melt or in the presence of boric acid.
The reaction of the o-diaminobenzenes with the tetrahydrothiophene-2,5-dicarboxylic acid or the functional derivative of this dicarboxylic acid to imidazole tetrahydrothiophene compounds of the formula (1) can advantageously also be achieved by heating the two starting materials in aqueous mineral acids such as hydrochloric acid or Phosphoric acid, and especially in aqueous sulfuric acid, to higher temperatures, e.g. B. to about 90 to 110 ° C.
The imidazole tetrahydrothiophene derivatives of the formula (1) which can be obtained according to the invention, in which X is the group
EMI0003.0025
represents, can optionally be acylated, alkylated, alkenylated, oxyalkylated or aralkylated. The acylation can be carried out by methods known per se, eg. B. by treating the latter tetrahydrothiophene- derivatives with carboxylic acid chlorides, such as acetyl or benzoyl chloride, in an anhydrous inert organic solvent. The alkylation, alkenylation, oxyalkylation or aralkylation can also be carried out in a conventional manner, for.
B. by treatment with alkyl halides such as ethyl chloride, alkenyl halides such as allyl bromide, alkylene halohydrins, for example ethylene or propylene chlorohydrin, or, with aralkyl halides such as benzyl chloride, expediently with the addition of acid-binding agents. Dialkyl sulfates, such as dimethyl sulfate, can also be used for the alkylation.
To the quaternary ammonium salts of the imidazole tetrahydrothiophene derivatives of the formula (1), one reaches z. B. if the imidazole tetrahydrothiophene derivatives or the salts of these derivatives are reacted with compounds that are able to quaternize an imidazole nitrogen atom. The usual alkylating and aralkylating agents are primarily suitable as such compounds. Examples include: methyl iodide, ethyl bromide, isopropyl chloride, sec. Butyl bromide, dodecyl bromide, epichlorohydrin, dimethyl sulfate, benzyl halides and in the core z.
B. substituted by halogen, alkyl or alkoxy groups benzyl halides, such as benzyl chloride, p-chlorobenzyl chloride, also menaphthyl chloride, toluenesulfonic acid ester, arylsulfonic acid alkyl ester, especially those of the benzene series with low molecular weight alkyl groups, such as toluenesulfonic acid ethyl or methyl ester, further compounds that one Contain water-solubilizing group, such as haloalkyl or aralkylsulphonic acids, e.g. B.
Bromoethanesulfonic acid, benzylchloride disulfonic acid. If salts of the diimidazoles are used as the starting point, alkylene oxides such as ethylene oxide, propylene oxide or glycide can also be used for quaternization, or finally low molecular weight aliphatic alcohols such as methanol or ethanol. The quaternization can be carried out under the usual conditions for such a reaction, expediently by heating the components th to: higher temperature, e.g. B. 50 to 200, if necessary under pressure. The use of inert solvents such as alcohols, e.g. B.
Methanol, ethyl alcohol, dioxane, glycol, monomethylglycol, benzene, toluene, nitrobenzene or mixtures of such solvents can be advantageous. If necessary, it can be beneficial to use an excess of quaternization agent.
To sulfonate the tetrahydrothiophene derivatives, the compounds of the formula (1) can be used in a conventional manner, for. B. with sulfuric acid or sulfuric acid containing sulfur trioxide, if necessary in the heat treated.
The new, according to the invention obtainable tetra hydrothiophene derivatives of the composition described above can be used as intermediates, eg. B. for the production of textile auxiliaries or dyes.
